Abstract:Introduction: Gastric cancer is the second most common cause of cancer-related mortality worldwide. 5-fluorouracil (5-FU) is a commonly used anti-cancer drug. Various polyunsaturated fatty acids (PUFAs) are known to have tumoricidal action both in vitro and in vivo. Though PUFAs are known to augment the cytotoxic action of anti-cancer drugs, the exact mechanism is not clear.Material and methods: The human gastric cancer cell line MGC (undifferentiated) and human gastric cancer cell line SGC (semi-differentiated) were either 5-FU alone or a combination of 5-FU + PUFAs and their proliferation, and ability to secrete tumor necrosis factor-alpha (TNF-alpha) and vascular endothelial growth factor (VEGF) and lipid metabolism-related factors lipoprotein lipase (LPL), peroxisome proliferator-activated-gamma (PPAR-gamma), and CCAAT enhancer-binding protein (C/EBP) were investigated and analyzed.Results: It was noted that combined treatment of 5-FU + PUFAs on gastric carcinoma (MGC and SGC) cells produced a significant growth inhibitory action compared with either agent alone by inhibiting the production of TNF-alpha and VEGF and a simultaneous increase in the expression of LPL, PPAR-gamma, and C/EBP.Conclusions: Based on the results of the present study, it is concluded that PUFAs enhance the tumoricidal action of the anti-cancer drug 5-FU by acting on anti-angiogenic factors and enzymes involved in lipid metabolism.
Experimental Study of Euongmus Mupinensis Alkaloid on the Inhibition of Liver Cancer Cells

Abstract:OBJECTIVE To study the inhibition of Euongmus mupinensis alkaloid on liver cancer cells in vitro.METHODS Use the method of lactate dehydrogenase releasing to measure the toxic action of Euongmus mupinensis alkaloid, 5-fluorouracil and cyclophosphamide on liver cancer in vitro.RESULTS Euongmus mupinensis alkaloid can inhibit the growth of the tumor and the effect is improved when the concentration is increasing. The effect is similar to those of 5-fluorouracil and cyclophos phamide.CONCLUSION Euongmus mupinensis alkaloid can inhibit the growth of the tumor and it is promising that it may be an anti-cancer drug for clinical therapy.
5-Fluorouracil Nanoparticles Inhibit Hepatocellular Carcinoma Via Activation of the P53 Pathway in the Orthotopic Transplant Mouse Model

Abstract:Biodegradable polymer nanoparticle drug delivery systems provide targeted drug delivery, improved pharmacokinetic and biodistribution, enhanced drug stability and fewer side effects. These drug delivery systems are widely used for delivering cytotoxic agents. In the present study, we synthesized GC/5-FU nanoparticles by combining galactosylated chitosan (GC) material with 5-FU, and tested its effect on liver cancer in vitro and in vivo. The in vitro anti-cancer effects of this sustained release system were both dose- and time-dependent, and demonstrated higher cytotoxicity against hepatic cancer cells than against other cell types. The distribution of GC/5-FU in vivo revealed the greatest accumulation in hepatic cancer tissues. GC/5-FU significantly inhibited tumor growth in an orthotropic liver cancer mouse model, resulting in a significant reduction in tumor weight and increased survival time in comparison to 5-FU alone. Flow cytometry and TUNEL assays in hepatic cancer cells showed that GC/5-FU was associated with higher rates of G0-G1 arrest and apoptosis than 5-FU. Analysis of apoptosis pathways indicated that GC/5-FU upregulates p53 expression at both protein and mRNA levels. This in turn lowers Bcl-2/Bax expression resulting in mitochondrial release of cytochrome C into the cytosol with subsequent caspase-3 activation. Upregulation of caspase-3 expression decreased poly ADP-ribose polymerase 1 (PARP-1) at mRNA and protein levels, further promoting apoptosis. These findings indicate that sustained release of GC/5-FU nanoparticles are more effective at targeting hepatic cancer cells than 5-FU monotherapy in the mouse orthotropic liver cancer mouse model.
Andrographolide Enhances 5-Fluorouracil-induced Apoptosis Via Caspase-8-dependent Mitochondrial Pathway Involving P53 Participation in Hepatocellular Carcinoma (SMMC-7721) Cells.

Abstract:Despite recent significant advances in the treatment of human carcinoma (HCC), the results of chemotherapy to date remain unsatisfactory. 5-Fluorouracil (5-FU) still represents the cornerstone of treatment of carcinoma, and resistance to the actions of 5-FU is a major obstacle to successful chemotherapy. More effective treatment strategies may involve combinations of agents with activity against HCC. Andrographolide (ANDRO), a natural bicyclic diterpenoid lactone isolated from Andrographis paniculata, has been shown to suppress the growth of HCC cells and trigger apoptosis in vitro. To assess the suitability of ANDRO as a chemotherapeutic agent in HCC, its cytotoxic effects have been evaluated both as a single agent and in combination with 5-FU. ANDRO potentiates the cytotoxic effect of 5-FU in HCC cell line SMMC-7721 through apoptosis. ANDRO alone induces SMMC-7721 apoptosis with p53 expression, Bax conformation and caspase-3,8,9 activation. Surprisingly, the addition of ANDRO to 5-FU induces synergistic apoptosis, which could be corroborated to the increased caspase-8, p53 activity and the significant changes of Bax conformation in these cells, resulting in increased losses of mitochondrial membrane potential, increased release of cytochrome c, and activation of caspase-9 and caspase-3. Suppression of caspase-8 with the specific inhibitor z-IETD-fmk abrogates largely ANDRO/5-FU biological activity by preventing mitochondrial membrane potential disappearance, caspase-3,9 activation and subsequent apoptosis. The results suggest that ANDRO may be effective in combination with 5-FU for the treatment of HCC cells SMMC-7721.

Abstract:The uhp gene, which specifies the uptake of hexose phosphates, and several other genes in the vicinity of minute 81 on the E. coli linkage map have been located by phage-mediated transductions. The order found is mtl-gpsA-pyre-gltc-uhp-tna-dnaa. Alleles specifying the Uhp- and Uhp+ characters were separated from that specifying constitutivity of hexose phosphate uptake (Uhpc). Although cotransduction frequencies between gltC and uhp as high as 90%, and between uhp and tna as high as 80%, were observed, these frequencies were unusually strongly dependent on which marker was selected. This may be due to the proximity of the uhp region to the point of origin of chromosome replication.
FNDC5 induces M2 macrophage polarization and promotes hepatocellular carcinoma cell growth by affecting the PPARγ/NF-κB/NLRP3 pathway

Abstract:PURPOSE: The purpose of this study was to investigate the effect of FNDC5 expression levels in hepatocellular carcinoma on the phenotypic changes of macrophages in tumor tissues.METHODS: In this study, we established an in vitro co-culture system of hepatocellular carcinoma cells and macrophages. Then we performed overexpression or knockdown of FNDC5 gene in hepatocellular carcinoma cells to observe the effect of changes in FNDC5 expression level on the phenotypic changes of THP-1 macrophages. And the conclusions obtained in the in vitro assay were further validated by a subcutaneous tumorigenic nude mice model.RESULTS: Our findings suggest that elevated FNDC5 expression in hepatocellular carcinoma cells lead to an increased M2 phenotype and decreased M1 phenotype in macrophages. This effect may be achieved by elevating PPARγ levels in macrophages while decreasing NF-κB and NLRP3 levels. These changes could be reversed by using PPARγ inhibitors.CONCLUSION: We preliminarily demonstrated that FNDC5 in hepatocellular carcinoma cells promotes the polarization of M2 macrophages by affecting the PPARγ/NF-κB/NLRP3 pathway.

Relationship Between Tumor Associated Macrophages and Cytotoxic T Lymphocytes in Rat Liver Carcinoma

Abstract:Objective To investigate the influence of tumor associated macrophages on cytotoxic T lymphocytes (CTL) infiltration and apoptosis in rat liver carcinoma.Methods Walker-256 Wistar rat hepatoma models were established.Then rats were randomly divided into three groups:group A (clodronatelipsomes by intraperitoneal injection) ; group B (PBS-lipsomes by intraperitoneal injection) ; group C (normal saline by intraperitoneal injection).Animals were killed at the 12th day after treatment.The tumor volume was measured,and the number of positive cells of CD68,CD8,CD163,and Granzyme B was detected by using immunohistochemical method,respectively.The apoptosis of CD8 + T cells in tumor tissues was tested by the method of double staining with immunohistochemistry and TUNEL.Results ( 1 ) There was no significant difference in tumor volume among groups A,B and C ( P ＞ 0.05 ) ; (2) There was significant difference in CD68 positiv cells between groups A and B,CDE163 positive cells between groups A and B,CD8 positive cells between group A and B or between groups A and C; and Granzyme B positive cells between groups A and C (all P ＜0.05).The number of CD68 and CD163 positive cells in group A was less than in groups B and C.The number of CD8 and Granzyme B positive cells in group A was more than in groups B and C ; (3) There was a negative correlation not only between the positive cells of CD68 and CD8,but also between CD163 and Granzyme B positive cells.However,there was no correlation between the positive cells of CD68 and Granzyme B,as well as between the positive cells of CD163 and CD8 ;(4) There was significant difference in apoptosis rate of CD8 T cells between groups A and C group (P ＜0.05).Apoptosis rate of CD8 T cells in group A was lower than in groups B and C.Conclusion Clodronate liposome can deplete macrophages of rat liver carcinoma tissues effectively.After macrophages were depleted,the number of CTL was increased and the apoptosis rate of CTL was decreased in rat liver carcinoma.Tumor associated macrophages can promote apoptosis and inhibit killing activity of cytotoxicity T lymphocyte,and play an important role in tumor development.

Abstract:Abstract Background: Sorafenib prolonged survival in patients with unresectable hepatocellular carcinoma (HCC), but the effect is modest and tumor finally progressed even under continuous sorafenib therapy. The aim of this study is to investigate the mechanism of tumor progression under sorafenib treatment. Methods: Tumor growth, local invasion and lung metastasis were observed in MHCCLM3-R, a human HCC xenograft nude mouse model, when treated by sorafenib (30 mg/kg qd, n=6 per group) and vehicle as control. Macrophage infiltration was measured in the peripheral blood and macrophage in tumor after sorafenib treatment by immunohistochemistry and flow cytometry with F4/80 antibody, and the effect of macrophage depletion on tumor angiogenesis and metastasis after sorafenib treatment, using two drugs target macrophages, zoledronic acid (ZA) and clodrolip, in a mouse model of human hepatocellular carcinoma (HCC). Results: Sorafenib treatment significantly inhibited tumor growth and lung metastasis, but also induced significant increase in peripheral F4/80-positive cells and intratumoral macrophages infiltration, which was associated with elevation of colony stimulating factor-1, stromal-derived factor 1α and vascular endothelial growth factor in tumor and plasma VEGF of mouse origin in peripheral blood, suggesting the role of macrophage in tumor progression during sorafenib treatment. Depletion of macrophage by clodrolip or ZA in combination with sorafenib significantly inhibited tumor progression, tumor angiogenesis and lung metastasis compared to that of mice treated with sorafenib alone. ZA was more effective than clodrolip. Conclusions: Macrophage may have an important role in tumor progression under sorafenib treatment. M1‐like tumor‐associated macrophages enhance proliferation and anti‐apoptotic ability of liver cancer cells via activating the NF‐κB signaling pathway

Abstract:Liver cancer is the second leading cause of cancer‐associated death worldwide. The present study aimed to evaluate the infiltration of M1‐like tumor‐associated macrophages (TAMs) and explore the role of infiltration of M1‐like TAMs in the proliferation and apoptosis evasion of liver cancer cells. Furthermore, the association between M1‐like TAM and the efficacy of postoperative transcatheter arterial chemoembolization (TACE) for patients with liver cancer was investigated. The levels of CD68, human leukocyte antigen‐DR and phosphorylated NF‐κB (p‐)p65 were detected by western blot analysis and immunohistochemistry. Cell cycle analysis, MTT and clonogenic assays were utilized to investigate the proliferation of liver cancer cells. It was indicated that M1‐like TAM increased the p‐p65/p65 ratio in liver cancer cells and promoted cell proliferation. Furthermore, JSH‐23, an inhibitor that prevents p65 from entering the nucleus, decreased the proliferation of liver cancer cells in M1‐like TAM‐conditioned medium. In addition, M1‐like TAM increased the number of liver cancer cells in the S and G2/M phases of the cell cycle and also upregulated the expression levels of cyclin‐dependent kinase (CDK)1, CDK2 and cyclin D1. By contrast, M1‐like TAM decreased the expression level of p21. Through these effects, the anti‐apoptotic ability of liver cancer cells was enhanced. Of note, JSH‐23 reversed these changes related to the cell cycle, anti‐apoptotic ability and the expression levels of proteins induced by M1‐like TAM in liver cancer cells. In conclusion, the infiltration of M1‐like TAM in liver tissue negatively influenced the efficacy of postoperative TACE for patients with liver cancer.

Abstract:Dimethoxycurcumin (DMC) is a lipophilic analog of curcumin, an effective treatment for colon cancer, which has greater chemical and metabolic stability. Chemotherapy treatments, such as 5-fluorouracil (5-Fu), play a key role in the current management of colon cancer. In this study, we investigated the antitumor efficacy of DMC in combination with 5-Fu in SW480 and SW620 colon cancer cells. CCK-8 assay was used to evaluate the inhibitory effect of DMC and 5-Fu on cancer cells proliferation, and the combination index was calculated. The influence of DMC and 5-Fu on cell cycle, apoptosis, reactive oxygen species (ROS) production, and mitochondrial membrane potential in SW480 and SW620 cells was determined using flow cytometry, and the related signaling pathways were detected by western blot. Transmission electron microscopy was used to observe endoplasmic reticulum expansion. DMC- and/or 5-Fu-induced apoptosis, stimulated G0/G1 phase arrest, increased ROS levels, decreased mitochondrial membrane potential, and enhanced endoplasmic reticulum expansion. The induction of apoptosis is involved in the increasing of Bax and cytochrome c and decreasing of Bcl2 expressions. Increased production of ROS was accompanied by upregulation of CHOP and Noxa. Combination therapy of DMC and 5-Fu had increased efficacy on the above pathways compared with either drug alone. Based on the calculated IC50 , combination treatment with DMC and 5-Fu had an additive antitumor effect in both cell lines. Combined treatment with DMC and 5-Fu led to an additive antitumor effect in colon cancer cells that was related to apoptosis induction, G0/G1 phase arrest, increased ROS production, decreased mitochondrial membrane potential, and enhanced endoplasmic reticulum expansion.

Abstract:<p>Hepatic fibrosis is a common pathological process involving persistent liver injury with various etiologies and subsequent inflammatory responses that occur in chronic liver diseases. If left untreated, liver fibrosis can progress to liver cirrhosis, hepatocellular carcinoma, and eventually, liver failure. Unfortunately, to date, there is no effective treatment for liver fibrosis, with the exception of liver transplantation. Although the pathophysiology of liver fibrosis is multifactorial and includes the activation of hepatic stellate cells, which are known to drive liver fibrogenesis, hepatic macrophages have emerged as central players in the development of liver fibrosis and regression. Hepatic macrophages, which consist of resident macrophages (Kupffer cells) and monocyte‐derived macrophages, have been shown to play an intricate role in the initiation of inflammatory responses to liver injury, progression of fibrosis, and promotion of fibrosis resolution. These features have made hepatic macrophages uniquely attractive therapeutic targets in the fight against hepatic fibrosis. In this review, we synthesized the literature to highlight the functions and regulation of heterogeneity in hepatic macrophages. Furthermore, using the existing findings, we attempt to offer insights into the molecular mechanisms underlying the phenotypic switch from fibrogenic macrophages to restorative macrophages, the regulation of heterogeneity, and modes of action for hepatic macrophages. A better understanding of these mechanisms may guide the development of novel anti‐fibrotic therapies (e.g., macrophage subset‐targeted treatments) to combat liver fibrosis in the future.</p>

Effects of curcumin on 5-fluorouracil resistance of colon cancer cells through the PI3K/AKT/mTOR pathway via MACC1

Abstract:Introduction : Colon cancer (CC) represents a common malignancy of the digestive tract featured by high metastasis and recurrence rates. Despite the critical role of 5-fluorouracil (5-Fu) in tumor therapy, long-term and heavy use of 5-Fu leads to reduced treatment efficacy in CC patients. Curcumin can avert multidrug resistance as a sensitizing agent. This study sought to investigate the mechanism of curcumin in 5-Fu resistance of CC cells. Methods : Firstly, the LoVo/5-Fu CC cell line was constructed using gradient 5-Fu, and subjected to treatment with curcumin or oe-MACC1 and pathway inhibitor BEZ235. Cell proliferation was assessed by MTT and clone formation assays, and IC50 values were calculated. Cell apoptosis was evaluated by means of Annexin V-FITC/PI double staining. Levels of drug-resistance factors and Vimentin and E-cadherin were measured using RT-qPCR. Levels of PI3K/AKT/mTOR pathway-associated proteins were determined with a Western blot assay. Results : LoVo/5-Fu cells presented with increased IC50 value, enhanced proliferation, weakened apoptosis, augmented levels of drug-resistance factors and Vimentin, and diminished E-cadherin levels. MACC1 was highly-expressed and the PI3K/AKT/mTOR-pathway-associated proteins phosphorylated in LoVo/5-Fu cells. Curcumin partly averted 5-Fu resistance and decreased the ratios of p-PI3K/PI3K, p-AKT/AKT, and p-mTOR/mTOR. MACC1 over-expression mitigated the action of curcumin on inhibiting 5-Fu resistance and elevating phosphorylation levels of PI3K/AKT/mTOR proteins. BEZ235 treatment abolished the action of over-expression of MACC1 on 5-Fu resistance of CC cells. Conclusion : Our findings highlighted that curcumin suppressed the epithelial-mesenchymal transition of CC cells and thus reduced 5-Fu resistance of CC cells by regulating the PI3K/AKT/mTOR pathway via MACC1.

Abstract:Chronic inflammation is the hallmark of fibrotic disorders and is characterized by the activation of immune cells in the damaged tissues. Macrophages have emerged as central players in the fibrotic process since they initiate, sustain and amplify the inflammatory reaction. As regards the liver, distinct populations of phagocytic cells, like Kupffer cells and monocyte-derived macrophages, are indisputably key cells implicated in the pathogenesis of several chronic liver diseases. In this review, we summarize the current knowledge on the origin, role and functions of macrophages in fibrotic conditions, with a specific focus on liver fibrosis; then, we discuss some innovative therapeutic strategies targeting macrophages in fibrotic liver diseases.

Abstract:Inflammation is a hallmark of virtually all liver diseases, such as liver cancer, fibrosis, nonalcoholic steatohepatitis, alcoholic liver disease, and cholangiopathies. Liver macrophages have been thoroughly studied in human disease and mouse models, unravelling that the hepatic mononuclear phagocyte system is more versatile and complex than previously believed. Liver macrophages mainly consist of liver-resident phagocytes, or Kupffer cells (KCs), and bone marrow-derived recruited monocytes. Although both cell populations in the liver demonstrate principal functions of macrophages, such as phagocytosis, danger signal recognition, cytokine release, antigen processing, and the ability to orchestrate immune responses, KCs and recruited monocytes retain characteristic ontogeny markers and remain remarkably distinct on several functional aspects. While KCs dominate the hepatic macrophage pool in homeostasis ("sentinel function"), monocyte-derived macrophages prevail in acute or chronic injury ("emergency response team"), making them an interesting target for novel therapeutic approaches in liver disease. In addition, recent data acquired by unbiased large-scale techniques, such as single-cell RNA sequencing, unraveled a previously unrecognized complexity of human and murine macrophage polarization abilities, far beyond the old dogma of inflammatory (M1) and anti-inflammatory (M2) macrophages. Despite tremendous progress, numerous challenges remain in deciphering the full spectrum of macrophage activation and its implication in either promoting liver disease progression or repairing injured liver tissue. Being aware of such heterogeneity in cell origin and function is of crucial importance when studying liver diseases, developing novel therapeutic interventions, defining macrophage-based prognostic biomarkers, or designing clinical trials. In Vitro and Clinical Study of Association Between Macrophage and Epithelial-Mesenchymal Transition in Hepatocellular Carcinoma Cells.

Abstract:e15149 Background: Macrophages are a major component of the leukocyte infiltrate of tumors and play a pivotal role in the progression of hepatocellular carcinoma (HCC). However, the molecular mechanisms by which macrophages promote HCC invasion are poorly understood. Methods: Double-staining immunohistochemistry was used to observe the association between macrophages and epithelial–mesenchymal transition (EMT) markers in 40 clinical HCC samples. In addition, after HCC cells were cultured with macrophages for 24 hours, the effect upon HCC cell migration and invasion was assessed in vitro and the expression of EMT markers (Snail, E-Cadherin, and N-Cadherin) was investigated via western blot. The supernatant of these cultures was analyzed using a cytokine antibody array. Finally, the role of macrophage-derived IL-8 in the invasion of HCC cells was assayed by western blot, and downstream signaling pathways were also investigated. Results: In clinical HCC samples, EMT primarily occurred at the edge of the tumor nest, in which infiltrating macrophages were always observed. Double-staining immunohistochemistry indicated that CD68 expression which is the marker of macrophage was correlated with EMT marker levels in these samples. In addition, after being cultured with macrophages for 24 hours, the ability of HCC cells to migrate and invade increased, Snail and N-Cadherin expression was up-regulated, and E-Cadherin was down-regulated. An antibody array assay demonstrated that IL-8 increased significantly in the supernatant of the macrophage co-culture system. Furthermore, we found that IL-8 (1) may induce EMT and promote HCC cell migration and invasion and (2) is associated with the JAK2/STAT3/Snail signaling pathway. Conclusions: Macrophages that have infiltrated tumors may induce the EMT of HCC cells via the IL-8 activated JAK2/STAT3/Snail pathway. This finding offers a potential target for developing new HCC therapies.
Preparation of 5-Fluorouracil Nanoparticles of Galactosylated Chitosan and Its Inhibition of the Colon Cancer in Mice

Abstract:Objective:The 5-fluorouracil(5-Fu) nanoparticles of galactosylated chitosan was synthesized and the efficacy were observed in colon cancer model in vivo with 5-fluorouracil nanoparticles of galactosylated chitosan.Methods:C26 cells were cultured left lobe of the liver in mice,after tumor formation,models were treated with normal saline in NS group,empty nanoparticles in NP group,5-Fu nanoparticles in 5-Fu NP group and 5-Fu in 5-Fu group,respectively,via tail vein injection.Tumor weight were measured in the end of this experiment.The serum interferon-gamma(IFN-y) and interleukin-2(IL-2) were detected by enzyme-linked immunospot-based assays(ELISA),and natural killer(NK) cells and cytotoxic T lymphocytes(CTL) activity of mouse spleen were measured by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium(MTT) colorimetric assay. Results:Compared with NS group and NP group,the tumor weight in 5-Fu group and group was significantly lighter(P 0.01),the tumor weight decreased more obvious in 5-Fu NP group(P0.01).The levels of IFN-y and IL-2 of liver metastases of colon cancer model in mice decreased obviously in 5-Fu group than those in other groups(P0.01),however,the decrease of IFN-γand IL-2 in 5-Fu NP group was not obvious(P 0.05),compared with the saline group,while the levels in NP group was significantly higher than those in NS group(P0.01).The killing activity of NK and CTL in 5-Fu group was significantly lower than that in other groups(P0.01),however,the killing activity of NK and CTL in NP group increased compared with NS group(P0.05),and 5-Fu NP group was no lower compared with NS group(P 0.05).Conclusions: The 5-Fu nanoparticles have significant anti-tumor effect in colon cancer model,nanoparticle materials can stimulate immune function of mice and reduced the side effects of 5-Fu in immune suppression.

Abstract:Abstract Aims: To investigate the effect of colony stimulating factor 1 receptor (CSF1R) inhibitor on macrophages and tumor pathogenesis. Methods: CSF1 and CSF2 stimulated macrophages were obtained from mouse bone marrow derived monocytes cultured with CSF1 and CSF2. Mouse hepatoma cell line Hepa 1-6, and human hepatoma cell lines, HCCLM3 and HepG2 were used to established a allograft or xenograft tumor models to examine the effect of CSF1R inhibitor on tumor growth and distribution of macrophages. The characteristics of macrophages were determined by flow cytometry, immunohistochemistry, cell motility and Western blotting assays. Results: CSF1R inhibitor suppressed tumor growth in the tumor models derived from three hepatoma cell lines, and prolonged hosts’ survival, but CSF1R inhibitor did not decreased the number of macrophages in Hepa1-6 allograft tumors. in vitro study showed CSF1R inhibitor suppressed proliferation and motility of CSF1 stimulated macrophages but not CSF2 stimulated macrophages. Furthermore, we found CSF1R inhibitor treatment decreased the ability of THP-1 macrophages to promote migration and invasion of tumor cells. Conclusions: CSF1R inhibitor suppressed tumor growth in hepatoma animal models by modulating function of tumor associating macrophages. Abstract:Liver cancer is one of the leading causes of cancer-related mortality. Hepatocellular carcinoma and cholangiocarcinoma are the most common types, and despite numerous advances, therapeutic options still remain poor for these cancer patients. Tumor development and progression strictly depend on a supportive tumor microenvironment (TME). Tumor-associated macrophages (TAMs) are the most abundant immune cells population within a tumorigenic liver; they sustain cancer cells' growth and invasiveness, and their presence is correlated with a poor prognosis. Furthermore, TAM cross-talk with cells and components of the TME promotes immunosuppression, a desmoplastic response, and angiogenesis. In this review, we summarize the latest advances in understanding TAM heterogeneity and function, with a particular focus on TAM modulation of the TME. We also discuss the potential of targeting macrophage subpopulations and how this is now being exploited in current clinical trials for the treatment of liver cancer.

Abstract:Objective To study the expression of drug-resistance associated protein,such as ERK,JUK and P38,in hepatocellular carcinoma HepG2 cells treated with docosahexaenoic acid and 5-fluorouracil.Methods The expression of JNK,ERK and p38 were analyzed in HepG2 treated with 5-FU or 5-FU in combination with DHA for 48 hours by western blot.Results When compared with the group only treated with 5-FU,samples from the group treated with 5-FU and DHA showed different expression level of drugresistance associated-proteins.Down regulation of p-ERK,up regulation of p-JNK and no change of p-p38 could be found in the latter group.Conclusion With inhibiting the p-ERK and activating p-JNK,DHA could regulate hepatocellular carcinoma cells to significantly enhance the cytotoxic activity of 5-FU.So that it could play a role in the sensitization of chemotherapy drugs.

Abstract:Pancreatic ductal adenocarcinoma (PDAC) remains a clinically challenging cancer, mainly due to limited therapeutic options and the presence of a highly prominent tumor microenvironment (TME), facilitating tumor progression. The TME is predominated by heterogeneous populations of cancer-associated fibroblasts (CAFs) and tumor associated macrophages (TAMs), in constant communication with each other and with tumor cells, influencing many tumoral abilities such as therapeutic resistance. However how the crosstalk between CAFs and macrophages evolves following chemotherapeutic treatment remains poorly understood, limiting our capacity to halt therapeutic resistance.
