Abstract:In order to investigate the effect of deep brain stimulation on diseases such as epilepsy, we developed a closed-loop electrical stimulation system using LabVIEW virtual instrument environment and NI data acquisition card. The system was used to detect electrical signals of epileptic seizures automatically and to generate electrical stimuli. We designed a novel automatic detection algorithm of epileptic seizures by combining three features of field potentials: the amplitude, slope and coastline index. Experimental results of rat epileptic model in the hippocampal region showed that the system was able to detect epileptic seizures with an accuracy rate 91.3% and false rate 8.0%. Furthermore, the on-line high frequency electrical stimuli showed a suppression effect on seizures. In addition, the system was adaptive and flexible with multiple work modes, such as automatic and manual modes. Moreover, the simple time-domain algorithm of seizure detection guaranteed the real-time feature of the system and provided an easy-to-use equipment for the experiment researches of epilepsy control by electrical stimulation.

Abstract:Natural sensory inputs, such as speech and music, are often rhythmic. Recent studies have consistently demonstrated that these rhythmic stimuli cause the phase of oscillatory, i.e. rhythmic, neural activity, recorded as local field potential (LFP), electroencephalography (EEG) or magnetoencephalography (MEG), to synchronize with the stimulus. This phase synchronization, when not accompanied by any increase of response power, has been hypothesized to be the result of phase resetting of ongoing, spontaneous, neural oscillations measurable by LFP, EEG, or MEG. In this article, however, we argue that this same phenomenon can be easily explained without any phase resetting, and where the stimulus-synchronized activity is generated independently of background neural oscillations. It is demonstrated with a simple (but general) stochastic model that, purely due to statistical properties, phase synchronization, as measured by 'inter-trial phase coherence', is much more sensitive to stimulus-synchronized neural activity than is power. These results question the usefulness of analyzing the power and phase of stimulus-synchronized activity as separate and complementary measures; particularly in the case of attempting to demonstrate whether stimulus-synchronized neural activity is generated by phase resetting of ongoing neural oscillations.
Optimizing real-time phase detection in diverse rhythmic biological signals for phase-specific neuromodulation

Abstract:Closed-loop, phase-specific neurostimulation is a powerful method to modulate ongoing brain activity for clinical and research applications. Phase-specific stimulation relies on estimating the phase of an ongoing oscillation in real time and issuing a control command at a target phase. Phase detection algorithms based on Fast Fourier transform (FFT) are widely used due to their computational efficiency and robustness. However, it is unclear how algorithm performance depends on the spectral properties of the input signal and how algorithm parameters can be optimized. We used offline simulation to evaluate the performance of three algorithms (endpoint-corrected Hilbert Transform, Hilbert Transform and phase mapping) on three rhythmic biological signals with distinct spectral properties (rodent hippocampal theta potential, human EEG alpha and human essential tremor). First, we found that algorithm performance was more strongly influenced by signal amplitude and frequency variation compared with signal to noise ratio. Second, our simulations showed that the size of the data window for phase estimation was critical for the performance of FFT-based algorithms, where the optimal data window corresponds to the period of the oscillation. We validated this prediction with real time phase detection of hippocampal theta oscillations in freely behaving rats performing spatial navigation. Our findings define the relationship between signal properties and algorithm performance and provide a convenient method for optimizing FFT-based phase detection algorithms.

An Automatic HFO Detection Method Combining Visual Inspection Features with Multi-Domain Features

Abstract:As an important promising biomarker, high frequency oscillations (HFOs) can be used to track epileptic activity and localize epileptogenic zones. However, visual marking of HFOs from a large amount of intracranial electroencephalogram (iEEG) data requires a great deal of time and effort from researchers, and is also very dependent on visual features and easily influenced by subjective factors. Therefore, we proposed an automatic epileptic HFO detection method based on visual features and non-intuitive multi-domain features. To eliminate the interference of continuous oscillatory activity in detected sporadic short HFO events, the iEEG signals adjacent to the detected events were set as the neighboring environmental range while the number of oscillations and the peak-valley differences were calculated as the environmental reference features. The proposed method was developed as a MatLab-based HFO detector to automatically detect HFOs in multi-channel, long-distance iEEG signals. The performance of our detector was evaluated on iEEG recordings from epileptic mice and patients with intractable epilepsy. More than 90% of the HFO events detected by this method were confirmed by experts, while the average missed-detection rate was < 10%. Compared with recent related research, the proposed method achieved a synchronous improvement of sensitivity and specificity, and a balance between low false-alarm rate and high detection rate. Detection results demonstrated that the proposed method performs well in sensitivity, specificity, and precision. As an auxiliary tool, our detector can greatly improve the efficiency of clinical experts in inspecting HFO events during the diagnosis and treatment of epilepsy.
A Scalable Real-Time Architecture for Neural Oscillation Detection and Phase-Specific Stimulation

Abstract:Oscillations in the local field potential (LFP) of the brain are key signatures of neural information processing. Perturbing these oscillations at specific phases in order to alter neural information processing is an area of active research. Existing systems for phase-specific brain stimulation typically either do not offer real-time timing guarantees (desktop computer based systems) or require extensive programming of vendor-specific equipment. This work presents a real-time detection system architecture that is platform-agnostic and that scales to thousands of recording channels, validated using a proof-of-concept microcontroller-based implementation.

Abstract:Bandpass filters play a core role in ECoG signal processing. Commonly used frequency bands such as alpha, beta, and gamma bands can reflect the normal rhythm of the brain. However, the universally predefined bands might not be optimal for a specific task. Especially the gamma band usually covers a wide frequency span (i.e., 30-200 Hz) which can be too coarse to capture features that appear in narrow bands. An ideal option is to find the optimal frequency bands for specific tasks in real-time and dynamically. To tackle this problem, we propose an adaptive band filter that selects the useful frequency band in a data-driven way. Specifically, we leverage the phase-amplitude coupling (PAC) of the coupled working mechanism of synchronizing neuron and pyramidal neurons in neuronal oscillations, in which the phase of slower oscillations modulates the amplitude of faster ones, to help locate the fine frequency bands from the gamma range, in a task-specific and individual-specific way. Thus, the information can be more precisely extracted from ECoG signals to improve neural decoding performance. Based on this, an end-to-end decoder (PACNet) is proposed to construct a neural decoding application with adaptive filter banks in a uniform framework. Experiments show that PACNet can improve neural decoding performance universally with different tasks.

Abstract:Objective: Closed-loop neurostimulation (CLNS) procedures, aligning stimuli with electrical brain activity, are quickly gaining popularity in neuroscience. They have been employed to reveal causal links between neural activity patterns and function, and to explore therapeutic effects of electroencephalography (EEG-)guided stimulations during sleep. Most CLNS procedures are developed for a single purpose, detecting one specific pattern of interest in the EEG. Furthermore, most procedures have limited, if any, flexibility to adapt to temporal or interindividual variance in the signal, which means they wouldn't work optimally across the full physiological phenomenology. Approach: Here we present a new approach to CLNS, based on real-time signal modelling to predict brain activity, allowing targeting of a broad variety of oscillatory dynamics. Intrinsic to the modelling approach is adaptation to signal variance, such that no personalization steps prior to use are necessary. We systematically assess stimulus targeting performance of modelling-based CLNS (M-CLNS), across a wide range of brain oscillation frequencies and phases in human and rodent neurophysiological signals. Main results: Our results show high performance for all target phases and frequency bands, including slow oscillations, theta and alpha waves. Significance: These findings highlight the general applicability and adaptability of M-CLNS, which also favors its application in populations with an atypical oscillatory signature, like clinical or elderly populations. In conclusion, M-CLNS provides a promising new tool for neural activity-dependent stimulation in both experimental research and therapeutic applications, such as enhancing deep sleep in patients with sleep disorders.

Abstract:Neural oscillations have been linked to multiple behaviors and neuropsychiatric disorders. The individual contributions to behavior from both oscillations and non-oscillatory activity are still unclear, complicating efforts to link neurophysiology to cognition, hindering the discovery of novel biomarkers, and preventing the development of effective therapeutics. To overcome these hurdles, it will be critical to investigate the biological origins of neural oscillations by characterizing the dynamic properties of different brain regions. The dynamical regime for a population of neurons generating oscillations in neural recordings can be discovered by stimulating the population and recording its subsequent response to stimulation. There are different dynamical regimes that can produce population-level neural oscillations. For certain dynamical regimes, like that of a nonlinear oscillator, the phase response curve (PRC) can help differentiate the dynamic state of the population. The PRC can be measured by stimulating the population across different phases of its oscillatory state. However, neural dynamics are non-stationary, so neural oscillations will vary in frequency and amplitude across a recording and the PRC can change over time. This non-stationarity could bias a PRC estimated from an electrophysiological experiment, preventing accurate characterization of a neural population's dynamics. This necessitates tools that can operate online to trigger stimulation and update PRC estimates. To that end, we develop online methods for tracking non-stationary oscillations and recovering PRCs corrupted by estimation errors. We validate the performance of our non-stationary oscillation estimator compared to both a known ground truth model and an alternative phase estimation approach. We demonstrate that a PRC can be recovered online under different random error conditions in silico and that a similar amplitude response curve (ARC) can be estimated from physiologic data using online methods compared to offline approaches.

An Energy Efficient AdaBoost Cascade Method for Long-term Seizure Detection in Portable Neurostimulators.

Abstract:Responsive neurostimulation (RNS) is becoming a promising therapy in refractory epilepsy control. In a RNS system, a critical challenge is how to detect seizure onsets accurately with low computational costs. In this study, an energy efficient AdaBoost cascade method for robust long-term seizure detection from local field potential (LFP) signals was proposed and evaluated in a portable neurostimulator. The AdaBoost cascade method included two stages: a seizure candidate detection stage (stage1) and a false alarm rejection stage (stage2). Since seizure activities occurred occasionally in most cases, most normal signal segments can be efficiently classified in stage1. A small percent of suspicious segments were fed into stage2 for more strict examination, where more sophisticated features were extracted to precisely identify seizure activities and reduce false alarms. To further optimize energy efficiency for hardware implementation, we proposed a soft-cascade algorithm for stage2 to reduce the computational costs. Our method was implemented in a generalized neurostimulator and evaluated online with four rats with chronic temporal lobe epilepsy (TLE). A total of 2280.1 hours of LFP signals were recorded and analyzed. Our approach achieves a detection rate of 91.6%, 3.85/h false alarm rate, and 2.31 second detection delay. With the two-stage cascade approach, 98.13% of computational costs could be reduced, with respect to the time of calculation of all features. Our method can detect seizure onsets precisely with high energy efficiency, which is suitable for hardware implementation in portable neuro-stimulators. Therefore, this proposed approach is promising to provide effective and robust performances in long-term seizure detection in neurostimulators for responsive seizure control.
Response of neuronal populations to phase-locked stimulation: model-based predictions and validation

Abstract:Background: Modulation of neuronal oscillations holds promise for the treatment of neurological disorders. Nonetheless, stimulating neuronal populations in a continuous open-loop manner can lead to side effects and suboptimal efficiency. Closed-loop strategies such as phase-locked stimulation aim to address these shortcomings by offering a more targeted modulation. While theories have been developed to understand the neural response to stimulation, their predictions have not been thoroughly tested using experimental data. Objective: We aimed to test the predictions of a mathematical model regarding the response of neuronal populations to phase-locked stimulation. Methods: Using a coupled oscillator model, we expanded on two key predictions describing the response to stimulation as a function of the phase and amplitude of ongoing neural activity. To investigate these predictions, we analyzed electrocorticogram (ECoG) recordings from a previously conducted study in Parkinsonian rats, and extracted the corresponding phase and response curves. Results: We demonstrated that the amplitude response to stimulation is strongly correlated to the derivative of the phase response (𝜌 > 0.8) in all animals except one, thereby validating a key model prediction. The second prediction postulated that the stimulation becomes ineffective when the network synchrony is high, a trend that appeared missing in the data. Our analysis explained this discrepancy by showing that the neural populations in Parkinsonian rats did not reach the level of synchrony for which the theory would predict ineffective stimulation. Conclusions: Our results highlight the potential of fine tuning stimulation paradigms informed by mathematical models that consider both the ongoing phase and amplitude of the targeted neural oscillation.

Abstract:Closed-loop neuromodulation, especially using the phase of the electroencephalography (EEG) rhythm to assess the real-time brain state and optimize the brain stimulation process, is becoming a hot research topic. Because the EEG signal is non-stationary, the commonly used EEG phase-based prediction methods have large variances, which may reduce the accuracy of the phase prediction. In this study, we proposed a machine learning-based EEG phase prediction network, which we call EEG phase prediction network (EPN), to capture the overall rhythm distribution pattern of subjects and map the instantaneous phase directly from the narrow-band EEG data. We verified the performance of EPN on pre-recorded data, simulated EEG data, and a real-time experiment. Compared with widely used state-of-the-art models (optimized multi-layer filter architecture, auto-regress, and educated temporal prediction), EPN achieved the lowest variance and the greatest accuracy. Thus, the EPN model will provide broader applications for EEG phase-based closed-loop neuromodulation.

Novel Cyclic Homogeneous Oscillation Detection Method for High Accuracy and Specific Characterization of Neural Dynamics

Abstract:Detecting temporal and spectral features of neural oscillations is essential to understanding dynamic brain function. Traditionally, the presence and frequency of neural oscillations are determined by identifying peaks over 1/f noise within the power spectrum. However, this approach solely operates within the frequency domain and thus cannot adequately distinguish between the fundamental frequency of a non-sinusoidal oscillation and its harmonics. Non-sinusoidal signals generate harmonics, significantly increasing the false-positive detection rate — a confounding factor in the analysis of neural oscillations. To overcome these limitations, we define the fundamental criteria that characterize a neural oscillation and introduce the Cyclic Homogeneous Oscillation (CHO) detection method that implements these criteria based on an auto-correlation approach that determines the oscillation’s periodicity and fundamental frequency. We evaluated CHO by verifying its performance on simulated sinusoidal and non-sinusoidal oscillatory bursts convolved with 1/f noise. Our results demonstrate that CHO outperforms conventional techniques in accurately detecting oscillations. Specifically, we determined the sensitivity and specificity of CHO as a function of signal-to-noise ratio (SNR). We further assessed CHO by testing it on electrocorticographic (ECoG, 8 subjects) and electroencephalographic (EEG, 7 subjects) signals recorded during the pre-stimulus period of an auditory reaction time task and on electrocorticographic signals (6 SEEG subjects and 6 ECoG subjects) collected during resting state. In the reaction time task, the CHO method detected auditory alpha and pre-motor beta oscillations in ECoG signals and occipital alpha and pre-motor beta oscillations in EEG signals. Moreover, CHO determined the fundamental frequency of hippocampal oscillations in the human hippocampus during the resting state (6 SEEG subjects). In summary, CHO demonstrates high precision and specificity in detecting neural oscillations in time and frequency domains. The method’s specificity enables the detailed study of non-sinusoidal characteristics of oscillations, such as the degree of asymmetry and waveform of an oscillation. Furthermore, CHO can be applied to identify how neural oscillations govern interactions throughout the brain and to determine oscillatory biomarkers that index abnormal brain function.

Robust Adaptive Deep Brain Stimulation Control of Non-Stationary Cortex-Basal Ganglia-Thalamus Network Models in Parkinson’s Disease

Abstract:Closed-loop deep brain stimulation (DBS) is a promising therapy for Parkinson’s disease (PD) that works by adjusting DBS patterns in real time from the guidance of feedback neural activity. Current closed-loop DBS mainly uses threshold-crossing on-off controllers or linear time-invariant (LTI) controllers to regulate the basal ganglia (BG) beta band oscillation power. However, the critical cortex-BG-thalamus network dynamics underlying PD are nonlinear, non-stationary, and noisy, hindering the accurate and robust control of PD neural dynamics using current closed-loop DBS methods. Here, we develop a new robust adaptive closed-loop DBS method for regulating cortex-BG-thalamus network dynamics in PD. We first build an adaptive state-space model to quantify the dynamic, nonlinear, and non-stationary neural activity. We then construct an adaptive estimator to track the nonlinearity and non-stationarity in real time. We next design a robust controller to automatically determine the DBS frequency based on the estimated PD neural state while reducing the system’s sensitivity to high-frequency noise. We adopt and tune a biophysical cortex-BG-thalamus network model as a testbed to simulate various nonlinear and non-stationary neural dynamics for evaluating DBS methods. We find that under different nonlinear and non-stationary neural dynamics, our robust adaptive DBS method achieved accurate regulation of the BG beta band oscillation power with small control error, bias, and deviation. Moreover, the accurate regulation generalizes across different therapeutic targets and consistently outperforms state-of-the-art on-off and LTI DBS methods. These results have implications for future designs of clinically-viable closed-loop DBS systems to treat PD and other neurological and neuropsychiatric disorders. ### Competing Interest Statement The authors have declared no competing interest.
Design of a novel closed-loop neural electrical stimulation system

Abstract:In order to exert electrical stimulation at a given phase of brain electric signal waveforms, a closed-loop system was developed using a general NI data acquisition card together with LabVIEW virtual instrument environment. One of the A/D channels on the card is used to record the field potential signals from the hippocampal region in a rat brain. A customized LabVIEW program analyzes the amplitude and frequency features of the signals, and then predicts the upcoming peak or trough of the theta rhythms in the field potentials. Finally, stimulation waveforms are outputted from a digital I/O port or a D/A channel of the card exactly at the expected time phase to realize the closed-loop stimulation. Experimental results show that the percent prediction accuracies are 92% and 86% for peak and trough predictions, respectively. The electrical stimulation system is versatile and flexible, and can be used for predicting various rhythm phases and other features appeared in neural signals. It provides a novel closed-loop control method for neural stimulation both in neuroelectrophysiology researches and in electrical stimulation therapy of brain diseases.
Benchmarking real-time algorithms for in-phase auditory stimulation of low amplitude slow waves with wearable EEG devices during sleep

Abstract:Auditory stimulation of EEG slow waves (SW) during non-rapid eye movement (NREM) sleep has shown to improve cognitive function when it is delivered at the up-phase of SW. SW enhancement is particularly desirable in subjects with low-amplitude SW such as older adults or patients suffering from neurodegeneration such as Parkinson disease (PD). However, existing algorithms to estimate the up-phase suffer from a poor phase accuracy at low EEG amplitudes and when SW frequencies are not constant. We introduce two novel algorithms for real-time EEG phase estimation on autonomous wearable devices. The algorithms were based on a phase-locked loop (PLL) and, for the first time, a phase vocoder (PV). We compared these phase tracking algorithms with a simple amplitude threshold approach. The optimized algorithms were benchmarked for phase accuracy, the capacity to estimate phase at SW amplitudes between 20 and 60 microV, and SW frequencies above 1 Hz on 324 recordings from healthy older adults and PD patients. Furthermore, the algorithms were implemented on a wearable device and the computational efficiency and the performance was evaluated on simulated sleep EEG, as well as prospectively during a recording with a PD patient. All three algorithms delivered more than 70% of the stimulation triggers during the SW up-phase. The PV showed the highest capacity on targeting low-amplitude SW and SW with frequencies above 1 Hz. The testing on real-time hardware revealed that both PV and PLL have marginal impact on microcontroller load, while the efficiency of the PV was 4% lower than the PLL. Active auditory stimulation did not influence the phase tracking. This work demonstrated that phase-accurate auditory stimulation can be delivered during home-based sleep interventions with a wearable device also in populations with low-amplitude SW.
The Portiloop: a deep learning-based open science tool for closed-loop brain stimulation

Abstract:Closed-loop brain stimulation refers to capturing neurophysiological measures such as electroencephalography (EEG), quickly identifying neural events of interest, and producing auditory, magnetic or electrical stimulation so as to interact with brain processes precisely. It is a promising new method for fundamental neuroscience and perhaps for clinical applications such as restoring degraded memory function; however, existing tools are expensive, cumbersome, and offer limited experimental flexibility. In this article, we propose the Portiloop, a deep learning-based, portable and low-cost closed-loop stimulation system able to target specific brain oscillations. We first document open-hardware implementations that can be constructed from commercially available components. We also provide a fast, lightweight neural network model and an exploration algorithm that automatically optimizes the model hyperparameters to the desired brain oscillation. Finally, we validate the technology on a challenging test case of real-time sleep spindle detection, with results comparable to off-line expert performance on the Massive Online Data Annotation spindle dataset (MODA; group consensus). Software and plans are available to the community as an open science initiative to encourage further development and advance closed-loop neuroscience research.

Abstract:Summary The slow oscillation is a central neuronal dynamic during sleep, and is generated by alternating periods of high and low neuronal activity (ON‐ and OFF‐states). Mounting evidence causally links the slow oscillation to sleep's functions, and it has recently become possible to manipulate the slow oscillation non‐invasively and phase‐specifically. These developments represent promising clinical avenues, but they also highlight the importance of improving our understanding of how ON/OFF‐states affect incoming stimuli and what role they play in neuronal plasticity. Most studies using closed‐loop stimulation rely on the electroencephalogram and local field potential signals, which reflect neuronal ON‐ and OFF‐states only indirectly. Here we develop an online detection algorithm based on spiking activity recorded from laminar arrays in mouse motor cortex. We find that online detection of ON‐ and OFF‐states reflects specific phases of spontaneous local field potential slow oscillation. Our neuronal‐spiking‐based closed‐loop procedure offers a novel opportunity for testing the functional role of slow oscillation in sleep‐related restorative processes and neural plasticity.

Automatic Detection of High-Frequency Oscillations With Neuromorphic Spiking Neural Networks

Abstract:Interictal high-frequency oscillations (HFO) detected in electroencephalography recordings have been proposed as biomarkers of epileptogenesis, seizure propensity, disease severity, and treatment response. Automatic HFO detectors typically analyze the data offline using complex time-consuming algorithms, which limits their clinical application. Neuromorphic circuits offer the possibility of building compact and low-power processing systems that can analyze data on-line and in real time. In this review, we describe a fully automated detection pipeline for HFO that uses, for the first time, spiking neural networks and neuromorphic technology. We demonstrated that our HFO detection pipeline can be applied to recordings from different modalities (intracranial electroencephalography, electrocorticography, and scalp electroencephalography) and validated its operation in a custom-designed neuromorphic processor. Our HFO detection approach resulted in high accuracy and specificity in the prediction of seizure outcome in patients implanted with intracranial electroencephalography and electrocorticography, and in the prediction of epilepsy severity in patients recorded with scalp electroencephalography. Our research provides a further step toward the real-time detection of HFO using compact and low-power neuromorphic devices. The real-time detection of HFO in the operation room may improve the seizure outcome of epilepsy surgery, while the use of our neuromorphic processor for non-invasive therapy monitoring might allow for more effective medication strategies to achieve seizure control. Therefore, this work has the potential to improve the quality of life in patients with epilepsy by improving epilepsy diagnostics and treatment.
Null Mutation in PGAP1 Impairing Gpi-Anchor Maturation in Patients with Intellectual Disability and Encephalopathy

Abstract:Many eukaryotic cell-surface proteins are anchored to the membrane via glycosylphosphatidylinositol (GPI). There are at least 26 genes involved in biosynthesis and remodeling of GPI anchors. Hypomorphic coding mutations in seven of these genes have been reported to cause decreased expression of GPI anchored proteins (GPI-APs) on the cell surface and to cause autosomal-recessive forms of intellectual disability (ARID). We performed homozygosity mapping and exome sequencing in a family with encephalopathy and non-specific ARID and identified a homozygous 3 bp deletion (p.Leu197del) in the GPI remodeling gene PGAP1. PGAP1 was not described in association with a human phenotype before. PGAP1 is a deacylase that removes an acyl-chain from the inositol of GPI anchors in the endoplasmic reticulum immediately after attachment of GPI to proteins. In silico prediction and molecular modeling strongly suggested a pathogenic effect of the identified deletion. The expression levels of GPI-APs on B lymphoblastoid cells derived from an affected person were normal. However, when those cells were incubated with phosphatidylinositol-specific phospholipase C (PI-PLC), GPI-APs were cleaved and released from B lymphoblastoid cells from healthy individuals whereas GPI-APs on the cells from the affected person were totally resistant. Transfection with wild type PGAP1 cDNA restored the PI-PLC sensitivity. These results indicate that GPI-APs were expressed with abnormal GPI structure due to a null mutation in the remodeling gene PGAP1. Our results add PGAP1 to the growing list of GPI abnormalities and indicate that not only the cell surface expression levels of GPI-APs but also the fine structure of GPI-anchors is important for the normal neurological development.
A spiking neural network (SNN) for detecting high frequency oscillations (HFOs) in the intraoperative ECoG

Abstract:Abstract To achieve seizure freedom, epilepsy surgery requires the complete resection of the epileptogenic brain tissue. In intraoperative electrocorticography (ECoG) recordings, high frequency oscillations (HFOs) generated by epileptogenic tissue can be used to tailor the resection margin. However, automatic detection of HFOs in real-time remains an open challenge. Here we present a spiking neural network (SNN) for automatic HFO detection that is optimally suited for neuromorphic hardware implementation. We trained the SNN to detect HFO signals measured from intraoperative ECoG on-line, using an independently labeled dataset (58 min, 16 recordings). We targeted the detection of HFOs in the fast ripple frequency range (250-500 Hz) and compared the network results with the labeled HFO data. We endowed the SNN with a novel artifact rejection mechanism to suppress sharp transients and demonstrate its effectiveness on the ECoG dataset. The HFO rates (median 6.6 HFO/min in pre-resection recordings) detected by this SNN are comparable to those published in the dataset (Spearman’s $$\rho$$ ρ = 0.81). The postsurgical seizure outcome was “predicted” with 100% (CI [63 100%]) accuracy for all 8 patients. These results provide a further step towards the construction of a real-time portable battery-operated HFO detection system that can be used during epilepsy surgery to guide the resection of the epileptogenic zone.
