Abstract:In order to provide references for leaf nutrition diagnosis of fingered citron, the technique of near infrared reflectance spectroscopy (NIRS) was introduced to analyze nitrogen (N), phosphorus (P), potassium (K), iron (Fe), manganese (Mn), zinc (Zn), and copper (Cu) in the dry-leaf samples of fingered citron. The best calibration model for N was developed with high RSQ(CAL) (0.90), SD/SECV (2.73) and low SEC (1.06 mg g(-1)), good calibration models were obtained for P, K, Fe and Mn, and no significant correlations were found between the spectra and the individual amounts of Zn and Cu. When tested using a validation set (n = 38), N was well predicted with low values of SEP (1.21 mg g(-1)) and high RPD (2.5). The values of SEP and RPD were also acceptable for the external validation of P, Fe and Mn. Near-infrared spectroscopy analysis technique shows potential of diagnosing minerals in fingered citron, particularly for N, P, Fe and Mn.

Abstract:OBJECTIVE Evaluating the effect of preconditioning with simvastatin in acute kidney injury induced by sepsis. METHOD Male adult Wistar rats were divided into the following groups: SHAM (control); SHAM+Statin (0.5 mg/kg simvastatin, orally); Sepsis (cecal puncture ligation - CPL); Sepsis+Statin. Physiological parameters, peritoneal fluid culture, renal function, oxidative metabolites, severity of acute kidney injury and animal survival were evaluated. RESULTS The treatment with simvastatin in induced sepsis showed elevation of creatinine clearance with attenuation of generation of oxidative metabolites, lower severity of acute kidney injury and reduced mortality. CONCLUSION This investigation confirmed the renoprotection with antioxidant principle of the simvastatin in acute kidney injury induced by sepsis in an experimental model.

Abstract:Abstract Neutron Activation Analysis Techniques more used in determining macro and microcontent in food ingredients. The quality of the test results is needed to produce a valid analysis. To validate the result of the analysis of macro and micro mineral content in food ingredients, quantitative analysis of the elements in the reference standard is NIST SRM 1547 certified Peach Leaves, NIST SRM 1573a Tomato Leaves, and NIST SRM 1570a Spinach Leaves have been done. The Z-score parameter was used as a validation parameter. Elemental with a long half-life was determined quantitatively using the NAA k0 method. The elements were determined quantitatively through long-lived radioisotopes using the k0 Instrumental Neutron Activation Analysis in the G.A Siwabessy reactor. A number of 50 mg-120 mg samples were weighed and irradiated at the neutron flux of 2.5x10 13 n.cm−2.s−1. Irradiation was carried out for 3 hours on the rabbit system while counting with gamma spectrometry was carried out after cooling time of 23 weeks. Quantitative analysis was carried out using a soft K0-IAEA device. The results of quantitative analysis obtained elements of Ca, Fe, Zn, Rb, Sr on SRM NIST 1547 Peach Leaves; elements of Ca, Cr, Fe, Co on NIST SRM 1573a Tomato leaves and elements Ca, Sc, Zn, Rb, Sr, Co on NIST SRM 1570a Spinach Leaves with uncertainty <10% (3.26-8.68%). The Z-score value ranges from-1.25 to 0.69 and the relative bias ranges from 0.1 to 6.28%, so the results of the analysis of these elements are valid for testing elements in food.

Abstract:This paper presents the standardization of Proton Induced X-rays Emission (PIXE) technique for the trace element analysis of thick standard samples. Three standard reference materials (SRMs) viz-à-vis titanium, copper and iron base alloys were used for the study due to their availability. The protons beam was accelerated up to 2.57 MeV energy by 5UDH-II tandem Pelletron accelerator and samples were irradiated at different geometry and durations. Spectrum was acquired using a multi-channel spectrum analyzer while spectrum analysis was done using a GUPIXWIN model for determination of elemental concentrations of trace elements. The obtained experimental data was compared with theoretical data and results were found in close agreement.

Abstract:Due to the enormous interest in plants related to bioscience, environmental and toxicological research, analytical methods are expected with the ability of getting information on elemental transfer, distribution and contents in plants. In this work, a mixture of gelatin (GA) and hydroxypropyl methyl cellulose (HPMC) was prepared to simulate plant matrix, a method based on laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) with matrix-matching external calibration was proposed for direct quantification of multiple elements in plants. The pH and composition of GA&HPMC substrate was optimized, such as the concentration of spiked nitric acid, the mass fraction of both GA and HPMC in the substrate and the mass ratio of GA: HPMC. After spiking elemental solution, coating the mixture onto a glass slide and drying overnight at room temperature, GA&HPMC substrate was obtained. The substrate obtained with GA: HPMC of 8: 2 was used to fabricate the standard series, which exhibited good elemental homogeneity and similar elemental signal intensities in LA-ICP-MS detection to that obtained for plant Certified Reference Material (CRM). CRMs of different plants including Citrus leaf (GBW10019), Tea (GBW07605), Beans (GBW10021) and Scallions (GBW10049) were further pressed into pellets and subjected to the proposed method, and the quantification accuracy was demonstrated. The limits of detections of this method were found to be 0.003 (Ce)-104 (Ca) μg g −1 , with a wide linear range (0.01–10000 μg g −1 ) for 17 target elements. The application potential of the method was further demonstrated by performing elemental imaging in Trigonotis peduncularis leaves. Rapid in-situ quantitative imaging of Zn, Cu, Sr and Mn was achieved, and the elemental quantitative distributions were discussed. The constructed substrate helps direct elemental quantification in plants. It provided a powerful and efficient tool for the investigation of the distribution and transfer of elements in plants, favoring further exploration of elemental bioavailability, transport and toxicity mechanisms.

Abstract:Hyperbilirubinaemia is a well-recognized effect in dyserythropoietic anaemias (Lewis & Verwilghen. 1973). while Lewis (1 9 72) also noted the occurrence of acquired dyserythropoiesis as an early manifestation of liver cirrhosis. Since these early observations there has been scanty information on the association of acquired dyserythropoiesis with liver disease. In order to draw attention to this phenomenon we report the following cases. Case 1. Female aged 58 years. In 1956 the patient developed infective hepatitis, leading to chronic hepatitis in 1966. Ten years later whilst the patient was undergoing hysterectomy for fibromyoma the liver was seen to be cirrhotic. In February 1977 the spleen enlarged 6 cm below the left costal margin. At that time haemoglobin was 13.7 g/ dl, leucocyte and platelet counts were normal. Scintigraphy with radioactive gold suggested chronic hepatitis. Direct Coombs test was negative using anti IgG serum but positive with polyvalent antiglobulin serum and with anti C3D and C4 sera. There were no signs of haemolysis. Indirect serum bilirubin was 12-25 mg/l (20.7-43.1 pnol): there was increased fetal urobilinigen excretion of 356-408 mg per 24 h. The bone marrow showed marked dyserythropoiesis (Table I) which disappeared after 8 months of liver protection therapy. Abstract:MeV ion microprobe measurements of the lesser and trace life element concentrations in biological tissues are challenging because of complex spatial inhomogeneities in these types of samples. Measurements on ex vivo tissue sections require determination of the matrix element composition and the tissue section thickness. For these reasons, in this work, we adapted the Dynamic Analysis approach known from literature, to interpret the MeV ion microscopy data to determine concentrations of H, C, O, and N as well as the thickness of different tissue regions in Non Human Primate (NHP) mesenteric lymph node section. The results showed no strong variations of the matrix element contents regardless of section thickness variations in the tissue. The matrix information was used to quantify total-Ca molarities and a significant ∼ 30 mM Ca concentration hotspot was observed at the edge of sinus structure in the mesenteric lymph node as compared to the 3-4 mM total-Ca levels in the surrounding tissues. Thus, MeV ion microprobe imaging combined with dynamic analysis comprise a novel chemometric approach paving a way for quantitative analysis of similarly complicated animal and plant biological tissue sections.

Abstract:Abstract Distributed connectionist (DC) systems offer a set of processing features which are distinct from those provided by traditional symbol processing (SP) systems. In general, the features of DC systems are derived from the nature of their distributed representations. Such representations have a microsemantics -i.e. symbols with similar internal representations tend to have similar processing effects. In contrast, the symbols in SP systems have no intrinsic microsemantics of their own; e.g. SP symbols are formed by concatenating ASCII codes that are static, human engineered and arbitrary. Such symbols possess only a macrosemantics - i.e. symbols are placed into structured relationships with other symbols, via pointers, and bindings are propagated via variables. The fact that DC and SP systems each provide a distinct set of useful features serves as a strong research motivation for seeking a synthesis. Abstract:Cascabela Thevetia medicinal plant, known to possess anti-cancer properties, was probed for its elemental content with a proton beam of 3 MeV energy. The present Particle Induced X-ray Emission (PIXE) investigation was performed using the 3 MV Pelletron accelerator of the tandem type available at the Institute of Physics, Bhubaneswar, India. The purpose of this study was to identify and thereby quantify the levels of elements present in the bark of Cascabela Thevetia medicinal plant, collected from four environmentally different locations, and study their variation with respect to the geographical region. An attempt was also made to correlate the occurrence of elements in this medicinal plant to its pharmacological action and ascertain its efficacy in the treatment of cancer. The outcome of this study underlines the need for exercising precaution in prescribing safe and effective doses and deciding the appropriate duration for which herbal medicines can be prescribed to cancer patients based on their regional variation.

Abstract:Neem (Azadirachta indica) a member of Meliaceae plays an immense role in human health and disease which is attributed to its composition of Bioactive Secondary Metabolites (BASM). It has been widely used in Indian Traditional Systems of Medicine that includes Ayurveda, Siddha, Unani, Homeopathy and other Folklore Systems of Medicine practiced in the Indian Subcontinent for the treatment and prevention of various diseases. Current global health perspectives and medical practice in the post COVID era has no other way but to seek to merge alternative systems of medicine with evidence-based therapeutic aspects for a better understanding of the metabolic process and its effects in the human body. The studies based on animal model established that neem and its chief constituents play pivotal role in anticancer management through the modulation of various molecular pathways including p53, pTEN, NF-κB, PI3K/Akt, Bcl-2, and VEGF. Besides, NEEM plays a vital role in the management of diabetics and its associated long term complication through ROS scavenging and ameliorative potentials to restore oxidative injury/ inhibit enzymes linked to. Overall NEEM is considered as GRAS medicinal plant that modulates metabolic inflammations without side effects. Though it has been confirmed that neem and its constituents play role in the scavenging of free radical and prevention of disease pathogenesis, a clear scientific basis of its pharmacoinformatics is still lacking. Gas Chromatography–Mass Spectroscopy (GC–MS) analysis of the fractions revealed the presence of 62 metabolites. Abstract:Accurately determining the elemental composition of plant tissues is essential for physiological studies on plant stress, including salinity tolerance. However, high-throughput routine analysis of light elements (range of sodium to calcium) in plant samples is challenging due to the need for complete sample dissolution and expensive inductively coupled plasma-mass-spectrometry (ICP-MS) analysis. Lower costs method (ion chromatography, ion selective electrodes) exists, but also require sample dissolution and lack sensitivity for very small samples (<10 mg). This study reports on a new method for the quantitative analysis of light elements in plant tissues using monochromatic X-ray fluorescence (XRF) instrumentation and innovative sample preparation and mounting. We used this approach to assess elemental uptake, distribution, and accumulation in Arabidopsis thaliana and Oryza sativa plants subjected to salt stress. The method can be used on samples as small as 1 mg making it suitable for small Arabidopsis thaliana plants. We systematically evaluated different sample preparations methods, repeatability, and measurement times to confirm the robustness of the technique. The results show that the monochromatic XRF method delivers rapid, non-destructive, and extraction-free analysis, strongly correlating with ICP-MS acquired data. As such, the monochromatic XRF method is a reliable and efficient alternative for studying salinity tolerance ideally suited for investigating elemental composition of early plant developmental stages, offering new possibilities for research into early stimuli sensing, perception and nutrient efficiency.

Abstract:Medicinal plants play a vital role in herbal medical field and allopathic medicine field industry. Chemical and spectroscopic studies of Taraxacum officinale, Hyoscyamus niger, Ajuga bracteosa, Elaeagnus angustifolia, Camellia sinensis, and Berberis lyceum are conducted in this paper by using a 532-nm Nd:YAG laser in an open air environment. These medicinal plant's leaves, roots, seed, and flowers are used to treat a range of diseases by the locals. It is crucial to be able to distinguish between beneficial and detrimental metal elements in these plants. We demonstrated how various elements are categorized and how roots, leaves, seeds and flowers of same plants differ from each other on the basis of elemental analysis. Furthermore, for classification purpose, different classification models, partial least square discriminant analysis (PLS-DA), k-nearest neighbors (kNN), and principal component analysis (PCA) are used. We found silicon (Si), aluminum (Al), iron (Fe), copper (Cu), calcium (Ca), magnesium (Mg), sodium (Na), potassium (K), manganese (Mn), phosphorous (P), and vanadium (V) in all of the medicinal plant samples with a molecular form of carbon and nitrogen band. We detected Ca, Mg, Si, and P as primary components in all of the plant samples, as well as V, Fe, Mn, Al, and Ti as essential medicinal metals, and additional trace elements like Si, Sr, and Al. The result's findings show that the PLS-DA classification model with single normal variate (SNV) preprocessing method is the most effective classification model for different types of plant samples. The average correct classification rate obtained for PLS-DA with SNV is 95%. Moreover, laser-induced breakdown spectroscopy (LIBS) was successfully employed to perform rapid, sensitive, and quantitative trace element analysis on medicinal herbs and plant samples.

Abstract:Azadirachta indica has several medicinal uses, especially its leaves. Over 4000 years ago, Ayurvedic medicine used it for its therapeutic benefits. This study examined the biological activity of Neem crude extracts and green-chemically produced Ag-NPs. TPCs and TFCs were measured for polyphenolic burden in consecutive extracts. DPPH, ABTS, and FRAP experiments measured antioxidant and antimicrobial activity against seven strains of food-borne pathogenic bacteria and eight mycotoxigenic fungi. At 1000 μg/mL, ethanolic and aqueous extracts of Neem leaves had 80.10% and 69.41% in DPPH and 71.42% and 74.61% in ABTS assays for the antioxidant activity, compared to 93.58% for BHT. At 800 μg/mL, both extracts showed antioxidant activity with 57.52 and 57.87 μM in the FRAP assay, compared to 139.97 μM for Ascorbic acid. Both extracts demonstrated antimicrobial activity with 0.02 to 0.35 mg/mL as antibacterials, 0.03 to 2.17 mg/mL as antifungals, and 0.04 to 0.42 mg/mL as antibacterials. Compared to Neem crude extract, Neem Ag-NPs had the lowest MIC values as antibacterials and antifungals at 0.05 to 0.07 mg/mL and 0.07 to 0.20 mg/mL, respectively. Neem Ag-NPs and crude extract boost antioxidant and antibacterial properties.

Abstract:Objective: Development and validation of a technique for preparation of a reference sample for elemental microanalysis using the XRF technique in terms of repeatability, reproducibility, and optimization of the technique for rapid determination of the elemental composition of medicinal plants based on X-ray fluorescence analysis. Methods: Samples: fresh shoots of Kalanchoe daigremontiana, ready reference sample "Birch Leaf" LB-1 (A. P. Vinogradov Institute of Geochemistry, Siberian Branch of the Russian Academy of Sciences, Irkutsk, Russia), and IAEA reference sample SRM 2976 (IAEA, MEL, Monaco). The dispersed fraction was analyzed using a Master Sizer 2000 instrument (Malvern Panalytical, Worcestershire, UK). Elemental analysis using an energy dispersive X-ray fluorescence spectrometer EDX-7000 Shimadzu (Shimadzu Corporation, Kyoto, Japan), GZ-AAS using an Agilent instrument, model 240Z AA instrument (Agilent Technologies, Inc., Santa Clara, USA) with electrothermal atomization and Zeeman background correction, and ICP-MS using an Agilent 7500 CE instrument (Agilent Technologies, Inc., Santa Clara, USA). Results: By the LALLS method, they were separated by the maximum distribution, which was 63 microns, and a minor fraction of 39 microns. This indicates sufficient homogeneity in the sample. Further, homogeneity was proved by the XRF method by measuring six independent samples obtained by the quartering method. Also, the elemental composition of the reference samples was determined: completely dried, homogenized before sifting, and homogenized after sifting. Further, the obtained reference sample of K. daigremontiana was compared with reference samples: IAEA SRM 2976 and "birch leaf methods: ICP-MS, GZ-AAS, XRF. Conclusion: The reference sample will allow for rapid analysis of medicinal plant raw materials. Standardization of medicinal plants by the content of microelements will allow observing species differences as well as adjusting the concentrations of microelements for therapeutic purposes using medicinal plants.

This study has revealed the occurrence of nutrients, phytochemicals, with biological value, and also the GC-MS analysis exposed 20 peaks through 20 individual chemical compounds, and all the compounds are deliberated as energetic medicinal bioactive compounds. Likewise the FE-SEM is used to find out the topographical characteristics of biomaterial and the FTIR analysis.

Abstract:Inorganic element composition of genetically transformed hairy root cultures (HRCs) of a medicinal plant, Sida acuta was determined using PIXE technique. HRCs had a higher accumulation of pro-health trace elements compared to natural roots. It was estimated that < 160 g d.wt. of a selected rhizoclone could suffice to provide nearly all tested essential elements catering to per diem requirement of the human body. The ideal multi-elemental profile ushers a new possibility of integrating hairy root extracts in modern therapeutics (“rhizotherapy”) as complementary medicine or a dietary nutraceutical supplement.

Abstract:The use of medicinal plants is integral to addressing liver, heart, lung, and other metabolic issues. These plants are rich in vitamins, minerals, flavonoids, and alkaloids, which collectively help in lowering intestinal glucose absorption and increasing insulin secretion by pancreatic tissues. Elemental analysis, encompassing major, minor, and trace elements, was performed on various parts (leaves, roots, and seeds) of 16 anti-diabetic medicinal plants collected from different regions of Swat and Peshawar in Khyber Pakhtunkhwa (KPK), Pakistan. This analysis utilized proton-induced X-ray emission (PIXE) and inductively coupled plasma optical emission spectroscopy (ICP-OES) techniques. Our PIXE and ICP-OES analysis revealed the presence of major (Ca, K, S, P), minor (Si, Cl), and trace (Al, Mn, Fe, Ni, Cu, Zn, Se, Cr, and Sc) elements in various parts (leaves, roots, and seeds) of the 16 anti-diabetic medicinal plants studied. Specifically, elements such as Ca, K, Cr, Cu, Mn, Zn, and Se were detected, all of which are known to contribute in maintaining normal glucose metabolism. Notably, Zn and Se are crucial trace elements for the synthesis, secretion, and action of insulin. Significant Zn concentrations were observed in ten anti-diabetic medicinal plants: Albizia lebbeck (AL), Atropa acuminata (AA), Avena fatua (AF), Citrus medica (CM), Commiphora wightii (CW), Cymbopogon citratus (CC), Daucus carota (DC), Ziziphus mauritiana (FM), Hyoscyamus niger (HN), and Martynia annua (MA), and significant Se concentrations were observed in twelve medicinal plants, i.e., Albizia lebbeck (AL), Allium sativum (AS), Atropa acuminata (AA), Avena fatua (AF), Cannabis sativa (CS), Capparis spinosa (CaS), Commiphora wightii (CW), Cymbopogon citratus (CC), Datura alba (DA), Daucus carota (DC), Ziziphus mauritiana (FM), and Hyoscyamus niger (HN). Our study's elemental analysis using PIXE and ICP-OES on various parts of 16 medicinal plants identified a significant number of useful elements. Elements such as Ca, K, S, P, Al, Si, Cl, Mn, Fe, Ni, Cu, Zn, Se, and Cr were identified and quantified. These findings support the potential use of these plants in managing diabetes and highlight the importance of elemental profiling in understanding their therapeutic properties.

Abstract:Seven plant certified reference materials (NIST SRM1515 Apple Leaves, NIST SRM1547 Peach Leaves, BCR-129 Hay Powder, BCR-670 Aquatic Plant, GBW07603 Bush Twigs and Leaves, GBW10015 Spinach Leaves and NCS ZC73036a Green Tea) were analysed for their mass fractions of 48 elements by inductively coupled plasma tandem-mass spectrometry (ICP-MS/MS): Li, Be, Na, Mg, Al, Ca, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, Ge, As, Se, Rb, Sr, Y, Nb, Mo, Ag, Cd, Sb, Te, Ba, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu, Ta, Tl, Pb, Bi, Th, U. Special focus was put on the determination of technology-critical elements (TCEs), to which, e.g. Li, Be, Ga, Ge, Nb, Sb, Ta, Tl, Bi, and the rare-earth elements (REEs, lanthanides and Y) are counted. Closed-vessel microwave digestion was performed using HNO 3 , H 2 O 2 and HBF 4 . The average bias for certified values is − 1% ± 13% ( SD ). Limits of detection ( x L ) in the measured solutions lie between 13 fg g −1 (Tb) and 52 ng g −1 (Ca). This article seeks to provide an optimised measurement procedure for the determination of element mass fractions of emerging importance in environmental samples, which are challenging to analyse with more traditional techniques such as single-quad ICP-MS. In addition, it aims to improve the characterisation of commonly used plant reference materials by providing mass fraction data for rarely studied elements.

Abstract:There is considerable controversy regarding whether angiotensin II (AngII) stimulates hypertrophy or hyperplasia of vascular smooth muscle cells (SMC). The purpose of the present study was to determine whether stretch of the vessel wall or AngII treatment increased protein or DNA synthesis in intact aortic rings in vitro and whether stretch of the vessel wall altered the growth responses to AngII. Rat aortic rings were mounted on steel supports in serum-free medium for 16 h and subjected to 0 or 1.5 g of preload (i.e. passive stretch). Fetal bovine serum (13%, FBS) or AngII [1 microM, in the presence or absence of angiotensin receptor antagonist, losartan (DuP753) 10 microM] was administered and isometric tension development was measured. 35S-methionine (3 microCi/ml) was added to the baths at 14-16 h for measurement of protein synthesis. Passive stretch did not increase protein synthesis as compared to vessels mounted under no-preload conditions. AngII and FBS elicited similar increases in isometric tension development, but tension development in FBS-treated rings was sustained 4 times longer than in rings treated with AngII. AngII and FBS increased protein synthesis by 35 and 121%, respectively, but there was no difference in the extent of contractile agonist-induced protein synthesis between rings subjected to 0 or 1.5 g of passive stretch. Losartan totally abolished AngII-induced tension development and protein synthesis. AngII and FBS did not stimulate increased DNA synthesis in aortic rings, as measured by 3H-thymidine incorporation. These results suggest that AngII stimulates hypertrophy rather than hyperplasia of fully contractile SMC in an intact vessel.
