Abstract:The enigmatic association of photosynthetically active chloroplasts from algae and some sacoglossan sea slugs, called functional kleptoplasty, is a functional unique system of photosymbioses observed in metazoans. Besides the specific adaptations of the slugs necessary to incorporate and maintain the plastids, the organelles need to ensure optimal photosynthesis. Photoprotective mechanisms in the plastids, namely the xanthophyll cycle (XC) and the high-energy dependent quenching (qE) part of the non-photochemical quenching (NPQ), and the repair of the D1 protein of Photosystem II (PSII) are considered crucial for kleptoplast longevity in the slugs. Here, we studied the sea slugs Elysia viridis fed with the naturally occurring XC and qE-deficient Bryopsis hypnoides, and E. timida fed on Acetabularia acetabulum, an alga that possesses both mechanisms. The aim of the study was to understand (i) whether qE remains active after ingestion of kleptoplasts by E. timida, (ii) how different light intensities affect the photosynthetic activity of kleptoplasts with and without photoprotection mechanisms, and (iii) if the kleptoplasts are able to repair photodamaged D1 protein. With regard to NPQ, freshly incorporated kleptoplasts responded to different light stress in the same manner as the chloroplasts in their native host algae. Even after 3 weeks of incorporation the qE part of NPQ was present in the kleptoplasts of E. timida. However, the presence of the qE component of NPQ did not prevent the kleptoplasts from significant PSII photoinactivation under high light intensities. This is probably due to the fact that the kleptoplasts have a reduced PSII repair capacity, despite plastid encoded repair mechanisms in every sacoglossan food source. Hence, photoprotective mechanisms are probably not a key factor explaining kleptoplast longevity in Sacoglossa sea slugs.

Abstract:Photosynthesizers face a trade-off regarding light: they need enough to maintain high photosynthetic rates, yet excess leads to oxidative stress. Despite this, light and its detrimental effects are chronically underestimated. Solar-powered sea slugs (Sacoglossa: Gastropoda) provide the ideal lens with which to study this trade-off, since they steal chloroplasts from algae but do not inherit photoacclimation and photorepair capacities. We collected three slug species in Curaçao during March and December 2022, comparing the amount of light they received in nature to their optimal light intensities for photosynthesis, and their preferred light intensities. We then investigated behavioral and physiological photoprotection mechanisms to determine if and how they limit light. Finally, we examined oxidative activity under optimal and excess light. All three species were naturally exposed to more light (> 1000 μmol m −2 s −1 ) than is optimal or preferred. Elysia crispata (kleptoplast retention for > 3 months) is fully exposed to light in nature but reduces the light reaching its kleptoplasts via parapodial shading. Elysia velutinus retains kleptoplasts for ~ 2 weeks and hides in its macroalgal food, limiting light exposure. Both species displayed low amounts of oxidative activity under optimal light, which increased slightly under excess light. Elysia ornata retained chloroplasts for ~ 3 days, lacked observable photoprotection and always displayed high levels of oxidative activity, potentially explaining its limited capacity for kleptoplast retention. Furthermore, both E. velutinus and E. ornata display strong light-avoidance behaviors. This study clearly demonstrates links between high light intensities, photoprotection, and oxidative stress, highlighting the need for future studies that examine aquatic photosynthesizers under natural lighting.

Abstract:Sacoglossan sea slugs are unique in the animal kingdom in that they sequester and maintain active plastids that they acquire from the siphonaceous algae upon which they feed, making the animals photosynthetic. Although most sacoglossan species digest their freshly ingested plastids within hours, four species from the family Plakobranchidae retain their stolen plastids (kleptoplasts) in a photosynthetically active state on timescales of weeks to months. The molecular basis of plastid maintenance within the cytosol of digestive gland cells in these photosynthetic metazoans is yet unknown but is widely thought to involve gene transfer from the algal food source to the slugs based upon previous investigations of single genes. Indeed, normal plastid development requires hundreds of nuclear-encoded proteins, with protein turnover in photosystem II in particular known to be rapid under various conditions. Moreover, only algal plastids, not the algal nuclei, are sequestered by the animals during feeding. If algal nuclear genes are transferred to the animal either during feeding or in the germ line, and if they are expressed, then they should be readily detectable with deep-sequencing methods. We have sequenced expressed mRNAs from actively photosynthesizing, starved individuals of two photosynthetic sea slug species, Plakobranchus ocellatus Van Hasselt, 1824 and Elysia timida Risso, 1818. We find that nuclear-encoded, algal-derived genes specific to photosynthetic function are expressed neither in P. ocellatus nor in E. timida. Despite their dramatic plastid longevity, these photosynthetic sacoglossan slugs do not express genes acquired from algal nuclei in order to maintain plastid function.

Abstract:Green macroalgae within the order Bryopsidales lack the fundamental photoprotective mechanisms of green algae, the xanthophyll cycle and energy-dependent dissipation of excess light. Here, by measuring chlorophyll fluorescence at 77K after specific light treatments, we show that Bryopsidales algae also lack state transitions, another ubiquitous photoprotection mechanism present in other green algae. Certain Sacoglossa sea slugs can feed on Ulvophyceae algae, including some Bryopsidales, and steal chloroplasts -kleptoplasts- that remain functional inside the animal cells for months without the support of the algal nucleus. Our data reveal that the state transition capacity is not retained in the kleptoplasts of the sea slugs, and we provide evidence that the loss is caused by structural changes during their incorporation by the animals. This enforced chloroplast sphericity was observed in all studied kleptoplastic associations, and we propose that it is a fundamental property supporting long-term retention of kleptoplasts in photosynthetic sea slugs.

Abstract:Eukaryotic organelles depend on nuclear genes to perpetuate their biochemical integrity. This is true for mitochondria in all eukaryotes and plastids in plants and algae. Then how do kleptoplasts, plastids that are sequestered by some sacoglossan sea slugs, survive in the animals' digestive gland cells in the absence of the algal nucleus encoding the vast majority of organellar proteins? For almost two decades, lateral gene transfer (LGT) from algae to slugs appeared to offer a solution, but RNA-seq analysis, later supported by genome sequencing of slug DNA, failed to find any evidence for such LGT events. Yet, isolated reports continue to be published and are readily discussed by the popular press and social media, making the data on LGT and its support for kleptoplast longevity appear controversial. However, when we take a sober look at the methods used, we realize that caution is warranted in how the results are interpreted. There is no evidence that the evolution of kleptoplasty in sea slugs involves LGT events. Based on what we know about photosystem maintenance in embryophyte plastids, we assume kleptoplasts depend on nuclear genes. However, studies have shown that some isolated algal plastids are, by nature, more robust than those of land plants. The evolution of kleptoplasty in green sea slugs involves many promising and unexplored phenomena, but there is no evidence that any of these require the expression of slug genes of algal origin.

Abstract:Photosynthetic animals produce oxygen, providing an ideal lens for studying how oxygen dynamics influence thermal sensitivity. The algivorous sea slug, Elysia viridis, can steal and retain chloroplasts from Bryopsis sp. for months when starved, but chloroplast retention is mere weeks when fed Chaetomorpha sp. To examine plasticity in thermal tolerance and changes in net oxygen exchange when fed and starving, slugs fed each alga were acclimated to 17°C (the current maximum temperature to which they are exposed in nature) and 22°C (the increase predicted for 2100) and measured at different points during starvation. We also examined increased illumination to evaluate a potential tradeoff between increased oxygen production but faster chloroplast degradation. Following acclimation, we subjected slugs to acute thermal stress to determine their thermal tolerance. We also measured net oxygen exchange before and after acute thermal stress. Thermal tolerance improved in slugs acclimated to 22°C, indicating they can acclimate to temperatures higher than they naturally experience. All slugs exhibited net oxygen uptake, and rates were highest in recently fed slugs before exposure to acute thermal stress. Oxygen uptake was suppressed following acute thermal stress. Under higher light, slugs exhibited improved thermal tolerance, possibly because photosynthetic oxygen production alleviated oxygen limitation. Accordingly, this advantage disappeared later in starvation when photosynthesis ceased. In conclusion, E. viridis can cope with heat waves by suppressing metabolism and plastically adjusting heat tolerance, however, starvation influences a slug's thermal tolerance and oxygen uptake such that continuous access to algal food for its potential nutritive and oxygenic benefits is critical when facing thermal stress.

Abstract:The sea slug Elysia chlorotica acquires plastids by ingestion of its algal food source Vaucheria litorea. Organelles are sequestered in the mollusc's digestive epithelium, where they photosynthesize for months in the absence of algal nucleocytoplasm. This is perplexing because plastid metabolism depends on the nuclear genome for >90% of the needed proteins. Two possible explanations for the persistence of photosynthesis in the sea slug are (i) the ability of V. litorea plastids to retain genetic autonomy and/or (ii) more likely, the mollusc provides the essential plastid proteins. Under the latter scenario, genes supporting photosynthesis have been acquired by the animal via horizontal gene transfer and the encoded proteins are retargeted to the plastid. We sequenced the plastid genome and confirmed that it lacks the full complement of genes required for photosynthesis. In support of the second scenario, we demonstrated that a nuclear gene of oxygenic photosynthesis, psbO, is expressed in the sea slug and has integrated into the germline. The source of psbO in the sea slug is V. litorea because this sequence is identical from the predator and prey genomes. Evidence that the transferred gene has integrated into sea slug nuclear DNA comes from the finding of a highly diverged psbO 3' flanking sequence in the algal and mollusc nuclear homologues and gene absence from the mitochondrial genome of E. chlorotica. We demonstrate that foreign organelle retention generates metabolic novelty ("green animals") and is explained by anastomosis of distinct branches of the tree of life driven by predation and horizontal gene transfer.

Abstract:Several species of sacoglossan sea slugs possess the incredible ability to sequester chloroplasts from the algae they consume. These "photosynthetic animals" incorporate stolen chloroplasts, called kleptoplasts, into the epithelial cells of tubules that extend from their digestive tracts throughout their bodies. The mechanism by which these slugs maintain functioning kleptoplasts in the absence of an algal nuclear genome is unknown. Here, we report a draft genome of the sacoglossan slug Elysia crispata morphotype clarki, a morphotype native to the Florida Keys that can retain photosynthetically active kleptoplasts for several months without feeding. We used a combination of Oxford Nanopore Technologies long reads and Illumina short reads to produce a 786-Mb assembly (N50 = 0.459 Mb) containing 68,514 predicted protein-coding genes. A phylogenetic analysis found no evidence of horizontal acquisition of genes from algae. We performed gene family and gene expression analyses to identify E. crispata genes unique to kleptoplast-containing slugs that were more highly expressed in fed versus unfed developmental life stages. Consistent with analyses in other kleptoplastic slugs, our investigation suggests that genes encoding lectin carbohydrate-binding proteins and those involved in regulation of reactive oxygen species and immunity may play a role in kleptoplast retention. Lastly, we identified four polyketide synthase genes that could potentially encode proteins producing UV- and oxidation-blocking compounds in slug cell membranes. The genome of E. crispata is a quality resource that provides potential targets for functional analyses and enables further investigation into the evolution and mechanisms of kleptoplasty in animals.
Chlorophyll a synthesis by an animal using transferred algal nuclear genes

Abstract:Chlorophyll synthesis is an ongoing requirement for photosynthesis and a ubiquitous, diagnostic characteristic of plants and algae amongst eukaryotes. However, we have discovered that chlorophyll a (Chla) is synthesized in the symbiotic chloroplasts of the sea slug, Elysia chlorotica, for at least 6 months after the slugs have been deprived of the algal source of the plastids, Vaucheria litorea. In addition, using transcriptome analysis and PCR with genomic DNA, we found 4 expressed genes for nuclear-encoded enzymes of the Chla synthesis pathway that have been horizontally transferred from the alga to the genomic DNA of the sea slug. These findings demonstrate the first discovery of Chla production in an animal using transferred nuclear genes from its algal food.

Abstract:Analysis of the transcriptome of the kleptoplastic sea slug, Elysia chlorotica, has revealed the presence of at least 101 chloroplast-encoded gene sequences and 111 transcripts matching 52 nuclear-encoded genes from the chloroplast donor, Vaucheria litorea. These data clearly show that the symbiotic chloroplasts are translationally active and, of even more interest, that a variety of functional algal genes have been transferred into the slug genome, as has been suggested by earlier indirect experiments. Both the chloroplast- and nuclear-encoded sequences were rare within the E. chlorotica transcriptome, suggesting that their copy numbers and synthesis rates are low, and required both a large amount of sequence data and native algal sequences to find. These results show that the symbiotic chloroplasts residing inside the host molluscan cell are maintained by an interaction of both organellar and host biochemistry directed by the presence of transferred genes.

Abstract:The marine slug Elysia chlorotica (Gould) forms an intracellular symbiosis with photosynthetically active chloroplasts from the chromophytic alga Vaucheria litorea (C. Agardh). This symbiotic association was characterized over a period of 8 months during which E. chlorotica was deprived of V. litorea but provided with light and CO2. The fine structure of the symbiotic chloroplasts remained intact in E. chlorotica even after 8 months of starvation as revealed by electron microscopy. Southern blot analysis of total DNA from E. chlorotica indicated that algal genes, i.e., rbcL, rbcS, psaB, psbA, and 16S rRNA are present in the animal. These genes are typically localized to the plastid genome in higher plants and algae except rbcS, which is nuclear-encoded in higher plants and green (chlorophyll a/b) algae. Our analysis suggests, however, that similar to the few other chromophytes (chlorophyll a/c) examined, rbcS is chloroplast encoded in V. litorea. Levels of psbA transcripts remained constant in E. chlorotica starved for 2 and 3 months and then gradually declined over the next 5 months corresponding with senescence of the animal in culture and in nature. The RNA synthesis inhibitor 6-methylpurine reduced the accumulation of psbA transcripts confirming active transcription. In contrast to psbA, levels of 16S rRNA transcripts remained constant throughout the starvation period. The levels of the photosystem II proteins, D1 and CP43, were high at 2 and 4 months of starvation and remained constant at a lower steady-state level after 6 months. In contrast, D2 protein levels, although high at 2 and 4 months, were very low at all other periods of starvation. At 8 months, de novo synthesis of several thylakoid membrane-enriched proteins, including D1, still occurred. To our knowledge, these results represent the first molecular evidence for active transcription and translation of algal chloroplast genes in an animal host and are discussed in relation to the endosymbiotic theory of eukaryote origins.

Unraveling the Phidiana paradox: Phidiana lynceus can retain algal symbionts but its nocturnal tendencies prevent benefits from photosynthesis

Abstract:Numerous marine invertebrates form symbiotic relationships with single-celled algae, termed "photosymbioses", and the diversity of these interactions is likely underestimated. We examined Phidiana lynceus , a cladobranch sea slug that feeds on photosymbiotic hydrozoans. We assessed its ability to acquire/retain algal symbionts by examining specimens in starvation, finding that P. lynceus is able to incorporate and retain symbionts for up to 20 days. Examining body size during starvation revealed that P. lynceus does not receive enough energy from hosting symbionts to maintain its body mass let alone grow. Intact symbionts were still present in deceased specimens, indicating that P. lynceus does not digest all of its symbionts, even when starving to death. We also examined slug behavior in the field and lab to determine if it seeks light to facilitate photosynthesis, which could provide energetic and oxygenic benefits. In the field, slugs were always observed hiding under stones during the day and they displayed light avoidance in the lab, suggesting this species actively prevents photosynthesis and the benefits it could receive. Lastly, we measured their metabolic rates during the day and night and when treated with and without a photosynthetic inhibitor. Higher metabolic rates at night indicate that this species displays nocturnal tendencies, expending more energy when it emerges at night to forage. Paradoxically, P. lynceus has evolved all of the requisite adaptations to profit from photosymbiosis but it chooses to live in the dark instead, calling into question the nature of this symbiosis and what each partner might receive from their interaction.

Abstract:Some cladobranch sea slugs host algae (Symbiodiniaceae), forming stable or unstable photosymbiotic relationships. Although some benefits from retaining symbionts have been described in stable photosymbioses, unstable photosymbioses remain largely uninvestigated. We examined two potential benefits – nutrition and oxygen produced via photosynthesis – in the unstable cladobranch model species, Berghia stephanieae . To investigate potential nutritive benefits, we conducted transmission electron microscopy and observed both partially digested symbionts and lipid droplets, indicating that B. stephanieae benefits energetically from hosting zooxanthellae through digestion. Since increased temperatures can cause oxygen limitation, any oxygenic benefits B. stephanieae receives from photosynthesis could influence their thermal tolerance, allowing photosynthetic slugs to withstand higher temperatures than specimens where photosynthesis is limited or absent. To assess this, we measured the maximum temperature they can withstand before succumbing to heat-shock under three light intensities (0, 100 and 700 μmol m -2 s -1 ). Oxygen uptake was measured before and after heat-shock to determine whether uptake was affected by thermal stress. Slugs exposed to high light displayed significantly lower thermal limits than those at zero or moderate light, indicating exposure to acute high light negatively impacts thermal tolerance. Lastly, we assessed if and how light affects juvenile development and survival. Juveniles exposed to moderate light survived longest, while both other light intensities reduced survival. These investigations demonstrate that unstable photosymbiosis provides B. stephanieae with nutritive benefits during different ontogenetic stages. Oxygenic benefits are less clear, as slugs exposed to thermal stress in dark and under moderate light did not display different thermal tolerances.

Abstract:The first molecular evidence of horizontal gene transfer between multicellular eukaryotes was our discovery of the presence of three Vaucheria litorea nuclear-encoded genes [fucoxanthin chlorophyll a/c-binding protein (fcp) and light-harvesting complex 1 and 2 (Lhcv1 and 2)] in the genomic DNA of the sea slug, Elysia chlorotica, which are used to support the chloroplast endosymbiosis in the slug. These genes are translated and transcribed in the host cell, and vertically transmitted to subsequent generations of the host species. In order to provide a database of native V. litorea sequences to facilitate the search for additional transferred genes between these two species, we have partially sequenced and annotated the transcriptome of V. litorea, using 454 Life Science’s next generation pyrosequencing technology. Preliminary analysis of the sequence data has led to the discovery of six additional algal nuclear genes in E. chlorotica cDNA and genomic DNA, which encode enzymes in the chlorophyll synthesis pathway as well as additional light-harvesting and metabolic enzymes. Abstract:Abstract Although light is the driving force of photosynthesis, excessive light can be harmful. One of the main processes that limits photosynthesis is photoinhibition, the process of light-induced photodamage. When the absorbed light exceeds the amount that is dissipated by photosynthetic electron flow and other processes, damaging radicals are formed that mostly inactivate photosystem II (PSII). Damaged PSII must be replaced by a newly repaired complex in order to preserve full photosynthetic activity. Chlorella ohadii is a green micro-alga, isolated from biological desert soil crusts, that thrives under extreme high light and is highly resistant to photoinhibition. Therefore, C. ohadii is an ideal model for studying the molecular mechanisms underlying protection against photoinhibition. Comparison of the thylakoids of C. ohadii cells that were grown under low light versus extreme high light intensities, found that the alga employs all three known photoinhibition protection mechanisms: i) massive reduction of the PSII antenna size; ii) accumulation of protective carotenoids; and iii) very rapid repair of photo-damaged reaction center proteins. This work elucidated the molecular mechanisms of photoinhibition resistance in one of the most light-tolerant photosynthetic organisms and shows how photoinhibition protection mechanisms evolved to marginal conditions, enabling photosynthesis-dependent life in severe habitats. Adaptation of light-harvesting functions of unicellular green algae to different light qualities

Abstract:Oxygenic photosynthetic organisms perform photosynthesis efficiently by distributing captured light energy to photosystems (PSs) at an appropriate balance. Maintaining photosynthetic efficiency under changing light conditions requires modification of light-harvesting and energy-transfer processes. In the current study, we examined how green algae regulate their light-harvesting functions in response to different light qualities. We measured low-temperature time-resolved fluorescence spectra of unicellular green algae Chlamydomonas reinhardtii and Chlorella variabilis cells grown under different light qualities. By observing the delayed fluorescence spectra, we demonstrated that both types of green algae primarily modified the associations between light-harvesting chlorophyll protein complexes (LHCs) and PSs (PSII and PSI). Under blue light, Chlamydomonas transferred more energy from LHC to chlorophyll (Chl) located far from the PSII reaction center, while energy was transferred from LHC to PSI via different energy-transfer pathways in Chlorella. Under green light, both green algae exhibited enhanced energy transfer from LHCs to both PSs. Red light induced fluorescence quenching within PSs in Chlamydomonas and LHCs in Chlorella. In Chlorella, energy transfer from PSII to PSI appears to play an important role in balancing excitation between PSII and PSI.

Abstract:As a common macroalga living in the intertidal zone, Sargassum thunbergii (Sargassaceae, Phaeophyta) is often exposed to drastic changes in solar photosynthetically active radiation during a diurnal cycle; thus, the potential photosynthetic adaptation processes deserve attention. In this work, we examined the photosynthetic performance and xanthophyll cycle activity of this alga in response to high light (1,200 μmol photons m –2 s –1 , the average in-situ light intensity at noon) by using chlorophyll fluorescence and high-performance liquid chromatography (HPLC). On exposure to high light, a rapid decrease in the effective quantum yield of photosystem II (PSII) (Y(II)) occurred, indicating down-regulation of PSII activity; a corresponding increase in non-photochemical quenching (NPQ) indicated the existence of energy-dissipating cycles. After turning off the light, Y(II) gradually increased to 0.7, accompanied by a decrease in NPQ. However, no complete recovery of NPQ was observed, and its value remained at ≈4, even after an 80-min dark treatment. The size of the xanthophyll cycle pigments pool was quantified using HPLC and was found to be ≈16 mol mol −1 Chl a × 100. The activity of the xanthophyll cycle, characterized by a de-epoxidation state (DPS), could reach up to ≈0.5. Such a large pigments pool and rapid accumulation of zeaxanthin may allow S. thunbergii to induce high values of NPQ (≈10). These results were further complemented by inhibitor (dithiothreitol, DTT) and pre-illumination experiments showing that (1) both NPQ and the xanthophyll cycle could be inhibited by DTT, and there was always a strong positive correlation between NPQ and DPS; (2) the previously formed antheraxanthin exhibited a long retention time and a slow epoxidation; and (3) the long retention of antheraxanthin contributed to a rapid accumulation of zeaxanthin. In conclusion, our present study demonstrated that xanthophyll cycle-induced NPQ can significantly protect S. thunbergii from light fluctuations in the intertidal zone.
