Abstract:Coherent fluctuations of spontaneous brain activity are present in distinct functional-anatomic brain systems during undirected wakefulness. However, the behavioral significance of this spontaneous activity has only begun to be investigated. Our previous studies have demonstrated that successful memory formation requires coordinated neural activity in a distributed memory network including the hippocampus and posteromedial cortices, specifically the precuneus and posterior cingulate (PPC), thought to be integral nodes of the default network. In this study, we examined whether intrinsic connectivity during the resting state between the hippocampus and PPC can predict individual differences in the performance of an associative memory task among cognitively intact older individuals. The intrinsic connectivity, between regions within the hippocampus and PPC that were maximally engaged during a subsequent memory fMRI task, was measured during a period of rest prior to the performance of the memory paradigm. Stronger connectivity between the hippocampal and posteromedial regions during rest predicted better performance on the memory task. Furthermore, hippocampal-PPC intrinsic connectivity was also significantly correlated with episodic memory measures on neuropsychological tests, but not with performance in non-memory domains. Whole-brain exploratory analyses further confirmed the spatial specificity of the relationship between hippocampal-default network posteromedial cortical connectivity and memory performance in older subjects. Our findings provide support for the hypothesis that one of the functions of this large-scale brain network is to subserve episodic memory processes. Research is ongoing to determine if impaired connectivity between these regions may serve as a predictor of memory decline related to early Alzheimer's disease.

Abstract:When reading a narrative, comprehension and retention of information benefit considerably from the use of situation models--coherent representations of the characters, locations, and activities described in the text. Here we used functional magnetic resonance imaging (fMRI) to explore the neural mechanisms supporting situation model processing. Participants read blocks of sentences that were either unrelated to one another or formed coherent narratives. A timecourse-based approach was used to identify regions that differentiated narrative-level comprehension from sentence-level comprehension. Most brain regions that showed modulation of activation during narrative-level comprehension were also modulated to a lesser extent during sentence-level comprehension, suggesting a shared reliance on general coherence-building mechanisms. However, tentative evidence was found for narrative-specific activation in dorsomedial prefrontal cortex. Additional analyses identified spatiotemporally distinct neural contributions to situation model processing, with posterior parietal regions supporting situation model construction and frontotemporal regions supporting situation model maintenance. Finally, a set of subsequent memory analyses demonstrated that the boost in comprehension and memory performance observed for coherent materials was attributable to the use of integrative situation models rather than lower-level differences in sentence-level or word-level encoding. These results clarify the functional contributions of distinct brain systems to situation model processing and their mapping onto existing psychological models of narrative comprehension.
Verbal working memory and syntactic comprehension segregate into the dorsal and ventral streams

Abstract:Syntactic processing and verbal working memory are both essential components to sentence comprehension. Nonetheless, the separability of these systems in the brain remains unclear. To address this issue, we performed causal-inference analyses based on lesion and connectome network mapping using MRI and behavioral testing in 103 individuals with chronic post-stroke aphasia. We employed a rhyme judgment task with heavy working memory load without articulatory confounds, controlling for the overall ability to match auditory words to pictures and to perform a metalinguistic rhyme judgment, isolating the effect of working memory load. We assessed noncanonical sentence comprehension, isolating syntactic processing by incorporating residual rhyme judgment performance as a covariate for working memory load. Voxel-based lesion analyses and structural connectome-based lesion symptom mapping controlling for total lesion volume were performed, with permutation testing to correct for multiple comparisons (4,000 permutations). We observed that effects of working memory load localized to dorsal stream damage: posterior temporal-parietal lesions and frontal-parietal white matter disconnections. These effects were differentiated from syntactic comprehension deficits, which were primarily associated with ventral stream damage: lesions to temporal lobe and temporal-parietal white matter disconnections, particularly when incorporating the residual measure of working memory load as a covariate. Our results support the conclusion that working memory and syntactic processing are associated with distinct brain networks, largely loading onto dorsal and ventral streams, respectively.

Expanding the language network: Domain-specific hippocampal recruitment during high-level linguistic processing

Abstract:Abstract Language processing requires us to encode linear relations between acoustic forms and map them onto hierarchical relations between meaning units. Such relational binding of linguistic elements might recruit the hippocampus given its engagement by similar operations in other cognitive domains. Historically, hippocampal engagement in online language use has received little attention because patients with hippocampal damage are not aphasic. However, recent studies have found that these patients exhibit language impairments when the demands on flexible relational binding are high, suggesting that the hippocampus does, in fact, contribute to linguistic processing. A fundamental question is thus whether language processing engages domain-general hippocampal mechanisms that are also recruited across other cognitive processes or whether, instead, it relies on certain language-selective areas within the hippocampus. To address this question, we conducted the first systematic analysis of hippocampal engagement during comprehension in healthy adults ( n =150 across three experiments) using fMRI. Specifically, we functionally localized putative “language-regions” within the hippocampus using a language comprehension task, and found that these regions (i) were selectively engaged by language but not by six non-linguistic tasks; and (ii) were coupled in their activity with the cortical language network during both “rest” and especially story comprehension, but not with the domain-general “multiple-demand (MD)” network. This functional profile did not generalize to other hippocampal regions that were localized using a non-linguistic, working memory task. These findings suggest that some hippocampal mechanisms that maintain and integrate information during language comprehension are not domain-general but rather belong to the language-specific brain network. Significance statement According to popular views, language processing is exclusively supported by neocortical mechanisms. However, recent patient studies suggest that language processing may also require the hippocampus, especially when relations among linguistic elements have to be flexibly integrated and maintained. Here, we address a core question about the place of the hippocampus in the cognitive architecture of language: are certain hippocampal operations language-specific rather than domain-general? By extensively characterizing hippocampal recruitment during language comprehension in healthy adults using fMRI, we show that certain hippocampal subregions exhibit signatures of language specificity in both their response profiles and their patterns of activity synchronization with known functional regions in the neocortex. We thus suggest that the hippocampus is a satellite constituent of the language network.
Differentiable Processing of Objects, Associations, and Scenes within the Hippocampus

Abstract:The hippocampus is known to be important for a range of cognitive functions, including episodic memory, spatial navigation, and thinking about the future. However, researchers have found it difficult to agree on the exact nature of this brain structure's contribution to cognition. Some theories emphasize the role of the hippocampus in associative processes. Another theory proposes that scene construction is its primary role. To directly compare these accounts of hippocampal function in human males and females, we devised a novel mental imagery paradigm where different tasks were closely matched for associative processing and mental construction, but either did or did not evoke scene representations, and we combined this with high-resolution functional MRI. The results were striking in showing that different parts of the hippocampus, along with distinct cortical regions, were recruited for scene construction or nonscene-evoking associative processing. The contrasting patterns of neural engagement could not be accounted for by differences in eye movements, mnemonic processing, or the phenomenology of mental imagery. These results inform conceptual debates in the field by showing that the hippocampus does not seem to favor one type of process over another; it is not a story of exclusivity. Rather, there may be different circuits within the hippocampus, each associated with different cortical inputs, which become engaged depending on the nature of the stimuli and the task at hand. Overall, our findings emphasize the importance of considering the hippocampus as a heterogeneous structure, and that a focus on characterizing how specific portions of the hippocampus interact with other brain regions may promote a better understanding of its role in cognition.SIGNIFICANCE STATEMENT The hippocampus is known to be important for a range of cognitive functions, including episodic memory, spatial navigation, and thinking about the future. Abstract:During discourse comprehension, every new word adds to an evolving representation of meaning that accumulates over consecutive sentences and constrains the next words. To minimize repetition and utterance length, languages use pronouns, like the word 'she', to refer to nouns and phrases that were previously introduced. It has been suggested that language comprehension requires that pronouns activate the same neuronal representations as the nouns themselves. Here, we test this hypothesis by recording from individual neurons in the human hippocampus during a reading task. We found that cells that are selective to a particular noun are later reactivated by pronouns that refer to the cells' preferred noun. These results imply that concept cells contribute to a rapid and dynamic semantic memory network which is recruited during language comprehension. This study uniquely demonstrates, at the single-cell level, how memory and language are linked.

The Neural Architecture of the Language Comprehension Network: Converging Evidence from Lesion and Connectivity Analyses

Abstract:While traditional models of language comprehension have focused on the left posterior temporal cortex as the neurological basis for language comprehension, lesion and functional imaging studies indicate the involvement of an extensive network of cortical regions. However, the full extent of this network and the white matter pathways that contribute to it remain to be characterized. In an earlier voxel-based lesion-symptom mapping analysis of data from aphasic patients (Dronkers et al., 2004), several brain regions in the left hemisphere were found to be critical for language comprehension: the left posterior middle temporal gyrus, the anterior part of Brodmann's area 22 in the superior temporal gyrus (anterior STG/BA22), the posterior superior temporal sulcus (STS) extending into Brodmann's area 39 (STS/BA39), the orbital part of the inferior frontal gyrus (BA47), and the middle frontal gyrus (BA46). Here, we investigated the white matter pathways associated with these regions using diffusion tensor imaging from healthy subjects. We also used resting-state functional magnetic resonance imaging data to assess the functional connectivity profiles of these regions. Fiber tractography and functional connectivity analyses indicated that the left MTG, anterior STG/BA22, STS/BA39, and BA47 are part of a richly interconnected network that extends to additional frontal, parietal, and temporal regions in the two hemispheres. The inferior occipito-frontal fasciculus, the arcuate fasciculus, and the middle and inferior longitudinal fasciculi, as well as transcallosal projections via the tapetum were found to be the most prominent white matter pathways bridging the regions important for language comprehension. The left MTG showed a particularly extensive structural and functional connectivity pattern which is consistent with the severity of the impairments associated with MTG lesions and which suggests a central role for this region in language comprehension.

Abstract:Mild cognitive impairment (MCI) patients report memory problems greater than those normally expected with ageing, but do not fulfil criteria for clinically probable Alzheimer's disease. Accumulating evidence demonstrates that impaired performance on the Paired Associates Learning (PAL) test from the Cambridge Neuropsychological Test Automated Battery (CANTAB) may be sensitive and specific for early and differential diagnosis of Alzheimer's disease. We adapted the basic CANTAB PAL task for functional magnetic resonance imaging (fMRI) in order to examine the functional brain deficits, at encoding and retrieval separately, in patients with MCI compared to healthy matched volunteers. As well as investigating the main effects of encoding and retrieval, we characterized neural responses in the two groups to increasing memory load. We focused on changes in BOLD response in the hippocampus and related structures, as an a priori region of interest based on what is known about the neuropathology of the early stages of Alzheimer's disease and previous information on the neural substrates of the PAL task. We also used structural MRI in the same patients to assess accompanying structural brain abnormalities associated with MCI. In terms of the BOLD response, the bilateral hippocampal activation in the MCI and control groups depended upon load, the MCI patients activating significantly more than controls at low loads and significantly less at higher loads. There were no other differences between MCI patients and controls in terms of the neural networks activated during either encoding or retrieval of the PAL task, including the prefrontal, cingulate and temporal cortex. The functional deficit in hippocampal activation in the MCI patients was accompanied by structural differences in the same location, suggesting that the decrease in hippocampal activation may be caused by a decrease in the amount of grey matter. This is one of the first studies to have used both encoding and retrieval phases of a memory paradigm for fMRI in MCI patients, and to have shown that the BOLD response in MCI patients can show both hyperactivation and hypoactivation in the same individuals as a function of memory load and encoding/retrieval. The findings suggest that performance on PAL might be a useful cognitive biomarker for early detection of Alzheimer's disease, especially when used in conjunction with neuroimaging.

Abstract:Purpose of review: The neural basis of semantic memory has not only theoretical interest, but also implications for several neurodegenerative disorders such as Alzheimer's disease and semantic dementia. Recent findings: The main focus of functional imaging studies is to disentangle the contribution of several interacting factors to the landscape of cerebral activation observed in normal individuals performing tasks requiring access to semantic knowledge (e.g. picture naming, word comprehension sentence verification). The main factors found to play a role are the modality of stimulus presentation (e.g. visual, verbal); the domain to which the stimuli belong (e.g. concrete or abstract entities; natural kinds or artifacts); and the type of knowledge accessed (e.g. visual properties vs. action properties). Another area of investigation is the neural correlates of semantic processes, such as selection among alternatives or information retrieval. Summary: The results of imaging studies confirm the neuropsychological concept that semantic knowledge and its usage depend upon a widely distributed network of brain areas. Specificity within this extended network, which includes a number of areas involved in perceptual and action processing, as well as the language areas, is related both to the type of knowledge and to different conceptual domains. The heterogeneous pattern of semantic memory dysfunction in neurological disorder may be a consequence of this complex neural organization.
Hippocampal Functions Modulate Transfer-Appropriate Cortical Representations Supporting Subsequent Memory

Abstract:The hippocampus plays a central role as a coordinate system or index of information stored in neocortical loci. Nonetheless, it remains unclear how hippocampal processes integrate with cortical information to facilitate successful memory encoding. Thus, the goal of the current study was to identify specific hippocampal-cortical interactions that support object encoding. We collected fMRI data while 19 human participants (7 female and 12 male) encoded images of real-world objects and tested their memory for object concepts and image exemplars (i.e., conceptual and perceptual memory). Representational similarity analysis revealed robust representations of visual and semantic information in canonical visual (e.g., occipital cortex) and semantic (e.g., angular gyrus) regions in the cortex, but not in the hippocampus. Critically, hippocampal functions modulated the mnemonic impact of cortical representations that are most pertinent to future memory demands, or transfer-appropriate representations Subsequent perceptual memory was best predicted by the strength of visual representations in ventromedial occipital cortex in coordination with hippocampal activity and pattern information during encoding. In parallel, subsequent conceptual memory was best predicted by the strength of semantic representations in left inferior frontal gyrus and angular gyrus in coordination with either hippocampal activity or semantic representational strength during encoding. We found no evidence for transfer-incongruent hippocampal-cortical interactions supporting subsequent memory (i.e., no hippocampal interactions with cortical visual/semantic representations supported conceptual/perceptual memory). Collectively, these results suggest that diverse hippocampal functions flexibly modulate cortical representations of object properties to satisfy distinct future memory demands.Significance Statement The hippocampus is theorized to index pieces of information stored throughout the cortex to support episodic memory. Yet how hippocampal processes integrate with cortical representation of stimulus information remains unclear. Using fMRI, we examined various forms of hippocampal-cortical interactions during object encoding in relation to subsequent performance on conceptual and perceptual memory tests. Our results revealed novel hippocampal-cortical interactions that utilize semantic and visual representations in transfer-appropriate manners: conceptual memory supported by hippocampal modulation of frontoparietal semantic representations, and perceptual memory supported by hippocampal modulation of occipital visual representations. These findings provide important insights into the neural mechanisms underlying the formation of information-rich episodic memory and underscore the value of studying the flexible interplay between brain regions for complex cognition.
The Human Posterior Cingulate, Retrosplenial, and Medial Parietal Cortex Effective Connectome, and Implications for Memory and Navigation

Abstract:The human posterior cingulate, retrosplenial, and medial parietal cortex are involved in memory and navigation. The functional anatomy underlying these cognitive functions was investigated by measuring the effective connectivity of these Posterior Cingulate Division (PCD) regions in the Human Connectome Project-MMP1 atlas in 171 HCP participants, and complemented with functional connectivity and diffusion tractography. First, the postero-ventral parts of the PCD (31pd, 31pv, 7m, d23ab, and v23ab) have effective connectivity with the temporal pole, inferior temporal visual cortex, cortex in the superior temporal sulcus implicated in auditory and semantic processing, with the reward-related vmPFC and pregenual anterior cingulate cortex, with the inferior parietal cortex, and with the hippocampal system. This connectivity implicates it in hippocampal episodic memory, providing routes for "what," reward and semantic schema-related information to access the hippocampus. Second, the antero-dorsal parts of the PCD (especially 31a and 23d, PCV, and also RSC) have connectivity with early visual cortical areas including those that represent spatial scenes, with the superior parietal cortex, with the pregenual anterior cingulate cortex, and with the hippocampal system. This connectivity implicates it in the "where" component for hippocampal episodic memory and for spatial navigation. The dorsal-transitional-visual (DVT) and ProStriate regions where the retrosplenial scene area is located have connectivity from early visual cortical areas to the parahippocampal scene area, providing a ventromedial route for spatial scene information to reach the hippocampus. These connectivities provide important routes for "what," reward, and "where" scene-related information for human hippocampal episodic memory and navigation. The midcingulate cortex provides a route from the anterior dorsal parts of the PCD and the supracallosal part of the anterior cingulate cortex to premotor regions.
Exploring anterograde memory: a volumetric MRI study in patients with mild cognitive impairment

Abstract:Abstract Background The aim of this volumetric study was to explore the neuroanatomical correlates of the Free and Cued Selective Reminding Test (FCSRT) and the Delayed Matching-to-Sample—48 items (DMS-48), two tests widely used in France to assess verbal and visual anterograde memory. We wanted to determine to what extent the two tests rely on the medial temporal lobe, and could therefore be predictive of Alzheimer’s disease, in which pathological changes typically start in this region. Methods We analysed data from a cohort of 138 patients with mild cognitive impairment participating in a longitudinal multicentre clinical research study. Verbal memory was assessed using the FCSRT and visual recognition memory was evaluated using the DMS-48. Performances on these two tests were correlated to local grey matter atrophy via structural MRI using voxel-based morphometry. Results Our results confirm the existence of a positive correlation between the volume of the medial temporal lobe and the performance on the FCSRT, prominently on the left, and the performance on the DMS-48, on the right, for the whole group of patients (family-wise error, P < 0.05). Interestingly, this region remained implicated only in the subgroup of patients who had deficient scores on the cued recall of the FCSRT, whereas the free recall was associated with prefrontal aspects. For the DMS-48, it was only implicated for the group of patients whose performances declined between the immediate and delayed trial. Conversely, temporo-parietal cortices were implicated when no decline was observed. Within the medial temporal lobe, the parahippocampal gyrus was prominently involved for the FCSRT and the immediate trial of the DMS-48, whereas the hippocampus was solely involved for the delayed trial of the DMS-48. Conclusions The two tests are able to detect an amnestic profile of the medial temporal type, under the condition that the scores remain deficient after the cued recall of the FCSRT or decline on the delayed recognition trial of the DMS-48. Strategic retrieval as well as perceptual/attentional processes, supported by prefrontal and temporo-parietal cortices, were also found to have an impact on the performances. Finally, the implication of the hippocampus appears time dependent, triggered by a longer delay than the parahippocampus, rather than determined by the sense of recollection or the encoding strength associated with the memory trace.
Computational Analysis of the Role of the Hippocampus in Memory.

Abstract:The authors draw together the results of a series of detailed computational studies and show how they are contributing to the development of a theory of hippocampal function. A new part of the theory introduced here is a quantitative analysis of how backprojections from the hippocampus to the neocortex could lead to the recall of recent memories. The theory is then compared with other theories of hippocampal function. First, what is computed by the hippocampus is considered. The hypothesis the authors advocate, on the basis of the effects of damage to the hippocampus and neuronal activity recorded in it, is that it is involved in the formation of new memories by acting as an intermediate-term buffer store for information about episodes, particularly for spatial, but probably also for some nonspatial, information. The authors analyze how the hippocampus could perform this function, by producing a computational theory of how it operates, based on neuroanatomical and neurophysiological information about the different neuronal systems contained within the hippocampus. Key hypotheses are that the CA3 pyramidal cells operate as a single autoassociation network to store new episodic information as it arrives via a number of specialized preprocessing stages from many association areas of the cerebral cortex, and that the dentate granule cell/mossy fiber system is important, particularly during learning, to help to produce a new pattern of firing in the CA3 cells for each episode. The computational analysis shows how many memories could be stored in the hippocampus and how quickly the CA3 autoassociation system would operate during recall. The analysis is then extended to show how the CA3 system could be used to recall a whole episodic memory when only a fragment of it is presented. It is shown how this recall could operate using modified synapses in backprojection pathways from the hippocampus to the cerebral neocortex, resulting in reinstatement of neuronal activity in association areas of the cerebral neocortex similar to that present during the original episode. The recalled information in the cerebral neocortex could then be used by the neocortex in the formation of long-term memories. (C) 1994 Wiley-Liss, Inc.

Abstract:The cortical connections of the human hippocampal memory system are fundamental to understanding its operation in health and disease, especially in the context of the great development of the human cortex. The functional connectivity of the human hippocampal system was analyzed in 172 participants imaged at 7T in the Human Connectome Project. The human hippocampus has high functional connectivity not only with the entorhinal cortex, but also with areas that are more distant in the ventral 'what' stream including the perirhinal cortex and temporal cortical visual areas. Parahippocampal gyrus TF in humans has connectivity with this ventral 'what' subsystem. Correspondingly for the dorsal stream, the hippocampus has high functional connectivity not only with the presubiculum, but also with areas more distant, the medial parahippocampal cortex TH which includes the parahippocampal place or scene area, the posterior cingulate including retrosplenial cortex, and the parietal cortex. Further, there is considerable cross connectivity between the ventral and dorsal streams with the hippocampus. The findings are supported by anatomical connections, which together provide an unprecedented and quantitative overview of the extensive cortical connectivity of the human hippocampal system that goes beyond hierarchically organised and segregated pathways connecting the hippocampus and neocortex, and leads to new concepts on the operation of the hippocampal memory system in humans.
Brain Network for the Core Deficits of Semantic Dementia: A Neural Network Connectivity-Behavior Mapping Study

Abstract:Individuals with semantic dementia (SD) typically suffer from selective semantic deficits due to degenerative brain atrophy. Although some brain regions have been found to be correlated with the semantic impairments of SD patients, it is unclear if the damage is actually responsible for SD patients' semantic disorders because these findings were primarily obtained by examining the roles of local individual regions themselves without considering the influence of other regions that are functionally or structurally connected to the local individual regions. To resolve this problem, we investigated, from the brain network perspective, the relationship between the brain-network measures of regions and connections with semantic performance in 17 SD patients. We found that the severity of semantic deficits of SD patients was significantly correlated with the degree centrality values of the left anterior hippocampus (aHIP). Moreover, the semantic performance of the patients was also significantly correlated with the strength of gray matter functional connectivity of this region and two other regions: the left temporal pole/insula (TP/INS) and the left middle temporal gyrus. We further observed that the strength of the white matter structural connectivity of the left aHIP-left TP/INS tract could effectively predict the semantic performance of SD patients. When we controlled for a wide range of potential confounding factors (e.g., total gray matter volume), the above effects still held well. These findings revealed the critical brain network with the left aHIP as the center that could be contributing to the semantic impairments of SD.
Semantic representations in inferior frontal and lateral temporal cortex during picture naming, reading, and repetition

Abstract:The current multivariate pattern analysis (MVPA) study revealed a neurobiological basis for the distinction between task‐dependent and task‐independent retrieval of word meaning. Whereas the left BA45 and supramarginal gyrus showed a semantic similarity effect during picture naming only, the anterior STG and ventral BA44/frontal operculum, among other regions, showed a task‐independent semantic similarity effect. Reading, naming, and repetition are classical neuropsychological tasks widely used in the clinic and psycholinguistic research. While reading and repetition can be accomplished by following a direct or an indirect route, pictures can be named only by means of semantic mediation. By means of fMRI multivariate pattern analysis, we evaluated whether this well‐established fundamental difference at the cognitive level is associated at the brain level with a difference in the degree to which semantic representations are activated during these tasks. Semantic similarity between words was estimated based on a word association model. Twenty subjects participated in an event‐related fMRI study where the three tasks were presented in pseudo‐random order. Linear discriminant analysis of fMRI patterns identified a set of regions that allow to discriminate between words at a high level of word‐specificity across tasks. Representational similarity analysis was used to determine whether semantic similarity was represented in these regions and whether this depended on the task performed. The similarity between neural patterns of the left Brodmann area 45 (BA45) and of the superior portion of the left supramarginal gyrus correlated with the similarity in meaning between entities during picture naming. In both regions, no significant effects were seen for repetition or reading. The semantic similarity effect during picture naming was significantly larger than the similarity effect during the two other tasks. In contrast, several regions including left anterior superior temporal gyrus and left ventral BA44/frontal operculum, among others, coded for semantic similarity in a task‐independent manner. These findings provide new evidence for the dynamic, task‐dependent nature of semantic representations in the left BA45 and a more task‐independent nature of the representational activation in the lateral temporal cortex and ventral BA44/frontal operculum.

Integrating episodic and spatial context signals in the hippocampus

Abstract:Episodic and spatial memory are the two key components of the mnemonic system in humans. Episodic memory enables us to remember events from the past whereas spatial memory enables us to form a map-like representation of the environment. Interestingly, these seemingly different functions rely on the same brain structure: the hippocampus. Yet, how the hippocampus supports both at the same time remains unclear. Here, we tested the hypotheses that the hippocampus supports these two systems either via a common coding mechanism or via a parallel processing mechanism. To this end, we combined functional magnetic resonance imaging (fMRI) with an episodic life-simulation task and a spatial virtual reality task to manipulate episodic and spatial context associations of objects. We then investigated fMRI adaptation effects between these objects as a result of shared contexts. We found that the fMRI signal in the anterior hippocampus scaled with the combined prediction of shared episodic and spatial contexts, in line with the idea of a common coding mechanism. We found no evidence for a parallel processing mechanism, as there were no differences between episodic and spatial effects. The common coding effect for episodic and spatial memory dovetails with the broader notion of domain-general hippocampal cognitive maps.

Persistent schema-dependent hippocampal-neocortical connectivity during memory encoding and postencoding rest in humans

Abstract:The hippocampus is thought to promote gradual incorporation of novel information into long-term memory by binding, reactivating, and strengthening distributed cortical-cortical connections. Recent studies implicate a key role in this process for hippocampally driven crosstalk with the (ventro)medial prefrontal cortex (vmPFC), which is proposed to become a central node in such representational networks over time. The existence of a relevant prior associative network, or schema, may moreover facilitate this process. Thus, hippocampal-vmPFC crosstalk may support integration of new memories, particularly in the absence of a relevant prior schema. To address this issue, we used functional magnetic resonance imaging (fMRI) and prior schema manipulation to track hippocampal-vmPFC connectivity during encoding and postencoding rest. We manipulated prior schema knowledge by exposing 30 participants to the first part of a movie that was temporally scrambled for 15 participants. The next day, participants underwent fMRI while encoding the movie's final 15 min in original order and, subsequently, while resting. Schema knowledge and item recognition performance show that prior schema was successfully and selectively manipulated. Intersubject synchronization (ISS) and interregional partial correlation analyses furthermore show that stronger prior schema was associated with more vmPFC ISS and less hippocampal-vmPFC interregional connectivity during encoding. Notably, this connectivity pattern persisted during postencoding rest. These findings suggest that additional crosstalk between hippocampus and vmPFC is required to compensate for difficulty integrating novel information during encoding and provide tentative support for the notion that functionally relevant hippocampal-neocortical crosstalk persists during off-line periods after learning.

Abstract:Words, unlike images, are symbolic representations. The associative details inherent within a word's meaning and the visual imagery it generates, are inextricably connected to the way words are processed and represented. It is well recognised that the hippocampus associatively binds components of a memory to form a lasting representation, and here we show that the hippocampus is especially sensitive to abstract word processing. Using fMRI during recognition, we found that the increased abstractness of words produced increased hippocampal activation regardless of memory outcome. Interestingly, word recollection produced hippocampal activation regardless of word content, while the parahippocampal cortex was sensitive to concreteness of word representations, regardless of memory outcome. We reason that the hippocampus has assumed a critical role in the representation of uncontextualized abstract word meaning, as its information-binding ability allows the retrieval of the semantic and visual associates that, when bound together, generate the abstract concept represented by word symbols. These insights have implications for research on word representation, memory, and hippocampal function, perhaps shedding light on how the human brain has adapted to encode and represent abstract concepts.
