Abstract:Activation of mitogen-activated protein kinases (MAPKs) is one of the earliest responses after plants sense an invading pathogen. Here, we show that MPK3 and MPK6, two Arabidopsis thaliana pathogen-responsive MAPKs, and their upstream MAPK kinases, MKK4 and MKK5, are essential to both stomatal and apoplastic immunity. Loss of function of MPK3 and MPK6, or their upstream MKK4 and MKK5, abolishes pathogen/microbe-associated molecular pattern-and pathogen-induced stomatal closure. Gain-of-function activation of MPK3/MPK6 induces stomatal closure independently of abscisic acid (ABA) biosynthesis and signaling. In contrast, exogenously applied organic acids such as malate or citrate are able to reverse the stomatal closure induced by MPK3/MPK6 activation. Gene expression analysis and in situ enzyme activity staining revealed that malate metabolism increases in guard cells after activation of MPK3/MPK6 or inoculation of pathogen. In addition, pathogen-induced malate metabolism requires functional MKK4/MKK5 and MPK3/MPK6. We propose that the pathogen-responsive MPK3/MPK6 cascade and ABA are two essential signaling pathways that control, respectively, the organic acid metabolism and ion channels, two main branches of osmotic regulation in guard cells that function interdependently to control stomatal opening/closure.

Abstract:Abstract Photosynthesis is co-limited by multiple factors depending on the plant and its environment. These include biochemical rate limitations, internal and external water potentials, temperature, irradiance, and carbon dioxide (CO2). Amphis- tomatous leaves have stomata on both abaxial and adaxial leaf surfaces. This feature is considered an adaptation to alleviate CO2 diffusion limitations in pro- ductive environments as the diffusion path length from stomate to chloroplast is effectively halved in amphistomatous leaves. Plants may also reduce CO2 limitations through other aspects of optimal stomatal anatomy: stomatal den- sity, distribution, patterning, and size. A number of studies have demonstrated that stomata are overdispersed compared to a random distribution on a sin- gle leaf surface; however, despite their prevelance in nature and near ubiquity among crop species, much less is known about stomatal anatomy in amphis- tomatous leaves, especially the coordination between leaf surfaces. Here we use novel spatial statistics based on simulations and photosynthesis modeling to test hypotheses about how amphistomatous plants may optimize CO2 diffusion in the model angiosperm Arabidopsis thaliana grown in different light environ- ments. We find that 1) stomata are overdispersed, but not ideally dispersed, on both leaf surfaces across all light treatments; 2) the patterning of stomata on abaxial and adaxial leaf surfaces is independent; and 3) the theoretical im- provements to photosynthesis from abaxial-adaxial stomatal coordination are miniscule (≪ 1%) across the range of feasible parameter space. However, we also find that 4) stomatal size is correlated with the mesophyll volume that it supplies with CO2, suggesting that plants may optimize CO2 diffusion limita- tions through alternative pathways other than ideal, uniform stomatal spacing. We discuss the developmental, physical, and evolutionary constraits which may prohibit plants from reaching this theoretical adaptive peak of uniform stomatal spacing and inter-surface stomatal coordination. These findings contribute to our understanding of variation in the anatomy of amphistomatous leaves.

Fighting for Survival at the Stomatal Gate

Abstract:Stomata serve as the battleground between plants and plant pathogens. Plants can perceive pathogens, inducing closure of the stomatal pore, while pathogens can overcome this immune response with their phytotoxins and elicitors. In this review, we summarize new discoveries in stomata-pathogen interactions. Recent studies have shown that stomatal movement continues to occur in a close-open-close-open pattern during bacterium infection, bringing a new understanding of stomatal immunity. Furthermore, the canonical pattern-triggered immunity pathway and ion channel activities seem to be common to plant-pathogen interactions outside of the well-studied Arabidopsis-Pseudomonas pathosystem. These developments can be useful to aid in the goal of crop improvement. New technologies to study intact leaves and advances in available omics data sets provide new methods for understanding the fight at the stomatal gate. Future studies should aim to further investigate the defense-growth trade-off in relation to stomatal immunity, as little is known at this time.
Relationships between nodal status and cell kinetics, DNA ploidy pattern and histopathology of the deeply infiltrating sites in colorectal adenocarcinoma

Abstract:Tumor cell kinetics and DNA contents were investigated by in vitro labeling with bromodeoxyuridine (BrdU) and flow cytometry using paraffin‐embedded specimens in both superficially (Sup) and deeply (Deep) infiltrating sites of 28 colorectal adenocarcinomas. Eleven were well differentiated and 17 were moderately differentiated adenocarcinomas. In moderately differentiated adenocarcinoma, histologic low grade types of Deep were frequently associated with lymph node metastasis, but the BrdU labeling indices (LI) or tumor DNA ploidy pattern did not correlate with nodal status. Among the seven aneuploid tumors of moderately differentiated adenocarcinoma, the BrdU LI and histology of Deep tended to correlate with nodal status. These findings suggest that the histology of Deep should have a greater significance than tumor cell kinetics or DNA contents. They also suggest that the tumor DNA ploidy pattern may be heterogeneous and divided into subgroups according to its cell kinetics or histology of Deep in relation to lymph node metastasis.

Recent studies by Hu et al. and Roussin-Léveillée et al. demonstrate that, following entry, Pseudomonas syringae closes stomata and, thus, reduces transpiration in infected leaves, adding another layer of complexity to the stomata-pathogen interaction.

Abstract:Phyllosphere microbes have received much attention. More and more studies have been conducted to know about the microbes species and how they colonize and interact with plants. Stomata were considered as passive portal of entry for plant pathogenic bacteria. However, recent studies suggest that stomata can play an active role in restricting bacterial invasion as one part of the plant innate immune system. The paper has summarized recent developments in molecular ecology on stomata-based immunity. And on the base of reports on the relationship between stomata closure and plant water use efficiency, studies to develop crop water-saving and product- increasing technique by making use of the stomata-based defense are feasible. Plant stomata-pathogen interaction will become the next focus of molecular ecology and agricultural ecology.
Leaf temperature and its dependence on atmospheric CO2 and leaf size

Abstract:There is general concern that the rapid increase in atmospheric CO2 concentration will lead to reduced stomatal conductance and subsequent increases in leaf temperature. Such an increase in leaf temperature is expected to adversely impact a plethora of processes connected to leaf metabolism and microbial/fungal communities on leaves. A model is proposed that combines the leaf energy balance with leaf gas exchange and photosynthesis to explore such issues. The model represents a hybrid ecological/physiological approach described by systems of equations based on steady‐state leaf‐gas exchange theories and leaf energy/radiation balance, equilibrium thermodynamics within the leaf, stomatal data, and atmospheric CO2 concentration. The model allows separating air from leaf temperatures thereby permitting exploration of the dependence of leaf cooling or heating for any combination of environmental conditions (e.g., wind velocity, atmospheric humidity, and atmospheric CO2 level), anatomic leaf properties (e.g., leaf size), and physiologic quantities (e.g., assimilation rate and transpiration rate). The model permits to distinguish whether leaf cooling or heating is to be expected if these parameters are varied. Based on model calculations, it is shown that leaf temperature is far more impacted by leaf size or wind speed than reduction in stomatal conductance caused by elevated atmospheric CO2. The model results are consistent with measurements of leaf cooling and heating.

A Whole Leaf Comparative Study of Stomatal Conductance Models

Abstract:We employed a detailed whole leaf hydraulic model to study the local operation of three stomatal conductance models distributed on the scale of a whole leaf. We quantified the behavior of these models by examining the leaf-area distributions of photosynthesis, transpiration, stomatal conductance, and guard cell turgor pressure. We gauged the models' local responses to changes in environmental conditions of carbon dioxide concentration, relative humidity, and light irradiance. We found that a stomatal conductance model that includes mechanical processes dependent on local variables predicts a spatial variation of physiological activity across the leaf: the leaf functions of photosynthesis and transpiration are not uniformly operative even when external conditions are uniform. The gradient pattern of hydraulic pressure which is needed to produce transpiration from the whole leaf is derived from the gradient patterns of turgor pressures of guard cells and epidermal cells and consequently leads to nonuniform spatial distribution patterns of transpiration and photosynthesis via the mechanical stomatal model. Our simulation experiments, comparing the predictions of two versions of a mechanical stomatal conductance model, suggest that leaves exhibit a more complex spatial distribution pattern of both photosynthesis and transpiration rate and more complex dependencies on environmental conditions when a non-linear relationship between the stomatal aperture and guard cell and epidermal cell turgor pressures is implemented. Our model studies offer a deeper understanding of the mechanism of stomatal conductance and point to possible future experimental measurements seeking to quantify the spatial distributions of several physiological activities taking place over a whole leaf.

Comparing Methodologies for Stomatal Analyses in the Context of Elevated Modern CO2

Abstract:Leaf stomata facilitate the exchange of water and CO2 during photosynthetic gas exchange. The shape, size, and density of leaf pores have not been constant over geologic time, and each morphological trait has potentially been impacted by changing environmental and climatic conditions, especially by changes in the concentration of atmospheric carbon dioxide. As such, stomatal parameters have been used in simple regressions to reconstruct ancient carbon dioxide, as well as incorporated into more complex gas-exchange models that also leverage plant carbon isotope ecology. Most of these proxy relationships are measured on chemically cleared leaves, although newer techniques such as creating stomatal impressions are being increasingly employed. Additionally, many of the proxy relationships use angiosperms with broad leaves, which have been increasingly abundant in the last 130 million years but are absent from the fossil record before this. We focus on the methodology to define stomatal parameters for paleo-CO2 studies using two separate methodologies (one corrosive, one non-destructive) to prepare leaves on both scale- and broad-leaves collected from herbaria with known global atmospheric CO2 levels. We find that the corrosive and non-corrosive methodologies give similar values for stomatal density, but that measurements of stomatal sizes, particularly guard cell width (GCW), for the two methodologies are not comparable. Using those measurements to reconstruct CO2 via the gas exchange model, we found that reconstructed CO2 based on stomatal impressions (due to inaccurate measurements in GCW) far exceeded measured CO2 for modern plants. This bias was observed in both coniferous (scale-shaped) and angiosperm (broad) leaves. Thus, we advise that applications of gas exchange models use cleared leaves rather than impressions.

Abstract:Earth system models are incorporating plant trait diversity into their land components to better predict vegetation dynamics in a changing climate. However, extant plant trait distributions will not allow extrapolations to novel community assemblages in future climates, which will require a mechanistic understanding of the trade-offs that determine trait diversity. In this study, we show how physiological trade-offs involving leaf mass per unit area (LMA), leaf lifespan, leaf nitrogen, and leaf respiration may explain the distribution patterns of evergreen and deciduous trees in the temperate and boreal zones based on (1) an evolutionary analysis of a simple mathematical model and (2) simulation experiments of an individual-based dynamic vegetation model (i.e., LM3-PPA). The evolutionary analysis shows that these leaf traits set up a trade-off between carbon- and nitrogen-use efficiency at the scale of individual trees and therefore determine competitively dominant leaf strategies. As soil nitrogen availability increases, the dominant leaf strategy switches from one that is high in nitrogen-use efficiency to one that is high in carbon-use efficiency or, equivalently, from high-LMA/long-lived leaves (i.e., evergreen) to low-LMA/short-lived leaves (i.e., deciduous). In a region of intermediate soil nitrogen availability, the dominant leaf strategy may be either deciduous or evergreen depending on the initial conditions of plant trait abundance (i.e., founder controlled) due to feedbacks of leaf traits on soil nitrogen mineralization through litter quality. Simulated successional patterns by LM3-PPA from the leaf physiological trade-offs are consistent with observed successional dynamics of evergreen and deciduous forests at three sites spanning the temperate to boreal zones.

An Inverse Scaling Relationship between Stomatal Density and Mean Nearest Neighbor Distance: Evidence from a Photinia Hybrid and One of Its Parents

Abstract:Stomata are involved in transpiration and CO2 uptake by mediating gas exchange between internal plant tissues and the atmosphere. The capacity for gas exchange depends on stomatal density (SD), stomatal size, and pore dimensions. Most published work on stomatal quantification has assumed that stomatal distribution and stomatal density are spatially homogeneous across the leaf, but this assumption has been seldom tested. We selected 32 leaves from a Photinia hybrid, Photinia × fraseri 'Red Robin', and one of its parents, P. serratifolia. For each leaf, the leaf surface was divided into three or four equidistant layers along the apical-basal axis, and, in each layer, two positions, one closer to the midrib and the other closer to the leaf margin, were further selected. We calculated SD and mean nearest neighbor distance (MNND) for each lamina section and tested the scaling relationship between SD and MNND of the sampled stomatal centers using reduced major axis protocols. In addition, we calculated the stomatal aggregation index (SAI) for each lamina section to examine the spatial arrangement of stomata at the given size of field of view of 1.2 mm × 0.9 mm. We observed that SD decreased from the lamina apex towards the base for central lamina areas but varied little at leaf margins. An inverse scaling relationship between SD and MNND was observed for both species. This relationship could be used for SD estimation using the rapidly estimated trait, MNND. SAI did not vary significantly throughout leaf lamina, and the numerical values of SAI for all fields of view were greater than one, which indicates significant spatial repulsion between stomata. The study suggests that SD varies across leaf lamina to fine-tune plant water use and maximize carbon gain. However, spatial structures of stomata from different lamina sections exhibit similar patterns (i.e., spatial inhibition between stomata at small scales), probably due to hierarchical leaf vein patterns.

Abstract:The study tried to understand the meaning of hospitalization for children and teenagers with chronic diseases substantiated on Symbolic Interactionism and developed from the qualitative Assistential Converging Research whose participants were a child and three teenagers. Data collection took place through a nursing interaction process, participating observation, sensible listening and child drawing. During the analysis, the meaning of hospitalization was expressed as being ill, away from home, feeling pain, sadness, suffering, wishing to cry, nervousness, aggressiveness and losing freedom. It was also understood as treatment, cure, support, friends, reciprocity with nature and "pleasant experiences" through care, passion, love, happiness and games. The participants' ability to show their view of the situation in which they are inserted was highlighted in their drawings, imbued with meanings and life lessons.

Abstract:Stomata are tiny pores on the underside of a plant leaf that allow for gas exchange. The pores are formed by a pairs of guard cells that are responsible for regulating the size of the opening. Abstract:Many plant pathogens gain entry to their host via stomata. On sensing attack, plants close these pores to restrict pathogen entry. Here, we show that plants exhibit a second longer‐term stomatal response to pathogens. Following infection, the subsequent development of leaves is altered via a systemic signal. This reduces the density of stomata formed, thus providing fewer entry points for pathogens on new leaves. <i>Arabidopsis thaliana</i> leaves produced after infection by a bacterial pathogen that infects through the stomata (<i>Pseudomonas syringae</i>) developed larger epidermal pavement cells and stomata, and consequently had up to 20% reductions in stomatal density. The bacterial peptide flg22 or the phytohormone salicylic acid induced similar systemic reductions in stomatal density suggesting that they might mediate this effect. In addition, flagellin receptors, salicylic acid accumulation, and the lipid transfer protein AZI1 were all required for this developmental response. Furthermore, manipulation of stomatal density affected the level of bacterial colonisation, and plants with reduced stomatal density showed slower disease progression. We propose that following infection, development of new leaves is altered by a signalling pathway with some commonalities to systemic acquired resistance. This acts to reduce the potential for future infection by providing fewer stomatal openings.

Abstract:Models disagree on how to represent effects of drought stress on plant gas exchange. Some models assume drought stress affects the marginal water use efficiency of plants (marginal WUE=∂A/∂E; i.e. the change in photosynthesis per unit of change in transpiration) whereas others assume drought stress acts directly on photosynthetic capacity. We investigated drought stress in an analysis of results from 22 experimental data sets where photosynthesis, stomatal conductance and predawn leaf water potential were measured at increasing levels of water stress. Our analysis was framed by a recently developed stomatal model that reconciles the empirical and optimal approaches to predicting stomatal conductance. The model has single parameter g 1, a decreasing function of marginal WUE. Species differed greatly in their estimated g 1 values under moist conditions, and in the rate at which g 1 declined with water stress. In some species, particularly the sclerophyll trees, g 1 remained nearly constant or even increased. Photosynthesis was found almost universally to decrease more than could be explained by the reduction in g 1, implying a decline in apparent carboxylation capacity (V cmax). Species differed in the predawn water potential at which apparent V cmax declined most steeply, and in the steepness of this decline. Principal components analysis revealed a gradient in water relation strategies from trees to herbs. Herbs had higher apparent V cmax under moist conditions but trees tended to maintain more open stomata and higher apparent V cmax under dry conditions. There was also a gradient from malacophylls to sclerophylls, with sclerophylls having lower g 1 values under well-watered conditions and a lower sensitivity of apparent V cmax to drought. Despite the limited amount of data available for this analysis, it is possible to draw some firm conclusions for modeling: (1) stomatal and non-stomatal limitations to photosynthesis must both be considered for the short-term response to drought and (2) plants adapted to arid climate respond very differently from others.

Abstract:Stomatal pores in leaves mediate CO2 uptake into the plant and water loss via transpiration. Most plants are hypostomatous with stomata present only in the lower leaf surface (abaxial epidermis). Many herbs, including the model plant Arabidopsis thaliana, have substantial numbers of stomata also on the upper (adaxial) leaf surface. Studies of stomatal development have mostly focused on abaxial stomata and very little is known of adaxial stomatal formation. We addressed the role of leaf number in determination of stomatal density and stomatal ratio, and studied adaxial and abaxial stomatal patterns in mutants deficient in known abaxial stomatal development regulators. We found that stomatal density in some genetic backgrounds varies between different fully developed leaves and recommend using defined leaves for analyses of stomatal patterning. Our results indicate that stomatal development is at least partly independently regulated in adaxial and abaxial epidermis, as i) plants deficient in ABA biosynthesis and perception have increased stomatal ratios, ii) the epf1epf2, tmm and sdd1 mutants have reduced stomatal ratios, iii) erl2 mutants have increased adaxial but not abaxial stomatal index, and iv) stomatal precursors preferentially occur in abaxial epidermis. Further studies of adaxial stomata can reveal new insights into stomatal form and function.

Abstract:Leaf surface conductance to water vapor and CO2 across the epidermis (gleaf) strongly determines rates of gas exchange. Thus, clarifying the drivers of gleaf has important implications for resolving mechanisms of photosynthetic productivity and leaf and plant responses and tolerance to drought. It is well recognized that gleaf is a function of the conductances of the stomata (gs) and of the epidermis + cuticle (gec). Yet, controversies have arisen around the relative roles of stomatal density (d) and size (s), fractional stomatal opening (α; aperture relative to maximum) and gec in determining gleaf. Resolving the importance of these drivers is critical across the range of leaf surface conductances, from strong stomatal closure under drought (gleaf, min), to typical opening for photosynthesis (gleaf, op), to maximum achievable opening (gleaf, max). We derived equations and analyzed a compiled database of published and measured data for approximately 200 species and genotypes. On average, within and across species, higher gleaf, min was determined ten times more strongly by α and gec than by d, and negligibly by s; higher gleaf, op was determined approximately equally by α (47%) than by stomatal anatomy (45% by d, and 8% by s), and negligibly by gec; and higher gleaf, max was determined entirely by d. These findings clarify how diversity in stomatal functioning arises from multiple structural and physiological causes with importance shifting with context. The rising importance of d relative to α, from gleaf, min to gleaf, op, enables even species with low gleaf, min, which can retain leaves through drought, to possess high d and thereby achieve rapid gas exchange in periods of high water availability.

Abstract:In terrestrial plants, stomata in the leaf epidermis formed by guard cells are the major pathways for gas exchange. However, opened stomata also provide a major channel for pathogen entry. At the pre‐invasive stage, plants actively close stomata to prevent pathogen attack, which is termed stomatal immunity, and plant hormones are involved in this process. Here, we summarize recent advances in the role of abscisic acid (ABA) in promoting stomatal immunity to prevent pathogen entry. Additionally, salicylic acid shares common downstream elements with ABA to promote stomatal immunity, whereas reactive oxygen species and Ca2+ act as critical signals to cross‐talk with ABA signalling to regulate stomatal movement, and they also enhance the effect of ABA in stomatal immunity. On the other hand, at the post‐invasive stage, closed stomata create a water‐soaked environment that allows pathogen multiplication, and ABA signalling is hijacked by pathogens to reduce stomatal aperture. Here, we propose a model of dual roles of ABA‐mediated stomatal closure during plant‐pathogen interaction and offer emerging consequences and questions for ABA‐mediated stomatal immunity.

Installation, system-level control strategy and commissioning of the Nanjing UPFC project

Abstract:The Nanjing unified power flow controller (UPFC) project, which is the world's first modular multilevel converter based UPFC (MMC-UPFC) project, has been put into operation in the Nanjing Western Power Grid by State Grid Corporation of China since December 14, 2015. The project comprises three 60 MVA converters and is applied to a 220 kV ac system. The installation of the project is reported in this paper, including the optimized structure and the layout. Besides, the UPFC systemlevel control strategy is discussed. With the proposed system-level control strategy, a key transmission section of the Nanjing Western Power Grid will be operated at the preset operating point under the normal condition. While, the power flows of the transmission lines in the transmission section can be controlled within the line loading limits under fault conditions. The commissioning test results of the Nanjing UPFC project and the proposed system-level control strategy are also presented. The waveforms recorded show that the project performs well under the normal operation condition and the fault conditions. The security of the Nanjing Western Power Grid is enhanced by the installation of the Nanjing UPFC project.
