Abstract:Pecking behavior is used to measure psychomotor stimulant effect of addictive drugs in present study. Each day after chicks were interperitoneally injected 0.5,1.0,2.0 mg•kg-1•morphine for one hour, number of pecking was counted for two minutes with a blue bead. After four days, a two-minute-test recorded the number of pecking in drug-free state using blue and red bead respectively. The result revealed that 0.5 mg•kg-1 morphine group exhibited significant more number of pecking compared with control group during the fourth day and test, indicating morphine induced pecking behavioral sensitization and conditioned pecking behavioral sensitization.
Effect of Morphine Dependence on the Ultrastructure of Sprague-Dawley Rats

Abstract:Objective: To investigate the ultrastructural changes of morphine dependent rat in the brain area of ventral tegmental area (VTA) and locus coeruleus (LC). Methods: The rats were given (sc) increasing doses of morphine to form morphine dependent models. The ultrastructural changes of the brain area of the above were observed by transmission electron microscope . Results: In the morphine dependent group,the neuronal nucleus of LC was irregular and swelling, there were vesiculations and dissolution of nuclear matrix in the nucleus of LC neuron , the rough endoplasmic reticulum was decreased, the free ribosomes were sparseness and the myelin figure was observed in cytoplasm of LC. Neuronal microfilament was abundant and myelinic degeneration of neuronal fibra was observed in nerve cell and neurogliocyte of VTA. Conclusion: Chronic exposure to morphine might induce ultrastructural changes in the brain area of VTA and LC which might contribute to the development of morphine dependence.

Abstract:The membrane attack complex of complement (C5b-9) is identical in composition regardless of which pathway of activation was instrumental in its formation. Band V protein was consistently a subunit of the soluble complex. Since band V protein is not required for complement-dependent cytolysis, it probably represents a membrane site equivalent in serum of the nascent C5b-9 complex.

Abstract:Molecular Radar (MR) is a new method to detect biological processes in living cells at the level of molecular, it is also the newest means to get intracellular information. In this paper we study the effect of morphine on PC12 cells using MR. The results show that the effect of morphine on PC12 cells is time- and concentration-dependent. Morphine treating for short time induces the increase and fluctuation of intracellular (CA2+), while morphine treating for long time induces chromatin condensation, loss of mitochondria membrane potential apoptosis.

Abstract:In the avian forebrain, the medial striatum and the intermediate medial mesopallium are thought to be important structures for associative learning in chicks, where the role of dopaminergic systems in learning processes has been verified in various behavioral paradigms, such as one-trial passive avoidance learning. However, it is not yet clear whether the dopaminergic system of these regions is responsible for associative learning underlying cue-elicited drug reward. In this study, a 6-day conditioning schedule in day-old chicks with i.p. morphine (2mg/kg) was used to compare the effects of intracerebrally injected dopamine D1 receptor antagonist, SCH23390, and D2 antagonist, spiperone. The antagonists were injected bilaterally (3µg/site) into either the medial striatum or the intermediate medial mesopallium, and tests were conducted on morphine-induced conditioned place preference or locomotor activity. The acquisition of place preference was significantly inhibited by SCH23390 in either the medial striatum or the intermediate medial mesopallium, but not by spiperone. However, in the medial striatum, but not in the intermediate medial mesopallium, the locomotor activity was blocked by both SCH23390 and spiperone. These data suggest that the medial striatum and the intermediate medial mesopallium in birds are differentially involved in the rewarding effects of morphine, and similarly to mammals, the dopamine D1 system may play an important role in the development of opiate reward.

Abstract:A novel means of measuring and analysing behavioural effects of dopamine agonists is described and illustrated by a comparison of the effects of d-amphetamine and apomorphine in the rat. d-Amphetamine (0–15 mg/kg IP) produced significant dose- and time-dependent changes in responses such as locomotion, rearing and sniffing, but not in licking or gnawing. In contrast, apomorphine (0–5 mg/kg SC) produced significant increases in licking and gnawing, as well as in locomotion and sniffing, but no changes in rearing. The results are discussed in comparison with those obtained by other methods, such as photocell beam interruptions or stereotypy rating scales, and may be of importance in elucidating the functions of the forebrain dopamine projections.
PET Imaging Reveals Brain Metabolic Changes in Adolescent Rats Following Chronic Escalating Morphine Administration

Abstract:Purpose Non-medical use of prescription opioids, especially among adolescents, has been substantially increased in recent years. However, the neuromechanism remains largely unexplored. In the present study, we aimed to investigate the brain metabolic changes in adolescent rats following chronic escalating morphine administration using positron emission tomography (PET). Procedures 2-Deoxy-2-[ 18 F]Fluoro- d -glucose ([ 18 F]FDG) microPET imaging was performed, and statistical parametric mapping (SPM) was used for image analysis. Glucose transporter 3 (Glut-3), dopamine D 2 receptor (D 2 R), and Mμ-opioid receptor (μ-OR) were used for immunostaining analysis. Results Cerebral glucose metabolism was increased in the corpus callosum (CC) and right retrosplenial dysgranular cortex (rRSD), while it was decreased in the right ventral pallidum (rVP). The expressions of Glut-3, D 2 R, and μ-OR were increased in CC and rRSD, while they were decreased in rVP. Furthermore, glucose metabolism and Glut-3 expression were positively correlated with the expressions of D 2 R or μ-OR in CC, rRSD, and rVP. Conclusions [ 18 F]FDG microPET brain imaging study in combination with immunohistological investigation revealed that CC, rRSD, and rVP were specifically involved in opioid dependence in adolescents. Our findings provided valuable insights into the neuromechanism of adolescent addiction of prescription opioids and might have important implications for the development of prevention and intervention approaches.
Dopamine Denervation Leads to an Increase in the Intramembrane Interaction Between Adenosine A2 and Dopamine D2 Receptors in the Neostriatum

Abstract:We have previously found, in striatal membrane preparations from young (2 months old) rats, that stimulation of adenosine A2 receptors (with the selective adenosine A2 agonist CGS 21680) increases the dissociation constants of high- (Kh) and low-affinity (Kl) dopamine D2 binding sites (labelled with the selective dopamine D2 antagonist [3H]raclopride) without changing the proportion of high affinity binding sites (Rh). In the present study in striatal preparations from adult (6 months old) rats, it was found that in addition to the increase in both Kh and Kl values, stimulation of adenosine A2 receptors is associated with an increase in Rh. These result suggest that, in the adult rat, adenosine A2 stimulation may inhibit the behavioural effects induced by dopamine D2 stimulation both by decreasing the affinity and the transduction of dopamine D2 receptors. We have also studied the intramembrane A2-D2 receptor interaction in an experimental model of Parkinson's disease, namely in rats with a unilateral 6-OH-dopamine-induced lesion of the nigro-striatal dopamine pathway. It was found that a unilateral dopamine denervation is associated with a higher density of striatal dopamine D2 receptors in the order of 20%, without any change in their affinity compared with the unlesioned neostriatum. Furthermore, the density (Bmax values) of dopamine D2 receptors in the contralateral neostriatum was significantly higher (about 20%) than in the striatum from native animals. This finding suggests that an unilateral dopamine denervation also induces compensatory long-lasting changes of dopamine D2 receptors in the contralateral neostriatum. In addition to the hightened sensitivity to dopamine agonists, it is known that the dopamine denervated striatum is more sensitive to adenosine antagonists like methylxanthines. If the adenosine A2-dopamine D2 interaction is the main mechanism of action mediating the central effects of methylxanthines, the dopamine denervation might also potentiate this interaction, i.e., dopamine D2 receptors could be not only more sensitive to dopamine but also to adenosine A2 receptor activation. Our results support this hypothesis, since membrane preparations from the denervated neostriatum are more sensitive to the effect of CGS 21680 on dopamine D2 receptors. Thus a low dose of CGS 21680 (3 nM), which is not effective in membrane preparations from the neostriatum of naive animals, is still effective in membranes from the denervated neostriatum. These results underline the potential antiparkinsonian activity of adenosine A2 antagonists.

Abstract:Dosing‐time–dependent changes in the effect and toxicity of morphine were examined in mice housed under alternating 12 h light (07:00 to 19:00 h) and dark (19:00 to 07:00 h) cycles. Morphine (0.5 mg/kg) was injected intraperitoneally (i.p.) in animals to assess its beneficial effect (i.e., protection against the kaolin‐induced, bradykinin‐mediated, writhing reaction) and its toxicity (i.e., alteration of the hepatic enzymes of aspartate aminotransferase [AST] alanine aminotransferase [ALT], and glutathione [GSH] in separate experiments). The magnitude of the analgesic effect of morphine depended on dosing time, with minimum effect at 02:00 h and maximum effect at 14:00 h. The serum hepatic enzyme levels of AST and ALT increased after dosing morphine (100 mg/kg) at 02:00 and 14:00 h. Time courses of these enzymes did not differ between the two trials. However, hepatic GSH, which is involved in the detoxification of chemical compounds, significantly decreased after i.p. morphine injection at 02:00 but not at 14:00 h. Overall, the results suggest that the analgesic effect of morphine is greater after dosing during the resting than during the activity phase of mice that have been induced with bradykinin‐mediated pain. Drug‐induced hepatic damage as inferred by GSH alteration, however, may be greater after dosing during the active phase.

Effects of Morphine on Rheological Properties of Rat Red Blood Cells.

Abstract:To evaluate the effect of morphine on red blood cells, in vivo and in vitro rat models of morphine dependence were established. Rheological properties of rat red cells were measured by ektacytometry; the biophysical changes in the membrane of rat red cells were measured by the Fourier-Transformed Infrared technique (FT-IR) and the fluorescence depolarization method. The results show that the membrane fluidity of red cells was greatly reduced by morphine and the secondary structure of membrane proteins was changed. This suggests that morphine affects the rat red cell membrane directly, rather than through opioids-receptors.

Abstract:We studied the effects of self‐administered (SA) vs. experimenter‐administered (EA) morphine on dendritic spines in the hippocampal formation (CA1 and dentate), nucleus accumbens shell (NAcc‐s), sensory cortex (Par1 and Oc1), medial frontal cortex (Cg3), and orbital frontal cortex (AID) of rats. Animals in the SA group self‐administered morphine in 2‐h sessions (0.5 mg/kg/infusion, i.v.) for an average of 22 sessions and animals in the EA group were given daily i.v. injections of doses that approximated the total session dose for matched rats in Group SA (average cumulative dose/session of 7.7 mg/kg). Control rats were given daily i.v. infusions of saline. One month after the last treatment the brains were processed for Golgi‐Cox staining. In most brain regions (Cg3, Oc1, NAcc‐s) morphine decreased the density of dendritic spines, regardless of mode of administration (although to a significantly greater extent in Group SA). However, only SA morphine decreased spine density in the hippocampal formation and only EA morphine decreased spine density in Par1. Interestingly, in the orbital frontal cortex morphine significantly increased spine density in both Groups SA and EA, although to a much greater extent in Group SA. We conclude: 1) Morphine has persistent (at least 1 month) effects on the density of dendritic spines in many brain regions, and on many different types of cells (medium spiny neurons, pyramidal cells, and granule cells); 2) The effect of morphine on spine density (and presumably synaptic organization) varies as a function of both brain region and mode of drug administration; and 3) The ability of morphine to remodel synaptic inputs in a regionally specific manner may account for the many different long‐term sequelae associated with opioid use. Ultra‐high‐field pharmacological functional MRI of dopamine D1 receptor‐related interventions in anesthetized rats

Abstract:Dopamine D1 receptor manipulation induced BOLD signal changes in anesthetized rat brain which detected using pharmacological functional MRI. D1‐like receptor agonist increased, while the antagonist decreased the BOLD signal intensity in the striatum, thalamus, and cerebellum. Pharmacological functional MRI can be used for assessment of dopamine receptors‐related neurophysiological actions in live animals. The dopamine D1 receptor (D1R) is associated with schizophrenia, Parkinson's disease, and attention deficit hyperactivity disorder. Although the receptor is considered a therapeutic target for these diseases, its neurophysiological function has not been fully elucidated. Pharmacological functional MRI (phfMRI) has been used to evaluate regional brain hemodynamic changes induced by neurovascular coupling resulting from pharmacological interventions, thus phfMRI studies can be used to help understand the neurophysiological function of specific receptors. Herein, the blood oxygenation level‐dependent (BOLD) signal changes associated with D1R action in anesthetized rats was investigated by using a preclinical ultra‐high‐field 11.7‐T MRI scanner. PhfMRI was performed before and after administration of the D1‐like receptor agonist (SKF82958), antagonist (SCH39166), or physiological saline subcutaneously. Compared to saline, the D1‐agonist induced a BOLD signal increase in the striatum, thalamus, prefrontal cortex, and cerebellum. At the same time, the D1‐antagonist reduced the BOLD signal in the striatum, thalamus, and cerebellum by evaluating temporal profiles. PhfMRI detected D1R‐related BOLD signal changes in the brain regions associated with high expression of D1R. We also measured the early expression of c‐fos at the mRNA level to evaluate the effects of SKF82958 and isoflurane anesthesia on neuronal activity. Regardless of the presence of isoflurane anesthesia, c‐fos expression level was increased in the region where positive BOLD responses were observed with administration of SKF82958. These findings demonstrated that phfMRI could be used to identify the effects of direct D1 blockade on physiological brain functions and also for neurophysiological assessment of dopamine receptor functions in living animals.

Investigating the Influence of Morphine and Cocaine on the Mesolimbic Pathway Using a Novel Microimaging Platform

Abstract:Dopamine (DA)'s relationship with addiction is complex, and the related pathways in the mesocorticolimbic system are used to deliver DA, regulating both behavioral and perceptual actions. Specifically, the mesolimbic pathway connecting the ventral tegmental area (VTA) and the nucleus accumbens (NAc) is crucial in regulating memory, emotion, motivation, and behavior due to its responsibility to modulate dopamine. To better investigate the relationship between DA and addiction, more advanced mapping methods are necessary to monitor its production and propagation accurately and efficiently. In this study, we incorporate dLight1.2 adeno-associated virus (AAV) into our latest CMOS (complementary metal-oxide semiconductor) imaging platform to investigate the effects of two pharmacological substances, morphine and cocaine, in the NAc using adult mice. By implanting our self-fabricated CMOS imaging device into the deep brain, fluorescence imaging of the NAc using the dLight1.2 AAV allows for the visualization of DA molecules delivered from the VTA in real time. Our results suggest that changes in extracellular DA can be observed with this adapted system, showing potential for new applications and methods for approaching addiction studies. Additionally, we can identify the unique characteristic trend of DA release for both morphine and cocaine, further validating the underlying biochemical mechanisms used to modulate dopaminergic activation.
Sub-chronic exposure to morphine alters general anesthetic potency by differentially regulating the expression of neurotransmitter receptor subunits in mice

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3>Sub-chronic exposure to morphine can increase the efficacy of propofol but decrease the efficacy of ketamine by unknown mechanisms. The present study was designed to investigate the effects of sub-chronic exposure to morphine on the expression of neurotransmitter receptor subunits, which might contribute to the efficacy changes of ketamine and propofol in vivo.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3>Sub-chronic exposure to morphine was established by administering subcutaneous injections of morphine for 5 consecutive days. The median effective dose (ED50) of ketamine and/or propofol was measured on day 1, day 3, day 7 and day 15, after the last morphine dosage. Mice in the sham group received an equal volume of normal saline. The expressions of N-methyl D-aspartate (NMDA) receptor and γ-aminobutyric acid A (GABAA) receptor subunits in the forebrain were measured. Knockdown or overexpression of a subunit was used to determine the causality between the change in anesthetic potency and the expression of an identified receptor subunit.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3>After sub-chronic exposure of mice to morphine, the expression of NMDA receptor 1 (NR1) was most elevated in the forebrain on day 1 (P < 0.0001 vs. sham). In contrast, the expression of GABAA receptor β3 (GABAARβ3) gradually decreased to its lowest level on day 7 (P = 0.005 vs. sham) in the forebrain. Regression analysis revealed that the expression of NR1 in the forebrain was relevant to the increased ED50 of ketamine (P = 0.0002), while the expression of GABAARβ3 in the forebrain was relevant to the decreased ED50 of propofol (P = 0.0051) after morphine exposure. Knockdown expression of NR1 in the forebrain reversed the elevated ED50 of ketamine after morphine treatment. Overexpression of GABAARβ3 in the forebrain increased the ED50 of propofol to the sham-level after morphine treatment.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3>Sub-chronic exposure to morphine can differentially modulate the expressions of NR1 and GABAARβ3 in mice, which may contribute to the changes in ED50 of ketamine and propofol in vivo.

A comparison of D1 receptor binding and mRNA in rat brain using receptor autoradiographic and in situ hybridization techniques

Abstract:D1, a subtype of the dopamine receptors, is widely distributed in the nervous system and has been shown to be positively coupled to adenylate cyclase. Using a combination of in vitro receptor autoradiographic and in situ hybridization techniques, the present study examines the co-distribution of D1 receptor binding sites and D1 receptor messenger RNA in adjacent rat brain sections. D1 receptor binding sites were labeled using the selective antagonist [3H]SCH23390 (4.6 nM) in the presence of 1 μM ketanserin, while the D1 receptor messenger RNA was visualized with a35S-labeled riboprobe corresponding to a region between transmembrane domains III and VI of the rat D1 receptor (bp 383–843). Analysis of serial sections suggested a good agreement between D1 receptor binding and messenger RNA in several brain regions, including the paleocortex, caudate-putamen, nucleus accumbens, amygdala and suprachiasmatic nucleus. Marked discrepancies between D1 receptor binding and messenger RNA were observed in other brain regions including the entopeduncular and subthalamic nuclei, substantia nigra (pars reticulata), hippocampus and cerebellum. While technical considerations may contribute to these results, much of the discordance between the distributions is likely due to the differential localization D1 receptor messenger RNA in cell bodies and receptor binding sites on fibers and may provide insights into receptor synthesis, transport and membrane insertion. In the basal ganglia, for instance, D1 receptors are synthesized in the striatum and are either transported to efferent projections in areas such as the substantia nigra, or remain localized in striatal cells bodies. Ibotenic acid lesions in the striatum are consistent with these conclusions and demonstrate a coordinate loss of D1 receptor binding and messenger RNA in the caudate-putamen that is accompanied by a degeneration of fibers projecting to substantia nigra and a loss of D1 binding in the pars reticulata. Neurons in the dentate gyrus and in the granular layer of the cerebellum, on the other hand, synthesize D1 receptors and transport them entirely to either their dendritic or axonal fields, respectively, in the molecular layer.

Abstract:Dynorphin and catecholamine were measured in ischemic rat produced by four-vessel (2 vertebral arteries and 2 common carotid arteries) occlusion for 10 min. The results showed that: (1) The contents of dynorphine (pg/mg tissue) in cerebral cortex were 5.5 +/- 0.6 (n = 7) in normal rats and decreased to 4.9 +/- 0.5 (n = 9, P less than 0.05) in cerebral ischemic rats; with immediate ip phencyclidine (1-(1-phenylcyclophexyl)piperidine, PCP, 1 mg.kg-1), the contents of dynorphin were increased to 5.3 +/- 0.4 (n = 5, P less than 0.05 vs the ischemic rats). (2) The contents of DOPAC (pg/mg tissue) in cerebral cortex were 38 +/- 6 (n = 7) and increased to 120 +/- 60 (n = 5, P less than 0.05) in 10 min cerebral ischemic rats; with immediately ip PCP (1 mg.kg-1), the contents of DOPAC were decreased to 26 +/- 13 (n = 7, P less than 0.05 vs the ischemic rats). (3) The release of DA (pg/mg tissue) in cortical slices in vitro, in high K+ solution were 24 +/- 3 (n = 5) and significantly increased to 57 +/- 15 (n = 5, P less than 0.05) in ischemic rat brain slices; with immediate ip PCP (1 mg.kg-1), the contents of DA were decreased to 38 +/- 10 (n = 5, P less than 0.05 vs the ischemic rats). These results suggest PCP play an antagonistic role in cerebral ischemic damage of rats.
The effect of chronic administration of oxycodone on the behavioral functions and histopathology in the cerebellum and striatum of adult male rats

Abstract:Oxycodone is widely used for pain management and acts via binding to mu- and kappa opioid receptors. It was shown that extended oxycodone usage can result from the demyelination and degeneration of neurons through the stress response, which triggers apoptotic signaling pathways. The striatum and cerebellum are recognized as significant contributors to addiction; however, there is no report on the effect of oxycodone on the cerebellum and striatum and motor coordination. We treated rats daily with oxycodone at 15 mg/kg doses for thirty days. Motor performance and electromyography activity were then evaluated. Stereological methods were performed to assess the number of neurons in the cerebellum and striatum as well as immunohistochemistry for microgliosis and astrogliosis. Furthermore, the Sholl analysis method was utilized to evaluate the cellular structure of both microglia and astrocytes. Results of the rotarod test for motor coordination show no significant (P < 0.05) difference between the oxycodone subjects and those in the control group. In addition, open-field assessments indicated that the application of oxycodone did not alter the amount of distance covered (as an indicator of locomotion) or time spent in the central area (as an indicator of anxiety) (P < 0.001). The electromyography (EMG) test result showed that oxycodone caused a delay in the reaction of the muscular nerves (P < 0.001). Data and results from our experiment revealed that administering oxycodone did not affect astrogliosis and the number of neurons in the cerebellum and striatum (P < 0.05). In contrast, it altered neuromuscular function. In addition, oxycodone administration activated microglia in the cerebellum and striatum. In conclusion, we encourage more research on the adverse effects of oxycodone on the brain.
Morphine Differentially Alters the Synaptic and Intrinsic Properties of D1R- and D2R-Expressing Medium Spiny Neurons in the Nucleus Accumbens

Abstract:Exposure to opioids reshapes future reward and motivated behaviors partially by altering the functional output of medium spiny neurons (MSNs) in the nucleus accumbens shell. Here, we investigated how morphine, a highly addictive opioid, alters synaptic transmission and intrinsic excitability on dopamine D1-receptor (D1R) expressing and dopamine D2-receptor (D2R) expressing MSNs, the two main output neurons in the nucleus accumbens shell. Using whole-cell electrophysiology recordings, we show, that 24 h abstinence following repeated non-contingent administration of morphine (10 mg/kg, i.p.) in mice reduces the miniature excitatory postsynaptic current (mEPSC) frequency and miniature inhibitory postsynaptic current (mIPSC) frequency on D2R-MSNs, with concomitant increases in D2R-MSN intrinsic membrane excitability. We did not observe any changes in synaptic or intrinsic changes on D1R-MSNs. Last, in an attempt to determine the integrated effect of the synaptic and intrinsic alterations on the overall functional output of D2R-MSNs, we measured the input-output efficacy by measuring synaptically-driven action potential firing. We found that both D1R-MSN and D2R-MSN output was unchanged following morphine treatment.

Kinetic modeling of receptor-ligand binding applied to positron emission tomographic studies with neuroleptic tracers

Abstract:Positron emission tomography (PET) with labeled neuroleptics has made possible the study of neurotransmitter-receptor systems in vivo. In this study we investigate the kinetics of the 3,4-dihydroxyphenylethylamine (dopamine) receptor-ligand binding using PET data from a series of experiments in the baboon with the 18F-labeled drugs spiperone, haloperidol, and benperidol. Models used to describe these systems are based on first-order kinetics which applies at high specific activity (low receptor occupancy). The parameters governing the uptake and loss of drug from the brain were found by fitting PET data from regions with little or no receptor concentration (cerebellum) and from experiments in which specific binding was blocked by pretreatment with the drug (+)-butaclamol. Receptor constants were determined by fitting data from receptor-containing structures. Correcting the arterial plasma activities (the model driving function) for the presence of drug metabolites was found to be important in the modeling of these systems.
