Abstract:Anthracnose of chili (Capsicum spp.) causes major production losses throughout Asia where chili plants are grown. A total of 260 Colletotrichum isolates, associated with necrotic lesions of chili leaves and fruit were collected from chili producing areas of Indonesia, Malaysia, Sri Lanka, Thailand and Taiwan. Colletotrichum truncatum was the most commonly isolated species from infected chili fruit and was readily identified by its falcate spores and abundant setae in the necrotic lesions. The other isolates consisted of straight conidia (cylindrical and fusiform) which were difficult to differentiate to species based on morphological characters. Taxonomic analysis of these straight conidia isolates based on multi-gene phylogenetic analyses (ITS, gapdh, chs-1, act, tub2, his3, ApMat, gs) revealed a further seven known Colletotrichum species, C. endophyticum, C. fructicola, C. karsti, C. plurivorum, C. scovillei, C. siamense and C. tropicale. In addition, three novel species are also described as C. javanense, C. makassarense and C. tainanense, associated with anthracnose of chili fruit in West Java (Indonesia); Makassar, South Sulawesi (Indonesia); and Tainan (Taiwan), respectively. Colletotrichum siamense is reported for the first time causing anthracnose of Capsicum annuum in Indonesia and Sri Lanka. This is also the first report of C. fructicola causing anthracnose of chili in Taiwan and Thailand and C. plurivorum in Malaysia and Thailand. Of the species with straight conidia, C. scovillei (acutatum complex), was the most prevalent throughout the surveyed countries, except for Sri Lanka from where this species was not isolated. Colletotrichum siamense (gloeosporioides complex) was also common in Indonesia, Sri Lanka and Thailand. Pathogenicity tests on chili fruit showed that C. javanense and C. scovillei were highly aggressive, especially when inoculated on non-wounded fruit, compared to all other species. The existence of new, highly aggressive exotic species, such as C. javanense, poses a biosecurity risk to production in countries which do not have adequate quarantine regulations to restrict the entry of exotic pathogens.

Abstract:Thirty-four isolates of Colletotrichum spp. including 2 species, C. gloeosporioides and C. capsici, from anthracnose on Bell pepper, Long cayenne pepper and Bird's eye chilli were isolated and their pathogenicity was proven via fruit inoculation. Pathogenicity tests divided pathogenic potential into low, medium and high virulence groups. It is clearly revealed that C. capsici from the three tested hosts expressed the highest virulent isolates. Cross-inoculation of three high virulent isolates of C. capsici in accordance with three chilli varieties showed that all isolates could produce anthracnose symptom in the same lesions. All tested isolates developed lesions after co-inoculation of all hosts. Inter simple sequence repeat (ISSR) analysis indicated that there are two distinct groups of C. gloeosporioides and C. capsici. Furthermore, genetic diversity was correlated with geographic distribution, while there was no clear relationship between genetic diversity and pathogenic variability. But it is clearly demonstrated that whereas C. gloeosporioides appears in the same geographic area as C. capsici, it causes lower disease incidence.

Abstract:Strawberry anthracnose, caused by Colletotrichum spp., is a major disease of cultivated strawberry. This study identifies 31 isolates of Colletotrichum spp. which cause strawberry anthracnose in Zhejiang Province and Shanghai City, China. Eleven isolates were identified as C. acutatum, 10 as C. gloeosporioides and 10 as C. fragariae based on morphological characteristics, phylogenetic and sequence analyses. Species-specific polymerase chain reaction (PCR) and enzyme digestion further confirmed the identification of the Colletotrichum spp., demonstrating that these three species are currently the causal agents of strawberry anthracnose in the studied regions. Based on analysis of rDNA internal transcribed spacers (ITS) sequences, sequences of all C. acutatum were identical, and little genetic variability was observed between C. fragariae and C. gloeosporioides. However, the conservative nature of the MvnI specific site from isolates of C. gloeosporioides was confirmed, and this site could be used to differentiate C. gloeosporioides from C. fragariae.

Abstract:Colletotrichum is an important plant pathogenic fungi that causes anthracnose/-twister disease in onion. This disease was prevalent in the monsoon season from August to November months and the symptoms were observed in most of the fields. This study aimed to investigate the pathogenicity and cumulative effect, if any of Colletotrichum gloeosporioides and Fusarium acutatum . The pot experiment was laid out to identify the cause responsible for inciting anthracnose-twister disease, whether the Colletotrichum or Fusarium or both, or the interaction of pathogens and GA3. The results of the pathogenicity test confirmed that C. gloeosporioides and F. acutatum are both pathogenic. C. gloeosporioides caused twisting symptoms independently, while F.acutatum independently caused only neck elongation. The independent application of GA3 did not produce any symptoms, however, increased the plant height. The combined treatment of C. gloeosporioides and F. acutatum caused twisting, which enhanced upon interaction with GA3 application giving synergistic effect. The acervuli were found in lesions infected with C. gloeosporioides after 8 days of inoculation on the neck and leaf blades. Symptoms were not observed in untreated control plants. Koch's postulates were confirmed by reisolating the same pathogens from the infected plants.

Abstract:A total of nine fungal species were isolated from the anthracnose infected chilli fruits. The fungi associated with infected chilli fruits were Alternaria alternata (Fr.) Keissler, A. tenuissima (Kunze ex Pers.), Aspergillus flavus Link, A. niger van Tieghhm, Colletotrichum dematium (Pers.) ex Fr., C. gloeosporioides (Penz.) Sacc., Curvularia clavata Wakker, Fusarium semitectum Berk. & Rav. and Rhizopus stolonifer (Ehrenb.) Vull. Among the isolated fungi, Colletotrichum dematium (49.03), C. gloeosporoides (18.06) and Fusarium semitectum (20.12) showed the higher per cent frequency. Abstract:Chilli is one of the most significant spice crops cultivated with numerous culinary, medicinal, and industrial applications. Colletotrichum have a wide host range causing anthracnose disease in various crops whereas in chili anthracnose caused by Colletotrichum capsici is the major constraint in chili production. Under severe environmental conditions, the symptoms appear all over the plant including fruit which lead to complete crop damage. The particular disease has a huge economic impact on yield reduction and also debars the export criteria due to the less quality of chili crop. Management of the disease is difficult under field conditions and majority of the management strategies are highly dependent on the use of chemicals. Solely rely on chemicals is harmful to environmental and human health so integrated disease management approach can help to manage the disease at the initial stage. Detection and identification of pathogen play a major role in pathogen management. Early identification and detection of the pathogen based on conventional and molecular methods are very important to reduce yield loss under field conditions. The current review consists of a detailed understanding of the pathogen, identification, detection and diagnosis, and the most recent research updates.

Abstract:The increased phosphorylation and activity of protein kinase B (PKB/Akt) was found early upon butyrate treatment of HT-29 cells with a potent differentiating agent, sodium butyrate. It was accompanied by the increased phosphorylation of glycogen synthase kinase-3 (GSK-3) and the inhibition of the activity of GSK-3beta to catalyze phosphorylation of its substrate, translation initiation factor eIF2B. Phosphorylation of PKB and GSK-3 in HT-29 cells was reduced by wortmannin, the inhibitor of phosphatidylinositol-3' kinase (PI3'-kinase), which is upstream activator of PKB and GSK-3 in the intracellular signalling. Modulation of the activity and phosphorylation of these protein kinases during transient in vitro differentiation of HT-29 cells indicates that control of the PI3'-kinase/PKB-dependent signalling pathway may be implicated very early in the changes of malignant phenotype of HT-29 cells.

Abstract:Anthracnose fruit rot caused by Colletotrichum spp. is a serious production constraint causing severe marketable yield loss in chilli. Field evaluation of chilli accessions for resistance to Colletotrichum spp. depends on various factors affecting disease expression such as edaphic conditions, temperature, rainfall, humidity and other variables that are difficult to control, therefore considered less accurate. Also, high chances of cross contamination with different Colletotrichum species leads to inconclusive assays for specific pathogen species and isolate. To identify a stable and reliable screening method, various chilli accessions were subjected to in vitro pin-prick and non-wounding spray methods using a specific pathogen isolates. When chilli accessions were screened against C. gloeosporioides isolate ‘IHRCg-1’, the in vitro pin- prick method showed positive correlation with the non- wounding spray method, except in the accession PBC80. The change in bioassay influenced the disease reaction pattern in the accession PBC 80, probably the pin pricks break the basal cuticle defense mechanism that was retained in spray inoculation method indicating varied resistance pattern. However, in the accession PBC 81 stable resistance pattern was observed against isolates of both species viz., C. truncatum ‘IIHR Ct-1’ and C. gloeosporioides ‘IIHR Cg-1’ and in the accession PBC 80 against C. truncatum ‘IIHR Ct-1’ in both the inoculation methods that depicted the expression of resistance genes during both methods of inoculation. Based on disease development pattern, the red ripe chilli expressed a variant reaction to infection by C. truncatum and C. gloeosporioides. The peak anthracnose infection at 10 DAI and 14 DAI is an accurate duration to record ‘IIHR Cg-1’ and ‘IIHR Ct-1’ infection, respectively on chilli ripe fruit for assaying the resistance.
Systemic Resistance in Chilli Pepper against Anthracnose (Caused by Colletotrichum truncatum) Induced by Trichoderma harzianum, Trichoderma asperellum and Paenibacillus dendritiformis

Abstract:In the present study, Paenibacillus dendritiformis, Trichoderma harzianum, and Trichoderma asperellum were appraised as potential biocontrol agents that induce resistance in chilli (Capsicum annuum) against the devastating pathogen Colletotrichum truncatum, which causes anthracnose. Bright-field and scanning electron micrographs showed the hyphal degradation, lysis, and abnormal swelling in C. truncatum against P. dendritiformis in a dual plate assay. Under greenhouse conditions, chilli seeds pretreated with P. dendritiformis, T. asperellum, T. harzianum, and T. asperellum + T. harzianum by soil soak method inflicted an induced systemic resistance (ISR) in chilli against a C. truncatum-challenged condition. In chilli, the disease index percentage was significantly reduced in the T. asperellum + T. harzianum-treated seeds, followed by the T. harzianum-, T. asperellum-, and P. dendritiformis-treated seeds as compared to the untreated and challenged, respectively. Chilli seeds were primed with T. asperellum + T. harzianum (78.67%), which revealed maximum disease protection under the challenged condition, followed by T. harzianum (70%), T. asperellum (64%), and P. dendritiformis (56%) as compared to untreated and C. truncatum-challenged (6%) condition served as control. The seeds that were pretreated with biocontrol agents (BCAs) inflicted ISR against C. truncatum by enhancing the activity of defence-related enzymes (superoxide dismutase (SOD), peroxidase (POX), polyphenol oxidase (PPO), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX) and phenylalanine ammonia-lyase (PAL)), accumulating phenolic compounds, and increasing the relative chlorophyll content in chilli. Nitroblue tetrazolium (NBT) and 3,3′-Diaminobenzidine (DAB) stains were used to detect the accumulation of superoxide anion and hydrogen peroxide that appeared nearby the fungal infection sites. The accumulation of reactive oxygen species (O2− and H2O2) in the pathogen-inoculated leaves was a maximum of 48 hpi, followed by P. dendritiformis, T. asperellum, T. harzianum, and T. asperellum + T. harzianum treated tissue upon C. truncatum-challenged condition as compared to the control. Overall, our results showed the potential of T. harzianum, T. asperellum, and P. dendritiformis as biocontrol agents that prevent infection by C. truncatum and inflict an induced systemic resistance in chilli by enhancing the biosynthesis of phenolic compounds, defence and antioxidative enzymes, and reducing the lesion development and reactive oxygen species accumulation. This is the first report of induced systemic resistance against anthracnose in chilli obtained by application of T. harzianum, T. asperellum and P. dendritiformis, through seed priming.

The comparative study of capsicum anthracnose pathogens from Korea withthat of China

Abstract:Anthracnose of capsicum casued by Colletotrichum spp. (C. spp. ) has been a great problems for capsicum production in Korea and China. Based on morphological characters, the capsicum anthracnose pathogen isolates K1 and C1, K2 and C2, K3 and C3 were identified as Colletotrichurn gloeosporioides G type, C. gloeosporioides R type and C. coccodes, respectively. Pathogenicity of Korea isolate and China isolate in capsicum lines of Korea and China did not show a difference. Results of RAPD-PCR analyses indicate that the similarities between K1 and C1, K2 and C2, and K3 and C3 were 85.71％, 71.43％ and 50.0％, respectively.
Comparison of associations to vowel speech sounds by English and Spanish speakers.

Abstract:Many psycholinguists have studied associations to vowel speech sounds. It appears that associations involving brightness and size are related to the manner in which the vowels are articulated. That is, high front vowels are judged to be bright and small, and low back vowels are judged to be dim and large. In an extension of a study by Greenberg and Jenkins (1966), 40 English-speaking and 40 Spanish-speaking adults rated nine audiotaped vowel sounds on 23 dimensions. The front-back distinction was again found for both groups. In addition, ratings for all nine vowels were similar for the two groups, which has implications for the cross-cultural universality of these associations.

Abstract:Members of the genus Colletotrichum include some of the most economically important fungal pathogens in the world. Accurate diagnosis is critical to devising disease management strategies. Two species, Colletotrichum gloeosporioides and C. truncatum, are responsible for anthracnose disease in papaya (Carica papaya L.) and bell pepper (Capsicum annuum L.) in Trinidad. The ITS1–5.8S–ITS2 region of 48 Colletotrichum isolates was sequenced, and the ITS PCR products were analyzed by PCR-RFLP analysis. Restriction site polymorphisms generated from 11 restriction enzymes enabled the identification of specific enzymes that were successful in distinguishing between C. gloeosporioides and C. truncatum isolates. Species-specific restriction fragment length polymorphisms generated by the enzymes AluI, HaeIII, PvuII, RsaI, and Sau3A were used to consistently resolve C. gloeosporioides and C. truncatum isolates from papaya. AluI, ApaI, PvuII, RsaI, and SmaI reliably separated isolates of C. gloeosporioides and C. truncatum from bell pepper. PvuII, RsaI, and Sau3A were also capable of distinguishing among the C. gloeosporioides isolates from papaya based on the different restriction patterns that were obtained as a result of intra-specific variation in restriction enzyme recognition sites in the ITS1–5.8S–ITS2 rDNA region. Of all the isolates tested, C. gloeosporioides from papaya also had the highest number of PCR-RFLP haplotypes. Cluster analysis of sequence and PCR-RFLP data demonstrated that all C. gloeosporioides and C. truncatum isolates clustered separately into species-specific clades regardless of host species. Phylograms also revealed consistent topologies which suggested that the genetic distances for PCR-RFLP-generated data were comparable to that of ITS sequence data. ITS PCR-RFLP fingerprinting is a rapid and reliable method to identify and differentiate between Colletotrichum species.

Identification and Genetic Diversity Analysis of the Pathogen of Anthracnose of Pepper in Guizhou

Abstract:Anthracnose of pepper is a significant disease caused by Colletotrichum spp. In 2017 and 2021, 296 isolates were obtained from 69 disease samples. Through morphological analysis, pathogenicity detection, and polygenic phylogenetic analysis, the above strains were attributed to 10 species: C. scovillei, C. fructicola, C. karstii, C. truncatum, C. gloeosporioides, C. kahawae, C. boninense, C. nymphaeae, C. plurivorum, and C. nigrum. C. scovillei had the most strains (150), accounting for 51.02% of the total isolates; C. fructicola came in second (72 isolates), accounting for 24.49%. Regarding regional distribution, Zunyi City has the highest concentration of strains—92 strains total, or 34.18%—across seven species. Notably, this investigation showed that C. nymphaeae infected pepper fruit for the first time in China. Genetic diversity analysis showed that C. fructicola could be divided into seven haplotypes, and the population in each region had apparent genetic differentiation. However, the genetic distance between each population was not significantly related to geographical distance. Neutral detection and nucleotide mismatch analysis showed that C. fructicola might have undergone population expansion.

Abstract:ABSTRACT The present studies were undertaken to investigate the effect of bioprotectant, plant based chemical like Salicylic acid and silicon based nutrient Potassium silicate against the anthracnose or ripe fruit rot of chilli (Capsicum annuum L.) incited by Colletotrichum capsici (Sydow.) Butler and Bisby in chilli var. K2. The chilli anthracnose disease susceptible variety Kovilpatti-2 (K2) grown in pots and field trial were used for the study. The plants were given artificial inoculation by spraying the spore suspensions after pinpricking the fruits with adequate spore load at 90-days after transplanting for fruit rot incidence assessment. The crop was maintained in a poly house with frequent spraying of water to provide adequate moisture and relative humidity to enable successful infection by the pathogen. Three replications for each treatment and a control were maintained. The fungicide mancozeb @ 0.25 per cent was used for comparison and the standard agronomic practices as recommended by the State Agricultural Department were followed. The plant activator and silicon-based nutrient were sprayed as individual as well as combined approach manner. Seed treatment with Pseudomonas fluorescens @ 10 g/kg of seeds in all the treatments except comparison fungicide and control treatments. Among the various treatments, seed treatment with Bio protectant, foliar application of Salicylic acid @ 50 ppm on 40 days after transplanting (40 DAT) and foliar application of Potassium silicate @ 3 % on 60 DAT recorded the minimum disease incidence and increased fruit length, fruit weight, fruit per plant, branches per plant, plant height and germination percentage when compared to control and Comparison fungicide Mancozeb. The activity of plant defense enzymes like Peroxidase, polyphenol oxidase and Phenylalanine ammonia lyase increased up to 5th day of sampling in plants treated with Bio-protectant, resistance inducing chemical along with potassium silicate and challenged inoculated with the test pathogen. Also, the same treatment significantly decreased the disease severity and increased the growth and yield parameters when compared to control and comparison fungicide under the field trials.
Soybean Bacterial Endophytes against Anthracnose Disease Collected from Karnataka State: An in-vitro Study

Abstract:Endophytes trigger various defence mechanisms within their host plants, engaging primary and secondary protective pathways. This investigation primarily aimed to isolate bacterial endophytes from diverse agroecological regions in Karnataka. Subsequently, these endophytes were assessed for their inhibition against Colletotrichum truncatum using the in-vitro streak plate technique. A total of 43 bacteria isolated from soybean plants and key endophytes showing the inhibition against C. truncatum were in different zones namely, DHW-9(87%), BID-2(85%), BID-13(85%), BID-14(82.50%), DHW-15(80%), BID-15(75%),and BID-16(75%) exhibited notable efficacy against C. truncatum in decreasing order. Among these, the DHW-9 (Stenotrophomonas maltophilia strain P4-32) bacterial endophytes isolated from the North Transition Zone (Dharwad) were highly effective against the pathogen, possibly due to employing many direct and indirect mechanisms. Furthermore, the inhibition potential of the bacterial endophytes varies with and within the place of agroecological zones. In conclusion, it has been observed that the bacterial endophyte DHW-9 inhibited the progression of anthracnose disease caused by C. truncatum in controlled in vitro. Hence, it is imperative to conduct additional experiments, including pot and field studies, to explore its potential to enhance the growth and yield of soybean plants.

Abstract:The root-knot nematode infestation is a significant challenge in chilli cultivation, necessitating the development of resistant cultivars. This study aimed to screen 200 chilli germplasm lines and identify resistant sources for root-knot nematode using multivariate analysis at ICAR-Indian Institute of Horticultural Research, Bangalore. The parameters such as shoot length, root length, shoot weight, root weight, shoot girth were assessed, and nematode infestation was quantified by counting galls and egg masses. The germplasm lines were categorized based on gall index and egg mass index into highly resistant, resistant, moderately resistant, moderately susceptible, susceptible, and highly susceptible groups. The analysis of variance revealed substantial variation among the germplasm lines. The Pearson correlation analysis revealed significant positive associations between nematode infestation and growth parameters. The cluster analysis identified three distinct groups of genotypes based on root knot nematode tolerance. The factor analysis 1 is associated with nematode infestation, and factor analysis 2 is related to growth parameters. The multi-trait ideotype genotype distance index analysis integrated multiple traits to identify top-performing genotypes. The germplasm lines EC771549-A, IHR3226 and IHR3575 showed high resistance, while Pusa Sadabahar was susceptible. The PCR-specific markers were used for validation of Me genes for root-knot nematode resistance. The simple sequence repeat (SSR) marker GPMS 171 and sequence characterized amplified region (SCAR) marker SCAR_CD showed clear polymorphism, correlating with phenotypic resistance. The germplasm line EC771549-A, is consistently resistant at phenotypic, molecular and selected based on multi-trait ideotype genotype distance index values, while Pusa Sadabahar was confirmed as susceptible. This study highlights the effectiveness of integrating molecular marker analysis with phenotypic evaluation and multivariate analysis to identify root-knot nematode resistant sources. The findings provide valuable insights into the genetic architecture of root-knot nematode resistance in chilli and candidate lines for breeding programme aimed at developing resistant cultivars, thereby contributing to sustainable chilli production and nematode management.

Abstract:Northeastern states of India are known for unique landraces of Capsicum spp. with geographical indications. However, little information is available about these valuable landraces of chillies. Surveys and collections were carried out in niche areas to find out their ecology and diversity through morphological traits and molecular analysis using microsatellite markers. Our result characterized the ecology of niche areas as cool (11.0°C–20.7°C) and humid (>60% relative humidity) climates for dalle-chilli ( Capsicum annuum L.); mild-warm (12.2°C–28.6°C) and humid for king-chilli ( C. chinense Jacq.); and cool to warm (11.3°C–33.1°C) and humid for bird's eye chilli ( C. frutescens L.) during the crop period. The canonical correspondence analysis has shown the significant impact of temperature on the agro-morphological traits and distribution of the landraces in their niche areas. A wide variability was observed for different quantitative traits and yield attributing characters (fruit length, diameter, weight, and yield), showing high heritability (97.0%–99.0%), and genetic advance as a percentage of the mean (119.8%–434.0%). A total of 47 SSR markers used for the molecular analysis generated 230 alleles, ranging from 2 (HPMSE-7) to 10 (HPMSE-5), with an average of 4.89 alleles per locus. The average polymorphism information content was also high (0.61) and ranged from 0.20 (HPMSE-7) to 0.85 (CAMS-91). The observed average heterozygosity was lower than the expected value. Analysis of molecular variance has shown significant variation within (69%) and between (31%) of the populations of Capsicum spp. Based on Nei's genetic distance, bird's eye chilli and king-chilli were found to be closer to each other, whereas dalle-chilli, a tretraploid species, was closer to hot pepper ( C. annuum ). However, the flower size of dalle-chilli was large and found closer to king-chilli in color and differs from C. chinense due to the presence of calyx teeth. For quality traits, landraces king-chilli, dalle-chilli, and bird's eye chilli have shown 2.8, 2.0, and 1.4 times higher average capsaicin and 0.46, 0.25, and 0.22 times higher average oleoresin content over the hot pepper, respectively. The knowledge of ecology and diversity can be used in identifying new areas for production, selection of elite lines, conservation, and crop improvement.

Abstract:Anthracnose leaf spot caused by Colletotrichum gloeosporioides is an important disease of apples, resulting in serious damage to the fruit production. In this paper, the pathogenic and physiological characters of sixteen isolates and the wild isolate “Stj16” were studied. In the current study, we generated C. gloeosporioides strains expressing green fluorescence by introducing a GFP gene via an Agrobacterium tumefaciens-mediated transformation (ATMT). To confirm the subcellular localisation of the A2799 gene, an A2799gfp fusion expression mutant was constructed. After observation of the fusion expression, the A2799gfp fusion expression protein was located in the peroxisomes of the cell. The pathogenicity results showed that the mutants A4204, M44, A1919, A3638 and A1598 lost the pathogenic capability and virulence, however, the virulence of the mutants A1764, A439, A3885, G1183, A3144, A1649 and A2675 increased significantly to the apple fruits compared to the wild isolate “Stj16”. The biological study indicated that a Rose Bengal Agar (RBA) medium decreased the mycelium growth, but it can increase the sporulation for most of the isolates. The mutant A4204 does not grow well at pH 4.0 and pH 8.0, and mutant M44 just has the optimum growth at pH 8.0, and a 12 h light and 12 h dark condition stimulates the sporulation for most of the tested mutants, but the A1764 mutant more sporulated at regular dark conditions. All the mutants and “Stj16” grew vigorously at 25 °C–30 °C, for “Stj16”, it produced the highest number of conidia at 30 °C compared with the other temperatures. Based on the biological study, we found the best growing and sporulation conditions for all the tested isolates. The information generated in the present study will facilitate molecular research on this devastating fungus.
