Abstract:Type 2 diabetes is a major public health problem, accounting for significant premature mortality and morbidity. It has been reported to be associated with several chronic complications which involve coronary artery disease and renal and liver diseases that are considered the primary cause of disability and mortality in diabetic patients. Different studies stated that vitamin D have functional role in glucose tolerance through its effects on insulin secretion and sensitivity and ameliorate type 2 diabetes complication. For that this study was carried out to investigate the effect of dietary vitamin D on type 2 diabetes complication on the liver, pancreas, lipid profile, and brain function. Rats were divided into 5 groups (control, diabetic, Vitamin D treated, Vitamin D preventive, and vitamin D control). Blood and tissue samples were collected after 6 weeks for measurement of fasting blood glucose (FBG), HOMA-IR, fasting serum insulin (FSI), serum lipid profile, blood antioxidants [reduced glutathione (GSH),superoxide dismutase (SOD), and catalase], serum lipid peroxidation product (malondialdehyde (MDA)), and histopathological examination of the pancreas and liver, besides brain acetylcholinesterase (AChE) gene expression. Our results indicated that dietary vitamin D supplementation to diabetic rats significantly increased FSI, decreased FBG and HOMA-IR, enhancing lipid profile by increase HDL-C and decrease LDL-C, TAG, and VLDL-C; Vitamin D also ameliorate GSH level and catalase activity and decrease the level of serum MDA, in addition to depress expression of brain AChE gene and improving liver and pancreas architecture. From these results, it can be concluded that vitamin D assured great protection against type 2 diabetes complications in rats.
Vitamin D regulates insulin and ameliorates apoptosis and oxidative stress in pancreatic tissues of rats with streptozotocin-induced diabetes

Abstract:This study was designed to evaluate the potential therapeutic efficacy of vitamin D (Vit D) in averting the harmful effects of type 2 diabetes mellitus (T2D). Forty male Wistar rats were allotted into four groups: (1) the control, (2) Vit D, (3) streptozotocin (STZ), and (4) STZ + Vit D groups. Rats co-treated with Vit D had significantly ( p < 0.05) decreased levels of cortisol; proinflammatory cytokines, including interleukin-6 (IL-6); and malondialdehyde (MDA). Meanwhile, the levels of insulin significantly ( p < 0.05) increased, whereas the activity of the antioxidant system, including glutathione (GSH), superoxide dismutase (SOD), catalase, and total antioxidant capacity (TAC), significantly ( p < 0.05) decreased. Histopathological examination revealed the destruction of beta cells in the islets of Langerhans in rats with diabetes. Meanwhile, immunoexpression revealed an increase in the immunoreactivity of caspase-3 and endothelial nitric oxide synthase and a reduction in the immunoreactivity of insulin in rats with diabetes. In conclusion, Vit D ameliorated the harmful biochemical impact of diabetes mellitus, probably by increasing insulin secretion and sensitivity, ameliorating β-cell function, and decreasing cortisol levels; also, the anti-inflammatory effect of Vit D reduces the number of proinflammatory cytokines (e.g., IL-6) and increases the activity of the antioxidant system, such as GSH, SOD, TAC, and catalase while reducing lipid peroxidation enzymes (e.g., MDA).

Abstract:Purpose: The high morbidity and mortality associated with type 2 diabetes mellitus (T2DM) pose a significant global health challenge, necessitating the development of more efficient anti-diabetic drugs with fewer side effects. This study investigated the intervention of vitamin D3 combined with glibenclamide in rats with T2DM to elucidate its effects on pancreatic β-cells through the NF-κB pathway. Methods: Twenty-four healthy male Sprague-Dawley (SD) rats were randomly assigned to four groups: the control group (CG), the model group (MG), the glibenclamide group (GG), and the glibenclamide + vitamin D3 group (GDG). After inducing the T2DM model using high-fat and high-sugar diet and intraperitoneal injection of streptozotocin, the rats in the GG group were administered glibenclamide orally (0.6 mg/kg/day), while those in the GDG group received both glibenclamide (0.6 mg/kg/day) and vitamin D3 (500 IU/kg/day) in corn oil for a duration of 8 weeks. Biochemical indices were measured, and histopathological changes in pancreatic tissue and islet β cells were observed using hematoxylin and eosin staining. The expression of pancreatic nuclear factor κB (NF-κB), islet β-cells, and inflammatory cytokines were assessed using the TUNEL method and PCR. Results: According to the data from this current study, the GDG group showed significant positive differences in plasma biochemical indices, as well as in the expression of β cells, NF-κB p65, TNF-α, IL-1β, INF-γ, and Fas, compared to the GG and CG groups (P < 0.05). Conclusion: The results suggest that vitamin D has beneficial effects on T2DM by improving the functions of islet β cells through inhibition of the NF-κB signaling pathway. Therefore, it is suggested that vitamin D supplementation, when used alongside antidiabetic drugs, may more effectively prevent and treat T2DM.
Potential cardioprotective effect of Vitamin D and sodium-glucose transport protein 2 inhibitor in improving cardiac hypertrophy and fibrosis in Type 2 diabetic rats

Abstract:Background: Diabetes mellitus (DM) is a major risk factor for cardiovascular diseases. The progression of myocardial abnormalities due to DM occurs slowly but is progressive and asymptomatic. Sodium-glucose transport protein 2 inhibitors (SGLT-2i) and Vitamin D have potential cardioprotective properties that inhibit cardiomyocyte fibrosis and hypertrophy, which are early structural changes that occur in the heart of DM patients. Aim: The study aimed to determine the potential protective effects of SGLT-2i and Vitamin D administration on cardiac hypertrophy and fibrosis in Type 2 diabetic rats.Methods: This is an experimental study with a post-test-only control group design. Thirty-two male Wistar rats were given a high-fat/high-glucose (HF/HG) diet. After 3 weeks, rats were given an injection of streptozotocin (STZ 35 mg/kg) to induce pancreatic damage. The diabetic rats were then divided into four groups (n = 8 per group): untreated diabetic group (HF/HG/STZ), the diabetic group treated with empagliflozin (EMPA) 10 mg/kg body weight (BW) (HF/HG/ STZ+EMPA), the diabetic group treated with Vitamin D 225 IU/day (HF/HG/STZ+VitD), and the diabetic group treated with a combination of EMPA 10 mg/kg BW and Vitamin D 225 IU/day (HF/ HG/STZ+EMPA+VitD). Treatments were given by oral gavage for 8 weeks. Left ventricular biopsy was performed at week 13 to examine collagen deposition, the cardiomyocyte cross-sectional area (CSA), and the mRNA expression of β-myosin heavy chain (β-MHC) and transforming growth factor-β (TGF-β). All the obtained data were analyzed statistically. Results: The administration of EMPA, Vitamin D, and combination therapy of EMPA and Vitamin D reduced the mRNA expression of β-MHC and TGF-β in diabetic rats compared to the untreated diabetic group. The administration of EMPA, Vitamin D, and combination therapy also resulted in a decrease in both the cardiomyocyte CSA and collagen deposition. Compared to monotherapy, combination therapy led to significantly better parameter reduction. Conclusion: Administration of EMPA, Vitamin D, and combination therapy improved cardiac hypertrophy and fibrosis in type 2 diabetic rats.Relevance for Patients: The combination of Vitamin D and SGLT-2i may be proposed as a cardioprotective strategy and preventive measure to reduce the incidence of cardiovascular disease in patients with Type 2 DM.

Abstract:Previous studies have demonstrated that Ganoderma lucidum polysaccharides (Gl-PS) exhibited potential anti-hyperglycemic effect in rats. The aim of the present study was to investigate the mechanism of the hypoglycemic effect of a low-molecular-weight Gl-PS in streptozotocin (STZ)-induced diabetic Sprague-Dawley (SD) rats. Gl-PS was extracted and purified from Ganodema lucidum fruiting body. 50 male SD rats were included in the study; 10 were taken as healthy controls; 40 were induced to diabetes by a single injection of 65 mg/kg STZ, of which 30 were selected as successful diabetic rat models. The 30 diabetic rats were divided into three groups: Gl-PS (200 mg/kg Gl-PS), metformin (100 mg/kg metformin) and diabetic control (n = 10 per group). After eight weeks' oral administration, plasma concentrations of fasting glucose, triacylglyceride, total cholesterol and nitric oxide were significantly decreased in Gl-PS and metformin groups. Pancreatic superoxide dismutase, catalase and glutathione peroxidase were significantly increased in Gl-PS and metformin groups. Histopathological results showed that Gl-PS and metformin had protective effect on beta-cells. The mRNA expressions of Bcl-2 and PDX-1 in pancreas were up-regulated, but Bax, iNOS and Casp-3 down-regulated in Gl-PS and metformin groups compared to diabetic control group. The present results suggested that Gl-PS had a hypoglycemic effect in STZ-induced diabetic rats through preventing apoptosis of pancreatic beta-cells and enhancing beta-cells regeneration.

Abstract:Objectives: Testicular dysfunction has been associated with chronic hyperglycemia in diabetes mellitus patients. We investigated taurine's possible mechanisms and protective effects against testicular damage using a rat model of streptozotocin-induced diabetes. Methods: Wistar rats (N = 56) were divided into seven equal groups. Untreated control rats received saline, and treated control rats received taurine 50 mg/kg orally. To induce diabetes, rats received a single dose of streptozotocin. Metformin-treated diabetic rats received metformin at a dose of 300 mg/kg. Taurine-treated groups received 10, 25, or 50 mg/kg. All treatments were provided orally once a day for 9 weeks following the streptozotocin injection. Levels of blood glucose, serum insulin, cholesterol, testicular tumour necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), interleukin-1beta (IL-1β), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione (GSH), and catalase (CAT) were examined. Sperm count, progressive sperm motility, and sperm abnormalities were examined. Body and relative reproductive gland weights were assessed. Histopathological examinations of the testes and epididymis were performed. Results: Metformin as well as taurine (in a dose-dependent manner) resulted in significant improvements in body and relative reproductive gland weights, blood glucose, serum cholesterol, and insulin levels, as well as cytokine and oxidative parameters. These findings were associated with significant improvement in sperm count, progressive sperm motility, sperm abnormalities, and histopathological lesions in the testes and epididymis. Conclusion: Taurine can potentially improve hyperglycemia, hypercholesterolemia, and testicular damage associated with diabetes mellitus, possibly by controlling inflammation and oxidative stress.

Abstract:BACKGROUND Type 2 diabetes is one of the most prevalent chronic diseases worldwide, significantly impacting patients' quality of life. Current treatment options like metformin (MET) effectively counteract hyperglycemia but fail to alleviate diabetes-associated complications such as retinopathy, neuropathy, nephropathy, hepatopathy, and cardiovascular diseases. AIM To propose the supplementation of cholecalciferol (CHO) and taurine (TAU) to enhance MET efficacy in controlling diabetes while minimizing the risk of associated complications. METHODS The study involved sixty rats, including ten non-diabetic control rats and fifty experimental rats with type 2 diabetes induced by streptozotocin. The experimental rats were further subdivided into positive control and treatment subgroups. The four treatment groups were randomly allocated to a single MET treatment or MET combined with supplements either CHO, TAU, or both. RESULTS Diabetic rats exhibited elevated levels of glucose, insulin, Homeostasis Model Assessment of Insulin Resistance (HOMA-IR), glycated hemoglobin%, lipid markers, aspartate aminotransferase, and malondialdehyde, along with reduced levels of antioxidant enzymes (catalase and superoxide dismutase). The administration of CHO and TAU supplements alongside MET in diabetic rats led to a noticeable recovery of islet mass. The antioxidative, anti-inflammatory, and anti-apoptotic properties of the proposed combination therapy significantly ameliorated the aforementioned abnormalities. CONCLUSION The supplementation of CHO and TAU with MET showed the potential to significantly improve metabolic parameters and protect against diabetic complications through its antioxidative, anti-inflammatory, and anti-apoptotic effects.

Abstract:Diabetic nephropathy is the one of the major complication of diabetes mellitus which is the main cause of renal failure. The treatment of diabetic nephropathy includes different nonpharmacological and pharmacological approach, but poor prognosis. Our study was designed to investigate the combined effect of resveratrol and vitamin D3 on the glucose homeostasis, histological changes, and cell damage biomarkers in streptozotocin-induced diabetic rat kidneys. Oral treatment with resveratrol (10 mg/kg/day), vitamin D3 (0.5 µgm/kg), and the combination for 10 weeks showed an increase in the average body weights compared to the untreated diabetic rats. The combination showed a highly significant decrease in fasting blood glucose levels, serum cholesterol levels compared to the untreated diabetic rats, there is a significant increase in serum HDL cholesterol levels. There is a highly significant decrease in serum creatinine, urea, albumin levels compared to the untreated diabetic rats. However, the combination treatment provides superior efficacy than individual drug treatments. Histopathological examination of the kidney tissue section revealed that the diabetic rats treated with resveratrol, vitamin D3, and the combination showed noticeable improvement in the histopathological parameters compared to untreated diabetic rats. However, a marked reduction of TNF-α and IL-6 expression was observed in combination treated rats compared to individual drug treatments. Combination therapy was significantly more effective in treating diabetic nephropathy than resveratrol and vitamin D3 alone. Resveratrol and vitamin D3 has a protective role of diabetic kidneys.
Comparative Analysis of the Effects of Selected Medicinal Plants on Gut Microbiota and Glycemic Control in Streptozotocin-Induced Diabetic Rats

Abstract:The study focused on conducting a comparative analysis of the impact of selected medicinal plants on intestinal flora during diabetes management in rats induced with streptozotocin. Thirty male albino rats were utilised for the experiment, with 36 rats subjected to the induction of diabetes using streptozotocin. By injecting streptozotocin intraperitoneally at a dose of 75 mg/kg body weight dissolved in normal saline, diabetes type 2 was induced. Following the induction, the rats were administered a 5% glucose solution to prevent hypoglycemic shock. Blood glucose levels were monitored regularly, and rats above 200 mg/dl were deemed diabetic and added to the study. Glibenclamide 45 mg/kg b.w., Bitter melon, Okra, and akuamma leaves. 200 mg/kg b.w., and leaves were administered daily for 28 days in diabetic rats. Some intestinal taxa changed as a result of streptozotocin-induced diabetes. The percentage of Enterobacteriaceae increased versus Control. Pseudomonas aeruginosa, influenced the quantitative proportion of some intestinal species (Bacillus subtilis, proteus mirabilis, E. coli, shigella flauor). Glibenclamide stimulates the growth of Lactobacilli and Eubacterium spp. But suppresses the G+/G- ratio in small intestinal samples. The results indicated that administering plant extracts significantly reduced blood glucose and a notable presence of microbes suitable for insulin secretion and sugar fermentation. Additionally, the study highlighted the potential of plant extracts to positively impact intestinal flora, suggesting a promising avenue for managing diabetes in streptozotocin-induced rats.

Abstract:Diabetes mellitus (DM) is a serious health issue for the general public. In this work, male diabetic rats were used to determine the impact of Citrullus colocynthis and Phyllanthus Emblica fruits ethanolic extract (EECC and EEPE). Six groups of thirty mature rats (male Sprague Dawley) were examined. In the first group, normal rats have been fed a balanced diet served as the negative control group (C-); in the second group, diabetic rats served as the control positive (C+). Diabetic rats in groups 3, 4, 5, and 6 received daily treatments of (200 and 400 mg kg-1 body weight (BW) EECC and EEPE, respectively. After four weeks of treatment, Biochemical indicators were examined in all blood samples. Streptozotocin was injected and resulted in a significant reduction in feed intake (FI), feed efficiency (FER) body weight gain (BWG), serum insulin, high-density lipoprotein (HDL-c), globulin, albumin, and total protein, as well as antioxidant enzymes, glutathione (GSH) and superoxide dismutase (SOD) in pancreatic tissue, whereas a significant increment was noted in serum glucose, triglyceride (TG), total cholesterol (TC), low & very low-density lipoprotein cholesterol (LDL-c and VLDL-c), atherogenic index (AI), Alkaline phosphatase (ALP), Aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, uric acid, creatinine, tumor necrosis factor-α, creatine phosphate kinase (CPK) and lactate dehydrogenase (LDH) and lipid peroxidation (MDA) in pancreatic tissue. The treatment of diabetic animals with different ethanolic extracts caused improvements in all previously measured parameters, with EEPE (400 mg kg-1 BW) showing the greatest improvement.
Potential Effect of Curcumin in Lowering Blood Glucose Level in Streptozotocin-Induced Diabetic Rats

Abstract:Purpose: The prevalence of diabetes mellitus has significantly increased, with 537 million individuals living with diabetes in 2021. Curcumin, a natural compound present in turmeric, has anti-inflammatory and antioxidant properties that aid in controlling diabetes. Curcumin can lower blood glucose levels, increase pancreatic cell function, and reduce insulin resistance. The pathophysiology of diabetes involves oxidative stress and endoplasmic reticulum stress, which can lead to cell death. This study aimed to evaluate the antidiabetic activity of curcumin in rats by administering it for a month and evaluating pancreatic tissue histology. Patients and methods: STZ-induced diabetic rats were fed a high-fat diet containing glibenclamide, 200 mg/kg body weight (BW) curcumin, 400 mg/kg BW curcumin, or a placebo for 4 weeks. After intervention, blood glucose levels were measured, and the pancreatic tissue was examined. Blood glucose levels were measured at 0, 2, 4, 6, and 8 h. Results: One-way ANOVA was performed to measure the mean difference among the groups at 0, 2, 4, 6, and 8 h of observation, which reported a statistically significant difference (p < 0.05). The blood glucose levels decreased after 4 h in the group receiving curcumin. Histological evaluation of the pancreas showed slight hydropic degeneration after 4 weeks of curcumin treatment. Conclusion: Our study indicates that curcumin has a beneficial effect in diabetic rats by reducing blood glucose levels and a protective effect on the pancreas.
Effects of streptozotocin-induced diabetes and of insulin treatment on homocysteine metabolism in the rat.

Abstract:An elevation in the concentration of total plasma homocysteine is known to be an independent risk factor for the development of vascular disease. Alterations in homocysteine metabolism have also been observed clinically in diabetic patients. Patients with either type 1 or type 2 diabetes who have signs of renal dysfunction tend to exhibit elevated total plasma homocysteine levels, whereas type 1 diabetic patients who have no clinical signs of renal dysfunction have lower than normal plasma homocysteine levels. The purpose of this study was to investigate homocysteine metabolism in a type 1 diabetic animal model and to examine whether insulin plays a role in its regulation. Diabetes was induced by intravenous administration of 100 mg/kg streptozotocin to Sprague-Dawley rats. We observed a 30% reduction in plasma homocysteine in the untreated diabetic rat. This decrease in homocysteine was prevented when diabetic rats received insulin. Transsulfuration and remethylation enzymes were measured in both the liver and the kidney. We observed an increase in the activities of the hepatic transsulfuration enzymes (cystathionine beta-synthase and cystathionine gamma-lyase) in the untreated diabetic rat. Insulin treatment normalized the activities of these enzymes. The renal activities of these enzymes were unchanged. These results suggest that insulin is involved in the regulation of plasma homocysteine concentrations by affecting the hepatic transsulfuration pathway, which is involved in the catabolism of homocysteine.

We performed a meta-analysis of controlled clinical trials that assessed glycemic outcome measures among adults at risk for type 2 diabetes, including prediabetes, overweight, or obesity. We searched PUBMED/ MEDLINE, CINAHL, and Google Scholar databases for trials published prior to April 2017. Placebo-controlled clinical trials with random allocation to vitamin D with or without calcium supplementation were selected. Data collection included country, study design, inclusion criteria, sample size, form, and dose of vitamin D, supplementation interval, control group, duration, participant characteristics, comorbidities, baseline and follow-up serum 25-hydroxyvitamin D [25(OH)D] concentration, and available outcome measures [glycosylated hemoglobin (HbA1c), fasting plasma glucose, plasma glucose after 2-hour oral glucose tolerance test, and homeostatic model assessment of insulin resistance (HOMA-IR)]. Data synthesis was conducted using random-effect models (PROSPERO registration no. CRD42017055326). Twenty-eight trials, representing 3848 participants, met the eligibility criteria. Compared with the control group, vitamin D supplementation significantly reduced HbA1c level by -0.48% (95% CI, -0.79 to -0.18), fasting plasma glucose level by -0.46 mmol/L (95% CI, -0.74 to -0.19), and HOMA-IR level by -0.39 (95% CI, -0.68 to -0.11). Subgroup analysis revealed that the effects of vitamin D supplementation on different glycemic measures were influenced by age, calcium coadministration, vitamin D deficiency, serum 25(OH)D level after supplementation, and duration of supplementation. Vitamin D supplementation and improved vitamin D status improved glycemic measures and insulin sensitivity and may be useful as part of a preventive strategy for type 2 diabetes.

Abstract:Background & objectives: The combination of drugs with herbs is a prevalent treatment for metabolic and chronic diseases. This study combined glibenclamide with vitexin and performed in vivo and in vitro investigations, aiming to evaluate their synergistic effect on type 2 diabetes mellitus (T2DM). Materials and methods: T2DM rat models were established by injecting streptozotocin, and the sham group was injected with a normal saline solution as the control. A single dosage was orally performed with 10 mg/kg glibenclamide, while the co-administrated was conducted by the pre-treatment of 20 mg/kg vitexin for 7 d to avoid potential chemical interactions. The pharmacokinetics of glibenclamide was evaluated in both sham and T2DM rats. The blood glucose of T2DM rats was detected in the presence of different administration strategies. The metabolic stability of glibenclamide was assessed in rat liver microsomes. Results: Glibenclamide showed a significant hypoglycemic effect on T2DM rats, which was enhanced by the co-administration of vitexin. Vitexin significantly affected the pharmacokinetics of glibenclamide in both sham and T2DM rats, where the AUC0-t, Cmax, and t1/2 of glibenclamide significantly increased, and the clearance rate decreased. Moreover, vitexin increased the in vitro half-life and decreased intrinsic clearance of glibenclamide in rat liver microsomes. The previously reported inhibitory effect of vitexin on CYP2C9 activity was hypothesized as the mechanism underlying the interaction. Conclusion: Co-administration of glibenclamide and vitexin would increase the systemic exposure and metabolic stability of glibenclamide and exert a synergistic effect on T2DM.
