Abstract:Asynchronous electrical activation, induced by bundle branch block (BBB), can cause reduced ventricular function. However, the effects of BBB on the mechanical function of heart are difficult to assess experimentally. Many heart models have been developed to investigate cardiac properties during BBB but have mainly focused on the electrophysiological properties. To date, the mechanical function of BBB has not been well investigated. Based on a three-dimensional electromechanical canine heart model, the mechanical properties of complete left and right bundle branch block (LBBB and RBBB) were simulated. The anatomical model as well as the fiber orientations of a dog heart was reconstructed from magnetic resonance imaging (MRI) and diffusion tensor MRI (DT-MRI). Using the solutions of reaction-diffusion equations and with a strategy of parallel computation, the asynchronous excitation propagation and intraventricular conduction in BBB was simulated. The mechanics of myocardial tissues were computed with time-, sarcomere length-dependent uniaxial active stress initiated at the time of depolarization. The quantification of mechanical intra- and interventricular asynchrony of BBB was then investigated using the finite-element method with an eight-node isoparametric element. The simulation results show that (1) there exists inter- and intraventricular systolic dyssynchrony during BBB; (2) RBBB may have more mechanical synchrony and better systolic function of the left ventricle (LV) than LBBB; (3) the ventricles always move toward the early-activated ventricle; and (4) the septum experiences higher stress than left and right ventricular free walls in BBB. The simulation results validate clinical and experimental recordings of heart deformation and provide regional quantitative estimates of ventricular wall strain and stress. The present work suggests that an electromechanical heart model, incorporating real geometry and fiber orientations, may be helpful for better understanding of the mechanical implications of congestive heart failure (CHF) caused by BBB.
A Research about Single-Point Ligated Ischemic Heart Failure Rabbits in Physiology and Biomechanics

Abstract:Objective Animal models are important to study basic mechanism and biomechanics of ischemic heart failure (HF). Among a four heart chambers, the left ventricular (LV) plays the most significant mechanical part of pumping blood to the entire body. Biomechanical and morphological analyses can reflect the function of LV. The single‐point ligated HF model is a typical rabbit heart failure model, which can reveal the mechanism of heart failure. Left ventricular myofiber stress and strain analyzing is an effective biomechanical method in HF study. We aim at studying the physiological and biomechanical behave of the ischemic heart failure model. Method By single‐point ligation method, we establish the ischemic heart failure model. We ligate the left circumflex coronary arteries of 6 New Zealand white rabbits. The rabbits are scanned by Computed Tomography (CT), with short‐axis slice separated by 1.25 mm, after 2‐weeks and 4‐weeks. To determine regional stress and strain distributions in rabbit ventricular myocardium, an anatomically detailed nonlinear finite element model was used, which can solve the equations of stress equilibrium during passive filling of the left ventricle. At same time, the paraffin sections have be surveyed in pathology. Result The volume can be calculated by 3‐D reconstructed model, which range from 1528.33 mm 3 (SD 286.23) at 2‐weeks to 1990.72 mm 3 (SD 178.71) at 4‐weeks. The strain and stress get bigger from 2‐weeks to 4‐weeks. The huge change can be found at the pathology sections. Conclusions This research find the changing regulation of the strain and stress. Based on biomechanics and physiology, a new method for evaluating the function of the left ventricular is presented. 3D Echo-Based Patient-Specific Computational Left Ventricle Models to Quantify Material Properties and Stress/Strain Differences between Ventricles with and without Infarct.

Abstract:Identifying ventricle material properties and its infarct area after heart attack noninvasively is of great important in clinical applications. An echo-based computational modeling approach was proposed to investigate left ventricle (LV) mechanical properties and stress conditions using patient-specific data. Echo data was acquired from one healthy volunteer (male, age: 58) and a male patient (age: 60) who had an acute inferior myocardial infarction one week before echo image acquisition. Standard echocardiograms were obtained using an ultrasound machine (E9, GE Mechanical Systems, Milwaukee, Wisconsin) with a 3V probe and data were segmented for model construction. Finite element models were constructed to obtain ventricle stress and strain conditions. A pre-shrink process was applied so that the model ventricle geometries under end-of-systole pressure matched in vivo data. Our results indicated that the modeling approach has the potential to be used to determine ventricle material properties. The equivalent Young's modulus value from the healthy LV (LV1) was about 30% softer than that of the infarct LV (LV2) at end of diastole, but was about 100% stiffer than that of LV2 at end of systole. This can be explained as LV1 has more active contraction reflected by stiffness variations. Using averaged values, at end-systole, longitudinal curvature from LV2 was 164% higher than that from LV1. LV stress from LV2 was 82% higher than that from LV1. At end-diastole, L-curvature from LV2 was still 132% higher than that from LV1, while LV stress from LV2 was only 9% higher than that from LV1. Longitudinal curvature and stress showed the largest differences between the two ventricles, with the LV with infarct having higher longitudinal curvature and stress values. Large scale studies are needed to further confirm our findings.
The Structure-Function Remodeling In Rabbit Hearts Of Myocardial Infarction

Abstract:Animal models are of importance to investigate basic mechanisms for ischemic heart failure (HF). The objective of the study was to create a rabbit model through multiple coronary artery ligations to investigate the postoperative structure-function remodeling of the left ventricle (LV) and coronary arterial trees. Here, we hypothesize that the interplay of the degenerated coronary vasculature and increased ventricle wall stress relevant to cardiac fibrosis in vicinity of myocardial infarction (MI) precipitates the incidence and progression of ischemic HF. Echocardiographic measurements showed an approximately monotonic drop of fractional shortening and ejection fraction from 40% and 73% down to 28% and 58% as well as persistent enlargement of LV cavity and slight mitral regurgitation at postoperative 12weeks. Micro-CT and histological measurements showed that coronary vascular rarefaction and cardiac fibrosis relevant to inflammation occurred concurrently in vicinity of MI at postoperative 12weeks albeit there was compensatory vascular growth at postoperative 6weeks. These findings validate the proposed rabbit model and prove the hypothesis. The post-MI rabbit model can serve as a reference to test various drugs for treatment of ischemic HF.
The interplay of cardiac dysfunctions and hemodynamic impairments during the progression of myocardial infarction in male rats.

Abstract:Myocardial infarction (MI) can induce heart failure with reduced ejection fraction (HFrEF). Systemic circulation is of importance to interact with MI-induced HFrEF. Hence, we studied the changes of myofiber stresses and peripheral hemodynamics during the progression from MI to HFrEF. MI was induced in Wistar male rats by the coronary artery ligation surgery. Physiological and hemodynamic measurements were carried out in the LV and peripheral arteries at postoperative 3 and 6 weeks, based on which LV myofiber stresses were computed and peripheral hemodynamic analysis was demonstrated. This study showed that MI significantly impaired cardiac functions and peripheral hemodynamics and altered the corresponding histological properties of cardiac wall and peripheral arterial wall, which deteriorated with time after operation. In summary, the interplay of cardiac dysfunctions and hemodynamic impairments accelerates the progression of MI-induced HF.

Abstract:Although cardiac wall mechanics is of importance for understanding heart failure with preserved ejection fraction (HFpEF), there is a lack of relevant mechanics studies. The aim of this study was to analyze the changes in stress and strain in the left ventricle (LV) in hypertension-induced HFpEF rats. Based on experimental measurements in DSS rats fed with high-salt (HS) and low-salt (LS) diets, LV stress and strain were computed throughout the cardiac cycle using Continuity software. HS-feeding increased myofiber stress and strain along both the transmural and longitudinal directions at the end-diastolic state but resulted in a lower absolute value of strain and relatively unchanged stress at the end-systolic state. Moreover, the end-diastolic stress and strain decreased with increasing radial position from the endocardial towards the epicardial walls despite negligible changes along the longitudinal direction. The changes in LV wall mechanics characterized the elevated diastolic LV stiffness and slow LV relaxation in HS-fed rats of HFpEF. These findings denote that a vicious cycle of increased stress and strain and diastolic dysfunction can prompt the development of HFpEF.
Comparative Study on the Ways of Cardiac Function Evaluation in Rat Myocardial Infarction Models

Abstract:Objective To compare the characteristics of two tools for evaluating the rat's cardiac function after myocardial infarction,and to confirm the stage when stable heart failure is established after myocardial infarction.Methods We established rat myocardial infarction models with ligature of the left coronary artery.The rats were divided into five groups,four of the groups were fed for 2,4,6,or 8 weeks after the surgery(operation group),while another group was set as a sham-operated group,and was fed for 4 weeks.After the process,the rats received intraperitoneal injection of anesthetics,and a small ultrasonography instrument was used to carry out two-dimensional,M-mode echocardiography examination and determine the mitral flow spectrum.To measure the left ventricular end-systolic pressure(LVSP) and other hemodynamic parameters,a catheter was inserted through the carotid artery into the left ventricle.Finally,the heart of the rats was resected for weighing.The cardiac function of the 5 groups were analyzed and compared. Results The cardiac function of the rat models did not change within the first 2 weeks after the ligature of the left coronary artery(P>0.05).After that,both systolic and diastolic function began to decrease.In 4 weeks after the operation,both the the maximum rate of left ventricular isovolumic systolic pressure + dp/dtmax and the maximum rate of left ventricular isovolumic diastolic pressure-dp/dtmax decreased to the lowest levels [(1249.89 ± 95.82) mm Hg/s and(-1316.40 ± 58.31) mm Hg/s,respectively];and then in 6 weeks after the operation,echocardiography showed that the left ventricular short axis fractional shortening FS reached the lowest level [(18.70 ± 3.83)%].Moreover,we found that the FS was highly related with the + dp/dtmax(r=0.864,P<0.001).The hemodynamic examination showed that the diastolic function decreased 2 weeks after the surgery,while echocardiography showed no difference at the time.Conclusions The first 2 weeks after the ligature of the left coronary artery was the early stage of heart failure in the rat models.After this period,either the systolic or the diastolic function begins to decrease.Both echocardiography and hemodynamics can be used to measure the cardiac systolic function of rat,whereas the latter is a better choice for measuring diastolic function.

Abstract:Objective To identify the different proteins expressed in the left ventricular hypertrophy(LVH) induced by abdominal aortic constriction(AAC) rats using proteomic approach and analyze the functions of these various proteins associated with the mechanism of hypertensive hypertrophy.Methods AAC and sham operation were performed on 8 week old male Sprague Dawley rats.Echocardiography,hemodynamie,cardiac morphologi- cal changes were investigated 9 weeks after operation.Differential proteins between AAC and sham operation rats were separated by 2DE and identified by MODI-TOF-MS.Changed protein were further verified by West- ern-blot.Results ACC rats had significant higher,HR,SBP,DBP,LVESP,dP/dt,IVSd,PWTd,HM, HM/BM in relative to sham operated rats(P0.05).Histological examination revealed that myocardial cells were disarranged and cell nuclei were enlarged,malformed and densely stained.Twenty-one differential proteins in myocardium were identified with 14 proteins up-regulation,4 proteins down-regulation,and 3 proteins were newly found.Myosin light polypeptide,cardiac muscle a actin 1 proprotein,β-myosin heavy chain,mitochondri- al ribosomal protein L15,G protein-coupled receptor 34,tyrosine-protein kinase ZAP-70,aspartyl-and tRNA synthetase were up-regulated while ATP synthase,L 3-hydroxyacyl-Coenzyme A dehydrogenase,3-hydroxybu- tyrate dehydrogenase were down-regulated.Conclusion Various proteins involved in glycolysis,cell signaling and communication,gene expression and regulation,myocardial remodeling were increased left ventricular hyper- trophy.

Abstract:Objective To explore the biomechanical mechanism development of left ventricular wall in the patients with myocardial infarction using wall stress and strain.Methods Quantitative tissue velocity imaging was applied for obtaining 5 segments of left ventricle(LV)anterior wal long axis peak systolic strain(PSS)in 30 cases with myocardial infarction(MI)and 30 normal subjects.LV wall meridional peak systolic stress(Pσm)was calculating by brachial blood pressure.Results ① The PSS of segments in LV anterior wall was decreased in MI group,compared with the corresponding segments,and the PSS were lower in the patients with myocardial infarction(P<0.05).② Pσm was higher in the patients with myocardial infarction(16.64±0.52)than that in the control group(20.68±0.95),and there was significant difference between groups(P<0.05).Conclusion The formation of wall aneurysm is correlated with wall stress and strain.
Evaluation of Left Ventricular Diastolic Dysfunction at the Early Stage of Pressure Overload-Induced Myocardial Remodeling in Rats

Abstract:AIM: To investigate the evaluation method of left ventricular diastolic function and myocardial hypertrophy induced by pressure overload in rats.METHODS: Male Sprague-Dawley rats were subject to left ventricular pressure overload by transverse aortic constriction(TAC).Cardiac structure and diastolic function were evaluated by echocardiography,hemodynamic analysis and examination of hydroxyproline concentration in the myocardial tissues.RESULTS: Compared with the sham-operated controls,left ventricular wall dimension in diastole significantly increased in the rats 3 weeks after TAC [anterior wall:(1.71±0.11) mm vs(1.43±0.06) mm,P0.01;posterior wall:(1.78±0.03) mm vs(1.44±0.02) mm,P0.01].Left ventricular early diastolic posterior wall motion velocity(E′) significantly decreased in the rats 3 weeks after TAC[(52.1±0.8) mm·s-1 vs(63.8±1.5) mm·s1,P0.01],and was much lower than that in the rats 6 weeks after TAC.Left ventricular mass to tibia length in TAC rats was much higher than that in sham-operated controls [3 weeks after TAC:(19.6±1.1) mg·mm-1 vs(14.8±0.6) mg·mm-1,P0.01;6 weeks after TAC:(23.2±0.9) mg·mm-1 vs(15.6±0.7) mg·mm-1,P0.01].The ratio of maximum rate of degression of left ventricular pressure(dp/dtmin) to left ventricular systolic pressure(dp/dtmin/LVSP) started to decrease in TAC rats in the 3rd week(48.9±5.9 vs 63.5±9.9) and significantly decreased in TAC rats in the 6th week as compared with sham-operated controls(35.4±4.0 vs 54.4±2.9,P0.01).Sirius red-stained collagen in cardiac interstitium,especially around the blood vessels,was increased in TAC rats.Six weeks after TAC,a significant increase in the content of myocardial hydroxyproline[(465.3±17.3) μg vs(351.4±8.2) μg,P0.05] was observed.CONCLUSION: The early diastolic posterior wall motion velocity(E′) detected by tissue Doppler imaging is a sensitive indicator of diastolic dysfunction at the early stage of myocardial remodeling induced by pressure overload in rats.
Development and Evaluation for Volume Overload- Induced and Pressure Overload- Induced Cardiac Hypertrophy in Rats

Abstract:AIM: To compare the evaluations for the structure and function of the hypertrophic hearts induced by volume overload or pressure overload in rats. METHODS: Volume overload-induced cardiac hypertrophy was established by abdominal aortacaval fistula (ACF) and pressure overload-induced cardiac hypertrophy was developed by constriction of aorta (CA). The cardiac structure and function were analyzed by echocardiography, hemodynamic determination, heart weight measurement and histological examination. RESULTS: Heart weight of rats in all the operated groups was increased compared to the sham-operated groups. In 1-week ACF group, the internal diameter (0.67±0.03)cm vs (0.60±0.02)cm, P0.01 and volume of left ventricle increased (0.69±0.10)mL vs (0.50±0.04)mL, P0.01,relative wall thickness (RWT) decreased (0.46±0.05 vs 0.55±0.05, P0.01), compared with the sham-operated group. In 1-week CA group, interventricular septal thickness (0.20±0.03)cm vs (0.16±0.02)cm, P0.05, left ventricular posterior wall thickness (0.20±0.03)cm vs (0.16±0.02)cm, P0.01, RWT (0.71±0.17 vs 0.56±0.12, P0.05) and +dp/dt_ max ( 4 886 ± 1 304 vs 3 674 ±325, P0.05) were all increased compared with the sham-operated group. In 2-week-groups, these parameters changed more significiantly. CONCLUSION: Cardiac structure and function could be evaluated by echocardiography and hemodynamic determination. RWT is a sensitive index for the cardiac hypertrophy induced by both volume overload and pressure overload.
The Physiological and Biomechanical Study about the Passive Diastolic Modelling of the Heart Failure Rabbits Causing by Aortic Arch Banding

Abstract:Objective Cardiovascular disease (CVD) is a major health problem world‐wide and can lead to congestive heart failure (HF). Aortic arch banding is a classic experimental scheme to establish the congestive HF model, when hypertension and cardiac hypertrophy occur to the rabbits. Left ventricular myofiber stress and strain analyzing is an effective biomechanical method to study congestive HF. Our target is to study the physiological and biomechanical behave of the congestive heart failure model. Method By aortic arch banding, we establish the heart failure model. 6 New Zealand white rabbits have been banded 30% at aortic arch. The rabbits are scanned by Computed Tomography after 2‐weeks and 4‐weeks separately. We developed a three‐dimensional finite element model, which can describe the rabbit ventricular geometry and fiber architecture in detail. At same time, the paraffin sections have be surveyed in pathology. Result The volumes of the hearts can be calculated by 3‐D reconstructed model, which range from 2338.99 mm 3 (SD 84.06) at 2‐weeks to 3339.07 mm 3 (SD 701.91) at 4‐weeks. The strain and stress get lower from 2‐weeks to 4‐weeks. A huge change can be found in pathology sections. Conclusions We find the changing pattern of the strain and stress. The law of heart failure in physiology and biomechanics were discussed. Passive mechanical properties of the left ventricular myocardium and extracellular matrix in hearts with chronic volume overload from mitral regurgitation

Abstract:Cardiac volume overload from mitral regurgitation (MR) is a trigger for left ventricular dilatation, remodeling, and ultimate failure. While the functional and structural adaptations to this overload are known, the adaptation of myocardial mechanical properties remains unknown. Using a rodent model of MR, in this study, we discern changes in the passive material properties of the intact and decellularized myocardium. Eighty Sprague-Dawley rats (350-400 g) were assigned to two groups: (1) MR (n = 40) and (2) control (n = 40). MR was induced in the beating heart by perforating the mitral leaflet with a 23G needle, and rats were terminated at 2, 10, 20, or 40 weeks (n = 10/time-point). Echocardiography was performed at baseline and termination, and explanted hearts were used for equibiaxial mechanical testing of the intact myocardium and after decellularization. Two weeks after inducing severe MR, the myocardium was more extensible compared to control, however, stiffness and extensibility of the extracellular matrix did not differ from control at this timepoint. By 20 weeks, the myocardium was stiffer with a higher elastic modulus of 1920 ± 246 kPa, and a parallel rise in extracellular matrix stiffness. Despite some matrix stiffening, it only contributed to 31% and 36% of the elastic modulus of the intact tissue in the circumferential and longitudinal directions. At 40 weeks, similar trends of increasing stiffness were observed, but the contribution of extracellular matrix remained relatively low. Chronic MR induces ventricular myocardial stiffening, which seems to be driven by the myocyte compartment of the muscle, and not the extracellular matrix.

Abstract:The assessment by speckle tracking echocardiography (STE) provides useful information on regional and global left ventricular (LV) functions. The aim of the study is to investigate if STE-based strain analysis could detect the difference of pressure overload-induced myocardial remodelling between young and adult rats. Physiological, haemodynamic, histological measurements were performed post-operatively in young and adult rats with transverse aortic constriction (TAC) as well as the age-matched shams. Two-way ANOVA was used to detect the statistical difference of various measured parameters. Pressure overload decreased the ejection fraction, fractional shortening, dp/dt(max) and |dp/dtmin|, but increased the LV end-diastolic (ED) pressure in adult rat hearts for nine weeks after TAC operation than those in young rat hearts. Pressure overload also resulted in different changes of peak strain and strain rate in the free wall, but similar changes in the interventricular septum of young and adult rat hearts. The changes in myocardial remodelling were confirmed by the histological analysis including the increased apoptosis rate of myocytes and collagen area ratio in the free wall of adult rat hearts of LV hypertrophy when compared with the young. Pressure overload alters myocardial components in different degrees between young and adult animals. STE-based strain analysis could detect the subtle difference of pressure overload-induced myocardial remodelling between young and adult rats.

Abstract:Objective: To investigate the stiffness change of hypertension-associated hypertrophied cardiomyocytes and the arrhythmogenic mechanism at micro scales.Methods:(1) Separating respectively ventricular myocytes from spontaneously hypertensive rats(SHR) and wistar-Kyoto rats(WKY) at different ages;(2) collecting force distance curves with atomic force microscope;(3) comparing the Young's moduli.Results: For 12-week-old and 21-week-old SHR,the Young's moduli of ventricular myocytes were significantly larger than that from WKY of the same age(4.61 kPa ± 2.08 kPa vs.3.55 kPa ± 1.35 kPa,5.95 kPa ±1.98 kPa vs.3.79 kPa ± 1.66 kPa,P 0.01).Comparing the Young's moduli of ventricular myocytes from SHR of 21-week-old and12-week-old,the former were higher than the latter(5.95 kPa ± 1.98 kPa vs.4.61 kPa ± 2.08 kPa,P 0.01).However,the Young's moduli of ventricular myocytes had not visible difference between WKY of 21-week-old and 12-week-old(3.79 kPa ± 1.66kPa vs.3.55 kPa ± 1.35 kPa,P 0.05).Conclusions: The intrinsic mechanical property of hypertension-associated hypertrophied ventricular myocytes has changed.Young's moduli increased,in another word,hypertrophied ventricular myocytes were stiffer.In the meanwhile,as the degree of cardiac hypertrophy deteriorated gradually,the stiffness of ventricular myocytes increased accordingly.These changes were probably due to the occurrence and maintenance of cardiac arrhythmias.
Identification of Early Cardiac Dysfunction and Heterogeneity after Pressure and Volume Overload in Mice by High-Frequency Echocardiographic Strain Imaging

Abstract:ObjectAortic stenosis and regurgitation are clinically important conditions characterized with different hypertrophic types induced by pressure or volume overload, respectively, but with comparable cardiac function in compensated stage. Speckle-tracking based strain imaging has been applied to assess subtle alterations in cardiac abnormality, but its application in differentiating these two types of ventricular hypertrophy is still sparse. Here, we performed strain imaging analysis of cardiac remodeling in these two loading conditions.MethodsC57BL/6J mice were subjected to transverse aortic constriction (TAC)-induced pressure overload or aortic regurgitation (AR)-induced volume overload. Conventional echocardiography and strain imaging were comprehensively assessed to detect stimulus-specific alterations in TAC and AR hearts.ResultsConventional echocardiography did not detect significant changes in left ventricular systolic (ejection fraction and fractional shortening) and diastolic (E/E’) function in either TAC or AR mice. On the contrary, global strain analysis revealed global longitudinal strain and strain rate were remarkably impaired in TAC while preserved in AR mice, although global radial, and circumferential strain and strain rate were significantly reduced in both models. Regional strain analysis in the long axis demonstrated that longitudinal strain and strain rate in all or most segments were decreased in TAC but maintained or slightly dented in AR mice, while radial strain and strain rate indicated overt decline in both models. Moreover, decreased radial and circumferential strain and strain rate were observed in most segments of TAC and AR mice in the short axis.ConclusionStrain imaging is superior to conventional echocardiography to detect subtle changes in myocardial deformation, with longitudinal strain and strain rate indicating distinct functional changes in pressure versus volume overload myocardial hypertrophy, making it potentially an advanced approach for early detection and differential diagnosis of cardiac dysfunction.
Mitral regurgitation increases systolic strains in remote zone and worsens left ventricular dyssynchrony in a swine model of ischemic cardiomyopathy

Abstract:Background: Ischemic mitral regurgitation (IMR) imposes volume overload on the left ventricle (LV), accelerating adverse LV remodeling. In this study, we sought to investigate the impact of volume overload due to IMR on regional myocardial contractile mechanics. Methods: Ten Yorkshire swine were induced with myocardial infarction (MI) by occluding the left circumflex coronary artery (LCx). Cardiac MRI was performed at baseline (BL) and 2.5 months (2.5M) post-MI. IMR was quantified with epicardial echocardiography 3 months post-MI. The animals were then assigned to 2 groups: no/mild MR (nmMR, n = 4) and moderate/severe MR (msMR, n = 6). MRI images were analyzed to assess infarction size, end-diastolic and end-systolic volume (EDV and ESV, respectively), ejection fraction (EF), longitudinal strain (LS), circumferential strain (CS), and systolic dyssynchrony index (SDI). The myocardial region was divided into infarction, border, and remote zones based on the LCx-supplied region. Results: There was no difference in the infarction size. Group-wise comparison of LS and CS between BL and 2.5M demonstrated that LS and CS in the infarction zone and the border zone decreased at 2.5M in both groups. However, LS and CS in the remote zone were elevated only in the msMR group (LS: −9.81 ± 3.96 vs. −12.58 ± 5.07, p < 0.01; CS; −12.78 ± 3.81 vs. −16.09 ± 3.33, p < 0.01) at 2.5M compared to BL. The SDI of CS was significantly elevated in the msMR group (0.1255 vs. 0.0974, p = 0.015) at 2.5M compared to BL. Conclusions: Elevated LS and CS in the remote zone were observed in moderate/severe MR and ventricular dyssynchrony. These elevated cardiac strains, coupled with ventricular dyssynchrony, may contribute to the progression of MR, thereby accelerating heart failure.

Abstract:Introduction: Surgically induced, combined volume and pressure overload has been used in rabbits to create a simplified and reproducible model of acute left ventricular (LV) failure. Materials and methods: New Zealand white male rabbits (n = 24, mean weight 3.1 ± 0.2 kg) were randomly assigned to either the Control group (n = 10) or to the Heart Failure group (HF, n = 14). Animals in the Control group underwent "sham" procedures. Animals in the HF group underwent procedures to induce LV volume overload by inducing severe aortic valve regurgitation with aortic cusp disruption and pressure overload using an occlusive silver clip positioned around the pre-renal abdominal aorta. Results: Following Procedure-1 (volume overload) echocardiography confirmed severe aortic regurgitation in all animals in the HF group, with increased mean pulse pressure difference from 18 ± 3 to 38 ± 3 mmHg (P < 0.0001). After Procedure-2 (pressure overload) all animals in the HF group showed clinical and echocardiographic signs of constriction of the abdominal aorta and echocardiography confirmed progressively declining LV function. At the end of the protocol there was a significant increase of the heart/body weight ratio in the HF group vs. Control group (4.6 ± 0.2 vs. 2.9 ± 0.1 g/kg, P < 0.05), and echocardiography showed in HF group significant increase of the LV end-diastolic diameter (2.15 ± 0.09 vs. 1.49 ± 0.03 cm, P < 0.001) and reduction of the LV shortening fraction (26.3 ± 3.8 vs. 41.3 ± 1.6%, P < 0.001). Conclusion: This experimental model: (a) consistently produces LV hypertrophy/dilatation and subsequent congestive heart failure, (b) provides new data on the time course of LV dilatation, hypertrophy and failure, (c) allows study of the progress and evolution of LV systolic and diastolic dysfunction in the presence of induced LV failure, (d) is suitable to study intervention or pharmacological administration to reduce the negative effects of acute LV failure.
Fine‐scale population structure in a desert amphibian: landscape genetics of the black toad (Bufo exsul)

Abstract:Environmental variables can strongly influence a variety of intra‐ and inter‐population processes, including demography, population structure and gene flow. When environmental conditions are particularly harsh for a certain species, investigating these effects is important to understanding how populations persist under difficult conditions. Furthermore, species inhabiting challenging environments present excellent opportunities to examine the effects of complex landscapes on population processes because these effects will often be more pronounced. In this study, I use 16 microsatellite loci to examine population structure, gene flow and demographic history in the black toad, Bufo exsul, which has one of the most restricted natural ranges of any amphibian. Bufo exsul inhabits four springs in the Deep Springs Valley high desert basin and has never been observed more than several meters from any source of water. My results reveal limited gene flow and moderately high levels of population structure (FST = 0.051–0.063) between all but the two closest springs. I found that the geographic distance across the arid scrub habitat between springs is significantly correlated with genetic structure when distance accounts for topography and barriers to dispersal. I also found very low effective population sizes (Ne = 7–30) and substantial evidence for historical population bottlenecks in all four populations. Together, these results suggest that the desert landscape and B. exsul’s high habitat specificity contribute significantly to population structure and demography in this species and emphasize the importance of considering behavioural and landscape data in conservation genetic studies of natural systems.
Myocardial Infarction and Cardiac Remodelling in Mice

Abstract:We established a mouse model of cardiac dysfunction due to myocardial infarction (MI). For this we ligated the left anterior descending coronary artery in male C57BL/6J mice and assessed healing and left ventricular (LV) remodelling at 1, 2 and 4 days and 1, 2 and 4 weeks after MI. Echocardiography was performed at 1 and 2 weeks and 1, 2, 4 and 6 months after MI. We found that neutrophil infiltration of the infarct border was noticeable at 1-2 days. Marked macrophage infiltration occurred at day 4, while lymphocyte infiltration was apparent at 7-14 days. Massive proliferation of fibroblasts and collagen accumulation began by day 7-14, and scar formation was completed by day 21. LV diastolic dimension increased markedly at 2 weeks and remained at the same level thereafter. LV shortening fraction decreased significantly at 2 weeks and then slowly decreased. In non-infarcted areas of the LV, myocyte cross-sectional area and interstitial collagen fraction increased progressively, reaching a maximum at 4 months. This study provides important qualitative and quantitative information about the natural history of cardiac remodelling after MI in mice.
