Abstract:Photosystem II (PSII), especially the D1 protein, is highly sensitive to the detrimental impact of heat stress. Photoinhibition always occurs when the rate of photodamage exceeds the rate of D1 protein repair. Here, genetically engineered codA-tomato with the capability to accumulate glycinebetaine (GB) was established. After photoinhibition treatment at high temperature, the transgenic lines displayed more thermotolerance to heat-induced photoinhibition than the control line. GB maintained high expression of LeFtsHs and LeDegs and degraded the damaged D1 protein in time. Meanwhile, the increased transcription of synthesis-related genes accelerated the de novo synthesis of D1 protein. Low ROS accumulation reduced the inhibition of D1 protein translation in the transgenic plants, thereby reducing protein damage. The increased D1 protein content and decreased phosphorylated D1 protein (pD1) in the transgenic plants compared with control plants imply that GB may minimize photodamage and maximize D1 protein stability. As D1 protein exhibits a high turnover, PSII maybe repaired rapidly and efficiently in transgenic plants under photoinhibition treatment at high temperature, with the resultant mitigation of photoinhibition of PSII.

Glycinebetaine accumulation is more effective in chloroplasts than in the cytosol for protecting transgenic tomato plants against abiotic stress

Abstract:Tomato (Lycopersicon esculentum Mill. cv. Moneymaker) plants were transformed with a gene for choline oxidase (codA) from Arthrobacter globiformis. The gene product (CODA) was targeted to the chloroplasts (Chl-codA), cytosol (Cyt-codA) or both compartments simultaneously (ChlCyt-codA). These three transgenic plant types accumulated different amounts and proportions of glycinebetaine (GB) in their chloroplasts and cytosol. Targeting CODA to either the cytosol or both compartments simultaneously increased total GB content by five- to sixfold over that measured from the chloroplast targeted lines. Accumulation of GB in codA transgenic plants was tissue dependent, with the highest levels being recorded in reproductive organs. Despite accumulating, the lowest amounts of GB, Chl-codA plants exhibited equal or higher degrees of enhanced tolerance to various abiotic stresses. This suggests that chloroplastic GB is more effective than cytosolic GB in protecting plant cells against chilling, high salt and oxidative stresses. Chloroplastic GB levels were positively correlated with the degree of oxidative stress tolerance conferred, whereas cytosolic GB showed no such a correlation. Thus, an increase in total GB content does not necessarily lead to enhanced stress tolerance, but additional accumulation of chloroplastic GB is likely to further raise the level of stress tolerance beyond what we have observed.
Polyamine Accumulation in Transgenic Tomato Enhances the Tolerance to High Temperature Stress

Abstract:Polyamines play an important role in plant response to abiotic stress. S-adenosyl-l-methionine decarboxylase (SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines. In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato, SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation. Transgene and expression was confirmed by Southern and Northern blot analyses, respectively. Transgenic plants expressing yeast SAMDC produced 1.7- to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress, and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed. This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress, which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress (38 degrees C) compared with wild-type plants. Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.

Abstract:To evaluate the biological tolerance of the human liver to prolonged warm ischemia, two groups of extensive hepatic resection for tumor were compared. Group 1 (11 patients) performed with short hepatic inflow occlusion (7 [mean] +/- 2 [SEM] minutes), and group 2 (nine patients) operated with use of complete hepatic vascular exclusion and prolonged warm liver ischemia (38 [mean] +/- 5 [SEM] minutes). Comparison of biological values, such as transaminase, bilirubin, total protein, albumin, and fibrinogen levels, the platelet count, prothrombin complex, and proaccelerin level, did not show statistically significant differences between the two groups. Therefore, the hepatic warm ischemia period may be, if needed, safely extended beyond the classical 15 minutes. It lasted 65 minutes in one case without adverse effect. These clinical observations parallel recent experimental work and should destroy the myth of the high sensitivity of the liver to warm ischemia.

The protective mechanisms of GB on photosystem I (PSI) and photosystem II (PSII) against chilling stress, however, remain poorly understood. Here, we address this problem through exogenous GB application and generation of transgenic tomatoes (Moneymaker) with a gene encoding betaine aldehyde dehydrogenase (BADH), which is the key enzyme in the synthesis of GB, from spinach. Our results demonstrated that GB can protect chloroplast ultramicrostructure, alleviate PSII photoinhibition and maintain PSII stability under chilling stress. More importantly, GB increased the electron transfer between QA and QB and the redox potential of QB and maintained a high rate of cyclic electron flow around PSI, contributing to reduced production of reactive oxygen species, thereby mitigating PSI photodamage under chilling stress. Our results highlight the novel roles of GB in enhancing chilling tolerance via the protection of PSI and PSII in BADH transgenic and GB-treated tomato plants under chilling stress. Thus, introducing GB-biosynthetic pathway into tomato and exogenous GB application are effective strategies for improving chilling tolerance.

Abstract:GDP-mannose pyrophosphorylase (GMPase: EC 2.7.7.22) plays a crucial role in the synthesis of l-ascorbate (AsA) and the consequent detoxification of reactive oxygen species (ROS). Herein, a GMPase (accession ID DQ449030) was identified and cloned from tomato. The full-length cDNA sequence of this gene contains 1,498 bp nucleotides encoding a putative protein with 361 amino acid residues of approximate molecular weight 43 kDa. Northern blot analysis revealed that the GMPase was expressed in all examined tomato tissues, but its expression level was up-regulated in tomato plants subjected to abnormal temperatures. We then overexpressed this tomato GMPase in tobacco plants and observed that the activity of GMPase and the content of AsA were significantly increased by two- to fourfold in the leaves of transgenic tobacco plants. The effect of this gene overexpression was superimposed by the treatments of high or low temperature in tobacco, since the activities of both chloroplastic SOD (superoxide dismutase EC 1.15.1.1), APX (ascorbate peroxidase EC 1.11.1.7) and the content of AsA in leaves were significantly higher in transgenic plants than those of WT, while the contents of H2O2 and O2 −· were reduced. Meanwhile, relative electric conductivity increased less in transgenic plants than that in WT, and the net photosynthetic rate (P n) and the maximal photochemical efficiency of PSII (F v/F m) of transgenic plants were notably higher than those of WT under temperature stresses. In conclusion, the overexpression of GMPase increased the content of AsA, thereby leading to the increase in tolerance to temperature stress in transgenic plants.
Exogenous application of glycinebetaine increases chilling tolerance in tomato plants

Abstract:Tomato (Lycopersicon esculentum Mill. cv. Moneymaker) plants are chilling sensitive, and do not naturally accumulate glycinebetaine (GB), a metabolite that functions as a stress protectant. We reported previously that exogenous GB application enhanced chilling tolerance in tomato. To understand its protective role better, we have further evaluated various parameters associated with improved tolerance. Although its effect was most pronounced in younger plants, this benefit was diminished 1 week after GB application. When administered by foliar spray, GB was readily taken up and translocated to various organs, with the highest levels being measured in meristematic tissues, including the shoot apices and flower buds. In leaves, the majority of endogenous GB was found in the cytosol; only 0.6-22.0% of the total leaf GB was localized in chloroplasts. Immediately after GB application, levels of H(2)O(2), catalase activity and expression of the catalase gene (CAT1) were all higher in GB-treated than in control plants. One day after exposure to chilling stress, the treated plants had significantly greater catalase activity and CAT1 expression, although their H(2)O(2) levels remained unchanged. During the following 2 d of this chilling treatment, GB-treated plants maintained lower H(2)O(2) levels but had higher catalase activity than the controls. These results suggest that, in addition to protecting macromolecules and membranes directly, GB-enhanced chilling tolerance may involve the induction of H(2)O(2)-mediated antioxidant mechanisms, e.g. enhanced catalase expression and catalase activity.

Abstract:Genetically engineered tomato (Lycopersicon esculentum) with the ability to synthesize glycinebetaine was generated by introducing the codA gene encoding choline oxidase from Arthrobacter globiformis. Integration of the codA gene in transgenic tomato plants was verified by PCR analysis and DNA blot hybridization. Transgenic expression of gene was verified by RT-PCR analysis and RNA blot hybridization. The codA-transgenic plants showed higher tolerance to salt stress during seed germination, and subsequent growth of young seedlings than wild-type plants. The codA transgene enhanced the salt tolerance of whole plants and leaves. Mature leaves of codA-transgenic plants revealed higher levels of relative water content, chlorophyll content, and proline content than those of wild-type plants under salt and water stresses. Results from the current study suggest that the expression of the codA gene in transgenic tomato plants induces the synthesis of glycinebetaine and improves the tolerance of plants to salt and water stresses.

Abstract:Abiotic stress such as drought seriously endangers maize growth and production. Glycine betaine (GB) accumulation in transgenic crops following heterologous overexpression of the BADH gene has been shown to dramatically improve the tolerance to salt, cold, and oxidative stresses, but it is unclear if GB accumulation leads to improved drought tolerance of maize. In this study, we analyzed the drought tolerance of high-generation BADH transgenic maize inbred lines at different growth stages using the hyperosmotic solution and water-withholding methods. Molecular detection revealed that exogenous BADH was successfully introduced into the maize plant genome and overexpressed in three transgenic maize inbred lines. Under osmotic stress, transgenic maize held better germination ability than the unmodified Dan988 (WT) line. In addition, transgenic maize contained higher levels of antioxidant enzymes and osmotic regulatory substances compared with WT, and thus accumulated less harmful substances and this alleviated the negative effects of drought. We used the membership function method to quantify the drought tolerance and found that Dan988-BADH-4 showed the best tolerance, followed by Dan988-BADH-2 and Dan988-BADH-1 while WT ranked lowest. This was consistent with the statistical analysis of the experiments. From the indoor and field survey, we observed that the agronomic traits of transgenic maize were not affected by the overexpression of BADH. In conclusion, BADH overexpression in maize is beneficial for drought tolerance and the three transgenic maize lines can be used for further breeding experiments.

Abstract:A chloroplast-localized tomato (Lycopersicon esculentum Mill.) omega-3 fatty acid desaturase gene (LeFAD7) was isolated and characterized with regard to its sequence, response to various temperatures, and function in antisense transgenic tomato plants. The deduced amino acid sequence had four histidine-rich regions, of which three regions were highly conserved throughout the whole omega-3 fatty acid desaturase gene family. Southern blotting analysis showed that LeFAD7 was encoded by a single copy gene and had two homologous genes in the tomato genome. Northern blot showed that LeFAD7 was expressed in all organs and was especially abundant in leaf tissue. Meanwhile, expression of LeFAD7 was induced by chilling stress (4 degrees C), but was inhibited by high temperature (45 degrees C), in leaves. Transgenic tomato plants were produced by integration of the antisense LeFAD7 DNA under the control of a CaMV35S promoter into the genome. Antisense transgenic plants with lower 18:3 content could maintain a higher maximal photochemical efficiency (F-v/F-m) and O-2 evolution rate than wild-type plants. These results suggested that silence of the LeFAD7 gene alleviated high-temperature stress. There was also a correlation between the low content of 18:3 resulting from silence of the LeFAD7 gene and tolerance to high-temperature stress.

Relative electric conductivity increased less in transgenic plants than in WT. Under heat stress, the maximal photochemical efficiency of photosystem II (Fv/Fm) and the O(2) evolution rate decreased more in WT than in transgenic plants. Enhanced tolerance of transgenic potato plants expressing choline oxidase in chloroplasts against water stress

Abstract:Background: Glycinebetaine, whose biosynthesis could be catalyzed by choline oxidase (COD), is an extremely efficient compatible solute for scavenging oxidative stress-inducing molecules and protecting the photosynthetic system in plants. To study the effects of the codA transgene for choline oxidase on the drought resistance and recovery, a transgenic potato cultivar (SC) bearing codA gene and a non-transgenic (NT) control cultivar were raised in pots under moderate and severe drought stress. The experiment was constituted by a two-day-pretreatment with 20% PEG and a four-day-water stress combined with two-day-recovery treatment. Results: Under the four-day-water stress, plants were provided with normal water condition, 10% or 20% polyethylene glycol. The results of pretreatment showed an expression of codA gene in transgenic potato and an accumulation of glycine betaine (GB); leaf water potential was higher in SC than in NT. In the stress-recovery-treatment, SC showed stronger antioxidant ability, more efficient photosynthetic system, higher chlorophyll content, lower malondialdehyde content and better recovery from water deficit stress than NT. Conclusion: Although this work concentrated on the short-term water stress and recover treatments on transgenic potato plants with the over-expression of CodA gene and its control line. The datas shows that the exogenous codA gene provided potato a stronger drought resistance and recovery ability.

Abstract:Genetically engineered tomato hairy root lines with the ability to synthesize glycine betaine were established by introducing the betaine aldehyde dehydrogenase gene (BADH-1) from sorghum plants. The lines showed a mikimopine activity, confirming the transformation with pRi plasmid. The stable integration of the BADH gene into the tomato genome was confirmed by Southern blotting analysis. The expression of BADH mRNA could be detected in the tomato hairy roots by Northern analysis. The osmotic potential (Ψ s) of the transgenic hairy roots was sufficient to allow water uptake. The accumulation of betaine was detected by 1H-NMR analysis. The accumulation of betaine in the transgenic hairy roots contributed to the maintenance of the osmotic potential of the cells.

Abstract:Cold stress is known to influence tomato growth, development, and yield. In this study, we analyzed the germination of tomato seeds treated with exogenous glycine betaine (GB) at a low temperature (14 °C). The results showed that cold stress inhibited tomato seed germination, and pretreatment with exogenous GB reduced this inhibition and enhanced the germination rate (GR), germination index (GI), and viability of tomato seeds at low temperatures. Analysis of gene expression and metabolism revealed that GB positively regulated endogenous hormone gibberellin (GA) content and negatively regulated abscisic acid (ABA) content, while GB reduced the starch content in the seeds by up-regulating the amylase gene expression. Gene expression analysis showed that the key genes (SlSOD, SlPOD, and SlchlAPX) involved in reactive oxygen species (ROS) scavenging systems were up-regulated in GB-pretreated tomato seeds compared with the control. At the same time, levels of malondialdehyde and hydrogen peroxide were significantly lower, while the proline content and peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT) levels were elevated compared with those in the control. These results demonstrate that exogenous GB as a positive regulator effectively alleviated the inhibition of tomato seed germination under cold stress by different signal pathways.

Abstract:Plant genetic modification in order to increase their tolerance to various abiotic stresses has been of exceptional importance in recent years. Heterologous expression of glycine betaine (GB) biosynthetic genes leads to increased salt and drought tolerance in various plant species by maintaining the osmotic balance with the environment and stabilizing the quaternary structure of complex proteins. However, GB biosynthesis in transgenic plants is limited by choline availability. Members of the Plumbaginaceae family accumulate -alanine betaine (AB) instead [1]. The synthesis of AB is not limited by the availability of choline, as it follows the methylation pathway of the aproteinogenic amino acid -alanine. For the first time, we have generated Nicotiana tabacum plants expressing the -alanine N-methyltransferase (LlBANMT) gene of Limonium latifolium. Transgenic plants were much less affected by such abiotic stresses as increased salinity, excessive illumination, and low temperature. The experimental Nicotiana tabacum lines had lower rates of chlorophyll degradation under stress conditions compared to the control plants. LlBANMT expression also resulted in less biomass loss under stress conditions, which was associated with higher activities of reactive oxygen species detoxification systems and healthier cell membranes. Increase of glycinebetaine synthesis improves drought tolerance in cotton

Abstract:The tolerance to drought stress of the homozygous transgenic cotton (Gossypium hirsutum L.) plants with enhanced glycinebetaine (GB) accumulation was investigated at three development stages. Among the five transgenic lines investigated, lines 1, 3, 4, and 5 accumulated significantly higher levels of GB than the wild-type (WT) plants either before or after drought stress, and the transgenic plants were more tolerant to drought stress than the wild-type counterparts from young seedlings to flowering plants. Under drought stress conditions, transgenic lines 1, 3, 4, and 5 had higher relative water content, increased photosynthesis, better osmotic adjustment (OA), a lower percentage of ion leakage, and less lipid membrane peroxidation than WT plants. The GB levels in transgenic plants were positively correlated with drought tolerance under water stress. The results suggested that GB may not only protect the integrity of the cell membrane from drought stress damage, but also be involved in OA in transgenic cotton plants. Most importantly, the seedcotton yield of transgenic line 4 was significantly greater than that of WT plants after drought stress, which is of great value in cotton production.

Gly Betaine Surpasses Melatonin to Improve Salt Tolerance in Dalbergia odorifera

Abstract:Salinity is one of the most serious factors limiting plant growth which can provoke significant losses in agricultural crop production, particularly in arid and semi-arid areas. This study aimed to investigate whether melatonin (MT; 0.05 and 0.1 mM), which has pleiotropic roles, has a better effect than glycine betaine (GB; 10 and 50 mM) on providing salt tolerance in a woody plant Dalbergia odorifera T. Chen. Also, the alternative oxidase activity (AOX) in plant subjected to MT or GB under salinity (150 and 250 mM) was evaluated given that the effect of exogenous MT or GB on AOX has not been reported yet. The results showed that the exogenous application of GB on the seedlings of D. odorifera increased the plant growth parameters, relative water content, total of chlorophyll content, and carotenoid content compared with well-watered and MT treatments. Under severe salinity, the seedlings subjected to GB showed, a significant enhancement in water use efficiency, transpiration, and net photosynthetic rate regardless to MT-treated seedlings. The levels of proline and soluble sugar in the seedlings treated with MT or GB decreased significantly under mild and severe salinity correlated with those in salt-stressed seedlings. Furthermore, GB-treated plants exhibited a significant inhibition of malondialdehyde content compared with MT-treated plants. The concentration of thiols and phenolic compounds were significantly enhanced in the leaves of seedlings treated with MT compared with those treated with GB. Under salt stress condition, GB scavenged significantly higher levels of hydrogen peroxide than MT; while under severe salinity, plants subjected to MT showed better scavenging ability for hydroxyl radicals compared with GB-treated seedlings. The results demonstrated also an enhancement of the levels of superoxide dismutase (SOD), guaiacol peroxidase, and AOX activities in seedlings treated with GB or MT compared with salt-stressed plants. The catalase activity (CAT) was increased by 0.05 mM MT and 0.1 mM GB under mild salinity. Meanwhile, the AOX activity under severe salinity was enhanced only by GB 50 mM. The findings of this study suggested that GB-treated seedlings possessed a better salt tolerance in comparison with MT-treated seedlings.

Abstract:In order to investigate the relationship between the lipid composition in thylakoid membrane and thermostability of photosynthetic apparatus, tobacco transformed with sweet pepper sense glycerol-3-phosphate acyltransferase (GPAT) gene were used to analyze the lipid composition in thylakoid membrane, the net photosynthetic rate and chlorophyll fluorescence parameters under high temperature stress. The results showed that the saturated extent of monogalactosyldiacylglycerol (MGDG), sulfoquinovosyldiacylglycerol, digalactosyldiacylglycerol and phosphatidylglycerol in thylakoid membrane of transgenic tobacco T(1) lines increased generally. Particularly, the saturated extent in MGDG increased obviously by 16.2% and 12.0% in T(1)-2 and T(1)-1, respectively. With stress temperature elevating, the maximum efficiency of photosystem II (PSII) photochemistry (Fv/Fm), actual photochemical efficiency of PSII in the light (Phi(PSII)) and net photosynthetic rate (Pn) of the two lines and wild type tobacco plants decreased gradually, but those parameters decreased much less in transgenic plants. Even though the recovery process appeared differently in the donor and acceptor side of PSII in transgenic tobacco compared with wild-type plants, the entire capability of PSII recovered faster in transgenic tobacco, which was shown in the parameters of PI, Fv/Fm and Phi(PSII), as a result, the recovery of Pn was accelerated. Conclusively, we proposed that the increase in saturated extent of thylakoid membrane lipids in transgenic plants enhanced the stability of photosynthetic apparatus under high temperature stress.

Novel DnaJ Protein Facilitates Thermotolerance of Transgenic Tomatoes

Abstract:DnaJ proteins, which are molecular chaperones that are widely present in plants, can respond to various environmental stresses. At present, the function of DnaJ proteins was studied in many plant species, but only a few studies were conducted in tomato. Here, we examined the functions of a novel tomato (Solanum lycopersicum) DnaJ protein (SlDnaJ20) in heat tolerance using sense and antisense transgenic tomatoes. Transient conversion assays of Arabidopsis protoplasts showed that SlDnaJ20 was targeted to chloroplasts. Expression analysis showed that SlDnaJ20 expression was induced by chilling, NaCl, polyethylene glycol, and H2O2, especially via heat stress. Under heat stress, sense plants showed higher fresh weights, chlorophyll content, fluorescence (Fv/Fm), and D1 protein levels, and a lower accumulation of reactive oxygen species (ROS) than antisense plants. These results suggest that SlDnaJ20 overexpression can reduce the photoinhibition of photosystem II (PSII) by relieving ROS accumulation. Moreover, higher expression levels of HsfA1 and HsfB1 were observed under heat stress in sense plants, indicating that SlDnaJ20 overexpression contributes to HSF expression. The yeast two-hybrid system proved that SlDnaJ20 can interact with the chloroplast heat-shock protein 70. Our results indicate that SlDnaJ20 overexpression enhances the thermotolerance of transgenic tomatoes, whereas suppression of SlDnaJ20 increases the heat sensitivity of transgenic tomatoes.
