Abstract:Calcium phosphate materials are widely used as bone-like scaffolds or coating for metallic hip and knee implants due to their excellent biocompatibility, compositional similarity to natural bone and controllable bioresorbability. Local delivery of drugs or osteogenic factors from scaffolds and implants are required over a desired period of time for an effectual treatment of various musculoskeletal disorders. Curcumin, an antioxidant and anti-inflammatory molecule, enhances osteoblastc activity in addition to its anti-osteoclastic activity. However, due to its poor solubility and high intestinal liver metabolism, it showed limited oral efficacy in various preclinical and clinical studies. To enhance its bioavailability and to provide higher release, we have used poly (ε-caprolactone) (PCL), poly ethylene glycol (PEG) and poly lactide co glycolide (PLGA) as the polymeric system to enable continuous release of curcumin from the hydroxyapatite matrix for 22 days. Additionally, curcumin was incorporated in plasma sprayed hydroxyapatite coated Ti6Al4V substrate to study in vitro cell material interaction using human fetal osteoblast (hFOB) cells for load bearing implants. MTT cell viability assay and morphological characterization by FESEM showed highest cell viability with samples coated with curcumin-PCL-PEG. Finally, 3D printed interconnected macro porous β-TCP scaffolds were prepared and curcumin-PCL-PEG was loaded to assess the effects of curcumin on in vivo bone regeneration. The presence of curcumin in TCP results in enhanced bone formation after 6 weeks. Complete mineralized bone formation increased from 29.6 % to 44.9% in curcumin-coated scaffolds compared to pure TCP. Results show that local release of curcumin can be designed for both load bearing or non-load bearing implants with the aid of polymers, which can be considered an excellent candidate for wound healing and tissue regeneration applications in bone tissue engineering.

Abstract:Aim: In this study, a nanosponge structure was synthesised with capability of encapsulating curcumin as a model polyphenolic compound and one of the herbal remedies that have widely been considered due to its ability to treat cancer.Methods: FTIR, DSC and XRD techniques were performed to confirm the formation of the inclusion complex of the nanosponge-drug.Results: DSC and XRD patterns showed an increasing stability and a decreasing crystallinity of curcumin after formation of inclusion complex. Encapsulation efficiency was 98% (w/w) and a significant increase was observed in loading capacity (184% w/w). The results of cytotoxicity assessments demonstrated no cell toxicity on the healthy cell line, while being toxic against cancer cells. Haemolysis test was performed to evaluate the blood-compatibility characteristic of nanosponge and complex and the results showed 0.54% haemolysis in the lowest complex concentration (50μgml−1) and 5.09% at the highest concentration (200μgml−1).Conclusions: Thus, the introduced system could be widely considered in cancer treatment as a drug delivery system.

Controlled Release of Curcumin and Hypocrellin A from Electrospun Poly(l-Lactic Acid)/Silk Fibroin Nanofibers for Enhanced Cancer Cell Inhibition

Abstract:Nanofibers have emerged as a highly effective method for drug delivery, attributed to their remarkable porosity and ability to regulate drug release rates while minimizing toxicity and side effects. In this study, we successfully loaded the natural anticancer drugs curcumin (CUR) and hypocrellin A (HA) into pure poly(l-lactic acid) (PLLA) and PLLA-silk protein (PS) composite nanofibers through electrospinning technology. This result was confirmed through comprehensive analysis involving SEM, FTIR, XRD, DSC, TG, zeta potential, and pH stability analysis. The encapsulation efficiency of all samples exceeded 85%, demonstrating the effectiveness of the loading process. Additionally, the drug release doses were significantly higher in the composites compared to pure PLLA, owing to the enhanced crystallinity and stability of the silk proteins. Importantly, the composite nanofibers exhibited excellent pH stability in physiological and acidic environments. Furthermore, the drug-loaded composite nanofibers displayed strong inhibitory effects on cancer cells, with approximately 28% (HA) and 37% (CUR) inhibition of cell growth and differentiation within 72 h, while showing minimal impact on normal cells. This research highlights the potential for controlling drug release through the manipulation of fiber diameter and crystallinity, paving the way for wider applications of electrospun green nanomaterials in the field of medicine.

Abstract:This study investigates the impact of curcumin extract incorporation on the properties of PVA/Collagen/Chitosan/HAp nanofiber scaffolds for bone tissue engineering applications. Nanofibers were fabricated via electrospinning using PVA, collagen, chitosan, hydroxyapatite (HAp), and curcumin. Their morphology, mechanical properties, wettability, degradation rate, and antibacterial activity were comprehensively evaluated. Fourier transform infrared (FTIR) spectroscopy confirmed the successful incorporation of all components into the nanofibers and the potential for interactions between their functional groups. Scanning Electron Microscopy (SEM) revealed bead-free, fine nano-fibers with a decrease in diameter ranging from 139 nm to 108 nm. Simultaneously, enhanced mechanical properties were observed, with ultimate strength increasing from 2.8 MPa to 8.8 MPa as the diameter decreased. Notably, 15% curcumin extract maintained favorable hydrophilicity and degradation rates, crucial factors for tissue regeneration. Antibacterial activity was also enhanced, with an inhibition zone of 6.71 mm against E.coli observed at 15% curcumin treatment. Staphylococcus aureus exhibited the largest zone of inhibition (8.74 mm) at 15% curcumin concentration. This research demonstrates the feasibility of incorporating curcumin into nanofiber scaffolds for bone tissue engineering applications.

Abstract:Gas chromatography/multiphoton ionization/time-of-flight mass spectrometry (GC/MPI/TOF-MS) was applied to a soil sample to survey several groups of polychlorinated aromatic hydrocarbons (polychlorinated dibenzo- p -dioxins/dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), polychlorinated naphthalenes (PCNs), polychlorinated diphenylethers (PCDEs), and polychlorinated terphenyls (PCTs)). The signal peaks in the two-dimensional display of GC/MS could be easily and accurately assigned from the intensity distribution of the isotope peaks, even in the presence of numerous interfering species. Using this technology, mutual interferences between organochlorine compounds can be readily recognized from the data of the two-dimensional display after a measurement, although the separation of these compounds is sometimes difficult using high-resolution magnetic-sector-type mass spectrometry. This approach, based on MPI, results in less fragmentation, and is useful for the identification of analytes. Thus, GC/MPI/TOF-MS allows for the simultaneous determination of PCDD/Fs and related compounds in real samples containing numerous interfering species.

Abstract:ABSTRACT We previously optimized imipenem and tobramycin combination regimens against a double-resistant clinical Pseudomonas aeruginosa isolate by using in vitro infection models, mechanism-based pharmacokinetic/pharmacodynamic modeling (MBM), and Monte Carlo simulations. The current study aimed to evaluate these regimens in a neutropenic murine thigh infection model and to characterize the time course of bacterial killing and regrowth via MBM. We studied monotherapies and combinations of imipenem with tobramycin in vivo against the double-resistant clinical P. aeruginosa isolate by using humanized dosing schemes. Viable count profiles of total and resistant populations were quantified over 24 h. Tobramycin monotherapy (7 mg/kg every 24 h [q24h] as a 0.5-h infusion) was ineffective. Imipenem monotherapies (continuous infusion of 4 or 5 g/day with a 1-g loading dose) yielded 2.47 or 2.57 log10 CFU/thigh killing at 6 h. At 24 h, imipenem at 4 g/day led to regrowth up to the initial inoculum (4.79 ± 0.26 log10 CFU/thigh), whereas imipenem at 5 g/day displayed 1.75 log10 killing versus the initial inoculum. The combinations (i.e., imipenem at 4 or 5 g/day plus tobramycin) provided a clear benefit, with bacterial killing of ≥2.51 or ≥1.50 log10 CFU/thigh compared to the respective most active monotherapy at 24 h. No colonies were detected on 3×MIC agar plates for combinations, whereas increased resistance (at 3×MIC) emerged for monotherapies (except imipenem at 5 g/day). MBM suggested that tobramycin considerably enhanced the imipenem target site concentration up to 2.6-fold. The combination regimens, rationally optimized via a translational modeling approach, demonstrated substantially enhanced bacterial killing and suppression of regrowth in vivo against a double-resistant isolate and are therefore promising for future clinical evaluation.

Abstract:A pH-sensitive nanocomposite based on halloysite nanotubes (HNT) coated with polyacrylic acid (PAA)/carboxymethyl cellulose (CMC) hydrogel was synthesized for controlled delivery of curcumin (CUR) to MCF-7 cancer cells. Hazelnut oil was added to minimize the nanocomposite size and increase its uniformity via water/oil/water (W/O/W) double emulsion method. Likewise, Span 80 surfactant was incorporated to produce a nanoniosomal emulsion that increased the stability and retention of CUR as an anticancer drug. The molecular interactions between the nanocomposite components, its crystalline structure, surface morphology, size distribution and surface charge were analyzed through Fourier transformed infrared (FT-IR) spectroscopy, field emission-scanning electron microscope (FE-SEM), X-ray diffraction (XRD), dynamic light scattering (DLS) and zeta potential measurements, respectively. The resulting drug delivery system showed a 45% drug loading and an 87% encapsulation efficiency, which are amongst the highest values reported up to date for CUR nanocarriers. The zeta potential of the nanocomposite was about 36 mV, indicating good stability. A sustained drug release was observed, with 61% and 96% CUR released at pH 7.4 and 5.4 after 96 h, respectively, which confirms the pH-sensitivity of the developed nanocarrier. MTT assay demonstrated the strong toxicity of the nanocomposites against MCF-7 cancer cells. Also, 46% of cell death rate in the apoptotic phase was observed by flow cytometry. The results obtained herein support the great potential of PAA/CMC/HNT/CUR hydrogel nanocomposites as smart drug delivery vehicles for breast cancer treatment.

Dual drug release mechanisms through mesoporous silica nanoparticle/electrospun nanofiber for enhanced anticancer efficiency of curcumin

Abstract:Electrospun nanofibers (NFs)-based drug delivery approaches are of particular interest as a hopeful implantable nanoplatform for localized cancer therapy and treating tissue defect after resection, allowing the on-site drug delivery with minimal side effect to healthy cells. To maintain therapeutic concentrations of anticancer molecules for a relatively long time through a combination of burst and sustained drug release mechanisms, a hybrid of polycaprolactone and gelatin (PCL/GEL) was used for co-encapsulation of free curcumin (CUR) and CUR-loaded mesoporous silica nanoparticles (CUR@MSNs) via electrospinning, resulting in a novel drug-loaded nanofibrous scaffold, CUR/CUR@MSNs-NFs. The as-prepared MSNs and composite NFs were characterized via TGA, FTIR, FE-SEM, TEM, and BET. In vitro release profile of CUR from CUR/CUR@MSNs-NFs was examined, and the in vitro antitumor efficacy against MDA-MB-231 breast cancer cells was also evaluated through MTT, scratch assay, DAPI staining, and real-time PCR. The results disclosed that the smooth, bead-free, and randomly oriented CUR/CUR@MSNs-NFs displayed a combination of initial rapid discharge and sustained release for CUR, which led to higher cytotoxicity, lower migration as well as a more pronounced effect on apoptosis induction than CUR-NFs and CUR@MSNs-NFs. The present study illustrated that the dual drug release mechanisms through MSN/NF-mediated drug delivery systems might have a highly hopeful application as a localized implantable scaffold for potential postoperative breast cancer therapy.

Abstract:Oral bone defects occur as a result of trauma, cancer, infections, periodontal diseases, and caries. Autogenic and allogenic grafts are the gold standard used to treat and regenerate damaged or defective bone segments. However, these materials do not possess the antimicrobial properties necessary to inhibit the invasion of the numerous deleterious pathogens present in the oral microbiota. In the present study, poly(ε-caprolactone) (PCL), nano-hydroxyapatite (nHAp), and a commercial extract of Humulus lupulus L. (hops) were electrospun into polymeric matrices to assess their potential for drug delivery and bone regeneration. The fabricated matrices were analyzed using scanning electron microscopy (SEM), tensile analysis, thermogravimetric analysis (TGA), FTIR assay, and in vitro hydrolytic degradation. The antimicrobial properties were evaluated against the oral pathogens Streptococcus mutans, Porphyromonas gingivalis, and Aggregatibacter actinomycetemcomitans. The cytocompatibility was proved using the MTT assay. SEM analysis established the nanostructured matrices present in the three-dimensional interconnected network. The present research provides new information about the interaction of natural compounds with ceramic and polymeric biomaterials. The hop extract and other natural or synthetic medicinal agents can be effectively loaded into PCL fibers and have the potential to be used in oral applications.

Stark effect of intrinsic and extrinsic charge-transfer excitons in a linear donor-acceptor stack: anthracene-pyromellitic dianhydride.

Abstract:Anthracene-PMDA single crystals display at 2K about 70 meV below a well-known intrinsic charge-transfer exciton three narrow absorption lines, which are attributed to CT excitons bound to defects of a few 10(-5) concentration. All excitons respond very sensitively to electric fields along the molecular stack because of the large dipole moment, about 2 eA, of an ionized donor-acceptor pair, but only intrinsic excitons observe an optical selection rule. Although the triclinic unit cell contains only one pair of molecules, excitons appear in field-modulated spectra as near-degenerate doublets of different parity with very small splitting. The line shape of the EA spectra and selection rules with respect to the polarization of light and orientation of the field are consistent with the inversion symmetry of the lattice and the molecules. The simple crystal structure enables identification of the defects that are responsible for extrinsic excitons. Symmetry consideration based on translation invariance lead to a new interpretation of intrinsic excitons as true crystal states with charge transfer from the donor to the acceptor sublattice.
Bronchoalveolar Lavage plus Surfactant in a Piglet Model of Meconium Aspiration Syndrome

Abstract:Introduction: Meconium aspiration produces airway obstruction and surfactant inhibition. Bronchoalveolar lavage (BAL) and surfactant replacement have been proposed as treatments for the syndrome. Objective: To evaluate the effect of BAL with normal saline followed by a supplementary dose of surfactant in a piglet model of meconium aspiration syndrome. Methods: 15 newborn piglets were used in the study. The animals were ventilated with fixed settings. After inhalation of 4 ml/kg of diluted meconium, the piglets were randomized into three groups: group I (n = 5) – tracheal aspiration without BAL; group II (n = 5) – BAL with normal saline (15 ml/kg), and group III (n = 5) – BAL with normal saline (15 ml/kg) followed by a supplementary dose of surfactant (Curosurf® 100 mg/kg). Arterial blood gas samples were obtained 30 min and 6 h after the inhalation of meconium. Results: A significant increase of PaO2 values at 6 h after treatment was only observed in group III (from 51 ± 13 to 189 ± 115 mm Hg; p = 0.04). At this time, PaO2 in group III was significantly higher compared to group II (189 ± 115 and 37 ± 11 mm Hg, respectively; p = 0.023) and showed a borderline significance when compared to group I (p = 0.066). Conclusion: BAL with normal saline followed by a supplementary dose of surfactant may improve oxygenation in an experimental piglet model of meconium aspiration syndrome.
Development and Characterization of Polyester and Acrylate-Based Composites with Hydroxyapatite and Halloysite Nanotubes for Medical Applications

Abstract:We aimed to study the distribution of hydroxyapatite (HA) and halloysite nanotubes (HNTs) as fillers and their influence on the hydrophobic character of conventional polymers used in the biomedical field. The hydrophobic polyester poly (ε-caprolactone) (PCL) was blended with its more hydrophilic counterpart poly (lactic acid) (PLA) and the hydrophilic acrylate poly (2-hydroxyethyl methacrylate) (PHEMA) was analogously compared to poly (ethyl methacrylate) (PEMA) and its copolymer. The addition of HA and HNTs clearly improve surface wettability in neat samples (PCL and PHEMA), but not that of the corresponding binary blends. Energy-dispersive X-ray spectroscopy mapping analyses show a homogenous distribution of HA with appropriate Ca/P ratios between 1.3 and 2, even on samples that were incubated for seven days in simulated body fluid, with the exception of PHEMA, which is excessively hydrophilic to promote the deposition of salts on its surface. HNTs promote large aggregates on more hydrophilic polymers. The degradation process of the biodegradable polyester PCL blended with PLA, and the addition of HA and HNTs, provide hydrophilic units and decrease the overall crystallinity of PCL. Consequently, after 12 weeks of incubation in phosphate buffered saline the mass loss increases up to 48% and mechanical properties decrease above 60% compared with the PCL/PLA blend.

Influence of molecular interactions on structure, controlled release and cytotoxicity of curcumin encapsulated chitosan - Silica nanostructured microspheres

Abstract:Curcumin possesses numerous medicinal benefits including anti-cancer and anti-viral properties. However, its wide scale use as a drug is often hindered owing to the dearth of suitable drug-delivery systems which can solubilise it for long-term sustained-release and safeguard its beneficial properties. In this work, a fast, one-step method, employing evaporation induced assembly of colloids, has been employed for the synthesis of curcumin encapsulated organic-inorganic hybrid micron-sized spheres. Detailed physical properties of the microspheres, with scaffolds of silica nanoparticles (∼8.5 nm) cross linked by chitosan, are studied to trace the underlying mechanism of structural assembly in such systems, by tuning the polymer matrix with solubilizing agents, DMSO and Tween 20. A systematic modification in the hydrogen bonding network, conformations and interactions between macromolecules is revealed upon tuning the organic matrix. This in turn is found to control the assembly vis-à-vis the granular morphology, drug entrapment and packing fraction of nanoparticles in the microspheres, which have direct influence on the biological properties. Consequently, the microspheres are found to follow a first order drug release kinetics with a tunable rate constant which follows the ordering of packing fraction of silica nanoparticles in the micro-granules. A sustained curcumin release for a period extending up to 24 h has been achieved. Further studies using human lung and cervical cancer cell lines assert good anti-cancer properties of these nanostructured microspheres in cancer cells, while showing no toxicity towards normal cells. Thus, such hybrid organic-inorganic formulations achieved using multi-component colloidal assembly approach, with enhanced stability against degradation, are promising candidates for future clinical applications of water-insoluble drugs.

Abstract:A casein-methyl cellulose nanocomplex, loaded with curcumin and coated with dextran (DX-CasCur-MC), is designed to enhance curcumin's oral delivery and inhibit cancer growth. Its physicochemical properties reveal chemical bonding between protein and polysaccharides, transforming curcumin from crystalline into amorphous state to improve water solubility. The encapsulation efficiency of curcumin reaches 92%, and its release profile in physiological and tumor microenvironments exhibits controlled and sustained release. In vitro studies confirm the significant therapeutic efficacy of DX-CasCur-MC in inducing cancer cell death and DNA damage compared to free curcumin. The effectiveness of DX-CasCur-MC for oral drug delivery is validated in simulated gastrointestinal fluids, with 23 and 69% release in gastric and intestinal fluids, respectively. In vivo studies demonstrate a significant reduction in tumor volume in mice treated with DX-CasCur-MC compared to those treated with free curcumin or untreated, confirming DX-CasCur-MC's ability to improve curcumin's pharmacological properties and inhibit tumor growth via repeated oral administration. The conjugation of the two polysaccharides with the hydrocolloidal casein nanomicelles improves the nanocomplexes stability, making DX-CasCur-MC a promising natural candidate for oral curcumin delivery with a significant cancer therapeutic efficacy.

Abstract:In this study, the feasibility of the production of electrospun nanofibre composite mainly for biomedical applications is reported. Biocompatible polyether-based polyurethane, natural proteinaceous polymer silk sericin (SS) and natural inorganic nanoclay halloysite as a drug carrier with a model drug chlorhexidine acetate were used to produce nanofibres by electrospinning technique. Sericin was extracted from Bombyx mori silk cocoons by high pressure high temperature (HT-HP) degumming. Chlorhexidine acetate (CA), an antimicrobial agent, was loaded into halloysite nanotubes (HNTs) at different weight ratios, and 1:1 weight ratio showed the maximum loading which was confirmed by TGA and XRD analysis. Electrospinning of polymer solution with different compositions of polyurethane, sericin, CA and CA-HNTs was conducted at 10% w/v concentration, 20 kV voltage, 15 μl min −1 flow rate and 10 cm distance which resulted in the formation of bead-free uniform fibres. Antimicrobial activity of nanofibrous webs was evaluated by the disc diffusion method (AATCC 90) and it was found that CA and CA-HNT loaded nanofibres show sustained antibacterial action against both the Gram-positive (S. aureus) and Gram-negative (E. coli) bacteria. The CA-HNT and sericin/CA-HNT loaded nanofibres showed controlled release of CA. In addition, the cytocompatibility assessment of developed nanofibrous composites showed good biocompatibility. Hence the produced composite nanofibre can serve as an excellent material for sustained drug release for various biomedical applications.
Fabrication and characterization of silk fibroin-derived curcumin nanoparticles for cancer therapy

Abstract:Biologically derived nanoparticles (<100 nm) were fabricated for local and sustained therapeutic curcumin delivery to cancer cells. Silk fibroin (SF) and chitosan (CS) polymers were blended noncovalently to encapsulate curcumin in various proportions of SF and CS (75:25, 50:50, and 25:75 SF:CS) or pure SF at two concentrations (0.1% w/v and 10% w/v) using the devised capillary-microdot technique. Curcumin-polymer conjugates were frozen, lyophilized, crystallized, suspended in phosphate-buffered saline for characterization, and tested for efficacy against breast cancer cells. All nanoparticle formulations except 0.1% w/v 50:50 SFCS were less than 100 nm in size as determined with the transmission electron microscopy. The entrapment and release of curcumin over eight days was highest for SF-derived nanoparticles as compared to all SFCS blends. The uptake and efficacy of SF-coated curcumin was significantly higher (p < 0.001) than SFCS-coated curcumin in both low and high Her2/neu expressing breast cancer cells. Interestingly, the uptake of curcumin was highest for the high Her2/neu expressing breast cancer cells when delivered with a 10% w/v SF coating as compared to other formulations. In conclusion, SF-derived curcumin nanoparticles show higher efficacy against breast cancer cells and have the potential to treat in vivo breast tumors by local, sustained, and long-term therapeutic delivery as a biodegradable system.

Abstract:Herein, we report a drug eluting scaffold composed of a composite nanofibers of poly(ε-caprolactone) (PCL) and poly(glycerol sebacate) (PGS) loaded with Hydroxyapatite nanoparticles (HANPs) and simvastatin (SIM) mimicking the bone extracellular matrix (ECM) to improve bone cell proliferation and regeneration process. Indeed, the addition of PGS results in a slight increase in the average fiber diameter compared to PCL. However, the presence of HANPs in the composite nanofibers induced a greater fiber diameter distribution, without significantly changing the average fiber diameter. The in vitro drug release result revealed that the sustained release of SIM from the composite nanofiber obeying the Korsemeyer–Peppas and Kpocha models revealing a non-Fickian diffusion mechanism and the release mechanism follows diffusion rather than polymer erosion. Biomineralization assessment of the nanofibers was carried out in simulated body fluid (SBF). SEM and EDS analysis confirmed nucleation of the hydroxyapatite layer on the surface of the composite nanofibers mimicking the natural apatite layer. Moreover, in vitro studies revealed that the PCL-PGS-HA displayed better cell proliferation and adhesion compared to the control sample, hence improving the regeneration process. This suggests that the fabricated PCL-PGS-HA could be a promising future scaffold for control drug delivery and bone tissue regeneration application.

Abstract:Composite electrospun nanofibers based on polycaprolactone (PCL) have shown promise in various biomedical applications due to their unique properties. This study investigates the effects of incorporating hydroxyl (–OH)- or carboxylic acid (–COOH)-functionalized multiwall carbon nanotubes (MWCNTs) into PCL matrices. Two types of functionalized additives, MWCNT-OH and MWCNT-COOH, were used at different concentrations (0.06 and 0.12 wt%). Various characterization techniques including FTIR, XRD, AFM, SEM, water contact angle analysis, and tensile strength testing were employed to evaluate changes in nanofiber morphology, crystallinity, surface topography, wettability, and mechanical properties. In addition, in vitro cytotoxicity assays were conducted using HUVECs and L929 fibroblasts over 1-, 3-, and 5-day intervals. This study represents a novel examination of (–OH)- and (–COOH)-functionalized MWCNTs as additives in electrospun PCL biopolymer matrices. The findings indicate that incorporating small amounts of (–OH)- or (–COOH)-functionalized MWCNTs enhances the physicochemical characteristics of PCL nanofibers, making them more suitable for biomedical applications. While both types of functionalized MWCNT additives improved properties compared to pure PCL nanofibers, (–COOH)-functionalized MWCNT-incorporated nanofibers exhibited the most favorable features. In conclusion, this research highlights the potential of tailored PCL-based composite nanofibers containing functionalized MWCNTs as advanced biomaterial systems for biomedical applications, contributing to the development of innovative biomaterials for diverse biomedical contexts.

Abstract:Herein, a biodegradable and biocompatible composite comprising of support membrane based on crosslinked PVA/PEG film and curcumin loaded electrospun poly(lactic acid) (PLA) nanofiber mat is introduced. The membrane film was prepared from PVA/PEG blend followed by crosslinking with an optimum amount of citric acid, 15 wt.%. After then, PLA solutions with different curcumin content, 0-11 wt.%, were electrospinned on the prepared membrane substrate. The prepared film showed high water absorption, water vapor transmission rate and superior mechanical properties with improved elastic modulus, tensile strength and with an elongation of around 320 % with respect to the non-crosslinked one. Also, the scanning electron microscopy was revealed uniformly dispersed pores throughout the membrane film with a nearly narrow in size distribution centered at 36 μm. As well, a nanostructure porous morphology was found for the electrospun fibrous curcumin loaded PLA from the scanning electron microscopy micrographs and the average fiber diameter was decreased with curcumin content. In vitro drug release from the prepared flexible composite into the vertical diffusion cell was recorded by the measuring curcuminoids content using high performance liquid chromatography and drug release kinetic evaluations were revealed that the release pattern of all prepared samples, containing different content of curcumin, well fitted to the Higuchi’s model signifying diffusion-controlled release mechanism. As well, the determined release rate at the second release stages, i.e. steady state flux (J), was varied from 0.31 to 43.53 μg·cm-2·h-1 with increasing drug content from 1 to 11 wt.%. Regarding this results, this flexible composite by providing the moist environment along with miraculous healing properties of curcumin, can be potential candidate for transdermal drug delivery.
