Abstract:The invention of the optical microscope is significant to the development of many aspects of science and technology,including novel materials,life sciences and medicine,for its superiorities in resolving fine structures.But,restrained by the Abbe diffraction limit,the best-resolving capability that can be obtained by conven-tional linear optical microscopy is～200 nm laterally and～500 nm axially using an objective lens(numerical aperture=1.4)at 550 nm wavelength.Over the past decades,to break the resolution limit and meet the increasing demand for super-resolution ability,numerous attempts were made,and fruitful developments emerged[1-8].Among all the approaches,one significant class is based on the integrated spatial frequency engineering to extend the collectable spatial frequency region via spatial frequency shift(SFS)method[3-13].Through the combination of SFS with spe-cially designed photonic chips,highly sophisticated optical sys-tems in traditional super-resolution instruments are moved out.
On-chip Microscopy Using Random Phase Mask Scheme

Abstract:In this study, a simple and novel phase-retrieval scheme is implemented using multi-angle illumination to enhance the resolution of lensless microscopy. A random-phase mask (from 0 to 2π) precedes the sample to encode the information at the sensor plane. The sample is illuminated with multiple angles that are symmetrical along the optical axis of the system. The system is initially calibrated while recording the images without any sample at the corresponding multi angles. The two types of image are mutually subtracted, and the resultant images are summed at the sensor plane and backpropagated to the sample plane. The final image is free of the twin-image effect, and has a high signal-to-noise ratio owing to the multi angles of the illumination scheme. This scheme gives a resolution of ~4 micron for a large field-of-view (~15 mm 2 ). The scheme is useful for robust imaging owing to the fast phase-retrieval method, and it enables a straightforward analytical reconstruction instead of using complicated iterative algorithms in a lensless microscopic setup.
Wide-field 3D Nanoscopy on Chip Through Large and Tunable Spatial-Frequency-shift Effect

Abstract:Linear super-resolution microscopy via synthesis aperture approach permits fast acquisition because of its wide-field implementations, however, it has been limited in resolution because a missing spatial-frequency band occurs when trying to use a shift magnitude surpassing the cutoff frequency of the detection system beyond a factor of two, which causes ghosting to appear. Here, we propose a method of chip-based 3D nanoscopy through large and tunable spatial-frequency-shift effect, capable of covering full extent of the spatial-frequency component within a wide passband. The missing of spatial-frequency can be effectively solved by developing a spatial-frequency-shift actively tuning approach through wave vector manipulation and operation of optical modes propagating along multiple azimuthal directions on a waveguide chip to interfere. In addition, the method includes a chip-based sectioning capability, which is enabled by saturated absorption of fluorophores. By introducing ultra-large propagation effective refractive index, nanoscale resolution is possible, without sacrificing the temporal resolution and the field-of-view. Imaging on GaP waveguide material demonstrates a lateral resolution of lamda/10, which is 5.4 folds above Abbe diffraction limit, and an axial resolution of lamda/19 using 0.9 NA detection objective. Simulation with an assumed propagation effective refractive index of 10 demonstrates a lateral resolution of lamda/22, in which the huge gap between the directly shifted and the zero-order components is completely filled to ensure the deep-subwavelength resolvability. It means that, a fast wide-field 3D deep-subdiffraction visualization could be realized using a standard microscope by adding a mass-producible and cost-effective spatial-frequency-shift illumination chip.

Abstract:In article number 1900126, Xu Liu, Qing Yang, and co-workers present a waveguide chip design that permits label-free superresolution imaging. The method captures all sample spatial frequencies over a large bandwidth, permitting artifact-free reconstruction. Two-dimensional sample structures are imaged without distortion and reconstructed over large field of views at sub-wavelength resolution. The results match well with data obtained via SEM and AFM imaging.

Abstract:Wide field-of-view (FOV), label-free, super-resolution imaging is demonstrated using a specially designed waveguide chip that can illuminate a sample with multicolor evanescent waves travelling along different directions. The method is enabled by a polymer fluorescent film that emits over a broad wavelength range. Its polygonal geometry ensures coverage over all illumination directions, enabling high-fidelity image reconstruction while minimizing distortion and image blurring. By frequency shifting and iterative stitching of different spatial frequencies in Fourier space, the reconstruction of 2D samples is achieved without distortion over wide FOVs. The fabrication process is facile and compatible with conventional semiconductor-fabrication methods. The super-resolution chip (SRC) can thus be produced with high yield, offering opportunities for potential conjunction of super-resolution techniques integrated optical circuits or for the development of single-use diagnostic kits.

Abstract:The resolution of lensless on-chip microscopy is mainly limited by the pixel size of the image sensor. Many superresolution techniques have emerged to solve the problem of insufficient imaging resolution. Most existing pixel super-resolution technologies rely on precise electric translation stage for hundreds of high-precision displacements, or on expensive tunable lasers to generate diffraction diversity. Conventional ptychography iterative engine (PIE) is considered an effective method for improving imaging resolution, but it is prone to oscillations in the early stage of iteration. In this paper, we propose a ptychography imaging technique based on scattering multiplexing, which involves coating the surface of the image sensor with a layer of polystyrene microspheres, and utilizing a 4 x 3 LED array to sequentially illuminate the sample. An innovative ptychography reconstruction algorithm based on dual amplitude gradient descent (DAGD) is designed for the reconstruction from the holograms, which effectively avoids the problems of slow convergence speed and obvious oscillation. Compared with other similar technologies, our system has no moving parts and uses inexpensive partially coherent light illumination. It only records 12 holograms and reaches the imaging resolution of 1.23 mu m, which is 1.36 times pixel super-resolution compared with the pixel size of the sensor.

Abstract:Achieving high spatial resolution over a wide field of view (FOV) is the goal of many imaging systems. In a traditional lens-based microscope, designing a complex objective with high numerical aperture (NA) to achieve this goal is a tough and challenging task. The lens-free wide-field imaging method based on phase retrieval provides a new way to bypass the trade-off between the spatial resolution and FOV of conventional microscopy. However, the typical lens-free microscopy usually requires mechanical devices with high precision and repeatability. In this paper, we report a robust and cost-effective pixel super-resolved lens-free imaging method based on dual laterally shifting modulation. A thin diffuser is inserted between the object and the image sensor to be used as the modulator. The diffuser and the object are transversely scanned at the same time to add diversities for phase retrieval and pixel super-resolution, respectively. In this way, the positional shifts of the diffuser and the object can be directly recovered with the registration algorithm, thus addressing the low stability and inaccuracy issues of translation stages. We also propose a pixel super-resolution phase-retrieval algorithm to recover the object and the unknown diffuser. We first use numerical simulations to evaluate the proposed scheme. Then, we validate this approach by imaging a resolution target and a pollen sample, thus achieving an FOV of similar to 30 mm(2) and a half-pitch resolution of 0.78 mu m, which surpasses 2.14 times the theoretical Nyquist-Shannon sampling resolution limit. The Dynamic Super-Resolution Phase Imaging Based on Low-Cost Lensfree System

Abstract:In this Letter, we present a new active micro-scanning setup and associated super-resolution (SR) phase reconstruction method in lensfree microscopy to achieve SR dynamic phase imaging. By rotating two orthogonal parallel plates to achieve controllable micro-scanning, a set of low-resolution (LR) intensity images with relative sub-pixel displacements can be acquired. These LR intensity images are then processed sequentially based on the matched imaging pixel transfer model and SR reconstruction algorithm to obtain the super-resolved quantitative phase images. The reconstructed result of Benchmark Quantitative Phase Microscopy Target (QPT (TM)) demonstrates the resolution enhancement quantitatively, which achieves a half-pitch lateral resolution of 775 nm over a large field-of-view (FOV) of similar to 29:84mm(2), surpassing 2.15 times of the theoretical NyquistShannon sampling resolution limit imposed by the pixel-size of the sensor (1.67 mu m). Investigations of unstained Hela cells are then presented, suggesting that the method developed can provide promising application in the dynamic study as well as morphological analysis of the subcellular features for unlabelled biological samples.

Abstract:Lens-free on-chip microscopy has many advantages compared with lens-based microscopy such as lightweight of hardware, low cost and improved resolution etc. A microscopy algorithm is proposed in this paper and its merits include high resolution, strong robustness, insensitivity to initial state of the object. Theories related to diffraction are introduced as the premise. 2D log-Gabor filters with different scales and orientations are constructed to extract the features of the captured photo. The reconstructed photo is determined by the extracted features and theoretical calculation. The algorithm is verified with 0.95×0.95 μm2 pixel size image sensor. The experimental results illustrate the effectiveness and high resolution of this algorithm.

Abstract:Lensless imaging systems have gained significant attention recently due to their advantages in terms of reduced size and weight compared to traditional lens-based systems. However, like other imaging methods, lensless imaging encounters challenges in resolving scenes with more details. In this article, we propose a novel four-frame super-resolution method specifically tailored for lensless imaging systems. Our approach shares similarities with previous lensless imaging systems, involving a sensor and a modulation device placed in front of the image sensor. We develop an explicit degradation downsampling model with sub-pixel shifts and provide the solution to corresponding inverse problem, may offering valuable guidance for other super-resolution imaging algorithms based on spatial displacements. By applying random lateral sub-pixel shifts, acquiring four low-resolution (LR) images, and fusing their spatial information, we achieve high-resolution (HR) sensor recordings, enabling super-resolution reconstruction of the imaging scene. Numerical simulations demonstrate approximately an improvement in spatial resolution compared to single-measurement methods. Furthermore, we evaluate the performance of our method across various lensless imaging systems utilizing different masks, validating its versatility and effectiveness in achieving higher resolution outcomes. Experimental results also support our proposed scheme's ability to achieve higher spatial resolution reconstruction in a real system.

Pixel super-resolved lensless on-chip sensor with scattering multiplexing

Abstract:Lensless on-chip microscopy has shown great potential for biomedical imaging due to its large-area and high-throughput imaging capabilities. By combining the pixel super-resolution (PSR) technique, it can improve the resolution beyond the limit of the imaging detector. However, existing PSR techniques are restricted to the feature size and crosstalk of modulation components (such as spatial light modulator), which cannot efficiently encode target information. Besides, the reconstruction algorithms suffer from the trade-off between image quality, reconstruction resolution and computational efficiency. In this work, we constructed a novel integrated lensless on-chip sensor via scattering multiplexing, and reported a robust PSR algorithm for sample reconstruction. The sensor employed a scattering layer as a modulator, which was permanently integrated with the detector. Benefiting from the high-degree-of-freedom reconstruction of the scattering layer, we realized fine wavefront modulation with a small feature size. The integration engineering avoided repetitious calibration and reduce the measurement complexity. The reported PSR algorithm combines both model-driven and data-driven strategies to efficiently exploit the high-frequency information from the fine modulation. A series of experiments validated that the reported sensor provides a low-cost solution for large-scale microscopic imaging, with significant advantages in resolution, image contrast and noise robustness.

Abstract:We present a wavelength-scanning-based lensfree microscopy that generates high-resolution reconstructions from undersampled raw measurements captured at multiple wavelengths.The reconstruction result of the standard 1951 USAF achieves a half-pitch lateral resolution of 775 nm, corresponding to a numerical aperture of ∼ 1.0, across a large field of view (∼ 29.85 mm2). Compared with other super-resolution methods such as lateral or axial shift-based device and illumination source rotation design, wavelength scanning avoids the need for shifting/rotating mechanical components. This wavelength-scanning super-resolution method would benefit the research and development of more stable lensfree microscopy system.

Abstract:We present a data-efficient framework to achieve a super-resolved reconstruction in lensfree on-chip microscopy. Instead of using conventional 3D-stack holograms, our method accomplishes a 5 x reduction of data size without the loss of the imaging resolution, in which merely 30 raw holograms are recorded. The experiment of the resolution target demonstrates a half-pitch lateral resolution of 780 nm across a large field-of-view (FOV) of 28.6 mm(2). For the view of H&E stained intestine pathology slide, our method shows an equivalent resolution but a 20-fold expansion of FOV in comparison with a 10 x commercial wide-field microscope. This work will be a useful tool for the miniaturization of whole slide pathological microscopy.

Abstract:We report an ingenious method of super-resolution optical microscopy utilizing scannable cantilever-combined microsphere. By scanning the microsphere over the sample surface in a cantilever-combined microsphere-sample contact state, super-resolution images can be acquired at arbitrary sample regions through near-field information collection by the microsphere. In addition, such a state can effectively reduce the possibility of breaking the cantilever and damaging the microsphere or sample surface. This work has developed a new method and technique of sub-diffraction-limit optical microscopy, and can be practically applied in various fields of micro/nanoscopy.

The reconstruction result of the benchmark quantitative phase microscopy target (QPTTM) demonstrates a half-pitch lateral resolution of 775 nm across a large field-of-view of ∼29.84 mm2, surpassing 2.15 times that of the theoretical Nyquist-Shannon sampling resolution limit imposed by the pixel size of the imaging sensor (1.67 μm). The proposed approach is also evaluated by imaging unstained HeLa cells, suggesting it is a promising toolset for high-throughput monitoring and quantitative analysis of unlabeled biological samples.

Abstract:The microlens-assisted microscope realizes super-resolution imaging and observation, and has the advantages of no marking, no damage, real-time, localization, and good environmental compatibility. Liquid microlens arrays with uniformity and easy manipulation can realize super-resolution imaging without complicated mechanical scanning and driving. However, simply and efficiently controlling the imaging distance is a key technical challenge to the realization of super-resolution imaging of microlens. In this paper, the uniform depths of photoresist microholes on light disk are fabricated by ultraviolet exposure technology. Using liquid self-assembly technology, the microholes are filled with glycerol droplets, and thus ensuring the near-field imaging distance of the microlens. The reconfigurable super-resolution of 226-nm-wide grating line and the imaging magnification of 1.59 times are observed under the optical microscope. At present, the theory of super-resolution imaging based on microlens is not unified and perfect. In this paper, the Abbe imaging principle is used to explain the imaging magnification and super-resolution characteristics. Therefore, the liquid microlens arrays integrated on the light disk show great potential application in optical nanometer measurements and sensing devices.

Abstract:Lensless imaging is a technique that records diffraction patterns without using lenses and recovers the complex field of object via phase retrieval. Robust lensless phase retrieval process usually requires multiple measurements with defocus variation, transverse translation or angle-varied illumination. However, making such diverse measurements is time-consuming and limits the application of lensless setup for dynamic samples. In this paper, we propose a single-shot lensless imaging scheme via simultaneous multi-angle LED illumination. Diffraction patterns under multi-angle lights are recorded by different areas of the sensor within a single shot. An optimization algorithm is applied to utilize the single-shot measurement and retrieve the aliasing information for reconstruction. We first use numerical simulations to evaluate the proposed scheme quantitatively by comparisons with the multi-acquisition case. Then a proof-of-concept lensless setup is built to validate the method by imaging a resolution chart and biological samples, achieving ∼ 4.92 μm half-pitch resolution and ∼ 1.202mm2 field of view (FOV). We also discuss different design tradeoffs and present a 4-frame acquisition scheme (with ∼ 3.48 μm half-pitch resolution and ∼ 2.35 × 2.55 mm2 FOV) to show the flexibility of performance enhancement by capturing more measurements.

The reconstruction result of the Benchmark Quantitative Phase Microscopy Target (QPTTM) demonstrates the resolution enhancement quantitatively, which achieves a half-pitch lateral resolution of 691 nm across a large field of view (~29.85 mm2), surpassing 2.41 times of the theoretical NyquistShannon sampling resolution limit imposed by the pixel-size of the sensor (1.67 µm). Compared with other superresolution methods such as lateral or axial shift-based device and illumination source rotation design, wavelength multiplexed avoids the need for shifting/rotating mechanical components. This multi-wavelength multiplexed super-resolution method would benet the research and development of a more stable lensless microscopy system.
