Urinary concentrations of parabens and their association with demographic factors: A population-based cross-sectional study

Abstract:Parabens are broad-spectrum antimicrobial agents used in a range of consumer products, including personal care products, cosmetics, and food. Recently, the widespread use of parabens has raised concerns about the potential health risks associated with their endocrine-disrupting effect. In the present study, 2541 urine samples were collected and analyzed by liquid chromatography-mass spectrometry for the determination of the concentrations of methyl paraben (MeP), ethyl paraben (EtP), propyl paraben (PrP) and butyl paraben (BuP). The detection rate and geometric mean concentrations of parabens in the general population followed the order MeP (97.7%; 116ng/mL)>EtP (97.2%; 24.7ng/mL)>PrP (96.7%; 11.0ng/mL)>BuP (83.5%; 1.13ng/mL). The composition profiles showed that MeP and EtP accounted for >90% of the urinary paraben concentration. We performed statistical analysis in order to evaluate differences between demographic variables and urinary concentrations. Our results showed that adjusted proportional change of MeP, PrP, and BuP in adults were 2.67-6.13 times higher in females than in males. The urinary concentrations of PrP in adults increased significantly with age. The adjusted proportional changes of MeP and PrP in adults were associated with increased body mass index (BMI). The adjusted proportional changes of BuP and PrP in children and adolescents were 1.44 and 1.69 times higher in females than in males. However, there was no clear association between paraben concentrations and demographic variables in the children and adolescents groups. The estimated daily intake (EDIurine) of MeP and EtP in adults were 301μg/kg bw/day, which is lower than the acceptable daily intake (ADI; 10mg/kg bw/day). In summary, our results revealed that the general population in Korea was exposed to parabens during 2009-2010, and most Koreans are exposed to parabens. The urinary levels of parabens varied by age group with demographic factors in the Korean population. The results of study may be used to establish a nationally representative baseline of exposure to parabens in risk assessment.
Parabens As Chemicals of Emerging Concern in the Environment and Humans: A Review

Abstract:Parabens are one of the most widely used preservatives in food, pharmaceuticals and personal care products (PCPs) because of their advantageous properties and low toxicity based on the early assessments. However, recent research indicates that parabens may act as endocrine-disrupting chemicals (EDCs) and thus, are considered as chemicals of emerging concern that have adverse human health effects. To provide the basis for future human health studies, we reviewed relevant literature, published between 2005 and 2020, regarding the levels of parabens in the consumer products (pharmaceuticals, PCPs and food), environmental matrices and humans, including susceptible populations, such as pregnant women and children. The analysis showed that paraben detection rates in consumer products, environmental compartments and human populations are high, while the levels vary greatly by country and paraben type. The concentrations of parabens reported in pregnant women (~20–120 μg/L) were an order of magnitude higher than in the general population. Paraben concentrations in food and pharmaceuticals were at the ng/g level, while the levels in PCPs reached mg/g levels. Environmental concentrations ranged from ng/L–μg/L in surface waters to tens of μg/g in wastewater and indoor dust. The levels of human exposure to parabens appear to be higher in the U.S. and EU countries than in China and India, which may change with the increasing production of parabens in the latter countries. The review provides context for future studies to connect paraben exposure levels with human health effects.

Abstract:Parabens, known as ubiquitous preservatives, have been linked to adverse health outcomes in humans. This study aimed to examine urinary paraben concentrations of children at 3 years of age and evaluate their associations with anthropometric parameters. Urinary parabens including methylparaben (MeP), ethylparaben (EtP), propylparaben (PrP), butylparaben (BuP) and benzylparaben (BeP) were measured among 436 children in a birth cohort using gas chromatography with tandem mass spectrometry. Generalized linear models were performed to evaluate associations of paraben exposures with age- and sex-specific z scores, including weight, height, weight for height and body mass index. MeP, EtP and PrP were the dominant parabens in urinary samples, with the median concentrations of 6.03 μg/L, 3.17 μg/L, 2.40 μg/L, respectively. The median values of estimated daily intake (EDIurine) of five urinary paraben concentrations were 12.10, 5.68, 4.50, 0.06 and 0.17 μg/kg-body weight/day, respectively. Urinary EtP concentrations were positively associated with weight z scores [regression coefficient β = 0.16, 95% confidence interval (CI): 0.04, 0.29; p = 0.01] and height z scores (β = 0.15, 95% CI: 0.03, 0.27; p = 0.01). Positive associations were found between the sum of molar concentrations of five parabens and height z scores among all children (β = 0.24, 95% CI: 0.04, 0.45; p = 0.02). These significant associations were only observed in boys. Our findings suggest that exposure to parabens may be adversely associated with physical growth in 3-year-old boy children. Further prospective studies are warranted to understand the toxicological mechanisms of paraben exposures and potential risk of children.

Abstract:OBJECTIVE To evaluate the veracity of the C1 lateral mass screw insertion in the atlantoaxial fixation using plate and screw method without A-P fluoroscopic guiding. METHODS In the atlantoaxial fixation, without A-P fluoroscopic guiding, we probe lateral mass edge and identify the entrance point of C1 lateral mass screw indirectly. 159 patients treated with atlantoaxial fixation using plate and screw method were studied. Postoperative CT was used for analysis, and the position of the lateral mass screws in atlas was identified by coronary and axial CT scan. Three areas were delimited in and around the lateral mass of atlas in the axial CT scan, so as to analyze the location of screws: area A (inside the joint face), area B (outside the joint face but still in lateral mass), and area C (outside the lateral mass). RESULTS Among the 318 screws, 308 (96.9%) were located in area A, 5 in area B (1.6%), and 5 in area C (1.6%). All cases got atlantoaxial union at 4 months after operation. CONCLUSION Probing lateral mass edge and identifying the lateral mass indirectly is reliable for identifying the path of screw in atlas.

Abstract:The usage of parabens, bisphenol A and triclosan in diverse consumer products is in widespread. Nevertheless, there are limited data concerning exposure to these chemicals in human being, especially in primiparas. Biomonitoring of chemicals in primiparas is useful for the estimation of chemical exposure risks for both primiparas and their offspring. This study aims to investigate urinary levels of parabens, bisphenol A and triclosan of 84 primiparas from Shenzhen, China and to evaluate their potential health risks. Methyl, ethyl, and n-propyl parabens bisphenol A and triclosan exhibited high detection rates (DRs) (> 97%) in urine samples, suggesting that primiparas are exposed to them widely. The median concentrations of methyl, ethyl, and n-propyl parabens, bispenol A and triclosan in urine were 2.14, 4.10, 0.46, 1.30 and 3.00 µg/L, respectively. Ethyl paraben was the predominant paraben accounting for nearly half of Σ3parabens (The sum concentrations of methyl, ethyl, n-propyl parabens). Positive associations with significance (p < 0.05) were found between the usage of plastic containers and urinary concentrations of ethyl paraben or BPA, indicating plastic containers might be an important factor influencing primipara exposure to these two chemicals. Urinary concentrations of methyl paraben were positively associated (p < 0.05) with the time of computer use by participant, suggesting that indoor dust might constitute an important source of parabens. The estimated daily intakes of parabens, bisphenol A and triclosan contrasted with the acceptable daily intakes in a comparatively low level. The hazard quotients (HQs) of these chemicals were all less than 1, suggesting no health risks for primiparas from South China.

Abstract:The adverse effects of parabens raise concerns about their extensive use as preservatives in consumer products, especially in cosmetics. Until now, their distribution and excretion in humans have attracted little attention. Here, we quantified various agents including, for the first time, methyl-; ethyl-; n-propyl-; n-butyl-, and i-butylparaben (MeP, EtP, PrP, n-BuP, i-BuP); methyl- and ethyl-protocatechuate (OH-MeP and OH-EtP); hydroxybenzoic acid (4-HB); and 3,4-dihydroxybenzoic acid (3,4-DHB) in urine, serum, and seminal plasma samples from 50 healthy Chinese men in Beijing, China. Urine paraben concentrations were 1–2 orders of magnitudes higher than those in serum and seminal plasma. MeP and PrP were predominant and correlated with each other in the urine, serum, and seminal plasma. In urine, we observed a significant correlation between MeP and OH-MeP; EtP and OH-EtP; and 4-HB and 3,4-DHB concentrations. All these results provide new information on parabens as biomarkers for the assessment of exposure.Graphical abstract

Abstract:Parabens are added into foodstuffs, pharmaceuticals and personal care products (PCPs) as additives extensively due to their excellent antiseptic and antibacterial effects. In the past decade, parabens have raised great concerns on their potential harm to humans. Existing studies have suggested positive correlations between PCP application and urinary paraben concentrations in females, but little is known about paraben exposure levels and health risks arising from PCP use. In this study, 150 PCP samples covering eleven categories were collected from South China and measured for the concentrations of five parabens, including methyl paraben (MeP), ethyl paraben (EtP), propyl paraben (PrP), butyl paraben (BuP) and benzyl paraben (BeP). Parabens were widely detected in PCPs, with a detection frequency of 100%, 99.3%, 80.0%, 74.0% and 13.3%, for MeP, EtP, PrP, BuP and BeP, respectively. The median concentration of Σ5parabens was 126 μg/g with a range of 6.38-424 μg/g across all PCP samples. The contents of MeP, EtP and PrP measured in leave-on PCPs were obviously higher than those in the rinse-off ones (p < 0.05). MeP and PrP were the main paraben analogues, together accounting for 93.6% of Σ5parabens in all PCPs. The daily intakes of parabens through dermal absorption by Chinese adult females estimated by measurements obtained in the present study were 0.15 and 83.2 μg/kg-bw/day on basis of the application of rinse-off and leave-on PCPs, respectively. Among the eleven categories, sunscreen, body lotion and mask constituted the main exposure sources of parabens to females. The hazard quotients of parabens were far less than 1, indicating no considerable health risk for Chinese adult females.

Abstract:Parabens are widely used as preservatives in personal care products, foodstuffs, and pharmaceuticals. Concerns have been raised regarding the potential endocrine disruption effects of parabens. In the present study, the urinary concentration of four common parabens, including methylparaben (MP), ethylparaben (EP), propylparaben (PP), and butylparaben (BP), in 100 Iranian adolescents randomly referring to health services centres were analyzed using GC/MS. The association of sociodemographic and lifestyle variables, collected through questionnaire, with the concentration of parabens also were studied. Median concentrations of MP, EP, PP, and BP were 92.21, 8.46, 12.26, and 8.42 µg/g creatinine, respectively. There was a strong positive significant correlation between MP and PP (r = 0.694) and moderate to a weak correlation between the other parabens. The concentration of urinary MP in females was significantly higher than those in male (p = 0.021). There was a significant negative association between different BMI groups and MP and EP. There also was a positive significant association between the MP and age, and between MP, EP, and PP, and tobacco use. Although the estimated daily intake of the parabens was lower than the Acceptable Daily Intake, it was higher than those reported in other countries. This confirms the widespread exposure of Iranian adolescents to the paraben compounds and their association with sociodemographic factors. This was the first study reporting the urinary parabens level in Iranian adolescents, and the data can be used as a basis for assessing the risk of exposure to parabens in the Iranian population in future studies.

Abstract:Parabens, alkyl esters of p-hydroxybenzoic acid, are widely used in cosmetics, pharmaceuticals, personal care products and as food additives to inhibit microbial growth and extend product shelf life. Consumers of these compounds are frequently exposed via the skin, lips, eyes, oral mucosa, nails, and hair. Parabens are estrogenic molecules but exert weaker activity than natural estrogens, which would imply a low risk. Consistent with this idea, a number of recent commission reports from different countries suggested that parabens pose a negligible endocrine-disrupting risk at the recommended doses. However, individuals are not routinely exposed to a single paraben, and most of the available paraben toxicity data, reviewed in these reports, are from single-exposure studies. Further, assessing the additive and cumulative risk of multiple paraben exposure from daily use of multiple cosmetic and/or personal care products is presently not possible based on current studies. In this review, current and recent studies of paraben exposure and public health policies as well as critical gaps in the knowledge are discussed and new research directions regarding multiple exposures and novel target cohorts are recommended.

Abstract:Previous studies implied that elevated exposure to parabens may result in increased oxidative stress. However, the association between exposure to paraben derivatives and oxidative stress biomarkers in children has been rarely studied. This study examined the associations between exposure to paraben derivatives and oxidative stress biomarkers in Chinese children. Nine targeted compounds of parabens and their derivatives including methyl paraben (MeP), ethyl paraben (EtP), propyl paraben (PrP), butyl paraben (BuP), p-hydrox4ybenzoic acid (p-HB), 3,4-dihydroxy benzoic acid (3,4-DHB), benzoic acid, methyl 3,4-dihydroxybenzoate (rOH-MeP), and ethyl 3,4-dihydroxybenzoate (rOH-EtP) were detected in urine collected from 139 children from South and Central China. Additionally, 8-hydroxy-2'-deoxyguanosine (8-OHdG), 8-hydroxyguanosine (8-OHG), and 4-hydroxy-2-nonenal mercapturic acid (HNE-MA) were measured as oxidative stress biomarkers. All targeted compounds (except for BuP) were frequently detected in urine (detection frequencies ranged 80.8%-100%). Linear mixed effects model revealed that all targeted compounds (with detection frequencies >50%), except for EtP, were significantly associated with an increase in 8-OHdG. rOH-EtP was found to be significantly associated with 8-OHG (β = 0.12; 95% confidence interval [95% CI]: 0.08, 0.16) positively. In addition, PrP and benzoic acid were associated with elevated levels of HNE-MA. Weighted quantile sum regression revealed that co-exposure to the targeted compounds was positively associated with 8-OHdG (β = 0.17; 95% CI: 0.12, 0.22), 8-OHG (β = 0.14; 95% CI: 0.10, 0.18), and HNE-MA (β = 0.43; 95% CI: 0.27, 0.59); rOH-EtP and benzoic acid were the major contributors for the combined effects on oxidative stress of nucleic acids and lipid, respectively. Our findings provide new evidence for the effects of exposure to paraben derivatives on nucleic acid oxidative damage and lipid peroxidation in children.

Abstract:Parabens, a group of endocrine disrupting chemicals (EDCs), are well known preservatives in pharmaceuticals and personal care products (PPCPs). However, studies on parabens exposure and their cumulative effects in Asian population are limited. This study aimed to identify the exposure characteristics and estimate the cumulative risk of four parabens in the general Taiwanese. We have collected urine samples including 271 adults (18-97 yrs old) and 95 minors (7-17 yrs old), from Taiwan Environmental Survey for Toxicants 2013, and analyzed for four urinary parabens including methyl (MeP)-, ethyl (EtP)-, propyl (PrP)-, and butylparaben (BuP) by using ultraperformance liquid chromatography-tandem mass spectrometry. The health-based guidance value (HBGV) and the antiandrogenic properties of parabens were used to calculate the hazard index (HI) for cumulative risk. MeP and PrP were most abundant compounds and startlingly higher than those in other countries. Adults had a higher geometric mean level of four parabens than minors (adults: MeP, 381.7; PrP, 108.6; EtP, 39.6 and BuP 6.3 ng/mL; minors: MeP, 65.7; PrP, 7.9, EtP, 2.6 and BuP 2.2 ng/mL). Participants who used a higher number of personal care products had a significantly higher risk with higher concentrations of PrP (above 75th %tile) [adjusted odds ratio (aOR): 1.79, 95 % CI: 1.01-3.15] and BuP [aOR: 1.78, 95 % CI: 1.03-3.07]. The median and 95th %tile HI (the sum of the HQs of each paraben) was as 1.10 and 4.39-fold higher than acceptable cumulative threshold (HI <1) and PrP accounted for 90 % of the HI. Our results indicate omnipresent exposure to parabens among the Taiwanese population, which might cause certain level of concerns. These significant increasing trends of HI with age dependence were observed, which mainly driven by PPCPS used. Routine survey of parabens in PPCPs and continued biomonitoring needs to be urgently addressed.

Abstract:Parabens are widely used as preservatives in food, pharmaceuticals, and cosmetics due to their excellent antimicrobial activities, cost-effectiveness, and stability. Previous studies have demonstrated their harmful potential and ubiquity in the environment and human tissues. This study revealed profiles of parabens and their metabolites in urine samples from a general population of different ages in China using non-target screening. Metabolism of parabens in human bodies was further explored through the identified metabolites in combination of molecular networking. A total of 34 paraben compounds were screened in the urine samples. In addition to 3 identified confidence level 1 (CL1) parent parabens, 3 CL2 compounds were also identified, namely 4-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, and ethylparaben sulfate. Furthermore, 6 CL3 compounds were tentatively identified, five of which were sulfonated and sulfated metabolites of parabens. The remaining 22 were CL4 features without certain chemical structures. Hazardousness assessment suggested toxic potential of the identified metabolites. Distribution of the parabens and metabolites in the urines showed age-dependent differences. Sulfonation and sulfation were potentially significant metabolic pathways of the parabens in human bodies. This study provides a new insight into understanding metabolism of parabens in human bodies and potential risks of human exposure to parabens.

Abstract:Cosmetics are a source of lifetime exposure to various substances including parabens, being the most popular synthetic preservatives. Because the use of cosmetics shows an increasing trend and some adverse health outcomes of parabens present in these products have been reported, the present review focused on the safety of dermal application of these compounds. Special attention has been paid to the absorption of parabens and their retention in the human body in the intact form, as well as to their toxicological characteristics. Particular emphasis has been placed on the estrogenic potential of parabens. Based on the available published data of the concentrations of parabens in various kinds of cosmetics, the average ranges of systemic exposure dose (SED) for methylparaben, ethylparaben, propylparaben, and butylparaben have been calculated. Safety evaluations [margin of safety (MoS)] for these compounds, based on their aggregate exposure, have also been performed. Moreover, evidence for the negative impact of methylparaben on skin cells has been provided, and the main factors that may intensify dermal absorption of parabens and their impact on the skin have been described. Summarizing, the use of single cosmetics containing parabens should not pose a hazard for human health; however, using excessive quantities of cosmetic preparations containing these compounds may lead to the development of unfavorable health outcomes. Due to the real risk of estrogenic effects, as a result of exposure to parabens in cosmetics, simultaneous use of many cosmetic products containing these preservatives should be avoided.

Abstract:The present study assessed the exposure to methylparaben (MP) and propylparaben (PP) from antiperspirants in serum of 24 women aged 20-30 years old and an in vitro skin assay. An effective liquid chromatography-tandem mass spectrometry method for the determination of MP and PP levels in serum was developed and validated in the range of 10-100 μg/L; the method was fast, simple, sensitive, linear, precise, and accurate. In addition, a simple and rapid liquid chromatography-ultraviolet detection method for the determination of MP and PP levels in antiperspirants was developed and validated in the range of 2-26 mg/L, which presented satisfactory linearity, precision, and accuracy. Using these two methods, 20 commercial antiperspirants were evaluated, and only three showed MP and PP in the formulation. The antiperspirant containing 0.2% and 0.1% w/w MP and PP, respectively, was given to the volunteers, to estimate the internal dose, and submitted to a pig ear skin permeation assay in Franz diffusion cells, presenting a permeation flux of 32% for MP and 71% for PP. In this assay, both MP and PP permeated the skin; however, there was no correlation between antiperspirant use and paraben serum concentration in the volunteers. Graphical abstract.

Abstract:Parabens are widely used in the food, cosmetics and pharmaceutical industry and are widespread in the environment. As endocrine disruptors, parabens have adverse effects on living organisms. However, knowledge of the exposure of domestic animals to parabens is extremely scarce. Therefore, this study assessed the exposure level of dogs to three parabens commonly used in industry (i.e. methylparaben - MeP, ethylparaben - EtP and propylparaben - PrP) using fur sample analysis in liquid chromatography-tandem mass spectrometry. The presence of parabens has been noted in the samples collected from all dogs included in the study (n = 30). Mean concentrations of MeP, EtP and PrP in the fur of dogs were 176 (relative standard deviation - RSD = 127.48%) ng/g dry weight (dw), 48.4 (RSD = 163.64%) ng/g dw and 79.8 ng/g dw (RSD = 151.89%), respectively. The highest concentrations were found for MeP (up to 1023 ng/g dw). Concentrations of MeP and EtP in males were statistically higher than those in females (p < 0.05). Statistically significantly higher concentration levels of PrP in young animals (up to three years old) were also found. This is the first study concerning the use of fur samples to evaluate the exposure of domestic animals to parabens. The results indicate that an analysis of the fur may be a useful tool of paraben biomonitoring in dogs. The presence of parabens in the canine fur also suggests that these substances may play a role in veterinary toxicology. However, many aspects connected with this issue are not clear and require further study.
Exposure to parabens and associations with oxidative stress in adults from South China

Abstract:Parabens are widely applied as preservatives in cosmetics, drugs and food. Previous studies suggested that parabens could exhibit potential risks to human health. However, data on human exposure levels and health effects of parabens remain limited, especially in the potential effects on DNA oxidative stress. This study aimed to investigate urinary levels of parabens in adults from South China and explore the relationships between urinary parabens and DNA oxidative stress. Five short chain parabens, including methyl paraben (MeP), ethyl paraben (EtP), n-propyl paraben (PrP), butyl paraben (BuP) and benzyl paraben (BzP), were determined in urine from 319 adults in Shenzhen, China. MeP, EtP and PrP were frequently detected in urine samples (detection frequencies >66.5%), suggesting broad exposure in South China adults. Median concentrations of MeP, EtP, PrP, BuP and BzP were 5.78, 0.39, 0.35, 0.01 and 0.02 μg/L, respectively. A significantly positive correlation was observed between the urinary concentrations of MeP and PrP (p < 0.01), suggesting similar sources for these two chemicals. In addition, participants with alcohol consumption exhibited significantly lower paraben concentrations in urine than those without alcohol drinking (p < 0.05). Significant association was observed between urinary concentrations of parabens and 8-hydroxy-2′-deoxyguanosine (8-OHdG) levels (p < 0.01), while no significant dose-response relationship was found (p > 0.05). A potential risk from PrP exposure was found in South China adults.
