Abstract:Drought stress hinders the growth and development of crop plants and ultimately, its productivity. It is expected that drought stress will be frequent and intense in the future due to drastic changes in the global climate. It is necessary to make crop plants more resilient to drought stress through various techniques; drought‐hardening is one of them. Defining various metabolic strategies used by tobacco plants to confer drought tolerance will be important for maintaining plant physiological functions, but studies addressing this topic are limited. This study was designed to elucidate the drought tolerance and adaptation strategies used by tobacco via the application of different circular drought‐hardening cycles (control: no drought‐hardening, T1: one cycle of drought hardening, T2: two cycles of drought‐hardening, and T3: three cycles of drought‐hardening) to two tobacco varieties namely Honghuadajinyuan (H) and Yun Yan‐100 (Y). The results revealed that drought‐hardening decreased the fresh and dry biomass of the plant. The decrease was more pronounced in the T3 treatment for both H (23 and 29%, respectively) and Y (26 and 31%, respectively) under drought stress. The MDA contents, especially in T1 and T2 in both varieties, were statistically similar compared with control under drought stress. Similarly, higher POD, APX, and GR activities were observed, especially in T3, and elevated amounts of AsA and GSH were also observed among the different circular drought‐hardening treatments under drought stress. Thus drought‐hardening mitigated the oxidative damage by increasing the antioxidant enzyme activities and elevated the content of antioxidant substances, a key metabolic strategy under drought stress. Similarly, another important plant metabolic strategy is the osmotic adjustment. Different drought‐hardening treatments improved the accumulation of proline and soluble sugars contents which contributed to osmoregulation. Finally, at the molecular level circular drought‐hardening improved the transcript levels of antioxidant enzyme‐related genes (CAT, APX1, and GR2), proline and polyamines biosynthesis‐related genes (P5CS1 and ADC2), and ABA signalling (SnRK2), and transcription factors (AREB1 and WRKY6) in response to drought stress. As a result, circular drought‐hardening (T2 and T3 treatments) promoted tolerance to water stress via affecting the anti‐oxidative capacity, osmotic adjustment, and regulation of gene expression in tobacco.

Abstract:The drought tolerance of 20 tobacco cultivars were investigated by determining traits of germination characteristics,seedling quality and some physiological and biochemical characteristics under normal conditions and simulated drought stress.The ratios(relative values) of trait values under drought stress to those under normal conditions were used to assess drought tolerance.The correlation analyses showed that the relative values of germination index,vigor index,seedling fresh and dry weight were significantly correlated to each other,these parameters were suitable for evaluating drought tolerance.After the cluster analysis,20 tobacco cultivars were grouped into three classes: drought tolerance,intermediate drought tolerance and drought sensitivity,the results were consistent with that of the correlation analysis.The results would provide theory basis for research of drought tolerance mechanism,cultivation and breeding in tobacco.

Abstract:Drought is one of the most severe forms of abiotic stresses that threaten the survival of plants, including crops. In turn, plants dramatically change their physiology to increase drought tolerance, including reconfiguration of proteomes. Here, we studied drought-induced proteomic changes in leaves of cultivated tobacco (Nicotiana tabacum), a solanaceous plant, using the isobaric tags for relative and absolute quantitation (iTRAQ)-based protein labeling technology. Of identified 5570 proteins totally, drought treatment increased and decreased abundance of 260 and 206 proteins, respectively, compared with control condition. Most of these differentially regulated proteins are involved in photosynthesis, metabolism, and stress and defense. Although abscisic acid (ABA) levels greatly increased in drought-treated tobacco leaves, abundance of detected ABA biosynthetic enzymes showed no obvious changes. In contrast, heat shock proteins (HSPs), thioredoxins, ascorbate-, glutathione-, and hydrogen peroxide (H2O2)-related proteins were up- or down-regulated in drought-treated tobacco leaves, suggesting that chaperones and redox signaling are important for tobacco tolerance to drought, and it is likely that redox-induced posttranslational modifications play an important role in modulating protein activity. This study not only provides a comprehensive dataset on overall protein changes in drought-treated tobacco leaves, but also shed light on the mechanism by which solanaceous plants adapt to drought stress.

Abstract:Drought stress usually causes huge economic losses for tobacco industries. Drought stress exhibits multifaceted impacts on tobacco systems through inducing changes at different levels, such as physiological and chemical changes, changes of gene transcription and metabolic changes. Understanding how plants respond and adapt to drought stress helps generate engineered plants with enhanced drought resistance. In this study, we conducted multiple time point-related physiological, biochemical,transcriptomic and metabolic assays using K326 and its derived mutant 28 (M28) with contrasting drought tolerance. Through integrative analyses of transcriptome and metabolome,we observed dramatic changes of gene expression and metabolic profiles between M28 and K326 before and after drought treatment. we found that some of DEGs function as key enzymes responsible for ABA biosynthesis and metabolic pathway, thereby mitigating impairment of drought stress through ABA signaling dependent pathways. Four DEGs were involved in nitrogen metabolism, leading to synthesis of glutamate (Glu) starting from NO−3 /NO−2 that serves as an indicator for stress responses. Importantly, through regulatory network analyses, we detected several drought induced TFs that regulate expression of genes responsible for ABA biosynthesis through network, indicating direct and indirect involvement of TFs in drought responses in tobacco. Thus, our study sheds some mechanistic insights into how plant responding to drought stress through transcriptomic and metabolic changes in tobacco. It also provides some key TF or non-TF gene candidates for engineering manipulation for breeding new tobacco varieties with enhanced drought tolerance.

Abstract:Chilling is one of the major abiotic stresses limiting yield and quality of many important crops. For better understanding of chilling stress responses in tobacco (Nicotiana tabacum), growth rate and antioxidant enzymes of seedlings in 2 tobacco cultivars, viz., MSk326 (chilling sensitive variety) and Honghuadajinyuan (HHDJY, chilling tolerant variety) at chilling temperature (5÷C) were studied. The results showed that the relative growth rate in chilling period to that in recovery period was significantly higher in roots than that in shoots for both cultivars, suggesting that shoots growth was more easily affected by chilling stress. Chilling stress increased peroxidase (POD) activity and reduced superoxide dismutase (SOD) activity in shoots of HHDJY, and catalase (CAT) activity was little affected. In the roots of HHDJY, chilling stress increased SOD and CAT activities, and had little effect on POD activity. For MSk326, chilling treatment increased SOD activity in shoots and declined CAT activity in roots. MDA concentration in both varieties was increased under the chilling stress, while it was decreased after seedlings were recovered growth for 4 d at normal temperature (25÷C). It showed that tobacco seedlings might have the capacity of recovering from chilling injury for a short term. The relationship between the growth rate and antioxidant enzyme activity was analyzed by stepwise regression. It was found that there was a close relationship between relative growth rate of tobacco seedlings and CAT activity under chilling stress condition and regression equations containing CAT could be used in predicting seedling growth rate of tobacco under chilling stress condition.

Abstract:Background: In recent years, more severe droughts have occurred frequently in many parts of the world, drought stress is the primary abiotic stress factor restricting the growth and quality of flue-cured tobacco. Therefore, screening dryland cultivation-compatible flue-cured tobacco varieties will help reduce the negative impact of drought. Methods: Tobacco varieties were selected: Qinyan 96 (Q96), Zhongyan 101 (Z101), Yunyan 87 (Y87), and Yunyan 116 (Y116). A pot experiment was conducted with four water supply gradients: sufficient, mild stress, moderate stress, and severe stress. The aim was to analyze inter-varietal differences in agronomic traits, photosynthetic traits, reactive oxygen species (ROS) metabolism, and antioxidant enzyme system under drought stress. Additionally, the drought resistance of four flue-cured tobacco varieties was evaluated using principal component analysis and membership function analysis. Results: The results showed that drought intensification inhibited seedling growth and development across all varieties, with Q96 showing the least decrease and Y116 the greatest. With the increasing degree of drought stress, photosynthetic rates (Pn), transpiration rate (Tr), and stomatal conduction (Gs) have shown gradually decreasing trends, while substomatal cavity CO2 concentration (Ci) showed a growing trend. Severe drought corresponded with lower chlorophyll content and decreased the maximal photochemical efficiency (Fv/Fm), photosystem II (PSII), and photochemical quenching coefficient (qP) in all varieties, while steady-state non-photochemical quenching (NPQ) increased. Increased drought stress led to significantly higher reactive oxygen species (ROS) and malondialdehyde (MDA) content accumulation in tobacco seedlings. The antioxidant enzyme activities in, Q96, Z101, and Y87 increased under mild drought stress, whereas Y116 showed decreased activity. Conclusion: The drought resistance ranking among the four varieties is as follows: Q96 > Z101 > Y87 > Y116. Therefore, Q96 is a promising drought-tolerant breeding material that can be used as a reference for dryland cultivation of flue-cured tobacco.
Co-inoculation of Arbuscular Mycorrhizal Fungi and the Plant Growth-Promoting Rhizobacteria Improve Growth and Photosynthesis in Tobacco Under Drought Stress by Up-Regulating Antioxidant and Mineral Nutrition Metabolism

Abstract:Drought stress is a major environmental concern that limits crop growth on a large scale around the world. Significant efforts are required to overcome this issue in order to improve crop production. Therefore, the exciting role of beneficial microorganisms under stress conditions needs to be deeply explored. In this study, the role of two biotic entities, i.e., Arbuscular mycorrhizal fungi (AMF, Glomus versiforme) and plant growth-promoting rhizobacteria (PGPR, Bacillus methylotrophicus) inoculation in drought tolerance of tobacco (Nicotiana tabacum L.), was investigated. The present results showed that drought stress considerably reduced tobacco plant's growth and their physiological attributes. However, the plants co-inoculated with AMF and PGPR showed higher drought tolerance by bringing up significant improvement in the growth and biomass of tobacco plants. Moreover, the co-inoculation of AMF and PGPR considerably increased chlorophyll a, b, total chlorophylls, carotenoids, photosynthesis, and PSII efficiency by 96.99%, 76.90%, and 67.96% and 56.88%, 53.22%, and 33.43% under drought stress conditions, respectively. Furthermore, it was observed that drought stress enhanced lipid peroxidation and electrolyte leakage. However, the co-inoculation of AMF and PGPR reduced the electrolyte leakage and lipid peroxidation and significantly enhanced the accumulation of phenols and flavonoids by 57.85% and 71.74%. Similarly, the antioxidant enzymatic activity and the plant nutrition status were also considerably improved in co-inoculated plants under drought stress. Additionally, the AMF and PGPR inoculation also enhanced abscisic acid (ABA) and indole-3-acetic acid (IAA) concentrations by 67.71% and 54.41% in the shoots of tobacco plants. The current findings depicted that inoculation of AMF and PGPR (alone or in combination) enhanced the growth and mitigated the photosynthetic alteration with the consequent up-regulation of secondary metabolism, osmolyte accumulation, and antioxidant system.

Abstract:Drought stress inhibits seed germination, plant growth and development of tobacco, and seriously affects the yield and quality of tobacco leaves. However, the molecular mechanism underlying tobacco drought stress response remains largely unknown. In this study, integrated analysis of transcriptome and metabolome was performed on the germinated seeds of a cultivated variety K326 and its EMS mutagenic mutant M28 with great drought tolerance. The result showed that drought stress inhibited seed germination of the both varieties, while the germination rate of M28 was faster than that of K326 under drought stress. Besides, the levels of phytohormone ABA, GA19, and zeatin were increased by drought stress in M28. Five vital pathways were identified through integrated transcriptomic and metabolomic analysis, including zeatin biosynthesis, aspartate and glutamate synthesis, phenylamine metabolism, glutathione metabolism, and phenylpropanoid synthesis. Furthermore, 20 key metabolites in the above pathways were selected for further analysis of gene modular-trait relationship, and then four highly correlated modules were found. Then analysis of gene expression network was carried out of Top30 hub gene of these four modules, and 9 key candidate genes were identified, including HSP70s, XTH16s, APX, PHI-1, 14-3-3, SCP, PPO. In conclusion, our study uncovered some key drought-responsive pathways and genes of tobacco during seeds germination, providing new insights into the regulatory mechanisms of tobacco drought stress response.

Effects of Drought Stress on Photosynthesis and Antioxidant Properties of Tobacco in Budding Stage

Abstract:In order to study the response of different tobacco cultivars to drought stress,NC89 and Yunyan 97(YY 97) were used as the plant material to analyze the physiological and biochemical changes of photosynthetic indicators and antioxidant enzyme(SOD,POD) activities and the content of chlorophyll,protein,MDA,Pro,GSH through pot experiment under two kinds of drought stress treatments in the budding stage.The results showed that under drought stress,each index changed greatly,but only the chlorophyll and Pro content,and the MDA content of NC89,GSH content of YY97 reached the significant difference level.The transpiration rate,stomatal conductance and net photosynthetic rate of tobacco decreased;intercellular CO2 concentration decreased firstly and then increased;SOD activity,chlorophyll,protein and Pro content,increased;POD activity,MDA and GSH content showed different responses in different tobacco cultivars.As a conclusion,NC89 was more drought-resistant than YY97.

Arbuscular mycorrhizal fungi improve growth, essential oil, secondary metabolism, and yield of tobacco (Nicotiana tabacum L.) under drought stress conditions

Abstract:Drought is a major environmental threat limiting worldwide crop production. Drought stress affects the tobacco quality and yield; therefore, the current research studies were undertaken to investigate the effectiveness of arbuscular mycorrhizal fungi (AMF) under drought stress on morphological and biochemical attributes of tobacco (Nicotiana tabacum L. variety Yunyan 87). AMF-inoculated and AMF-non-inoculated plants were maintained in a greenhouse and irrigated with a half-strength Hoagland solution (100 mL pot-1) once a week. At harvesting, the plant height, number of leaves, fresh and dry weights, mycorrhizal colonization, and concentration of leaf photosynthetic pigments and photosynthetic rate were measured. Data were statistically analyzed by ANOVA and the principal component (PCA) analyses. The effect of root colonization significantly increased biomass production and essential oil accumulation. Results showed that drought at mild and severe stressed levels significantly affected tobacco growth by decreasing plant height, biomass, and a number of leaves. However, inoculation of AMF considerably increased plant height, fresh and dry weights, chlorophyll (a, b), total chlorophyll, and carotenoid content by 43.84, 40.87 and 49.76, 185.29, 325.60, 173.12, and 211.49%, respectively. Compared with non-inoculated plants, AMF inoculation significantly enhanced the essential oil yield and the uptake of nitrogen, phosphorus, and potassium with the increase of 257.36, 102.71, and 90.76, 62.32, and 84.51%, respectively, in mild drought + AMF-treated plants. Similarly, the antioxidant enzymatic activity, glomalin-related soil protein (GRSP), and accumulation of phenols and flavonoids and osmolytes content were also significantly improved in inoculated plants under drought stress. Additionally, AMF inoculation significantly upregulated the lipoxygenase (LOX) and phenylalanine ammonia-lyase (PAL) enzymes by 197 and 298.44% under drought conditions. These findings depicted that the symbiotic association of AMF improved the overall growth pattern and secondary metabolism in tobacco plants under severe drought stress conditions and may be used as an approaching source of important drugs in the field of pharmacology.

Abstract:Heat stress is a prevalent factor that significantly damages crops, especially with the ongoing global warming and increasing frequency of extreme weather events. Tobacco is particularly sensitive to temperature fluctuations, experiencing reduced yield and quality under high temperatures. However, the underlying molecular mechanisms of heat resistance in tobacco remain poorly understood. This study comprehensively analyzed biochemical, transcriptomic, and metabolomic responses to heat stress on the root and shoot of the tobacco cultivar K326 compared to control conditions. Heat stress significantly increased the activities of antioxidant enzymes (CAT, POD, and SOD) and levels of osmotic mediators (soluble sugars, sucrose, and proline) in the shoot. Furthermore, transcriptome analysis identified 13,176 differentially expressed genes (DEGs) in the root (6,129 up-regulated and 7,047 down-regulated) and 12,283 DEGs (6,621 up-regulated and 5,662 down-regulated) in the shoot. The root had 24 enriched KEGG pathways, including phenylpropanoid metabolism, while the shoot had 32 significant pathways, such as galactose metabolism and MAPK signaling. The metabolomic data identified 647 metabolites in the root and 932 in the shoot, with carbohydrates and amino acids being the main categories. The root had 116 differentially abundant metabolites (DAMs) (107 up-regulated and 9 down-regulated), and the shoot contained 256 DAMs (251 up-regulated and 5 down-regulated). Joint transcriptome and metabolome analysis showed that galactose metabolism and starch and sucrose metabolism were co-enriched in both tissues. In contrast, amino sugar and nucleotide sugar metabolism was enriched in the root, and purine metabolism in the shoot. The purine metabolic pathway in the shoot can modulate the expression of MYB transcription factors by influencing ABA synthesis and signaling, thereby controlling the accumulation of HSPs, raffinose, sucrose, and trehalose to enhance heat tolerance. Furthermore, NtMYB78, an MYB transcription factor, enhances tolerance for heat stress in tobacco. This research offers a foundational framework for investigating and implementing heat-resistant genes and metabolic pathways in the root and shoot of tobacco seedlings.

Abstract:The development and growth of plants are significantly impacted by adverse surroundings, particularly drought conditions. The yield and quality of plants, in particular, are heavily reliant on the presence of favorable growth conditions. Here, we performed comprehensive research to investigate phenotype, physiological characteristics, transcriptomic and metabolomic changes in Nicotiana tabacum (N. tabacum) in responses to drought stress (DS). This work aimed to investigate the detailed responses of N. tabacum to DS under different drought conditions (CK, well-watered; LD, light drought; MD, moderate drought and SD, severe drought). N. tabacum grew normally under CK but was inhibited under LD, MD and SD stress; the relative water content, transpiration rate and protective enzyme activity significantly influenced under DS. In the LD/CK, MD/CK and SD/CK comparison groups, there were 7483, 15558 and 16876 differentially expressed genes (DEGs), respectively, and 410, 485 and 523 differentially accumulated metabolites (DAMs), respectively. The combined analysis of transcriptomic and metabolomic data unveiled the significant involvement of phenylpropanoid biosynthesis in the N. tabacum's response to drought stress. These findings characterized the key metabolites and genes in responses to drought stress in N. tabacum, hence offering valuable insights into the underlying mechanisms driving these responses to DS and maintaining plant health under climate change.

Abstract:Drought stress is one of the primary environmental stress factors that gravely threaten crop growth, development, and yields. After drought stress, plants can regulate the content and proportion of various hormones to adjust their growth and development, and in some cases to minimize the adverse effects of drought stress. In our previous study, the tobacco cis -abienol synthesis gene ( NtCPS2 ) was found to affect hormone synthesis in tobacco plants. Unfortunately, the role of NtCPS2 genes in the response to abiotic stress has not yet been investigated. Here, we present data supporting the role of NtCPS2 genes in drought stress and the possible underlying molecular mechanisms. NtCPS2 gene expression was induced by polyethylene glycol, high-temperature, and virus treatments. The results of subcellular localization showed that NtCPS2 was localized in the cell membrane. The NtCPS2 -knockdown plants exhibited higher levels of gibberellin (GA) content and synthesis pathway genes expression but lower abscisic acid (ABA) content and synthesis pathway genes expression in response to drought stress. In addition, the transgenic tobacco lines showed higher leaf water loss and electrolyte loss, lower soluble protein and reactive oxygen species content (ROS), and lower antioxidant enzyme activity after drought treatment compared to wild type plants (WT). In summary, NtCPS2 positively regulates drought stress tolerance possibly by modulating the ratio of GA to ABA, which was confirmed by evidence of related phenotypic and physiological indicators. This study may provide evidence for the feedback regulation of hormone to abiotic and biotic stresses.

Pot Level Drought Stress Tolerance Assay in Tobacco through Plant Phenotyping and Antioxidant Assay

Abstract:[Abstract] Drought is an important abiotic factor which has a huge detrimental impact on crop productivity. Study of plant responses towards drought stress and investigating the mechanism of drought tolerance is crucial for achieving the target of developing drought-tolerant plants. Phenotyping is a cost effective approach which can be adopted to evaluate the severity of drought stress in a plant. Next to phenotyping parameters, biochemical parameters such as the study of antioxidant enzyme activity play significant roles in assessing the extent of drought stress caused injury in a plant. Among the antioxidant enzymes, ascorbate peroxidase is an enzyme which plays a crucial role in drought tolerance in plants. It has been well established that the activity of this enzyme increases under drought stress. Here, we present a simple and reproducible protocol to investigate the response of tobacco plants towards drought stress through measurement of phenotypic parameters and antioxidant enzyme activity. Though, these experiments have been conducted with tobacco plants, this protocol could be adopted for other crop species.

Combined application of biochar and peatmoss for mitigation of drought stress in tobacco

Abstract:Drought poses a significant ecological threat that limits the production of crops worldwide. The objective of this study to examine the impact of soil applied biochar (BC) and peatmoss (PM) on the morpho-biochemical and quality traits of tobacco plants under drought conditions. In the present experiment work, a pot trial was conducted with two levels of drought severity (~ well-watered 75 ± 5% field capacity) and severe drought stress (~ 35 ± 5% field capacity), two levels of peatmoss (PM) @ 5% [PM+ (with peatmoss) and PM- (without peatmoss)] and three levels of rice straw biochar (BC 0 = no biochar; BC 1 = 150 mg kg − 1 ; and BC 2 = 300 mg kg − 1 of soil) in tobacco plants. The results indicate that drought conditions significantly impacted the performance of tobacco plants. However, the combined approach of BC and PM significantly improved the growth, biomass, and total chlorophyll content (27.94%) and carotenoids (32.00%) of tobacco. This study further revealed that the drought conditions decreased the production of lipid peroxidation and proline accumulation. But the synergistic approach of BC and PM application increased soluble sugars (17.63 and 12.20%), soluble protein (31.16 and 15.88%), decreased the proline accumulation (13.92 and 9.03%), and MDA content (16.40 and 8.62%) under control and drought stressed conditions, respectively. Furthermore, the combined approach of BC and PM also improved the leaf potassium content (19.02%) by limiting the chloride ions (33.33%) under drought stressed conditions. Altogether, the balanced application of PM and BC has significant potential as an effective approach and sustainable method to increase the tolerance of tobacco plants subjected to drought conditions. This research uniquely highlights the combined potential of PM and BC as an eco-friendly strategy to enhance plant resilience under drought conditions, offering new insights into sustainable agricultural practices.

Abstract:Key message The expression of BdWRKY36 was upregulated by drought treatment. BdWRKY36 -overexpressing transgenic tobacco increased drought tolerance by controlling ROS homeostasis and regulating transcription of stress related genes. Abstract WRKY transcription factor plays important roles in plant growth, development and stress response. However, the function of group IIe WRKYs is less known. In this study, we cloned and characterized a gene of group IIe WRKY , designated as BdWRKY36 , from Brachypodium distachyon . Transient expression of BdWRKY36 in onion epidermal cell suggested its localization in the nucleus. Transactivation assay revealed that the C-terminal region, instead of full length BdWRKY36, had transcriptional activity. BdWRKY36 expression was upregulated by drought. Overexpression of BdWRKY36 in transgenic tobacco plants resulted in enhanced tolerance to drought stress. Physiological–biochemical indices analyses showed that BdWRKY36 -overexpressing tobacco lines had lesser ion leakage (IL) and reactive oxygen species (ROS) accumulation, but higher contents of chlorophyll, relative water content (RWC) and activities of antioxidant enzyme than that in control plants under drought condition. Meanwhile expression levels of some ROS-scavenging and stress-responsive genes were upregulated in BdWRKY36 -overexpressing tobacco lines under drought stress. These results demonstrate that BdWRKY36 functions as a positive regulator of drought stress response by controlling ROS homeostasis and regulating transcription of stress related genes.
Mechanistic basis for mitigating drought tolerance by selenium application in tobacco (Nicotiana tabacum L.): a multi-omics approach

Abstract:The lack of irrigation water in agricultural soils poses a significant constraint on global crop production. In-depth investigation into microRNAs (miRNAs) has been widely used to achieve a comprehensive understanding of plant defense mechanisms. However, there is limited knowledge on the association of miRNAs with drought tolerance in cigar tobacco. In this study, a hydroponic experiment was carried out to identify changes in plant physiological characteristics, miRNA expression and metabolite profile under drought stress, and examine the mitigating effects of selenium (Se) application. The shoot dry weight of drought-stressed plants was approximately half (50.3%) of that in non-stressed (control) conditions. However, plants supplied with Se attained 38.8% greater shoot dry weight as compared to plants with no Se supply under drought stress. Thirteen miRNAs were identified to be associated with drought tolerance. These included 7 known (such as nta-miR156b and nta-miR166a) and 6 novel miRNAs (such as novel-nta-miR156-5p and novel-nta-miR209-5p) with the target genes of squamosa promoter-binding-like protein 4 ( SPL4 ), serine/threonine protein phosphatase 2A ( PPP2A ), cation/calcium exchanger 4-like ( CCX4 ), extensin-1-like ( EXT1 ) and reduced wall acetylation 2 ( RWA2 ). Further investigation revealed that the expression levels of Ext1 and RWA2 were significantly decreased under drought stress but increased with Se addition. Moreover, key metabolites such as catechin and N-acetylneuraminic acid were identified, which may play a role in the regulation of drought tolerance. The integrated analysis of miRNA sequencing and metabolome highlighted the significance of the novel-nta-miR97-5p- LRR-RLK - catechin pathway in regulating drought tolerance. Our findings provide valuable insights into the molecular mechanisms underlying drought tolerance and Se-induced stress alleviation in cigar tobacco.

Abstract:Main conclusion TaGF14b enhances tolerance to multiple stresses through ABA signaling pathway by altering physiological and biochemical processes, including ROS-scavenging system, stomatal closure, compatible osmolytes, and stress-related gene expressions in tobaccos. The 14-3-3 proteins are involved in plant growth, development, and in responding to abiotic stresses. However, the precise functions of 14-3-3s in responding to drought and salt stresses remained unclear, especially in wheat. In this study, a 14 - 3 - 3 gene from wheat, designated TaGF14b , was cloned and characterized. TaGF14b was upregulated by polyethylene glycol 6000, sodium chloride, hydrogen peroxide, and abscisic acid (ABA) treatments. Ectopic expression of TaGF14b in tobacco conferred enhanced tolerance to drought and salt stresses. Transgenic tobaccos had longer root, better growth status, and higher relative water content, survival rate, photosynthetic rate, and water use efficiency than control plants under drought and salt stresses. The contribution of TaGF14b to drought and salt tolerance relies on the regulations of ABA biosynthesis and ABA signaling, as well as stomatal closure and stress-related gene expressions. Moreover, TaGF14b expression could significantly enhance the reactive oxygen species (ROS) scavenging system to ameliorate oxidative damage to cells. In addition, TaGF14b increased tolerance to osmotic stress evoked by drought and salinity through modifying water conservation and compatible osmolytes in plants. In conclusion, TaGF14b enhances tolerance to multiple abiotic stresses through the ABA signaling pathway in transgenic tobaccos by altering physiological and biochemical processes.

Abstract:Drought stress is an abiotic stressor that impacts photosynthesis, plant growth, and development, leading to decreased crop yields. Sodium hydrosulfide (NaHS), an exogenous additive, has demonstrated potential regulatory effects on plant responses to polyethylene glycol-induced drought stress in tobacco seedlings. Compared to the control, drought stress induced by 15 g/L PEG-6000 significantly reduced several parameters in tobacco seedlings: shoot dry weight (22.83%), net photosynthesis (37.55%), stomatal conductance (33.56%), maximum quantum yield of PSII (Fv/Fm) (11.31%), photochemical quantum yield of PSII (ΦPSII) (25.51%), and photochemical quenching (qP) (18.17%). However, applying NaHS, an H 2 S donor, mitigated these effects, ultimately enhancing photosynthetic performance in tobacco seedlings. Furthermore, optimal NaHS concentration (0.4 mM) effectively increased leaf stomatal aperture, relative water content (RWC) and root activity, as well as facilitated the absorption of N, K, Mg and S. It also enhanced the accumulation of soluble sugar and proline content to maintain osmotic pressure balance under drought stress. Compared to drought alone, pretreatment with NaHS also bolstered the antioxidant defense system in leaves, leading to 22.93% decrease in hydrogen peroxide (H 2 O 2 ) content, a 22.19% decrease in malondialdehyde (MDA) content and increased activities of ascorbate peroxidase (APX) by 28.13%, superoxide dismutase (SOD) by 17.07%, peroxidase (POD) by 46.99%, and catalase (CAT) by 65.27%. Consequently, NaHS protected chloroplast structure and attenuated chlorophyll degradation, thus mitigating severe oxidative damage. Moreover, NaHS elevated endogenous H 2 S levels, influencing abscisic acid (ABA) synthesis and the expression of receptor-related genes, collaboratively participating in the response to drought stress. Overall, our findings provide valuable insights into exogenous NaHS's role in enhancing tobacco drought tolerance. These results lay the foundation for further research utilizing H 2 S-based treatments to improve crop resilience to water deficit conditions.

Abstract:Organic acids play a pivotal role in improving plant response to long-term drought stress. External application of organic acids has been reported to improve drought resistance in several species. However, whether organic acids have similar effects in tobacco remains unknown. A screening study of the protective function of organic acids in tobacco and understanding the underlying molecular mechanism would be useful in developing a strategy for drought tolerance. Several physiological and molecular adaptations to drought including abscisic acid, stomatal closure, reactive oxygen species homeostasis, amino acid accumulation, and drought-responsive gene expression were observed by exogenous citric acid in tobacco plants. Exogenous application of 50 mm citric acid to tobacco plants resulted in higher chlorophyll content, net photosynthesis, relative water content, abscisic acid content and lower stomatal conductance, transpiration and water loss under drought conditions. Moreover, reactive oxygen species homeostasis was better maintained through increasing activity of antioxidant enzymes and decreasing hydrogen peroxide content after citric acid pretreatment under drought. Amino acids involved in the TCA cycle accumulated after external application of citric acid under drought stress. Furthermore, several drought stress-responsive genes also dramatically changed after application of citric acid. These data support the idea that external application of citric acid enhances drought resistance by affecting physiological and molecular regulation in tobacco. This study provides clear insights into mechanistic details of regulation of amino acid and stress-responsive gene expression by citric acid in tobacco in response to drought, which is promising for minimizing growth inhibition in agricultural fields.
