Abstract:Drought is the most important environmental factor inhibiting plant photosynthesis. In this study, the morphological characteristics, biomass allocation, and physiological and biochemical characteristics of four potted mulberry plants under drought stress were analyzed. The study revealed the drought tolerance differences of four mulberry potted seedlings in semi-arid sandy areas of China. Combined with the results of two−way ANOVA, under normal growth conditions, Shensang No. 1 and Ji'an grew well and produced higher benefits, which was attributed to their larger leaf areas, biomass, and total Chl contents, and there were significant differences between their other traits (p < 0.05). Drought stress led to a decrease in the photosynthetic capacity of the mulberry leaves, and the drought resistance capabilities of the four mulberry trees were different. Among the trees, Aerxiang and Fujia were less affected by drought, and their cultivation in a naturally arid environment was able to achieve certain drought resistance effects. The branch length, total leaf area, and specific leaf area were significantly differently correlated with the biomass components' dry leaf weight, dry branch weight, dry root weight, total biomass, and root−to−crown ratio (p < 0.05), and there was also a significant positive correlation with the photosynthetic fluorescence parameters GS, PIABS, ABS/RC, and TRo/RC and the biochemical parameters NSC (p < 0.05). Studies have shown that plant biomass and physiological and biochemical characteristics jointly affect plant growth. Our research results will help in the screening of mulberry trees, providing data support for the strategic planning of subsequent breeding, and maximizing the quality and resource benefits of mulberry trees.

Abstract:The objective of this study is to evaluate drought resistance of citrus rootstocks and select drought resistant resources. This experiment was done with seedlings of local citrus rootstock resources of Jiangxi province in China, named 'Sanhuhongju', 'Choupigan', 'Sanhuhuahong', 'Chongyiyeju' and the common rootstocks, 'Swingle' citrumelo, 'Trifoliate' orange, 'Goutoucheng', 'Carrizo' citrangeand 'Sour' pummelo. The rate and index of drought injury was determined by field observations and principal component and subordinate function analysis was applied to analysis physiological parameters such as relative water content (RWC), the activity of superoxide dismutase (SOD), catalase (CAT), peroxydase (POD) and the content of malonaldehyde (MDA), proline (Pro) and soluble sugars (SSC). The results showed that 'Sanhuhongju' and 'Goutoucheng' had stronger drought resistance having latest time, lowest rates and indexes on drought injury. 'Sour' pummelo and 'Sanhuhuahong' had the lowest drought resistance. The order of drought resistance from the comprehensive subordinate function values was: 'Sanhuhongju' > 'Goutoucheng' > 'Carrizo' citrange > 'Choupigan' > 'Trifoliate' orange > 'Swingle' citrumelo > 'Chongyiyeju' > 'Sanhuhuahong' > 'Sour' pummelo. This study suggests that 'Sanhuhongju', a local citrus cultivar of Jiangxi province, had stronger drought resistance and thus has the potential to be exploited as drought-resistant rootstock.

Abstract:Drought affects plant growth and has become a serious problem worldwide. Populus species have different drought tolerance and sensitivity abilities. In order to evaluate the mechanism of drought stress tolerance in different cultivated Populus species distributed in China, the response to drought in the morphology and physiology of three poplars clones, NL-895, NL-351 and NL-3412 was investigated. The growth, gas exchange capacity, water use efficiency, antioxidant enzyme activity and lipid peroxidation were measured in order to evaluate differences among different genotypes in responses to two watering regimes (80% and 30% of field capacity). Our results indicated that drought stress had serious adverse effects on the seedlings as the biomass, leaf area, nett photosynthetic rate (Pn) and chlorophyll content were decreased while the activities of antioxidative enzymes were increased and the relative electric conductivity and malondialdehyde (MDA) content were enhanced. The results showed Populus possessed an effective self-protective mechanism to relieve the inhibition of growth induced by water deficit through increasing waster use efficiency, adjusting osmotic membrane and decreasing plant growth. These findings deepened our understanding of drought-tolerance mechanism in Populus.
Drought tolerance in citrus rootstocks is associated with better antioxidant defense mechanism

Abstract:Citrus is one of the major fruit crops grown worldwide. Citrus trees are affected by different abiotic stresses including drought which decrease its yield. In this study, six different citrus rootstocks (Volkameriana lemon, Brazilian sour orange, Carrizo citrange, Eureka lemon, Gada dahi, and Rangpur lime) were subjected to drought stress (24 days) and leaf relative water content (LRWC), chlorophyll ‘a’ and ‘b’, antioxidant capacity (AC), total phenolic content, and proline content (PRO) were measured. Hydrogen peroxide (H2O2), lipid peroxidation (MDA), total soluble proteins (TSP), and enzymatic antioxidant activities, such as superoxidase dismutase (SOD), catalase (CAT), and peroxidase (POD), were measured in leaves and roots of the rootstocks. Drought-stressed Volkameriana lemon had the minimum LRWC but higher SOD activity in its leaves. Carrizo citrange possessed the minimum amount of Chl ‘a’ and ‘b’ in leaves and exhibited lesser MDA and H2O2 in roots but greater TSP content in leaves and roots. SOD, CAT, POD, PRO, MDA, H2O2, and AC decreased in the leaves of Eureka lemon and Rangpur lime under drought stress. MDA and H2O2 contents were observed higher in leaves of Brazilian sour orange (a drought sensitive rootstock). However, higher SOD activity in roots and higher CAT and POD activities in leaves and roots were recorded in Carrizo citrange (a drought-tolerant rootstock). The results indicated that all the studied rootstocks were different in their defense mechanism. Tolerant rootstocks exhibited less amount of MDA and H2O2 and more antioxidant enzymatic activities (SOD, CAT, and POD) to cope with reactive oxygen species produced during drought stress.

Abstract:Seedlings of three selected Juglans rootstocks (Juglans mandshurica Maxim, Juglans nigra L. and Juglans regia L. cv. Jizhaomian) were analyzed in terms of growth, water use efficiency (WUE), chlorophyll fluorescence and photosynthetic response to drought stress. Significant differences among seedlings of the three analyzed Juglans rootstocks were observed on performance in response to water deficit. Juglans mandshurica Maxim. and Juglans regia L. cv. Jizhaomian showed larger increase in WUE and smaller reduction in growth rate, chlorophyll fluorescence and gas exchange parameters than Juglans nigra L. under drought-stressed condition. It suggests that Juglans mandshurica Maxim. and Juglans regia L. cv. Jizhaomian are more tolerant to drought stress and more suitable as walnut rootstock in water deficit regions as compared with Juglans nigra L.

Abstract:The objective of this study was to investigate the effects of drought stress on the activity of antioxidant enzymes and osmotic adjustment substance content in the tillering period of drought-sensitive and drought-tolerant rice cultivars. The results showed that the superoxide dismutase (SOD), peroxidase (POD), catalase activity (CAT), hydrogen peroxide content, soluble protein content and soluble sugar content increased with the accumulation of time and intensity of drought stress. Compared with the drought-sensitive cultivar, drought-resistant cultivar had a smaller photosynthetic affected area, longer CAT enzyme activity duration, and lower H2O2 accumulation. Unlike POD and CAT enzymes, which maintain the ability to scavenge hydrogen peroxide under long drought conditions, ascorbate peroxidase (APX) enzymes seem to be a rapid response mechanism to scavenge hydrogen peroxide under drought stress. Under a −10 kPa water potential, using soluble sugars on the osmotic adjustment ability of the drought-resistant cultivars was more efficient; under −40 kPa water potential, drought-resistant cultivars can maintain relative high levels of ascorbate (ASA) content in the short term. After the restoration of irrigation, the indices gradually returned to control levels. The ASA content showed faster accumulation ability in drought-resistant cultivars and faster recovery. The soluble protein content recovered more slowly in drought-sensitive cultivars under the −40 kPa treatment. Drought-resistant cultivars showed stronger resistance to drought in the −10 kPa treatment and obtained similar yield to the control, while the drought-sensitive cultivars were more obviously affected by the drought stress.

Abstract:Citrus plants are sensitive to waterlogging, which can cause yield reduction. Their production heavily depends on the rootstock being used for grafting of scion cultivars, and the rootstock is the first organ to be affected by waterlogging stress. However, the underlying molecular mechanisms of waterlogging stress tolerance remain elusive. In this study we investigated the stress response of two waterlogging-tolerant citrus varieties ( Citrus junos Sieb ex Tanaka cv. Pujiang Xiangcheng and Ziyang Xiangcheng), and one waterlogging-sensitive variety (red tangerine) at the morphological, physiological, and genetic levels in leaf and root tissues of partially submerged plants. The results showed that waterlogging stress significantly decreased the SPAD value and root length but did not obviously affect the stem length and new root numbers. The malondialdehyde (MDA) content and the enzyme activities of superoxide dismutase (SOD), guaiacol peroxidase (POD), and catalase (CAT) were enhanced in the roots. The RNA-seq analysis revealed that the differentially expressed genes (DEGs) were mainly linked to 'cutin, suberine, and wax biosynthesis', 'diterpenoid biosynthesis', and 'glycerophospholipid metabolism' in the leaves, whereas were linked to 'flavonoid biosynthesis', 'biosynthesis of secondary metabolites and metabolic pathways' in the roots. Finally, we developed a working model based on our results to elucidate the molecular basis of waterlogging-responsive in citrus. Therefore, our data obtained in this study provided valuable genetic resources that will facilitate the breeding of citrus varieties with improved waterlogging tolerance.

Abstract:Water deficit episodes impact apple (Malus domestica) productivity through challenging the trees' water status, the influence of extreme high temperature climate has become increasingly prominent in recent years. Rootstocks can bestow specific properties on the fruit trees such as the resistance to drought stress. However, the related hydraulic mechanisms in response to water deficit have not been fully understood. Herein, five rootstocks (SH6, GM256, M9, M26, and MM106) were examined under water limitation. The hydraulic conductance of root (Kroot), shoots (Kshoot), and stems (Kstem‐shoot) in the five rootstocks reduced slightly during drought stress. Whereas the leaf water potential and photosynthesis of five rootstocks decreased dramatically when they were exposed to drought stress. Additionally, the Kshoot and Kstem‐shoot were strongly correlated with the total plant leaf area. Aquaporins (AQPs) involved in the symplastic water transport pathway, the PIP2:1, TIP1:1, and TIP2:2 mRNA levels of all genotypic rootstocks showed significant regulation under drought stress. We examined the relationships among photosynthesis, apoplastic, and symplastic water movement pathways to achieve a comprehensive understanding of rootstocks' hydraulic strategy for improving drought adaptation. The PIP2:1 and TIP2:1 in leaves were more sensitive to root hydraulic conductance in response to drought stress. Furthermore, the coordinated relationship existed in leaf‐specific conductance of shoot (Kl‐shoot) and transpiration rate (Tr) under drought stress in the rootstocks. Overall, the drought resistance in the five dwarfing rootstocks is associated with the rapid re‐establishment of water‐related traits, and the effect of the canopy on the drought resistance in apple rootstocks merits much more attention.

[Responses of Wheat Seedlings Root Growth and Leaf Photosynthesis to Drought Stress].

Abstract:Taking drought-sensitive wheat cultivar Wangshuibai and drought-tolerance cultivar Luohan 7 as test materials, a hydroponic experiment was conducted to study the effects of drought stress on root system morphology, physiological characteristics and leaf photosynthesis of wheat seedlings, aimed to elucidate the adaptation mechanisms to drought stress. Under drought stress, the root vitality of the cultivars increased markedly, but the root number and root surface area decreased. Drought stress decreased relative water content and increased the ratio of bound water to free water in leaves of Wangshuibai, but had less effects on Luohan 7. Drought stress decreased, the leaf chlorophyll content, Pn g(s), Ci, and transpiration rate of the two cultivars, but had no significant effects on leaf chlorophyll content and Pn of Luohan 7. Drought stress decreased the leaf area of the two cultivars and the root biomass, shoot biomass, and plant biomass of Wangshuibai, but had no significant effects on Luohan 7. The results indicated that under drought stress, drought-tolerant wheat cultivar was able to compensate decreased root absorption area and retain higher root water uptake capability via enhancing root vitality and maintaining higher root biomass, and further, to keep higher leaf photosynthetic area and Pn to mitigate the inhibition of drought on wheat seedlings growth.
Different responses of photosystem II and antioxidants to drought stress in two contrasting populations of Sour jujube from the Loess Plateau, China

Abstract:Abstract Sour jujube ( Zizyphus jujuba Mill. var. spinosus (Bge.) Hu) has gained considerable attention for its adaptation to drought prone environments. To characterize the physiological and biochemical basis of this drought adaptation, the effects of drought stress on Sour jujube seedlings were investigated in a greenhouse. Two contrasting populations were employed in our study, which were from the wet (YL) and dry (SB) climatic regions in the Loess Plateau of China. Results showed that SB exhibited lower water consumption and growth inhibition, but higher water use efficiency than YL under drought stress, indicating that growth of the wet‐climate population is more sensitive to drought stress. SB exhibited higher non‐photochemical quenching (NPQ) during progressive soil drying, higher photochemical quenching (qP) during the sustained water supply stage, and higher Δ F / and qP during a re‐watering period than YL. These results further indicate that the dry‐climate population possesses better PSII efficiency under adverse conditions. YL showed larger increases in the production rate of superoxide anions, hydrogen peroxide, and hydroxyl radicals than SB during the progressive soil drying stage, indicating that SB suffered from less oxidative damage than YL. Antioxidant enzymes including catalase, ascorbate peroxidase, peroxidase and glutathione reductase, and antioxidants including carotenoids, flavonoids and proline; when these interact, they contribute greatly to the antioxidant capacity of the dry‐climate population. Taken together, the better photosynthetic potential and antioxidant capacity contribute to the better performance of Sour jujube from the dry‐climate, providing useful information for understanding the drought tolerance mechanisms of Sour jujube.

Morphological and Physiological Responses of Melia azedarach Seedlings of Different Provenances to Drought Stress

Abstract:Melia azedarach Linn. is a deciduous tree of the Melia genus in the Meliaceae family that is native to China. To study the mechanism of drought resistance in Melia azedarach and evaluate the drought resistance capacity of each provenance, we selected eight provenances (Shandong Kenli, Jiangsu Pizhou, Hubei Shayang, Jiangsu Xuanwu, Jiangxi Xihu, Jiangsu Jurong, Guangdong Luogang, and Henan Shihe) as the research subjects and set four levels of drought stress treatment (CK: 75% of field capacity, mild drought: 60% of field capacity, moderate drought: 45% of field capacity, and severe drought: 30% of field capacity). The results showed that the growth in the seedling height and the ground diameter, the leaf relative water content, transpiration rate (Tr), net photosynthetic rate (Pn), stomatal conductance (Gs), and the content of chlorophyll (Chl) decreased with the increasing stress levels, while the root–shoot ratio, water saturation deficit, and the contents of malondialdehyde (MDA) increased. The SOD in most provenances initially increased and then decreased, reaching a peak during moderate drought. At the late stage of treatment, the magnitude of the changes in the photosynthetic indicators was more pronounced than in the physiological indicators. Principal component analysis showed that the contribution of all four principal components under the three drought stresses was above 85%, which represented the majority of the original data. Combined with the affiliation function method and weights, the comprehensive evaluation value (D value) of the drought resistance was calculated for the eight provenances. Then, we obtained the order of drought resistance of the test materials under the three drought stresses, respectively. The combined results revealed that the drought resistance of Henan Shihe and Jiangxi Xihu was stronger, while the drought resistance of Guangdong Luogang and Hubei Shayang was weaker. Based on the above findings, we can select provenances with strong and weak drought resistance for transcriptome sequencing to screen drought-resistant genes for an in-depth study at the molecular level.

Abstract:The compounds of rootstock and scion might be a mechanism for resistance to the environmental stresses such as drought in almonds. Considering photosynthetic rate (PN), stomatal conductance (gs), transpiration rate (E), sub-stomatal CO2 (Ci), mesophyll conductance (gm), leaf surface temperature (LST), fluorescence variable/fluorescence maximum (Fv/Fm) and water use efficiency (WUE) in some commercial almond cultivars grafted on different rootstocks, a factorial study based on randomized complete block design with three replications was carried out to explore the most resistant compound. The factors included cultivars in 5 levels ('Supernova', 'Texas', 'Marcona' 'Shokoufeh' and 'K13-40'), rootstocks in 3 levels [(Peach, Almond hybrids 'GF677', 'GN22' and 'seedling bitter almond No.32'] and drought stress in 4 levels (No: − 0.1 MPa (NS), Moderate: − 0.7 MPa (MS), Severe: − 1.1 MPa (SS) and very severe: Ψt ≤ − 1.8 MPa (VSS) of soil water potential (Ψt) (3, 5, 10 and 15 days after irrigation, respectively). The results revealed that increasing drought stress leads to a decrease in the Fv/Fm. Related to the control (0.808) and under the VSS stress, the lowest and highest decreases in Fv/Fm were observed in the "Sh" on 'GF677' (0.789) and 'Texas' on 'No.32' rootstock (0.667), respectively. Moreover, drought stress reduced gs and E, but increased gm, Ci and LST. In essence, all cultivars, especially 'Sh' grafted on 'GF677' rootstock, were more resistant to the VSS in comparison with the compound of the same cultivars on the 'No.32' and 'GN22' rootstocks, noticing that rootstock-scion compound was superior in this study.
Different Adaptive Responses of Leaf Physiological and Biochemical Aspects to Drought in Two Contrasting Populations of Seabuckthorn

Abstract:Two contrasting populations of seabuckthorn (Hippophae rhamnoides L.) from western China were employed to study their differences in adaptive responses to drought. The Daofu population was from a wetter upland climate region, whereas the Dingxi populations was from a drier lowland climate region. A completely randomized design with two factors, two populations and two watering regimes (100% and 25% of the soil water holding capacity), was used. In both populations, drought significantly decreased growth and the net photosynthesis rate (A), and significantly increased the root/shoot ratio (RS), catalase (CAT), peroxidase (POD), glutathione peroxidase (GPX) and ascorbate peroxidase (APX) activities, and abscisic acid (ABA) and proline contents. Compared with the Daofu population, drought induced a greater RS value, higher CAT, GPX, and APX activities, and a higher ABA content in the Dingxi population, whereas the gas exchange traits, for example, the stomatal limitation value (L-S) and intercellular CO2 concentration (C-i), were less responsive to drought in the Dingxi population. The two populations may have developed different strategies to tolerate drought, such as different pathways to dissipate excess absorbed light energy, to resist oxidative stress, and to keep water status. Such factors enable the Dingxi population to tolerate drought better than the Daofu population.
LEAF PHYSIOLOGICAL AND ANATOMICAL CHARACTERISTICS OF TWO INDICATOR SPECIES IN THE LIMESTONE REGION OF SOUTHERN CHINA UNDER DROUGHT STRESS

Abstract:Drought is a critical limiting factor of plant growth, and plants living in arid areas must develop adaptation mechanisms to resist drought stress. In this study, we studied the drought resistance mechanisms of two tree species, Triadica rotundifolia and Cinnamomum burmannii, based on their foliar physiological and anatomical characteristics. These trees are indicator plants in the limestone region of southern China. We evaluated and compared the changes in the contents of chlorophyll, soluble protein, soluble sugars, proline, malondialdehyde (MDA), proline, activity of superoxide dismutase (SOD) and peroxidase (POD), chlorophyll fluorescence parameters, and leaf anatomical structure between these two species. The results showed the following. (1) The content of Chla+b, soluble protein, soluble sugar and SOD activity of the two species’ seedlings had an increasing trend at first and then a decrease over the entire drought experiment. The change in MDA content was more sensitive in T. rotundifolia, and the peak (33.05 mmol·ml) appeared on the 28 day of the drought experiment. Comparatively, the proline content and POD activity were more sensitive in C. burmannii, and the proline peak (217.11 μg·g) appeared on the 24 day. (2) Under the continuous drought stress, the electron transport rate of the seedlings decreased, whereas the non-photochemical quenching showed different trends for the different species. Severe drought stress in T. rotundifolia caused an increasing trend in the light energy capture efficiency (Fv/Fm), while it declined continuously in C. burmannii. (3) Under the drought stress, the leaves of T. rotundifolia became thicker. The arrangement of spongy tissues became loose, and the ratio of palisade to spongy tissue decreased. However, the leaves of C. burmannii showed a few different features, such as inconspicuous differentiation between palisade and spongy tissues, a high loose degree of leaves, and low packing. (4) The leaf physiological indices had a strong relationship with the anatomy characteristics, as revealed by principal component analysis (PCA); the adaptability to drought tolerance of T. rotundifolia was stronger than C. burmannii. In conclusion, T. rotundifolia is better able to adapt to drought conditions due to its physiological and anatomical characteristics, which allow the species to better buffer drought-induced physiological responses and variability in leaf structure.

Influence of rootstock on drought response in young 'Gale Gala' apple (Malus domestica Borkh.) trees

Abstract:Background: Drought is a major environmental stress limiting plant growth, productivity, and survival worldwide. Rootstocks are widely used to enhance plants resistance to drought stresses. This study determined influence of rootstock on drought responses in 1-year-old 'Gale Gala' apple trees grafted onto Malus sieversii or M. hupehensis. Results: Choice of rootstock resulted in differential response to drought stress. Specifically, M. sieversii caused less drought-induced reduction in relative growth rate, biomass accumulation, leaf area, leaf chlorophyll content, relative water content, photosynthesis rate and maximum chlorophyll fluorescence yield but greater increase in whole-plant water use efficiency compared to M. hupehensis. Secondly, compared with M. hupehensis, M. sieversii caused less drought-induced accumulation of reactive oxygen species but more increase in activities of antioxidant enzymes. In addition, xylem sap abscisic acid concentration was greater in trees grafted onto M. hupehensis than in those grafted onto M. sieversii under drought stress. Conclusion: 'Gale Gala' trees' response to drought stress was associated with the rootstock's genotype onto which it was grafted. Trees with M. sieversii as rootstock are more drought resistant than trees with M. hupehensis as rootstock, which suggests that M. sieversii can be widely used as rootstock in arid and semi-arid regions.

Abstract:We examined tolerance to soil drying in clonally propagated apple (Malus domestica Borkh.) rootstocks used to control shoot growth of grafted scions. We measured leaf conductance to water vapor (g(L)) and leaf water potential (Psi(L)) in a range of potted, greenhouse-grown rootstocks (M9, M26, M27, MM111, AR69-7, AR295-6, AR360-19, AR486-1 and AR628-2) as the water supply was gradually reduced. Irrespective of the amount of available water, rootstocks that promoted scion shoot growth (M26 and MM111) generally had higher g(L) and more negative Psi(L) than rootstocks that restricted scion shoot growth (M27 and M9). After about 37 days of reduced water supply, there were significant decreases in g(L) and Psi(L) in all rootstocks compared with well-watered controls. In all treatments, the slope of the relationship between log (g(L)) and Psi(L) was positive, except for rootstocks AR295-6, AR628-2 and AR486-1 in the severe-drought treatment, where the drought-induced change in the relationship suggests that rapid stomatal closure occurred when leaf water potentials fell below -2.0 MPa. This drought response was associated with increased root biomass production. Rootstock M26 showed little stomatal closure even when its water potential fell below -2.0 MPa, and there was no effect of drought on root biomass production. We conclude that differences among rootstocks in the way that g(L) and Psi(L) respond to drought reflect differences in the mechanisms whereby they tolerate soil drying. We suggest that these differences are related to differences among the rootstocks in their ability to control shoot growth.

Abstract:Abstract G. sinensis is a crucial tree species in China, possessing important economic and ecological value, and having a wide geographical distribution. G. sinensis seedlings is highly vulnerable to the drought-rehydration-drought cycle during their growth, and there is a lack of quantitative and systematic research on the physiological mechanisms of drought resistance and rehydration in G. sinensis . There is also a lack of good drought-resistant families and reliable methods for evaluating drought resistance, which severely hinders the selection and promotion of drought-resistant G. sinensis families and the industry's development. Therefore, this study selection 58 families seedlings of G. sinensis to drought stress and rehydration using an artificial simulated water control method in potted seedlings. The aim was to compare the effects of different levels of drought and rehydration on the growth and physiological indices of seedlings from different families. Identification of drought-resistant families and dependable drought related indices and techniques, the explanation of divergence in drought stress effects on various drought-resistant seedlings and the mechanisms underpinning growth and physiological responses, and the provision of theoretical reference for G. sinensis drought-resistant variety selection and cultivation. The Drought Resistance Index (DRI) served as the primary indicator, supplemented by growth, leaf morphology, and photosynthetic physiological indicators, to thoroughly assess and identify five distinct drought tolerant taxa while also selecting five representative families. Soluble protein (SP), proline (Pro), and malondialdehyde (MDA) contents, as well as the activities of catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) in seedlings from the five families, increased as the degree of drought intensified. The highest values were appeared during periods of severe drought, and gradually decreased after subsequent rehydration. Principal component analysis (PCA) revealed MDA and soluble sugars (SS) as the primary predictors of drought and rehydration response in G. sinensis seedlings respectively. Changes in osmoregulatory substance content and increased antioxidant enzyme activity may be crucial for responding to drought tolerance mechanisms. Leaf morphological indicators, seedling height, soil plant analysis development (SPAD) value, photosynthetic indicators, and MDA are dependable parameters for assessing the drought tolerance of G. sinensis seedlings. When assessing the drought-resistance of seedlings using physiological indicators such as photosynthesis, a comprehensive analysis should incorporate multiple indicators and methods. This evaluation approach could serve as a reference for screening exceptional drought-resistant families of G. sinensis .

Physiological and PIP Transcriptional Responses to Progressive Soil Water Deficit in Three Mulberry Cultivars

Abstract:Although mulberry cultivars Wubu, Yu711, and 7307 display distinct anatomical, morphological, and agronomic characteristics under natural conditions, it remains unclear if they differ in drought tolerance. To address this question and elucidate the underlying regulatory mechanisms at the whole-plant level, 2-month old saplings of the three mulberry cultivars were exposed to progressive soil water deficit for 5 days. The physiological responses and transcriptional changes of PIPs in different plant tissues were analyzed. Drought stress led to reduced leaf relative water content (RWC) and tissue water contents, differentially expressed PIPs, decreased chlorophyll and starch, increased soluble sugars and free proline, and enhanced activities of antioxidant enzymes in all plant parts of the three cultivars. Concentrations of hydrogen peroxide (H2O2), superoxide anion (O2•−), and malonaldehyde (MDA) were significantly declined in roots, stimulated in leaves but unaltered in wood and bark. In contrast, except the roots of 7307, soluble proteins were repressed in roots and leaves but induced in wood and bark of the three cultivars in response to progressive water deficit. These results revealed tissue-specific drought stress responses in mulberry. Comparing to cultivar Yu711 and 7307, Wubu showed generally slighter changes in leaf RWC and tissue water contents at day 2, corresponding well to the steady PIP transcript levels, foliar concentrations of chlorophyll, O2•−, MDA, and free proline. At day 5, Wubu sustained higher tissue water contents in green tissues, displayed stronger responsiveness of PIP transcription, lower concentrations of soluble sugars and starch, lower foliar MDA, higher proline and soluble proteins, higher ROS accumulation and enhanced activities of several antioxidant enzymes. Our results indicate that whole-plant level responses of PIP transcription, osmoregulation through proline and soluble proteins and antioxidative protection are important mechanisms for mulberry to cope with drought stress. These traits play significant roles in conferring the relatively higher drought tolerance of cultivar Wubu and could be potentially useful for future mulberry improvement programmes.

Abstract:Drought stress is one of the major threats to rice production. The weakening of leaf photosynthesis due to drought is the main reason for the reduction of grain yield, but its mechanism is still obscure. The objectives of this study were to reveal the physiological mechanism of drought stress affecting photosynthetic capacity and grain yield. Pot experiments were conducted with drought-tolerant cultivars Hanyou113 (HY113) and Zhonghan3 (ZH3) and drought-sensitive cultivar Huanghuazhan (HHZ) under four water management treatments (traditional flooding (CK), mild drought stress (LD), moderate drought stress (MD) and severe drought stress (HD)) at heading stage in 2013 and 2014. Compared with CK, grain yield was significantly reduced by 14.9%, 30.8%, and 12.8% in HY113, HHZ, and ZH3 under LD, 32.9%, 33.7%, and 22.9% in HY113, HHZ and ZH3 under MD and 53.6%, 45.6%, and 30.7% in HY113, HHZ, and ZH3 under HD, respectively. The photosynthetic rate (Pn) decreased by 49.0% from 20.0 to 10.2 µmol m−2 s−1 in HY113, and 67.6% from 23.4 to 7.58 µmol m−2 s−1 in HHZ, and 39.3% from 23.4 to 14.2 µmol m−2 s−1 in ZH3 under HD. The Pn of HHZ was similar to that of ZH3 under CK conditions. During the drought periods from LD to HD at heading stage, the leaf water potential (LWP) reduced 31.9%, 54.8%, and 15.7% in HY113, HHZ, and ZH3, respectively. The non-photochemical quenching (NPQ) of HY113, HHZ, and ZH3 flag leaves increased by 150%, 97.6%, and 218%, respectively. The effective quantum yield of PSII photochemistry (ΦPSII) of flag leaves reduced by 20.3%, 11.9%, and 22.1% in HY113, HHZ, and ZH3, respectively. The enzymatic activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) increased by 11.4%, 18.0%, and 21.8% in HY113, and 13.2%, 14.3%, and 30.9% in HHZ, and 13.4% 21.7%, and 17.6% in ZH3 under MD on average across two seasons. The yield reduction of drought-resistant cultivars (HY113, ZH3) was smaller than that of conventional cultivars (HHZ). Maintaining leaf water potential (LWP), Pn, photosystem II (PSII) original light energy conversion efficiency, non–photochemical quenching coefficient (NPQ), and increasing in the ratio of photochemical reaction energy in fluorescence and antioxidant enzyme activity is the physiological basis to achieve a relatively high photosynthesis. These traits could be the target for breeder to develop drought-tolerant varieties.

Abstract:Take the seedlings of Cirsium japonicum as test materials, we studied the influence of relative water content (RWC), malondialdehyde (MDA) content, osmotic regulation substance contents and protective enzyme activities of leaves with PEG-6000 gradient concentrations(5%, 10%, 15%, 20%, 25%, 30%) under the stress time of 24 h, 48 h and 72 h to study the drought resistance and drought resistance mechanism preliminarily in the paper.The results showed that:(1)at stress of 24 h, 48h and 72 h,with the extension of stress time and the increase of treatment concentration, the relative water content showed a trend of decrease with the sharpest drop of 55.86%, while the MDA content significantly increased with the biggest increase of 186.21%.(2)With the extension of stress time, the soluble sugar and free proline contents of leaves increased under PEG-6000 ≤10%,while showed decrease trend after the first increase, and with the increase of treatment concentration the two indexes increased firstly and decreased afterward.The peak value of soluble sugar content appeared orderly at stress of 24 h, 48 h and 72 h on 20%, 20%,10% of PEG-6000 concentration, and the free proline appeared orderly on 20%, 15%, 15% of PEG-6000 concentration.At the stress 48 h, the above two indexes get the maximum increased nearly 4.7 times and 10.7 times on 20% and 15% of PEG-6000 concentration respectively compared with CK.(3)Expect SOD activity increased at stress 24 h, the protective enzyme activities all showed a trend of increase firstly and then decreased.The maximum increase was 370.14%, 248.91%and 118.78% respectively for SOD, POD and CAT activities.SOD and POD both appeared on stress 72h, 15% of PEG-6000 concentration, while CAT appeared on 48 h, 10% of PEG-6000 concentration.Above all, the C.japonicum has a stronger osmotic adjustment ability and higher enzyme activity with 15% of PEG-6000 concentration on a long time (72 h) stress, which shows a strong ability to drought tolerance.If over the concentration, the osmotic adjustment ability would be down, the enzyme activity weaken, the relative water content continuously reduced and the MDA content continuously increased.The physiological metabolism of C.japonicum was restrained.
