Abstract:Fe3O4@C nanocapsules were synthesized via a sacrificial-template method by coating SiO2 nanospheres with an Fe3O4@C double-shell structure, followed by dissolving SiO2 nanospheres through ammonia water under hydrothermal conditions. The nanocapsules show a loading capacity as high as 1300 mg g-1 for doxorubicin (DOX), and the DOX loaded on the surface of the carbon shells displays pH-sensitive release behavior. Drug release experiments were carried out at pH 7.4, 6.2 and 5.0. It was found that the drug release rate at pH 6.2 was about two times as fast as that at pH 7.4, and even faster at pH 5.0. A MTT assay was used to test the cytotoxicity of the DOX, nanocapsules and DOX-nanocapsules, which indicated the low cytotoxicity of the nanocapsules towards cells. The DOX-nanocapsules can be taken up by cancer cells through endocytosis, releasing DOX into the cytoplasm, which was observed by both transmission electron microscopy and confocal microscopy. In vitro magnetic resonance imaging (MRI) experiments validated the potential use of these nanocapsules as MRI contrast agents.
Calcium Carbonate End-Capped, Folate-Mediated Fe3O4@mSiO(2) Core-Shell Nanocarriers As Targeted Controlled-Release Drug Delivery System

Abstract:Magnetic mesoporous silica nanospheres (MMSN) were prepared and the surface was modified with cancer cell-specific ligand folic acid. Calcium carbonate was then employed as acid-activated gatekeepers to cap the mesopores of the MMSN, namely, MMSN-FA-CaCO3. The formation of the MMSN-FA-CaCO3 was proved by several characterization techniques, viz. transmission electron microscopy, zeta potential measurement, Fourier transform infrared spectroscopy, BET surface area measurement, and UV-Vis spectroscopy. Daunomycin was successfully loaded in the MMSN-FA-CaCO3 and the system exhibited sensitive pH stimuli-responsive release characteristics under blood or tumor microenvironment. Cellular uptake by folate receptor (FR)-overexpressing HeLa cells of the MMSN-FA-CaCO3 was higher than that by non-folated-conjugated ones. Intracellular-uptake studies revealed preferential uptake of these nanoparticles into FR-positive [FR(+)] HeLa than FR-negative [FR(-)]A549 cell lines. DAPI stain experiment showed high apoptotic rate of MMSN-FA-DNM-CaCO3 to HeLa cells. The present data suggest that the CaCO3 coating and folic acid modification of MMSN are able to create a targeted, pH-sensitive template for drug delivery system with application in cancer therapy.
Calcium Carbonate/Carboxymethyl Chitosan Hybrid Microspheres and Nanospheres for Drug Delivery

Abstract:Calcium carbonate/carboxymethyl chitosan (CaCO3/CMC) hybrid microspheres and nanospheres were prepared by the precipitation of calcium carbonate in an aqueous solution containing CMC. Through adjusting the preparation conditions, the size of CaCO3/CMC hybrid particles could be easily controlled at micro- to nanometer ranges with relatively narrow size distributions. The obtained microspheres and nanospheres were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, thermogravimetric analysis, and differential scanning calorimetry. The size and size distribution were measured by a particle size analyzer. Doxorubicin hydrochloride (DOX center dot HCl), a water-soluble anticancer drug, was loaded in the hybrid microspheres and nanospheres with a high encapsulation efficiency. The in vitro drug release showed that the release of DOX center dot HCl from the microspheres and nanospheres could be effectively sustained.

Abstract:Although various drug delivery systems that regulated Ca 2+ concentration has been developed for tumor therapy, their application still presented significant challenges due to the complex preparation and introduction of a large number of inorganic molecules that might cause serious toxic effects. To solve these problems, a folate-functionalized carboxymethyl chitosan (CMCS)/calcium phosphate hybrid nanoparticle (CF/CaP) with Ca 2+ production was designed to treat breast cancer combined with the Ca 2+ inhibitory effect of encapsulated curcumin (Cur). It was demonstrated that the optimal CF/CaP nanoparticles loaded with Cur (C@CF/CaP) were spherical nanoparticles, which exhibited a smaller size at about 179 nm than non-targeted nanoparticles with size at about 234 nm. C@CF/CaP had good biocompatibility, high stability and acid responsive drug release. Compared with the neutral environment, the cumulative release of Cur was >70 % after culture for 36 h at pH 5.0. Compared with non-targeted nanoparticles, C@CF/CaP could specifically target tumor tissues and then enter tumor cells through folate receptor-mediated endocytosis. C@CF/CaP could cause mitochondrial Ca 2+ overload, trigger the mitochondrial apoptotic pathway, destroy the mitochondrial structure and finally have good anti-tumor efficiency. The results proved that Ca 2+ nanomodulators based on CMCS might provide a potential organelle targeting strategy for cancer therapy.

Abstract:In this work, we report the fabrication and functional demonstration of a kind of dually responsive nanoparticles (NPs) as a potential drug delivery vector. The pH value, corresponding to the acidic microenvironment at the tumor site, and mannitol, to the extracellular trigger agent, were employed as the dually responsive factors. The function of dual responses was achieved by breaking the dynamic covalent bonds between phenylboronic acid (PBA) groups and diols at low pH value (pH 5.0) and/or under the administration of mannitol, which triggered the decomposition of the complex NPs and the concomitant release of anticancer drug of doxorubicin (DOX) loaded inside the NPs. The NPs were composed of modified chitosan (PQCS) with quaternary ammonium and PBA groups on the side chains, heparin (Hep), and poly(vinyl alcohol) (PVA), in which quaternary ammonium groups offer the positive charge for the cell-internalization of NPs, PBA groups serve for the formation of dynamic bonds in responding to pH change and mannitol addition, PVA furnishes the NPs with diol groups for the interaction with PBA groups and the formation of dynamic NPS, and Hep plays the roles of reducing the cytotoxicity of highly positively-charged chitosan and forming of complex NPs for DOX up-loading. A three-step fabrication process of drug-loaded NPs was described, and the characterization results were comprehensively demonstrated. The sustained drug release from the drug-loaded NPs displayed obvious pH and mannitol dependence. More specifically, the cumulative DOX release was increased more than 1.5-fold at pH 5.0 with 20 mg mL−1 mannitol. Furthermore, the nanoparticles were manifested with effective antitumor efficient and apparently enhanced cytotoxicity in response to the acidic pH value and/or mannitol.

Abstract:Zeolitic imidazolate framework-8 (ZIF-8) is a promising material for drug delivery, but its poor stability limits its wide application. In this work, homogeneous dodecahedral ZIF-8 was prepared by the room-temperature solution stirring method, and to improve the stability of ZIF-8, ZIF-8@mSiO 2 and DZIF-8 (ZIF-8 after calcination) @PMO composites with the core-shell structure were obtained using mesoporous silica (mSiO 2 ) and periodic mesoporous organic silica (PMO) coatings. Folic acid-coupled chitosan (CS-FA) was modified on nanoparticles, and curcumin (CUR) was used as a model anti-tumor drug to obtain the drug delivery systems with targeting and pH stimulation response: ZIF-8@mSiO 2 @CUR@CS-FA and DZIF-8@PMO@CUR@CS-FA, which exhibited good dispersion and high drug loading capability. In addition, we used the MTT assay to assess the cytotoxicity of the drug-carrying systems and selected human cervical cancer cells (Hela) as model cells. The results of the in vitro experiments suggested that the final CUR-loaded drug delivery systems had significantly increased anticancer efficiency compared with the free drug CUR, which improved the therapeutic effect of the hydrophobic CUR on tumors. In conclusion, the pH-responsive nanocarriers provided potential for the targeted delivery of anticancer medication, resulting in a platform for drug-controlled release and cancer therapy.

Abstract:'' Nano-metamaterials '', rationally designed novel class metamaterials with multilevel microarchitectures and both characteristic sizes and whole sizes at the nanoscale, are introduced into the area of drug delivery system (DDS), and the relationship between release profile and treatment efficacy at the single-cell level is revealed for the first time. Fe3+-core-shell-corona nano-metamaterials (Fe3+-CSCs) are synthesized using a dual-kinetic control strategy. The hierarchical structure of Fe3+-CSCs, with a homogeneous interior core, an onion-like shell, and a hierarchically porous corona. A novel polytonic drug release profile occurred, which consists of three sequential stages: burst release, metronomic release, and sustained release. The Fe3+-CSCs results in overwhelming accumulation of lipid reactive oxygen species (ROS), cytoplasm ROS, and mitochondrial ROS in tumor cells and induces unregulated cell death. This cell death modality causes cell membranes to form blebs, seriously corrupting cell membranes to significantly overcome the drug-resistance issues. It is first demonstrated that nano-metamaterials of well-defined microstructures can modulate drug release profile at the single cell level, which in turn alters the downstream biochemical reactions and subsequent cell death modalities. This concept has significant implications in the drug delivery area and can serve to assist in designing potential intelligent nanostructures for novel molecular-based diagnostics and therapeutics.

Abstract:In many applications the chip-embedded core of miniature devices is a micro-structured planar spiral power transformer (MPST), and operational and electromagnetic compatibility are key issues to its sizing. Numerical simulation aided design may then be used to solve them. This paper presents mathematical models and numerical simulation results related to sizing the ferrite core, the lumped parameters of the MPST, and their sensitivity with respect to the core thickness (the spacing between the planar windings) and the magnetic permeability of the ferrite core. These outcomes may be of interest to the electrical design of the device that uses the MPST and in analyzing its electromagnetic compatibility with the interfering working environment. Finally, we address the high-frequency noise reduction in DC/DC flyback power sources with MPST.
PEGylated chitosan decorated UiO-66 nanoscale metal–organic frameworks as promising carriers for drug delivery

Abstract:Nanoscale metal organic frameworks (NMOFs) have become excellent drug carriers for the treatment of cancer or other diseases, due to the advantages of large surface area, high drug loading, tunable structure and easy functionalization, with potential application in the field of drug delivery. In this paper, UiO-66 NMOF was synthesized using terephthalic acid as organic ligand and zirconium chloride as metal salt, and then biocompatible chitosan (CS) and PEGylated chitosan (CS-g-mPEG)-decorated NMOFs as drug carriers were constructed by surface modification, using 5-FU as the drug model. The structures, size and morphology were characterized by IR, XRD, DLS, SEM and TEM respectively. The results showed that when the concentration was 0.0872 M, the molar ratio was 1:1 and the acetic acid amount was 2.9 mL, the average particle size of NMOFs was 145.60 nm and 171.73 nm, respectively. The drug loading capacity and encapsulation rate could reach 15.7% and 52.8%. The in vitro controlled drug release results indicated that the release rate accelerated with increasing temperature and decreasing initial amount, greatly reduced under acidic conditions, and the coating of CS and CS-g-mPEG on the surface of UiO-66 had the effect of controlling drug release, further improved the bioavailability of 5-FU. The decorated UiO-66 NMOFs show excellent biocompatibility as nanodrug carriers and can avoid rapid degradation under gastric acid, which are more suitable for oral drug delivery.

pH-responsive chitosan-mediated spherical mesoporous silica microspheres for high loading and controlled delivery of 5-Fluorouracil

Abstract:The objective of this study is to develop a drug carrier to overcome the inherent drawbacks of 5-Fluorouracil (5-Fu), including low bioavailability, short half-life, and systemic toxicity. In the present work, mesoporous silica nanoparticles (MSNs) capped by chitosan (CS) to encapsulate 5-Fu (5-Fu MSNs/CS) were fabricated by the sol-gel process, ultrasonic impregnation, and emulsion cross-linking. The 5-Fu MSNs/CS microspheres exhibit pH-responsive drug release and remarkable drug encapsulation capacity, as well as perfect sphericity, high specific surface area (680.62 cm2/g), and uniform particle size (2.64 ± 0.05 μm). The drug-loading content and encapsulation efficiency are 14.12 ± 0.53 % and 82.21 ± 2.13 %, respectively. The cumulative release of 5-Fu from MSNs/CS microspheres is fast and sustained at pH 5.0 (89.56 ± 0.97 %) compared to that at pH 7.4 (57.88 ± 0.91 %) in 96 h, and it is Fickian diffusion controlled. In conclusion, the MSNs/CS microspheres prepared in this study could be potential carriers for 5-Fu delivery.

Abstract:Nowadays, the Magnetically Targeted Drug Delivery System (MTDDS) is among the most attractive and promising strategies for delivering drugs to the target site. The present study aimed to obtain a biopolymer–magnetite–drug nanosystem via a double crosslinking (ionic and covalent) technique in reverse emulsion, which ensures the mechanical stability of the polymer support in the form of original hybrid nanospheres (NSMs) loaded with biologically active principles (the 5-Fluorouracil (5-FU)) as a potential treatment for cancer. Obtained NSMs were characterized in terms of structure (FT-IR), size (DLS), morphology (SEM), swelling, and 5-FU entrapment/release properties, which were dependent on the synthesis parameters (polymer concentration, dispersion speed, and amount of ionic crosslinking agent). SEM analysis results revealed that NSMs presented a spherical shape and are homogeneous and separated. Moreover, NSMs’ ability to load/release 5-FU was tested in vitro, the results confirming, as expected, their dependence on the varied synthesis process and NSM swelling ability in physiological liquids. The drug transport mechanism through the polymer matrix of its release is the Fickian type. The morphological, bio-material characteristics and the ability to include and release an antitumor drug highlight the utility of the NSMs obtained for targeting and treating some tumor diseases.

Efficient Design to Monitor the Site-specific Sustained Release of a Non-Emissive Anticancer Drug

Abstract:A pH-responsive smart nanocarrier with significant components was synthesized by conjugating the non-emissive anticancer drug methyl orange and polyethylene glycol derived folate moiety to the backbone of polynorbornene. Complete synthesis procedure and characterization methods of three monomers included in the work: norbornene-derived Chlorambucil (Monomer 1), norbornene grafted with polyethylene glycol, and folic acid (Monomer 2) and norbornene attached methyl orange (Monomer 3) connected to the norbornene backbone through ester linkage were clearly discussed. Finally, the random copolymer CHO PEG FOL METH was synthesized by ring-opening metathesis polymerization (ROMP) using Grubbs' second-generation catalyst. Advanced polymer chromatography (APC) was used to find the final polymer's molecular weight and polydispersity index (PDI). Dynamic light scattering, scanning electron microscopy (SEM), and transmission electron microscopy (TEM) were utilized to explore the prodrug's size and morphology. Release experiments of the anticancer drug, Chlorambucil and the coloring agent, methyl orange, were performed at different pH and time. Cell viability assay was carried out for determining the rate of survived cells, followed by the treatment of our final polymer named CHO PEG FOL METH.
Preparation, optimisation, and properties of O-carboxymethyl chitosan-g-cholesterol succinic acid monoester polymer nanomicelles

Abstract:Polymer nanomicelles have the advantages of small particle size, improved drug solubility, retention effect and enhanced permeability, so they can be used in the treatment of tumour diseases. The aim of this study was to prepare and optimise a nanomicelle which can improve the solubility of insoluble drugs. Firstly, the carboxyl group of cholesterol succinic acid monoester was grafted with the side chain amino group of O-carboxymethyl chitosan-g-cholesterol succinic acid monoester (CCMC), and its structure was characterized by fourier transform infrared spectroscopy (FTIR) and proton nuclear magnetic resonance (1H-NMR). Particle size has an important impact on tissue distribution, cell uptake, permeability and inhibition of tumour tissue. In this study, particle size and polydispersity index (PDI) were selected as indexes to optimise the preparation process of CCMC nanomicelles through single factor experiment, Plackett-Burman experiment, the steepest climbing experiment and response surface design experiment. The optimised CCMC nanomicelles showed an average particle size of 173.9 ± 2.3 nm and a PDI of 0.170 ± 0.053. The Cell Counting Kit-8 assay showed no significant effect on cell viability in the range of 0-1000 μg ml-1concentration. Coumarin-6 (C6) was used as a fluorescent probe to investigate the drug-carrying ability of CCMC nanomicelles. C6-CCMC showed 86.35 ± 0.56% encapsulation efficiency with a drug loading of 9.18 ± 0.32%. Both CCMC and C6-CCMC demonstrated excellent stability in different media. Moreover, under the same conditions, the absorption effect of C6 in C6-CCMC nanomicelles was significantly higher than that of free C6 while also exhibiting good sustained-release properties. Therefore, this study demonstrates CCMC nanomicelles as a promising new drug carrier that can significantly improve insoluble drug absorption.
Multifunctional Fe3O4@C-based Nanoparticles Coupling Optical/mri Imaging and Ph/photothermal Controllable Drug Release As Efficient Anti-Cancer Drug Delivery Platforms

Abstract:Multifunctional nanomedicines featuring high drug loading capacity, controllable drug release and real-time self-monitoring are attracting increasing attention due to their potential to improve cancer therapeutic efficacy. Herein, a new kind of Fe3O4@C-based nanoparticles modified with isoreticular metal organic frameworks (IRMOF-3), folic acid (FA) and detachable polyethylene glycol (PEG) under tumor microenvironment was developed. The core-shell structured Fe3O4@C was synthesized via the one-pot solvothermal reaction and the IRMOF-3 layers were coated on the outer shell of Fe3O4@C through layer-by-layer coating method. The FA and PEG were conjugated on the surface of nanoparticles by reacting with the amine groups provided by IRMOF-3. The as-synthesized nanoparticles showed stable photothermal effect, superparamagnetic properties and blue fluorescence characteristic under 360 nm irradiation. The in vitro experiments showed that the drug loaded nanoparticles exhibit pH-dependent drug release property, and PEGylation was proved effective in suppressing burst drug release (only 8.0% of drugs were released within 95 h). The confocal laser scanning microscopy study revealed that the as-synthesized nanoparticles could serve as a cell imaging agent and the cell internalization can be significantly enhanced after FA modified. The IRMOF-3 modified nanoparticles showed negligible cytotoxicity and the drug loaded nanoparticles showed pH/photothermal-stimuli enhanced cytotoxicity in vitro. It is believed that the present smart drug delivery platforms will hold great potential in imaging guided drug delivery and cancer therapy.

Abstract:Mesoporous silica nanoparticles (MSNs) are promising drug carriers for cancer therapy. Their functionalization with ligands for specific tissue/cell targeting and stimuli-responsive cap materials for sealing drugs within the pores of MSNs is extensively studied for biomedical and pharmaceutical applications. The objective of the present work was to establish MSNs as ideal nanocarriers of anticancer drugs such as 5-FU and silymarin by exploiting characteristics such as their large surface area, pore size, and biocompatibility. Furthermore, coating with various biopolymeric materials such as carboxymethyl chitosan-dopamine and hyaluronic acid-folic acid on their surface would allow them to play the role of ligands in the process of active targeting to tumor cells in which there is an overexpression of specific receptors for them. From the results obtained, it emerged, in fact, that these hybrid nanoparticles not only inhibit the growth of glioblastoma and breast cancer cells, but also act as pH-responsive release systems potentially useful as release vectors in tumor environments.

Abstract:It is urgent to develop functional nanocarriers for targeted drug therapy of cancer. This work developed a smart drug controlled release system based on the dendritic mesoporous silica nanoparticle (DMSN) for dual gatekeepers drug delivery. The DMSN-Cys grafted with disulfide bonds appeared as spheres of uniform size with a porous structure, which enabled a drug loading efficiency of 68.32%. Since the protonated amino groups of chitosan were responsive to pH and the disulfide bonds were sensitive to glutathione, the modified 5-Fu@DMSN-Cys/CS can be used for pH/GSH responsive drug delivery. Release behaviors showed excellent drug controlled release effects. Good biocompatibility and pronounced cytotoxicity of 5-Fu@DMSN-Cys/CS were confirmed by cytotoxicity test. Noted specially, the microcosmic adsorption interaction between drug molecules and DMSN-Cys has been evaluated through molecular dynamic simulation. Electrostatic potential and adsorption simulation confirmed that the driving force of 5-Fu adsorption was originated from electrostatic force and hydrogen bonding interaction between the nanocarriers and 5-Fu molecules. These results indicated that 5-Fu@DMSN-Cys/CS could be potential carriers for anticancer drug delivery.

Abstract:We aimed to investigate whether the combination of the modification of short chain amylose (SCA) with chitosan (CS) through the electrostatic interaction could be considered as a candidate for oral delivery of bioactive ingredients. Carboxymethyl short chain amylose (CMSCA) was synthesized by reacting SCA with monochloroacetic acid. The changes in SCA levels after the reaction were investigated by zeta-potential determination, Fourier transform infrared (FTIR) spectroscopy, differential scanning calorimetry, thermogravimetry and derivative thermogravimetry. Complex nanoparticles (NPs) were then synthesized using CMSCA and CS by ionic gelation. FTIR spectral analysis revealed that the complex NPs were synthesized by hydrogen bonding and electrostatic interactions between CMSCA and CS. CMSCA/CS NPs show an insulin encapsulation efficiency of 85.2% and exhibit sustained release of insulin in vitro. CMSCA/CS NPs were observed to show excellent cytocompatibility by cell culture. These findings demonstrated that CMSCA/CS NPs constructed by the ionic gelation method could be further exploited as a potential oral delivery system for peptide drugs.

Preparation and characterization of sodium alginate/phosphate-stabilized amorphous calcium carbonate nanocarriers and their application in the release of curcumin

Abstract:Phosphate-stabilized amorphous calcium carbonate (ACCP) has excellent biocompatibility, bioactivity, and biodegradability, and can be easily synthesized and stored. However, unmodified ACCP, as a controlled drug release carrier, decomposes rapidly in an acidic environment and highly depends on the system's pH value, which can not meet the need for long-term release of active substances, thus limiting its application scope. To realize the specific pH responsiveness of ACCP nanoparticles, we designed and synthesized monodisperse sodium alginate/ACCP (Alginate/ACCP) composite nanoparticles in this paper. After ultrasonic treatment, nanoparticles with an average particle size less than 200 nm could form stable water dispersion that could be dispersed for up to 10 d. Based on the specific pH sensitivity of sodium alginate, the drug-controlled release performance of composite nanoparticles and the therapeutic effect of drug-loaded nanoparticles on A549 cancer cells were studied. The results indicated that under the same pH condition, the curcumin (Cur) release rate of composite nanoparticles gradually decreased with sodium alginate addition. When the dosage of sodium alginate was 1.0 mg ml−1, the cumulative drug release rate of nanoparticles in 40 h was only about 35%. Besides, the drug-loaded nanoparticles showed the excellent killing ability of cancer cells, and the survival rate of cancer cells decreased in a concentration-dependent manner. Therefore, through reasonable optimization design, we can synthesize composite nanoparticles with excellent sustained-release properties to provide a new strategy for cancer cells' long-term treatment.

Abstract:Zinc-phosphate/hydroxyapatite hybrid form (ZP/HP) in core-shell nanostructure was developed and functionalized with both chitosan (CS@ZP/HP) and β-cyclodextrin (CD@ZP/HP) as bio-composite of enhanced physicochemical and biological properties. These structures were assessed as potential deliveries of 5-fluorouracil, exhibiting enhanced loading, release, and anti-cancer behaviors. The functionalization strongly prompted the loading effectiveness to be 301.3 mg/g (CS@ZP/HP) and 342.8 mg/g (CD@ZP/HP) instead of 238.9 mg/g for ZP/HP. The loading activities were assessed based on the hypotheses of traditional kinetic and isotherm models, alongside the computational variables of the monolayer model with a single energetic site as an advanced isotherm model. The functionalized versions exhibit much greater loading efficacy compared to ZP/HP as a result of the increment in the density of the existing loading sites [Nm(5-Fu) = 78.85 mg/g (ZP/HP), 93.87 mg/g (CS@ZP/HP), and 117.8 mg/g (CD@ZP/HP)]. Furthermore, the loading energies of approximately 40 kJ/mol, together with the loading potential of each receptor (n > 1) and Gaussian energies of approximately 8 kJ/mol, indicate the physical entrapment of 5-Fu molecules according to a vertical orientation. The materials mentioned verify long-term and continuous release characteristics. Following the modification processes, this behavior became faster as both CS@ZP/HP and CD@ZP/HP displayed complete release within 120 h at pH 1.2. The kinetic studies and diffusing exponent (>0.45) indicate that release characteristics are controlled by both diffusion and erosion processes. These carriers also markedly increase the cytotoxicity of 5-Fu against HCT-116 colorectal cancer cell lines: 5-Fu-ZP/HP (3.2% cell viability), 5-Fu-CS@ZP/HP (1.12% cell viability), and 5-Fu-CD@ZP/HP (0.63% cell viability).

Abstract:This study aimed to combine the active targeting function of folate (FA) receptor-mediated endocytosis with the pH-responsive drug delivery of poly (ethylene glycol)-grafted-poly (−amino ester) copolymers (PEG-PAE) in cancer targeting therapy. Herein, O -carboxymethylated chitosan (OCMC) was grafted with hydrophobic deoxycholic acid (DOCA). Further, PEG-PAE and FA-conjugated DOCA modified OCMC were synthesized to develop the potential cancer-targeted carrier (PEG-PAE-DOMC-FA), for which the structure was investigated by 1 H NMR and FTIR. Then riccardin D (RD) was successfully loaded for tumor-targeted drug delivery. The particle size, zeta potential, encapsulating efficiencies, and loading content profiles of PEG-PAE-DOMC-FA/RD showed a strong dependence on the environmental pH values. The cumulative release of PEG-PAE-DOMC-FA/RD at pH 5.0 (90.63 %) was higher than pH 7.4 (51.12 %), which also indicated the pH sensitivity. Moreover, a lower IC50 and higher coumarin-6 uptake were found because of the folate-receptor-mediated endocytosis. In pharmacokinetic study, PEG-PAE-DOMC-FA/RD significantly improved the mean retention time (MRT) and AUC (0-∞) from 7.89 h and 36.1 mg/L·h of control group to 10.03 h and 123.8 mg/L·h. In the xenograft mice model, stronger antitumor efficacy and lower toxicity were confirmed. In conclusion, the multi-functional micelles could be considered as a promising vehicle for delivering hydrophobic drugs to tumors.
