Abstract:Due to their superparamagnetism and enzyme-like activity, iron oxide (Fe3O4) nanozymes can be readily used for sample pretreatment and the generation of detection signals, and have, thus, attracted much attention in the field of bioanalysis and diagnosis. However, the low catalytic activity of Fe3O4 nanozymes does reduce the sensitivity of Fe3O4-based methods, limiting their application. In this study, Fe3O4@Cu@poly(pyrrole-2-carboxylic acid) yolk-shell nanozymes (Fe3O4@Cu@PCPy YSNs) were synthesized using a facile approach and selective chemical etching technology. Compared with Fe3O4 nanozymes, the Fe3O4@Cu@PCPy YSNs demonstrated a three-fold increase in the peroxidase-like activity, good dispersity and strong superparamagnetism. In addition, the flower-shaped structure of aptamer-complementary strand (Apt-CS) conjugates was designed on the surface of the Fe3O4@Cu@PCPy YSNs, which effectively inhibited their peroxidase-like activity by creating a physical barrier that hindered the access of substrates to the center of the Fe3O4@Cu@PCPy YSNs. Based on this principle, a robust and facile colorimetric aptasensor was developed for detecting Salmonella Typhimurium. The flower-shaped Apt-CS were dissociated in the presence of S. Typhimurium, promoting the recovery of Fe3O4@Cu@PCPy YSN catalytic activity. Under optimized conditions, this proposed aptasensor successfully detected S. Typhimurium in a linear range of 3 to 3 × 106 CFU/mL, achieving a detection limit of 1 CFU/mL. Finally, the feasibility of this novel aptasensor was further validated by three actual samples, with recoveries of between 84.3% and 102%, thereby demonstrating the huge potential of the proposed aptasensor for detecting S. Typhimurium in foods.

Abstract:A simple, fast and convenient colorimetric aptasensor was fabricated for the detection of Salmonella typhimurium (S. typhimurium) which was based on the color change effect of gold nanoparticles (GNPs). S. typhimurium is one of the most common causes of food-associated disease. Aptamers with specific recognition toward S. typhimurium was modified to the surface of prepared GNPs. They play a role for the protection of GNPs from aggregation toward high concentrations of NaCl. With the addition of S. typhimurium, aptamers preferably combined to S. typhimurium and the protection effect was broken. With more S. typhimurium, more aptamers detached from GNPs. In such a situation, the exposed GNPs would aggregated to some extent with the addition of NaCl. The color changed from red, purple to blue which could be characterized by UV–Vis spectrophotometer. The absorbance spectra of GNPs redshifted constantly and the intensity ratio of A700/A521 changed regularly. This could be calculated for the basis of quantitative detection of S. typhimurium from 102cfu/mL to 107cfu/mL. The obtained linear correlation equation was y=0.1946x–0.2800 (R2=0.9939) with a detection limit as low as 56cfu/mL. This method is simple and rapid, results in high sensitivity and specificity, and can be used to detect actual samples.

Abstract:A simple, rapid, selective, and sensitive biosensor was constructed for detection of Salmonella typhimurium (S. typhimurium), based on enhanced Raman intensity using spiny gold nanoparticles (SGNPs). The SGNPs were respectively functionalized with 4-mercaptobenzoic acid (p-MBA, Raman signal molecules) and thiolated S. typhimurium aptamers as SERS nanoprobes. On the other side, the biotinylated aptamer was anchored on the microtiter plate which could realize specific recognition and capture of S. typhimurium. Gradient dilutions of S. typhimurium captured gradient amounts of the SERS nanoprobes. The SERS intensity of p-MBA at 1586 cm(-1) was used to successfully measure S. typhimurium (y = 50.6155 + 1044.8341x, R-2 = 0.9958) in the range 10(1) cfu/mL to 10(5) cfu/mL, with a limit of detection (LOD) of 4 cfu/mL. This method was also used successfully to detect S. typhimurium inoculated into pork samples. This method, using aptamers and the SGNPs-based SERS biosensor may be useful to detect S. typhimurium for food safety analysis and clinical diagnosis. (C) 2017 Published by Elsevier Ltd.

Abstract:This paper presents a sensitive and convenient visual methodology for Salmonella typhimurium detection using gold nanoparticles (AuNPs) as colorimetric probes and magnetic nanoparticles (MNPs) as concentration elements. In the protocol, the aptamers were first immobilized onto the surface of AuNPs and MNPs, respectively. Then, S. typhimurium were added into the above solution and incubated for 45 min. During the incubation, aptamer on the surface of nanoparticles could specifically bind to the target and form a MNPs-aptamer-S. typhimurium-aptamer-AuNPs sandwich structure complex. In a magnetic field, the formed complexes were easily separated from the solution, resulting in a fading of the AuNPs suspension and a decrease of the ultraviolet visible (UV/Vis) signal. The assay shows a linear response toward S. typhimurium concentration through a range of 25 to 10(5) cfu/mL, and the detection limit was improved to 10 cfu/mL. The applicability of the bioassay in real food samples was also investigated; the results were consistent with the experimental results obtained from plate-counting methods. It is believed that the developed aptasensor will broaden the application in bioassays.

Abstract:In this work, we developed an ultrasensitive colorimetry for Salmonella typhimurium detection with multifunctional Au-Fe3O4 dumbbell-like nanoparticles (DBNPs) which possessed easy bio-modifiability, excellent LSPR characteristics, superparamagnetic properties and super peroxidase-like activity. In the detection, the anti-S. typhimurium antibody modified DBNPs (IDBNPs) bound with S. typhimurium and aggregated on their surfaces in a large number, which showed much quicker magnetic response than free IDBNPs. By controlling appropriate separation conditions, IDBNPs@S. typhimurium composites were captured, while free IDBNPs were remained in the supernatant. Therefore, by detecting the absorbance of the supernatant, quantitative detection was achieved from 10 to 1000 CFU/mL. Moreover, utilizing the peroxidase-like activity of IDBNPs, we further realized semi-quantitative naked-eye detection. By adding ABTS into the above supernatant, which was oxidized to green chelate (OxABTS), colorimetric signal was amplified significantly, and meanwhile, the green chelates and the wine-red IDBNPs engendered mixed color, enhancing the range of color gradation and greatly improving the visual resolution. Finally, a detection limit (10 CFU/mL) comparable with that of above spectrum measurement was achieved. Besides, our method exhibited efficient capture capability (nearly 100% even for rare S. typhimurium), and had good stability and specificity, and acceptable anti-interference ability in fetal bovine serum and milk samples.

Abstract:A sensitive steady-state chemiluminescent aptasensor based on rolling circle amplification (RCA) was fabricated for the detection of Salmonella typhimurium. The sensor utilized aptamer modified Fe3O4 magnetic nanoparticles (MNPs) as capture probes, aptamer as recognition molecules, and Co2+ enhanced N‑(aminobutyl)-N-(ethylisoluminol) (ABEI) functional flowerlike gold nanoparticles (AuNFs) and complementary strand (cDNA) complex (Co2+/ABEI-AuNFs-cDNA) as signal probes. And P-Iodophenol (PIP) was also added to form a typical ABEI- AuNFs-PIP-H2O2 steady-state CL system. By virtue of Fe3O4 MNPs based solid-phase RCA strategy, S. typhimurium can be first captured by the aptamer immobilized on the surface of Fe3O4 MNPs then complex with RCA products to form a sandwich complex. Co2+/ABEI-AuNFs-cDNA signal probes were then assembled on the RCA products to produce and enhance CL signals. Under optimal conditions, the logarithmic correlation between the concentration of S. typhimurium and the CL signal was found to be linear within the range of 32cfumL−1 to 3.2×106cfumL−1 (R2 =0.9921). The limits of detection of the developed method were found to be 10cfumL−1 for S. typhimurium. The method was also used to detect S. typhimurium in real pork samples. The results were compared with those obtained from the plate-counting methods and showed good consistency. Therefore, this detection aptasnesor can be a good candidate for sensitive and selective detection of S. typhimurium with simple and effective operations.

Abstract:Pathogens detection is a crucial measure in the prevention of foodborne diseases. This study developed a novel multicolor colorimetric assay to visually detect Salmonella Typhimurium ( S. Typhimurium ), by utilizing the etching process of gold nanorods (AuNRs) with TMB 2+ . The strategy involved the construction of nanozyme by assembling magnetic covalent organic framework (MCOF) with aptamer-conjugated AuNPs (Apt-AuNPs), exhibiting remarkable peroxidase-like activity to catalyze the oxidation of TMB/H 2 O 2 and inducing the etching of AuNRs. The presence of S. Typhimurium could inhibit this process, resulting in the generation of vivid colors. The multicolor colorimetric assay could specifically determine S. Typhimurium from 10 2 to 10 8 CFU mL −1 in 60 min with visual detection limit of 10 2 CFU mL −1 , and instrumental detection limit of 2.3 CFU mL −1 . Moreover, detecting S. Typhimurium in chicken, milk, pork and lettuce samples has shown promise in practical applications.

Abstract:A rapid, specific and visible detection method for Salmonella Typhimurium was developed based on the recognition of aptamers coupled with nanogold labeling and silver signal amplification. Briefly, biotinylated aptamer 1, which was specific to Salmonella Typhimurium, was immobilized onto the surface of microtiter plate-wells modified with streptavidin through the binding of biotin and streptavidin. Then, the target bacteria, Salmonella Typhimurium, and the aptamer 2-gold nanoparticle (aptamer 2-AuNPs) conjugates were incubated in the wells to form sandwich-type aptamer/bacteria/aptamer-AuNPs complexes. Under optimal conditions, the correlation between the concentration of Salmonella Typhimurium and the intensity of the signals was observed to be linear within the range of 10–106 cfu mL−1 (R2 = 0.9913), and the detection limit of the proposed method was observed to be 7 cfu mL−1 This developed method offers the potential for rapid, sensitive and visible detection of Salmonella Typhimurium in samples.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has been used in a variety of biological applications due to its high sensitivity and specificity. Here, we report a SERS-based aptasensor approach for quantitative detection of pathogenic bacteria. A SERS substrate bearing Au@Ag core/shell nanoparticles (NPs) is functionalized with aptamer 1 (apt 1) for the capture of target molecules. X-rhodamine (ROX)-modified aptamer 2 (apt 2) is used as recognition element and Raman reporter. Salmonella typhimurium specifically interacted with the aptamers to form Au@Ag-apt 1-target-apt 2-ROX sandwich-like complexes. As a result, the concentration of S. typhimurium was determined using this developed aptasensor structure, and a calibration curve is obtained in the range of 15 to 1.5 × 10(6) cfu/mL with a limit of detection of 15 cfu/mL. Our method was successfully applied to real food samples, and the results are consistent with the results obtained using plate counting methods. We believe that the developed method shows potential for the rapid and sensitive detection of pathogenic bacteria in food safety assurance.
A dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for Salmonella Typhimurium detection

Abstract:Background Salmonella Typhimurium poses a serious threat to human health worldwide, necessitating the development of a rapid, sensitive, and convenient method for S . Typhimurium detection. Nanozymes are considered ideal signal report elements, which are extensively used for developing colorimetric methods. However, single-component nanozymes display low catalytic activity, and colorimetric methods are susceptible to environmental interference, reducing the sensitivity and accuracy of detection results. To address these drawbacks, this study constructed a dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for S . Typhimurium detection in food. Results In this study, the composite Fe 3 O 4 @MIL-100(Fe) nanozymes were successful synthesized and demonstrated substantial peroxide-like activity, with 4.76-fold higher specificity activity (SA) than that of Fe 3 O 4 nanozymes. Then, a glucose oxidase (GOx)-Fe 3 O 4 @MIL-100(Fe) cascade reaction was developed for colorimetric detection via an aptamer to facilitate the formation of Fe 3 O 4 @MIL-100(Fe)/ S . Typhimurium/carboxylated g-C 3 N 4 (CCN)-GOx sandwich complexes. Meanwhile, the fluorescence mode was achieved by measuring the fluorescence intensity of the sandwich complexes. In optimum conditions, the dual-mode detection limits (LOD) were 1.8 CFU/mL (colorimetric mode) and 1.2 CFU/mL (fluorescence mode), respectively, with the S . Typhimurium concentration ranging from 10 CFU/mL to 10 7 CFU/mL. Finally, the feasibility of the dual-mode colorimetric-fluorescence method was validated via three actual samples, yielding recovery rates of 77.32 % to91.17 % and 82.17 % to 103.7 %, respectively. Significance and novelty This study successfully develops a composite nanozyme-based cascade colorimetric and fluorescence dual-mode aptasensor for S . Typhimurium detection. It presents several distinct benefits, such as a broader linear range (10–10 7 CFU/mL), a lower LOD value (1.2 CFU/mL), and more accurate results. More importantly, the proposed dual-mode method displays a low LOD in colorimetric mode, demonstrating considerable potential for S . Typhimurium on-site detection in food.

Abstract:Salmonella Typhimurium (S. Typhimurium) is a globally distributed foodborne pathogen, which can lead to outbreaks of foodborne infectious diseases. It is essential to guarantee food safety by timely and correct detection of S. Typhimurium. In this investigation, an original fluorescence aptasensor was constructed to detect S. Typhimurium rapidly and sensitively. Through the coupling of magnetic beads, aptamer, and gold nanoparticles (AuNPs), a fluorescence quenching system with a “sandwich structure” was established. The aptamer acted as a link, and its specific binding to S. Typhimurium could release AuNPs from the system. Meanwhile, fluorescent DNA-stabilized silver nanoclusters (DNA-AgNCs) were synthesized. The fluorescence intensity changes caused by the fluorescence resonance energy transfer between DNA-AgNCs and AuNPs were utilized to detect S. Typhimurium. The purposed aptasensor exhibited high selectivity and sensitivity with a linear response to S. Typhimurium, ranging from 3.7 × 102 to 3.7 × 105 cfu/mL. The limit of detection (LOD) was estimated to be 98 cfu/mL within 2 h 10 min. In addition, this method showed excellent application for detection of S. Typhimurium in artificially contaminated milk, with LOD reaching 3.4 × 102 cfu/mL. Therefore, the developed fluorescence aptasensor has great potential to identify S. Typhimurium in foodstuffs.

Gold Nanoparticles Enhanced SERS Aptasensor for the Simultaneous Detection of Salmonella Typhimurium and Staphylococcus Aureus.

Abstract:Salmonella typhimurium and Staphylococcus aureus are most common causes of food-associated disease. A Raman based biosensor was developed for S. typhimurium and S. aureus detection simultaneously. The biosensor was based on nanoparticles enhanced Raman intensity and the specific recognition of aptamer. The Raman signal probe and the capture probe are built. Gold nanoparticles (GNPs) modified with Raman molecules (Mercaptobenzoic acid and 5,5'-Dithiobis(2-nitrobenzoic acid)) and aptamer are used as the signal probe for S. typhimurium and S. aureus, respectively. Fe3O4 magnetic gold nanoparticles (MGNPs) immobilized with both aptamer of S. typhimurium and S. aureus are used as the capture probe. When S. typhimurium and S. aureus are added in the reaction system, the capture probe will capture the target bacteria through the specific binding effect of aptamer. And then the signal probe will be connected to the bacteria also by the effect of aptamer to form the sandwich like detection structure. The Raman intensified spectrum was measured to quantify S. typhimurium and S. aureus. Under optimal conditions, the SERS intensity of MBA at 1582 cm(-1) are used to measure S. typhimurium (y=186.4762+704.8571x, R(2)=0.9921) and the SERS intensity of DNTB at 1333 cm(-1) are used to measure S. aureus (y=135.2381+211.4286x, R(2)=0.9946) in the range of 10(2)-10(7) cfu mL(-1). The LOD is 35 cfu mL(-1) for S. aureus and 15 cfu mL(-1) for S. typhimurium. This method is simple and rapid, results in high sensitivity and specificity, and can be used to detect actual samples.

Abstract:A new colorimetric aptasensor platform was fabricated to detect Salmonella enterica serovar typhimurium based on the peroxidase-like activity of ZnFe2O4-reduced graphene oxide (ZnFe2O4/rGO) nanostructures. The synthesized ZnFe2O4/rGO can catalytically oxidize 3,3',5,5'-tetramethylbenzidine (TMB) by H2O2 and generate a typical blue product, which was detected by micro-reader at 652nm. Therefore, ZnFe2O4/rGO was conjugated with aptamer to act as the signal probe. Biotin modified aptamer was immobilized onto the micro-plate to act as the capture probe. In the presence of target, a "sandwich-type" complex of aptamer (on micro-plate)-target-aptamer-ZnFe2O4/rGO complexes was formed through specific recognition of aptamers and corresponding target. The limit of detection was 11cfu/mL and the detection range was 11 to 1.10×105cfu/mL of S. typhimurium in buffer solution. Actual samples analysis indicated that this colorimetric aptasensor produced results consistent with plate-counting analysis. The developed method was simple and sensitive, which may pave the way for the detection of other pathogenic bacteria with suitable aptamers.
Silver/copper bimetallic nanoclusters integrating with cryonase-assisted target recycling amplification detection of Salmonella typhimurium

Abstract:An unsophisticated fluorescence-enabled strategy is brought forward to process the highly sensitive fluorescence detection of Salmonella typhimurium (S. typhimurium) which based on polyethyleneimine (PEI)-templated silver/copper nanoclusters (Ag/CuNCs) (λ excitation = 334 nm and λ emission = 466 nm) with cryonase-assisted target recycling amplification. The Ag/CuNCs nanoclusters are synthesized as fluorescent materials due to their strong and stable fluorescence characteristics and are modified with S. typhimurium aptamers to form aptamer-Ag/CuNCs probes. The probes can be adsorbed on the surface of quenching agents-polydopamine nanospheres (PDANSs), thereby inducing fluorescence quenching of the probes. Once the aptamers are bound to the target, the aptamers/targets complexes are separated from the PDANSs surface, and the Ag/CuNCs recover the fluorescence signal. The released complexes will immediately be transformed into a substrate digested by cryonase (an enzyme that can digest all types of nucleic acids), and the released targets are bound to another aptamers to initiate the next round of cleavage. This reaction will be repeated continuously until all relevant aptamers are consumed and all Ag/CuNCs are released, resulting in a significant amplification of the fluorescence signal and improved sensitivity. Using Ag/CuNCs as fluorescent probes combined with cryonase-assisted amplification strategy, the fluorescence aptasensor is constructed with detection limits as low as 3.8 CFU mL-1, which is tenfold better than without the cryonase assistance. The method developed has been applied to milk, orange juice, chicken, and egg white samples with excellent selectivity and accuracy providing an approach for the early and rapid detection of S. typhimurium in food.

Abstract:An aptamer-based assay is described for the determination of Salmonella typhimurium (S. typh). Carboxymethyl chitosan was loaded with amino-modified aptamer against S. typh, and then adsorbed on gold nanoparticles by electrostatic interaction to form a composite that acts as the molecular recognition element. In the presence of S. typh, it will be bound by the aptamer, and this changes the structure of the recognition element. On addition of salt solution, the gold nanoparticles agglomerate so that the color of the solution changes from red to blue. S. typh can be detected via measurement of the absorbance at 550 nm. Absorbance increases linearly with the logarithm of the S. typh concentration in the range from 100 to 109 cfu·mL−1. The limit of detection is 16 cfu·mL−1. The specificity and practicability of the assay were evaluated. The recoveries of S. typh from spiked milk samples are between 92.4 and 97.2%. The analytical results are basically consistent with those of a plate counting method.
Aptamer Based SERS Detection of Salmonella Typhimurium Using DNA-assembled Gold Nanodimers

Abstract:The authors describe a surface-enhanced Raman scattering (SERS) based aptasensor for Salmonella typhimurium (S. typhimurium). Gold nanoparticles (AuNPs; 35 nm i.d.) were functionalized with the aptamer (ssDNA 1) and used as the capture probe, while smaller (15 nm) AuNPs were modified with a Cy3-labeled complementary sequence (ssDNA 2) and used as the signalling probe. The asymmetric gold nanodimers (AuNDs) were assemblied with the Raman signal probe and the capture probe via hybridization of the complementary ssDNAs. The gap between two nanoparticles is a “hot spot” in which the Raman reporter Cy3 is localized. It experiences a strong enhancement of the electromagnetic field around the particle. After addition of S. typhimurium, it will be bound by the aptamer which therefore is partially dehybridized from its complementary sequence. Hence, Raman intensity drops. Under the optimal experimental conditions, the SERS signal at 1203 cm−1 increases linearly with the logarithm of the number of colonies in the 102 to 107 cfu·mL−1 concentration range, and the limit of detection is 35 cfu·mL−1. The method can be performed within 1 h and was successfully applied to the analysis of spiked milk samples and performed very well and with high specificity.

Abstract:Nanozyme-based detection methods have gained increasing attention in the field of food safety. However, the effect of the food matrix on the nanozyme activity requires tedious sample pre-treatment for nanozyme-based detection. Therefore, a difunctional histidine-modified magnetic hybrid nanozymes (His-Fe3O4@Cu) was used as capture probes and signal amplifiers to develop a simple colorimetric aptasensor for Salmonella Typhimurium detection. The prepared His-Fe3O4@Cu nanozymes demonstrated a 5.0-fold increase in peroxidase-like activity compared to Fe3O4 nanozymes and exhibited superparamagnetism with saturation magnetization values of 63.5 emu/g. By virtue of the strong superparamagnetism of the His-Fe3O4@Cu nanozymes, aptamer-conjugated His-Fe3O4@Cu nanozymes first enriched and separated S. Typhimurium from food matrix. Then, aptamer-conjugated His-Fe3O4@Cu nanozymes employed an etching solution to generate a detection signal via the peroxidase-like activity. In optimal conditions, this proposed colorimetric aptasensor successfully detected S. Typhimurium with a limit of detection (LOD) of 8 CFU/mL in a linear range of 1.2 × 101 to 1.2 × 106 CFU/mL. Finally, the proposed aptasensor was further validated in three actual samples, with a capture efficiency ranging from 88.6 % to 96.5 % and recoveries between 88.2 % and 104 %. These results revealed the significant potential of this method for S. Typhimurium detection in food samples.

Abstract:Sensitive, accurate, simple and quick monitoring of Salmonella typhimurium ( S. typhimurium ) in food is significant for preventing food poisoning, but still remains a challenge. Herein, a colorimetry/fluorescence dual-mode sensing strategy was fabricated to detect S. typhimurium by integrating the self-assembly of magnetic covalent organic framework (MCOF) with gold nanoparticles (Au NPs) as the peroxidase-mimicking nanozyme and aggregation-induced emission luminogen (AIEgen). S. typhimurium could competitive bind to aptamer conjugated Au NPs (Au NPs@apt), inhibit the self-assembly of MCOF with Au NPs, and shield the catalytic activity of AuNPs. After adding H 2 O 2 and TPE-4A, the dark green solution changed to light with increasing S. typhimurium concentration, on the contrary, the fluorescent signals were generated. As a result, in colorimetry/fluorescence modes, S. typhimurium could be detected in the linear ranges of 10 3 –10 8 CFU mL −1 and 10 1 –10 7 CFU mL −1 , with LODs of 1000 and 10 CFU mL −1 , respectively. Importantly, different colors consistent with various S. typhimurium concentrations can be accurately classified by smartphone app and linear discriminant analysis (LDA). The smartphone-assisted data interpretation can generate complementary colorimetry and fluorescence signals without any sophisticated equipment and achieve on-site detection. Moreover, the proposed strategy could be explored for S. typhimurium monitoring in milk with satisfactory recoveries (97.6–100.4 %) in colorimetry and fluorescence mode and good classification and prediction performance in smartphone/LDA system, suggesting the feasibility and potential applications of the sensing platform.
