Abstract:Novel structurally intriguing heterocycles embedded with spiropyrrolidine, quinoxaline and chromanone units were synthesized in good yields using a [Bmim]Br accelerated multicomponent reaction strategy. The key step of the reaction is 1,3-dipolar cycloaddition involving highly functionalized dipolarophile, viz. 3-benzylidenechroman-4-one, to afford spiroquinoxalinopyrrolidine embedded chromanone hybrid heterocycles. The formation of spiro products occurs via two C–C, two N–C and one C–N bonds possessing four adjoining stereogenic centers, two of which are spiro carbons. The newly synthesized spiro compounds showed potent acetylcholinesterase and butyrylcholinesterase inhibitory activities. Moreover, compounds with fluorine displayed the highest AChE (3.20 ± 0.16 μM) and BChE (18.14 ± 0.06 μM) inhibitory activities. Further, docking studies, followed by all-atom molecular dynamics, showed results that are consistent with in vitro experimental findings. Although docking scores for the synthesized derivatives were higher than those of the standard drug, MD MMPBSA results showed better binding of synthesized derivatives (−93.5 ± 11.9 kcal mol −1 ) compared to the standard drug galantamine (−66.2 ± 12.3 kcal mol −1 ) for AChE but exhibited similar values (−98.1 ± 11.2 and −97.9 ± 11.5 kcal mol −1 ) for BChE. These differences observed in drug binding with AChE/BChE are consistent with RMSD, RMSF, LIG plots, and FEL structural analysis. Taken together, these derivatives could be potential candidates as inhibitors of AChE and BChE.

Abstract:Macrocyclic compounds play an important role in supramolecular chemistry accounting for their unique recognition and self-assembly properties and potential applications in biomedicine and material science. Azo compounds display promising capability in fields of molecular switches, polymers, smart materials and molecular machines because of their photoactive and electroactive properties. Introducing azo groups into macrocycles gives them signal moieties, additional recognition sites, photo-responsive properties, and so on. Herein, we comprehensively review the structures of azo-containing macrocyclic compounds reported up to now. Then we describe representative works on azo macrocycles with their molecular recognition, self-assembly and application emphasized.
One-Pot Synthesis of Diverse Anion-Binding Macrocycles

Abstract:A one-pot synthetic protocol to access a diverse library of diamide–diester macrocycles from the same starting materials is reported. The molecular symmetry can be readily tuned based on the reaction sequence, while the core structure can be varied using amino acids and aromatic building blocks. The first class of macrocycles with C 2 axes in their molecular plane were obtained in 24 h with 40–70% yield, while another class with C 2 axes perpendicular to the plane were synthesized in 18 h in 10–30% yield. The phenyl- and serine-derived macrocycles of the first class could bind acetate, chloride, and phosphate ions. These macrocycles can be functionalized with hydrophobic groups and potentially be used as ion transporters. Abstract:Macrocyclic compounds, characterized by cyclic structures, often originate from either modified forms of unicyclic canonical molecules or natural products. Within the field of medicinal chemistry, there has been a growing fascination with drug-like macrocycles in recent years, primarily due to compelling evidence indicating that macrocyclization can significantly influence both the biological and physiochemical properties, as well as the selectivity, when compared to their acyclic counterparts. The approval of contemporary pharmaceutical agents like Lorlatinib underscore the notable clinical relevance of drug-like macrocycles. Nonetheless, the synthesis of these drug-like macrocycles poses substantial challenges, primarily stemming from the complexity of ring-closing reactions, which are inherently dependent on the size and geometry of the bridging linker, impacting overall yields. Nevertheless, macrocycles offer a promising avenue for expanding the synthetic toolkit in medicinal chemistry, enabling the creation of bioactive compounds. To shed light on the subject, we delve into the clinical prowess of established macrocyclic drugs, spanning various therapeutic areas, including oncology, and infectious diseases. Case studies of clinically approved macrocyclic agents illustrate their profound impact on patient care and disease management. Abstract:Background:: Nowadays, macrocyclic compounds constitute a privileged source for the development of compounds with interesting biological properties. Ring-closing olefin me-tathesis has received great attention for the synthesis of small, medium, and larger ring systems. Methods:: In the present work, we described the synthesis of eight original pyridyl macrocyclic triarylmethanes using an efficient 3-step synthetic strategy. The bisalkylated 4,4'-(pyridin-X-ylmethylene) diphenols (X=2-4) were prepared by ring-closing metathesis as macrocyclization key step, using Grubbs second generation catalyst. Results:: The pyridyl macrocyclic triarylmethanes were obtained with moderate to good yields. The introduction of a pyridine N-oxide moiety before the macrocyclization proved to be interest-ing to afford a higher yield of the corresponding metathesis product. FT-IR, 1 H NMR, 13C NMR, and X-ray diffraction analysis have been used for the characterization of the synthesized compounds. Conclusion:: The synthetic strategy used here proposes an efficient alternative to prepare macro-cyclic triarylmethanes of different sizes.

Abstract:A novel class of cyclometalated macrocycles [(Cp*Ir)(2)(R-N=C-C(6)H(2)-C=N-R)(2)](2)(pyrazine)(2)·(OTf)(4) [R = Ph (4a), p-MeOC(6)H(4) (4b), p-MeC(6)H(4) (4c), p-ClC(6)H(4) (4d), Me (4e)]; [(Cp*Rh)(2)(R-N=C-C(6)H(2)-C=N-R)(2)](2)(pyrazine)(2)·(OTf)(4) [R = Ph (4a'), p-MeOC(6)H(4) (4b'), p-MeC(6)H(4) (4c')] and [(Cp*Ir)(2)(R-C=N-C(6)H(4)-N=C-R)(2)](2)(pyrazine)(2)·(OTf)(4) [R = Ph (5a), p-MeOC(6)H(4) (5b)] was stepwise constructed through the double-site C-H activation of aromatic bis-imine substrates. The structures of binuclear complexes and tetranuclear macrocycles were confirmed by single-crystal X-ray diffraction. Isomers were found both in binuclear species and macrocyclic complexes. Flexible substrates led to the existence of isomers for binuclear species, yet gave no isomers after macrocyclic constructions; rigid ones, in contrast, led to isomers only for macrocyclic species. The isomers of tetranuclear macrocycles were thermodynamically stable to reversible transformation on a scale of days. Robust bonding and a certain degree of rigidity were invoked to explain the existence of isomers. This is the first example, to our knowledge, in which coordinated macrocycles containing half-sandwich Cp*M (M = Ir, Rh) fragments have been constructed, without a dynamic reversible process.

Abstract:Small macrocycles with four or fewer amino acids are among the most potent natural products known, but there is currently no way to systematically generate such compounds. We describe a computational method for identifying ordered macrocycles composed of alpha, beta, gamma, and 17 other amino acid backbone chemistries, which we used to predict 14.9 million closed cycles composed of >42,000 monomer combinations. We chemically synthesized 18 macrocycles predicted to adopt single low-energy states and determined their x-ray or nuclear magnetic resonance structures; 15 of these were very close to the design models. We illustrate the therapeutic potential of these macrocycle designs by developing selective inhibitors of three protein targets of current interest. By opening up a vast space of readily synthesizable drug-like macrocycles, our results should considerably enhance structure-based drug design.

Abstract:This study introduces a novel solid-phase macrocyclization method generating 2-pyridone rings. This method relies on the intramolecular condensation between secondary and tertiary dimethoxy-propionic amide (DMPA) units. This selective reaction leads to the formation of a single, well-defined regioisomer. The method demonstrates remarkable efficiency in producing diverse peptidic and non-peptidic bioactive targets, paving the way for the development of innovative macrocycle libraries featuring the 2-pyridone unit.

Abstract:A series of tetraamino-bisthiourea chiral macrocycles containing two diarylthiourea and two chiral diamine units were synthesized by a fragment-coupling approach in high yields. Different chiral diamine units, including cyclohexanediamines and diphenylethanediamines were readily incorporated by both homo and hetero [1 + 1] macrocyclic condensation of bisamine and bisisothiocyanate fragments. With the easy synthesis, gram-scale of macrocycle products can be readily obtained. These chiral macrocycles were applied in catalyzing bioinspired decarboxylative Mannich reactions. Only 5 mol % of the optimal macrocycle catalyst efficiently catalyzed the decarboxylative addition of a broad scope of malonic acid half thioesters to isatin-derived ketimines with excellent yields and good enantioselectivity. The rigid macrocyclic framework and the cooperation between the thiourea and tertiary amine sites were found to be crucial for achieving efficient activation and stereocontrol. As shown in control experiments, catalysis with the acyclic analogues having the same structural motifs were non-selective.

Abstract:In a sustained search for novel potential drug candidates with multispectrum therapeutic application, a series of novel spirooxindoles was designed and synthesized via regioselective three-component reaction between isatin derivatives, 2-phenylglycine and diverse arylidene-imidazolidine-2,4-diones (Hydantoins). The suggested stereochemistry was ascertained by an X-ray diffraction study and NMR spectroscopy. The resulting tetracyclic heterocycles were screened for their in vitro and in vivo anti-inflammatory and analgesic activity and for their in vitro antimicrobial potency. In vitro antibacterial screening revealed that several derivatives exhibited remarkable growth inhibition against different targeted microorganisms. All tested compounds showed excellent activity against the Micrococccus luteus strain (93.75 μg/mL ≤ MIC ≤ 375 μg/mL) as compared to the reference drug tetracycline (MIC = 500 μg/mL). Compound 4e bearing a p-chlorophenyl group on the pyrrolidine ring exhibited the greatest antifungal potential toward Candida albicans and Candida krusei (MIC values of 23.43 μg/mL and 46.87 μg/mL, respectively) as compared to Amphotericin B (MIC = 31.25 and 62.50 μg/mL, respectively). The target compounds were also tested in vitro against the lipoxygenase-5 (LOX-5) enzyme. Compounds 4i and 4l showed significant inhibitory activity with IC50 = 1.09 mg/mL and IC50 = 1.01 mg/mL, respectively, more potent than the parent drug, diclofenac sodium (IC50 = 1.19 mg/mL). In addition, in vivo evaluation of anti-inflammatory and analgesic activity of these spirooxindoles were assessed through carrageenan-induced paw edema and acetic acid-induced writhing assays, respectively, revealing promising results. In silico molecular docking and predictive ADMET studies for the more active spirocompounds were also carried out.

Abstract:A biological impact of the fused heterocyclic rings as anti-enzymatic agents was explored through experimental, theoretical and computational studies. The targeted hybrids of 1,2,4-triazole ( 8a–q ) were synthesized through multistep protocol. Initially, the carboxylate 3 was synthesized through stirring of 4-chlorobenzenesulfonyl chloride ( 1 ) and 4-(ethoxycarbonyl)piperidine ( 2 ) at room temperature. Carboxylate was converted into carbohydrazide 4 , which was refluxed with phenyl isothiocyanate and KOH to synthesize 1,2,4-triazole ( 6 ) through the formation of an intermediate hydrazinecarbothioamide (5) . A series of alkyl/aralkyl halides ( 7a–q ) were stirred at room temperature with compound 6 in the presence of Li(I) catalyst to avail the targeted library of hybrids ( 8a–q ). All the designed hybrids were screened for their acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), urease, hemolysis and biofilm inhibition potential and were found active with variable potential against all the evaluations. The best compound for AChE inhibition was 8c having 89.24 ± 0.78 % inhibition at 0.5 mM with IC 50 16.43 ± 0.62 μM, while the best against BChE was 8 g with 87.25 ± 0.47 % inhibition and IC 50 31.42 ± 0.34 μM. The whole series of compounds remained potent inhibitors against urease enzyme with better potential as compared to standard thiourea. The potent compounds of the series were 8o and 8i with IC 50 values of 1.32 ± 1.03 μM and 1.38 ± 0.34 μM, respectively. Half of the whole library of compounds ( 8a–q ) possessed outstanding biofilm inhibition potential against the tested bacterial strains and confirmed themselves as better anti-microbial agents. The compounds were also screened for hemolytic activity and results exhibited medium to excellent hemolytic activity. The compound 8b showed the least toxicity among the synthesized compounds, 8a–q . The computational studies of all the synthesized compounds were also carried out to elaborate the binding forces of synthesized compounds with the active sites of enzymes to establish SAR. In silico data supported the in vitro profiles. Further work is suggested for in vivo studies of the most of compounds in search for their anti-cholinesterase and anti-urease potential. Work is in progress on these lines.

Abstract:A diminutive chemical library of acyl thiotriazinoindole (ATTI) based bioactive scaffolds was synthesized, instigated by taking the economical starting material Isatin, through a series of five steps. Isatin was first nitrated followed by the attachment of pentyl moiety via nucleophilic substitution reaction. The obtained compound was reacted with thiosemicarbazide to obtain thiosemicarbazone derivative, which was eventually cyclized using basic conditions in water as solvent. Finally, the reported series was obtained through reaction of nitrated thiotriazinoindole moiety with differently substituted phenacyl bromides. The synthesized compounds were characterized using NMR spectroscopy and elemental analysis. Finally, the synthesized motifs were scrutinized for their potential to impede urease, α -glucosidase, DPPH, and α -amylase. Compound 5 h with para cyano group manifested the most pivotal biological activity among all, displaying IC 50 values of 29.7 ± 0.8, 20.5 ± 0.5 and 36.8 ± 3.9 μM against urease, α -glucosidase, and DPPH assay, respectively. Simultaneously, for α- amylase compound 5 g possessing a p -CH 3 at phenyl ring unfolded as most active, with calculated IC 50 values 90.3 ± 1.1 μM. The scaffolds were additionally gauged for their antifungal and antibacterial activity. Among the tested strains, 5d having bromo as substituent exhibited the most potent antibacterial activity, while it also demonstrated the highest potency against Aspergillus fumigatus . Other derivatives 5b , 5e, 5i, and 5j also exhibited dual inhibition against both antibacterial and antifungal strains. The interaction pattern of derivatives clearly displayed their SAR, and their docking scores were correlated with their IC 50 values. In molecular docking studies, the importance of interactions like hydrogen bonding was further asserted. The electronic factors of various substituents engendered variety of interactions between the ligands and targets implying their importance in the structures of the synthesized heterocyclic scaffolds. To conclude, the synthesized compounds had satisfactory biological activity against various important targets. Further studies are therefore encouraged by attachment of different substitutions in the structure at various positions to enhance the activity of these compounds.

Abstract:A series of hydroxy‐quinoline-based sulfonohydrazide derivatives ( 1–16 ) were synthesized and their structures were elucidated by using various spectroscopic tools including 1 HNMR, 13 CNMR and HREI-MS and evaluated against acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) enzymes. All synthesized derivatives showed the varied range of AChE and BuChE activities having IC 50 values ranging from 0.20 ± 0.01 to 11.60 ± 0.20 μM (against AChE) and 0.80 ± 0.05 to 22.70 ± 0.30 μM (against BuChE) as compared to standard drug Donepezil (IC 50 = 2.16 ± 0.12 μM & 4.5 ± 0.11 μM, respectively). Among the series, compounds 1, 8, and 10 were identified to be most potent, even more, active than standard drug having IC 50 values of 0.40 ± 0.05, 0.20 ± 0.01, 0.70 ± 0.05 μM (against AChE) and 0.80 ± 0.05, 0.80 ± 0.05, 2.10 ± 0.10 μM (against BuChE) respectively. Based on the substitution pattern around the aryl ring, structure-activity relationship (SAR) analysis was conducted for all synthesized derivatives. Additionally, the molecular docking method was created to investigate the mechanism of interactions between the scaffolds that are most active and the active sites of specific AChE and BuChE enzymes.

Abstract:Two o-carborane-based π-conjugated macrocycles are synthesized via oxidative coupling of acetylene-terminated o-carborane precursors. The single-crystal X-ray diffraction analysis unambiguously revealed their unique porous supramolecular structures, which display micropore characteristics. The novel macrocycles possess aggregation-induced emission both in aggregate state and solid state. This work provides a new design strategy for developing o-carborane macrocyclic molecules with excellent photophysical properties. Based on the obvious fluorescence quenching phenomenon of macrocycle by 1,4-dinitrobenzene and 2,4,6-trinitrophenol, the macrocyclic compounds have promising potential as sensors for nitroaromatics.

Abstract:The present study aims to synthesize new thiadiazole/oxadiazole fused thiadiazole derivatives, incorporating pyridine moiety as effective inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) to treat Alzheimer disease. Initially, pyridine based thiosemicarbazone was synthesized by treating pyridine 2,6-dicarboxaldehyde with thiosemicarbazide. Thiosemicarbazone was cyclized into bis-thiadiazole, which further react with different isothiocyanates to give pyridine-based bis-(thiadiazole-fused-thiadiazole) derivatives (4a-h) . Next pyridine based semicarbazone was synthesized by treating pyridine 2,6-dicarboxaldehyde with semicarbazide. Then semicarbazone was cyclized into bis-oxadiazole which further react with different isothiocyanates to give pyridine-based bis-(oxadiazole-fused-thiadiazole) derivatives (7a-h) .The synthetic analogs were structurally elucidated through 1 H NMR, 13 C NMR and HRMS (EI) analysis and assessed for acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitory potential. All synthetic analogues showed varying degrees of inhibitory effects on AChE and BuChE, with IC 50 value ranging from 12.89 ± 1.10 μM - 61.76 ± 2.40 μM for AChE and 14.79 ± 1.20 μM - 56.87 ± 2.50 μM for BuChE in comparison of standard inhibitor Galantamine (IC 50 = 37.40 ± 1.20 μM for AChE and IC 50 = 41.20 ± 1.90 μM for BuChE). Among the series, analogues 4c IC 50 = 13.32 ± 1.30 μM (against AChE), and 15.17 ± 1.40 μM (against BuChE), and 7c IC 50 = 12.89 ± 1.10 μM (against AChE) and 14.79 ± 1.20 μM (against BuChE) exhibited notable inhibitory activity against AChE and BuChE enzymes, Moreover, molecular docking studies were conducted on potential analogues to investigate their binding interactions with the targeted enzyme's active sites.

Abstract:A multifunctional molecular design of macrocycles (E 1 -E 4 ) based on pillar [5] arene-appended cinnamate ester linking group was synthesized and well characterized. All four pillar [5] arene-based macrocycles exhibit a columnar hexagonal phase over a broad temperature range and higher thermal stability. The optical and thermal properties of these macrocycles have been extensively investigated through polarized optical microscopy, differential scanning calorimetry, and thermogravimetric analysis. The arrangement of molecules in the liquid crystalline state has been probed using X-ray diffraction techniques. All four macrocycles demonstrate enantiontropic liquid crystalline properties. The tubular-shaped molecular system of pillar[5]arene-derived derivatives display columnar self-assembly, which is further stabilized upto below 50 °C. An intriguing aspect is the blue fluorescence displayed by all four materials in solution and in the solid thin film state. The potential utility of macrocycle E 4 as a staining agent for the human colorectal cancer cell line HCT116 has been demonstrated, thus expanding the scope of applications for these supramolecular macrocycles.

Abstract:A series of novel macroacyclic Schiff base ligands and their Cu (II) complexes were synthesised via reacting dicarbonyls of varying chain lengths with S-methyl dithiocarbazate (SMDTC) and S-benzyl dithiocarbazate (SBDTC) followed by coordination with Cu (II) ions. X-ray crystal structures were obtained for compound 4, an SBDTC-diacetyl analogue, and Cu7, an SMDTC-hexanedione Cu (II) complex. Anticancer evaluation of the compounds showed that Cu1, an SMDTC-glyoxal complex, demonstrated the highest cytotoxic activity against MCF-7 and MDA-MB-231 breast cancer cells with IC50 values of 1.7 μM and 1.4 μM, respectively. There was no clear pattern observed between the effect of chain length and cytotoxic activity; however, SMDTC-derived analogues were more active than SBDTC-derived analogues against MDA-MB-231 cells. The antibacterial assay showed that K. rhizophila was the most susceptible bacteria to the compounds, followed by S. aureus. Compound 4 and the SMDTC-derived analogues 3, 5, Cu7 and Cu9 possessed the highest antibacterial activity. These active analogues were further assessed, whereby 3 possessed the highest antibacterial activity with an MIC of <24.4 μg/mL against K. rhizophila and S. aureus. Further antibacterial studies showed that at least compounds 4 and 5 were bactericidal. Thus, Cu1 and 3 were the most promising anticancer and antibacterial agents, respectively.

Abstract:Macrocycles are essential in protein‐protein interactions and the preferential intake of bioactive scaffolds. Macrocycles are commonly synthesized by late‐stage macrolactonizations, macrolactamizations, transition metal‐catalyzed ring‐closing metathesis, S‐S bond‐forming reactions, and copper‐catalyzed alkyne–azide cycloaddition. Recently, transition metal‐catalyzed C‐H activation strategies have gained significant interest among chemists to synthesize macrocycles. This article provides a comprehensive overview of the transition metal‐catalyzed macrocyclization via C‐H bond functionalization of heterocycle‐containing peptides, annulations, and heterocycle‐ring construction through direct C‐H bond functionalization. In the first part, palladium salt catalyzed coupling with indolyl C(sp3)‐H and C(sp2)‐H bonds for macrocyclization is reported. The second part summarizes rhodium‐catalyzed macrocyclizations via site‐selective C‐H bond functionalization. Earth‐abundant, less toxic 3d metal salt Mn‐catalyzed cyclizations are reported in the latter part. This summary is expected to spark interest in emerging methods of macrocycle production among organic synthesis and chemical biology practitioners, helping to develop the discipline. We hope that this mini‐review will also inspire synthetic chemists to explore new and broadly applicable C‐C bond‐forming strategies for macrocyclization via intramolecular C‐H activation.

Abstract:β-Elemene is the major constituent of the antitumor drugs elemene extract approved in China. By incorporating macrocyclization strategy into the β-elemene skeleton, we designed a series of novel macrocycles retaining three key carbon–carbon double bonds. Four different methods have been successfully developed for these challenging ring systems. A total of twenty-eight 14- to 24-membered macrocycles were synthesized. Most of these macrocycles exhibited good antitumor activity against several cancer cell lines (PC-3, A549, U87MG, U251 and HCT116), with up to 40 folds improvement of activity comparing to β-elemene. Additionally, X-ray single crystal structures of compounds Ic, Ip, and IIh were successfully solved as the proof of macrocycle formation. The results warrant the further investigation of this novel class macrocycles in pharmacokinetic and pharmacodynamics studies, which will be reported in due course.
