Abstract:The deposition, dissipation, and metabolism of difenoconazole as well as reduced application dose were studied in the open field and greenhouse pepper system. The deposition amount of difenoconazole gradually increased with the increasing dosages, which followed an order with upper leaf > lower leaf > upper stem > lower stem > cultivated soil > fruit. The dissipation half-lives of difenoconazole in different parts of pepper plant and cultivated soil were higher in the greenhouse than those in the open field. Difenoconazole was transformed to metabolite I (C16H13Cl2N3O2) in pepper plant, but metabolite II (C13H14ClN3O3) in cultivated soil. Additionally, the field control efficacy of difenoconazole at the 0.75-fold minimum recommended dosage could meet the basic control requirement after the first application for 7 d. The results revealed deposition distribution and metabolism characteristics of difenoconazole and the application dose may be effectively decreased in the open field and greenhouse pepper-planted ecosystem.

Abstract:The initial deposition amount, dissipation dynamics, retention rate, and field control efficacy of difenoconazole in pepper-soil system were studied with different application dosages, planting regions and patterns. The initial deposition amount of difenoconazole under the same application dosage showed the following order: fruits < cultivated soils < lower stems < upper stems < lower leaves < upper leaves, open field < greenhouse, and Changjiang < Cixi < Hefei < Langfang, respectively, which increased with increasing application dosage. The dissipation rates in leaves, stems, fruits and cultivated soils exhibited an initially fast and then slow trend, while the retention rates displayed a tendency of first increasing and then stabilizing with increasing application dosages. After 7 d of difenoconazole application, the retention rates at five concentrations were 10.3%-39.1%, and the field efficacy mostly reached the minimum effective dose. These results suggested that difenoconazole could be reduced by 25% based on the minimum recommended dose meeting the requirements of field control efficacy for controlling pepper anthracnose.

Abstract:Triazole-resistance in Aspergillus fumigatus has been frequently found in agricultural soils. We determined if triazole-resistance in A. fumigatus can be acquired in soil and earthworm guts after exposure to environmentally relevant concentrations of formulated propiconazole (FPRO) and difenoconazole (FDIF). Acquired triazole-resistance was found in two strains of A. fumigatus after exposure to 2.5 mg/L of FPRO and three strains of A. fumigatus after induction by 0.1–2.5 mg/L of FDIF for 40 d in liquid medium. Resistance development depended upon exposure time and substrate concentration. There were one, three, and two resistant strains of A. fumigatus in soils receiving FPRO, FDIF, and their combination, respectively. Resistance was identified in two strains of A. fumigatus isolated from the guts of earthworms exposed in soils contaminated by FDIF and two strains of A. fumigatus isolated from the guts of earthworms exposed to the combination of FDIF and FPRO. We conclude that normal applications of FPRO, FDIF, and their combination are likely to induce triazole-resistance in A. fumigatus in agricultural fields. Earthworms may act as disseminators of triazole-resistant A. fumigatus.
Reduced Bacterial Network Complexity in Agricultural Soils after Application of the Neonicotinoid Insecticide Thiamethoxam.

Abstract:Pesticides may alter soil microbial community structure or diversity, but their impact on microbial co-occurrence patterns remains unclear. Here, the effect of the widely used neonicotinoid insecticide thiamethoxam on the bacterial community in five arable soils was deciphered using the 16S rRNA gene amplicon sequencing technique. The degradation half-life of thiamethoxam in nonsterilized soils was significantly lower than that in sterilized soils, suggesting a considerable contribution from biodegradation. Soil bacterial community diversity diminished in high concentration thiamethoxam treatment and its impact varied with treatment concentration and soil type. Bacterial co-occurrence network complexity significantly decreased after exposure to thiamethoxam. Under thiamethoxam stress, the relative changes in bacterial co-occurrence networks were closely related (the majority of p-values < 0.05) to the soil physicochemical properties, yet the diversity and dominant phyla were slightly related (the majority of p-values > 0.05). Additionally, three bacterial genera, Sphingomonas, Streptomyces, and Catenulispora, were identified to be relevant to the degradation of thiamethoxam in soils. This finding deciphers the succession of the bacterial community under thiamethoxam stress across multiple soils, and emphasizes the potential role of physicochemical properties in regulating the ecotoxicological effect of pesticides on the soil microbiome.

Abstract:Triazole resistance in Aspergillus fumigatus is a growing public health concern. In addition to its emergence in the therapy of invasive aspergillosis by triazole medicines, it has been frequently detected in agricultural fields all over the world. Here, we explore the potential link between residues of azole fungicides with similar chemical structure to triazole medicines in soil and the emergence of resistant A. fumigatus (RAF) through 855 500 km(2) monitoring survey in Eastern China covering 6 provinces. In total, 67.3%, 15.2%, 12.3%, 2.9%, 1.5%, 0.4%, and 0.3% of the soil samples contained these five fungicides (tebuconazole, difenoconazole, propiconazole, hexaconazole, and prochloraz) of 0-100, 100-200, 200-400, 400-600, 600-800, 800-1000, and >1000 ng/g, respectively. The fractions of samples containing RAF isolates were 2.4%, 5.2%, 6.4%, 7.7%, 7.4%, 14.3%, and 20.0% of the samples with total azole fungicide residues of 0-100, 100-200, 200-400, 400-600, 600-800, 800-1000, and >1000 ng/g, respectively. We find that the prevalence of RAFs is positively (P < 0.0001) correlated with residual levels of azole fungicides in soils. Our results suggest that the use of azole fungicides in agriculture should be minimized and the intervals between treatments expanded to reduce the selective pressure toward the development of resistance in A. fumigatus in agricultural fields.

Abstract:Propiconazole (PPC) and difenoconazole (DFC) are often combined for field applications. The resulted co-exsistence of PPC and DFC may have an effect on the fate of their individuals in soil. In this study, adsorption, desorption and leaching of PPC and DFC alone and their combinations were investigated in five different soils. Adsorption of PPC and DFC was significantly different on each soil with the Freundlich adsorption coefficients of 2.86–28.69 and 14.86–98.93 negatively correlated with soil pH, respectively. In addition, adsorption of PPC and DFC was declined by 27.12–37.59% and 17.28–25.35% with the presence of coexisting DFC and PPC, respectively. Mobility of PPC and DFC in tested soils was enlarged in coexisting system. The results indicate that adsorption, desorption and mobility of PPC and DFC were mainly affected by soil pH, and these behaviors of individual PPC and DFC were obviously altered by their co-existence.

Abstract:Degradation dynamics of carbendazim in soil and its effects on community diversity of soil microorganisms were investigated in a microcosm mimicking soil ecosystem.Half-life of carbendazim in soil was measured to be 8.6 and 6.8 d at levels of 2.0 mg kg-1 and 4.0 mg kg-1,respectively.Values of average well color development(AWCD) of soil microorganisms were significantly inhibited within the initial periods after treatments of carbendazim at levels of 2.0 mg kg-1 and 4.0 mg kg-1 soil.Meanwhile,significant reduction in the richness,evenness and superiority of soil microorganisms was also observed,which indicated that the inhibitory effect of carbendazim on soil microorganisms was increased with its concentration in soil.After the initial variation,soil microorganisms adapted gradually to carbendazim and recovered to levels similar to that in the control without carbendazim.

Abstract:The fungicide difenoconazole has been frequently detected in agricultural products, soils and surface waters, causing increasingly public concern due to toxicological properties. Although systemic triazole fungicides can inhibit the enzymatic activity of many CYP450 isozymes, how difenoconazole affects the human CYP3A4 still remains largely unknown. We thus investigated the effect of difenoconazole on normal mRNA expression and protein expression of human CYP3A4 by real-time quantitative PCR and western blot, respectively. Results show that the exposure to difenoconazole from 0.01 to 0.5 μM for 24 h down-regulates mRNA expression levels of human CYP3A4 in HepG2 cells. We also found that difenoconazole could inhibit the enzymatic activity of human CYP3A4 in a concentration-dependent mode. The IC50 of difenoconazole for inhibition of CYP3A4 activity is 0.41 µM, showing a stronger inhibitor in comparison with ketoconazole. Overall, our findings indicate the potential risk of difenoconazole for the disruption of human CYP3A4.

Abstract:The dissipation of carbendazim and chloramphenicol alone and in combination and their effects on soil fungal:bacterial ratios and soil enzyme activities were investigated. The results revealed that carbendazim dissipation was little affected by chloramphenicol, whereas chloramphenicol dissipation was found to be retarded significantly by the presence of carbendazim. The inhibitory effect of carbendazim on the fungal:bacterial ratios was increased by the presence of chloramphenicol, and the inhibitory effect of chloramphenicol on neutral phosphatase was increased by the presence of carbendazim. Carbendazim increased soil catalase and urease activities, but this increase was partially diminished by the presence of chloramphenicol. Little interaction was observed between carbendazim and chloramphenicol with regard to their influence on soil invertase. The results obtained in this study suggest that combinations of fungicides and antibiotics may alter the compounds’ individual behaviors in soil and their effects on soil enzymes.

Abstract:Pesticides, protect crops but can harm the environment and human health when used without caution. This study evaluated the impact of propiconazole, a fungicide that acts on fungal cell membranes, on soil microbiome abundance, diversity, and functional profile, as well as soil dehydrogenase activity (DHA). The study conducted microcosm experiments using soil samples treated with propiconazole and employed next-generation sequencing (MiSeq) and chromatographic approaches (GC–MS/MS) to analyze the shift in microbial communities and propiconazole level, respectively. The results showed that propiconazole significantly altered the distribution of microbial communities, with notable changes in the abundance of various bacterial and fungal taxa. Among soil bacterial communities, the relative abundance of Proteobacteria and Planctomycetota increased, while that of Acidobacteria decreased after propiconazole treatment. In the fungal communities, propiconazole increased the abundance of Ascomycota and Basidiomycota in the treated soil, while that of Mortierellomycota was reduced. Fungicide application further triggered a significant decrease in DHA over time. Analysis of the functional profile of bacterial communities showed that propiconazole significantly affected bacterial cellular and metabolic pathways. The carbon degradation pathway was upregulated, indicating the microbial detoxification of the contaminant in the treated soil. Our findings suggest that propiconazole application has a discernible impact on soil microbial communities, which could have long-term consequences for soil health, quality, and function.

Abstract:Rhizosphere soil microorganisms have great agricultural importance. To explore the relationship between rhizosphere microorganisms and the disease incidence, and to optimize the concentration of difenoconazole fungicide for the control of wheat dwarf bunt, caused by Tilletia controversa Kühn, the rhizosphere microorganisms were characterized based on sequencing methods. We found that the disease incidence correlated with the relative abundance of some microbial communities, such as Acidobacteria, Nocardioides, Roseiflexaceae, Pyrinomonadaceae, and Gemmatimonadaceae. Actinobacteria showed significant differences in the infected soils when compared to the control soils, and the relative abundance of Acidobacteria, Pyrinomonadaceae, Gemmatimonadaceae, and Saccharimonadales populations was distinctly higher in the T. controversa -inoculated group than in the control group. The members of Dehalococcoidia, Nitrosomonadaceae, and Thermomicrobiales were found only in T. controversa -inoculated soils, and these taxa may have potential effects against the pathogen and contribute to disease control of wheat dwarf bunt. In addition, for T. controversa -infected plants, the soil treated with difenoconazole showed a high relative abundance of Proteobacteria, Actinobacteria, Ascomycota, Basidiomycota, Mortierellomycota, and Olpidiomycota based on the heatmap analysis and ANOVA. Our findings suggest that the optimized concentration of fungicide (5% recommended difenoconazole) exhibits better control efficiency and constant diversity in the rhizosphere soil.

Abstract:The lethal factor of Bacillus anthracis is central to the pathogenesis of anthrax. Its mechanism of action is still unknown. Recently, on the basis of sequence similarities, we suggested that lethal factor might act similarly to leukotriene A4 hydrolase (LTA4), a bifunctional enzyme also endowed with a metallopeptidase activity. Here we show that some inhibitors of the LTA4 hydrolase and metallopeptidase activities of LTA4 hydrolase also affect the cytotoxicity of the anthrax lethal factor on macrophage cell lines, without interfering with the ability of the lethal factor to enter cells. These results support the proposal that anthrax lethal factor might display in the cytosol of intoxicated cells a peptidase activity similar to that of LTA4 hydrolase.
The widespread presence of triazole fungicides in greenhouse soils in Shandong Province, China: A systematic study on human health and ecological risk assessments

Abstract:Triazole fungicides (TFs) are extensively used on greenhouse vegetables and are ubiquitously detected in the environment. However, the human health and ecological risks associated with the presence of TFs in the soil are unclear. In this study, ten widely used TFs were measured in 283 soil samples from vegetable greenhouses across Shandong Province, China, and their potential human health and ecological risks were assessed. Among all soil samples, difenoconazole, myclobutanil, triadimenol, and tebuconazole were the top detected TFs, with detection rates of 85.2–100%; these TFs had higher residues, with average concentrations of 5.47–23.8 μg/kg. Although most of the detectable TFs were present in low amounts, 99.3% of the samples were contaminated with 2–10 TFs. Human health risk assessment based on hazard quotient (HQ) and hazard index (HI) values indicated that TFs posed negligible non-cancer risks for both adults and children (HQ range, 5.33 × 10 −10 to 2.38 × 10 −5 ; HI range, 1.95 × 10 −9 to 3.05 × 10 −5 , <1). Ecological risk assessment based on the toxicity exposure ratio (TER) and risk quotient (RQ) values indicated that difenoconazole was a potential risk factor for soil organisms (TER max = 1 for Eisenia foetida , <5; RQ mean = 1.19 and RQ max = 9.04, >1). Moreover, 84 of the 283 sites showed a high risk (RQ site range, 1.09–9.08, >1), and difenoconazole was the primary contributor to the overall risk. Considering their ubiquity and potential hazards, TFs should be continuously assessed and prioritized for pesticide risk management.

Sub-Chronic Difenoconazole Exposure Induced Gut Microbiota Dysbiosis in Mice

Abstract:Difenoconazole (DIF) is a widely separated triazole fungicide in many countries. The excessive usage of DIF increases the high volume of residues in agriculture production and water bodies. Some previous studies demonstrated the toxic effects of DIF on non-target animals, however, there were still some gaps in the knowledge of the potential hazards of DIF to mammals and human health. Herein, 7-week-old male mice were exposed to 30 and 100 mg/kg/day DIF for 14 and 56 days. We observed that 56 days of DIF exposure decreased the colonic mucus expression of alcin blue-periodic acid-schiff (AB-PAS) stain and the immunochemical stain of muc2 protein. The transcript levels of mucin protein (muc1, muc2 and muc3) decreased significantly in the gut of mice followed 56 days of 100 mg/kg/day DIF exposure. In addition, the gut microbiota composition was also affected after 14 or 56 days of DIF exposure. Although the mucus expression after 14 days of DIF exposure only decreased slightly, the gut microbiota composition compared with the control group was changed significantly. Moreover, the DIF-30 and DIF-100 caused respectively different changes on the gut microbiota. The relative abundance of Bacteroidetes decreased significantly after 14 days and 56 days of DIF exposure. After 14 days of DIF exposure, there were 35 and 18 differential genera in the DIF-30 and DIF-100 group, respectively. There were 25 and 32 differential genera in the DIF-30 and DIF-100 group after 56 days of exposure, respectively. Meanwhile, the alpha diversity indexes, including observed species, Shannon, Simpson, Chao1 and ACE, in gut microbiota decreased significantly after 56 days of DIF exposure. Interestingly, the relative abundance of Akkermansia increased significantly after 56 days of 100 mg/kg/d DIF exposure. Although Akkermansia was considered as one probiotic, the phenomenon of dramatic Akkermansia increase with the decrease in gut microbiota diversity needed further discussion. These results provided some new insights on how DIF exposure impacts the mucus barrier and induces gut microbiota dysbiosis.

Abstract:The toxicological study of pesticides at sub-lethal and environment-relevant concentrations has become increasingly crucial for human and environmental health. Toxic mechanisms of agrochemicals contribute to discovering green pesticides, assessing the hazards of pesticides comprehensively, and supporting legitimate regulatory decisions. However, the toxicological effects of difenoconazole are not yet fully understood despite being frequently detected in fruits, vegetables, waters, and soils and posing hazards to humans and the environment. This lack of knowledge could lead to flawed risk assessment and administrative oversight. Thus, the review aimed to provide some investigation perspectives for clarifying the toxicological effects of difenoconazole by synthesizing the toxic data of difenoconazole on various organisms, such as bees, Daphnia magna , fish, earthworms, mammals, and plants and summarizing the toxicological mechanisms of difenoconazole, especially in fish and mammals from peer-reviewed publications. Evidence revealed that difenoconazole caused multiple toxicological effects, including developmental toxicity, reproductive toxicity, endocrine disruption effects, neurotoxicity, and transgenerational toxicity. The toxic mechanisms involved in metabolic disturbance, oxidative stress, inflammation, apoptosis, and autophagy by activating reactive oxygen species-mediated signaling pathways and mitochondrial apoptosis routes, disturbing amino acids, lipid, and nucleotide metabolism, and regulating gene transcription and expression in mammals and fish. Based on the review, further studies better focus on the toxic differences of difenoconazole stereoisomers, the toxicological effects of transformation products of difenoconazole, and the mechanism of action of difenoconazole on sex-specific endocrine disruption effects, intestinal damage, and gut dysbacteriosis for its hazard assessment and management synthetically.

Abstract:The enantioselective environmental behavior of difenoconazole, a widely utilized triazole fungicide commonly detected in agricultural soils, has yet to be comprehensively explored within the earthworm-soil system. To address this research gap, we investigated the bioaccumulation and elimination kinetics, degradation pathways, biotransformation mechanisms, spatial distribution, and toxicity of chiral difenoconazole. The four stereoisomers of difenoconazole were baseline separated and analyzed using SFC-MS/MS. Pronounced enantioselectivity was observed during the uptake phase, with earthworms exhibiting a preference for (2 R ,4 R )-difenoconazole and (2 R ,4 S )-difenoconazole. A total of five transformation products (TPs) were detected and identified using UHPLC-QTOF/MS in the earthworm-soil system. Four of the TPs were detected in both earthworm and soil, and one TP was produced only in eaerthwroms. Hydrolysis and hydroxylation were the primary transformation pathways of difenoconazole in both earthworms and soil. Furthermore, a chiral TP, 3-chloro, 4-hydroxy difenoconazole, was generated with significant enantioselectivity, and molecular docking results indicate the greater catalytic bioactivity of (2 R ,4 R )- and (2 R ,4 S )-difenoconazole, leading to the preferential formation of their corresponding hydroxylated TPs. Furthermore, Mass Spectrometry Imaging (MSI) was applied for the first time to explore the spatial distribution of difenoconazole and the TPs in earthworms, and the "secretory zone" was found to be the dominant region to uptake and biodegrade difenoconazole. ECOSAR predictions highlighted the potentially hazardous impact of most difenoconazole TPs on aquatic ecosystems. These findings are important for understanding the environmental fate of difenoconazole, evaluating environmental risks, and offering valuable insights for guiding scientific bioremediation efforts.

Abstract:Fungicides are used worldwide to improve crop yields, but they can affect non-target soil microorganisms which are essential for ecosystem functioning. Microorganisms form complex communities characterized by a myriad of interspecies interactions, yet it remains unclear to what extent non-target microorganisms are indirectly affected by fungicides through biotic interactions with sensitive taxa. To quantify such indirect effects, we fragmented a soil microbial community by filtration to alter biotic interactions and compared the effect of the fungicide hymexazol between fractions in soil microcosms. We postulated that OTUs which are indirectly affected would exhibit a different response to the fungicide across the fragmented communities. We found that hymexazol primarily affected bacterial and fungal communities through indirect effects, which were responsible for more than 75% of the shifts in relative abundance of the dominant microbial OTUs after exposure to an agronomic dose of hymexazol. However, these indirect effects decreased for the bacterial community when hymexazol doses increased. Our results also suggest that N-cycling processes such as ammonia oxidation can be impacted indirectly by fungicide application. This work sheds light on the indirect impact of fungicide exposure on soil microorganisms through biotic interactions, which underscores the need for higher-tier risk assessment. Environmental Implication In this study, we used a novel approach based on the fragmentation of the soil microbial community to determine to which extent fungicide application could indirectly affect fungi and bacteria through biotic interactions. To assess off-target effects of fungicide on soil microorganisms, we selected hymexazol, which is used worldwide to control a variety of fungal plant pathogens, and exposed arable soil to the recommended field rate, as well as to higher rates. Our findings show that at least 75% of hymexazol-impacted microbial OTUs were indirectly affected, therefore emphasizing the importance of tiered risk assessment.
