Abstract:Objective. We investigated the efficacy of astragalus polysaccharide (APS) combined with budesonide and the effect on expressions of peripheral NK cells and Treg cells and the molecular mechanism in mice with bronchial asthma. Methods. In this study, we established a mouse model of asthma. Four groups of BaLB/C mice were developed; control group had no asthma induction, and the other three groups of mice were sensitized by OVA (Ovalbumin), OVA + budesonide, and OVA + APS + budesonide. Flow cytometry was used to determine the proportion of NK cells and Treg cells. Levels of cytokines IL-4 and IL-10 were detected using RT-PCR and ELISA. Results. Asthma mice treated with APS + budesonide showed alleviated airway resistance compared to model mice (P<0.05). The percentage of dendritic cells (DCs) was reduced (P<0.05), while anti-inflammatory NK cells and Treg cells significantly increased after APS + budesonide treatment (P<0.05). Further, APS + budesonide treatment resulted in improvements in IL-4 and IL-10 mRNA and protein levels (P<0.05). Conclusion. APS combined with budesonide medication may regulate expressions of DCs cells and related cytokines, reliving clinical symptom of bronchial asthma.

Abstract:OBJECTIVE:To develop a method of establishment of airway allergic inflammation model of C57BL/6 mice sensitized and challenged with the spores of Alternaria alternate.METHODS:C57BL/6 mice were divided into negative control group, positive control group and model group, twelve mice in each group. Mice of model group were sensitized with spores at day 0, 5, 10 by intraperitoneal injection, and challenged four times by intranasal drip from day 14 to day 17. OVA instead of spores were used in the positive control group, PBS used in the negative control group. Bronchoalveolar lavage fluid (BALF) was taken at 24h after the last time of challenge, and the pathological manifestation of the lung, cell counts and differential count and IL-4, IFN-gamma total protein levels in BALF were assessed. The levels of total IgE and specific IgE in plasma were identified, the airway resistance and lung compliance were determined.RESULTS:There was pulmonary eosinophilic inflammation in the positive control group and model group. Total cells count, differential cells count and IL-4 and total protein levels in BALF were enhanced significantly while IFN-gamma declined remarkably. The levels of total IgE and specific IgE were increased remarkably in plasma in positive control group and model group, the airway resistance was enhanced and lung compliance decreased.CONCLUSION:An airway allergic inflammation model of C57BL/6 mice has been established by sensitizing and challenge with spores of Alternaria alternate.
Establishment and characterization of an experimental mouse model of allergic rhinitis

Abstract:Allergic rhinitis (AR) is a common worldwide disease. Animal studies on AR were adopted in various investigations. However, animal studies simply aimed at establishing an animal model for AR have been seldom seen. The purpose of this study was to introduce an easy-to-establish experimental mouse model of AR. To develop a mouse model of AR, 38 Balb/c mice were randomly assigned to two groups. Mice in the study group were sensitized by intraperitoneal (IP) injection of ovalbumin (OVA) on day 1 and 6, followed by continuous inhalation (IH) of OVA solution for 1 week (day 8–14) using a newly designed inhalation box. The control group mice received sensitization of IP normal saline and IH sterilized distilled water instead of OVA. Before and after sensitization, the frequencies of nasal symptoms (sneezing, nasal rubbing) were recorded and the serum levels of total immunoglobulin E (IgE) were evaluated using ELISA. Finally, the murine nasal mucosal tissues were stained by Giemsa solution to estimate the degree of mast cell infiltration. After sensitization by IP and IH OVA, the study group showed significant phenotypic changes including increased sneezing and rubbing. Pathological and cytological findings also confirmed significant elevated serum total IgE titer and local mast cell infiltration in the study group statistically. We successfully developed a workable experimental animal model for AR that was more easily sensitized using our new-designed inhalation box, with less stress and more precisely to be observed.

Abstract:Abstract The present study aims to investigate the effect of immunotherapy in a mouse model of allergic rhinitis (AR) and to explore the possible molecular mechanisms of action. An animal model of AR was established by sensitization and challenge of BALB/c mice with house dust mite (HDM) extract. The mice were injected subcutaneously with HDM for immunotherapy. AR nasal symptoms were evaluated according to the frequencies of nose rubbing and sneezing and the degree of rhinorrhea. The nasal mucosa and lung tissue architecture and inflammatory status by histological analysis; the infiltration of eosinophils in nasal lavage fluid (NALF) of mice was observed by Diff-Quik stain; ELISA-based quantification of serum HDM-specific IgE and TH1/TH2 cytokine concentration; and flow cytometry detected the number of serum CD4+/CD8+ cells to evaluate the mechanism of immunotherapy. It was found that after immunotherapy, the AR symptom score was reduced, the number of eosinophils in NALF was reduced, and the infiltration of inflammatory cells and tissue damage in the nasal mucosa and lung tissue were alleviated. Immunotherapy can increase the number of CD4+ T cells in the peripheral blood, increase the ratio of CD4+/CD8+ cells, increase the expression of Th1 cytokines such as IL-2 and IFN-γ, reduce the expression of Th2 cytokines such as IL-4 and IL-5. The results showed that repeated intraperitoneal injection of crude extract of HDM for sensitization, followed by nasal drops can effectively construct a mouse model of AR, and subcutaneous injection of immunotherapy in mice can reduce allergic inflammation in model mice and improve the inflammatory infiltration of the nasal cavity in allergic rhinitis. Immunotherapy can reduce the expression of inflammatory factors in AR, improve Th1/Th2 balance, and may play a role in the treatment of AR by improving the function of immune cells.

Abstract:Abstract The reduction of dioxygen in the presence of sodium cations can be tuned to give either sodium superoxide or sodium peroxide discharge products at the electrode surface. Control of the mechanistic direction of these processes may enhance the ability to tailor the energy density of sodium–oxygen batteries (NaO2: 1071 Wh kg−1 and Na2O2: 1505 Wh kg−1). Through spectroelectrochemical analysis of a range of non‐aqueous solvents, we describe the dependence of these processes on the electrolyte solvent and subsequent interactions formed between Na+ and O2 −. The solvents ability to form and remove [Na+‐O2 −]ads based on Gutmann donor number influences the final discharge product and mechanism of the cell. Utilizing surface‐enhanced Raman spectroscopy and electrochemical techniques, we demonstrate an analysis of the response of Na‐O2 cell chemistry with sulfoxide, amide, ether, and nitrile electrolyte solvents.
The roles of thymic stromal lymphopoietin receptor and its antibody in airway inflammatory response in asthmatic mice

Abstract:Objective To determine the roles of thymic stromal lymphopoietin receptor (TSLPR) and its antibody in airway inflammatory response in asthmatic mice, and to promote maturation and activation of dendritic cells (DCs) in mouse airway. Methods BALB/c mice were randomly divided into group A, B and C. The mice in group B and C were intraperitoneally injected with OVA for allergization while the mice in group A were intraperitoneally injected with PBS as the normal control. The mice in group B and C were treated by inhalation of non-specific IgG and TSLPR IgG respectively, before provocation of asthma using OVA. The bronchoalveolar lavage fluid (BALF) of the mice in different groups were collected for cell differential counts and quantitative detection of IL-4, IL-5, IFN-γand IL-10 levels by ELISA. Moreover, the pulmonary tissue specimens of the mice were collected for pathological examination, and the numbers and phenotypes of DCs from the local lymph nodes and pulmonary tissue were determined by flow cytometry. Results The levels of all the tested cytokines in the BALF from mice in group B and C were remarkably higher compared to those from mice in group A (P<0.01). However, both the IL-4 and IL-5 levels in the BALF from group C mice that pre-blocked with TSLPR IgG were lower than those from group B (P<0.05, P<0.01), whereas both the IFN-γ and IL-10 levels in the BALF from group C mice were higher than those from group B (P<0.05, P<0.01). Furthermore, the numbers of total cells, eosinophils and lymphocytes in the BALF from group C mice were also lower than those from group B (P<0.01). A large number of inflammatory cell infiltration around the bronchus, beaker cell proliferation and mucous secretion reinforcement could be found in the samples from group B mice, while slight inflammatory cell infiltration and beaker cell proliferation in the samples from group C mice. The numbers of DCs in mediastinal lymph node and the levels of I-Ad, CD40, CD80 and CD86 expression of pulmonary DCs from group B mice were higher than those from group C mice (P<0.05). Conclusion TSLP/TSLPR have an effect on promoting asthma, which is closely relative to its regulation of DCs activation. And the interference of TSLPR antibody can decrease the effect of TSLP/TSLPR which indicating a potential of the antibody as a novel anti-asthma drug.
Preparation and evaluation of a high level of IgE of mouse asthma model induced by Dermatohpagoides farinae extracts

Abstract:Objective: To construct a mouse asthma model sensitized and challenged by Dermatohpagoides farinae extracts with high level of IgE. Methods: C57BL /6 mice were randomly divided into control group and asthma group. Mice in asthma group were sensitized by Dermatohpagoides farinae extracts through intraperitoneal injection at day point 0,7 and 14,PBS instead for the control group. On day 28,the mice of asthma group were challenged by intranasal Dermatohpagoides farinae extracts for seven consecutive days and the ones of control group were administrated by PBS instead. Observe and record the symptoms of asthma in each group. After the final allergen challenging,the total and sorting cell count of bronchoalveolar lavage fluid( BALF),histological examination,serum IgE and IL-4 and IFN-γ in BALF were carried out. Results: The number of total cells and eosinophils in BALF of asthma group was significantly higher than that in the control group( P 0. 01). Inflammatory cells were obviously observed in the lung tissues of the asthma group. The level of total IgE( P 0. 001) and rDer f 2-sIgE( P 0. 01) in serum enhanced significantly. The level of IL-4 in BALF was significantly increased( P 0. 05),while the INF-γ was decreased( P 0. 01). Conclusion: It is possible to construct a mouse model of asthma through repeatedly sensitizing and challenging by Dermatohpagoides farinae extracts to product high level of IgE.
Comparison of the Roles of House Dust Mite Allergens, Ovalbumin and Lipopolysaccharides in the Sensitization of Mice to Establish A Model of Severe Neutrophilic Asthma

Abstract:Neutrophilic asthma (NA) is associated with a severe disease course and poor response to corticosteroids. The present study aimed to compare the effects of various concentrations of house dust mite (HDM) allergens, ovalbumin (OVA), the major egg allergen, and lipopolysaccharide (LPS) in combination on the onset of severe NA. Female C57BL/6 mice were grouped according to a random number table and intranasally sensitized with HDM/LPS/OVA extracts on days 0, 1 and 2 of the study. In group 1, mice received 50 µg HDM + 50 µg OVA + 15 µg LPS, mice in group 2 received 50 µg HDM + 100 µg OVA + 15 µg LPS, mice in group 3 received 100 µg HDM + 50 µg OVA + 15 µg LPS and those in group 4 received 100 µg HDM + 100 µg OVA + 15 µg LPS, while mice in the control group received saline only. The mice were then challenged by OVA solution with atomized excitation on days 14, 15, 18, 19 and 20 for 30 min each. Ethology, airway hyperresponsiveness (AHR), immune cell distributions in bronchoalveolar lavage fluid (BALF), and specific cytokines interleukin 17A (IL-17A) and IL-4 in serum were assessed. Histological examination of inflammation by hematoxylin and eosin staining and immunohistochemical assessment of neutrophils (NEU), eosinophils (EOS), IL-17A and IL-4 were also performed. Compared with the control group, the HDM/OVA/LPS-sensitized groups 1-4 had markedly increased BALF cells, serum interleukin IL-17A and IL-4, inflammatory cell infiltration, EOS as well as IL-17A and IL-4 by immunohistochemical staining (all P<0.05). Among the four HDM/OVA/LPS-sensitized groups, mice of group 4 had higher AHR, a significantly higher total cell number, NEU and EOS in BALF as well as significantly higher NEU and NEU/EOS ratios according to immunohistochemical staining when compared to groups 1-3 (P<0.05 for all). In conclusion, sensitization with 100 µg HDM + 100 µg OVA + 15 µg LPS successfully established a severe asthma model with a predominantly neutrophilic inflammatory phenotype.
Yupingfeng Pulvis Regulates the Balance of T Cell Subsets in Asthma Mice.

Abstract:Background. Yupingfeng Pulvis (HFBP) had played an active role in many diseases, especially respiratory tract infections. Exploring the possible prevention mechanism of HFBP may provide new ideas in clinical applications for this well-known herbal formula. Purpose. To study the possible mechanisms of therapy effect of HFBP on asthma mice via regulating the balance of Tregs and Th17 cells. Method. The female BALB/c mice were divided into five groups: control group, model group, prednisone (5.5 mg/kg) group, and 22 g/kg HFBP and 44 g/kg HFBP groups. Ovalbumin was used to make the asthma model of mice; the drug was ig administered daily after atomization for consecutive 15 d. The mice were killed after the last administration. The paraffin-embedded tissue sections of the lungs were stained by H&E. Tregs and Th17 cells in bronchoalveolar lavage fluid were detected by flow cytometry. IL-4, TGF-β, and TNF-α in the serum were detected by ELISA assay. Results. HFBP could alleviate the inflammation in the lung tissue of mice, decrease the proportion of Th17 cells, and increase the proportion of Treg cells in bronchoalveolar lavage fluid. HFBP could decrease IL-4 and TNF-α level and increase TGF-β level in blood. Conclusion. HFBP could treat the asthma through impacting the balance of Th17 cells and Treg cells as well as the levels of related inflammatory cytokines in asthma mice.
Enhanced Local Foxp3 Expression in Lung Tissue Attenuates Airway Inflammation in A Mouse Model of Asthma

Abstract:Objective: Bronchial asthma is a chronic inflammatory disease of the airway mediated by a Th2 immune response. A great deal of data has demonstrated that regulatory T cells (Tregs) have the ability to suppress Th2 immune responses and the transcription factor fork-head box protein 3 (Foxp3) is indispensable for the development of CD4 + CD25 + Tregs. In this study, we hypothesized that enhanced local Foxp3 expression in lung tissue could suppress Th2-mediated allergic asthma. Methods: Foxp3/PMX retroviruses containing the mouse Foxp3 gene were constructed and administered into asthmatic mice through intra-tracheal instillation before ovalbumin challenging. Foxp3 expression, airway hyper-responsiveness (AHR), bronchoalveolar lavage fluid (BALF) and tissue inflammatory cell and cytokine profiles were characterized. Results: Foxp3 mRNA and protein were increased in the lung tissue of asthmatic mice. Enhanced expression of Foxp3 locally in the lung tissue reduced the airway AHR, inflammatory cell infiltration and mucus production. It also attenuated Th2 and Th17 immune responses as evidenced by reduced IL-4, IL-13 and IL-17 levels. Conclusions: This study demonstrates that enhanced Foxp3 expression in the airway by intra-tracheally instilled Foxp3/ PMX retroviruses alleviates allergic airway inflammation by reducing the Th2 immune response.
Establishing a High Microbial Load Maternal-Offspring Asthma Model in Adult Mice

Abstract:BACKGROUND:Although it is widely accepted that the "hygiene hypothesis" explains the increased incidence of asthma, the lack of suitable animal models hinders further in-depth studies of the underlying molecular immune mechanisms. Therefore, we aimed to develop a robust mouse asthma model to investigate the role of bacteria in preventing asthma.METHODS:BALB/c female mice were fed a mixture of eight common bacterial lysates (BL; Broncho-Vaxom®) and a commercial probiotic (Bifidobacterium tetravaccine tablets) at different concentrations before and during pregnancy to simulate different microbial load levels. Faeces from the mother mice were subjected to bacterial 16S rRNA sequencing to quantify the maternal microbial load. TLR2/4 expression and the proportion of regulatory T cells (Tregs) in the intestinal tract of female mice were determined, and the safety of the microbial load was evaluated. An asthma model was established in the offspring mice after weaning, and the extent of pulmonary pathological changes and Treg proportion were evaluated.RESULTS:A BL concentration of 1 mg/kg enriched the intestinal flora, increased the proportion of Tregs, and increased the expression of TLR2/4 in the maternal mice. The proportion of peripheral blood Tregs was increased, whereas the risk of asthma decreased only in the offspring from mothers with a high microbial load relative to control mice.CONCLUSION:This study established a safe and stable high microbial load maternal-offspring mouse asthma model, laying the foundation for a study on the molecular mechanism underlying the protective effects of a high microbial load against asthma.
Alteration of lung tissues proteins in birch pollen induced asthma mice before and after SCIT

Abstract:Subcutaneous immunotherapy (SCIT) is a classic form of allergen-specific immunotherapy that is used to treat birch pollen induced allergic asthma. To investigate the underlying molecular mechanisms of SCIT, we aimed to profile lung samples to explore changes in the differential proteome before and after SCIT in mice with allergic asthma. Fresh lungs were collected from three groups of female BALB/c mice: 1) control mice, 2) birch pollen-induced allergic mice, and 3) birch pollen-induced allergic mice with SCIT. Tandem mass tag (TMT) labelling coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to analyze the lung proteome in the mice. Ingenuity pathway analysis (IPA) and Gene Ontology (GO) classification analysis were applied to identify differentially expressed proteins (DEPs) and crucial pathways. The screened DEPs were validated by immunohistochemistry analysis. A total of 317 proteins were upregulated and 184 proteins were downregulated in the asthma group compared to those of the control group. In contrast, 639 DEPs (163 upregulated and 456 downregulated proteins) were identified after SCIT in comparison with those of the asthma group. Among the 639 DEPs, 277 proteins returned to similar levels as those of the relative non-asthma condition. Bioinformatic analysis revealed that the 277 proteins played a significant role in the leukocyte extravasation signaling pathway. The leukocyte extravasation signaling pathway and related DEPs were of crucial importance in birch pollen SCIT.

A Mouse Model of Atopic March Induced by Epicutaneous Immunization with House Dust Mite Allergen: a Preliminary Study

Abstract:Objective To establish a mouse model of atopic march induced by epicutaneous sensitization and nasal challenge with dust mite allergen.Methods C57BL/6J mice were subjected to repeated epicutaneous sensitization with dust mite allergen or saline and then challenged with intranasal administration of mite allergen to induce airway inflammation.The mouse model was evaluated in terms of pathology of skin and lung,quantitative mRNA expression of inflammatory molecules,count of inflammatory cells in bronchoalveolar lavage fluid,and airway hyperresponsiveness.Results Mite allergen sensitized mice exhibited typical pathological changes of eczematous dermatitis and significantly higher expression levels of Th2 cytokines in lesions while Thl cytokines remained unchanged in comparison with normal controls.The number of total cells,lymphocytes and eosinophils in the bronchial alveolar lavage fluid were significantly higher in model mice than in the controls.Pulmonary histopathology showed peribronchial infiltration of eosinophils.The expression levels of Th2 cytokine mRNA in lung tissue also significantly increased.Methacholine dose-dependently induced increase in the lung resistance and decrease in the dynamic compliance of the lungs in mouse model.Conclusion In this study,we use epicutaneous sensitization and nasal challenge to successfully establish the mite allergen induced atopic march model.
The Effects Of Overexpression Of Histamine Releasing Factor (Hrf) In A Transgenic Mouse Model

Abstract:Background: Asthma is a disease that affects all ages, races and ethnic groups. Its incidence is increasing both in Westernized countries and underdeveloped countries. It involves inflammation, genetics and environment and therefore, proteins that exacerbate the asthmatic, allergic phenotype are important. Our laboratory purified and cloned a histamine releasing factor (HRF) that was a complete stimulus for histamine and IL-4 secretion from a subpopulation of allergic donors' basophils. Throughout the course of studying HRF, it was uncovered that HRF enhances or primes histamine release and IL-13 production from all anti-IgE antibody stimulated basophils. In order to further delineate the biology of HRF, we generated a mouse model.Methodology/Principal Findings: We constructed an inducible transgenic mouse model with HRF targeted to lung epithelial cells, via the Clara cells. In antigen naive mice, overproduction of HRF yielded increases in BAL macrophages and statistical increases in mRNA levels for MCP-1 in the HRF transgenic mice compared to littermate controls. In addition to demonstrating intracellular HRF in the lung epithelial cells, we have also been able to document HRF's presence extracellularly in the BAL fluid of these transgenic mice. Furthermore, in the OVA challenged model, we show that HRF exacerbates the allergic, asthmatic responses. We found statistically significant increases in serum and BAL IgE, IL-4 protein and eosinophils in transgenic mice compared to controls.Conclusions/Significance: This mouse model demonstrates that HRF expression enhances allergic, asthmatic inflammation and can now be used as a tool to further dissect the biology of HRF.
A 4-Week Model of Allergic Airways Inflammation That Exhibits the 3 Salient Features of Asthma

Abstract:Background: Mouse models of allergic airways inflammation are useful experimental tools to study asthma. Numerous models vary widely, depending on the type, dose and frequency of allergen used to induce airways inflammation phenotype. Many models exhibit some, but not all, of the salient features of asthma. Models that exhibit all 3 features, namely hyperresponsiveness (AHR), airway inflammation and airway remodeling, are most representative of asthma. However, airway remodeling requires prolonged allergen exposure, typically 8-12 weeks, adding time and cost. Purpose: To develop an allergic airways inflammation model of asthma in a shorter time. Methods: We exposed BALB/c mice to house dust mite (HDM), a clinically relevant allergen, for 2, 4 or 8 weeks, and compared lung function, methacholine (MCh) responsiveness, airway inflammation and airway remodeling 24h after the last exposure. Results: Enhanced AHR to MCh of the total and central respiratory resistance was seen in all HDM groups compared to Saline control. Within the HDM groups, we found a progressive increase in the total cell, neutrophil, eosinophil and lymphocyte counts amongst the 2-, 4- and 8-week groups. Serum HDM-specific IgE levels increased significantly at 4 and 8 week with a peak at 4 weeks. Airway remodeling was observed in the 4 and 8 weeks, but absent at 2 weeks. Conclusion: The 4-week HDM model exhibits all 3 salient features of asthma and is phenotypically similar to the 8-week model. The shorter development offers an advantage in experimental models that aim to evaluate features of asthma.
Establishment of a mouse model mimicking neutrophilic steroid-refractory asthma

Abstract:Objective To establish a murine model of neutrophilic steroid-refractory asthma for a better exploration of its pathogenesis and treatment.Methods A mouse model of neutrophilic steroid-refractory asthma was established by intratracheal administration of a mixture of house dust mite (HDM) and lipopolysaccharide (LPS).Eighteen female C57BL/6 mice were randomly divided into a control group,an asthma group (HDM + LPS) and a dexamethasone (Dex) group (HDM + LPS + Dex).The airway resistance was measured by pulmonary function test.Airway inflammation,hypersecretion and inflammatory cell recruitment were observed by HE,PAS and Wright's staining,respectively.Inflammatory mediators in lung and bronchoalveolar lavage fluid (BALF) were detected by PCR and ELISA.Th17 cell differentiation in the lung was analyzed by flow cytometry.Results HE staining showed a significantly increased airway inflammation in the asthma group than that in the control group (P＜0.05),and a slightly decreased airway inflammation in the Dex group than that in the asthma group (P＞0.05).Cell counts in BALF showed a sharply elevated recruitment of inflammatory cells in the asthma group (except eosinophils) than that in the control group (total number of inflammatory cells (2797±400)× 106/L vs (105±75)× 106/L,neutrophiis (1151±395)×106/L vs (12±6)×106/L,lymphocytes(897± 135)× 106/L vs (11±5)×106/L,macrophages (215±51)× 106/L vs (34± 16)× 106/L,P＜0.05).Compared with the asthma group,Dex group significantly reduced the recruitment of total number of inflammatory cells and macrophages [total number of inflammatory cells (1140±418) × 106/L vs (2797±400)× 106/L,macrophages (117±31)×106/L vs (215±51)×106/L,all P＜0.05]albeit no significances in lymphocytes and neutrophils [lymphocytes (587±208)× 106/L vs (897± 135)× 1 06/L,neutrophils (294± 134)×106/L vs (1151 ± 395)× 106/L,all P＞0.05].Immunohistochemical staining confirmed that Dex could not effectively alleviate the neutrophilic inflammation in asthmatic mice.Although there was no significant difference in airway hyperreactivity between the asthma group and the Dex group (P＞0.05),both had a higher airway hyperreactivity than that of the control group (P＜0.05).The Dex group exhibited a significantly lower Th2-related inflammatory mediators in the lung than the asthma group(all P＜0.05),together with an increased trend in Th17 cell-related inflammatory mediators and no improvement in Th1-related inflammatory mediators.Flow cytometry showed an obviously increased percentage of Th 17 cells in the Dex group than that in the asthma group [(5.8 ± 1.9)％ vs (2.3 ±0.8)％,P＜0.01].Conluusion A novel murine model mimicking clinical neutrophilic asthma was elegantly established,in which Dex could not attenuate the neutrophilic airway inflammation and hyperreactivity,but rather augmented Th17 cell differentiation,demonstrating the presence of steroid-resistance.
Effect of intralymphatic allergen-specific immunotherapy on house dust mite in a murine model of allergic rhinitis

Abstract:Background: Intralymphatic immunotherapy (ILIT) is a promising alternative for the treatment of patients with allergic rhinitis, providing similar therapeutic efficacy to conventional allergen-specific immunotherapy (AIT). However, the allergic mechanism of ILIT is not completely known. Aim: The aim of this study was to determine the efficacy of ILIT in a house dust mite (HDM) mouse model of allergic rhinitis. Methods: BALB/c mice were divided into four groups: G1, control without allergy; G2, allergy sensitized with HDM; G3, allergy with ILIT (starting with HDM 1.25 μg/mL); and G4, allergy with ILIT (starting with HDM 2.5 μg/mL). After the murine model of allergic rhinitis with HDM was established, mice were administered an intralymphatic injection through the inguinal lymph nodes with HDM. Results: ILIT decreased serum total IgE level and eosinophil infiltration in the nasal mucosa. ILIT also decreased the expression levels of IL-13, IL-25, IL-33, IFN-γ, IL-6, and IL-17, and increased the expression of FoxP3(+) T reg cells. Conclusions and significance: Our results suggest that ILIT regulates the specific immunotherapy immunologic mechanism by downregulating Th1, Th2, and Th17 cytokines and upregulating FoxP3(+) T reg cells in the HDM allergic mouse model.

Abstract:IL-5/IL-5R signaling plays a key role in the maturation, activation, survival, and recruitment of eosinophils to sites of inflammation. Targeting this pathway is an attractive therapeutic strategy for certain inflammatory diseases, such as asthma. Here, we generated a novel humanized IL5/IL5RA model (B-hIL5/hIL5RA) to examine the utility of human IL-5/IL5R-targeting therapeutic candidates. Specifically, the entire murine Il5 coding region was replaced by the corresponding human IL5 sequence, and the human IL5RA sequence coding the extracellular domain was inserted to replace the corresponding murine sequence. Successful generation of the model was confirmed by expression of human IL-5 by ELISA and IL5RA by flow cytometry. Furthermore, the proportion of eosinophils in the blood of B-hIL5/hIL5RA mice increased after stimulation with human IL5 recombinant protein, indicating that the humanized IL5RA protein is functional. Next, the ability of the humanized mice to establish asthma-like disease was tested using ovalbumin: following a 14 day sensitization period, mice were challenged with nebulized OVA daily from day 21 until the endpoint; treatment with mepolizumab or benralizumab analog was performed. Flow cytometric analysis of the bronchioalveolar fluid indicates that treatment with mepolizumab or benralizumab analog significantly lowered the number of eosinophils when compared to the isotype control in homozygous B-hIL5/hIL5RA mice. Blood counts and quantification of lung H&E sections also confirm that the treatments lowered the number of eosinophils in circulation and lung tissue, respectively. Taken together, these data indicate that B-hIL5/hIL5RA mice are a suitable preclinical model to test novel modulators of human IL-5/IL-5RA signaling.
