Abstract:Developing polymers with switchable glass transition temperature (Tg) can solve scientific challenges such as fracture healing of polymers with high Tg and processing of hard polymers without plasticizer at room temperature. The azobenzene-containing polymers, or the so-called azopolymers, are smart materials that show photoswitchable Tg and photoinduced motions, which can be used in fabrication of photoactuators. Here, healable and reprocessable models were constructed by using linear azopolymers. Molecular dynamics (MD) simulation was used to study the behaviors of photo-induced bending, self-healing, and directional photofluidization of the azopolymers. A temporary modification of the C–N=N–C dihedral potential was applied to achieve the isomerization of azopolymer by using polymer consistent force-field (PCFF). The simulation results show that isomerization of azobenzene groups can lead to photoswitchable Tg of the azopolymers, which resulting in shape changes and different mechanical responses. Furthermore, the photoswitchable Tg makes the azopolymers much easier to heal under UV irradiation, and restore the mechanical properties under visible light irradiation. Finally, the mechanism of photoliquefaction was investigated, our results imply that the photoinduced trans–cis-trans cycles can reduce the mechanical properties and Tg values of azopolymers.

Abstract:The reversible photoisomerization of thirty azobenzene derivatives, their cis to trans thermal isomerization and supramolecular interactions have been controlled by various substitutions at different positions. This perspective article features the recent work published in this journal on functionalized azobenzene photoswitches, studied by Venkataramani and coworkers [Grewal, S. et al (2024) Photochem. Photobiol., 1–16]. Azobenzene‐based photochromic molecules undergo reversible trans‐to‐cis isomerization under exposure to light. The reverse cis‐to‐trans isomerization also takes place under thermal conditions in the dark. The study explores how different functional groups at various positions affect the responses of these compounds to light, their thermal stabilities, the isomeric ratios in the photostationary states, and their supramolecular behavior. Results show varied thermal half‐lives, influenced by the terminal groups and their positions. Aggregation studies reveal the formation of supramolecular architectures, from microcrystals to gels with these systems. Despite complexities with the large volume of work with 30 different compounds, the research provides insights into designing efficient photochromic supramolecular motifs.

Abstract:Benzimidazole heterocycles are of great importance in medicinal chemistry due to their applicability to a wide range of pharmacological targets, therefore representing a prototypical "privileged structure". In photopharmacology, azoheteroarene photoswitches have emerged as valuable tools for a variety of applications due to the high tuneability of their photophysical properties. Benzimidazole-based photoswitches should therefore enable the optically-controlled investigation of many pharmacological targets. Here we report a combined experimental and computational investigation of such arylazobenzimidazoles, which allowed us to identify derivatives with near-quantitative bidirectional photoswitching using visible light and highly tuneable Z-isomer stability. We further demonstrate that arylazobenzimidazoles bearing a free benzimidazole N-H group not only exhibit efficient bidirectional photoswitching, but also excellent thermal Z isomer stability, contrary to previously reported fast-relaxing Z-isomers of N-H azoheteroarenes. Finally, we describe derivatives which can be reversibly isomerized with cyan and red light, thereby enabling significantly "red-shifted" photocontrol over prior azoheteroarenes. The understanding gained in this study should enable future photopharmacological efforts by employing photoswitches based on the privileged benzimidazole structure.

A platform for the development of highly red-shifted azobenzenes

Abstract:Azobenzenes are versatile photoswitches that can be used to generate elaborate optical tools including photopharmaceuticals. However, the targeted application-guided design of new photoswitches with specific properties, such as the ability to use tissue-penetrating red light combined with defined thermal relaxation rates and stability towards biological reducing agents, remains challenging. We have developed synthetic protocols for derivatives of the dfdc (di-ortho-fluoro di-ortho-chloro) azobenzene scaffold with various chemical alterations in the para-/ortho-positions and performed an in-depth study into the effects of their structures on their photo-physical properties with an emphasis on the n → pi* absorption band using NMR, UV-vis and X-ray analysis. The data was used to establish and validate a computational approach that, among other parameters, allows to compute realistic UV-vis spectra by combining TD-DFT excited-state calculations from 6000 thermally accessible structures generated through ab-initio MD simulations while taking the high structural flexibility of ortho-substituted azobenzenes into account. With our platform, we added 15 new visible light-operated photoswitches to the toolbox for the development of optical devices that harbor relaxation rates across multiple orders of magnitude. We have identified several examples with stronger bathochromic shifts than the dfdc azobenzene lead structure and a high stability towards the biological reducing agent GSH, which is a prerequisite for in vivo applications. An intriguing example is dfdb (di-ortho-fluoro di-ortho-bromo) azobenzene which demonstrates accelerated photoswitching with deep red light at increased temperatures. Taken together, our combined experimental and computational study forms the foundation for the advanced in silico design and synthesis of new highly red-shifted azobenzene photoswitches.

Visible-Light-Induced Exciton Dynamics and Trans-to-Cis Isomerization in Azobenzene Aggregates: Insights from Surface Hopping / Semiempirical Configuration Interaction Molecular Dynamics Simulations

Abstract:Assemblies of photochromic molecules feature exciton states, which govern photochemical and photophysical processes in multichromophoric systems. Understanding photoinduced dynamics of the assemblies requires nonadiabatic treatment involving multiple exciton states and numerous nuclear degrees of freedom, thus posing a challenge for simulations. In this work, we address this challenge for aggregates of azobenzene, a prototypical molecular switch, performing on-the-fly surface hopping calculations combined with semiempirical configuration interaction electronic structure and augmented with transition density matrix analysis to characterize exciton evolution. Specifically, we consider excitation of azobenzene tetramers in the nπ* absorption band (located in the visible (blue) part of the electromagnetic spectrum) thus extending our recent work on dynamics after ππ* excitation (corresponding to the ultraviolet region) [Titov, J. Phys. Chem. C 2023, 127, 13678–13688]. We find that the nπ* excitons, which are initially strongly localized by ground state conformational disorder, undergo further (very strong) localization during short-time photodynamics. This excited-state localization process is extremely ultrafast, occuring within first 10 fs of photodynamics. We observe virtually no exciton transfer of the localized excitons in the nπ* manifold. However, the transfer may occur via secondary pathways involving ππ* states or the ground state. Moreover, we find that nπ* quantum yields of the trans-to-cis isomerization are reduced in the aggregated state.

Ab initio study of photoisomerization mechanisms of push–pull p,p′-disubstituted azobenzene derivatives on S1 excited state

Abstract:In this study, the potential energy surfaces (PESs) of the ground-state (S0) and the first singlet excited-state (S1) along the CNNC rotational coordinate were mapped for 4-amino-4′-nitroazobenzene (ANAB) and 4-amino-4′-cyanoazobenzene (ACAB) on the basis of CASSCF calculations. The result indicates that the photoisomerization on the S1 state mainly involves the rotation mechanism for both ANAB and ACAB. A conical intersection with a twisted configuration (CIrot) locates on the middle of the PESs along the CNNC rotational coordinate. Another decay channel (inversion pathway or concerted inversion pathway), characterized by the evolution of two CNN angles bending to deactivate the excited molecules, also exists for both ANAB and ACAB. The energy of the conical intersection on the inversion channel (CIinv) is even higher than the E-FC point of S1 state, which is too high for cold S1 excited molecules to overcome. However, upon being excited to the S2 state or vibrationally hot S1 state, the inversion channel could be accessible to decay the excited electron energy. The differences between the CIinv of ANAB and ACAB can be seen by the analysis of the geometries, the gradient difference vector (X1), and the derivative coupling vector (X2). The calculation indicates that different electron-withdrawing groups (–NO2, –CN) on p-positions can modify the conical intersection lying on the inversion decay pathway of the azobenzene derivatives.

Abstract:A series of phenylene ethynylene azobenzenes bearing different symmetrical peripheral chromophores were synthesized by Sonogashira coupling and characterized by UV–vis absorption, fluorescence emission, cyclic voltammetry and cis–trans photoisomerization. All compounds exhibit strong 1π,π* absorption bands in the UV region, which pronouncedly blue-shifts when the π-electron donating chromophore attached on the para-position of phenyl group. When excited at the 1π,π* band, the compounds exhibit violet to blue emission (363 ∼ 430nm), which is attributed to a 1π,π* emission. All of these compounds show reversible trans–cis isomerization in dichloromethane solution, indicating that the π-conjugated phenylene ethynylene backbone does not prevent the occurrence of the photochemical processes of the azobenzene center. The photoisomerization properties were influenced by the peripheral chromophores in the conjugated system and the steric interactions. In addition, the kinetic investigation of the photoisomerization process was carried out.
Manipulating azobenzene photoisomerization through strong light-molecule coupling

Abstract:The formation of hybrid light-molecule states (polaritons) offers a new strategy to manipulate the photochemistry of molecules. To fully exploit its potential, one needs to build a toolbox of polaritonic phenomenologies that supplement those of standard photochemistry. By means of a state-of-the-art computational photochemistry approach extended to the strong-coupling regime, here we disclose various mechanisms peculiar of polaritonic chemistry: coherent population oscillations between polaritons, quenching by trapping in dead-end polaritonic states and the alteration of the photochemical reaction pathway and quantum yields. We focus on azobenzene photoisomerization, that encompasses the essential features of complex photochemical reactions such as the presence of conical intersections and reaction coordinates involving multiple internal modes. In the strong coupling regime, a polaritonic conical intersection arises and we characterize its role in the photochemical process. Our chemically detailed simulations provide a framework to rationalize how the strong coupling impacts the photochemistry of realistic molecules

Abstract:Herein, we report a series of azobenzene-substituted triptycenes. In their design, these switching units were placed in close proximity, but electronically separated by a sp3 center. The azobenzene switches were prepared by Baeyer-Mills coupling as key step. The isomerization behavior was investigated by 1 H NMR spectroscopy, UV/Vis spectroscopy, and HPLC. It was shown that all azobenzene moieties are efficiently switchable. Despite the geometric decoupling of the chromophores, computational studies revealed excitonic coupling effects between the individual azobenzene units depending on the connectivity pattern due to the different transition dipole moments of the π→π* excitations. Transition probabilities for those excitations are slightly altered, which is also revealed in their absorption spectra. These insights provide new design parameters for combining multiple photoswitches in one molecule, which have high potential as energy or information storage systems, or, among others, in molecular machines and supramolecular chemistry.

Abstract:Visible light driven azobenzene-based photoisomerization materials are expected to have potential applications in the fields of biological probe and sensor systems, photosensitive cells, energy storage materials and so on. The relationships between electronic structures and switching performance of o -R-substituted 4,4-diacetami-doazobenzene derivatives (R=F, Cl, Br or I) were investigated by using both density functional theory (DFT) and reactive molecular dynamics simulations. DFT calculations demonstrated that substituted groups of F, Cl, Br and I rendered the planar azo trans geometries distorted in different degree, resulting in the dipole-allowed excitations in the region of visible light. Furthermore, reactive MD simulations were carried out to simulate the reversible processes of o -R-substituted 4,4-diacetamidoazobenzene derivatives (R=F, Cl, Br or I), and 46%–86% trans isomers were demonstrated to translate to cis ones, which is in agreement with the experimental data.

Abstract:The opto-mechanical properties of a photo-responsive nematic polymer network (PRPN) are investigated using molecular dynamics simulation. For the implementation of the trans-to-cis isomerization of azo compounds, a switchable potential formalism for the N = N bond is applied to the crosslinked PRPN unit cell model. During the light switch-on and heating-up simulations at a wide range of temperatures, the scalar orientational order parameter for the mesogenic side group molecules, the effective photo-induced strain of the bulk polymer network, and the opto-mechanical properties are characterized. The correlation between the microstate which belongs to the molecular location and the macroscopically observed photostrain is identified according to the isomerization ratio of the diazene groups.

Abstract:The arylazopyrazole 3pzH as a novel photoswitch exhibits quantitative switching and high thermal stability. In this work, combined electronic structure calculations and ab initio multiple spawning (AIMS) dynamic simulations were performed to systemically investigate the cis ↔ trans photoisomerization mechanism and the chiral preference after photoexcitation of 3pzH to the first excited singlet state (S1). Unlike most of the azoheteroarene photoswitches reported previously, many twisted and T-shaped cis isomers were found to be stable for 3pzH in the S0 state, owing to the moderate interaction between the hydrogen atom and π electrons of the aromatic ring. Two twisted cis isomers with different chirality ((M)-Z1 and (P)-Z1), the most stable T-shaped cis isomer ((T)-Z2), and the most stable planar trans isomer (E2) were selected as the initial structures to carry out the AIMS nonadiabatic dynamic simulations. Following excitation to the S1 state, all of the cis isomers decayed to conical intersection (CI) regions via the same bicycle pedal mechanism, while the evolution of the trans isomers to their CI regions was achieved via rotation around the N═N bond. More importantly, chiral preferences were found for the twisted cis isomers in the S1 state through the AIMS dynamic simulations due to the steric effect and static electronic repulsion. Notably, chirality was also observed in S1 isomerization starting from the planar E2 isomer because of the dynamic effect. After the nonadiabatic transition to the S0 state, the bicycle pedal mechanism was found to play a crucial role in cis ↔ trans photoisomerization. The simulated photoisomerization productivities were generally consistent with past experimental observations. Our calculations not only uncover the underlying reason for the excellent photoswitching properties of 3pzH but also enrich the knowledge of photoisomerization for azoheteroarene photoswitches, which will surely benefit their rational design.

Photoisomerization dynamics and pathways of trans- and cis-azobenzene in solution from broadband femtosecond spectroscopies and calculations

Abstract:The photoisomerization of azobenzene in solution was studied experimentally and by calculations. trans-to-cis and cis-to-trans dynamics are described through broadband transient absorption, fluorescence, and stimulated Raman spectroscopy. Transient absorption was extended to cover not only the nπ* band but also the ππ* band in the ultraviolet. Isomerization yields are used for a quantitative comparison of trans and cis transient spectra under different excitation. For the trans-to-cis path upon nπ*(S(1)) excitation, the evolution develops with 0.3, 3, and 16 ps. The first two times reflect population relaxation to a local minimum S(1t )(L) and subsequent transition to a dark intermediate S(1t)(D) over an 8 kJ/mol barrier. The existence of stationary points S(1t)(L) and S(1t)(D) is confirmed by quantum-chemical calculations. The third time corresponds to S(1t) (D) → S0 relaxation to the ground state via an S1/S0 conical intersection over a 12 kJ/mol barrier. Thus, the 16 ps time constant is attributed to the isomerization process and not to vibrational cooling, contrary to the current view and in line with the previous interpretation by Lednev et al. (J. Phys. Chem. 1996, 100, 13338). The decay of the long-lived intermediate S(1t)(D) is consistent with the hula twist rather than with the inversion mechanism. For the cis-totrans reaction following nπ* excitation, signal decay is strongly nonexponential, with 0.1 and 1 ps. The latter (1 ps) is much shorter than the 16 ps decay of the trans isomer, implying different S1/S0 conical intersections and relaxation paths for the cis-totrans and trans-to-cis reaction. New results are also obtained with ππ*(Sn) excitation. Thus, for trans-azobenzene, 50% of the population relaxes to an S1 region, which is not accessible under nπ* excitation. For cis-azobenzene, up to 30% of the excited species isomerize to trans via an Sn/S1 intersection, resulting in a mixed cis/trans S1 population. The isomerization kinetics of azobenzene shows no viscosity dependence, putting into question the torsion mechanism and suggesting the hula-twist isomerization mechanism.
Bis‐azopyrazole Photoswitches for Efficient Solar Light Harvesting

Abstract:Although natural sunlight is one of the most abundant and sustainable energy resources, only a fraction of its energy is currently harnessed and utilized in photoactive systems. The development of molecular photoswitches that can be directly activated by sunlight is imperative for unlocking the full potential of solar energy and addressing the growing energy demands. Herein, we designed a series of 2-amino-1,3-bis-azopyrazoles that features a coupled πn system, resulting in a pronounced redshift in its spectral absorption, reaching up to 661 nm in the red region. By varying the amino substituents of these molecules, highly efficient E→Z photoisomerization under unfiltered sunlight can be achieved, with yields of up to 88.4%. Moreover, the Z,Z-isomers have high thermal stability with half-lives from days to years at room temperature. The introduction of ortho-amino substitutions and meta-bisazo units leads to a reversal of the n–π* and πn–π* transitions on the energy scale. This change provides a new perspective for further tuning the visible absorption of azo-switches by utilizing the πn–π* band instead of the conventional n–π* band. These results suggest that photoresponsive systems can be powered by sunlight instead of traditional artificial lights, thereby paving the way for sustainable smart materials and devices.

Designing a visible light-mediated double photoswitch: A combination of biradical and azobenzene structural motifs that can be switched independently

Abstract:A new molecular switch is presented that combines both biradical and azobenzene motifs to perform visible light-induced constitutional and stereo-isomerisation within the same molecule. The insertion of isonitrile-functionalized azobenzenes into the four-membered biradical [•P(μ-NTer)2P•] (1), yielding a phosphorus-centred cyclopentane-1,3-diyl (E-4B and E-5B), represents a straightforward method to generate the desired double switches (E-4B and E-5B) in excellent yields (>90%). The switching properties are demonstrated for the fluorinated species E-5B and, interestingly, can occur either stepwise or simultaneously, depending on the order in which the sample is irradiated with red and/or green light. All possible isomerisation reactions, i.e. housane formation in the phosphorus-centred cyclopentane-1,3-diyl fragment and E/Z isomerisation at the azobenzene, can be switched by irradiation and the reaction products E-5H, Z-5H and Z-5B (when performing the thermal reverse reaction in the dark) are identified using 19F{1H} and 31P{1H} NMR spectroscopy. Results from quantum chemical calculations contribute to the understanding and visualization of the different isomers of each of the observed compounds (E-5B, E-5H, Z-5H, and Z-5B) caused by the unique structure of the double switches.

Abstract:Light-triggered molecular switches are extensively researched for their applications in medicine, chemistry and material science and, if combined, particularly for their use in multifunctional smart materials, for which orthogonally, i.e. individually, addressable photoswitches are needed. Within this study, we compare the performance of the pure photoswitches, namely an azobenzene derivative (Azo) and a donor-acceptor Stenhouse adduct (DASA), with the switching properties of their mixture using time-resolved temperature-dependent UV/VIS absorption spectroscopy, time-resolved IR absorption spectroscopy at room temperature and quantum mechanical calculations to determine effective cross sections, switching kinetics as well as activation energies of thermally induced steps. We find slightly improved effective cross sections, percentages of switched molecules and no increased activation barriers of the equimolar mixture compared to the single compounds. Thus, the studied mixture Azo+DASA is very promising for future applications in multifunctional smart materials.

Abstract:The strong coupling between molecules and photons in resonant cavities offers a new toolbox to manipulate photochemical reactions. While the quenching of photochemical reactions in the strong coupling regime has been demonstrated before, their enhancement has proven to be more elusive. By means of a state-of-the-art approach, here we show how the \iso{trans}{cis} photoisomerization quantum yield of azobenzene embedded in a realistic environment can be higher in polaritonic conditions than in the cavity-free case. We characterize the mechanism leading to such enhancement and discuss the conditions to push the photostationary state towards the unfavoured reaction product. Our results provide a signature that the control of photochemical reactions through strong coupling can be extended from selective quenching to improvement of the quantum yields
