Abstract:Over the past decades, a tremendous rise in invasive fungal infection diseases attributed to the yeast Candida albicans in immunocompromised individuals poses a seriously challenging issue. Another concern is the emergence of multi-drug resistant pathogens to the existing medicines due to their overuse and misuse. It was recently reported that 25-55% of the mortality rate is caused by invasive infection. Despite a large variety of drugs being available to treat invasive candidiasis, only two of them contain a 1,2,4-triazole core, namely Fluconazole and itraconazole, which are efficient in treating infection induced by fungal Candida species. Moreover, long-term therapy associated with azole medications has led to an increase in azole resistance as well as a high risk of toxicity. Despite numerous outstanding achievements in antifungal drug discovery, development of novel, safer and potent antifungal agents while overcoming the resistance problem associated with the current drugs is becoming the main focus of medicinal chemists. Therefore, this review outlines the breakthroughs in medicinal chemistry research regarding 1,2,4- triazole-based derivatives as potential antifungal agents in the past decade. In addition, the structureactivity relationship of these compounds is also discussed.
Design, synthesis and in vitro biological studies of novel triazoles with potent and broad-spectrum antifungal activity

Abstract:A series of novel triazole derivatives containing aryl-propanamide side chains was designed and synthesised. In vitro antifungal activity studies demonstrated that most of the compounds inhibited the growth of six human pathogenic fungi. In particular, parts of phenyl-propionamide-containing compounds had excellent, broad-spectrum antifungal activity against Candida albicans SC5314, Cryptococcus neoformans 22-21, Candida glabrata 537 and Candida parapsilosis 22-20 with MIC values in the range of ≤0.125 μg/mL–4.0 μg/mL. In addition, compounds A1 , A2 , A6 , A12 and A15 showed inhibitory activities against fluconazole-resistant Candida albicans and Candida auris . Preliminary structure-activity relationships (SARs) are also summarised. Moreover, GC-MS analysis demonstrated that A1 , A3 , and A9 interfered with the C. albicans ergosterol biosynthesis pathway by inhibiting Cyp51. Molecular docking studies elucidated the binding modes of A3 and A9 with Cyp51. These compounds with low haemolytic activity and favourable ADME/T properties are promising for the development of novel antifungal agents.

Abstract:In order to develop new triazole derivatives, we optimized the lead compound a6 by structural modifications to obtain a series of (2R,3R)-3-((1-substituted-1H-1,2,3-triazol-4-yl) methoxy)-2-(2,4-difluorophenyl)-1-(1H-1,2,4-triazol-1-yl) butan-2-ol, compounds 5-36. Most of the target compounds exhibited excellent in vitro antifungal activity against Candida albicans 10231 and Candida glabrata 537 with MIC ≤ 0.125 µg/mL. Of particular note, compounds 6, 22, 28, 30 and 36 were highly active against Candida neoformans 32609 with MIC ≤ 0.125 µg/mL and showed broad-spectrum antifungal activity including against fluconazole-resistant Candida auris 891. In addition, compounds 6 and 22 demonstrated inhibitory effects on filamentation in the azole-resistant C. albicans isolate. Moreover, compounds 6 and 22 were minimally toxic to HUVECs and possessed weak inhibitory effects on the human CYP3A4 and CYP2D6. SARs and docking study further indicated that ortho-substituted groups in the terminal phenyl ring can promote the compounds to improve their antifungal activity.

Abstract:Twenty-eight novel triazole derivatives (compounds 1a-v, 2a-f) have been synthesized for structure-activity relationship studies as antifungal agents. The compounds were designed on the basis of the structure of fluconazole and molecular modeling of the active site of the cytochrome P450 14α-demethylase (CYP51). All of them are reported for the first time. Their chemical structures are characterized by (1) H NMR, (13) C NMR, LC-MS, and elemental analysis. The antifungal activities have been evaluated in vitro by measuring the minimal inhibitory concentrations (MICs). Compounds 1a-v exhibited higher activity against nearly all fungi tested except Aspergillus fumigatus (A. fum) than fluconazole (FCZ). The computational molecular docking experiments indicated that the inhibition of CYP51 involves a coordination bond with iron of the heme group, a hydrophilic H-bonding region, a hydrophobic region, and a narrow hydrophobic binding cleft.

Abstract:Life-threatening invasive fungal infections pose a serious threat to human health. A series of novel triazole derivatives bearing a pyrazole-methoxyl moiety were designed and synthesized in an effort to obtain antifungals with potent, broad-spectrum activity that are less susceptible to resistance. Most of these compounds exhibited moderate to excellent in vitro antifungal activities against Candida albicans SC5314 and 10,231, Cryptococcus neoformans 32,609, Candida glabrata 537 and Candida parapsilosis 22,019 with minimum inhibitory concentration (MIC) values of ≤0.125 μg/mL to 0.5 μg/mL. Use of recombinant Saccharomyces cerevisiae strains showed compounds 7 and 10 overcame the overexpression and resistant-related mutations in ERG11 of S. cerevisae and several pathogenic Candida spp. Despite being substrates of the C. albicans and Candida auris Cdr1 drug efflux pumps, compounds 7 and 10 showed moderate potency against five fluconazole (FCZ)-resistant fungi with MIC values from 2.0 μg/mL to 16.0 μg/mL. Growth kinetics confirmed compounds 7 and 10 had much stronger fungistatic activity than FCZ. For C. albicans, compounds 7 and 10 inhibited the yeast-to-hyphae transition, biofilm formation and destroyed mature biofilm more effectively than FCZ. Preliminary mechanism of action studies showed compounds 7 and 10 blocked the ergosterol biosynthesis pathway at Erg11, ultimately leading to cell membrane disruption. Further investigation of these novel triazole derivatives is also warranted by their predicted ADMET properties and low cytotoxicity.

Abstract:In order to search novel fungicides with higher activity, 28 new 1,2,4-triazolo[1,5-alpha]pyrimidine derivatives bearing 1,2,4-triazole heterocycle were synthesized. Their structures were characterized by H-1 NMR spectroscopy, mass spectrometry and elemental analyses. With triadimefon, validamycin and carbendazim as positive controls, the antifungal activities of 28 compounds against Fusarium oxysporum f. sp. vasinfectum, Gibberella sanbinetti, Cercospora beticola Sacc., Physaclospora piricola and Rhizoctonia solani were evaluated. Compound 2-[(5-(2,6-difluorobenzylthio)-4-phenyl-4H-1,2,4-triazol-3-yl)methylthio]-5,7-dimethyl[1,2,4]triazolo [1,5-a] pyrimidine (19) showed potent antifungal activities against G. sanbinetti, C. beticola, P piricola and R. solani. On the basis of the biological results, structure-activity relationships of these compounds were also discussed.

Abstract:A series of hybrid compounds with triazole and thiazolidine nuclei connected by a linker has been synthesized and extensively studied. Various synthetic methods for the target compounds have been tested. A microbiological assessment of the obtained compounds was carried out on strains of pathogenic fungi C. albicans, C. non-albicans, multidrug-resistant C. auris, Rhizopus arrhizus, Aspergillus spp. and some dermatophytes and other yeasts. The lowest obtained MIC values for target compounds lie between 0.003 μg/mL and 0.5 μg/mL and therefore the compounds are not inferior or several times better than commercial azole drugs. The length of the acylpiperazine linker has a limited effect on antifungal activity. Some bioisosteric analogues were tested in microbiological analysis, but turned out to be weaker than the leader in activity. The highest activity was demonstrated by a compound with para-chlorobenzylidene substituent in the thiazolidine fragment. Molecular modelling was used to predict binding modes of synthesized molecules and rationalize experimentally observed SAR. The leader compound is twice more effective in inhibiting the formation of germ tubes by Candida albicans yeast cells compared to voriconazole. An increased level of Pdr5, an azoles drug efflux pump was observed, but the increase is lower than that caused by azoles. The results can be useful for further development of more powerful and safe antifungal agents.

Abstract:Invasive fungal infections have high morbidity and mortality rates and have become one of the most serious threats to human health. In the present study, a series of triazole antifungal derivatives with phenylthiophene backbone were obtained by structural modification of the lead compound using Iodiconazole as the lead compound. Among them, compound 19g is a triazole antifungal compound with 4-chloro-2-fluoro phenylthiophene backbone, which showed optimal antifungal activity against Candida albicans, Cryptococcus neoformans, and Aspergillus, with a MIC80 value of 0.0625 μg/mL. In addition, compounds 19e, 19f, 19g, 19h, 19i and 19k exhibited different levels of inhibitory activity against fluconazole-resistant strains with MIC80 values ranging from 0.0625 μg/mL to 32 μg/mL. Since compound 19g had optimal in vitro antifungal activity, we selected 19g for human liver microsomal stability and CYP enzyme inhibition assays as well as further evaluated the inhibitory activity of compound 19g on normal and cancerous cells in humans. Finally, we verified the inhibitory effect of compound 19g on the filamentation of Candida albicans and determined the mechanism of action by sterol composition analysis.
Discovery of Novel Tetrazoles Featuring a Pyrazole Moiety as Potent and Highly Selective Antifungal Agents

Abstract:In pursuit of developing novel azole antifungals with potent activity and high selectivity, a series of (2R,3S)-3-(substituted-1H-pyrazol-3-yl)-2-(2,4-difluorophenyl)-1-(1H-tetrazol-1-yl)butan-2-ol derivatives were designed and synthesized based on our previous work. All compounds exhibited excellent in vitro antifungal activities against Candida spp. and Cryptococcus neoformans H99 with minimum inhibitory concentration values ranging from <0.008 to 4 μg/mL, with some even showing moderate activity against Aspergillus fumigatus 7544. The most active compounds (8, 11, 15, 24, and 25) displayed outstanding antifungal activity against six fluconazole-resistant C. auris clinical isolates and showed a potent inhibitory effect on biofilm formation of C. albicans SC5314 and C. neoformans H99. In addition, compounds 11 and 15 showed no inhibition of human CYP1A2 and low inhibitory activity against CYP3A4, indicating minimal risk of drug-drug interactions. Taken together, these promising tetrazoles with high in vitro potency and good safety profiles warrant further investigation.

Abstract:Fungal infections pose a significant threat to both morbidity and mortality, especially in immunocompromised patients, notably those with conditions such as AIDS. In the last two decades, the incidence of such fungal infections has increased significantly, especially due to Candida species. The increasing incidence of life-threatening fungal infections has increased the need for new drugs to treat these infections. In this study, 10 novel oxadiazole-triazole hybrid compounds were synthesized. Analysis of the obtained compounds provided by 1 H NMR (proton nuclear magnetic resonance), 13 C NMR (carbon nuclear magnetic resonance) spectroscopic methods and HRMS (high resolution mass spectrometry) spectrometric method. The inhibition effects of the obtained compounds on the 14α-demethylase enzyme were investigated. Among the synthesized compounds, 4a (contains 4-methyl substituent), 4c (contains 4-cyano substituent), 4d (contains 4-nitro substituent), 4e (contains 4-fluoro substituent), 4 g (contains 4‐bromo substituent) and 4j (contains 3,4-dichloro substituent) coded compounds were useful against C. parapsilosis strain; Compounds 4c, 4f and 4 g also showed significant inhibitory effect against C. albicans strain. Within the scope of the study, the interactions of 14α-demethylase enzyme active site and our compound were elucidated by molecular modeling and molecular dynamics studies. Binding points were determined by docking studies for the selected compounds in enzyme active site. The strongest interaction with 14α-demethylase enzyme active sites was observed in 4c and 4 g coded compounds.

Abstract:Due to increasing incidence of invasive fungal infections and severe drug resistance to triazole antifungal agents, a series of novel antifungal triazoles with substituted triazole-piperidine side chains were designed and synthesized. Most of the target compounds showed good inhibitory activity against a variety of clinically important fungal pathogens. In particular, compounds 8t and 8v were highly active against Candida albicans and Cryptococcus neoformans with MIC values in the range of 0.125 μg/mL to 0.0125 μg/mL. They represent promising leads for the development of new generation of triazole antifungal agents. Molecular docking studies revealed that the target compounds interacted with CACYP51 mainly through hydrophobic and Van der Waals interactions.

Abstract:Objective:To design and synthesize the triazole derivatives with the side chain containing tert-butyl and 4-substitued-piperazin-1-yl and to investigate their antifungal activities in vitro. Methods: The triazole derivatives were synthesized through several steps using 3, 3-dimethylbutyryl chloride as the starting material. All the target compounds were confirmed by 1HNMR to be 1-(1H-1, 2, 4-triazol-1-yl)-2-(3, 3-dimethyl)-3-(4-substituted)-piperazin-1-yl-2-propanols. Candida albicans (ATCC76615), Cryptococcus neoformans (ATCC32609), Candida tropicalis, Candida parapsilosis, Trichophyton rubrum, Microsporum lauosum, Fonsecaea compacta, and Aspergillus fumigatus were used to test the anti-fungal activities of the target compounds in vitro according to the National Committee for Clinical Laboratory Standards (NCCLS). Results: Eleven target compounds were obtained and all of them were firstly reported. We also found that all the 11 compounds had certain antifungal activities against the 8 tested strains. Conclusion: Compound 9 has a better effect against Cryptococcus neoformans than fluconazole and itraconazole; compound 8 has a better effect against Aspergillus fumigatus than fluconazole; these two compounds deserve further study.
Synthesis and Evaluation of the Antifungal Sensibility of Novel Thienopyridine 1,2,3-Triazole Derivatives

Abstract:The family of compounds known as azoles are part of most of the antimicrobial drugs used for the treatment of infections. Within this family triazoles have been extensively studied as pharmacophores with very promising results. In this work, four novel trisubstituted 1,2,3-triazole compounds with a thienopyridine moiety (1a,b; 2a,b) were synthesized through an azide-enolate 1,3-dipolar cycloaddition. Their cheminformatic properties were calculated using simulation software available online such as Molinspiration, Molsoft, Osiris Property Explorer, pkCSM, SwissADME, and GUSA. The results provided important information which allowed us to consider the evaluation of the antifungal activity of these novel compounds. Therefore, the antifungal activity of these compounds was evaluated in vitro against four filamentous fungi, including Aspergillus fumigatus ATCC 16907, Trichosporon cutaneum ATCC 28592, Rhizopus oryzae ATCC 10329, and Mucor hiemalis ATCC 8690; as well as six species of yeast from the Candida genus; C. albicans ATCC 10231, C. utilis ATCC 9226, C. tropicalis ATCC 13803, C. parapsilosis ATCC 22019, C. glabrata ATCC 34138 and C. krusei ATCC 14243 The sensibility studies suggest that compounds 1b, 2a and 2b can be considered candidates for complementary biological studies due to the exhibited antifungal activity. Resumen. La familia de compuestos conocidos como azoles forman parte de la mayoría de los medicamentos utilizados para el tratamiento de infecciones. Dentro de este grupo, los triazoles han sido extensamente estudiados como farmacóforos con resultados muy prometedores. En este trabajo, se sintetizaron cuatro nuevos 1,2,3-triazoles trisustituidos, que incluyen un anillo de tienopiridina en su estructura (1a,b; 2a,b) a través de una cicloadición 1,3-dipolar del tipo azida-enolato. Sus propiedades quimio informáticas fueron calculadas utilizando programas de simulación encontrados en línea como Molinspiration, Molsoft, Osiris Property Explorer, pkCSM, SwissADME y GUSAR. Los resultados obtenidos presentaron información importante que permitió considerar la evaluación de la actividad antifúngica de estos nuevos compuestos. Por lo tanto, esta actividad fue evaluada in vitro en cuatro cepas de hogos filamentosos, incluyendo Aspergillus fumigatus ATCC 16907, Trichosporon cutaneum ATCC 28592, Rhizopus oryzae ATCC 10329, and Mucor hiemalis ATCC 8690, así como también seis especies de levaduras del género Candida; C. albicans ATCC 10231, C. utilis ATCC 9226, C. tropicalis ATCC 13803, C. parapsilosis ATCC 22019, C. glabrata ATCC 34138 and C. krusei ATCC 14243. En estos estudios se observó que los compuestos 1a, 2a, y 2b pueden ser considerados para estudios posteriores de la evaluación biológica debido a la inhibición observada.

Abstract:The clinical prevalence of antifungal drug resistance has been increasing over recent years, resulting in the failure of treatments. In an attempt to overcome this critical problem, we sought novel synergistic enhancers to restore the effectiveness of fluconazole against resistant Candida albicans. Based on the structural optimization of hit compound 8 from our in-house library, a series of novel 1,3,5-triazines derivatives was designed, synthesized, and biologically evaluated for synergistic activity in combination with fluconazole. Among them, compounds 10a–o, which contain thiosemicarbazides side chains, exhibited excellent in vitro synergistic antifungal potency (MIC80 = 0.125–2.0 μg/mL, FICI range from 0.127 to 0.25). Interestingly, compound 10l exhibited moderate C. albicans activity as monotherapy with an MIC80 value of 4.0 μg/mL, and also on several Cryptococcus strains (MIC80 ranging from ≤ 0.125–0.5 μg/mL) and C. glabrata (MIC80 ≤ 0.125 μg/mL). These effects were fungal-selective, with much lower levels of cytotoxicity towards human umbilical vein endothelial cells. Here, we report a series of thiosemicarbazides containing 1,3,5-triazines derivatives as potent synergists with fluconazole, and have preliminarily validated compound 10l as a promising antifungal lead for further investigation.

Abstract:Aim To study antifungal activity of the triazole derivatives bearing the side chain containing tert-butyl and 4-(4-alkyloxyphenyl)-piperazin-1-yl and to compare their antifungal activities with that of fluconazole and itraconazole.Methods According to the structure of fluconazole,ten target compounds were designed and synthesized.All of them were confirmed by()~1H-NMR and IR spectra respectively.The MIC_(80) of all the target compounds were determined by the method recommended by the national committee for clinical laboratory standards(NCCLS) using the RPMI-1640 test medium.Results All the target compounds were firstly reported.The results of the preliminary antifungal test show that all the target compounds exhibit potent antifungal activities to a certain extent.The activities of four target compounds are 4 times as high as that of fluconazole and equal to that of itraconazole against Candida albicans in vitro.Conclusion More hydrophobic groups can be introduced to design triazole compounds and stereochemistry have important influence on the antifungal activities of the target compounds.

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:A series of triazole derivatives containing phenylethynyl pyrazole moiety as side chain were designed, synthesized, and most of them exhibited good in vitro antifungal activities. Especially, compounds 5k and 6c showed excellent in vitro activities against C. albicans (MIC = 0.125, 0.0625 μg/mL), C. neoformans (MIC = 0.125, 0.0625 μg/mL), and A. fumigatus (MIC = 8.0, 4.0 μg/mL). Compound 6c also exerted superior activity to compound 5k and fluconazole in inhibiting hyphae growth of C. albicans and inhibiting drug-resistant strains of C. albicans, and it could reduce fungal burdens in mice kidney at a dosage of 1.0 mg/kg. An in vivo efficacy evaluation indicated that 6c could effectively protect mice models from C. albicans infection at doses of 0.5, 1.0, and 2.0 mg/kg. These results suggested that compound 6c deserves further investigation.

Abstract:Objective: Present research communication was towards the investigation of antifungal minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) activity of some substituted clubbed thiazole-1,3,5-triazines derivatives and effect of physicochemical properties on bioactivity. Material and methods: MIC and MFC were evaluated against Candida albicans, Candida glabrata, Cryptococcus neoformans and Aspergillus niger using modified microdilution method recommended by CLSI. Cytotoxicity was determinate on the viability of marine shrimp larvaes. SAR and physicochemical correlations were studied by Molinspiration software. Results: The 5 and 9 derivatives showed an excellent antifungal activity with MIC lower than fluconazole and equivalent to amphotericin B specially against C. albicans and C. glabrata. The toxicity of these two derivatives was non-existent for 5 and moderate for 9 at the used concentration. SAR study around prototype molecule suggests that presence of di-hydrophobic fragment on 1,3,5-triazine is necessary for antifungal activity than halogen substituted aromatic amine. Conclusion: On the basis of selectivity, potency and non-toxicity, we have obtained two molecules (5 and 9) as prospective leads for further research work on 1,3,5-triazine as antifungal drug.

Abstract:In an effort to develop novel azole antifungals with potent activity and high selectivity, a series of (2 R ,3 R )-3-((3-substitutied-phenyl-isoxazol-5-yl)methoxy)-2-(2,4-difluorophenyl)-1-(1 H -tetrazol-1-yl)butan-2-ol derivatives were designed and synthesized based on our previously work. All compounds exhibited moderate to excellent in vitro antifungal activities against Candida albicans SC5314 and Cryptococcus neoformans H99, but inactive against Aspergillus fumigatus 7544. Among them, the most active compound 10h displayed outstanding antifungal activity against fluconazole-resistant C. albicans 103, C. glabrata 537 and C. auris 922 with MIC values of ≤0.008 μg/mL. In addition, compound 10h was superior to FLC in inhibiting the filamentation of FLC-resistant C. albicans 103. Notably, compound 10h showed no inhibition of human CYP3A4 with IC 50 values of >100 μM, low cytotoxicity at 32 μg/mL and low hERG inhibition with IC 50 values of 6.22 μM, suggesting a low risk of drug-drug interactions and good safety profiles. Furthermore, compound 10h exhibited excellent PK profiles and showed remarkable in vivo efficacy in a mouse model of C. albicans and C. neoformans infection. Taken together, compound 10h will be further investigated as a promising lead antifungal agent.

Abstract:Scheme: Synthesis of quinazolinone with 1,2,3‐triazole moieties and their 2D structure and Ligplot docking interactions Scheme: Synthesis of quinazolinone with 1,2,3‐triazole moieties. Novel 1,2,3‐triazole acetamide linked quinazolinone derivatives were synthesized via click reaction, by reacting of 3‐methyl‐2‐(prop‐2‐yn‐1‐ylthio)quinazolin‐4(3H)‐one with various aryl azides as prominent fungal pathogen Candida albicans interactions. The prepared triazole compounds were characterized using mass spectrometry, 1H NMR, 13C NMR, and IR spectroscopic techniques. Compounds were screened for their in vitro antibacterial and antifungal activity against a variety of microorganisms namely, Bacillus subtilis, Enterococcus faecalis, Klebsiella pneumonia, Escherichia coli, Aspergillus fumigatus, Candida albicans, and Aspergillus clavatus. Quinazolinone linked p‐hydroxyphenyl, o, p‐dihydroxyphenyl and o‐dimethylamino phenyl triazoles showed a remarkable antibacterial activity against E. faecalis with MIC values of 3.20±0.01, 4.01±0.04, and 4.10±0.01 μg mL−1, and screened compounds p‐nitrophenyl substituted triazole, and p‐methylphenyl substituted triazole displayed a significant antifungal activity against C. albicans with MICs of 3.16±0.01, 4.06±0.03 μg mL−1, compared to itraconazole (MIC=3.32±0.02 μg mL−1). For further exploration of the anti‐fungal mechanism of action, molecular docking was carried out for these compounds in C. albicans active site as one of the important antifungal inhibitors (PDB: 1A19). Furthermore, the ADMET profile was evaluated for all the final triazole compounds in contrast to reference drugs moxifloxacin and itraconazole. In conclusion, we discovered a novel quinazolinone linked 1,2,3‐triazoles with promising antimicrobial activity and a favorable pharmacokinetic profile.
