Abstract:Hypoxia/reoxygenation causes cellular injury and death associated with a number of pathophysiological conditions, including myocardial ischemia/reperfusion injury and stroke. The cell death pathways induced by hypoxia/reoxygenation and their underlying regulatory mechanisms remain poorly understood. Recent studies have shown that hypoxia/reoxygenation can induce Bax translocation and cytochrome c release. Using murine lung endothelial cells as a model, we found that the induction of apoptosis by hypoxia/reoxygenation involved the activation of both Bax-dependent and death receptor-mediated pathways. We demonstrated the activation of the death-inducing signal complex and Bid pathway after hypoxia/reoxygenation. Hepatocyte growth factor markedly inhibited hypoxia/reoxygenation-induced endothelial cell apoptosis. The cytoprotection afforded by hepatocyte growth factor was mediated in part by the stimulation of FLICE-like inhibiting protein expression, the attenuation of death-inducing signal complex formation, and the inhibition of Bid and Bax activation. Hepatocyte growth factor also prevented cell injury and death by increasing the expression of the antiapoptotic Bcl-XL protein. The inhibition of Bid/Bax-induced cell death by hepatocyte growth factor primarily involved p38 MAPK and in part Akt-dependent pathways but not ERK1/ERK2.
Synthesis of novel antibacterial and biocompatible polymer nanocomposite based on polysaccharide gum hydrogels

Abstract:According to recent studies on the benefits of natural polymer-based hydrogels in biomedical applications, gellan gum (GG)/acacia gum (AG) hydrogel was prepared in this study. In order to regulate the mechanical behavior of the hydrogel, graphite carbon nitride (g-C 3 N 4 ) was included in the hydrogel matrix. In addition, metal oxide nanoparticles ZnCuFe 2 O 4 were added to the composite for antibacterial activity. The prepared GG–AG hydrogel/g-C 3 N 4 /ZnCuFe 2 O 4 nanobiocomposite was characterized by using FE-SEM, FTIR, EDX, XRD and TGA. The nanobiocomposite exhibited spherical morphology, which was related to the incorporation of the metal oxide nanoparticles. GG–AG hydrogel/g-C 3 N 4 /ZnCuFe 2 O 4 nanobiocomposite showed 95.11%, 92.73% and 88.97% biocompatibility toward HEK293T cell lines within 24 h, 48 h and 72 h incubation, respectively, which indicates that this nanobiocomposite is completely biocompatible with healthy cells. Also, the nanobiocomposite was able to inhibit Pseudomonas aeruginosa biofilm growth on its surface up to 87%. Rheological studies showed that the nanobiocomposite has a viscoelastic structure and has a water uptake ratio of 93.2%. In comparison with other similar studies, this nanobiocomposite has exhibited superior antibacterial activity complete biocompatibility and proper mechanical properties, high swelling and water absorption capability. These results indicate that GG–AG hydrogel/g-C 3 N 4 /ZnCuFe 2 O 4 nanocomposite can be considered as a potential candidate for biomedical applications such as tissue engineering and wound healing.

Designing network structure hydrogels derived from carrageenan- phosphated polymers by covalent and supramolecular interactions for potential biomedical applications

Abstract:Recently, various efforts have been made to explore the potential of natural polysaccharides derived from sea weeds to promote sustainable development. Herein, carrageenan (CG), a polysaccharide extracted from red sea algae, was utilized to design network structures as hydrogels, aimed at significant applications in drug delivery (DD) systems. Hydrogels were designed by graft copolymerization reaction of poly(bis [2-methacryloyloxy] ethyl phosphate [poly(BMEP)] and poly(acrylic acid) [poly(AAc)] onto CG in the presence of a crosslinking agent. Hydrogels were developed by covalent linkage by graft copolymerization and supramolecular interactions, existing in the copolymers. Copolymers were characterized by Atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), 13C-nuclear magnetic resonance (NMR), and X-ray diffraction (XRD) instrumentations. The drug diffusion exhibited a sustained pattern due to polymer-drug interactions. The drug release followed non-Fickian diffusion mechanism and the release profile was most accurately depicted by first order kinetic model. The biocompatible nature of the copolymer was demonstrated from the hemolytic index value signifying minimal adverse interactions with blood component upon exposure. A protein adsorption test was performed using bovine serum albumin (BSA), exhibiting 8.15 ± 0.26 % albumin adsorption. Polymers exhibited mucoadhesive character, evidenced by their requirement of a detachment force measuring 195 ± 4.72 mN for separation from the membrane during interactions with the mucosal surface. The hydrogels exhibited antioxidant properties, evidenced by 2, 2'-Diphenylpicrylhydrazyl (DPPH) assay, revealing copolymer capable of scavenging 58.21 ± 2.26 % of free radical. The hydrogel revealed antimicrobial activity against P. aeruginosa and S. aureus bacteria, a property further enhanced in hydrogels with the drug doxycycline. These findings suggest suitability of these hydrogels for biomedical applications, with a significant emphasis on drug delivery.

Abstract:This study describes the fabrication and characterization of nanocomposite hydrogel beads based on Cellulose and synthesized silver nanoparticles (Ag-NPs) prepared by insitu and exsitu methods. The silver nanoparticles were synthesized by reducing silver nitrate (AgNO 3 ) by Sodium borohydrate (NaBH 4 ). Cellulose/Ag-NPs hydrogel beads were prepared using calcium chloride as the cross-linker. The hydrogel beads were characterized using FTIR and XRD. Moreover, swelling property of the cellulose /Ag-NPs hydrogel beads was investigated. The Ag release profile of the hydrogels was obtained for the amount of Ag released using Atomic Absorption spectroscopy (AAS). The cumulative release of Ag-NPs was estimated by measuring the absorbance at 405 nm of samples obtained at various time intervals. The exsitu and insitu nanocomposite hydrogels showed greater swelling behavior in comparison with virgin cellulose hydrogel. Both insitu and exsitu cellulose/Ag-NPs presented good antibacterial activities against Escherichia coli and Bacillus and with maximum zones of inhibition 12 ± 2 mm more than the pristine cellulose hydrogel thereby, have great pharmacological potential and a suitable level of safety for use in the biological systems.

Abstract:Polyacrylamide and dextran-graft-polyacrylamide hydrogels are prepared and used as nanoreactors and networks for the synthesis of silver nanoparticles (AgNPs). Photochemical generation of AgNPs is carried out under UV-irradiation of Ag+ ions in swollen hydrogels of different cross-linking densities. The obtained hydrogels and hydrogel/AgNPs composites are characterized by TEM, FTIR, and UV–Vis spectroscopy. Swelling studies have shown a relationship between the structure of the hydrogels and their ability to swell. It is shown that the presence of AgNPs in the polymer network leads to a decrease of the swelling capacity. An increase in the cross-linking density leads to an expansion of the AgNPs size distribution for both types of hydrogels. All synthesized hydrogel-silver nanoparticle composites have shown a high activity in the growth retardation of Staphylococcus aureus microorganisms.

Abstract:Versatility of the anionic algal polysaccharide carrageenan has long been discussed and explored, especially for their affinity towards water molecules. While this feature is advantageous in certain applications such as water remediation, wound healing, etc., the usefulness of this biopolymer is extremely limited when it comes to applications such as food packaging. Scientists around the globe are carrying out research works on venturing diverse methods to integrate a hydrophobic nature into these polysaccharides without compromising their other functionalities. Considering these foregoing studies, this review was designed to have an in-depth understanding of diverse methods and techniques adopted for tuning the hydrophobic nature of carrageenan-based bionanocomposites, both via surface alterations or by changes made to their chemical structure and attached functional groups. This review article mainly focused on how the hydrophobicity of carrageenan bionanocomposites varied as a function of the type and refinement of carrageenan, and with the incorporation of additives including plasticisers, nanofillers, bioactive agents, etc. Incorporation of nanofillers such as polysaccharide-based nanoparticles, nanoclays, bioceramic and mineral based nanoparticles, carbon dots and nanotubes, metal oxide nanoparticles, etc., along with their synergistic effects in hybrid bionanocomposites are also dealt with in this comprehensive review article.

Abstract:Algae biomass has been recognized as feedstock with diverse application including production of biofuel, biofertilizer, animal feed, wastewater treatment and bioremediation. In addition, algae species are a potential reservoir of metabolites and polymers with potential to be utilized for biomedicine, healthcare and industrial purposes. Carrageenan is one such medicinally and industrially significant polysaccharide which is extracted from red algae species ( Kappaphycus alvarezii and Eucheuma denticulatum , among the common species). The extraction process of carrageenan is affected by different environmental factors and the source of biomass, which can vary and significantly impact the yield. Diverse applications of carrageenan include hydrogel beads, bio-composites, pharmacological properties, application in cosmetics, food and related industries. Carrageenan biological activities including antioxidant, anti-inflammatory, antimicrobial, and antitumor activities are significantly influenced by sulfation pattern, yield percentage and molecular weight. In addition to natural biomedical potential of carrageenan, synergetic effect of carrageenan- nanocomposites exhibit potential for further improvisation of biomedical applications. Nanotechnology driven bio-composites of carrageenan remarkably improve the quality of films, food packaging, and drug delivery systems. Such nano bio-composites exhibit enhanced stability, biodegradability, and biocompatibility, making them suitable alternatives for drug delivery, wound-healing, and tissue engineering applications. The present work is a comprehensive study to analyze biomedical and other applications of Carrageenan along with underlying mechanism or mode of action along with synergetic application of nanotechnology.

Green Synthesis of Flower-Like Carrageenan-Silver Nanoparticles and Elucidation of Its Physicochemical and Antibacterial Properties

Abstract:Herein, we report the green synthesis of flower-like carrageenan-silver nanoparticles (c-AgNPs) through a facile hydrothermal reaction at 90 °C for 2 h. The reduction of silver nitrate (AgNO3) to c-AgNPs was evident by the colour change of the solution from colourless to dark brown and further confirmed by a UV-Vis surface plasmon resonance (SPR) peak at ~420 nm. The FTIR spectra showed that the abundance of functional groups present in the carrageenan were responsible for the reduction and stabilisation of the c-AgNPs. The XRD pattern confirmed the crystalline nature and face-centred cubic structure of the c-AgNPs, while the EDX analysis showed the presence of a high composition of elemental silver (85.87 wt%). Interestingly, the morphological characterisations by SEM and FE-SEM revealed the formation of flower-like c-AgNPs composed of intercrossed and random lamellar petals of approximately 50 nm in thickness. The growth mechanism of flower-like c-AgNPs were elucidated based on the TEM and AFM analyses. The c-AgNPs displayed promising antibacterial properties against E. coli and S. aureus, with zones of inhibition ranging from 8.0 ± 0.0 to 11.7 ± 0.6 mm and 7.3 ± 0.6 to 9.7 ± 0.6 mm, respectively, as the concentration of c-AgNPs increased from 0.1 to 4 mg/mL.

Abstract:In recent years, the development of multifunctional hydrogels has gained significant attention due to their potential in various biomedical applications, including antimicrobial, antioxidant, and anticancer therapies. By integrating biocompatible polymers and nanoparticles, these hydrogels can achieve enhanced activity and targeted therapeutic effects. In this study, carrageenan/2-dimethyl aminoethyl methacrylate/gelatin (CAR/DEMA/Gelt) composite hydrogel was synthesized using microwave radiation specifically for its efficiency in enhancing cross-linking and promoting uniform nanoparticle dispersion within the matrix. Zinc oxide (ZnO) nanoparticles were incorporated into the hydrogel to form the (CAR/DEMA/Gelt/ZnO) nanocomposite. The hydrogels were characterized using FT-IR, FE-SEM, XRD, TGA, and EDX, confirming successful cross-linking and structural integrity. The nanocomposite hydrogel exhibited more enhanced antimicrobial activity than the composite hydrogel against Gram-positive Staphylococcus aureus (S. aureus) and Bacillus subtilis (B. subtilis), with inhibition zones of 15 mm and 16 mm, respectively, while in case of the Gram-negative bacteria, Klebsiella pneumoniae (K. pneumoniae) and Escherichia coli (E. coli), the inhibition zones were 29 mm and 19 mm, respectively. In addition to the unicellular fungi, Candida albicans (C. albicans), the inhibition zone was 19 mm. Moreover, the nanocomposite showed anti-inflammatory activity comparable to those of Indomethacin and antioxidant activity, with an impressive IC50 value of 33.3 ± 0.05 μg/mL. In vitro cytotoxicity assays revealed significant anticancer activity. Against the MCF-7 breast cancer cell line, the CAR/DEMA/Gelt/ZnO nanocomposite showed 72.5 ± 0.02% cell viability, which decreased to 30.8 ± 0.01% after loading doxorubicin (DOX). Similarly, against the HepG2 liver cancer cell line, the free nanocomposite displayed 59.9 ± 0.006% cell viability, which depleted to 29.9 ± 0.005% when DOX was uploaded. This CAR/DEMA/Gelt/ZnO nanocomposite hydrogel demonstrates strong potential as a multifunctional platform for targeted biomedical applications, particularly in cancer therapy.

Abstract:The synthesis and specific surface functionalization of antimicrobial silver nanoparticles (AgNPs) and their incorporation into an alginate hydrogel is described. Divalent cation-mediated ionic crosslinking was used to disperse the AgNPs throughout the gel, made possible by -COO- cross-linking sites provided by the surface-enhanced nanoparticles, inspired by the classic egg-box model crosslinking of calcium alginate. An AgNP concentration, 10-20 μg g-1 increased hygrogel elasticity, viscosity, and shear resistance by 45, 30, and 31% respectively. Cryo-TEM revealed evenly distributed AgNP assemblies of discrete AgNPs throughout the gel matrices. FTIR-ATR indicated AgNPs were involved in alginate carboxylate-Ca2+-COO-AgNP crossbridging, which was not achieved through mixing of AgNPs into preformed gels. Live/dead fluorometric assays determined a minimal bactericidal concentration of 25 μg g-1 Ag for 6 microorganisms. Anti-biofilm assays showed species-dependent cell death of 44 -61%, with limited silver ion release of 0.41% and 1.1% after 7 days for Gram positive and negative bacteria, respectively.
Sustained-release, antibacterial, adhesive gelatin composite hydrogel with AgNPs double-capped with curdlan derivatives

Abstract:In this work, carboxymethylated curdlan (CMCD) was utilized as a capping and stabilizing agent for the green synthesis of silver nanoparticles. Subsequently, quaternized curdlan (QCD) was introduced as the second capping layer through electrostatic attraction, leading to the preparation of double-capped silver nanoparticles (AgNPs@CQ). The successful synthesis of silver nanoparticles was characterized using UV-vis, FTIR, XRD, TEM, and DLS. AgNPs@CQ were incorporated into gelatin and a AgNPs@CQ/Gel composite hydrogel was obtained. The incorporation of AgNPs@CQ imparts excellent antibacterial properties to the composite hydrogel, thereby enhancing its antimicrobial efficacy. The presence of double-capping layers significantly retards the release rate of silver, contributing to prolonged antimicrobial activity. The MTT and live/dead fluorescence staining results demonstrate that the gelatin hydrogel incorporating double-capped AgNPs exhibits enhanced cell viability compared to the one incorporating single-capped AgNPs. Additionally, the composite hydrogel exhibits remarkable mechanical strength and adhesive performance. The AgNPs@CQ/Gel composite hydrogel demonstrates a cost-effective and facile preparation, showing significant potential in the field of dressings.
Green Synthesis of Carbohydrate Polymer Based Gum Kondagogu/Hydroxypropyl Cellulose Blend Silver Nanocomposite Film and Their Antimicrobial Activity

Abstract:In the last few years, nanobiotechnology has undergone notable expansion, presenting a many applications driven by the practical viability of diverse synthesis techniques. The synthesis of silver nanocomposite through a green and eco-friendly approach is facilitated by polymer assistance, ensuring an efficient, cost-effective, and rapid process. This study specifically centers on creating eco-friendly and biodegradable nanocomposite films using hydroxypropyl (HP) cellulose and gum kondagogu (GK) through a simple casting process. In this method, silver nanocomposite (AgNPs) are synthesized by reducing AgNO 3 , with the biopolymer gum kondagogu serving dual roles as both a reducing and stabilizing agent. The resulting composite and Ag nanocomposite films underwent thorough characterization using techniques such as X-ray diffraction (XRD), fourier-transform infrared spectroscopy (FTIR), atomic force microscopy (AFM), field emission scanning electron microscopy, energy-dispersive X-ray spectroscopy (FESEM & EDX), transmission electron microscopy, and X-ray photoelectron spectroscopy. XRD and FTIR results revealed robust interactions between AgNPs and the polysaccharide chains. TEM and SEM images demonstrated the uniformity spherical, and smooth surface of HPGKP-Ag nanocomposite films, with EDAX confirming the presence of elemental Ag in the film matrix. AFM images disclosed AgNPs dispersed on the surface of composite films. XPS data clarified that silver existed solely in metallic form, with migration occurring during film formation. Furthermore, antibacterial assessments against strains such as Pseudomonas , and Streptococcus exhibited notable growth inhibition for both strains, highlighting the significant antibacterial potential of these nanocomposite films.

Abstract:Bacterial wound infections remain a significant health issue of great concern. Hence, there is a need to develop a novel material with antibacterial properties and smart functions. In this study, the effects of silver nanoparticles content (AgNPs) on properties of photothermal and pH-responsive nanocomposite hydrogels were investigated. The nanocomposite hydrogel samples were prepared using cassava starch waste modified by carboxymethylation (CMS), and mixed with poly vinly alcohol (PVA) and tannic acid (TA). The presence of AgNPs in the hydrogel samples enhanced antibacterial activities and photothermal conversion ability. The use of as-prepared hydrogel using 200 mM silver nitrate (H-AgNPs-200) combined with near infrared (NIR) radiation produced 100 % antibacterial efficiency for Escherichia coli (E.coli) and 98.2 % for Staphylococcus aureus (S.aureus). Furthermore, the H-AgNPs-200 also provided the highest storage modulus at 8.78 kPa. The obtained nanocomposite hydrogel was shown to exhibit pH-responsive release of TA. Under NIR radiation, higher release of TA at different pH was observed. The cytotoxicity study indicated that the nanocomposite hydrogels had good biocompatibility. Hence, the development of nanocomposite hydrogel-based CMS from cassava starch waste/PVA/AgNPs is a promising and sustainable approach where agro-waste product is used as the base material for medical application in wound dressing.
Hybrid gum tragacanth/sodium alginate hydrogel reinforced with silver nanotriangles for bacterial biofilm inhibition

Abstract:Biomaterial associated bacterial infections are indomitable to treatment due to the rise in antibiotic resistant strains, thereby triggering the need for new antibacterial agents. Herein, composite bactericidal hydrogels were formulated by incorporating silver nanotriangles (AgNTs) inside a hybrid polymer network of Gum Tragacanth/Sodium Alginate (GT/SA) hydrogels. Physico-chemical examination revealed robust mechanical strength, appreciable porosity and desirable in vitro enzymatic biodegradation of composite hydrogels. The antibacterial activity of AgNT-hydrogel was tested against planktonic and biofilm-forming Gram-negative ( Escherichia coli and Pseudomonas aeruginosa ) and Gram-positive ( Staphylococcus aureus ) bacteria. For all the strains, AgNT-hydrogel showed a dose-dependent decrease in bacterial growth. The addition of AgNT-hydrogels (40-80 mg ml −1 ) caused 87% inhibition of planktonic biomass and up to 74% reduction in biofilm formation. Overall, this study proposes a promising approach for designing antibacterial composite hydrogels to mitigate various forms of bacterial infection. Graphical

Abstract:This study aimed to explain the production and the characterization of gelatin/poly(2-hydroxyethylmethacrylate-co-2-Acrylamido-2-methyl-1-propanesulfonic acid), Gel/p(HEMA-co-AMPS), hydrogels including silver nanoparticles (AgNP) that were developed with a simple, cost-effective and environmentally-friendly process. Silver ions were added to the Gel/p(HEMA-co-AMPS) hydrogel matrix synthesized via free radical polymerization, and silver nanoparticles were developed by in-situ reduction of silver ions (Ag+) in the swollen hydrogel using Verbascum Longipedicellatum plant extract. The structure and morphology of the prepared nanocomposite hydrogel were confirmed by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), transmission electron microscopy-energy-dispersive X-ray spectroscopy (TEM-EDX), X-ray diffraction analysis (XRD) and thermogravimetric analysis (TGA). In addition, the swelling properties of hydrogels were investigated and AgNPs embedded in the hydrogel network were found to absorb more water, improving the swelling properties of hydrogels. Finally, the antibacterial activity of silver nanocomposites against S. aureus, E. faecalis, B. Subtilis, B. cereus, E. coli, P. aeruginosa as bacterial agents and C. albicans as fungal agent was examined. Therefore, the present study offers a multi-component hydrogel containing AgNPs with low-cost and environmentally-friendly fabrication process for application as bactericidal materials in biotechnology and biomedical fields.

Abstract:Modification of cellulose with silver nanoparticles produces various nanocomposites with significantly developed properties. This work aims to prepare a PVA hydrogel modified with cellulose/silver nanocomposites having potential applications in various fields including biomedical, antimicrobial inhibition, textile wears, etc. Microfibrillated cellulose/silver nanocomposites hydrogels were prepared in the aqueous medium with aid of microwave-assisted heating. Different percentages of nanocomposites were incorporated in PVA hydrogel to enhance the properties of PVA hydrogel. Prepared products were characterized by UV–Visible spectroscopy, FTIR, TGA, XRD, and SEM. The swelling (in water saline, acidic and alkaline solution), tensile, thermal, and antibacterial properties were also examined. The formation of Ag nanoparticles (AgNPs) in the (MFC-Ag)NC was confirmed by XRD and UV–Vis spectra. UV–Vis spectra showed the characteristic peaks of Ag in the UV–Vis spectra at 425 nm. Final products exhibited significant porosity and maximum swelling of 519.44%. The thermal stability of hydrogel increased with an increased percentage of (MFC-Ag)NC. Hydrogels exhibited significant antimicrobial inhibition against multidrug-resistant microorganisms, including Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.Graphical Abstract

Abstract:The recognized antibacterial properties of silver nanoparticles (AgNPs) characterize them as attractive nanomaterials for developing new bioactive materials less prone to the development of antibiotic resistance. In this work, we developed new composites based on self-assembling Fmoc-Phe3 peptide hydrogels impregnated with in situ prepared AgNPs. Different methodologies, from traditional to innovative and eco-sustainable, were compared. The obtained composites were characterized from a hydrodynamic, structural, and morphological point of view, using different techniques such as DLS, SEM, and rheological measurements to evaluate how the choice of the reducing agent determines the characteristics of AgNPs and how their presence within the hydrogel affects their structure and properties. Moreover, the antibacterial properties of these composites were tested against S. aureus, a major human pathogen responsible for a wide range of clinical infections. Results demonstrated that the hydrogel composites containing AgNPs (hgel@AgNPs) could represent promising biomaterials for treating S. aureus-related infections.

Abstract:ABSTRACT We previously optimized imipenem and tobramycin combination regimens against a double-resistant clinical Pseudomonas aeruginosa isolate by using in vitro infection models, mechanism-based pharmacokinetic/pharmacodynamic modeling (MBM), and Monte Carlo simulations. The current study aimed to evaluate these regimens in a neutropenic murine thigh infection model and to characterize the time course of bacterial killing and regrowth via MBM. We studied monotherapies and combinations of imipenem with tobramycin in vivo against the double-resistant clinical P. aeruginosa isolate by using humanized dosing schemes. Viable count profiles of total and resistant populations were quantified over 24 h. Tobramycin monotherapy (7 mg/kg every 24 h [q24h] as a 0.5-h infusion) was ineffective. Imipenem monotherapies (continuous infusion of 4 or 5 g/day with a 1-g loading dose) yielded 2.47 or 2.57 log10 CFU/thigh killing at 6 h. At 24 h, imipenem at 4 g/day led to regrowth up to the initial inoculum (4.79 ± 0.26 log10 CFU/thigh), whereas imipenem at 5 g/day displayed 1.75 log10 killing versus the initial inoculum. The combinations (i.e., imipenem at 4 or 5 g/day plus tobramycin) provided a clear benefit, with bacterial killing of ≥2.51 or ≥1.50 log10 CFU/thigh compared to the respective most active monotherapy at 24 h. No colonies were detected on 3×MIC agar plates for combinations, whereas increased resistance (at 3×MIC) emerged for monotherapies (except imipenem at 5 g/day). MBM suggested that tobramycin considerably enhanced the imipenem target site concentration up to 2.6-fold. The combination regimens, rationally optimized via a translational modeling approach, demonstrated substantially enhanced bacterial killing and suppression of regrowth in vivo against a double-resistant isolate and are therefore promising for future clinical evaluation.

Abstract:Gelatin, resulting from collagen, a naturally occurring protein found in ligaments and tissues, is formed through the boiling of connective tissues, bones and animal skins, commonly sourced from cows. Its exceptional properties, including the ability to form strong, transparent gels and flexible films, easy digestibility, solubility in hot water and positive binding action, make gelatin a highly valuable resource in various industries such as food processing, pharmaceuticals, photography and paper production. It is regarded as non-toxic, biocompatible, biodegradable and generally safe for use. Gelatin serves as a key ingredient in the manufacturing of a wide range of products, with a notable example being hard gelatin capsules, which come in varying levels of solubility in water. In this study, a Schiff base was synthesized by modifying gelatin with salicylaldehyde in ethanol at room temperature, a mixture of gelatin-Schiff base added to appropriate copper chloride sonicated in ethanol, the mixture was poured onto glass plates and allowed to dry. Modified gelatin film was produced and characterized using FTIR spectroscopy, while its surface properties were examined through Scanning Electron Microscopy (SEM) and Atomic Force Microscopy (AFM). Furthermore, the modified gelatin film’s potential biological activity was assessed against bacterial strains. The antibacterial screening tests revealed that the modified gelatin exhibited promising antibacterial activity against microorganisms, indicating an improvement in its antibacterial efficacy. These findings highlight the enhanced antibacterial potential of the modified gelatin film, further contributing to its versatility and potential applications in various fields. Different molecular weight distributions within the gelatin can lead to variations in film formation and subsequent surface features. Additionally, the composition of the gelatin film, including the presence of any crosslinking agents or modified functional groups, can further influence the resulting surface morphology. This study focuses on the preparation of modified gelatin films using suitable aldehyde compounds and explores their potential for pharmaceutical applications. Various techniques assessment of biological activity will be employed to investigate the properties and performance of the modified gelatin films. Abstract:Poly(acrylamide-co-acrylamidoglycolic acid)/guar gum@ Ag-nanocomposite (AgNC@PAAG) hydrogels has been fabricated by a green protocol utilizing rhubarb stem-extract as bioreductant. The prepared nanocomposites (NCs) are formulated by varying guar gum (GG) polymer and cross-linker content, and used remarkably to study the release of an anticancer drug, 5-fluorouracil (FU). The AgNC@PAAG has demonstrated its potential in bacterial inactivation and p-nitrophenol (PNP) reduction. The AgNC@PAAG hydrogels showed extended FU release time, which was up to 23 h in pH 7.4. The higher zone of inhibition was documented for AgNC@PAAG against B. subtilis and E. coli. It was noticed that, the inhibition activity of AgNC@PAAG, was directly proportional to cross-linker content than the GG polymer. The efficiency of AgNC@PAAG as a nanocatalyst was evaluated for a model reduction reaction of p-nitrophenol (PNP) reduction by aqueous sodium borohydride (NaBH4), with an apparent rate constant of 121.8 × 10-3 min-1 at ambient temperature. The proposed nanocatalysts are reliable and recyclable, demonstrated its catalytic recycle efficacy of 85% after the third successive run. These NCs robust its biological and catalytic activity after embedding silver nanoparticles (AgNPs) by the bioreduction process; these optimized nanocatalysts can be remarkably used in biomedical healthcare sectors and industrial catalysis.
