Abstract:Polyethylene glycol (PEG) has found tremendous applications in pharmaceutical products and has played a critical role in PEGylated drug modalities to improve pharmacokinetic properties and biological efficacy. The characterization and quantitation of PEGs are essential to control manufacture process and drug product quality. However, the assay value of PEG could change dramatically depending on the structures of the PEG and the detection techniques used. In this study, we developed a size exclusion chromatographic (SEC) method for quantitative PEG analysis, and we systematically evaluated the performance of three online detectors with different operating principles: a charged aerosol detector (CAD), a differential refractive index (dRI) detector, and a multi-angle light scattering detector (MALS). Fourteen PEG compounds covering a wide range of molecular weight (MW, 1 - 40 kDa) and molecular architectures (linear, branched, Y-shaped and multi-arm geometries) were evaluated by these three detection techniques. Our study revealed that the dRI showed the most universal responses among all the PEGs regardless of their molecular weight or geometries. In the contrast, CAD and MALS detector showed MW-dependent and semi-universal geometry-dependent responses. Another key finding is that the relative response factor for each multi-arm PEG in the CAD and the MALS were inversely correlated, suggesting both can be applied to qualitatively assess polymers of different architectures, including the ones with subtle differences in their core structures. The comparison of the three detectors not only provides the fundamental and comprehensive understanding of PEG quantitation but also enables the process development and control of high-quality PEGs in producing PEGylated therapeutics in the pharmaceutical industry.

Abstract:Polyethermethylsiloxanes (PEMSs) are silicone based polymers formed by attaching one or more ethylene oxide (EO)/propylene oxide (PO) chains to a siloxane chain. As the siloxane chain is lengthened, the polarity of the PEMSs are reduced. Little research has been conducted on the use of ultra-high performance liquid chromatography (UHPLC)-mass spectrometry (MS) to analyze PEMS oligomers with more than 4 siloxane groups because of their low polarity and poor solubility in water and acetonitrile (ACN). In this study, we developed a high chromatographic resolution method for PEMS and polyether oligomers using the water-ACN-isopropanol (IPA) tertiary solvents gradient. PEMSs oligomers containing one EO pendant chain and 3-13 silicone groups were analyzed. More than 102 PEMS oligomers and 21 polyether oligomers were separated within 45 min and identified by accurate molecular masses. During this gradient elution process, different chromatographic modes were used: both precipitation-redissolution and adsorption mode for PEMSs, adsorption mode for EO chains in poly-ethers, and exclusion mode for EO chains in PEMSs. This efficient separation method for PEMSs would broaden the characterization of silicone surfactants. Also, it is beneficial to establish the relationship between surfactant structure, synthetic route and performance optimization. (C) 2019 Elsevier B.V. All rights reserved.

Abstract:Mono-d-alpha-tocopheryl polyethylene glycol 1000 (TPGS 1000) and di-TPGS 1000 were prepared from the synthesized TPGS 1000 mixture. The key separation step was performed by a Simulating Moving Bed chromatographic process. The chemical structures and molecular weight distrubution were characterized by H-1-NMR and MALDI-TOF mass spectroscopy. NMR and MALDI-TOF MS data confirmed the occurrence of di-TPGS. Both NMR and MALDI-TOF MS results showed the degree of polymerization of the two esters and the molecular mass. The melting temperatures of the two polymers were investigated by DSC and the thermal decomposition temperatures have been determined by TGA. The melting temperatures of the two esters were 33 and 15 degrees C, separately. And the two separated TPGS esters exhibited different thermal decomposition courses. (C) 2010 Wiley Periodicals, Inc. Abstract:Water-soluble polymers (WSPs) are additives used as thickeners, stabilisers and flocculants in industry and in household products, including personal care products. Given their widespread use, it is likely WSPs enter the environment, particularly through wastewaters. This is of concern as there is little ecotoxicological research on their fate and behaviour once in the environment, which means their risk to aquatic life is not understood. The lack of suitable analytical techniques to detect, characterise and quantify WSPs hinders research on the potential impact of these polymers. A novel method has been developed that identifies polymers within a sample and separates them using gel-permeation chromatography (GPC). This is coupled with matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOF MS), to quantify the polymer fractions using molecular weight information. This process has been carried out on a range of aqueous media. Polyethylene glycol (PEG) ingredients were successfully separated from non-polymeric material in a commercial shaving gel personal care product (PCP), before being quantified at 1.62 wt%. This method was applied to a spiked wastewater influent sample to demonstrate the extraction and separation of PEG from organic constituents such as dissolved organic matter (DOM). This highlighted the additional challenges of analysing WSPs in the environment, as factors such as sorption and biodegradation affected the total recovery of PEG, with an extraction efficiency of 53%. Overall, this method was applied for the extraction of PEG from a PCP with accurate quantification, before a proof-of-concept extraction from wastewater demonstrated the difficulties associated with WSP analysis in environmental samples. This method provides opportunities to use tandem GPC/MALDI-TOF MS to quantify WSPs in a broad array of environmental samples. Additional studies could include its application to wastewater or freshwater monitoring.

Abstract:The characterization of the microstructure of in vivo degradable polyesters is gaining increased interest thanks to their high-performance applications, such as drug delivery systems. The design of such material requires a high level of understanding of the critical material attributes of the polyesters, such as molecular-weight distribution (MWD), chemical-composition distribution (CCD), and end-groups (functionality-type distribution, FTD). Size-exclusion chromatography (SEC) hyphenated with mass spectrometry (MS) yields SEC-MS, which is an effective method for analyzing the microstructure of polymers. While the MWD can be determined by size-exclusion chromatography hyphenated with ultraviolet spectrometry and refractive index (SEC-UV/RI), the CCD and FTD can be determined by SEC-MS. However, previous applications of SEC-MS have failed to address the risk of polymer fragmentation during the analysis process. It is crucial to establish whether SEC-MS methods can be applied to biodegradable polymers and to recognize if fragmentation processes occurred during the SEC separation or during the ESI-MS process. In this study, we investigate whether SEC-MS methods can be applied to PLGA biodegradable polyesters. The research demonstrates that the choice of alkali metal salt used during ionization strongly influences the stability of PLGA during SEC-MS analysis. CsI was found to minimize fragmentations during ESI-MS, simplifying the MS spectra and allowing isomeric PLGA structures to be distinguished. The resulting method facilitates end-group and CCD determination. Additionally, when combined with selective degradation, the described method can provide insights in the “blockiness” of the polymer and support the development of sequence-controlled PLGA synthesis.

Abstract:PEGylated lipids such as 1,2-distearoyl-glycero-3-phosphatidylethanolamine-N-methoxy-polyethyleneglycol-2000 (DSPE-mPEG) are essential components of lipid nanoparticle formulations. They have an impact on the efficacy and quality of the drug products they used in, and as such should be carefully characterized. The present work used liquid chromatography with charged aerosol detection and high resolution mass spectrometry to investigate the polydispersity and purity of several lots of DSPE-mPEG, revealing significant differences in the level and identity of impurities. Additionally, mass spectral data showed that there were variations in the polyethylene glycol chain length between lots of comparable purity, which could not be detected by charged aerosol detection alone. The obtained results may be of importance in the development of new lipid nanoparticle based drugs.

Abstract:The solubility of drugs in polyethylene glycol 400 (PEG 400) was estimated and rank ordered using a differential scanning calorimetry (DSC) method and the Fox Equation. Drug-polymer binary mixtures of six compounds (Ibuprofen, Indomethacin, Naproxen, and three proprietary compounds: PC-1 through PC-3) with PEG 400 were heat treated using a three-cycle DSC method to establish a correlation between equilibrium solubility and temperature. Thermal events such as heat of fusion, heat of recrystallization and glass transition temperature, T(g), were used to calculate the drug solubility at multiple higher temperatures through the Fox Equation. Subsequently, a van't Hoff plot was constructed to estimate the drug solubility at room temperature, and the values were compared with those measured by HPLC. With the exception of Naproxen, room temperature solubilities of the remaining drug compounds in PEG 400 were determined by this thermal method approach, and compared with those measured by HPLC: 26.7% vs. 24.7% for Ibuprofen, 5.8% vs. 9.6% for Indomethacin, 3.1 % vs. 1.5% for PC-1, 2.3% vs. 1.3% for PC-2, and 1.4% vs. 0.2% for PC-3 in PEG 400. There was good concordance in solubility rank order estimates between the two methods. These collective results support the potential utility of the thermal method as an alternative to other methods for estimation of drug solubility in Polymers which is an important determinant in the design of physically-stable amorphous systems.

Abstract:Background: Polyesters are applied in high-end products in many industrial applications, including resins and powder-coating applications. The characterization of the chemical heterogeneities within a polyester is of utmost interest to develop new products or improve existing applications. Unfortunately, characterization is a difficult task, as polyesters may feature distributions in end-group functionality, molecular weight, chemical composition, and degree of branching. Currently, no analytical method can characterize all these interdependent distributions in a single analysis. Results: We report the use of comprehensive normal-phase liquid chromatography × size-exclusion chromatography hyphenated with ultraviolet-light spectroscopy and high-resolution mass spectrometry in parallel (NPLC × SEC-UV/HRMS) to characterize polyesters according to their end-group-functionality and molecular-weight distributions. The chemical composition can be measured with HRMS, while relative quantitation can be performed with UV detection. A supercharging agent was used during ionization allowing to extend the molecular-weight range of the detected chemical species. Significance: The presented platform allows characterization of polyesters with varying fractions of carboxyl or hydroxyl end-group functionalities and varying distributions of molecular weight, degree of branching, and chemical compositions. The number-average and weight-average molar masses are obtained in the same analysis. This information cannot be obtained by any one-dimensional technique. The developed NPLC × SEC-UV/HRMS platform is a valuable tool for characterizing polyesters in an industrial setting.
Comprehensive ethoxymer characterization of complex alcohol ethoxy sulphate products by mixed-mode high-performance liquid chromatography coupled to charged aerosol detection

Abstract:The present work describes a simultaneous mixed-mode high performance liquid chromatography (HPLC) method combined with a universal and non-selective-response detector for the complete ethoxymer profiling of alcohol ethoxy sulphate mixtures. The optimized HPLC methodology combines the dual hydrophilic (HILIC) and reversed-phase selectivity of a surfactant-type column in order to render a comprehensive and simultaneous separation of more than 50 endogenous ethoxymers in a single analysis. Furthermore, an accurate quantitation of every single analyte was achieved using a final universal charged aerosol detector (CAD) including specific mathematical processing tools. Results obtained helped describing a complete alkyl chain and ethoxymer distribution of the investigated AES samples. Method validation evidences provided reliability of the individual ethoxymer contributions determined with the proposed HPLC-CAD methodology. Regarding accuracy including independent nuclear magnetic resonance (NMR) experiments, an excellent correlation was found between the structural information provided by a COSY NMR spectrum and the CAD results regarding the mono/polyethoxylated and the non-ethoxylated/ethoxylated distribution. Additional calculations including the average molecular weight and the degree of ethoxylation for the reference AES sample showed minimum differences (relative error < 1 %) between the two considered techniques. An outstanding precision and linearity along the working concentration range (r2>0.999) was also observed. The individual limit of detection for the target sulphate ethoxymers was determined to be in the low ppm range. Further validated distribution profiles for a large number of AES samples demonstrated the applicability of the optimized HPLC-CAD methodology to routine surfactant screenings. Therefore, the hereby developed methodology provided extensive information regarding the detailed individual ethoxymer profile of AES formulations, which can be extremely useful for the surfactant industry in order to gain information on specific synthesis routes and/or detergency properties.

Abstract:Polyethylene glycols are synthetic polymers composed of repeating oxyethylene subunits, which have been known for non-toxic, non-immunogenic, non-antigenic, good solubility in water and therefore approved for pharmaceutical applications. Recently, attachment or amalgamation of polyethylene glycols to therapeutic small molecules, peptides, proteins, or nanoparticles has become a mature technology for the sake of improving their pharmacokinetic and pharmacological profiles, also referred to as PEGylation. By comparison, there are only a few PEGylated pharmaceuticals have been registered for further clinical trials and even less was approved for marketing. High failure rate of PEGylated pharmaceuticals in pre-clinical and clinical trials could be majorly attributed to their unclear pharmacokinetic behaviors. Therefore, the in vivo fate of the PEGylated pharmaceuticals for the various routes of administration needs to be thoroughly investigated An accurate in vivo pharmacological study thereof highly depends on the precise detection of polyethylene glycols as well as their fragments in biological matrixes. The goal of this review is to highlight the analytical methods that were developed and applied to evaluate the polyethylene glycols in pharmaceutical ingredients and excipients, which bring us closer to bridging the gap between the development of polyethylene glycol-based drug delivery systems and their clinical application.
Development of a Robust Method for Simultaneous Quantification of Polymer (HPMC) and Surfactant (Dodecyl β-D-Maltoside) in Nanosuspensions

Abstract:This report describes the development of a chromatographic method for the simultaneous quantification of a polymer, hydroxypropyl methylcellulose (HPMC), and a surfactant, dodecyl β-D-maltoside (DM), that are commonly used in the physical stabilization of pharmaceutical formulations such as nanosuspensions and solid dispersions. These excipients are often challenging to quantify due to the lack of chromophores. A reverse phase size exclusion chromatography (SEC) with evaporative light scattering detector (ELSD) technique was utilized to develop an accurate and robust assay for the simultaneous quantification of HPMC and DM in a nanosuspension formulation. The statistical design of experiments was used to determine the influence of critical ELSD variables including temperature, pressure, and gain on accuracy, precision, and sensitivity of the assay. A robust design space was identified where it was determined that an increase in the temperature of the drift tube and gain of the instrument increased the accuracy and precision of the assay and a decrease in the nebulizer pressure value increased the sensitivity of the assay. In the optimized design space, response data showed that the assay could quantify HPMC and DM simultaneously with good accuracy, precision, and reproducibility. Overall, SEC-ELSD proved to be a powerful technique for the simultaneous quantification of HPMC and DM. This technique can be used to quantify the amount of HPMC and DM in nanosuspensions, which is critical to understanding their effects on the physical stability of nanosuspensions.

Abstract:Polyether polyols are often used in formulated systems, but their complete characterization is challenging, because of simultaneous heterogeneities in chemical composition, molecular weight and functionality. One-dimensional liquid chromatography-mass spectrometry is commonly used to characterize polyether polyols. However, the separation power of this technique is not sufficient to resolve the complexity of such samples entirely. In this study, comprehensive two-dimensional liquid chromatography hyphenated with high-resolution mass spectrometry (LC × LC-HRMS) was used for the characterization of (i) castor oil ethoxylates (COEs) reacted with different mole equivalents of ethylene oxide and (ii) a blended formulation consisting of glycerol ethoxylate, glycerol propoxylate and glycerol ethoxylate-random-propoxylate copolymers. Retention in the first (hydrophilic-interaction-chromatography) dimension was mainly governed by degree of ethoxylation, while the second reversed-phase dimension resolved the samples based on degree of propoxylation (blended formulation) or alkyl chain length (COEs). For different COE samples, we observed the separation of isomer distributions of various di-, tri- and tetra-esters, and such positional isomers were studied by tandem mass spectrometry (LC-MS/MS). This revealed characteristic fragmentation patterns, which allowed discrimination of the isomers based on terminal or internal positioning of the fatty-acid moieties and provided insight in the LC × LC retention behavior of such species.

Abstract:Polymer conjugation has risen in importance over the past three decades as a means of increasing the in vivo half-life of biotherapeutics, with benefits including better stability, greater drug efficacy, and lower toxicity. However, the intrinsic variability of polymer synthesis results in products with broad distributions in chain length and branching structure, complicating quality control for successful functionalization and downstream conjugation. Frequently, a combination of several analytical techniques is required for comprehensive characterization. While liquid chromatography-mass spectrometry (LC-MS) is a powerful platform that can provide detailed molecular features of polymers, the mass spectra are inherently challenging to interpret due to high mass polydispersity and overlapping charge distributions. Here, by leveraging Fourier transform-based deconvolution and macromolecular mass defect analysis, we demonstrate a new way to streamline pharmaceutical polymer analysis, shedding light on polymer size, composition, branching, and end-group functionalization with the capability for reaction monitoring.

Abstract:A majority of drugs marketed in amorphous formulations have a good glass-forming ability, while compounds less stable in the amorphous state still pose a formulation challenge. This work explores ternary solid dispersions of two model drugs with a polymer (i.e., hydroxypropyl cellulose) and a coformer as stabilizing excipients. The aim was to introduce a computational approach by preselecting additives using solubility parameter intervals (i.e., overlap range of solubility parameter, ORSP) followed by more advanced COSMO-RS theory modeling. Thus, a mapping of calculated mixing enthalpy and melting points is proposed for in silico evaluation prior to hot melt extrusion. Following experimental testing of process feasibility, the selected formulations were tested for their physical stability using conventional bulk analytics and by confocal laser scanning and atomic force microscopy imaging. In line with the in silico screening, dl-malic and l-tartaric acid (20%, w/w) in HPC formulations showed no signs of early drug crystallization after 3 months. However, l-tartaric acid formulations displayed few crystals on the surface, which was likely a humidity-induced surface phenomenon. Although more research is needed, the conclusion is that the proposed computational small-scale extrusion approach of ternary solid dispersion has great potential in the formulation development of challenging drugs.
Separation of Octylphenol Polyether Alcohols Surfactants by Capillary Column SFC and HPLC

Abstract:Separation of nonionic octylphenol polyether alcohols (OPA) by supercritical fluid chromatography (SFC) and HPLC is described. Using a density programming and a 50-μm i.d. capillary column, a total of 18 group oligomers was separated. The effects of the operating parameters, such as temperatures of mobile phase, flame ionization detector (FID) and injector, on SFC analysis were investigated. The results demonstrated that the separation efficiency is better when mobile phase temperature is above 100 °C, which must be due to the higher volatility of the oligomers. The results also demonstrated that the change of FID temperature from 300 °C to 420 °C has no effect on the total peak response. The study shows that injector cooling is not necessary when the analyte is in methanol. The total peak response was lower when the injector temperature was changed from 21 °C to 10 °C. A group of polar oligomers was found in OPA in HPLC analysis, not found in SFC. The result indicates that the polar oligomers present in OPA were not soluble in supercritical fluid carbon dioxide and, therefore, not observed in SFC.

Abstract:In this study, one aqueous two-phase system (ATPS) composed of polymer and salt was employed to extract sulfated polysaccharides (sulfated dextran) to polymer-rich phase from model polysaccharides mixture (dextran and sulfated dextran), and another ATPS was employed to back-extract sulfated polysaccharides to a salt phase. This process was further applied to separate sulfated Enteromorpha polysaccharides (EPs) from crude EPs. For optimal PEG1000/(NH4)2SO4 (TLL45) ATPS, the extraction efficiencies were 93.2 ± 0.1%, 95.4 ± 2.0%, 56.3 ± 4.4% for 5, 50, 500 kDa sulfated dextran in top PEG-rich phase and were 91.9 ± 1.4%, 97.1 ± 2.9%, 95.2 ± 0.9% for 5, 50, 500 kDa dextran in bottom salt-rich phase, respectively. The 500 kDa sulfated dextran presented precipitation at the interface, and the extraction efficiency was 26.9 ± 5.7% for 500 kDa sulfated dextran in the interface. Different ATPSs composed of PEG1000 and another salt (sodium citrate, K2HPO4, sodium tartrate, etc.) were performed to back-extract sulfated polysaccharides of PEG-rich phase to another salt phase. Under the PEG1000/sodium tartrate ATPS, the best back-extraction efficiencies of 5, 50, 500 kDa sulfated dextran were 75.1 ± 4.1%, 86.1 ± 0.2% and 88.3 ± 1.6%, respectively. Crude EPs was extracted by ultrasonic/microwave-assisted extraction from Enteromorpha powder with yield of 9.5% (w/w, dry algae powder). After that, crude EPs was separated by ATPSs, and analyzed using anion exchange chromatography and FT-IR. The results of anion exchange chromatography indicated that crude EPs contained three fractions (EP-1, EP-2, EP-3). The results of crude EPs separating by ATPSs demonstrated that majority of EP-3 concentrated in top PEG-rich phase, and EP-2 in bottom salt-rich phase of PEG1000/(NH4)2SO4 ATPS. FT-IR results showed that EP-3 presented SO and COS strength vibrations at 1254 and 848 cm−1 (SO4 2− group), while EP-2 had no correlative strength vibrations. These results revealed that an ATPS method was established preliminarily for sulfated polysaccharides separation from a natural polysaccharides mixture.

Abstract:Hydroxypropyl methylcellulose acetate succinate (HPMC-AS) is one of the most widely used polymers used in amorphous solid dispersions (ASD) for solubility and bioavailability enhancement of poorly water-soluble drugs. Once released from ASDs, HPMC-AS was often found to be highly effective in maintaining drug supersaturation, and this capability is dependent on the concentration and substitution types of this pH-dependent polymer. Therefore, accurate quantification of different grades of HPMC-AS allows us to better understand the release and supersaturation mechanisms of HPMC-AS based ASDs. Since previously reported analytical methods were unable to quantify HPMC-AS in a complex medium with enough sensitivity, we hereby developed a high-sensitivity HPLC-ELSD (evaporative light scattering detector) method with satisfactory specificity, linearity, accuracy and precision, to quantify HPMC-AS down to 20 μg/mL in dissolution media, with the presence of various commonly used pharmaceutical excipients. With the assistance of this method, we compared the intrinsic dissolution rates (IDR) of both the drug and the polymer of posaconazole ASDs based on different types of HPMC-AS. We observed that: 1) For ASDs that were spray dried and uniformly mixed, drug and polymer released simultaneously into the medium with practically identical IDRs slower than the IDR of pure HPMC-AS; 2) For ASDs that were heterogeneously mixed, IDRs of the drug and polymer were significantly slower or faster than the IDRs of the drug and polymer of the uniform ASDs, respectively. In summary, the high sensitivity HPLC-ELSD method established here can be readily applied to quantify HPMC-AS in various dissolution media, thus helps to reveal the release kinetics and mechanisms of different HPMC-AS based ASDs.
