Abstract:The changes in dissolved organic matter (DOM) throughout the treatment processes in a drinking water treatment plant in Sweden and the formation of disinfection byproducts (DBPs) were evaluated by using ultra-high-resolution mass spectrometry (resolution of ∼500,000 at m/z 400) and nuclear magnetic resonance (NMR). Mass spectrometric results revealed that flocculation induced substantial changes in the DOM and caused quantitative removal of DOM constituents that usually are associated with DBP formation. While half of the chromophoric DOM (CDOM) was removed by flocculation, ∼4-5 mg L(-1) total organic carbon remained in the finished water. A conservative approach revealed the formation of ∼800 mass spectrometry ions with unambiguous molecular formula assignments that contained at least one halogen atom. These molecules likely represented new DBPs, which could not be prevented by the flocculation process. The most abundant m/z peaks, associated with formed DBPs, could be assigned to C5HO3Cl3, C5HO3Cl2Br, and C5HO3ClBr2 using isotope simulation patterns. Other halogen-containing formulas suggested the presence of halogenated polyphenolic and aromatic acid-type structures, which was supported by possible structures that matched the lower molecular mass range (maximum of 10 carbon atoms) of these DBPs. 1H NMR before and after disinfection revealed an ∼2% change in the overall 1H NMR signals supporting a significant change in the DOM caused by disinfection. This study underlines the fact that a large and increasing number of people are exposed to a very diverse pool of organohalogens through water, by both drinking and uptake through the skin upon contact. Nontarget analytical approaches are indispensable for revealing the magnitude of this exposure and to test alternative ways to reduce it.

Evaluating gas chromatography with a halogen-specific detector for the determination of disinfection by-products in drinking water

Abstract:The occurrence of disinfection by-products (DBPs) in drinking water has become an issue of concern during the past decades. The DBPs pose health risks and are suspected to cause various cancer forms, be genotoxic, and have negative developmental effects. The vast chemical diversity of DBPs makes comprehensive monitoring challenging. Only few of the DBPs are regulated and included in analytical protocols. In this study, a method for simultaneous measurement of 20 DBPs from five different structural classes (both regulated and non-regulated) was investigated and further developed for 11 DBPs using solid-phase extraction and gas chromatography coupled with a halogen-specific detector (XSD). The XSD was highly selective towards halogenated DBPs, providing chromatograms with little noise. The method allowed detection down to 0.05 μg L-1 and showed promising results for the simultaneous determination of a range of neutral DBP classes. Compounds from two classes of emerging DBPs, more cytotoxic than the "traditional" regulated DBPs, were successfully determined using this method. However, haloacetic acids (HAAs) should be analyzed separately as some HAA methyl esters may degrade giving false positives of trihalomethanes (THMs). The method was tested on real water samples from two municipal waterworks where the target DBP concentrations were found below the regulatory limits of Sweden.
Formation potential and analysis of 32 regulated and unregulated disinfection by-products: Two new simplified methods

Abstract:Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic matter naturally present in water, leading to the formation of disinfection by-products (DBPs). Multi-analyte methods based on mass spectrometry (MS) are preferred to quantify multiple DBP classes at once however, most require extensive sample pre-treatment and significant resources. In this study, two analytical methods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap (P&T) coupled with gas chromatography mass spectrometry (GC-MS) method was optimized that automated sample pre-treatment and analyzed volatile and semi-volatile compounds, including trihalomethanes (THMs), iodinated trihalomethanes (I-THMs), haloacetonitriles (HANs), haloketones (HKTs) and halonitromethanes (HNMs). LOQs were between 0.02-0.4 μg/L for most DBPs except for 8 analytes that were in the low μg/L range. A second method with liquid chromatography (LC) tandem mass spectrometry (MS/MS) was developed for the quantification of 10 haloacetic acids (HAAs) with a simple clean-up and direct injection. The LC-MS/MS direct injection method has the lowest detection limits reported (0.2-0.5 μg/L). Both methods have a simple sample pre-treatment, which make it possible for routine analysis. Hyperchlorination and uniform formation conditions (UFC) formation potential tests with chlorine were evaluated with water samples containing high and low TOC. Hyperchlorination formation potential test maximized THMs and HAAs while UFC maximized HANs. Ascorbic acid was found to be an appropriate quencher for both analytical methods. Disinfected drinking water from four water utilities in Alberta, Canada were also evaluated.

Abstract:In this case study, high sensitivity simple methods for the analysis of trihalomethanes (THM4), iodinated-trihalomethanes (I-THMs), haloacetic acids (HAAs), bromide, iodide and iodate have been developed. A one-step procedure for the analysis of haloacetic acids by head-space GC-MS provides good reproducibility and low limits of quantification (≤50 ng L-1). These methods were applied to characterize the formation of disinfection by-products (DBPs) in a full scale drinking water treatment plant. In this treatment plant, the incorporation of bromine into THMs increases throughout the water treatment line, due to the formation of bromine reactive species favored by the decrease of competition between dissolved organic carbon (DOC) and bromide towards chlorine. A linear correlation has been observed between the bromine incorporation factor and the Br-/DOC mass ratio. The conversion of iodine to iodate by chlorination occurs in this water due to the relatively high bromide concentration. Moreover, a higher formation of iodate compared to iodide levels in the raw water is observed indicating a degradation of organic iodinated compounds. The formation of I-THMs was constant in terms of quantity and speciation between campaigns despite fluctuating concentrations of DOC and total iodine in the raw water. A preferential removal of DBPs formed by the intermediate chlorination in the order I-DBPs > Br-DBPs > Cl-DBPs occurs during the subsequent activated carbon filtration. The removal rates range from 25 to 36% for the regulated THM4, from 82 to 93% for the ∑I-THMs and 95% for haloacetic acids. The assessment of the relative toxicity shows that despite a much lower concentration of HAAs (<10% of the total mass of measured DBPs) compared to THMs, these compounds are responsible for 75% of the relative cytotoxicity of the treated water. Bromoacetic acid on its own accounts for more than 60% of the overall toxicity of the 17 compounds included in this study.

Abstract:Natural organic matter is a major source of precursors of hazardous chlorinated disinfection byproducts (Cl-DBPs) formed during water treatment, but the majority of Cl-DBPs are still unidentified. In this study, we used a self-written halogen extraction code to identify halogen isotopic patterns in combination with the R package MFAssignR, to identify Cl-DBPs from Orbitrap mass spectra. One hundred and eighty-nine Cl-DBPs were detected during chlorination of a Suwannee River natural organic matter solution, and the structures of 20 of these compounds are reported for the first time. Kendrick mass defect analysis and structural identification confirmed that chlorinated carboxylic acids are common and likely to form during chlorination. A toxicity prediction using quantitative structure-activity relationship models indicated that most of the chlorinated carboxylic acids may be highly toxic. Our analytical strategy can identify Cl-DBPs accurately from complex mixtures and may also be applicable to the identification of other halogenated disinfection byproducts formed during water treatment.

Abstract:Disinfection by-products (DBP) formed from natural organic matter and disinfectants like chlorine and chloramine may cause adverse health effects. Here, we evaluate how the quantity and quality of natural organic matter and other precursors influence the formation of DBPs during chlorination and chloramination using a comprehensive approach including chemical analysis of regulated and emerging DBPs, total organic halogen quantification, organic matter characterisation and bioanalytical tools. In vitro bioassays allow us to assess the hazard potential of DBPs early in the chain of cellular events, when the DBPs react with their molecular target(s) and activate stress response and defence mechanisms. Given the reactive properties of known DBPs, a suite of bioassays targeting reactive modes of toxic action including genotoxicity and sensitive early warning endpoints such as protein damage and oxidative stress were evaluated in addition to cytotoxicity. Coagulated surface water was collected from three different drinking water treatment plants, along with reverse osmosis permeate from a desalination plant, and DBP formation potential was assessed after chlorination and chloramination. While effects were low or below the limit of detection before disinfection, the observed effects and DBP levels increased after disinfection and were generally higher after chlorination than after chloramination, indicating that chlorination forms higher concentrations of DBPs or more potent DBPs in the studied waters. Bacterial cytotoxicity, assessed using the bioluminescence inhibition assay, and induction of the oxidative stress response were the most sensitive endpoints, followed by genotoxicity. Source waters with higher dissolved organic carbon levels induced increased DBP formation and caused greater effects in the endpoints related to DNA damage repair, glutathione conjugation/protein damage and the Nrf2 oxidative stress response pathway after disinfection. Fractionation studies indicated that all molecular weight fractions of organic carbon contributed to the DBP formation potential, with the humic rich fractions forming the greatest amount of DBPs, while the low molecular weight fractions formed more brominated DBPs due to the high bromide to organic carbon ratio. The presence of higher bromide concentrations also led to a higher fraction of brominated DBPs as well as proportionally higher effects. This study demonstrates how a suite of analytical and bioanalytical tools can be used to effectively characterise the precursors and formation potential of DBPs.

Abstract:With the rapid development of society, more and more unknown halogenated disinfection byproducts (DBPs) enter into drinking water and pose potential risks to humans. To explore the unknown halogenated DBPs in tap water, a selectively nontargeted analysis (SNTA) method was developed by conducting micro-liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (micro-LC-QTOFMS). In this method, two runs were employed: in the first run, the modes of TOFMS and precursor ion (the fragments were set as Cl35/Cl37, Br79/Br81, and I126.9) were performed, and the molecular ions or precursor ions of the halogenated organics could be obtained; in the second run, the product ion mode was conducted by setting the molecular ion screened above, and the MS/MS spectrums could be acquired to speculate concerning the structure. Two kinds of model DBPs (one kind had an aliphatic structure and the other was an aromatic compound) were used to optimize the parameters of the MS, and their MS characteristics were summarized. With this SNTA method, 15 halogenated DBPs were screened in two tap water samples and their structures were proposed. Of them, six DBPs had not been reported before and were assumed to be new DBPs. Overall, the detected halogenated DBPs were mostly acidic substances.

Unravelling High-Molecular-Weight DBP Toxicity Drivers in Chlorinated and Chloraminated Drinking Water: Effect-Directed Analysis of Molecular Weight Fractions

Abstract:As disinfection byproducts (DBPs) are ubiquitous sources of chemical exposure in disinfected drinking water, identifying unknown DBPs, especially unknown drivers of toxicity, is one of the major challenges in the safe supply of drinking water. While >700 low-molecular-weight DBPs have been identified, the molecular composition of high-molecular-weight DBPs remains poorly understood. Moreover, due to the absence of chemical standards for most DBPs, it is difficult to assess toxicity contributions for new DBPs identified. Based on effect-directed analysis, this study combined predictive cytotoxicity and quantitative genotoxicity analyses and Fourier transform ion cyclotron resonance mass spectrometry (21 T FT-ICR-MS) identification to resolve molecular weight fractions that induce toxicity in chloraminated and chlorinated drinking waters, along with the molecular composition of these DBP drivers. Fractionation using ultrafiltration membranes allowed the investigation of <1 kD, 1-3 kD, 3-5 kD, and >5 kD molecular weight fractions. Thiol reactivity based predictive cytotoxicity and single-cell gel electrophoresis based genotoxicity assays revealed that the <1 kD fraction for both chloraminated and chlorinated waters exhibited the highest levels of predictive cytotoxicity and direct genotoxicity. The <1 kD target fraction was used for subsequent molecular composition identification. Ultrahigh-resolution MS identified singly charged species (as evidenced by the 1 Da spacing in 13C isotopologues), including 3599 chlorine-containing DBPs in the <1 kD fraction with the empirical formulas CHOCl, CHOCl2, and CHOCl3, with a relative abundance order of CHOCl > CHOCl2 ≫ CHOCl3. Interestingly, more high-molecular-weight CHOCl1-3 DBPs were identified in the chloraminated vs chlorinated waters. This may be due to slower reactions of NH2Cl. Most of the DBPs formed in chloraminated waters were composed of high-molecular-weight Cl-DBPs (up to 1 kD) rather than known low-molecular-weight DBPs. Moreover, with the increase of chlorine number in the high-molecular-weight DBPs detected, the O/C ratio exhibited an increasing trend, while the modified aromaticity index (AImod) showed an opposite trend. In drinking water treatment processes, the removal of natural organic matter fractions with high O/C ratio and high AImod value should be strengthened to minimize the formation of known and unknown DBPs.

Abstract:Water disinfection can create an unintended chemical risk from the formation of disinfection byproducts (DBPs). Epidemiological studies have shown a potential association of water disinfection with an increased risk of bladder cancer. The DBPs responsible for the observed adverse health effects remain unclear. Identification of DBPs of health relevance presents a great analytical challenge. Halobenzoquinones (HBQs) are a unique class of emerging DBPs of potential toxicological relevance. This review presents the analytical characterization, occurrence, formation, transformation, and removal of HBQ DBPs in drinking water. Specifically, we will discuss the challenges of HBQ analytical method development and the resulting solid phase extraction and high performance liquid chromatography-tandem mass spectrometry techniques that achieve the determination of HBQs at ng/L levels in water. Other techniques for HBQ analysis, including ultraviolet-visible spectroscopy, electron spin resonance, and gas chromatography-mass spectrometry, will be briefly discussed.
Chemical characterization and relative toxicity assessment of disinfection byproduct mixtures in a large drinking water supply network

Abstract:In order to reduce the formation of disinfection by-products (DBPs) during potabilization of water it is necessary to explore the potential of the source water and the applied treatment to generate these chemicals. This is actually more challenging in large drinking water networks that use different source waters to satisfy drinking water demand. In this regard, this work investigated the formation of DBPs in water matrices that are commonly supplied to the city of Barcelona and its metropolitan area. The regulated trihalomethanes and haloacetic acids were the most abundant DBP classes in these waters, followed by haloacetamides and haloacetonitriles or trihalogenated acetaldehydes (THALs). On the contrary, the formation potential of iodo-DBPs was minor. Mixing of drinking water treatment plant finished waters with desalinated water decreased the overall DBP formation potential of the water but resulted in the increased formation of brominated DBPs after long chlorine contact time. The formation of most DBPs was enhanced at high water temperatures (except for Br-THALs) and increasing residence times. Potential cytotoxicity and genotoxicity of the DBP mixtures were mainly attributed to the presence of nitrogen-containing DBPs and HAAs.
Assessing the role of different dissolved organic carbon and bromide concentrations for disinfection by-product formation using chemical analysis and bioanalysis

Abstract:Concerns regarding disinfection by-product (DBP) formation during drinking water treatment have led water utilities to apply treatment processes to reduce the concentration of DBP precursor natural organic matter (NOM). However, these processes often do not remove bromide, leading to high bromide to dissolved organic carbon (DOC) ratios after treatment, which can increase the formation of more toxic brominated DBPs. In the current study, we investigated the formation and effect of DBPs in a matrix of synthetic water samples containing different concentrations of bromide and DOC after disinfection with chlorine. Trihalomethanes and haloacetic acids were analysed by chemical analysis, while effect was evaluated using in vitro bioassays indicative of the oxidative stress response and bacterial toxicity. While the addition of increasing bromide concentrations did not alter the sum molar concentration of DBPs formed, the speciation changed, with greater bromine incorporation with an increasing Br:DOC ratio. However, the observed effect did not correlate with the Br:DOC ratio, but instead, effect increased with increasing DOC concentration. Water samples with low DOC and high bromide did not exceed the available oxidative stress response effect-based trigger value (EBT), while all samples with high DOC, irrespective of the bromide concentration, exceeded the EBT. This suggests that treatment processes that remove NOM can improve drinking water quality, even if they are unable to remove bromide. Further, iceberg modelling showed that detected DBPs only explained a small fraction of the oxidative stress response, supporting the application of both chemical analysis and bioanalysis for monitoring DBP formation.

Abstract:Colony‐stimulating factor‐1 (CSF‐1) is a cytokine required for proliferation, differentiation, activity, and survival of cells of the mononuclear phagocytic system. The growth factor is synthesized as a soluble, matrix, or membrane associated molecule. The specific functions of these forms are not clear. However, some data suggest a dependence of the development of various populations of tissue macrophages on the locally expressed and presented cytokine. Deficiency in CSF‐1, as is the case in the murine mutant strain op/op, results in low numbers of macrophages and monocytes and, most striking, leads to osteopetrosis due to a virtual absence of osteoclasts. Using the op/op mutation as a model, CSF‐1 was established as one of the growth factors for osteoclasts. The expression of CSF‐1 receptors, encoded by the proto‐oncogene c‐fms, by osteoclast precursors and osteoclasts, suggested an effect of this cytokine not only during osteoclast formation but also on the mature cells. In fact, CSF‐1 was shown to inhibit the resorbing activity, to stimulate migration, and to support survival of isolated osteoclasts in vitro. By these actions on cells of the osteoclast lineage, CSF‐1 induces recruitment of new osteoclasts, leading to a net increase of bone resorption, and might govern the spatial distribution of resorption sites within the bone. During these processes, locally expressed and presented forms of the growth factor may play a crucial role, as will be discussed in this article. © 1994 Wiley‐Liss, Inc.

Abstract:Recently,13 new polar phenolic chlorinated and brominated disinfection byproducts (Cl-/Br-DBPs) have been identified in drinking water by using multiple reaction monitoring (MRM),precursor ion scan,and product ion scan of ultra-performance liquid chromatography/ electrospray ionization-triple quadrupole mass spectrometry (UPLC/ESI-tqMS).The new DBPs have attracted increasing concern since they were proved to have higher cytotoxicity,developmental toxicity,and growth inhibition than aliphatic DBPs,and form trihalomethanes and haloacetic acids via decomposition.In this study,a series of samples were prepared by reacting gallic acid with chlorine/monochloramine in the presence of bromide to simulate chlorinated/chloraminated drinking water samples with gallic acid as carbon source.The reaction products,intermediate compounds,and gallic acid were analyzed using UPLC/ESI-tqMS.Gallic acid was proved to exist in both simulat and real source waters,and 10 new polar phenolic Cl-/Br-DBPs were identified and quantified in the chlorinated/chloramined gallic acid reaction solutions,suggesting that gallic acid is one of the precursors of the 13 new phenolic DBPs.
Combination of high resolution mass spectrometry and a halogen extraction code to identify chlorinated disinfection byproducts formed from aromatic amino acids

Abstract:Chlorination can lead to the formation of hazardous chlorinated disinfection byproducts (Cl-DBPs). We identified tyrosine (Tyr) and tryptophan (Trp) as precursors of toxic Cl-DBPs and developed a halogen extraction code to complement ultra performance liquid chromatography in tandem with high resolution mass spectrometry (UPLC-HRMS) in detecting and identifying Cl-DBPs. We detected 20 and 11 Cl-DBPs formed from chlorination of Tyr and Trp, respectively, and identified the structures of 15 Cl-DBPs. Fourteen structures were previously unreported. We also proposed the tentative formation pathways of these newly identified Cl-DBPs. Their incidence in real water sources demonstrated that these Cl-DBPs are likely to form during chlorination of reclaimed water. We computationally predicted the toxicity of these Cl-DBPs, which was relatively high, indicating that these Cl-DBPs could be hazardous and were of valid concern. Combining analytical data with the halogen extraction code can identify Cl-DBPs accurately from complex compounds. This analytical method can be used to identify Cl-DBPs of water treatment procedures in further studies.

To address this, we have created a simple and sensitive method for simultaneously measuring 9 classes of DBPs. Haloacetic acids (HAAs) and iodo-acetic acids (IAAs) are determined using silylation derivatization, replacing diazomethane or acidic methanol derivatization with a more environmentally friendly and simpler treatment process that also offers greater sensitivity. Mono-/di-haloacetaldehydes (mono-/di-HALs) are directly analyzed without derivatization, along with trihalomethanes (THMs), iodo-THMs, haloketones, haloacetonitriles, haloacetamides, and halonitromethanes. Of the 50 DBPs studied, recoveries for most were 70-130%, LOQs for most were 0.01-0.05 μg/L, and relative standard deviations were <30%. We subsequently applied this method to 13 home tap water samples. Total concentrations of 9 classes of DBPs were 39.6-79.2 μg/L, in which unregulated priority DBPs contributed 42% of total DBP concentrations and 97% of total calculated cytotoxicity, highlighting the importance of monitoring their presence in drinking water. Br-DBPs were the dominant contributors to total DBPs (54%) and total calculated cytotoxicity (92%). Nitrogenous DBPs contributed 25% of total DBPs while inducing 57% of total calculated cytotoxicity. HALs were the most important toxicity drivers (40%), particularly four mono-/di-HALs, which induced 28% of total calculated cytotoxicity. This simple and sensitive method allows the synchronous analysis of 9 classes of regulated and unregulated priority DBPs and overcomes the weaknesses of some other methods especially for HAAs/IAAs and mono-/di-HALs, providing a useful tool for research on regulated and unregulated priority DBPs.
Effects of solid phase extraction conditions on molecular composition of unknown disinfection byproducts in chlorinated municipal wastewater based on FT-ICR-MS analysis

Abstract:Disinfection byproducts (DBPs) have been extensively investigated during the chlorination of water and wastewater. Although over 700 DBPs have been identified, more than 50% of the total organic halogen remains unknown. Solid phase extraction (SPE) has been emerged as a popular pretreatment approach for enrichment and desalting of unknown DBPs prior to the mass spectrometry analysis. However, the effects of SPE conditions on unknown DBPs in real wastewater have not yet been reported. Herein, three factors (acid types, pH values, and sorbent types) influencing the composition of DBPs in chlorinated municipal wastewater were systematically investigated by Fourier transform ion cyclotron resonance mass spectrometry and statistical analysis. The results indicated that the number of DBPs in different SPE conditions ranged from 280 to 706, and the majority ones were Br-DBPs and CHOX compounds. Compared with H2SO4, more common DBPs were found when using HCl and HCOOH to adjust the pH values of samples. The unique DBPs extracted at pH 1.0 and 2.0 generally owned higher modified aromaticity index (AImod) value and C number than at pH 3.0. The effect of acid types on the extracted DBPs was pH dependent, and the total number of extracted DBPs increased with the increasing of pH value. In terms of sorbent types, the unique DBPs in C18 sorbent possessed low O/C ratios (O/C < 0.6), whereas the unique ones in HLB sorbent owned high O/C ratios (O/C > 0.6). Compared with C18 and HLB sorbents, the unique DBPs extracted in PPL sorbent were characterized by relatively high AImod and DBE values. Based on mass difference analysis, 1496 precursors-DBPs pairs were identified in all extracted samples, with the highest number of bromine substitution reaction. Overall, the effects of SPE conditions on the composition of unknown DBPs should not be overlooked, and the amount and diversity of DBPs may be underestimated under a single SPE condition. This study provides new methodological references for the accurate identification of unknown DBPs with different characteristics in real wastewater.
Variations of disinfection byproduct precursors through conventional drinking water treatment processes and a real-time monitoring method

Abstract:In this investigation, raw water (RW), settled water (SW), and filtered water (FW) collected from a drinking water treatment plant were fractionated into 24 natural organic matter (NOM) fractions with varying molecular weights and hydrophobicity. The yields of disinfection byproducts (DBPs) obtained during the chlorination of the NOM fractions were explored. Results revealed that the 0-1 kDa, 5-10 kDa, and hydrophobic DBP precursors dominated RW. Hydrophobic fractions cannot be effectively removed, which contributed to the high DBP precursors remaining in the FW. The optional optical parameters, including UVA (UV340, UV360, and UV380), UVB (UV280, UV300, and UV310), and UVC (UV254, UV260, and UV272), were analyzed to determine the DBP yields during chlorination of different NOM fractions. Results revealed that UVC could be applied to indicate the regulated DBP yields of the humified precursors. Contrary to the generally accepted view, for biologically derived precursors, their regulated DBPs and dichloroacetonitrile correlated better with UVA (e.g. UV340). Moreover, PARAFAC analysis was applied to decompose an array of 24 EEM spectra. Good linear correlations were found between the PARAFAC components and most DBP yields. Furthermore, four fluorescence parameters were proposed via a modified fluorescence picking method, which can serve as excellent surrogates of PARAFAC components. These fluorescence parameters were found to be effective in indicating most DBP yields. Finally, the fluorescence intensity at excitation wavelength/emission wavelength = 310/416 nm was found to be a promising built-in parameter for the real-time monitoring of DBP precursors, regardless of the humification degree of the precursors.

Abstract:Formation of disinfection by-products (DBPs) in ten drinking source waters located in the United States was examined in this study. DBP generation was interpreted in the context of halogenation-induced changes of log-transformed absorbance spectra of dissolved organic matter (DOM) present in the waters. This approach allows probing the behavior of relatively minor structures that can be highly sensitive towards any process of interest, notably DOM halogenation. This concept was applied to examine effects of chlorination time on the kinetics of chlorine consumption and release of several DBP groups such as total trihalomethanes (THM4, including CHCl3, CHCl2Br, CHClBr2 and CHBr3), haloacetic acids (HAA(9), including MCAA, MBAA, DCAA, TCAA, BCAA, DBAA, BDCAA, DBCAA and TBAA), haloacetonitriles (THAN(4), including TCAN, DCAN, BCAN and DBAN), haloketones (HK2, including DCP and TCP), chloral hydrate (CH) and chloropicrin (CPN). Two alternative parameters, namely the differential logarithm of DOM absorbance at 350 nm (DLnA(350)) and change of the spectral slope in the range of wavelengths 325-375 nm (DSlope(325-375)) were introduced to quantify individual DBP species formed and Cl-2 consumption. DLnA(350) and DSlope(325-375), especially DLnA(350) were determined to be more reliable than differential absorbance at 272 nm that was utilized in prior applications of differential spectroscopy to characterize DBP formation. Strong linear relationships between DLnA(350) values and concentrations of major groups of and individual DBP species (e.g. THM4, HAA(9), HAN(4) and CPN were found to exist (mostly, R-2 > 0.95) and the intercept of these correlations with the y-axis was near zero for the examined water sources. Correlations between DLnA(350) values and concentrations of CH and HK2 were also strong but they were nonlinear. The slope of the correlations between the concentrations of major groups of DBP species vs-DLnA(350) were also well correlated with SUVA(254) and LnA(350) for all the examined source waters. It indicates that log-transformations of the absorbance spectra of surface water and parameters based on such transformations (e.g., DLnA(350) and DSlope(325-375)) have a potential to provide an alternative reliable approach to monitor the halogenation of DOM and attendant formation of individual DBP species. Abstract:Background: Clinical investigation of cancer immunotherapy has been very active and several approaches have been evaluated in Phase III trials. In particular, the characterisation at the molecular level of tumour-specific antigens, together with expert knowledge from GSK Biologicals in recombinant protein manufacturing and immunological Adjuvant Systems, has led the company to develop Antigen-Specific Cancer Immunotherapeutic (ASCI). Objective/methods: This paper reviews the different cancer immunotherapy approaches that have reached Phase III clinical development. A special attention is given to GSK's ASCI approach. Conclusion: Based on encouraging data in a double-blind Phase II trial in non-small-cell lung cancer, the selection of the most suitable adjuvant system in melanoma and the choice of the adequate clinical setting for the clinical development of cancer immunotherapy, the ASCI approach offers the perspective that the long quest towards a new cancer treatment approach is about to succeed.

Abstract:Background: Knowledge about human exposure and health effects associated with non-routinely monitored disinfection by-products (DBPs) in drinking water is sparse. Objective: To provide insights to estimate exposure to regulated and non-regulated DBPs in drinking water. Methods: We collected tap water from homes (N = 42), bottled water (N = 10), filtered tap water with domestic activated carbon jars (N = 6) and reverse osmosis (N = 5), and urine (N = 39) samples of participants from Barcelona, Spain. We analyzed 11 haloacetic acids (HAAs), 4 trihalomethanes (THMs), 4 haloacetonitriles (HANs), 2 haloketones, chlorate, chlorite, and trichloronitromethane in water and HAAs in urine samples. Personal information on water intake and socio-demographics was ascertained in the study population (N = 39) through questionnaires. Statistical models were developed based on THMs as explanatory variables using multivariate linear regression and machine learning techniques to predict non-regulated DBPs. Results: Chlorate, THMs, HAAs, and HANs were quantified in 98-100% tap water samples with median concentration of 214, 42, 18, and 3.2 μg/L, respectively. Multivariate linear regression models had similar or higher goodness of fit (R2) compared to machine learning models. Multivariate linear models for dichloro-, trichloro-, and bromodichloroacetic acid, dichloroacetonitrile, bromochloroacetonitrile, dibromoacetonitrile, trichloropropnanone, and chlorite showed good predictive ability (R2 = 0.8-0.9) as 80-90% of total variance could be explained by THM concentrations. Activated carbon filters reduced DBP concentrations to a variable extent (27-80%), and reverse osmosis reduced DBP concentrations ≥98%. Only chlorate was detected in bottled water samples (N = 3), with median = 13.0 µg/L. Creatinine-adjusted trichloroacetic acid was the most frequently detected HAA in urine samples (69.2%), and moderately correlated with estimated drinking water intake (r = 0.48). Significance: Findings provide valuable insights for DBP exposure assessment in epidemiological studies. Validation of predictive models in a larger number of samples and replication in different settings is warranted. Impact statement: Our study focused on assessing and describing the occurrence of several classes of DBPs in drinking water and developing exposure models of good predictive ability for non-regulated DBPs.
