Abstract:Lignin, a major component of lignocellulosic biomass, is crucial to plant growth and development but is a major impediment to efficient biomass utilization in various processes. Valorizing lignin is increasingly realized as being essential. However, rapid condensation of lignin during acidic extraction leads to the formation of recalcitrant condensed units that, along with similar units and structural heterogeneity in native lignin, drastically limits product yield and selectivity. Catechyl lignin (C-lignin), which is essentially a benzodioxane homopolymer without condensed units, might represent an ideal lignin for valorization, as it circumvents these issues. We discovered that C-lignin is highly acid-resistant. Hydrogenolysis of C-lignin resulted in the cleavage of all benzodioxane structures to produce catechyl-type monomers in near-quantitative yield with a selectivity of 90% to a single monomer.
Purity and structural composition of lignin isolated from Miscanthus x giganteus by sub-critical water extraction with associated modifiers

Abstract:Lignin is an industrial by-product produced from the pulping and paper industry, where the process generates lignin in the form of lignosulphonates. While there are many applications for lignin, there are all low value and attempts to add value to lignin are hindered by its complex physicochemical nature and the presence of sulphur. Adopting the biorefining concept, the study evaluates the impact of direct (DE) and sequential extraction (SE) of Miscanthus x giganteus using sub-critical water with associated modifiers; ethanol and carbon dioxide on the physical and chemical properties of the extracted lignin. Isolated lignins were characterised by a Fourier Transform Infrared Spectroscopy (FTIR). Although higher delignification was achieved by DE and SE about 81.5% and 58.0%, respectively, the lignin recovered from the SE process showed remarkably higher purity with 91.5%. Lignin recovery did not differ considerably for either processing method. FTIR revealed a qualitative reduction in the intensity of bonds corresponding to hydroxyl groups for the lignin derived from DE rather than SE processing routes. These indicated that the lignin derived from SE had potential for subsequent preparation in lignin value-added bio-based materials.
Assessment of Antioxidant and Antimicrobial Properties of Lignin from Corn Stover Residue Pretreated with Low-Moisture Anhydrous Ammonia and Enzymatic Hydrolysis Process.

Abstract:Lignin accounts for 15–35% of dry biomass materials. Therefore, developing value-added co-products from lignin residues is increasingly important to improve the economic viability of biofuel production from biomass resources. The main objective of this work was to study the lignin extracts from corn stover residue obtained from a new and improved process for bioethanol production. Extraction conditions that favored high lignin yield were optimized, and antioxidant and antimicrobial activities of the resulting lignin were investigated. Potential estrogenic toxicity of lignin extracts was also evaluated. The corn stover was pretreated by low-moisture anhydrous ammonia (LMAA) and then subjected to enzymatic hydrolysis using cellulase and hemicellulase. The residues were then added with sodium hydroxide and extracted for different temperatures and times for enhancing lignin yield and the bioactivities. The optimal extraction conditions using 4% (w/v) sodium hydroxide were determined to be 50 °C, 120 min, and 1:8 (w:v), the ratio between corn stover solids and extracting liquid. Under the optimal condition, 33.92 g of lignin yield per 100 g of corn stover residue was obtained. Furthermore, the extracts produced using these conditions showed the highest antioxidant activity by the hydrophilic oxygen radical absorbance capacity (ORAC) assay. The extracts also displayed significant antimicrobial activities against Listeria innocua. Minimal estrogenic impacts were observed for all lignin extracts when tested using the MCF-7 cell proliferation assay. Thus, the lignin extracts could be used for antioxidant and antimicrobial applications, and improve the value of the co-products from the biomass-based biorefinery.

Abstract:The efficient utilization of crop waste is limited because of plant cell wall recalcitrance. Solving this problem through sustainable processing while producing value-added products is crucial. Here we report a rapid lignocellulose fractionation method applied to common reed using milling under mild alkaline conditions without involving any organic solvents or other reagents. Lignin-containing cellulose nanofiber (LCNF) and native-like lignins were obtained in high yield. LCNF films prepared from the LCNF suspension could have their properties, such as transparency and hydrophobicity, adjusted by changing the pretreatment conditions. Overall this biomass fractionation provides value-added products and is an alternative to simply burning the crop waste.
Overcoming Cellulose Recalcitrance in Woody Biomass for the Lignin-First Biorefinery.

Abstract:Low-temperature swelling of cotton linter cellulose and subsequent gelatinization in trifluoroacetic acid (TFA) greatly enhance rates of enzymatic digestion or maleic acid–AlCl3 catalyzed conversion to hydroxymethylfurfural (HMF) and levulinic acid (LA). However, lignin inhibits low-temperature swelling of TFA-treated intact wood particles from hybrid poplar (Populus tremula × P. alba) and results in greatly reduced yields of glucose or catalytic conversion compared to lignin-free cellulose. Previous studies have established that wood particles from transgenic lines of hybrid poplar with high syringyl (S) lignin content give greater glucose yields following enzymatic digestion. Low-temperature (− 20 °C) treatment of S-lignin-rich poplar wood particles in TFA slightly increased yields of glucose from enzymatic digestions and HMF and LA from maleic acid–AlCl3 catalysis. Subsequent gelatinization at 55 °C resulted in over 80% digestion of cellulose in only 3 to 6 h with high-S-lignin wood, compared to 20–60% digestion in the wild-type poplar hybrid and transgenic lines high in guaiacyl lignin or 5-hydroxy-G lignin. Disassembly of lignin in woody particles by Ni/C catalytic systems improved yields of glucose by enzymatic digestion or catalytic conversion to HMF and LA. Although lignin was completely removed by Ni/C-catalyzed delignification (CDL) treatment, recalcitrance to enzymatic digestion of cellulose from the high-S lines was reduced compared to other lignin variants. However, cellulose still exhibited considerable recalcitrance to complete enzymatic digestion or catalytic conversion after complete delignification. Low-temperature swelling of the CDL-treated wood particles in TFA resulted in nearly complete enzymatic hydrolysis, regardless of original lignin composition. Genetic modification of lignin composition can enhance the portfolio of aromatic products obtained from lignocellulosic biomass while promoting disassembly into biofuel and bioproduct substrates. CDL enhances rates of enzymatic digestion and chemical conversion, but cellulose remains intrinsically recalcitrant. Cold TFA is sufficient to overcome this recalcitrance after CDL treatment. Our results inform a ‘no carbon left behind’ strategy to convert total woody biomass into lignin, cellulose, and hemicellulose value streams for the future biorefinery.
Natural Variation of Lignocellulosic Components in Miscanthus Biomass in China

Abstract:Lignocellulose content is an important factor affecting the conversion efficiency of biomass energy plants. In this study, 179 Miscanthus accessions in China were used to determine the content of lignocellulose components in stems via acid hydrolysis and high-performance liquid chromatography. Results showed that the average lignocellulose content of wild Miscanthus germplasm resources was 80.27 ± 6.51%, and the average content of cellulose, hemicellulose, lignin, extracts, and total ash was 38.38 ± 3.52, 24.23 ± 4.21, 17.66 ± 1.56, 14.50 ± 5.60, and 2.53 ± 0.59%, respectively. The average lignocellulose content of M. sinensis, M. floridulus, M. nudipes, M. sacchariflorus, M. lutarioriparius, and the hybrids was 77.94 ± 6.06, 75.16 ± 4.98, 75.68 ± 3.02, 83.71 ± 4.78, 81.50 ± 5.23, and 74.72 ± 7.13%, respectively. In all the tested materials, the highest cellulose content was 48.52%, and the lowest was 29.79%. Hemicellulose had the maximum content of 34.23% and a minimum content of 15.71%. The highest lignin content was 23.75%, and the lowest was 13.01%. The lignocellulosic components of different ploidy materials were compared. The content of lignocellulosic components of diploid M. sacchariflorus was higher than that of tetraploid M. sacchariflorus, and the content of lignocellulosic components of diploid M. lutarioriparius was lower than that of tetraploid M. lutarioriparius. Analysis of the relationship between the changes in lignocellulosic components and geographical locations of Miscanthus showed that the holocellulose and hemicellulose content was significantly positive correlated with the latitude of the original growth location. Results indicated that the lignocellulosic components of Miscanthus resources in China are rich in genetic diversity.

Abstract:Background: Miscanthus × giganteus is widely recognized as a promising lignocellulosic biomass crop due to its advantages of high biomass production, low environmental impacts, and the potential to be cultivated on marginal land. However, the high costs of bioethanol production still limit the current commercialization of lignocellulosic bioethanol. The lignin in the cell wall and its by-products released in the pretreatment step is the main component inhibiting the enzymatic reactions in the saccharification and fermentation processes. Hence, genetic modification of the genes involved in lignin biosynthesis could be a feasible strategy to overcome this barrier by manipulating the lignin content and composition of M. × giganteus. For this purpose, the essential knowledge of these genes and understanding the underlying regulatory mechanisms in M. × giganteus is required. Results: In this study, MgPAL1, MgPAL5, Mg4CL1, Mg4CL3, MgHCT1, MgHCT2, MgC3'H1, MgCCoAOMT1, MgCCoAOMT3, MgCCR1, MgCCR2, MgF5H, MgCOMT, and MgCAD were identified as the major monolignol biosynthetic genes in M. × giganteus based on genetic and transcriptional evidence. Among them, 12 genes were cloned and sequenced. By combining transcription factor binding site prediction and expression correlation analysis, MYB46, MYB61, MYB63, WRKY24, WRKY35, WRKY12, ERF021, ERF058, and ERF017 were inferred to regulate the expression of these genes directly. On the basis of these results, an integrated model was summarized to depict the monolignol biosynthesis pathway and the underlying regulatory mechanism in M. × giganteus. Conclusions: This study provides a list of potential gene targets for genetic improvement of lignocellulosic biomass quality of M. × giganteus, and reveals the genetic, transcriptional, and regulatory landscape of the monolignol biosynthesis pathway in M. × giganteus.

Abstract:Cell wall recalcitrance is a major constraint for the exploitation of lignocellulosic biomass as a renewable resource for energy and bio-based products. Transcriptional regulators of the lignin biosynthetic pathway represent promising targets for tailoring lignin content and composition in plant secondary cell walls. However, knowledge about the transcriptional regulation of lignin biosynthesis in lignocellulosic feedstocks, such as Miscanthus, is limited. In Miscanthus leaves, MsSCM1 and MsMYB103 are expressed at growth stages associated with lignification. The ectopic expression of MsSCM1 and MsMYB103 in N. benthamiana leaves was sufficient to trigger secondary cell wall deposition with distinct sugar and lignin compositions. Moreover, RNA-seq analysis revealed that the transcriptional responses to MsSCM1 and MsMYB103 overexpression showed an extensive overlap with the response to the NAC master transcription factor MsSND1, but were distinct from each other, underscoring the inherent complexity of secondary cell wall formation. Furthermore, conserved and previously described promoter elements as well as novel and specific motifs could be identified from the target genes of the three transcription factors. Together, MsSCM1 and MsMYB103 represent interesting targets for manipulations of lignin content and composition in Miscanthus towards a tailored biomass.

Comparison of Methodologies Used to Determine Monolignol Ratios in Lignocellulosic Biomass

Abstract:Abstract Background Multiple analytical methods have been developed to determine the ratios of monolignol monomers, particularly the syringyl/guaiacyl (S/G) ratio, of lignin biopolymers in plant cell walls. Chemical degradation methods yield monomers that are either selective of certain linkages, such as thioacidolysis, or induce chemical changes rendering it impossible to distinguish and determine the source of specific monomers, such as nitrobenzene oxidation. NMR methods provide powerful tools used to analyze cell walls for lignin monomeric composition and linkage information. Pyrolysis-mass spectrometry methods are also widely used, particularly as a high-throughput method. However, the different techniques used to analyze lignin monolignol ratios frequently yield different results within particular studies, making it difficult to interpret and compare results, and to obtain meaningful insights relating these measurements to other characteristics of plant cell walls that may impact biomass sustainability and conversion metrics for the production of bio-derived fuels and chemicals. Results The authors compared the S/G monolignol ratios of pine, several genotypes of poplar, and corn stover biomass obtained from thioacidolysis, pyrolysis-molecular beam mass spectrometry (py-MBMS), HSQC liquid-state NMR and solid-state (ss) NMR methodologies. An underutilized approach to deconvolute ssNMR spectra was implemented to derive S/G ratios. The S/G ratios obtained for the samples did not agree across the different methods, but trends were similar with the most agreement among the py-MBMS, HSQC NMR and deconvoluted ssNMR methods. The relationship between monolignol S/G, thioacidolysis yields, and linkage analysis determined by HSQC is also addressed. Conclusions This work demonstrates that different methods using chemical, thermal, and nondestructive NMR techniques to determine native monolignol S/G ratios in plant cell walls may yield different results depending on species and linkage abundances. Spectral deconvolution likely applies well to many hardwoods that are S and G dominant, but results may not be reliable for some woody and grassy species for which the lignin composition is more diverse. HSQC may be a better method for analyzing lignin in those species given the wealth of information provided on additional aromatic moieties and bond linkages. Careful consideration is required when choosing a method to measure S/G ratios and the benefits and shortcomings of each method discussed here are summarized.

Abstract:Bioenergy crops provide enormous renewable biomass resources convertible for biofuel production, but lignocellulose recalcitrance fundamentally determines its enzymatic saccharification at high cost and low efficiency. In this study, total 30 diverse Miscanthus lignocellulose substrates were incubated with T. reesei strain to secret lignocellulose-degradation enzymes, and their major wall polymers features (cellulose crystallinity, hemicellulose arabinose and lignin H-monomer) were meanwhile examined with distinct impacts on the enzyme activities. Using characteristic Miscanthus (Msi62) de-lignin residue as inducing substrate with the reesei strain, this study detected that the Msi62-induced enzymes were of consistently higher enhancements on enzymatic saccharification of various lignocellulose residues examined in 17 grassy and woody bioenergy crops, particularly for the hemicellulose hydrolyses, compared to other two reesei-secreted cellulases and three commercial enzymes. Notably, based on SDS-gel protein separation profiling and LC-MS/MS analysis, the Msi62-induced enzymes consist of distinct cellulases (CBHI, BG, EGII) compositions and high-activity xylanases. Therefore, this study has demonstrated an applicable approach to achieve the optimal cellulases and xylanases cocktails that enable for low-costly and high-efficient enzymatic saccharification of diverse lignocellulose sources, providing a potential strategy for large-scale biofuel production in all major bioenergy crops.

Abstract:To reduce the C footprint of human activities, there is a growing need for alternative energy sources including the production of bioenergy feedstocks. Miscanthus × giganteus is a high yielding grass with low environmental impact and high potential for feedstock use. Studying the composition of the aboveground tissues of Miscanthus is important for understanding feedstock quality for biofuel conversion and how crop residue quality may affect soil input management. Data on Miscanthus leaf and stem chemistry including carbon (C), nitrogen (N), macronutrient concentrations, and the optical characteristics of the water extractable organic matter (WEOM) was analyzed to identify differences in composition between aboveground tissues and modeled to identify soil variables that may be correlated with tissue chemistry. Leaves and stems were dominated by N, potassium (K), calcium (Ca), phosphorus (P), and magnesium (Mg), but overall, the leaves contained higher nutrient concentrations compared to the stems. The leaves displayed elevated Si:K (0.0935) and Ca:K (0.445) ratios and lower C:N (36) and C:P (323) ratios compared to the stems (0.0560, 0.145, 150, and 645, respectively). Leaf WEOM contained large, aromatic, and complex structures, while the stem WEOM was dominated by small, recently produced structures. Varying relationships were found between tissue C and the mobile C pool in surface (0–15 cm) and deep (45–60 cm) soils. Overall, Miscanthus leaves had a chemical composition indicative of reduced biofuel quality compared to the stems. The relationships with soil mobile C suggest a dynamic linkage between Miscanthus physiology and this active soil C pool. These results have implications for crop nutrient allocation and nutrient management practices.

Abstract:Forests being mostly non-renewable, miscanthus seems to be a promising alternative energy resource. This study investigates the enzymatic hydrolysis of pretreated miscanthus biomass. The research featured Miscanthus sinensis Strictus biomass. The methods included raw material pretreatment, enzymatic hydrolysis, matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF) of monosaccharide residues, and nuclear magnetic resonance (1H NMR) spectrometry of non-hydrolyzed lignin. A hammer mill with a particle size of 0.2–0.3 mm proved to be effective for preliminary mechanical processing of miscanthus. This type of treatment provided the maximal yield of carbohydrate-containing biomass (62.33 ± 1.87%). The optimal multi-enzyme composition included Cellulase Ultra obtained from Trichoderma reeseii, xylanase obtained from Thermomyces lanuginosus, and β-gluconase obtained from Myceliophtorafergusii. The fermentation temperature was 50 ± 1°C (72 ± 1 h). At these parameters, the conversion of holocellulose of miscanthus biomass was 96.0 ± 4.8%, and the yield of reducing substances was 97.00 ± 4.85%. The article introduces the optimal enzymatic hydrolysis parameters for pretreating Miscanthus sinensis Strictus biomass as a source of carbohydrate-containing substrates and describes their subsequent use for bacterial cellulose biosynthesis. The carbohydrate composition of hydrolysates included residual lignin, such polysaccharides as glucan and xylan, and various monosaccharides, namely arabinose, xylose, galactose, glucose, and uronic acids. Hydrolyzed miscanthus proved to be an effective renewable and environmentally friendly biodiesel.

Abstract:The chemical complexity of lignin remains a major challenge for lignin valorization into commodity and fine chemicals. A knowledge of the lignin features that favor its valorization and which plants produce such lignins can be used in plant selection or to engineer them to produce lignins that are more ideally suited for conversion. Sixteen biomass samples were compositionally surveyed by NMR and analytical degradative methods, and the yields of phenolic monomers following hydrogenolytic depolymerization were assessed to elucidate the key determinants controlling the depolymerization. Hardwoods, including those incorporating monolignol p-hydroxybenzoates into their syringyl/guaiacyl copolymeric lignins, produced high monomer yields by hydrogenolysis, whereas grasses incorporating monolignol p-coumarates and ferulates gave lower yields, on a lignin basis. Softwoods, with their more condensed guaiacyl lignins, gave the lowest yields. Lignins with a high syringyl unit content released elevated monomer levels, with a high-syringyl polar transgenic being particularly striking. Herein, we distinguish phenolic monomers resulting from the core lignin vs those from pendent phenolate esters associated with the biomass cell wall, acylating either polysaccharides or lignins. The basis for these observations is rationalized as a means to select or engineer biomass for optimal conversion to worthy phenolic monomers.

Abstract:Both annual (cotton, flax, hemp, etc.) and perennial (trees and grasses) plants can serve as a source of cellulose for fiber production. In recent years, the perennial herbaceous plant miscanthus has attracted particular interest as a popular industrial plant with enormous potential. This industrial crop, which contains up to 57% cellulose, serves as a raw material in the chemical and biotechnology sectors. This study proposes for the first time the utilization of miscanthus, namely Miscanthus Giganteus "KAMIS", to generate spinning solutions in N-methylmorpholine-N-oxide. Miscanthus cellulose's properties were identified using standard methods for determining the constituent composition, including also IR and atomic emission spectroscopy. The dry-jet wet method was used to make fibers from cellulose solutions with an appropriate viscosity/elasticity ratio. The structural characteristics of the fibers were studied using IR and scanning electron microscopy, as well as via X-ray structural analysis. The mechanical and thermal properties of the novel type of hydrated cellulose fibers demonstrated the possibility of producing high-quality fibers from miscanthus.

Abstract:The correlation between lignin structure, its properties, and performance is crucial for lignin engineering in high-value products. Currently, a widespread approach is to compare lignins which differ by more than one parameter (i.e., Kraft vs organosolv vs lignosulfonates) in various applications by attributing the changes in their properties/performance specifically to a certain variable (i.e., phenolic -OH groups). Herein, we suggest a novel approach to overcome this issue by changing only one variable at a time while keeping all others constant before investigating the lignin properties/performance. Indulin AT (Ind-AT), a softwood Kraft lignin, was chosen as the model substrate for this study. Selective (analytical) lignin modifications were used to mask/convert specific functionalities, such as aliphatic (AliphOH) including benzylic -OH (BenzOH) and phenolic -OH (PhOH) groups, carboxyl groups (-COOH) and carbonyl groups (CO) via methylation, acetylation, and reduction. The selectivity and completeness of the reactions were verified by comprehensive NMR analysis (31P and 2D HSQC) of the modified preparations together with state-of-the-art molar mass (MM) characterization. Methylene blue (MB) adsorption, antioxidant activity, and glass transition temperature (Tg) were used to demonstrate and compare the properties/performance of the obtained modified lignins. We found that the contribution of different functionalities in the adsorption of MB follows the trend BenzOH > -COOH > AlipOH > PhOH. Noteworthy, benzylic -OH contributes ca. 3 and 2.3 times more than phenolic and aliphatic -OH, respectively. An 11% and 17% increase of Tg was observed with respect to the unmodified Indulin by methylating benzylic -OH groups and through reduction, respectively, while full acetylation/methylation of aliphatic and phenolic -OH groups resulted in lower Tg. nRSI experiments revealed that phenolic -OH play a crucial role in increasing the antioxidant activity of lignin, while both aliphatic -OH groups and -COOHs possess a detrimental effect, most likely due to H-bonding. Overall, for the first time, we provide here a reliable approach for the engineering of lignin-based products in high value applications by disclosing the role of specific lignin functionalities.

Abstract:Miscanthus is a promising lignocellulosic bioenergy crop for bioethanol production. To identify candidate genes and regulation networks involved in secondary cell wall (SCW) development in Miscanthus , we performed de novo transcriptome analysis of a developing internode. According to the histological and in-situ histochemical analysis, an elongating internode of M. lutarioriparius can be divided into three distinct segments, the upper internode (UI), middle internode (MI) and basal internode (BI), each representing a different stage of SCW development. The transcriptome analysis generated approximately 300 million clean reads, which were de novo assembled into 79,705 unigenes. Nearly 65% of unigenes was annotated in seven public databases. Comparative profiling among the UI, MI and BI revealed four distinct clusters. Moreover, detailed expression profiling was analyzed for gene families and transcription factors (TFs) involved in SCW biosynthesis, assembly and modification. Based on the co-expression patterns, putative regulatory networks between TFs and SCW-associated genes were constructed. The work provided the first transcriptome analysis of SCW development in M. lutarioriparius . The results obtained provide novel insights into the biosynthesis and regulation of SCW in Miscanthus . In addition, the genes identified represent good candidates for further functional studies to unravel their roles in SCW biosynthesis and modification.
Extraction Of Lignin From Wood Biomass Using A New Ionic Liquid At Open Atmosphere

Abstract:Using wood biomass as opposed to petroleum/fossil feedstock to synthesize chemicals and monomers offers the value proposition of a reduced carbon foot print. A critical step in biomass utilization is to obtain the fermentable monomeric pentose and hexose sugar subunits from various lig-nocellulosic polymers. However, lignin is a three-dimensional amorphous polymer based on methoxylated phenylpropanoid units, and is crosslinks to different polysaccharides with covalent bond. The presence of lignin is a barrier to enzymatic hydrosis of cellulose by cellulases, as well as inhibiting fermentation to form products such as ethanol. Usually, there are three approaches to fractionate biomass into useful chemicals. The first one is extract lignin at high temperature and pressure. The second approach involves more severe conditions such as the temperatures and pressures used in gasification, or use the mineral acids at elevated temperatures to liberate sugars for fermentation. The third way is dissolution of biomass using ionic liquid to extract lignin, and then re-precipitation the cellulosic residue. In this paper, we investigate the lignin extraction uses a purpose designed ionic liquid (IL) 1 -butyl-3-methylimidazolium acesulfamate [BMIM]Ace at room temperature and open atmosphere. It was synthesized from the sugar acesulfamate potassium (Ace K), and with good property of greater thermal stability, increased viscosities, and inability to dissolve crystalline cellulose. In addition, it is of low cost and nontoxic, and is expected to interact with lignin owing to its aromatic character of anion. Organic solvents were firstly used in the extraction process, which reduce the viscosity of IL and increase the lignin yield. The lignin can be obtained by using acetone as the reconstitution solvent (IL/acetone 1:3, v/v), and approximately 11.5% and 96.5% of lignin and the reconstitution cellulose-rich material have been achieved in one dissolution/reconstitution cycle by fresh IL, respectively. It was found that the presence of some organic solvents mix with IL can increase the extraction efficiency. The regenerated ionic liquid showed good retention of structure and properties. The regenerated lignin fractions exhibited large molecular weights (M-w, 3700-7810 g mol(-1)). The physicochemical properties of the extracted lignin fractions were characterized by Fourier transform infrared (FT-IR), and high-performance anion exchange chromatography (HPAEC). The reconstitution cellulose-rich material can be serving as a valuable source for enzymatic saccharification for next step. These results indicate a potential approach in further optimization of the choice of low-cost Its combined with organic solvents for the efficient utilization of the wood biomass.
Extraction and Isolation of Different Lignins from Sugarcane Bagasse Biomass and Its Subsequent Thermal Degradation to Renewable Fuels

Abstract:The present work highlights the extraction and isolation of different lignins from hardwood biomass and their use as a sustainable feedstock to produce bio-based fuels. Four different kinds of lignins were extracted using different strategies namely Kraft, Soda Anthraquinone, Formic Acid and Acetic Acid and were isolated under desired operating conditions. Further, thermal pyrolysis was performed on all the extracted lignins, and a detailed investigation was done under different operating conditions. Structural characterization was explored using Fourier-transform infrared spectroscopy and 13 C Nuclear Magnetic Resonance ( 13 C NMR) which reveals the structural features of lignins. Maximum liquid yield was obtained at 700 °C for all the extracted lignins, i.e. 53.98% for Kraft lignin; 55.17% for Soda Anthraquinone; 35.69% for Acetic acid and 27.49% for Formic acid lignin. Moreover, phenol alkoxy ketones such as vanillin and dimethoxy-hydroxy benzaldehyde were found at lower temperatures (< 450 °C). At higher temperatures, phenol content in Kraft and Soda Anthraquinone lignin was higher compared to phenol alkoxy ketones. Further, catalytic thermal degradation with the incorporation of Cerium over Al 2 O 3 enhances the product yield. Maximum yield was found to be with Ce/Al 2 O 3 :15 catalyst, i.e. 69.71% (liquid yield), 27.74% (gaseous yield) using Soda AQ lignin. The proposed study resolves direct utilization of lignin and widens the correlation between thermal decomposition and its chemical structure.

Abstract:Lignin utilization in value-added co-products is an important component of enabling cellulosic biorefinery economics. However, aqueous dilute acid pretreatments yield lignins with limited applications due to significant modification during pretreatment, low solubility in many solvents, and high content of impurities (ash, insoluble polysaccharides). This work addresses these challenges and investigates the extraction and recovery of lignins from lignin-rich insoluble residue following dilute acid pretreatment and enzymatic hydrolysis of corn stover using three extraction approaches: ethanol organosolv, NaOH, and an ionic liquid. The recovered lignins exhibited recovery yields ranging from 30% for the ionic liquid, 44% for the most severe acid ethanol organosolv condition tested, and up to 86% for the most severe NaOH extraction condition. Finally, the fractional solubilities of different recovered lignins were assessed in a range of solvents and these solubilities were used to estimate distributions of Hildebrand and Hansen solubility parameters using a novel approach.

Abstract:Lignocellulose is mainly composed of hydrophobic lignin and hydrophilic polysaccharide polymers, contributing to an indispensable carbon resource for green biorefineries 1,2 . When chemically treated, lignin is compromised owing to detrimental intra- and intermolecular crosslinking that hampers downstream process 3,4 . The current valorization paradigms aim to avoid the formation of new C–C bonds, referred to as condensation, by blocking or stabilizing the vulnerable moieties of lignin 5,6,7 . Although there have been efforts to enhance biomass utilization through the incorporation of phenolic additives 8,9 , exploiting lignin's proclivity towards condensation remains unproven for valorizing both lignin and carbohydrates to high-value products. Here we leverage the proclivity by directing the C–C bond formation in a catalytic arylation pathway using lignin-derived phenols with high nucleophilicity. The selectively condensed lignin, isolated in near-quantitative yields while preserving its prominent cleavable β-ether units, can be unlocked in a tandem catalytic process involving aryl migration and transfer hydrogenation. Lignin in wood is thereby converted to benign bisphenols (34–48 wt%) that represent performance-advantaged replacements for their fossil-based counterparts. Delignified pulp from cellulose and xylose from xylan are co-produced for textile fibres and renewable chemicals. This condensation-driven strategy represents a key advancement complementary to other promising monophenol-oriented approaches targeting valuable platform chemicals and materials, thereby contributing to holistic biomass valorization.
