Abstract:Magnesium phosphate cement (MPC) has been evaluated as a novel bone substitute owing to its favorable biocompatibility, plasticity, and osteogenic potential. However, the low porosity of MPC prevents growth factors and osteoblasts from fully growing into the material, thereby limiting its clinical use. In this study, different concentrations (0-5%) of calcium carbonate and citric acid (CA) were used as foaming agents to prepare porous MPC. The MPC containing 3% CaCO3/CA exhibited the best physicochemical properties, including greater porosity, improved injectability, extended setting time, and decreased hydration temperature. The proliferation and adhesion of cells on 3%CaCO3/CA-MPC were higher than those on MPC alone. To explore its osteogenesis in vivo, 3% CaCO3/CA-MPC and Bio-Oss® bone powder were implanted into periodontal bone defects in rats for 4 weeks and 12 weeks, respectively. Micro-CT and histological analysis demonstrated the improved bone regeneration of 3%CaCO3/CA-MPC compared to the blank group (P < 0.05); it had slightly lower bone regeneration than the Bio-Oss® group but no statistical difference. The results indicated that porous MPC foamed with calcium carbonate and CA improved its physicochemical properties and enhanced its biocompatibility, making it a promising material for bone regeneration.
Injectable and strong nano-apatite scaffolds for cell/growth factor delivery and bone regeneration

Abstract:Objectives: Seven million people suffer bone fractures each year in the U.S., and musculoskeletal conditions cost $215 billion/year. The objectives of this study were to develop moldable/injectable, mechanically strong and in situ-hardening calcium phosphate cement (CPC) composite scaffolds for bone regeneration and delivery of osteogenic cells and growth factors. Methods: Tetracalcium phosphate [TTCP: Ca(4)(PO(4))(2)O] and dicalcium phosphate (DCPA: CaHPO(4)) were used to fabricate self-setting calcium phosphate cement. Strong and macroporous scaffolds were developed via absorbable fibers, biopolymer chitosan, and mannitol porogen. Following established protocols, MC3T3-E1 osteoblast-like cells (Riken, Hirosaka, Japan) were cultured on the specimens and inside the CPC composite paste carrier. Results: The scaffold strength was more than doubled via reinforcement (p<0.05). Relationships and predictive models were established between matrix properties, fibers, porosity, and overall composite properties. The cement injectability was increased from about 60% to nearly 100%. Cell attachment and proliferation on the new composite matched those of biocompatible controls. Cells were able to infiltrate into the macropores and anchor to the bone mineral-like nano-apatite crystals. For cell delivery, alginate hydrogel beads protected cells during cement mixing and setting, yielding cell viability measured via the Wst-1 assay that matched the control without CPC (p>0.1). For growth factor delivery, CPC powder:liquid ratio and chitosan content provided the means to tailor the rate of protein release from CPC carrier. Significance: New CPC scaffolds were developed that were strong, tough, macroporous and osteoconductive. They showed promise for injection in minimally invasive surgeries, and in delivering osteogenic cells and osteoinductive growth factors to promote bone regeneration. Potential applications include various dental, craniofacial, and orthopedic reconstructions.
Implementation of network traffic classifier using semi supervised machine learning approach

Abstract:Network Traffic Classification using classical techniques such as port number based and payload based is becoming very difficult because many applications use dynamic port number and encryption technique to avoid detection. To overcome the drawbacks of classical techniques various machine learning techniques were proposed. Machine learning technique faces the problem of labeled instances (in supervised learning) and time consuming manual work (in unsupervised learning). To address the above problems we proposed a semi supervised machine learning technique. The key idea of proposed technique is to build a classifier from training dataset consisting of both labeled and unlabeled instances. For experimental purpose KDD CUP 99 intrusion detection dataset and MATLAB tool is used. We evaluate and compare the performance of the classifier build with 10%, 20% and 30% labeled instances in training dataset. The result of experiments show that classifier build with 30% labeled instances in training dataset has better performance at number of clusters equals to 50.

Abstract:Modified macroporous structures and active osteogenic substances are necessary to overcome the limited bone regeneration capacity and low degradability of self-curing calcium phosphate cement (CPC). Curcumin (CUR), which possesses strong osteogenic activity and poor aqueous solubility/bioavailability, esterifies the side chains in hyaluronic acid (HA) to form a water-soluble CUR-HA macromolecule. In this study, we incorporated the CUR-HA and glucose microparticles (GMPs) into the CPC powder to fabricate the CUR-HA/GMP/CPC composite, which not only retained the good injectability and mechanical strength of bone cements, but also significantly increased the cement porosity and sustained release property of CUR-HA in vitro . CUR-HA incorporation greatly improved the differentiation ability of bone marrow mesenchymal stem cells (BMSCs) to osteoblasts by activating the RUNX family transcription factor 2/fibroblast growth factor 18 (RUNX2/FGF18) signaling pathway, increasing the expression of osteocalcin and enhancing the alkaline phosphatase activity. In addition, in vivo implantation of CUR-HA/GMP/CPC into femoral condyle defects dramatically accelerated the degradation rate of cement and boosted local vascularization and osteopontin protein expression, and consequently promoted rapid bone regeneration. Therefore, macroporous CPC based composite cement with CUR-HA shows a remarkable ability to repair bone defects and is a promising translational application of modified CPC in clinical practice.

Abstract:Background: Calcium phosphate cement (CPC) can be molded or injected to form a scaffold in situ, which intimately conforms to complex bone defects. Bioactive glass (BG) is known for its unique ability to bond to living bone and promote bone growth. However, it was not until recently that literature was available regarding CPC-BG applied as an injectable graft. In this paper, we reported a novel injectable CPC-BG composite with improved properties caused by the incorporation of BG into CPC. Materials and methods: The novel injectable bioactive cement was evaluated to determine its composition, microstructure, setting time, injectability, compressive strength and behavior in a simulated body fluid (SBF). The in vitro cellular responses of osteoblasts and in vivo tissue responses after the implantation of CPC-BG in femoral condyle defects of rabbits were also investigated. Results: CPC-BG possessed a retarded setting time and markedly better injectability and mechanical properties than CPC. Moreover, a new Ca-deficient apatite layer was deposited on the composite surface after immersing immersion in SBF for 7 days. CPC-BG samples showed significantly improved degradability and bioactivity compared to CPC in simulated body fluid (SBF). In addition, the degrees of cell attachment, proliferation and differentiation on CPC-BG were higher than those on CPC. Macroscopic evaluation, histological evaluation, and micro-computed tomography (micro-CT) analysis showed that CPC-BG enhanced the efficiency of new bone formation in comparison with CPC. Conclusions: A novel CPC-BG composite has been synthesized with improved properties exhibiting promising prospects for bone regeneration.

Abstract:Calcium phosphate cements (CPCs) have been widely used as bone graft substitutes for many years. The aim of this study was to evaluate the biocompatibility of two novel injectable, bioactive cements: beta-tricalcium phosphate (beta-TCP)/CPC and chitosan microsphere/CPC in vitro and in vivo. This was accomplished by culturing mouse pre-osteoblastic cells (MC3T3-E1) on discs and pastes of CPCs. Cell growth, adhesion, proliferation and differentiation were assessed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide and alkaline phosphatase assays as well as by scanning electron microscopy and fluorescence. The effect of CPC paste curing was also evaluated. Implantation of two materials into the muscle tissue of rabbits was also studied and evaluated by histological analysis. Cell analysis indicated good biocompatibility in vitro. The fluorescence assay suggested that the cured material discs had no obvious effect on cell growth, while the curing process did. Histological examination showed no inflammatory cell infiltration into soft tissue. These data suggest that beta-TCP/CPC and chitosan microsphere/CPC composites may be promising injectable material for bone tissue engineering.
Gold nanoparticles in injectable calcium phosphate cement enhance osteogenic differentiation of human dental pulp stem cells

Abstract:In this study, a novel calcium phosphate cement containing gold nanoparticles (GNP-CPC) was developed. Its osteogenic induction ability on human dental pulp stem cells (hDPSCs) was investigated for the first time. The incorporation of GNPs improved hDPSCs behavior on CPC, including better cell adhesion (about 2-fold increase in cell spreading) and proliferation, and enhanced osteogenic differentiation (about 2–3-fold increase at 14 days). GNPs endow CPC with micro-nano-structure, thus improving surface properties for cell adhesion and subsequent behaviors. In addition, GNPs released from GNP-CPC were internalized by hDPSCs, as verified by transmission electron microscopy (TEM), thus enhancing cell functions. The culture media containing GNPs enhanced the cellular activities of hDPSCs. This result was consistent with and supported the osteogenic induction results of GNP-CPC. In conclusion, GNP-CPC significantly enhanced the osteogenic functions of hDPSCs. GNPs are promising to modify CPC with nanotopography and work as bioactive additives thus enhance bone regeneration.

Abstract:Abstract Literature search revealed no systematic report on iron oxide nanoparticle-incorporating calcium phosphate cement scaffolds (IONP-CPC). The objectives of this study were to: (1) use γFe2O3 nanoparticles (γIONPs) and αFe2O3 nanoparticles (αIONPs) to develop novel IONP-CPC scaffolds, and (2) investigate human dental pulp stem cells (hDPSCs) seeding on IONP-CPC for bone tissue engineering for the first time. IONP-CPC scaffolds were fabricated. Physiochemical properties of IONP-CPC scaffolds were characterized. hDPSC seeding on scaffolds, cell proliferation, osteogenic differentiation and bone matrix mineral synthesis by cells were measured. Our data demonstrated that the osteogenic differentiation of hDPSCs was markedly enhanced via IONP incorporation into CPC. Substantial increases (about three folds) in ALP activity and osteogenic gene expressions were achieved over those without IONPs. Bone matrix mineral synthesis by the cells was increased by two- to three folds over that without IONPs. The enhanced cellular osteogenesis was attributed to: (1) the surface nanotopography of IONP-CPC scaffold, and (2) the cell internalization of IONPs released from IONP-CPC scaffold. Our results demonstrate that the novel CPC functionalized with IONPs is promising to promote osteoinduction and bone regeneration. In conclusion, it is highly promising to incorporate γIONPs and αIONPs into CPC scaffold for bone tissue engineering, yielding substantially better stem cell attachment, spreading and osteogenic differentiation, and much greater bone mineral synthesis by the seeded cells. Therefore, novel CPC scaffolds containing γIONPs and αIONPs are promising for dental, craniofacial and orthopaedic applications to substantially enhance bone regeneration.
Effects of adding resorbable chitosan microspheres to calcium phosphate cements for bone regeneration

Abstract:Calcium phosphate cements (CPCs) have been widely used as bone graft substitutes. However, the undesirable osteoinductivity and slow degradability of CPCs greatly hamper their clinical application. The aim of this study was to synthesize a type of injectable, bioactive cement. This was accomplished by incorporating chitosan microspheres into CPC. CPC containing chitosan microspheres was analyzed by X-ray diffraction (XRD) and scanning electron microscope (SEM). XRD showed that the hardened chitosan microsphere/CPC with different proportions of microspheres contained diffraction peaks of hydroxyapatite and chitosan. Compressive strength and dissolution in simulated body fluid were measured. The chitosan microsphere/CPC containing 10% (w/w) chitosan microspheres had a compressive strength of 14.78 ± 0.67 MPa. Cavity defects were created in both femoral condylar regions of New Zealand White rabbits. Chitosan microsphere/CPC (composite group) and α-TCP/CPC (control group) were implanted separately into the bone defects of both femurs. X-ray analysis was performed to observe the filling of these bone defects 3 days after surgery. The extent of bone substitute degradation and new bone formation were evaluated by SEM and histological examination at 8, 16, and 24 weeks after implantation. These results showed far more new bone formation and degradation of the chitosan microsphere/CPC composite in the bone defects. These data indicate that a chitosan microsphere/CPC composite might be considered as a promising injectable material for the generation of new bone tissue.
Nanocrystalline calcium sulfate/hydroxyapatite biphasic compound as a TGF-β1/VEGF reservoir for vital pulp therapy.

Abstract:OBJECTIVES:Vital pulp therapy aims to treat reversible pulpal injuries via protective dentinogenesis and to preserve more tooth structure. Mineral trioxide aggregate (MTA)-based capping materials demonstrate prolonged setting time increases the risk of pulpal infection during multi-visit treatment. Their non-degradable property occupies pulp space and limits dentin-pulp regeneration. This study reports an inorganic degradable biomaterial that presents a short initial setting time and acts as a growth factor reservoir to promote reparative dentinogenesis. METHODS:We synthesize nanocrystalline calcium sulfate hemihydrate (nCS), hydroxyapatite (HAp) and calcium sulfate hemihydrate (CS) as a reservoir to which transforming growth factor-beta 1 (TGF-β1) and vascular endothelial growth factor (VEGF) are added (denoted as nCS/HAp/CS/TGF-β1/VEGF). In vitro biocompatibility and mineralization (the activity and expression of alkaline phosphatase, ALP) were evaluated. Rat animal model was created to test in vivo efficacy. RESULTS:Cultured human dental pulp cells (HDPCs) showed that nCS/HAp/CS/TGF-β1/VEGF cement has excellent biocompatibility and the potential to elevate the activity and expression of ALP. The in vivo efficacy (rat animal model) indicates protective dentin by micro-computed tomography (μ-CT) measurements and histological analyses. The 3D μ-CT non-destructive analysis also determines volume changes during pulpotomy, suggesting that the degraded space of the nCS/HAp/CS/TGF-β1/VEGF cement is repaired by the formation of dentin-pulp tissue. SIGNIFICANCE:These findings demonstrate that nCS/HAp/CS cement acts as a potent reservoir for the sustained release of growth factors, and that nCS/HAp/CS/TGF-β1/VEGF cement has a high potential to form the reparative dentinogenesis in vivo.
Substance P incorporation in calcium phosphate cement for dental alveolar bone defect restoration

Abstract:A combination of osteoinductive neuropeptide substance P (SP) and osteoconductive bone cement of calcium phosphate (CPC) might provide an effective and lower-cost solution for complex alveolar bone defects restoration. The present study aims to investigate the key design considerations of SP delivery in CPC. In this study, CPC-based modular scaffolds were developed, where collagen type I was used as accessory organic ingredient to modulate the physical and biological characters. SP was directly mixed in the cement as free peptides, or was covalently immobilized with collagen component. The structural and mechanical properties of the scaffolds were assessed in vitro, and their osteogenic ability was observed in a rabbit model with alveolar bone defect. The results showed that SP could enhance the osteo-conductivity/inductivity of CPC. Collagen solution optimized biocompatibility of CPC, and meanwhile exhibited additive effects on the functions of SP. Nevertheless, immobilization of SP with collagen blocked their bioactivity in CPC. Collagen sponges created macro-porosity in CPC and achieved maximum bone ingrowth with the aid of SP. In conclusion, the present study primarily demonstrated that CPC scaffold can be functionalized by synthetic SP, and the biocompatibility and porosity of the scaffold are adaptable key factors determining their final osteogenic activities.
[In Vivo Study of Strontium-Doped Calcium Phosphate Cement for Biological Properties].

Abstract:OBJECTIVE:To evaluate the biocompatibility and osteogenic effect of new calcium phosphate cement (CPC) in vivo and to provide experimental basis for its further clinical application.METHODS:Thirty New Zealand white rabbits were randomly divided into four groups: CPC group, CPC+Bio-Oss group, Bio-Oss group and blank control group. Bone defect models of 6 mm in diameter and 7 mm in depth were made on the lateral condyle of bilateral hind legs of the rabbits. CPC, Bio-Oss and CPC+Bio-Oss mixture were implanted into the bone defect according to the group, and the mass ratio of CPC and Bio-Oss was 4 ∶ 1. The experimental animals were sacrificed the 4th, 12th and 24th week after operation. The tissue around the bone defect was taken for histological evaluation by H&E staining. Bone ingrowth fraction (BIF) was calculated. The expression of BMP-2 and COL-Ⅰ was detected by immunohis- tochemical staining by calculating the mean optical density (MOD) of the positive area the 4th week after operation, and the bone healing of each group was evaluated at different time points. The measurement data were analyzed by one-way ANOVA and LSD test was used for multiple comparison of the differences between the means by SPSS 19.0. P < 0.05 was considered to be statistically significant.RESULTS:The results of H&E staining showed that the BIF values of CPC group, CPC + Bio-Oss group and Bio-Oss group were significantly higher than those of blank control group at the same time point (P < 0.01). The BIF values of CPC group were lower than those of Bio-Oss group and CPC + Bio-Oss group (P < 0.01). There was no significant difference between CPC + Bio-Oss group and Bio-Oss group. Immunohistochemical staining showed that the MOD values of BMP-2 and COL-Ⅰ in CPC group were higher than those in blank control group, but lower than those in Bio-Oss group and CPC+Bio-Oss group (P < 0.01). There was no significant difference between BMP-2 and COL-Ⅰ in CPC+Bio-Oss group and Bio-Oss group.CONCLUSION:The new calcium phosphate cement has good biocompatibility and can promote early osteogenesis with stable and long-term effect.
A type of injectable porous calcium phosphate cement in vivo

Abstract:BACKGROUND: Calcium phosphate cement (CPC) has the disadvantages of brittleness, low waterborne (blood-soluble), poor mechanical property and conductibility which limit its clinical application in some degree. Therefore its modification research is in need. OBJECTIVE: To prepare a type of porous calcium phosphate cement with certain intensity and porosity for the bone growth. METHODS: This study was based on the CPC system. The liquid phase was the weak acidic solution of chitosan which had favorable biocompatibility and high viscosity to achieve injectable of CPC. It could strongly improve the application and comfort of CPC. The solid phase was biphasic calcium phosphate (four calcium phosphate + calcium hydrogen phosphate) system. The pores of mannitol and lactic-co-glycolic acid were added into solid phase to prepare the calcium phosphate scaffold. RESULTS AND CONCLUSION: The aperture of the prepared material could reach to 10-300 μm. The porosity of the solidified CPC could reach to (68.3±1.5)% when the proportion of the porogen was 60%. The increased porosity of CPC decreased the mechanical properties of the material, the compressive strength of CPC without porogen was (53.0±1.4) MPa initially, and dropped to (2.5±0.2) MPa with 60% porogen. The CPC material has better compressive strength and favorable biocompatibility, and can be degraded in vivo and used as a type of injectable scaffold materials.

Abstract:The integration of phosphate‐based glass into methylmethacrylate‐based cement (PIMC) presents promising characteristics for vital pulp treatment. PIMC demonstrates persistent strength and stability in wet conditions, effectively controls initial inflammation, and stimulates human dental pulp stem cells. Overall, PIMC supports mineralized tissue repair at the dentin‐pulp interface, making it an excellent candidate as a bioactive material for vital pulp therapy. Vital pulp treatment (VPT) is crucial for preserving the health and function of the tooth in cases where the pulp tissue remains vital despite exposure. Various materials are introduced for this purpose. However, challenges such as low strength, high solubility, and tooth discoloration persist. Methylmethacrylate‐based cement (MC) offers excellent sealing ability, feasibility, and mechanical properties, making it a promising alternative for VPT. Phosphate‐based glass (PBG) has the potential to promote hard tissue regeneration by releasing key inducers, phosphorus (P) and calcium (Ca), for reparative odontogenesis. This study investigates PBG‐integrated MC (PIMC) by characterizing its properties, assessing human dental pulp stem cell activity related to initial inflammatory adaptation and odontogenic differentiation, and evaluating hard tissue formation using an in vivo dog pulpotomy model. Results indicate that a 5% PBG‐integrated MC (5PIMC) maintains the physicochemical properties of MC. Furthermore, 5PIMC demonstrates cytocompatibility, excellent expression of osteo/odontogenic markers, and resistance to inflammatory markers, significantly outperforming MC. Enhanced hard tissue formation is observed in the dental pulp of mongrel dog teeth treated with 5PIMC. These findings suggest that 5PIMC could be an optimal and suitable material for reparative odontogenesis through VPT.

Abstract:As an injectable scaffold material for bone tissue engineering, calcium phosphate cement (CPC) has good biocompatibility, self-setting, and osteoconduction properties. Alginate-microencapsulated seed cells can pick up the degradation speed and bioactivity of CPC. The aim of this study was to explore the osteogenic ability of a composite of microencapsulated rabbit bone marrow mesenchymal stem cells (rBMMSCs) with β-tricalcium phosphate/calcium phosphate cement (β-TCP/CPC) in vivo. Cavity defects were created in both femoral condylar regions of New Zealand White rabbits. β-TCP/CPC (control group) and alginate microencapsulated rBMMSCs/β-TCP/CPC composite (composite group) were implanted separately into the bone defects of both femurs. Bone substitute degradation and new bone formation were evaluated by CBCT, and the defects were examined histologically 8, 16, and 24 weeks after implantation. In addition, fluorescent carbocyanine CM-Dil was used to track the rBMMSCs in vivo after implantation. The results showed that far more new bone and bone marrow grew into the bone defects in the composite group. Few CM-Dil labeled positive cells were observed postoperatively. However more native cells were detected in the graft areas of the composite group than those of the control group. The study indicates that a composite of microencapsulated seed cells/β-TCP/CPC might be considered as a promising injectable material for the generation of new bone tissue.
Promoting bone regeneration of calcium phosphate cement by addition of PLGA microspheres and zinc silicate via synergistic effect of in-situ pore generation, bioactive ion stimulation and macrophage immunomodulation

Abstract:Calcium phosphate cement (CPC) is an excellent bone graft material due to its self-setting, easiness to shape, good biocompatibility and osteoconductivity. However, low osteogenic activity and slow degradation rate of CPC severely influence its bone regeneration efficiency. Herein, zinc silicate (ZS) and poly (lactic-co-glycolic acid) (PLGA) microspheres were incorporated into CPC to overcome these issues. The addition of ZS obviously downregulated the expression of inflammatory-related genes (interleukin-1β, interleukin-6, and tumor necrosis factor α), while markedly upregulated the anti-inflammatory gene expression (interleukin-10). The incorporation of ZS significantly promoted the adhesion and proliferation of mouse bone marrow stem cells (mBMSCs). In addition, macrophage-conditioned ZS/PLGA/CPC extracts profoundly promoted osteogenic differentiation of mBMSCs, and evidently suppressed osteoclastogenesis of RAW264.7 cells. The optimum added amount of ZS was 10 wt.%. In vivo results also displayed that the addition of 10 wt.% ZS significantly improved bone repair effect of the composite. Therefore, the addition of ZS together with PLGA microspheres is a promising way for modifying the osteogenic activity and degradability of CPC via in-situ pore generation, promoting osteogenic differentiation and macrophage immunomodulation due to the simultaneous release of active silicon and zinc ions.

Abstract:Vital pulp preservation, which is a clinical challenge of aseptic or iatrogenic accidental exposure of the pulp, in cases direct pulp capping is the main technology. Human dental pulp stem cells (hDPSCs) play a critical role in pulp tissue repair, but their differentiative ability could be inhibited by the potential infection and inflammatory response of the exposed pulp. Therefore, inflammatory regulation and differentiated promotion of hDPSCs are both essential for preserving living pulp teeth. In this study, we constructed a functional dental pulp-capping hydrogel by loading cerium oxide nanoparticles (CNPs) and dentin matrix protein-1 (DMP1) into an injectable Fmoc–triphenylalanine hydrogel (Fmoc-phe3 hydrogel) as CNPs/DMP1/Hydrogel for in situ drugs delivery. With a view to long-term storage and release of CNPs (anti-inflammatory and antioxidant) to regulate the local inflammatory environment and DMP1 to promote the regeneration of dentin. Results of CCK-8, LDH release, hemolysis, and Live/Dead assessment of cells demonstrated the good biocompatibility of CNPs/DMP1/Hydrogel. The levels of alkaline phosphatase activity, quantification of the mineralized nodules, expressions of osteogenic genes and proteins demonstrated CNPs/DMP1/Hydrogel could protect the activity of hDPSCs' osteogenic/dentinogenic differentiation by reducing the inflammation response via releasing CNPs. The therapy effects were further confirmed in rat models, CNPs/DMP1/Hydrogel reduced the necrosis rate of damaged pulp and promoted injured pulp repair and reparative dentin formation with preserved vital pulps. In summary, the CNPs/DMP1/Hydrogel composite is an up-and-coming pulp-capping material candidate to induce reparative dentin formation, as well as provide a theoretical and experimental basis for developing pulp-capping materials.

Abstract:PURPOSE Hyperpolarized xenon-129 dissolved-phase MRI is the first imaging technique that allows 3-dimensional regional mapping of ventilation and gas uptake by tissue and blood the in human lung. Multiple outcome measures can be produced from this method. Existing studies in subjects with major lung diseases compared to healthy controls demonstrated high sensitivities of this method to pulmonary physiological factors including ventilation, alveolar tissue density, surface-to-volume ratio, pulmonary perfusion and gas-blood barrier thickness. The purpose of this study is to evaluate the utility of this new imaging tool to assess the lung function in patients with non-small cell lung cancer (NSCLC). METHODS Ten healthy controls (age: 63±10) and five patients (age: 62±13) with NSCLC underwent the xenon-129 dissolved-phase MRI, pulmonary function test (PFT) and CT for clinical purpose. Three outcome measures were produced from xenon-129 dissolved-phase MRI, including ventilation defect fraction (Vdef%) reflecting the airflow obstruction, tissue-to-gas ratio reflecting lung tissue density, and RBC-to-tissue ratio reflecting pulmonary perfusion and gas exchange. RESULTS Compared to healthy controls, patients with NSCLC showed more ventilation defects (NSCLC: 22±6%; control: 40±18%; P=0.01), lower tissue-to-gas (NSCLC: 0.82±0.31%; control: 1.07±0.13%; P=0.05) and RBC-to-tissue ratios (NSCLC: 0.82±0.31%; control: 1.07±0.13%; P=0.01). Maps for ventilation and gas uptake by tissue and blood were highly heterogeneous in the lungs of patients. Vdef% and RBC-to-tissue ratios in all 15 subjects correlated with corresponding global lung functional measures from PFT: FEV1/FVC (R=-0.91, P<0.001) and DLCO % predicted (R=0.54, P=0.03), respectively. The tissue-to-gas ratios correlated with tissue density (HU) measured by CT (R=0.88, P<0.001). CONCLUSION With the unique ability to provide detailed information about lung function including ventilation, tissue density, perfusion and gas exchange with 3D resolution, hyperpolarized xenon-129 dissolved-phase MRI has high potential to be used as an important reference for radiotherapy treatment planning and for evaluating the side effects of the treatment. Abstract:Autografts, allografts, and xenografts are the most widely used methods to enhance effective reconstruction of bone defects, but natural donor bone has some deficiencies, such as limited supply, excessive damage of transplant-supplied-parts, constrained growth and high complication rates.<a class="ejp-citation-link js-ejp-citation-link">[1]</a> Bioactive bone replacement materials provide an alternative for bone defect repair and regeneration. Augmented injectable bone scaffold material applied to bone replacement therapies in orthopedics and dentistry has generated much discussion in bone tissue engineering for bone defect repair. Injectable cement materials currently in clinical use and those under research can be classified into three main categories: polymethylmethacrylate (PMMA), calcium-phosphate-based bone cements (CPCs), and calcium-sulfate-based bone cements (CSCs). Of these, the chemical components of CPCs are most similar to the inorganic components of bones. CPCs mimic the mineral phase of bone to formulate a natural lattice for bone tissue to promote natural bone ingrowth and remodeling.<a class="ejp-citation-link js-ejp-citation-link">[2]</a> Furthermore, in comparison with the most widely used PMMA cements, CPCs have a lower exothermic reaction temperature and promote better osteointegration. For these reasons, they have become one of the most promising bone repair materials.<a class="ejp-citation-link js-ejp-citation-link">[3]</a> Nevertheless, CPCs have some limitations: (1) Their slow degradation rate and lack of macroporosity to promote the formation of new bone make their osteoinduction capacity insufficient for clinical needs.<a class="ejp-citation-link js-ejp-citation-link">[4]</a> Modification efforts include fabricating porous/nano structures and/or drug delivery CPC composites to promote bone growth; (2) Another drawback is that the mechanical strength of CPCs cannot match the strength of human cortical bone; thus, strength modification is another research direction.
