Abstract:Shallow CO2 vents are used to test ecological hypotheses about the effects of ocean acidification (OA). Here, we studied fish assemblages associated with Cymodocea nodosa meadows exposed to high pCO2/low pH conditions at a natural CO2 vent in the Mediterranean Sea. Using underwater visual census, we assessed fish community structure and biodiversity in a low pH site (close to the CO2 vent), a close control site and a far control site, hypothesising a decline in biodiversity and a homogenization of fish assemblages under OA conditions. Our findings revealed that fish diversity did not show a unique spatial pattern, or even significant relationships with pH, but correlated with seagrass leaf canopy. Among-site similarity was found in the abundance of juveniles, contrary to the expected impacts of OA on early life stages. However, pH seems an important driver in structuring fish assemblage in the low pH site, despite its high similarity with the close control site. This unexpected pattern may represent a combined response of fish mobility, enhanced food resources in the acidified site, and a 'recovery area' effect of the adjacent control site.

Abstract:Ocean acidification has been broadly recognised to have effects on the structure and functioning of marine benthic communities. The selection of tolerant or vulnerable species can also occur during settlement phases, especially for calcifying organisms which are more vulnerable to low pH–high pCO2 conditions. Here, we use three natural CO2 vents (Castello Aragonese north and south sides, and Vullatura, Ischia, Italy) to assess the effect of a decrease of seawater pH on the settlement of Mollusca in Posidonia oceanica meadows, and to test the possible buffering effect provided by the seagrass. Artificial collectors were installed and collected after 33 days, during April–May 2019, in three different microhabitats within the meadow (canopy, bottom/rhizome level, and dead matte without plant cover), following a pH decreasing gradient from an extremely low pH zone (pH < 7.4), to ambient pH conditions (pH = 8.10). A total of 4659 specimens of Mollusca, belonging to 57 different taxa, were collected. The number of taxa was lower in low and extremely low pH conditions. Reduced mollusc assemblages were reported at the acidified stations, where few taxa accounted for a high number of individuals. Multivariate analyses revealed significant differences in mollusc assemblages among pH conditions, microhabitat, and the interaction of these two factors. Acanthocardia echinata, Alvania lineata, Alvania sp. juv, Eatonina fulgida, Hiatella arctica, Mytilys galloprovincialis, Musculus subpictus, Phorcus sp. juv, and Rissoa variabilis were the species mostly found in low and extremely low pH stations, and were all relatively robust to acidified conditions. Samples placed on the dead matte under acidified conditions at the Vullatura vent showed lower diversity and abundances if compared to canopy and bottom/rhizome samples, suggesting a possible buffering role of the Posidonia on mollusc settlement. Our study provides new evidence of shifts in marine benthic communities due to ocean acidification and evidence of how P. oceanica meadows could mitigate its effects on associated biota in light of future climate change.

Abstract:Ocean acidification has been shown to have highly variable effects, with many negative and some positive responses from individual species, while community level effects are largely unknown. Although an overall loss of biodiversity is expected, predicting the effects of ocean acidification on whole assemblages can be problematic as both direct and indirect effects of acidification must be taken into consideration. This study demonstrates how invertebrate assemblages associated with the highly productive seagrass, Posidonia oceanica, respond to natural acidification that occurs at CO2 vents off the coast of Italy. We examined seasonal differences in invertebrate community structure between two distinct pH zones: control (pH8.1) and acidified (pH7.8) and show that many groups of invertebrate taxa were robust to acidification effects. Differences in community structure appeared to be driven by the indirect effects of acidification, such as changes to canopy structure and food availability, rather than physiological intolerance to low pH. The number of invertebrates collected in acidified stations was almost double that of control stations during the study and many heavily calcified species appeared to thrive. These results highlight how positive indirect effects may buffer the ecological impacts of acidification, and provide evidence that this highly productive, nearshore habitat may provide refuge to its associated communities from future ocean acidification.

Abstract:Ocean Acidification (OA), due to rising atmospheric CO2, can affect the seagrass holobiont by changing the plant's ecophysiology and the composition and functioning of its epiphytic community. However, our knowledge of the role of epiphytes in the productivity of the seagrass holobiont in response to environmental changes is still very limited. CO2 vents off Ischia Island (Italy) naturally reduce seawater pH, allowing to investigate the adaptation of the seagrass Posidonia oceanica L. (Delile) to OA. Here, we analyzed the percent cover of different epiphytic groups and the epiphytic biomass of P. oceanica leaves, collected inside (pH 6.9-7.9) and outside (pH 8.1-8.2) the CO2 vents. We estimated the contribution of epiphytes to net primary production (NPP) and respiration (R) of leaf sections collected from the vent and ambient pH sites in laboratory incubations. Additionally, we quantified net community production (NCP) and community respiration (CR) of seagrass communities in situ at vent and ambient pH sites using benthic chambers. Leaves at ambient pH sites had a 25% higher total epiphytic cover with encrusting red algae (32%) dominating the community, while leaves at vent pH sites were dominated by hydrozoans (21%). Leaf sections with and without epiphytes from the vent pH site produced and respired significantly more oxygen than leaf sections from the ambient pH site, showing an average increase of 47 ± 21% (mean ± SE) in NPP and 50 ± 4% in R, respectively. Epiphytes contributed little to the increase in R; however, their contribution to NPP was important (56 ± 6% of the total flux). The increase in productivity of seagrass leaves adapted to OA was only marginally reflected by the results from the in situ benthic chambers, underlining the complexity of the seagrass community response to naturally occurring OA conditions.
Phencyclidine dependence: the relationship of dose and serum concentrations to operant behavioral effects.

Abstract:The dependence-producing properties of 10 days of chronic i.v. infusions of phencyclidine (PCP) and the relationship between PCP serum concentrations and behavioral effects were studied in Sprague-Dawley rats. For dependence studies, rats were trained to respond for food under a fixed-ratio 30 schedule during half-hour response periods every 6 hr. After training, implantation of jugular catheters, and restabilization of behavior, the rats were infused with PCP.HCl at 3.2, 5.6, 10.0 or 17.8 mg/kg/day (n = 5 or 6 per dose). The two higher doses initially decreased response rates, but tolerance developed within 4 to 5 days. When PCP infusions were terminated, dose-dependent decreases in session response rate occurred in the three highest dose groups (P less than .05). Mild, overt signs of abstinence were observed only in the highest dose group. Response rates returned to base line within 2 to 3 days after stopping PCP infusions. PCP serum concentrations in rats infused with 10 mg of PCP.HCl/kg/day for 10 days were stable from hour 24 to day 10 (mean steady-state concentration (+/- S.D.) = 97 (+/- 20) ng of PCP/ml; n = 4). The average terminal elimination half-life after stopping infusions on day 10 was 4.6 hr. Comparison of the average response rates with the average serum concentrations showed that during the first 24 hr of infusions, the rate of responding for food decreased as PCP concentrations increased; however, once the animals became tolerant to PCP there was no relationship. In contrast, during the first 24 hr after stopping infusions, response rates decreased as serum concentrations decreased.

Abstract:Anthropogenic stressors are predicted to alter biodiversity and ecosystem functioning worldwide. However, scaling up from species to ecosystem responses poses a challenge, as species and functional groups can exhibit different capacities to adapt, acclimate, and compensate under changing environments. We used a naturally acidified seagrass ecosystem (the endemic Mediterranean Posidonia oceanica) as a model system to examine how ocean acidification (OA) modifies the community structure and functioning of plant detritivores, which play vital roles in the coastal nutrient cycling and food web dynamics. In seagrass beds associated with volcanic CO2 vents (Ischia, Italy), we quantified the effects of OA on seagrass decomposition by deploying litterbags in three distinct pH zones (i.e., ambient, low, extreme low pH), which differed in the mean and variability of seawater pH. We replicated the study in two discrete vents for 117 days (litterbags sampled on day 5, 10, 28, 55, and 117). Acidification reduced seagrass detritivore richness and diversity through the loss of less abundant, pH-sensitive species but increased the abundance of the dominant detritivore (amphipod Gammarella fucicola). Such compensatory shifts in species abundance caused more than a threefold increase in the total detritivore abundance in lower pH zones. These community changes were associated with increased consumption (52%-112%) and decay of seagrass detritus (up to 67% faster decomposition rate for the slow-decaying, refractory detrital pool) under acidification. Seagrass detritus deployed in acidified zones showed increased N content and decreased C:N ratio, indicating that altered microbial activities under OA may have affected the decay process. The findings suggest that OA could restructure consumer assemblages and modify plant decomposition in blue carbon ecosystems, which may have important implications for carbon sequestration, nutrient recycling, and trophic transfer. Our study highlights the importance of within-community response variability and compensatory processes in modulating ecosystem functions under extreme global change scenarios.

Abstract:Seagrass meadows form highly productive and diverse ecosystems in coastal areas worldwide, where they are increasingly exposed to ocean acidification (OA). Efficient nitrogen (N) cycling and uptake are essential to maintain plant productivity, but the effects of OA on N transformations in these systems are poorly understood. Here we show that complete N cycling occurs on leaves of the Mediterranean seagrass Posidonia oceanica at a volcanic CO 2 vent near Ischia Island (Italy), with OA affecting both N gain and loss while the epiphytic microbial community structure remains largely unaffected. Daily leaf-associated N 2 fixation contributes to 35% of the plant's N demand under ambient pH, while it contributes to 45% under OA. Nitrification potential is only detected under OA, and N-loss via N 2 production increases, although the balance remains decisively in favor of enhanced N gain. Our work highlights the role of the N-cycling microbiome in seagrass adaptation to OA, with key N transformations accelerating towards increased N gain.

Abstract:INTRODUCTION Investigations in both alcohol and smoking have provided preliminary evidence for the use of attentional training interventions to reduce vigilance toward salient drug cues. The current study evaluated the efficacy of an attentional training intervention for reducing attentional bias toward smoking cues and craving among cigarette smokers. METHODS Participants were randomized to receive an attentional training intervention or a control training. Sixty-four regular cigarette smokers (individuals smoking at least 10 cigarettes/day for at least 1 year) provided informed consent and were enrolled in the study. The dot probe paradigm was used both to measure attentional bias and for the training intervention. Attentional bias and craving were measured prior to and following training. RESULTS Results indicated that the randomized groups did not differ significantly in attentional bias as measured by novel stimuli and, correspondingly, in craving response to relevant cues following the training intervention. CONCLUSIONS These results for a single-session training suggest that this "dose" is insufficient for reducing attentional bias. Given the success of this intervention in other applications, research on the efficacy of multiple-session trainings is an important area for further study.

Abstract:Ocean acidification (OA) may have varied effects on fish eco-physiological responses. Most OA studies have been carried out in laboratory conditions without considering the in situ p CO 2 /pH variability documented for many marine coastal ecosystems. Using a standard otolith ageing technique, we assessed how in situ ocean acidification (ambient, versus end-of-century CO 2 levels) can affect somatic and otolith growth, and their relationship in a coastal fish. Somatic and otolith growth rates of juveniles of the ocellated wrasse Symphodus ocellatus living off a Mediterranean CO 2 seep increased at the high- p CO 2 site. Also, we detected that slower-growing individuals living at ambient p CO 2 levels tend to have larger otoliths at the same somatic length (i.e. higher relative size of otoliths to fish body length) than faster-growing conspecifics living under high p CO 2 conditions, with this being attributable to the so-called ‘growth effect’. Our findings suggest the possibility of contrasting OA effects on fish fitness, with higher somatic growth rate and possibly higher survival associated with smaller relative size of otoliths that could impair fish auditory and vestibular sensitivity.

Abstract:Although seagrasses are expected to thrive in future acidified oceans by overcoming low CO2 diffusion into plant tissues, the co‐occurrence of environmental stressors may affect their growth. Volcanic CO2 vents are often associated with toxic gases and metal‐rich fluids representing ideal sites to assess the effects of multiple stressors. We evaluated the response of Posidonia oceanica growing near shallow CO2 vents characterized by H2S spill‐out by comparing meadow structure and phenology to an area with no gas emissions. Seagrass descriptors at meadow, shoot and leaf level indicated that P. oceanica experienced stressful conditions at the vent area, in clear contrast to the flourishing features of P. oceanica previously described at CO2 vents with no evidence of toxic inputs. Furthermore, the reduction in both leaf δ34S and growth at the vent area indicates that sulfide intrusion occurs and affects seagrass growth performance, dampening the expected beneficial effects of high CO2 levels.

Abstract:Oceans' absorption of human-related CO2 emissions leads to a process called ocean acidification (OA), consisting of the decrease of the seawater pH with negative consequences for many marine organisms. In this study, we investigate the microbial community of two species of polychaetes found in naturally acidified CO2 vents: the nereid Platynereis massiliensis complex and the syllid Syllis prolifera. Animals were collected in the CO2 vents of Castello Aragonese (Gulf of Naples, Ischia, Italy) in three zones at decreasing pH. For the analysis of the microbiome, the V3-V4 hypervariable region of the 16S ribosomal RNA gene of 40 worm samples was sequenced on an Illumina MiSeq platform. No difference in the microbial alpha diversity of both species was highlighted. On the contrary, the microbial composition of worms collected in the site at normal pH was different from that of the individuals obtained from the sites at lower pH. This effect was evident also in samples from the site with a slight, but relevant, degree of acidification. Amplicon sequence variants showing a significant variation among the groups of samples collected from different pH zones were reported for both polychaetes, but no common trend of variation was observed. The present study deepens our knowledge about the composition of polychaete microbiome in marine naturally acidified sites. Our results stress the importance of future investigations about the connection between the variation of environmental and polychaete microbial communities induced by OA and about the effect of these variations on polychaete key biological and ecological traits.

Abstract:Altered ocean chemistry caused by ocean acidification (OA) is expected to have negative repercussions at different levels of the ecological hierarchy, starting from the individual and scaling up to the community and ultimately to the ecosystem level. Understanding the effects of OA on benthic organisms is of primary importance given their relevant ecological role in maintaining marine ecosystem functioning. The use of functional traits represents an effective technique to investigate how species adapt to altered environmental conditions and can be used to predict changes in the resilience of communities faced with stresses associated with climate change. Artificial supports were deployed for 1-y along a natural pH gradient in the shallow hydrothermal systems of the Bottaro crater near Panarea (Aeolian Archipelago, southern Tyrrhenian Sea), to explore changes in functional traits and metabolic rates of benthic communities and the repercussions in terms of functional diversity. Changes in community composition due to OA were accompanied by modifications in functional diversity. Altered conditions led to higher oxygen consumption in the acidified site and the selection of species with the functional traits needed to withstand OA. Calcification rate and reproduction were found to be the traits most affected by pH variations. A reduction in a community's functional evenness could potentially reduce its resilience to further environmental or anthropogenic stressors. These findings highlight the ability of the ecosystem to respond to climate change and provide insights into the modifications that can be expected given the predicted future pCO 2 scenarios. Understanding the impact of climate change on functional diversity and thus on community functioning and stability is crucial if we are to predict changes in ecosystem vulnerability, especially in a context where OA occurs in combination with other environmental changes and anthropogenic stressors.

Abstract:The oceans absorb ~25% of the annual anthropogenic CO2 emissions. This causes a shift in the marine carbonate chemistry termed ocean acidification (OA). OA is expected to influence metabolic processes in phytoplankton species but it is unclear how the combination of individual physiological changes alters the structure of entire phytoplankton communities. To investigate this, we deployed ten pelagic mesocosms (volume ~50 m3) for 113 days at the west coast of Sweden and simulated OA (pCO2 = 760 μatm) in five of them while the other five served as controls (380 μatm). We found: (1) Bulk chlorophyll a concentration and 10 out of 16 investigated phytoplankton groups were significantly and mostly positively affected by elevated CO2 concentrations. However, CO2 effects on abundance or biomass were generally subtle and present only during certain succession stages. (2) Some of the CO2-affected phytoplankton groups seemed to respond directly to altered carbonate chemistry (e.g. diatoms) while others (e.g. Synechococcus) were more likely to be indirectly affected through CO2 sensitive competitors or grazers. (3) Picoeukaryotic phytoplankton (0.2-2 μm) showed the clearest and relatively strong positive CO2 responses during several succession stages. We attribute this not only to a CO2 fertilization of their photosynthetic apparatus but also to an increased nutrient competitiveness under acidified (i.e. low pH) conditions. The stimulating influence of high CO2/low pH on picoeukaryote abundance observed in this experiment is strikingly consistent with results from previous studies, suggesting that picoeukaryotes are among the winners in a future ocean.

Abstract:Many studies addressed ocean acidification (OA) effects on marine life, whereas its effects on sedimentary organic matter (OM) have received less attention. We investigated differences in OM features in sediments from unvegetated and seagrass (Posidonia oceanica) beds in a shallow hydrothermal area (Aeolian Archipelago, Mediterranean Sea), under natural (8.1-8.0) and acidified (7.8-7.9) conditions. We show that a pH difference of -0.3 units have minor effects on OM features in unvegetated sediments, but relevant consequences within acidified seagrass meadows, where OM quantity and nutritional quality are lower than those under natural pH conditions. Effects of acidified conditions on OM biogeochemistry vary between unvegetated and seagrass sediments, with lower C degradation rates and longer C turnover time in the former than in the latter. We conclude that OA, although with effects not consistent between unvegetated and vegetated sediments, can affect OM quantity, composition, and degradation, thus having possible far-reaching consequences for benthic trophic webs.

Abstract:Foundation species have been widely documented to provide suitable habitats for other species by ameliorating stressful environmental conditions. Nonetheless, their role in rescuing stress-sensitive species from adverse conditions due to climate change remains often unexplored. Here, we performed a mesocosm experiment to assess whether the seagrass, Posidonia oceanica, through its photosynthetic activity, could mitigate the negative effects of ocean acidification on larval development and growth of the calcifying sea urchin, Paracentrotus lividus. Sea urchin larvae at early and late developmental stages that are generally associated to benthic habitats, were grown in aquaria with or without P. oceanica plants, under ambient or low pH conditions predicted by the end of the century under the worst climate scenario (RCP8.5). The percentage of abnormal larvae and their total body length under different experimental conditions were assessed on early- (i.e., pluteus; 72 h post-fertilization) and final-developmental stages (i.e., echinopluteus; 30 days post-fertilization), respectively. The presence of P. oceanica increased mean daily pH values of ∼0.1 and ∼0.15 units at ambient and low pH conditions, respectively, compared with tanks without plants. When grown at low pH in association with P. oceanica, plutei showed a ∼23 % reduction of malformations and echinoplutei a ∼34 % increase in total body length, respectively, compared with larvae developing in tanks without plants. Our results suggest that P. oceanica, by increasing pH and altering seawater carbonate chemistry through its metabolic activity, could buffer the negative effects of ocean acidification on calcifying organisms and could, thus, represent a tool against climate-driven loss of biodiversity.

Abstract:Despite the effects of ocean acidification (OA) on seagrasses have been widely investigated, predictions of seagrass performance under future climates need to consider multiple environmental factors. Here, we performed a mesocosm study to assess the effects of OA on shallow and deep Posidonia oceanica plants. The experiment was run in 2021 and repeated in 2022, a year characterized by a prolonged warm water event, to test how the effects of OA on plants are modulated by thermal stress. The response of P. oceanica to experimental conditions was investigated at different levels of biological organization. Under average seawater temperature, there were no effects of OA in both shallow and deep plants, indicating that P. oceanica is not limited by current inorganic carbon concentration, regardless of light availability. In contrast, under thermal stress, exposure of plants to OA increased lipid peroxidation and decreased photosynthetic performance, with deep plants displaying higher levels of heat stress, as indicated by the over-expression of stress-related genes and the activation of antioxidant systems. In addition, warming reduced plant growth, regardless of seawater CO2 and light levels, suggesting that thermal stress may play a fundamental role in the future development of seagrass meadows. Our results suggest that OA may exacerbate the negative effects of future warming on seagrasses.
Response of Subtropical Phytoplankton Communities to Ocean Acidification Under Oligotrophic Conditions and During Nutrient Fertilization

Abstract:The subtropical oceans are home to the largest phytoplankton biome on the planet. Yet, little is known about potential impacts of ocean acidification (OA) on phytoplankton community composition in the vast oligotrophic ecosystems of the subtropical gyres. To address this question, we conducted an experiment with 9 in situ mesocosms (~35 m3) off the coast of Gran Canaria in the eastern subtropical North Atlantic over a period of 9 weeks. By establishing a gradient of pCO2 ranging from ~350 to 1,025 μatm, we simulated carbonate chemistry conditions as projected until the end of the twenty-first century. Furthermore, we injected nutrient-rich deep water into the mesocosms halfway through the experiment to simulate a natural upwelling event, which regularly leads to patchy nutrient fertilization in the study region. The temporal developments of major taxonomic groups of phytoplankton were analyzed by flow cytometry, pigment composition and microscopy. We observed distinct shifts in phytoplankton community structure in response to high CO2, with markedly different patterns depending on nutrient status of the system. Phytoplankton biomass during the oligotrophic phase was dominated by picocyanobacteria (Synechococcus), which constituted 60–80% of biomass and displayed significantly higher cell abundances at elevated pCO2. The addition of deep water triggered a substantial bloom of large, chain-forming diatoms (mainly Guinardia striata and Leptocylindrus danicus) that dominated the phytoplankton community during the bloom phase (70–80% of biomass) and until the end of the experiment. A CO2 effect on bulk diatom biomass became apparent only in the highest CO2 treatments (>800 μatm), displaying elevated concentrations especially in the stationary phase after nutrient depletion. Notably, these responses were tightly linked to distinct interspecific shifts within the diatom assemblage, particularly favoring the largest species Guinardia striata. Other taxonomic groups contributed less to total phytoplankton biomass, but also displayed distinct responses to OA treatments. For instance, higher CO2 favored the occurrence of prymnesiophyceae (Phaeocystis globosa) and dictyochophyceae, whereas dinoflagellates were negatively affected by increasing CO2. Altogether, our findings revealed considerable shifts in species composition in response to elevated CO2 and/or lower pH, indicating that phytoplankton communities in the subtropical oligotrophic oceans might be profoundly altered by ocean acidification.

Abstract:Ocean acidification has been consistently evidenced to have profound and lasting impacts on marine species. Observations have shown seagrasses to be highly susceptible to future increased pCO2 conditions, but the responses of early life stages as seedlings are poorly understood. This study aimed at evaluating how projected Mediterranean Sea acidification affects the survival, morphological and biochemical development of Posidonia oceanica seedlings through a long-term field experiment along a natural low pH gradient. Future ocean conditions seem to constrain the morphological development of seedlings. However, high pCO2 exposures caused an initial increase in the degree of saturation of fatty acids in leaves and then improved the fatty acid adjustment increasing unsaturation levels in leaves (but not in seeds), suggesting a nutritional compound translocation. Results also suggested a P. oceanica structural components remodelling which may counteract the effects of ocean acidification but would not enhance seagrass seedling productivity.
