Abstract:Current medical countermeasures (MCMs) for nerve agent poisoning have limited efficacy, and can cause serious adverse effects, prompting the requirement for new broad-spectrum therapeutics. Human plasma-derived butyrylcholinseterase (huBChE) is a promising novel bioscavenger MCM which has shown potential in animal studies, however, is economically prohibitive to manufacture at scale. This study addresses current challenges for the economical production of a bioactive and long-acting recombinant huBChE (rBChE) in mammalian cells by being the first to directly compare novel rBChE design strategies. These include co-expression of a proline rich attachment domain (PRAD) and fusion of BChE with a protein partner. Additionally, a pre-purification screening method developed in this study enables parallel comparison of the expression efficiency, activity and broad-spectrum binding to nerve agents for ten novel rBChE molecular designs. All designed rBChE demonstrated functionality to act as broad-spectrum MCMs to G, V and A series nerve agents. Expression using the ExpiCHO™ Max protocol provided greatest expression levels and activity for all constructs, with most rBChE expressing poorly in Expi293™. Fc- or hSA-fused rBChE significantly outperformed constructs designed to mimic huBChE, including PRAD-BChE, and proved an effective strategy to significantly improve enzyme activity and expression. Choice of protein partner, directionality and the addition of a linker also impacted fusion rBChE activity and expression. Overall, hSA fused rBChE provided greatest expression yield and activity, with BChE-hSA the best performing construct. The purified and characterized BChE-hSA demonstrated similar functionality to huBChE to be inhibited by GD, VX and A-234, supporting the findings of the pre-screening study and validating its capacity to assess and streamline the selection process for rBChE constructs in a cost-effective manner. Collectively, these outcomes contribute to risk mitigation in early-stage development, providing a systematic method to compare rBChE designs and a focus for future development.

Abstract:Summary Carboxylesterase BioH of Escherichia coli is an important enzyme in the biotin synthesis.It contains a typical catalytic triad of Ser-His-Asp and displays significant carboxylesterase activities.The enzyme shows a broad substrate specificity with a preference for short acyl chain substrates.This property makes it a biocatalyst with a good application prospect.However,research on the use of this enzyme has been quite few to date.It shows great significance to get the protein and optimize its functions. In this study,BioH gene from E.coli K12 was cloned into pET30a(+) using double digestion method by BamHI and HindIII and was overexpressed in E.coli BL21(DE3) induced by isopropyl-β-D-thiogalactoside (IPTG).BioH protein content was calculated by image processing software Quantity One.Its hydrolysis activity was determined by the rate of hydrolyzing p-nitrophenyl butyrate(p-NPB).And its thermostability was detected after the enzyme was incubated at 60 ℃ for 40 min.In order to improve its hydrolysis activity,a directed evolution method was carried out.p-NPB was used as the screening substrate.The product,p-nitrophenol,showed the biggest absorption value at 405 nm.The mutant library was built by error-prone PCR method with adjusted Mn2+concentration. The complete length of the BioH gene was 771 bp and the molecular mass of the protein with 256 amino acids was about 28.2 ku.The targeted enzyme accounted for one quarter of the total expressed proteins.The enzyme??s hydrolysis activity was shown as 18 U/mg and it retained 70% of its hydrolysis activity after incubated at 60 ℃ for 40 min. The thermostability of the enzyme was the foundation in the enzyme application. The optimized concentration of Mn2+ in error-prone PCR was chosen as 0.1 mmol/L determined by agarose electrophoresis of the brightness of PCR product band.And mutant library with a mortality rate of 20% to 40% was appropriate for selection.Sequence detection showed that the mutation rate was kept as 2 3 changed bases in each gene on average.In each round of directed evolution,a library with 5 000 6 000 mutants was screened.In the first round,wild-type BioH was used as the parent and three mutants with improved hydrolysis activity were selected. These mutants were K213E,Q70L/M1 70T and V1 75A,and their hydrolysis activities were improved by 10%, 30% and 50% respectively.Subsequently,the two mutants with only one mutation site for each,K213E and V1 75A,were used as parents to undergo the next round of directed evolution.Three mutants from K213E were selected,namely K213E/M1 97L, K213E/L180M and K213E/C31Y/F50S, whose hydrolysis activities were improved by 40%,50% and 95% respectively.In the mutant library of V1 75A,one mutant,V1 75A/Q129R, was selected,which showed 100% improvement in hydrolysis activity compared to the wild type.Structure analysis by molecular 3D structure software PyMOL gave a clear view of the mutation sites.Sites 31,129,1 75, 180 were located in the α-helixes and sites 50,70,1 70,1 97,213 were located in the loops of protein structure. All of them were far away from the activity sites. This study clones and overexpresses successfully the carboxylesterase BioH in BL21(DH3).Seven mutants were selected by directed evolution in p-NPB method.The enzyme shows a good evolvability.The positive library can be used in further application of this enzyme.
[Construction and Verification of the Effectiveness of Pmbl: a Cloning Vector of Exported Proteins Encoding Genes].

Abstract:The beta-lactamase was used as the reporter of expression and transmembrane secretion in this paper. A fragment of Amp resistance gene encoding the mature part of beta-lactamase (delta P delta SP Amp, i.e. without promoter and signal peptide coding sequences) was amplified from pUC18 vector. The upstream primer has BglII, BclI, BamHI in three reading frame respectively, in order to in frame fuse and express target genes together with the downstream reporter in finally constructed vector. Meanwhile, pET-28 was digested with the restriction enzymes BglII and Bst1107 I. The 2.8 kb fragment with replication origin, Kan resistance gene and MCS was recovered, filled, self-ligated and resulted in a plasmid pKan-B. The Bgl II site on pKan-B was then filled and the plasmid pKan was obtained. The delta P delta SP Amp gene, which was first cloned into pGEM-T-EASY vector, was inserted into pKan between EcoR I and XbaI sites. A plasmid pMBL-E was selected, with which the bacteria host could grew on Kan plate but not on plate with both Amp and Kan. An EcoRI site beside HindIII on the plasmid pMBL-E was then filled, and the plasmid pMBL, a cloning vector of the exported proteins encoding genes was finally obtained. Both results of the restriction enzyme digestion and sequencing demonstrated the correctness of the construction. The Tet resistance gene, a transmemebrane protein encoding gene, was applied to verify the effectiveness of the reporter in the vector. Cut with EcoRI and BamHI, a 375 bp fragment including promoter and 96 animo acids coding sequence (including signal peptide) of Tet was obtained from pBR322 vector. The fragment was then ligated to the vector pMBL which had been cut with both enzymes of EcoRI and BglI, or EcoRI and BclI, or EcoRI and BamHI (as 0, +1, +2 respectively of the beta-lactamase gene reading frame). Kan and Amp double resistant colonies only grew with the EcoRI and BglII combination (0 position). Restriction enzyme digestion and sequencing results of the recombinant plasmid showed that Tet resistance gene, which promoted the expression and transmembrane secretion of downstream beta-lactamase, was inserted in a correct reading frame into the vector. Thus, the results verified the effectiveness of the constructed vector pMBL, which may be used effectively to clone genes encoding exported proteins with promoters and signal peptide sequences.

Abstract:A series of N,N-3-phenyl-3-benzylaminopropanamide derivatives were identified as novel CETP (cholesteryl ester transfer protein) inhibitors. In our previous study, lead compound L10 was discovered by pharmacophore-based virtual screening (Dong-Mei Zhao et al., 2014). Based on L10 (IC50 8.06μM), compound HL6 (IC50 10.7μM) was discovered following systematic structure variation and biological tests. Further optimization of the structure–activity relationship (SAR) resulted in N,N-3-phenyl-3-benzylaminopro panamides derivatives as novel CETP inhibitors. They were synthesized and evaluated against CETP by BODIPY-CE fluorescence assay. Among them, HL16 (IC50 0.69μM) was a highly potent CETP inhibitor in vitro. In addition, HL16 exhibited favorable HDL-C enhancement and LDL-C reduction in vivo by hamster. The molecular docking of HL16 into the CETP was performed. The binding mode demonstrated that HL16 occupied the CETP binding site and formed interactions with the key amino acid residues.
Enhanced Acylation Activity of Esterase BioH from Escherichia Coli by Directed Evolution Towards Improved Hydrolysis Activity

Abstract:Esterase BioH is a critical enzyme for Biotin synthesis in Escherichia coli, which has been previously found to be active in the acylation of secondary alcohols and amines. Directed evolution towards improved acylation activity requires a high-throughput screening method. The aim of this study is to explore whether the acylation activity of BioH can be improved by directed evolution of its hydrolysis activity. A colorimetric method based on p-nitrophenyl butyrate hydrolysis was adopted in the high-throughput determination of hydrolysis activity. The wild-type BioH showed a hydrolysis activity of 18 U/mg, and the specific activities for alpha-phenylethanol and alpha-phenylethylamine acylation were 12.8 U/mg and 3.5 U/mg, respectively. After two rounds of directed evolution, seven mutants with improved hydrolysis activity were obtained, among which, K213E, Q70L/M170T and M197L/K213E also showed improvement in acylation activity. To further improve the acylation activity, site mutations were generated in different combinations at the four hot spots Q70L, M170T, M197L and K213E. Among the resulting mutants, Q70L/M197L/K213E showed the highest activity in alpha-phenylethylamine acylation with a 120% improvement, while Q70L/K213E had the highest alpha-phenylethanol acylation activity, which was improved by 70%. The results demonstrated that directed evolution of the hydrolysis activity might be a possible approach to improve the acylation activity of the esterase BioH. (C) 2013 Elsevier B.V. High-level extracellular protein production in Bacillus subtilis using an optimized dual-promoter expression system

Abstract:BackgroundWe recently constructed a Bacillus subtilis strain (CCTCC M 2016536) from which we had deleted the srfC, spoIIAC, nprE, aprE and amyE genes. This strain is capable of robust recombinant protein production and amenable to high-cell-density fermentation. Because the promoter is among the factors that influence the production of target proteins, optimization of the initial promoter, PamyQ from Bacillus amyloliquefaciens, should improve protein expression using this strain. This study was undertaken to develop a new, high-level expression system in B. subtilis CCTCC M 2016536.ResultsUsing the enzyme β-cyclodextrin glycosyltransferase (β-CGTase) as a reporter protein and B. subtilis CCTCC M 2016536 as the host, nine plasmids equipped with single promoters were screened using shake-flask cultivation. The plasmid containing the PamyQ′ promoter produced the greatest extracellular β-CGTase activity; 24.1 U/mL. Subsequently, six plasmids equipped with dual promoters were constructed and evaluated using this same method. The plasmid containing the dual promoter PHpaII–PamyQ′ produced the highest extracellular β-CGTase activity (30.5 U/mL) and was relatively glucose repressed. The dual promoter PHpaII–PamyQ′ also mediated substantial extracellular pullulanase (90.7 U/mL) and α-CGTase expression (9.5 U/mL) during shake-flask cultivation, demonstrating the general applicability of this system. Finally, the production of β-CGTase using the dual-promoter PHpaII–PamyQ′ system was investigated in a 3-L fermenter. Extracellular expression of β-CGTase reached 571.2 U/mL (2.5 mg/mL), demonstrating the potential of this system for use in industrial applications.ConclusionsThe dual-promoter PHpaII–PamyQ′ system was found to support superior expression of extracellular proteins in B. subtilis CCTCC M 2016536. This system appears generally applicable and is amenable to scale-up.

Abstract:Effective gene therapy requires regulated gene expression and vector safety. We developed a strategy to exponentially increase native promoter activity while retaining inherent regulation by inserting multi-copy response elements (REs) into non-adjacent locations. For the hepatocyte nuclear factor (HNF) 4α-dependent Hnf1a (MODY3) gene, HNF4α stimulation increased from 5- to 90-fold by inserting 3 additional HNF4α REs (H4REs). Constructing a promoter with two 4xH4REs 0.25 kb apart by duplicating the 4xH4RE fragment increased stimulation to >1000-fold. HNF4α-induced protein expression by the duplicate 4xH4RE Hnf1a promoter was comparable to a viral promoter. Converting the two Apolipoprotein C3 (ApoC3) H4REs spaced 0.61 kb apart to 4xH4REs achieved a similar result. Increasing spacing to 2.1 kb with non-promoter DNA abolished the augmentation. Finally, converting the HNF1α RE of the HNF4A (MODY1) P2 promoter to 4xH1RE and adding a second 4xH1RE 0.84 kb upstream increased HNF1α stimulation from 26- to >200-fold. Deleting intervening DNA to produce 0.23-kb spacing increased stimulation to >500-fold. Spaced multi-copy RE motifs is a novel strategy for engineering promoters that boosts activity far beyond other techniques. Augmentation of three promoters suggests that this approach is potentially applicable to other promoters for gene therapy and might obviate the need for viral promoters.
Permeabilizing Escherichia Coli for Whole Cell Biocatalyst with Enhanced Biotransformation Ability from L-Glutamate to Gaba

Abstract:γ-Aminobutyric acid (GABA) is widely used as a pharmaceutical, nutraceutical, and as a precursor for synthesizing materials for industrial use. Bacterial cells can be exploited for use as biocatalysts with the potential to synthesize compounds such as GABA with greater efficiency, safety, and economy. However, efforts to use biocatalysts must overcome the permeability barrier of the cell envelope. Therefore, to produce a whole-cell biocatalyst with enhanced cell-associated glutamate decarboxylase (GAD) activity, we overexpressed the Glutamate (Glu)-GABA antiporter (GadC) in Escherichia coli (BL21(DE3)-pET28a-gadB). The cell-associated GAD activity of the transformants was higher by a factor of 2.6 in comparison; however, expression of GadC inhibited growth. Therefore, we permeabilized the cells using either organic solvents, surfactants, or heat. Permeabilization with organic solvents increased cell-associated GAD activity as a function of their hydrophobicity, and hexane was the most effective, increasing cell-associated GAD activity by a factor of 9.65 (6.72U/mg). The surfactants Triton X-100, CHAPS, NP-40, OGP, and Brij-35 enhanced cell-associated GAD activity, and Triton X-100 was the most effective, increasing cell-associated GAD activity by a factor of 10.8 (7.53U/mg). Heating BL21(DE3)-pET28a-gadB at 70°C for 30min increased cell-associated GAD activity by a factor of 13.1. GAD did not leak from the permeabilized cells under optimum conditions. When the heat-permeabilized cells were immobilized on Ca-alginate gel beads, the biotransformation ability of beads maintained over 60% of their initial ability after 20 consecutive batches, and the beads retained 90% of their initial activity after 30 days of storage. These approaches for improving cell permeability to enhance cell-associated GAD activity show great promise for decreasing the cost of industrial production of GABA.
A highly effective and stable butyrylcholinesterase inhibitor with multi-faceted neuroprotection and cognition improvement

Abstract:Butyrylcholinesterase (BChE) has been more and more attractive for treating neurodegenerative diseases, especially Alzheimer's disease (AD). In this study, we conducted activity and druggability optimization based on the structures that were previously reported. Most compounds exhibited pronounced BChE inhibitory capacity with nanomolar IC50 values. Based on the results of inhibiting activity and cyto-safety evaluations, two compounds (7, eqBChE IC50 = 2.94 nM, hBChE IC50 = 34.6 nM, and 20, eqBChE IC50 = 0.15 nM, hBChE IC50 = 45.2 nM) have been selected as candidates. High stability of compound 20 contributed to significantly improved blood concentration and tissue exposure, resulting in a reduced administration and effective dose in pharmacodynamic experiments. Two candidates exhibited remarkable neuroprotective properties and cognition improving activity, by benefiting cholinergic system, reducing the total Aβ amount and increasing the ghrelin content. Simultaneous modulation in the center and periphery greatly improves the efficiency of BChE inhibitors. Considering the regulation on ghrelin level, BChE inhibition could improve not only symptoms but also nutritional status of AD patients.

Abstract:The wider application of creatinase, an enzyme used to determine renal function, is limited by the yield problem and current production cannot meet the needs of society. Therefore, the work constructed an efficient B. subtilis expression strain for creatinase production based on cell leakage. Initial expression of creatinase in B. subtilis was achieved using the native promoter P-hpaII. Subsequently, the promoter was optimized, and the efficient expression of creatinase was realized by using the maltose-induced promoter P-glv, which increased the enzyme activity 5-fold and the properties of creatinase were analyzed. Meanwhile, it was found that creatinase without a signal peptide could be secreted into the extracellular medium, so the secretion of creatinase in B. subtilis was investigated. Disabling the classical secretory pathways didn't have an effect on the expression level of extracellular creatinase, indicating that the excretion of creatinase didn't depend on these pathways. Calcein-AM/PI double staining results showed that the membrane of the expression strain was damaged, while electron microscopy images revealed that leakage sites were present on the surface of the expressing bacteria, which led to the passive leakage of creatinase from B. subtilis. Finally, the enzyme activity was further improved to 141.9 U/mL in 5-L high-density fermentation.
Engineering and validation of a dual luciferase reporter system for quantitative and systematic assessment of regulatory sequences in Chinese hamster ovary cells

Abstract:Abstract Ongoing research efforts to identify potent regulatory sequences that deliver robust and sustained transgene expression are critical for Chinese hamster ovary (CHO) cell line development technologies to meet the growing demand for recombinant proteins. Here we report the engineering and validation of a highly customizable single vector toolkit that comprises an all-in-one dual luciferase reporter system for quantitative and systematic interrogation of transcriptional regulatory sequences in transient and stable transfectants of CHO cells. To model the execution of the reporter system, we implemented a battery of known constitutive promoters including human CMV-mIE, SV40, HSV-TK, mouse PGK, human EF1α, EF1α short (EFS), human UBC, synthetic CAG, and Chinese hamster EF1α (CHEF1α). Of the nine promoters, CMV-mIE yielded the highest transcriptional activity in transient transfection settings, while CHEF1α was the strongest among a select subset of promoters in stable transfectants of CHO-DG44 pools. Remodeling the vector toolkit to build a dual fluorescent reporter system featured an alternative to bioluminescence based reporters. We infer that the findings of this study may serve as a basis to establish new vectors with weak or strong constitutive promoters. Furthermore, the modular all-in-one architecture of the reporter system proved to be a viable tool for discovering novel regulatory sequences that ensure high levels of transient and stable transgene expression in CHO and perhaps other mammalian cell lines.

Construction of PTIE2-CREG-EGFP-N1 eukaryotic expression plasmid and its expression in mouse artery endothelial cells

Abstract:BACKGROUND: The molecular mechanism of blood vessel endothelium formation is the primary premise for cellular and gene replacement therapy. OBJECTIVE: To construct an eukaryotic expression plasmid for cellular repressor of E1A-stimulated genes (CREG) driven by an endothelial cell specific promoter TIE2 and reported by enhanced green fluorescence protein (EGFP), and then to observe its expression in mouse artery endothelial cells in vitro. METHODS: TIE2 promoter DNA sequence was synthesized according to GenBank, and was attached with Ase I and Nhe I restriction enzyme recognition sites at both ends, respectively. The TIE2 promoter sequence was released from pUC57-TIE2 plasmid by Ase I and Nhe I digestion enzymes, and then was inserted into pEGFP-N1 plasmid to form pTIE2-EGFP-N1 recombinant plasmid. CREG gene fragment was released from pcDNA3.1 myc-His/hCREG plasmid by BamH I and EcoR I digestion enzymes, and then was subcloned into pTIE2-EGFP-N1 plasmid in order to construct pTIE2-CREG-EGFP-N1 plasmid. The recombinant pTIE2-CREG-EGFP-N1 plasmid was then transfected into mouse artery endothelial cells by liposome. The EGFP expression was observed by fluorescence microscopy and the CREG expression was detected by western blot analysis. RESULTS AND CONCLUSION: Identification of pTIE2-CREG-EGFP-N1 by enzyme digestion and sequencing analysis showed that lengths of the target genes which were inserted into the recombinant plasmid were correct, and that the recombinant plasmid was transfected into mouse artery endothelial cells successfully. The strong expression of EGFP was observed by fluorescence microscopy, and the expression of CREG protein was detected by western blot analyis. The results showed that the pTIE2-CREG-EGFP-N1 eukaryotic expression plasmid was successfully constructed and the recombinant vector provides a practical tool in investigating the function and regulation of CREG in the development of cardiovascular diseases and also in producing transgenic mice with endothelial cell specific over-expression of CREG.

Abstract:OBJECTIVE To construct a eukaryotic expression vector for expressing hepatitis B virus (HBV) recombinant HBsAg-EGFP fusion protein and obtain a stable transfected Chang Liver cell line. METHODS The coding region of HBsAg gene of HBV was amplified by PCR and was digested by BamH I/EcoR I . This fragment was inserted into pEGFPN1 with T4 ligase and transformed E-coli TG1. The positive recombinant plasmid was selected, then the recombinant plasmid was transfected into Chang Liver cell by Lipofectamine 2000 cells containing stable transformants were selected by the ability of resistance to G418 and isolated with a limited dilution. The stable transfected cell line expressing high level HBsAg-EGFP fusion protein was obtained. RESULTS The eukaryotic expression vector named pEGFPN1-HBsAg was successfully constructed and the stable transfected Chang Liver cell line expressing pEGFPN1-HBsAg fusion protein was obtained. CONCLUSION The stable transfected Chang Liver cell line could express pEGFPN1-HBsAg fusion protein, could be used to screen the proteins differentially expressed in HBsAg expression Chang Liver cells, which brought some new clues for studying the potential molecular mechanism of HBsAg protein.

Abstract:(R)-2-Hydroxy-4-phenylbutyric acid ethyl ester ((R)-HPBE) is an essential chiral intermediate in the synthesis of angiotensin-converting enzyme (ACE) inhibitors. Its production involves the highly selective asymmetric reduction of ethyl 2-oxo-4-phenylbutyrate (OPBE), catalyzed by carbonyl reductase (CpCR), with efficient cofactor regeneration playing a crucial role. In this study, an in-situ coenzyme regeneration system was developed by coupling carbonyl reductase (CpCR) with glucose dehydrogenase (GDH), resulting in the construction of five recombinant strains capable of NADPH regeneration. Among these, the recombinant strain E. coli BL21-pETDuet-1-GDH-L-CpCR, where CpCR is fused to the C-terminus of GDH, demonstrated the highest catalytic activity. This strain exhibited an enzyme activity of 69.78 U/mg and achieved a conversion rate of 98.3%, with an enantiomeric excess (ee) of 99.9% during the conversion of 30 mM OPBE to (R)-HPBE. High-density fermentation further enhanced enzyme yield, achieving an enzyme activity of 1960 U/mL in the fermentation broth, which is 16.2 times higher than the volumetric activity obtained from shake flask fermentation. Additionally, the implementation of a substrate feeding strategy enabled continuous processing, allowing the strain to efficiently convert a final OPBE concentration of 920 mM, producing 912 mM of (R)-HPBE. These findings highlight the system's improved catalytic efficiency, stability, and scalability, making it highly suitable for industrial-scale biocatalytic production.

Abstract:Background: The Chinese hamster ovary (CHO) cell line is the main host for the high-titer production of therapeutic and diagnostic proteins in the biopharmaceutical industry. In most cases, plasmids for efficient protein expression in CHO cells are based on the cytomegalovirus (CMV) promoter. The autologous Chinese hamster eukaryotic translation elongation factor 1α (EEF1A1) promoter is a viable alternative to the CMV promoter in industrial applications. The EEF1A1 promoter and its surrounding DNA regions proved to be effective at maintaining high-level and stable expression of recombinant proteins in CHO cells. EEF1A1-based plasmids' large size can lead to low transfection efficiency and hamper target gene amplification. We hypothesized that an efficient EEF1A1-based expression vector with a long terminal repeat fragment from the Epstein-Barr virus (EBVTR) could be truncated without affecting promoter strength or the long-term stability of target gene expression. Methods: We made a series of deletions in the downstream flanking region of the EEF1A1 gene, and then in its upstream flanking region. The resulting plasmids, which coded for the enhanced green fluorescent protein (eGFP), were tested for the level of eGFP expression in the populations of stably transfected CHO DG44 cells and the stability of eGFP expression in the long-term culture in the absence of selection agents. Results: It was shown that in the presence of the EBVTR fragment, the entire downstream flanking region of the EEF1A1 gene could be excluded from the plasmid vector. Shortening of the upstream flanking region of the EEF1A1 gene to a length of 2.5 kbp also had no significant effect on the level of eGFP expression or long-term stability. The EBVTR fragment significantly increased expression stability for both the CMV and EEF1A1 promoter-based plasmids, and the expression level drop during the two-month culture was more significant for both CMV promoter-based plasmids. Conclusion: Target protein expression stability for the truncated plasmid, based on the EEF1A1 gene and EBVTR fragment, is sufficient for common biopharmaceutical applications, making these plasmid vectors a viable alternative to conventional CMV promoter-based vectors.

Abstract:Butyrylcholinesterase (BChE) is an essential human cholinesterase relevant to liver conditions and neurodegenerative diseases, which makes it a pivotal biomarker of health. It therefore remains challenging and highly desired to elaborate efficient chemical tools for BChE with simple operations and satisfactory working performance. In this work, a background-free detection strategy was built by virtue of the judicious coupling of a specific BChE-enzymatic reaction and in situ cyclization. High sensitivity with a low limit of detection (LOD) of 0.075 μg/mL could be readily achieved from the blank background and the as-produced emissive indicators, and the specific reaction site contributed to the high selectivity over other bio-species even acetylcholinesterase (AChE). In addition to the multifaceted spectral experiments to verify the sensing mechanism, this work assumed comprehensive studies on the application. The bio-investigation ranged from cells to an organism, declaring a noteworthy prospect in disease diagnosis, especially for Alzheimer's disease (AD), a common neurodegenerative disease with over-expressed BChE. Abstract:Butyrylcholinesterase (Eq-BChE) from equine (horse) serum has been used extensively to demonstrate the efficacy of cholinesterase as a single pretreatment drug (bioscavenger) for organophosphate toxicity in rodents and nonhuman primates. It has the longest mean retention time in animals among all ChE preparations examined. The authors describe the complete amino acid sequence of Eq-BChE. Like other mammalian BChEs, it is a glycosylated protein composed of four identical subunits. The apparent molecular weight of the subunit containing 574 amino acids and the estimated carbohydrate content of 26% is 84,551 Da. It has eight carbohydrate-modified sites at positions 57, 106, 241, 256, 341, 455, 481, and 486 as N-linked Asn residues. The active site charged triad aminoacids are Ser198, Glu325, and His438. The N-terminal aminoacids are GluGluAspIle, consistent with this being the mature form of the enzyme. In addition, a monomeric form of this enzyme that corresponds to 4% of the total BChE activity was isolated and purified. Comparison of Eq-BChE with other species shows that there is a high degree of primary sequence identity. There is a 90.1% identity with the human serum BChE sequence, although the equine enzyme has one less N-glycosylation site and Cys residue. If conserved amino acid substitutions are included then the proteins share 93.4% homology.
Enhancing Soluble Expression of Phospholipase B for Efficient Catalytic Synthesis of L-Alpha-Glycerylphosphorylcholine

Abstract:Phospholipase B (PLB) harbors three distinct activities with broad substrate specificities and application fields. Its hydrolyzing of sn-1 and sn-2 acyl ester bonds enables it to catalyze the production of L-alpha-glycerylphosphorylcholine (L-α-GPC) from phosphatidylcholine (PC) without speed-limiting acyl migration. This work was intended to obtain high-level active PLB and apply it to establish an efficient system for L-α-GPC synthesis. PLB from Pseudomonas fluorescens was co-expressed with five different molecular chaperones, including trigger factor (Tf), GroEL-GroES (GroELS), DnaK-DnaJ-GrpE (DnaKJE), GroELS and DnaKJE, or GroELS and Tf or fused with maltose binding protein (MBP) in Escherichia coli BL21(DE3) to improve PLB expression. PLB with DnaKJE-assisted expression exhibited the highest catalytic activity. Further optimization of the expression conditions identified an optimal induction OD600 of 0.8, IPTG concentration of 0.3 mmol/L, induction time of 9 h, and temperature of 25 °C. The PLB activity reached a maximum of 524.64 ± 3.28 U/mg under optimal conditions. Subsequently, to establish an efficient PLB-catalyzed system for L-α-GPC synthesis, a series of organic-aqueous mixed systems and surfactant-supplemented aqueous systems were designed and constructed. Furthermore, the factors of temperature, reaction pH, metal ions, and substrate concentration were further systematically identified. Finally, a high yield of 90.50 ± 2.21% was obtained in a Span 60-supplemented aqueous system at 40 °C and pH 6.0 with 0.1 mmol/L of Mg2+. The proposed cost-effective PLB production and an environmentally friendly PLB-catalyzed system offer a candidate strategy for the industrial production of L-α-GPC.

Abstract:During the emergence of infectious diseases, evaluating the efficacy of newly developed vaccines requires antigen proteins. Available methods enhance antigen protein productivity; however, structural modifications may occur. Therefore, we aimed to construct a novel transient overexpression vector capable of rapidly producing large quantities of antigenic proteins in mammalian cell lines. This involved expanding beyond the exclusive use of the human cytomegalovirus (CMV) promoter, and was achieved by incorporating a transcriptional enhancer (CMV enhancer), a translational enhancer (woodchuck hepatitis virus post-transcriptional regulatory element), and a promoter based on the CMV promoter. Twenty novel transient expression vectors were constructed, with the vector containing the human elongation factor 1-alpha (EF-1a) promoter showing the highest efficiency in expressing foreign proteins. This vector exhibited an approximately 27-fold higher expression of enhanced green fluorescent protein than the control vector containing only the CMV promoter. It also expressed the highest level of severe acute respiratory syndrome coronavirus 2 receptor-binding domain protein. These observations possibly result from the simultaneous enhancement of the transcriptional activity of the CMV promoter and the human EF-1a promoter by the CMV enhancer. Additionally, the synergistic effect between the CMV and human EF-1a promoters likely contributed to the further enhancement of protein expression.

Abstract:Bacillus licheniformis 2709 is a potential cell factory for the production of alkaline protease AprE, which has important value in industrial application but still lacks sufficient production capacity. To address this problem, we investigated the effects of the secretory viscous materials on the synthesis of AprE, which might seriously affect the industrial fermentation. Furthermore, an iterative chromosomal integration strategy at various chromosomal loci was implemented to achieve stable high-level expression of AprE in B. licheniformis 2709. The host was genetically modified by disrupting the native pgs cluster controlling the biosynthesis of viscous poly-glutamic acid identified in the study by GC/MS, generating a mutant with significantly higher biomass and better bioreactor performance. We further enhanced the expression of alkaline protease by integrating two additional aprE expression cassettes into the genome, generating the integration mutant BL ∆UEP-3 with three aprE expression cassettes, whose AprE enzyme activity in shake flasks reached 25,736 ± 997 U/mL, which was 136% higher than that of the original strain, while the aprE transcription level increased 4.05 times. Thus, an AprE high-yielding strain with excellent fermentation traits was engineered, which was more suitable for bulk-production. Finally, the AprE titer was further increased in a 5-L fermenter, reaching 57,763 ± 1039 U/mL. In summary, genetic modification is an enabling technology for enhancing enzyme production by eliminating the unfavorable characteristics of the host and optimizing the expression of aprE through iterative chromosomal integration. We believe that the protocol developed in this study provides a valuable reference for chromosomal overexpression of proteins or bioactive molecules in other Bacillus species.
