Abstract:Buzz-pollinated pollen flowers have pollen as the primary resource for pollinators and must deal with a conflict between the exploitation of pollen grains by bees and pollination success. It has been hypothesized that heterostemony allows division of labour between stamens as a solution to the pollen dilemma. To test the division of labour hypothesis, we chose Cassia fistula, which has a trimorphic androecium and analysed androecium development, pollen grain release mechanisms and visitor behaviour. We explored the reflectance of floral organs and carried out an exclusion experiment to test the attractiveness of each stamen morph to the bee species. Finally, we explored the structural, ultrastructural and functional variation between the pollen grains, including pollen viability across stamen morphs. The differences among the three stamen morphs, which is developed from two whorls of the stamen, are the first evidence of the division of labour in our study system. Large Bombus and Xylocopa bees actively and exclusively exploited the pollen grains from the central poricidal anthers generating pollen deposition on their bodies. The reflectance pattern of floral organs indicated a targeting of these large bees to the central anthers, corroborated by the anther manipulative experiment where only the exclusion of the anthers positioned in the flower centre, especially the intermediate stamens, reduced bee visits. Both results revealed a division of labour, in which the intermediate stamen morph was responsible for both floral attractiveness and pollen resources. Only the largest stamen morph produced germinable pollen grains, highlighting their role as pollinating stamens. The smallest stamen morph has a less clear function, likely representing an economy in pollen production for feeding function. Our findings suggest that the evolution of the trimorphic androecium is associated with division of labour in large pollen flowers and can represent a strong strategy for circumventing the pollen dilemma, optimizing the feeding function by reducing pollen grain investment from central anthers.

Abstract:Recent advances in microscopy have given us important clues as to the nature of chromosome segregation in bacteria. Most current observations favour the view that the process is co‐replicational: DNA replication forks are anchored at the cell centre, and the newly replicated DNA is moved towards the cell poles. This scheme can account for orderly segregation even at high growth rates where multiple replication cycles overlap. We argue that there are five distinct activities directly involved in co‐replicational segregation dynamics. These we refer to as Push, Direct, Condense, Hold and Clear. We attempt to assign one of these roles to each protein implicated in chromosome segregation. The proposed process is very different from mitosis in eukaryotic cells and perhaps more closely resembles the formation of separate sister chromatids during DNA replication.
Touch-sensitive stamens enhance pollen dispersal by scaring away visitors

Abstract:Animal-pollinated plants have to get pollen to a conspecific stigma while protecting it from getting eaten. Touch-sensitive stamens, which are found in hundreds of flowering plants, are thought to function in enhancing pollen export and reducing its loss, but experimental tests are scarce. Stamens of Berberis and Mahonia are inserted between paired nectar glands and when touched by an insect's tongue rapidly snap forward so that their valvate anthers press pollen on the insect's tongue or face. We immobilized the stamens in otherwise unmodified flowers and studied pollen transfer in the field and under enclosed conditions. On flowers with immobilized stamens, the most common bee visitor stayed up to 3.6× longer, yet removed 1.3× fewer pollen grains and deposited 2.1× fewer grains on stigmas per visit. Self-pollen from a single stamen hitting the stigma amounted to 6% of the grains received from single bee visits. Bees discarded pollen passively placed on their bodies, likely because of its berberine content; nectar has no berberine. Syrphid flies fed on both nectar and pollen, taking more when stamens were immobilized. Pollen-tracking experiments in two Berberis species showed that mobile-stamen-flowers donate pollen to many more recipients. These results demonstrate another mechanism by which plants simultaneously meter out their pollen and reduce pollen theft.

Abstract:Pollinator behavior is vital to plant-pollinator interactions, affecting the acquisition of floral rewards, patterns of pollen transfer, and plant reproductive success. During buzz pollination, bees produce vibrations with their indirect flight muscles to extract pollen from tube-like flowers. Vibrations can be transmitted to the flower via the mandibles, abdomen, legs, or thorax directly. Vibration amplitude at the flower determines the rate of pollen release and should vary with the coupling of bee and flower. This coupling often occurs through anther biting, but no studies have quantified how biting affects flower vibration. Here, we used high-speed filmography to investigate how flower vibration amplitude changes during biting in Bombus terrestris visiting two species of buzz-pollinated flowering plants: Solanum dulcamara and Solanum rostratum (Solanaceae). We found that floral buzzing drives head vibrations up to 3 times greater than those of the thorax, which doubles the vibration amplitude of the anther during biting compared with indirect vibration transmission when not biting. However, the efficiency of this vibration transmission depends on the angle at which the bee bites the anther. Variation in transmission mechanisms, combined with the diversity of vibrations across bee species, yields a rich assortment of potential strategies that bees could employ to access rewards from buzz-pollinated flowers.
Functional implications of the specialized staminal appendages in alpine ginger (Roscoea spp.: Zingiberaceae)

Abstract:Floral organs are widely believed to enhance the pollination and reproductive success of angiosperms. However, the functional implication of some floral structures is still unknown. In this study, we explored the functional role of staminal appendages on male and female reproductive success of Himalayan Roscoea spp. and tested if their function differed between species with biotic pollination and autonomous selfing. Phenotypic manipulation is a powerful approach to test the functional effect of a particular trait on plant fitness. We compared various proxies of pollination success between intact flowers and flowers with manually excised staminal appendages. We found that the rate of visitation did not differ between intact and manipulated flowers. Our results revealed that in outcrossing Roscoea spp., the staminal appendages act as triggering devices to facilitate pollen release and deposition and also to manipulate the foraging position of pollinators to ensure both male and female reproductive success. In contrast, in autonomously selfing Roscoea spp., the removal of staminal appendages did not affect any aspect of pollination processes. Our results suggest that the staminal appendages are an integral component of outcrossing in Roscoea spp. and are maintained by selection pressure through both male and female reproductive success. This study provides important insights on how variation in breeding systems can provoke changes in the structure and function of floral organs among congeners.
The fate of pollen in two morphologically contrasting buzz-pollinated Solanum flowers

Abstract:Study premise: Pollen transfer efficiency (PTE) and pollen deposition patterns on a pollinator's body significantly influence plant reproductive success. However, studies on pollen fates (i.e., the destination of pollen grains after being released) in animal-pollinated species, particularly those offering pollen as the sole reward, are limited. Here, we investigated pollen fates in two nectarless, buzz-pollinated Solanum species with contrasting floral morphology. Methods: We conducted experimental trials involving one pollen donor and four recipient flowers of Solanum rostratum and S. dulcamara, using captive Bombus terrestris as pollinator. After each trial, we assessed the amount of pollen remaining in the anthers, deposited on stigmas, placed on the pollinator, and falling to the ground. We then estimated pollen fates and PTE, and modelled their pollen deposition curves. Key results: We found that S. rostratum produced more pollen but had a more restricted pollen dispensing schedule compared to S. dulcamara. Although PTE was similar between species (0.72% vs. 1.07%, for S. rostratum and S. dulcamara, respectively), pollen loss mainly occurred due to falling to the ground in S. rostratum and bee grooming in S. dulcamara, potentially explained by their different floral architectures. Both species exhibited a typical exponential decay pattern in pollen deposition, with the first visited flowers receiving the most outcross pollen. Conclusions: Our results suggest that PTE in pollen-rewarding, buzz-pollinated species is quantitatively similar to that in nectar-rewarding flowers with single pollen units (PTE = 1-2%), and that different buzz-pollinated flower architectures achieve similar PTE although through different pollen-loss pathways.

Abstract:Novel pollen co‐transport networks reveal that plants (e.g., Delphinium uliginosum—DEUL shown at the centre of the figure) face different pollen‐pollen competitive landscapes [indicated, for example, by the variation in modularity (Q)] as they travel on the bodies of different pollinators with unexplored ecological and evolutionary consequences. Different node colour within networks indicates different modules of potential pollen‐pollen interactions. The patterns and drivers of pollen transport on insect bodies can have important consequences for plant reproductive success and floral evolution; however, they remain little studied. Recently, pollinator bodies have been further described as pollen competitive arenas, where pollen grains can compete for space, with implications for the evolution of pollen dispersal strategies and plant community assembly. However, the identity, strength, and diversity of pollen competitive interactions and how they vary across pollinator functional groups is not known. Evaluating patterns and drivers of the pollen co‐transport landscape and how these vary across different pollinator groups is central to further our understanding of floral evolution and co‐flowering community assembly. Here, we integrate information on the number and identity of pollen grains on individual insect pollen loads with network analyses to uncover novel pollen co‐transport networks and how these vary across pollinator functional groups (bees and bee flies). We further evaluate differences in pollen load size, species composition, diversity and phylogenetic diversity among insect groups and how these relate to body size and gender. Pollen co‐transport networks were diverse and highly modular in bees, with groups of pollen species interacting more often with each other on insect bodies. However, the number, identity and frequency of competitors that pollen grains encounter on insect bodies vary between some pollinator functional groups. Other aspects of pollen loads such as their size, richness and phylogenetical diversity were shaped by bee size or gender, with females carrying larger but less phylogenetically diverse pollen loads than males. Synthesis. Our results show that the number, identity and phylogenetic relatedness of pollen competitors changes as pollen grains travel on the body of different pollinators. As a result, pollinator groups impose vastly different interaction landscapes during pollen transport, with so far unknown consequences for plant reproductive success, floral evolution and community assembly.

Abstract:Buzz pollination involves the release of pollen from, primarily, poricidal anthers through vibrations generated by certain bee species. Despite previous experimental and numerical studies, the intricacies of pollen dynamics within vibrating anthers remain elusive due to the challenges in observing these small-scale, opaque systems. This research employs the discrete element method (DEM) to simulate the pollen expulsion process in vibrating anthers. By exploring various frequencies and displacement amplitudes, a correlation between the maximum jerk of anther walls and the initial rate of pollen expulsion is observed under translating oscillations. This study highlights that while increased vibration intensity enhances pollen release, the rate of increase diminishes at higher intensities. Our findings also reveal the significant role of pollen-pollen interactions, which account for upwards of one-third of the total collisions. Comparisons between poricidal and pseudoporicidal anther geometries suggest that pore size and shape also influence expulsion rates. This research provides a foundation for more comprehensive models that can incorporate additional factors such as cohesion, adhesion, and Coulomb forces, paving the way for deeper insights into the mechanics of buzz pollination and its variability across different anther types and vibration parameters.

Abstract:Several agriculturally valuable plants store their pollen in tube-like poricidal anthers, which release pollen through buzz pollination. In this process, bees rapidly vibrate the anther using their indirect flight muscles. The stiffness and resonant frequency of the anther are crucial for effective pollen release, yet the impact of turgor pressure on these properties is not well understood. Here, we perform three-point flexure tests and experimental modal analysis to determine anther transverse stiffness and resonant frequency, respectively. Dynamic nanoindentation is used to identify the anther's storage modulus as a function of excitation frequency. We subsequently develop mathematical models to estimate how turgor pressure changes after the anther is removed from a flower, thereby emulating zero water availability. We find that 30 minutes post-ablation, anther stiffness decreases 60%. Anther resonant frequency decreases 20% 60 minutes post-ablation. Models indicate turgor pressure in the fresh anther is about 0.2 - 0.3 MPa. Our findings suggest that natural fluctuations in turgor pressure due to environmental factors like temperature and light intensity may require bees to adjust their foraging behaviors. Interestingly, anther storage modulus increased with excitation frequency, underscoring the need for more sophisticated mechanical models that consider viscous fluid transport through plant tissue moving forward.

Abstract:ABSTRACT Specialization in plant pollination systems can arise from traits that function as filters of flower visitors. This may involve chemical traits such as floral volatiles that selectively attract favoured visitors and non-volatile nectar constituents that selectively deter disfavoured visitors through taste or longer-term toxic effects or both. We explored the functions of floral chemical traits in the African milkweed Gomphocarpus physocarpus, which is pollinated almost exclusively by vespid wasps, despite having nectar that is highly accessible to other insects such as honeybees. We demonstrated that the nectar of wasp-pollinated G. physocarpus contains cardenolides that had greater toxic effects on Apis mellifera honeybees than on Vespula germanica wasps, and also reduced feeding rates by honeybees. Behavioural experiments using natural compositions of nectar compounds showed that these interactions are mediated by non-volatile nectar chemistry. We also identified volatile compounds with acetic acid as a main component in the floral scent of G. physocarpus that elicited electrophysiological responses in wasp antennae. Mixtures of these compounds were behaviourally effective for attraction of V. germanica wasps. The results show the importance of both volatile and non-volatile chemical traits as filters that lead to specialization in plant pollination systems.

What keeps the style under tension? Experimental tests to understand the biomechanics of the explosive style movement in Marantaceae

Abstract:Pollination in Marantaceae is mediated by an explosive style movement. Before release, style tension is held by the hooded staminode. When a pollinator touches the trigger appendage of the hooded staminode the latter deforms and the style rapidly curls upwards. This movement has been interpreted as a turgor movement by some authors, but recent studies clearly indicate that setup, hold and release of tension are purely mechanical processes. However, in view of the high diversity of hooded staminodes, the question arises what keeps the tension in species with very thin staminodes. To test the holding mechanisms, we conducted mechanical and physico-chemical release experiments in four species with robust and four species with thin hooded staminodes in their natural tropical environment. We found almost the same response of all species to mechanical treatments, but species-specific reactions to different physico-chemical conditions. This indicates that style release follows the same mechanical principles in all species, but that the sensitivity of the explosive movement depends on material properties like tissue thickness and turgescence. As to the holding mechanisms, we found different degrees of floral synorganization. The hood of the hooded staminode formerly interpreted as an important holding structure does not play a noteworthy role. Instead, the basal plate of the hooded staminode antagonises the pressure of the style head against the holding point of the hooded staminode in species with robust hooded staminodes and well-developed basal plates. In some species with a thin hooded staminode, the latter is closely attached to the style and most likely stabilises tension by adhesive forces. In another species, a morphologically analogous structure adopts the function of the basal plate. We conclude that the holding mechanism of the style tension diversified during the evolution of Marantaceae whereas the release mechanism itself has been conserved throughout the family.

Armament imbalances: match and mismatch in plant-pollinator traits of highly specialized long-spurred orchids

Abstract:Background: Some species of long-spurred orchids achieve pollination by a close association with long-tongued hawkmoths. Among them, several Habenaria species present specialized mechanisms, where pollination success depends on the attachment of pollinaria onto the heads of hawkmoths with very long proboscises. However, in the Neotropical region such moths are less abundant than their shorter-tongued relatives and are also prone to population fluctuations. Both factors may give rise to differences in pollinator-mediated selection on floral traits through time and space. Methodology/principal findings: We characterized hawkmoth assemblages and estimated phenotypic selection gradients on orchid spur lengths in populations of three South American Habenaria species. We examined the match between hawkmoth proboscis and flower spur lengths to determine whether pollinators may act as selective agents on flower morphology. We found significant directional selection on spur length only in Habenaria gourlieana, where most pollinators had proboscises longer than the mean of orchid spur length. Conclusions/significance: Phenotypic selection is dependent on the mutual match between pollinator and flower morphologies. However, our findings indicate that pollinator-mediated selection may vary through time and space according to local variations in pollinator assemblages.

Abstract:Accelerated stability tests are indicated to assess, within a short time, the degree of chemical degradation that may affect an active substance, either alone or in a formula, under normal storage conditions. This method is based on increased stress conditions to accelerate the rate of chemical degradation. Based on the equation of the straight line obtained as a function of the reaction order (at 50 and 70 degrees C) and using Arrhenius equation, the speed of the reaction was calculated for the temperature of 20 degrees C (normal storage conditions). This model of accelerated stability test makes it possible to predict the chemical stability of any active substance at any given moment, as long as the method to quantify the chemical substance is available. As an example of the applicability of Arrhenius equation in accelerated stability tests, a 2.5% sodium hypochlorite solution was analyzed due to its chemical instability. Iodometric titration was used to quantify free residual chlorine in the solutions. Based on data obtained keeping this solution at 50 and 70 degrees C, using Arrhenius equation and considering 2.0% of free residual chlorine as the minimum acceptable threshold, the shelf-life was equal to 166 days at 20 degrees C. This model, however, makes it possible to calculate shelf-life at any other given temperature.
Nectary tracks as pollinator manipulators: The pollination ecology of Swertia bimaculata (Gentianaceae)

Abstract:Floral nectaries are closely associated with biotic pollination, and the nectar produced by corolla nectaries is generally enclosed in floral structures. Although some Swertia spp. (Gentianaceae), including S. bimaculata, evolved a peculiar form of corolla nectaries (known as "gland patches") arranged in a conspicuous ring on the rotate corolla and that completely expose their nectar, little is known about the pollination of these plants. Two hypotheses were made concerning the possible effects of gland patches: visual attraction and visitor manipulation. The floral traits, mating system, and insect pollination of S. bimaculata were examined, and the pollination effects of gland patches were evaluated. A comparative study was made using Swertia kouitchensis, a species with fimbriate nectaries. Swertia bimaculata flowers were protandrous, with obvious stamen movement leading to herkogamy in the female phase and to a significant reduction in nectary-anther distance. The species is strongly entomophilous and facultatively xenogamous. The daily reward provided per flower decreased significantly after the male phase. The most effective pollinators were large dipterans, and the visiting proportion of Diptera was significantly higher in S. bimaculata than in S. kouitchensis. Most visitors performed "circling behavior" in S. bimaculata flowers. Removing or blocking the nectaries caused no reduction in visiting frequency but a significant reduction in visit duration, interrupting the circling behavior. The circling behavior was encouraged by nectar abundance and promoted pollen dispersal. Visitor species with small body size had little chance to contact the anthers or stigma, revealing a filtration effect exerted by the floral design. These results rejected the "visual attraction" hypothesis and supported the "visitor manipulation" hypothesis. The nectary whorl within a flower acted like a ring-shaped track that urged nectar foragers to circle on the corolla, making pollination in S. bimaculata flowers more orderly and selective than that in classically generalist flowers.

Abstract:The pollination efficiency and their effects on floral morph variation of short‐tongued and long‐tongued pollinators were investigated in two Limonium myrianthum populations with short corolla tubular small flowers and considerable floral morph variation. We found that asymmetrical disassortative pollination affects floral morph variation. Compared with nectar‐seeking ST insects, pollen‐seeking ST insects and LT insects caused asymmetrical disassortative pollen transfer between high‐ and low‐level sexual organs due to the preference for reward and the position of stigmas/anthers within the flower. In heterostylous plants, short‐tongued pollinators are often ineffective/inefficient owing to the limitations imposed by a long corolla tube. However, it is unclear how disassortative pollen transfer is achieved in small flowers. We investigated the pollination pattern and floral morph variation by analyzing heterostylous syndrome, pollinator groups, and pollen deposition after a single visitation in two Limonium myrianthum populations with short‐corolla‐tubular small flowers. The predominant pollinators in the Hutubi population were pollen‐seeking short‐tongued syrphids, which can only transfer pollen between high‐level sexual organs. In the Xishan population, nectar‐seeking short‐tongued insects were efficient pollinators with symmetrical disassortative pollen transfer between high‐ and low‐level sexual organs, whereas long‐tongued pollinators had a low efficiency between high‐level sexual organs due to the low contact probability with the stigma of long‐styled flowers (L‐morph), which no longer offered the same advantage observed in tubular flowers. Asymmetrical disassortative pollination may cause the female fitness of short‐styled (S‐morph) individuals in the Hutubi and L‐morph individuals in the Xishan population to suffer greater selection pressure and exhibit a higher degree of floral morph variation. Limonium myrianthum exhibits an unusual pollination pattern in which the small flowers with short corolla tubes make it possible for short‐tongued insects to become effective pollinators. However, factors such as the position of stigma–anther within the flower, pollinator species and their preference further caused asymmetrical disassortative pollen transfer. Therefore, more factors should be considered when evaluating the effectiveness of short‐ and long‐tongued insects in pollination service.

Flowers and floral buds at different stages were prepared for observation under Scanning Electron Microscopy (SEM), Light Microscopy (ML), and X-ray Computed Tomography (XCT). Pollinator observations were conducted in populations in southern Africa and floral advertising and reward traits were quantified. Five (rarely six) sepals emerge helically, followed by the petal primordia in unidirectional order. The carpel arises synchronously with the stamens and expands rapidly. Stamen initiation occurs unidirectionally, with three abaxial antesepalous stamens fully developing while other stamen primordia remain sterile and underdeveloped (staminodial). The number of adaxial staminode tips can fluctuate by branching, regardless of their original whorl. Concentric collateral bundles supply all the staminodes. Swallowtail butterflies transfer pollen through their fluttering wings that touch the abaxial anthers and stigma while feeding on nectar in the hypanthium located on the adaxial side. Bauhinia galpinii exhibits an androecium characterised by secondary, unstable branching of staminodes, which form adaptive structures suited to its pollinators. The variation in androecium structure may be attributed to heterochronic changes during development, marked by an early carpel initiation and its basal expansion leading to spatial restriction. This ontogenetic contingency, associated with selective pressures from pollinators, notably swallowtail butterflies, seems to play a role in selecting this complex and distinct floral architecture.

Pollinator Preferences for Floral Volatiles Emitted by Dimorphic Anthers of a Buzz-Pollinated Herb

Abstract:Floral scents attract pollinators to plant rewards; in nectarless flowers, pollen grains are the only reward. Thus, pollen not only fertilizes ovules, but also feeds pollinators. This dilemma is resolved by specialization of anthers (i.e., heteranthery): feeding anthers that feed pollinators and pollinating anthers for fertilization. We hypothesized that the chemical composition of floral volatiles differs between the two types of anther and influences pollination preference for feeding anthers. We used Solanum rostratum as a study model because its heterantherous flowers produce a floral scent that suggests a close association with their pollinators. The main aim of this study was to determine the chemical composition of the two types of anther and to investigate how they influence foraging behaviour of pollinators. To characterize this composition, we used solid phase microextraction and hexane extraction followed by gas chromatography-mass spectrometry. We registered 12 volatile compounds in S. rostratum floral extracts, mainly aromatic and sesquiterpene compounds. The proportion of these compounds differed between feeding and pollinating anthers. Some of these compounds were probably emitted by osmophores located in both anther types. Also, we used electroantennography to investigate Melipona solani antennal response to floral volatiles. The M. solani antennae are receptive to the highest floral extract dose tested. Finally, we conducted two behavioural bioassays to test bee attraction for each type of floral extract: a) multiple-choice in a feeding arena using M. solani and b) Y-olfactometer bioassay using Bombus impatiens. Both bee species preferred feeding anthers in bioassays. In conclusion, heteranthery involves chemical differentiation (i.e., proportion of volatiles compounds) in anther specialization that influences bee preference for feeding anthers over pollinating anthers.
Complex floral morphology and scent chemistry mediate hawkmoth pollination of an African orchid in the diverse Habenaria clade

Abstract:Abstract One explanation for the evolution of complex floral morphology in orchids is that it can function as a component of ‘lock-and-key’ mechanisms that ensure pollen transfer by specific flower visitors. We investigated the roles of floral morphology and scent in the pollination of Bonatea antennifera, an African orchid that is phylogenetically embedded in the hyper-diverse Habenaria clade. Using camera traps and direct observations, we established that B. antennifera is pollinated by an assemblage of short-tongued hawkmoths with proboscides ~4 cm in length, corresponding to the spur length of the orchid. Pollinaria are attached between the palps of these hawkmoths via an intricate mechanism involving a wedge-shaped rostellum chamber and a tooth in the floral spur that deflect the head and proboscis of the hawkmoth to either side of the spur entrance. This functional subdivision of the flower results in removal of a single pollinarium per visit. Scent production is synchronized with the peak activity of hawkmoths at dusk, and electrophysiology experiments showed that hawkmoth antennae respond to several of the dominant volatile terpenoid compounds [e.g. R-(−)-linalool, caryophyllene epoxide, and (E)-nerolidol] emitted by the flowers. These results illustrate the functional significance of complex floral morphology and scent for pollen transfer in orchids.

Abstract:The stinger is a very small and efficient device that allows honeybees to perform two main physiological activities: repelling enemies and laying eggs for reproduction. In this study, we explored the specific characteristics of stinger penetration, where we focused on its movements and the effects of it microstructure. The stingers of Italian honeybees (Apis mellifera ligustica) were grouped and fixed onto four types of cubic substrates, before pressing into different substrates. The morphological characteristics of the stinger cross-sections were analyzed before and after penetration by microscopy. Our findings suggest that the honeybee stinger undergoes helical and clockwise rotation during penetration. We also found that the helical penetration of the stinger is associated directly with the spiral distribution of the barbs, thereby confirming that stinger penetration involves an advanced microstructure rather than a simple needle-like apparatus. These results provide new insights into the mechanism of honeybee stinger penetration.
