Abstract:Despite its functional significance in mammals and birds, the biological role of gonadotropin-inhibitory hormone (GnIH) in reproduction is still far from being fully understood in teleosts. In the current study, we have identified LPXRFa, the piscine ortholog of GnIH, and its cognate receptor (LPXRFa-R) in yellowtail kingfish (YTK), which is considered as a promising species for aquaculture industry worldwide. The YTK cDNA sequence of lpxrfa was 534 base pair (bp) in length and encoded a 178-amino acids (aa) preprohormone. The LPXRFa precursor comprised three putative peptide sequences that included –MPMRF, –MPQRF, or -LPERL motifs at the C-termini, respectively. The YTK lpxrfa-r cDNA sequence was composed of 1265 bp that gave rise to a LPXRFa-R of 420 aa, encompassing the characteristic seven hydrophobic transmembrane domains. In males, both lpxrfa and lpxrfa-r transcripts could be detected at high levels in the brain and testis. In females, a noteworthy expression of lpxrfa was observed in the brain and ovary, while the expression of lpxrfa-r was especially evident only in the brain. To study the ontogeny of LPXRFa system, transcript levels were also investigated during early life stages. Variable expression of the LPXRFa system was observed during all stages of YTK embryogenesis. The highest expression of lpxrfa and lpxrfa-r were noticed at 7 dph and 15 dph, respectively. Furthermore, LPXRFa peptides stimulated growth hormone (gh), luteinizing hormone (lhβ) and follicle-stimulating hormone (fshβ) gene expression from the pituitary. Taken together, our results provide initial evidence for the existence of the LPXRFa system in yellowtail kingfish and suggest its possible involvement at early development and reproductive functions.

Molecular identification, tissue distribution and in vitro functional analysis of growth hormone and its receptors in red-spotted grouper (Epinephelus akaara)

Abstract:Red-spotted grouper (Epinephelus akaara) is one of the high economic value grouper species, however, the knowledge regarding its growth is limited. In this study, full-length cDNAs of growth hormone (gh) and its receptors (ghr1 and ghr2) were cloned from the pituitary and liver of red-spotted grouper, respectively. Tissue distribution analysis showed that gh mRNA was predominantly expressed in the pituitary. ghr1 mRNA was highly expressed in the liver, muscle, fat and gonad, while ghr2 mRNA expression was ubiquitously high in the peripheral tissues. However, the mRNA expression of both ghr isoforms was relatively low in the central nervous system. Secretory recombinant grouper GH (rgGH) was expressed in yeast Pichia pastoris and verified. HEK293T cells transiently transfected with the GHR isoforms were used to elucidate the receptor-mediated signaling pathways related to growth regulation. rgGH activated rapid phosphorylation of Janus kinase 2, signal transducer and activator of transcription 5 (STAT5) and extracellular signal-regulated protein kinase 1/2 through GHR1, but only STAT5 was phosphorylated via GHR2. rgGH strongly activated STAT5 phosphorylation and significantly stimulated ghr1, ghr2 and insulin-like growth factor (igf1, igf2) mRNA expression in primary cultured hepatocytes. Data showed that the recombinant protein rgGH played effects on igf1/2 mRNA expression via GHR-mediated signaling pathways. Our findings provide essential information about GH and GHRs characteristics in red-spotted grouper.
Identification and expression analysis of two growth hormone receptors in zanzibar tilapia (Oreochromis hornorum)

Abstract:Growth hormone plays important roles in various physiological processes such as growth, metabolism, and reproduction. In this study, two cDNAs encoding growth hormone receptor (GHR) were isolated from the liver of zanzibar tilapia (Oreochromis hornornum). The two cDNAs were 2,831 and 2,044 bp in length and named GHR1 and GHR2, respectively. GHR1 and GHR2 shared 57.4% similarity in nucleotide sequences and 33.5% similarity in deduced amino acid sequences. Consequently, it was presumed that they were two different genes. Conserved regions of GHR1 and GHR2 in zanzibar tilapia were different from those of other vertebrates. For example, conserved box2 regions of GHR1 and GHR2 in zanzibar tilapia were, respectively, WVELM and WVEFT, while it was WVEFI for GHRs in other vertebrates. Similar to other fish species, GHR1 and GHR2 were expressed in brain, gill, liver, muscle, spleen, gonad, stomach, kidney, and pituitary in zanzibar tilapia. The expression levels were the highest in liver. Unlike fathead minnow (Pimephales promelas) and mossambique tilapia (O. mossambicus), the expression levels of GHR1 in most female fish tissues were higher than those in male fish. No significant difference in GHR2 expression was found in all the tissues in male and female of zanzibar tilapia. Under fasting condition, the expressions of GHRs and IGF-II were significantly up-regulated (P < 0.05) in liver, while the expression of IGF-I remained stable. This observation would contribute to understanding the evolution of the GHR family in further investigation of growth regulation of zanzibar tilapia.

Abstract:The cDNAs encoding two distinct growth hormone receptors (GHRs) were firstly cloned and sequenced from the liver of orange-spotted grouper (Epinephelus coioides). The cDNA of grouper GHR1 consisted of 2673-bp and encoded 658 amino acids, while the cDNA of grouper GHR2 consisted of 2989-bp and encoded 577 amino acids. The two cDNAs shared 78.6% identity in nucleotide sequence and 37.8% identity in deduced amino acid sequence. Northern blot analysis demonstrated a single GHR1 transcript of approximately 4.3 kb in liver and a single GHR2 transcript of approximately 3.9 kb in the liver and muscle. In the Real-time PCR assay, grouper GHR1 and GHR2 were expressed in all tissues tested. The expression of GHR2 was significantly higher than that of GHR1 in telencephalon, cerebellum, pituitary, heart and white muscle, whereas the expression of GHR1 was significantly higher in liver. These results indicated that there were two types of GHRs existing in orange-spotted grouper, and they had different structural features and tissues expression patterns. However, the functional differences between GHR1 and GHR2 in orange-spotted grouper remained further research.

Abstract:Growth hormone is an essential hormone that plays essential roles in growth, metabolism, cellular differentiation, immunity and reproduction in fish, by means of the growth hormone receptors. The encoding cDNA growth hormone receptors (GHR1 and GHR2) were cloned and characterized from Hybrid grouper (Epinephelus fuscoguttatus♀ × Epinephelus polyphekadion♂). Sequence analysis of the cloned GHR1 was observed as containing 2176, which comprised an ORF of 1842 bp, 5 UTR of 6 bp and 3 UTR of 328 bp, with 612 amino acids encoding proteins, while GHR2 was observed as containing 1824 bp that encompassed an ORF of 708 bp, 5 UTR of 48 bp and 3 UTR of 1068 bp with 235 amino acids encoding proteins. Relative mRNA expression of GHR1 and GHR2 in the liver and muscle was found to be highest respectively. Our findings provide vital statistics of GHRs likely to play a significant role in the growth of the fish.
Intragenic and Intergenic Sequences Regulating the Expression of the 5’-to-5’ Linked Adult Α-and Β-Globin Genes from Large Yellow Croaker Pseudosciaena Crocea

Abstract:One adult α-globin gene and one β-globin gene have been cloned from the large yellow croaker Pseudosciaena crocea. Linkage analysis indicated that the α-and β-globin genes were oriented head-to-head relative to each other. To identify the regulatory elements present in the intergenic and intragenic regions of the globin complex,the intergenic region alone or together with the β-globin gene first intron was cloned into the luciferase-reporter vector pGL3-Basic respectively,and the chimeric constructs were transiently transfected into Vero cells and primary fish erythrocytes. The intergenic region cannot support the high-level expression of luciferase. However,the promoter activity of the intergenic region was strongly stimulated by the positive regulatory elements （PRE） located in the β-globin gene intron 1. Thus,it is proposed that the intergenic promoters and intragenic PRE were necessary for the effective expression of the linked α-and β-globin genes.
Sequence and Structure of Encoding Eel Growth Hormone Gene

Abstract:The analysis of the sequence shows that the growth hormone gene (GH gene) of Anguilla anguilla contains 2393 bp from ATG to TAG, and consists of 4 extrons and 3 introns, also it was deduced to encode 209 amino acids. The first intron consists of 3 repeated monoploid forming its minisatellite sequence. Alignment analysis indicated that the GH gene can be divided into 5 structure domains. GD1 - GD4 domains may be relative to the GH activity and the specially binding field of the receptor, and GD5 domain to molecular structure and stability of the GH respectively. Evolution study demonstrated that there wereⅠ, Ⅱ and Ⅲgroups existing among the fishy GH genes, and the Anguilla anguilla GH belongs to group Ⅰ; group Ⅱ can be redivided to 2 subgroups which were 4 introns or 5 introns, and group 3Ⅲ has 5 introns only. It was presumed that the dismutation of the GH gene had happened through losing or adding introns during the evolution.
The co-existence of two growth hormone receptors in teleost fish and their differential signal transduction, tissue distribution and hormonal regulation of expression in seabream

Abstract:Two genomic contigs of putative growth hormone receptors (GHRs) were identified in fugu and zebrafish genomes by in silico analysis, suggesting the presence of two GHR subtypes in a single teleost species. We have tested this hypothesis by cloning the full-length cDNA sequence of a second GHR subtype from the black seabream in which the first GHR subtype had been previously reported by us. In addition, we had also cloned the sequences of both GHR subtypes from two other fish species, namely the Southern catfish and the Nile tilapia. Phylogenetic analysis of known GHR sequences from various vertebrates revealed that fish GHRs cluster into two distinct clades, viz. GHR1 and GHR2. One clade (GHR1), containing 6 to 7 extracellular cysteine residues, is structurally more akin to the non-teleost GHRs. The other clade (GHR2), containing only 4 to 5 extracellular cysteine residues, is unique to teleosts and is structurally more divergent from the non-teleost GHRs. In addition, we had examined the biological activities of both GHR subtypes from seabream using a number of reporter transcription assays in cultured eukaryotic cells and demonstrated that both of them were able to activate the Spi 2.1 and beta-casein promoters upon receptor stimulation in a ligand specific manner. In contrast, only GHR1 but not GHR2 in seabream could trigger the c-fos promoter activity, indicating that the two GHR subtypes possess some differences in their signal transduction mechanisms. Also, the expression of GHR2 is significantly higher than GHR1 in many tissues of the seabream including the gonad, kidney, muscle, pituitary and spleen. In vivo hormone treatment data indicated that cortisol upregulated hepatic GHR1 expression in seabream but not GHR2, whereas testosterone decreased hepatic GHR2 expression but not GHR1. On the other hand, hepatic expression of both GHR1 and GHR2 in seabream was decreased by estradiol treatment.

Abstract:The role of thyroid hormone (TH) and its receptors (TRs) in the regulation of body growth and muscle accretion is well established in mammals and birds, whereas the involvement of THs and TRs in fish growth, especially during the muscle accretion period of juvenile-adult transition, is unknown. This study describes the cloning of the partial cDNA sequences of TRα and TRβ in large yellow croaker, Pseudosciaena crocea (Richardson) and the patterns of TRα and TRβ mRNA expression in liver and muscle of 1- and 2-year-old large yellow croaker, associated with changes in body mass and muscle characteristics. Two TRα isoforms (TRα1, TRα2) and TRβ were identified in large yellow croaker. The deduced amino acid sequences showed high homology to the TRs of human and other teleosts. Hepatic TRβ mRNA expression was markedly lower in 2-year-old large yellow croaker compared with the 1-year-old, while no significant age difference was observed for hepatic TRα mRNA expression. Muscle expression of TRα mRNA was significantly higher in 2-year-old large yellow croaker, whereas TRβ exhibited no significant age difference. Meanwhile, serum concentration of T4 was significantly decreased in 2-year-old large yellow croaker, but no change was observed for T3. The body mass, fork length and body height of 2-year-old large yellow croaker were 4.7, 1.6 and 1.7 times greater, respectively compared with that of 1-year-old. Average diameters of skeletal muscle in 2-year-old large yellow croaker were remarkably larger than that in 1-year-old with no significant difference in muscle crude fat content. The down-regulation of hepatic TRβ expression was associated with the decrease in general growth rate and the increase in muscle expression of TRα was accompanied with muscle accretion and myofiber hypertrophy, implicating the different roles of TRs in the regulation of growth in large yellow croaker.

Cloning and Sequencing of the Growth Hormone Gene of Large Yellow Croaker and Its Phylogenetic Significance

Abstract:Using conserved primers and the PCR reaction, the growth hormone (GH) gene and the 3'-UTR of the large yellow croaker (Pseudosciaena crocea) were amplified and sequenced. The gene structure was analyzed and compared to the GH genes of 5 other percoid fish downloaded from Genbank. Also the GH gene of the large yellow croaker and the genes from 14 Percoidei and 2 Labroidei species were aligned using Clustal X. A matrix of 564 bp was used to construct the phylogenetic tree using maximum parsimony and neighbor-joining methods. Phylogenetic trees by the two methods are identical in most of the clades with high bootstrap support. The results are also identical to those from morphological data. In general, this analysis does not support the monophyly of the families Centropomidae and Carangidae. But our GH gene tree indicates that the representative species of the families Sparidae and Sciaenidae are a monophyletic group.
Molecular Cloning and Sequence Analysis of the Growth Hormone cDNAs from Seven Pleuronectiformes Fishes

Abstract:The growth hormone(GH) gene has been cloned from a number of fishes and was used to clarify phylogenetic relationships.The mRNA of Pituitary glands from seven important economic Pleuronectiformes Fishes was extracted,the cDNA sequences of GH gene were isolated from Cleisthenes herzensteini,Limanda yokohamae,Pleuronichthys Cornutus,Cynoglossus robustus,Platichthys bicoloratus,Zebrias zebra and Paralichthys dentatus with RT-PCR method by using designed specific primers based on the reported GH cDNA sequences of Pleuronectiformes.Alignment and homologous analysis of All 7 GH cDNA sequences was performed with other 8 Pleuronectiformes fishes and 7 outgroup species.The result of sequencing showed the lengths of the above cDNAs as follows: 479,564,519,440,564,440 and 522 bp,and the GH cDNA sequences encoding mature polypeptide of about 140 to 170 amino acid.The molecular weight and isoelectric point of GH protein from Platichthys bicoloratus,Limanda yokohamae,Pleuronichthys Cornutus and Paralichthys dentatus were calculated with software DNASTAR 7.0.After alignment of 7 GH with other Pleuronectiformes fishes,the 7 GH amino acid sequences all showed high homology to other known Pleuronectiformes fishes,especially to the sequences within the same family.Then the phylogenetic analysis on the GH for 15 species of Pleuronectiformes and 7 outgroup species was performed by PAUP software with the maximum parsimony method.The result was consistent with morphology of Pleuronectiformes on the whole.In the topology founded,Scophthalmus maximus and Solea senegalensis each formed an independent branch and showed far relationship with species of Bothidae and Pleuronectidae.The phlogenic position of Solea senegalensis is in accord with the result of analysis made by mitochondrial genome.It seems reasonable to speculate that GHs can be effectively used in analyzing genetic relationship and taxonomic status of Pleuronectiformes and other species of teleostean.

Abstract:Growth hormone (GH) is a member of the GH/Prolactin (PRL) protein super family. It is secreted by anterior pituitary and critically involved in the regulation of cell proliferation, differentiation and migration. Using RT-PCR (re-verse-transcription PCR), our previous study revealed that both gh and its receptor gene ghra (growth hormone receptor a) were maternally expressed in zebrafish. This suggested that the GH/GHR signaling pathway might play an important role at the early stage of zebrafish development. To further study the function of GH/PRL signaling pathway in zebrafish embryogenesis, here we characterized and compared the expression patterns of gh/prl and their receptor family members in early embryonic development using real-time PCR and in situ hybridization. We found that the ligand family mem-bers, gh and somatolactin (smtl), and the receptor gene family members, ghra and ghrb, were maternally expressed in zebrafish. The analysis of spi2.1 promoter activity indicated the cross-interaction between various ligand family mem-bers and GHRa in the early embryos of zebrafish. Our results demonstrated the important role of the GH/PRL family signaling pathway in the early development of zebrafish.
Isolation, characterization, and distribution of two cDNAs encoding for growth hormone receptor in rainbow trout (Oncorhynchus mykiss)

Abstract:Growth hormone (GH) plays important roles in a vast array of physiological processes, including growth, metabolism, and reproduction. In this study, cDNAs for two unique growth hormone receptor variants were cloned and sequenced from rainbow trout. The two cDNAs, one consisting of 2920 bp and the other of 2820 bp, share 87.2% identity in nucleotide sequence and 85.5% identity in deduced amino acid sequence and presumably arose through gene duplication. The cDNAs encode for putative 593- and 594-amino acid growth hormone receptors (designated GHR1 and GHR2, respectively), each containing a single transmembrane domain and other motifs characteristic of the receptor family. Both GHR1 and GHR2 mRNAs were present in all tissues examined. Trout GHR mRNAs are differentially expressed, both in terms of abundance among tissues and in terms of abundance within selected tissues. GHR1 was more abundant than GHR2 in the brain, whereas GHR2 was more abundant than GHR1 in pancreas and spleen. These findings expand our understanding of the evolution of the GH receptor family and suggest that independent mechanisms serve to regulate the tissue-specific expression of GHR mRNAs.

Advances in our understanding of the physiologic properties of oral agents has led to the discovery of methods for avoiding adverse effects. Additionally, new compounds and improved delivery by the aerosol route have mitigated side effects. Even with the new knowledge gained, systemic corticosteroids remain one of the most misunderstood medications utilized in the treatment of asthma and other pulmonary diseases. Abstract:The growth hormone(GH) gene has been cloned from a number of fishes and was used to clarify phylogenetic relationships.In order to better understand the physiological functions and evolution mechanism of the growth hormone of Turbot(Scophthalmus maximus),We cloned the full-length cDNA of the growth hormone(SmGH) gene by Switching Mechanism At 5' end of the RNA Transcript(SMART) RACE technology.The SmGH full-length cDNA of Turbot is 876 nucleotides long,codes for a polypeptide of 197 amino acids,including a signal peptide of 17 amino acids.The 5' and 3' UTR of the messenger RNA are 108 and 174 nucleotides long,respectively.Homologous analysis of SmGH cDNA only showed about 70% identity with the organisms of Pleuronectiformes,on the other hand,Paralichthys lethostigma showed 97.6% of similarity with Paralichthys orbignyanu and 94.8% with Paralichthys olivceus.The deduced amino acid sequences of SmGH of 7 species of Pleuronectiformes and 8 outgroup species were used for the phylogenetic analysis,which was performed by PAUP software with the maximum parsimony method.The result was consistent with morphology of Pleuronectiformes on the whole.In the topology founded,the Turbot formed an independent branch and showed far relationship with species of Bothidae and Pleuronectidae,it seems reasonable to speculate that Scophthalmus should be separated from Bothidae and become a new family of Pleuronectiformes.

Abstract:Growth hormone receptor (Ghr) is a single-transmembrane pass protein which is important in initiating the ability of growth hormone (Gh) to regulate development and somatic growth in vertebrates. In this study, molecular cloning, expression analysis of two different ghr genes (ghr1 and ghr2) in the tongue sole (Cynoglossus semilaevis) was conducted. As a result, the ghr1 and ghr2 cDNA sequences are 2364 bp and 3125 bp, each of which encodes a transmembrane protein of 633 and 561 amino acids (aa), respectively. Besides, the ghr1 gene includes nine exons and eight introns. The sex-specific tissue expression was analyzed by using 14 tissues from females, normal males and extra-large male adults. Both the ghr1 and ghr2 were predominantly expressed in the liver, and the ghr1 expression level in normal males was 1.6 and 1.4 times as much as those in females and extra-large males, while the ghr2 mRNA expression level in normal males was 1.1 and 1.2 times as much as those in females and extra-large males, respectively. Ontogenetic expression analysis at early life stages indicated that the ghr1 and ghr2 mRNAs were detected at all of the 35 sampling points (from oosphere to 410days-old). Furthermore, the sex differences in ghr mRNA expressions were also examined by using a full-sib family of C. semilaevis. Significantly higher levels of ghr1 mRNA were observed in males than in females at most stages of the sampling period (P<0.01). The ghr2 mRNA expression at most stages exhibited a significant sexual difference at each sampling point (P<0.01) without any variation trend related with the sexes during the whole sampling period.

Abstract:Yellow catfish(Pelteobagrus fulvidraco Richardson)is one of the most important freshwater farmed species in China.However,its small size and slow growth rate limit its economic value.Growth hormone is a protein secreted by pituitary.It promotes animal growth.Overexpression of growth hormone in fishes results in the growth hormone gene transgenic fishes with enlarged body size and accelerated growth.In order to increase its body size and growth rate,we performed transgenic research on yellow catfish.Employing RT-PCR and RACE(rapid amplification of cDNA ends)-PCR,we cloned 603 bp(base pairs)complete coding sequence and 485 bp 3′-UTR(untranslated region)of yellow catfish growth hormone gene,respectively.Performing overlapping PCR and molecular cloning with restriction endonucleases,we constructed an "all fish" growth hormone gene transgenic construct with 2105 bp in length.The construct comprises 1017 bp of yellow catfish β-actin proximal promoter,603 bp complete coding sequence and 485 bp 3′-UTR of yellow catfish growth hormone gene,respectively.Screening 301 individuals developed from the yellow catfish fertilized eggs microinjected with the growth hormone gene transgenic construct,we obtained 40 transgenic founders.Our results showed that we have established a yellow catfish transgenic platform that lays the foundation for our further molecular breeding to create new strains of yellow catfish with large body size and rapid growth.

Abstract:We developed a specific competitive enzyme-linked immunosorbent assay (ELISA) for yellowtail kingfish (Seriola lalandi) follicle stimulating hormone (FSH). We previously produced a full-length single chain recombinant yellowtail kingfish FSH using the Pichia pastoris expression system. We used the same method to produce the β subunit of the hormone, against which polyclonal antibodies were raised in rabbits. We first confirmed immunoreactivity of the polyclonal antibodies with the recombinant full length FSH and FSHβ as well as plasma and pituitary FSH of sexually immature and mature yellowtail kingfish by Western blot analysis. We then developed a precise and reproducible ELISA for yellowtail kingfish FSH and validated the assay in plasma and pituitary extracts. The intra- and inter-assay coefficients of variation was <2.2% and 10.2%, respectively. The sensitivity of the assay was 78 pg/ml. For further validation of the assay, we measured the plasma FSH in immature yellowtail kingfish treated with increasing doses (blank, 50, 100 and 150 µg/kg) of kisseptin2-10 peptide from a previous study. The dose response observed in treated females was not significant, however the increased plasma FSH levels coincided with the significantly higher estradiol levels we previously reported in the treated groups. We assessed the applicability of the assay in measuring circulating FSH in other species. We observed parallelism between the linearized FSH standard curve and displacement curves of serially diluted plasma from Atlantic bluefin tuna (Thunnus thynnus) and tilapia (Oreochromis niloticus). We also observed similar parallelism with full length recombinant giant grouper (Epinephelus lanceolatus) FSH. The ELISA we developed for yellowtail kingfish FSH will be useful in understanding the reproductive biology of the species as well as enhancing its aquaculture.

Abstract:The cDNA of growth hormone receptor (GHR) was cloned from the liver of 2-year common carp (Cyprinus carpio L.) by reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA end (RACE). Its open reading frame (ORF) of 1806 nucleotides is translated into a putative peptide of 602 amino acids, including an extracellular ligand-binding domain of 244 amino acids (aa), a single transmembrane domain of 24 aa and an intracellular signal-transduction domain of 334 aa. Sequence analysis indicated that common carp GHR is highly homologous to goldfish (Carassius auratus) GHR at both gene and protein levels. Using a pair of gene-specific primers, a GHR fragment was amplified from the cDNA of 2-year common carp, a 224 bp product was identified in liver and a 321 bp product in other tissues. The sequencing of the products and the partial genomic DNA indicated that the difference in product size was the result of a 97 bp intron that alternatively spliced. In addition, the 321 bp fragment could be amplified from all the tissues of 4-month common carp including liver, demonstrating the occurrence of the alternative splicing of this intron during the development of common carp. Moreover, a semi-quantitative RT-PCR was performed to analyze the expression level of GHR in tissues of 2-year common carp and 4-month common carp. The result revealed that in the tissues of gill, thymus and brain, the expression level of GHR in 2-year common carp was significantly tower than that of 4-month common carp.
