Abstract:Cold acclimation (CA) is a strategy which plants have evolved to increase freezing tolerance. Global climate change could obstruct CA and raise the probability of winter injury, especially for evergreens. Hence, understanding the regulatory mechanism of CA is crucial to improve freezing tolerance in evergreen plants. A comparative study on a pair of closely related evergreen and deciduous iris species in response to cold through CA was conducive to uncovering and complementing the knowledge of CA. We investigated morphological, physiological and biochemical changes, as well as the expression of associated genes in the functional leaves of both iris species from natural CA to deacclimation. Briefly, fast and strong CA in the evergreen iris might cause early expressions of BAM1, NCED3, GPX6, etc., which leads to strong enzyme activity of starch degradation, abscisic acid biosynthesis and reactive oxygen species scavenging. Additionally, genes belonging to the antioxidant system were mainly induced during deacclimation. These results suggest that interspecies differences in the leaf freezing tolerance of irises are associated with the rate and degree of CA, which activates multiple signaling networks with complex interactions and induces the transcription of cold-responsive genes. Moreover, the ICE–CBF–COR signaling cascade may integrate and initiate diverse cold-responsive pathways during CA of the evergreen iris. The findings of this study provide valuable insight to further research on CA mechanisms and implicate genes which could support breeding strategies in herbaceous perennials under climate changes.
Low temperature acclimation of photosynthetic capacity and leaf morphology in the context of phloem loading type

Abstract:Carbon export from leaf mesophyll to sugar-transporting phloem occurs via either an apoplastic (across the cell membrane) or symplastic (through plasmodesmatal cell wall openings) pathway. Herbaceous apoplastic loaders generally exhibit an up-regulation of photosynthetic capacity in response to growth at lower temperature. However, acclimation of photosynthesis to temperature by symplastically loading species, whose geographic distribution is particularly strong in tropical and subtropical areas, has not been characterized. Photosynthetic and leaf anatomical acclimation to lower temperature was explored in two symplastic (Verbascum phoeniceum, Cucurbita pepo) and two apoplastic (Helianthus annuus, Spinacia oleracea) loaders, representing summer- and winter-active life histories for each loading type. Regardless of phloem loading type, the two summer-active species, C. pepo and H. annuus, exhibited neither foliar anatomical nor photosynthetic acclimation when grown under low temperature compared to moderate temperature. In contrast, and again irrespective of phloem loading type, the two winter-active mesophytes, V. phoeniceum and S. oleracea, exhibited both a greater number of palisade cell layers (and thus thicker leaves) and significantly higher maximal capacities of photosynthetic electron transport, as well as, in the case of V. phoeniceum, a greater foliar vein density in response to cool temperatures compared to growth at moderate temperature. It is therefore noteworthy that symplastic phloem loading per se does not prevent acclimation of intrinsic photosynthetic capacity to cooler growth temperatures. Given the vagaries of weather and climate, understanding the basis of plant acclimation to, and tolerance of, low temperature is critical to maintaining and increasing plant productivity for food, fuel, and fiber to meet the growing demands of a burgeoning human population.

Abstract:Abstract A simple method is presented for converting an age distribution in any closed population into the stationary population corresponding to its current mortality conditions. The conversion only requires a set of age-specific growth rates, which will normally be available from successive censuses. From the stationary population, any life table mortality measure of interest can be computed. The index most robust to normal data errors in developing countries is life expectancy, and the paper focuses on its calculation. The sensitivity of results to various forms of data error is considered, and procedures are proposed for removing errors resulting from differential census coverage completeness and from age misstatement at older ages. Applications of the procedures are made to data from Sweden, India and South Korea. Because of the absence of a radix, estimation of life expectancy usually will begin at the fifth birthday.
Habitat Temperature and Precipitation of Arabidopsis thaliana Ecotypes Determine the Response of Foliar Vasculature, Photosynthesis, and Transpiration to Growth Temperature

Abstract:Acclimatory adjustments of foliar vascular architecture, photosynthetic capacity, and transpiration rate in Arabidopsis thaliana ecotypes (Italian, Polish [Col-0], Swedish) were characterized in the context of habitat of origin. Temperatures of the habitat of origin decreased linearly with increasing habitat latitude, but habitat precipitation was greatest in Italy, lowest in Poland, and intermediate in Sweden. Plants of the three ecotypes raised under three different growth temperature regimes (low, moderate, and high) exhibited highest photosynthetic capacities, greatest leaf thickness, highest chlorophyll a/b ratio and levels of β-carotene, and greatest levels of wall ingrowths in phloem transfer cells, and, in the Col-0 and Swedish ecotypes, of phloem per minor vein in plants grown at the low temperature. In contrast, vein density and minor vein tracheary to sieve element ratio increased with increasing growth temperature - most strongly in Col-0 and least strongly in the Italian ecotype - and transpirational water loss correlated with vein density and number of tracheary elements per minor vein. Plotting of these vascular features as functions of climatic conditions in the habitat of origin suggested that temperatures during the evolutionary history of the ecotypes determined acclimatory responses of the foliar phloem and photosynthesis to temperature in this winter annual that upregulates photosynthesis in response to lower temperature, whereas the precipitation experienced during the evolutionary history of the ecotypes determined adjustment of foliar vein density, xylem, and transpiration to temperature. In particular, whereas photosynthetic capacity, leaf thickness, and foliar minor vein phloem features increased linearly with increasing latitude and decreasing temperature of the habitats of origin in response to experimental growth at low temperature, transpiration rate, foliar vein density, and minor vein tracheary element numbers and cross-sectional areas increased linearly with decreasing precipitation level in the habitats of origin in response to experimental growth at high temperature. This represents a situation where temperature acclimation of the apparent capacity for water flux through the xylem and transpiration rate in a winter annual responded differently from that of photosynthetic capacity, in contrast to previous reports of strong relationships between hydraulic conductance and photosynthesis in other studies.

Abstract:In high-elevation grasslands, plants can encounter periods with high air temperature while the soil remains cold, which may lead to a temporary mismatch in the physiological activity of leaves and roots. In a climate chamber experiment with graminoid species from three elevations (4400, 2400, and 250 m a.s.l.), we tested the hypothesis that soil temperature can influence photosynthesis and stomatal conductance independently of air temperature. Soil monoliths with swards of Kobresia pygmaea (high alpine), Nardus stricta (lower alpine), and Deschampsia flexuosa (upper lowland) were exposed to soil temperatures of 25, 15, 5, and -2∘C and air temperatures of 20 and 10∘C for examining the effect of independent soil and air temperature variation on photosynthesis, leaf dark respiration, and stomatal conductance and transpiration. Soil frost (-2∘C) had a strong negative effect on gas exchange and stomatal conductance in all three species, independent of the elevation of origin. Leaf dark respiration was stimulated by soil frost in D. flexuosa, but not in K. pygmaea, which also had a lower temperature optimum of photosynthesis. Soil cooling from 15 to 5∘C did not significantly reduce stomatal conductance and gas exchange in any of the species. We conclude that all three graminoids are able to maintain a relatively high root water uptake in cold, non-frozen soil, but the high-alpine K. pygmaea seems to be especially well adapted to warm shoot – cold root episodes, as it has a higher photosynthetic activity at 10 than 20∘C air temperature and does not up-regulate leaf dark respiration upon soil freezing, as was observed in the grasses from warmer climates.

Abstract:Larrea divaricata, a desert evergreen shrub, has a remarkable ability to adjust its photosynthetic temperature response characteristics to changing temperature conditions. In its native habitat on the floor of Death Valley, California, plants of this C(3) species when provided with adequate water are able to maintain a relatively high and constant photosynthetic activity throughout the year even though the mean daily maximum temperature varies by nearly 30 C from winter to summer. The temperature dependence of light-saturated net photosynthesis varies in concert with these seasonal temperature changes whereas the photosynthetic rate at the respective optimum temperatures shows little change.Experiments on plants of the same age, grown at day/night temperatures of 20/15, 35/25, and 45/33 C with the same conditions of day length and other environmental factors, showed a similar photosynthetic acclimation response as observed in nature. An analysis was made of a number of factors that potentially can contribute to the observed changes in the temperature dependence of net CO(2) uptake at normal CO(2) and O(2) levels. These included stomatal conductance, respiration, O(2) inhibition of photosynthesis, and nonstomatal limitations of CO(2) diffusive transport. None of these factors, separately or taken together, can account for the observed acclimation responses. Measurements under high saturating CO(2) concentrations provide additional evidence that the observed adaptive responses are primarily the result of changes in intrinsic characteristics of the photosynthetic machinery at the cellular or subcellular levels. Two apparently separate effects of the growth temperature regime can be distinguished: one involves an increased capacity for photosynthesis at low, rate-limiting temperatures with decreased growth temperature, and the other an increased thermal stability of key components of the photosynthetic apparatus with increased growth temperature.

Abstract:Leaves and assimilative branches are crucial to the life cycle of Alhagi sparsifolia Shap. that is one of the main shrubs in the southern fringe of the Taklamakan desert. They reduce photosynthesis at midday during summer and exhibit different degrees of adaptability to severe environments. We investigated the changes in the PSII activity of leaves and assimilative branches of under elevated temperature and whether they have the same response and level of tolerance to different temperatures, thus to understand the roles of leaves and assimilative branches of A. sparsifolia in global warming. In this study, the kinetics of induced chlorophyll a fluorescence under heat-stress-induced inhibition of photosystem II in leaves and assimilative branches of A. sparsifolia after 15 min of treatment was investigated. The results show that both the activity and density of reaction centers of leaves are higher than assimilative branch at 35–48 °C, and both in the leaves and assimilative branches were decreased above 45 °C, the assimilative branches adapted better to the severe environment in terms of light energy transfer, light usage efficiency, and electron transport at 52 °C. Assimilative branches had better adaptability to elevated temperature than leaves of A. sparsifolia suggesting that assimilative branches might be more adaptive to severe environment.

Abstract:Climate change and invasive alien species threaten biodiversity. High temperature is a worrying ecological factor. Most responses of invasive plants aimed at coping with adversity are focused on the physiological level. To explore the molecular mechanisms underlying the response of an invasive plant (Sphagneticola trilobata L.) to high temperature, using a native species (Sphagneticola calendulacea L.) as the control, relevant indicators, including photosynthetic pigments, gas exchange, chlorophyll fluorescence, the antioxidant system, and related enzyme-coding genes were measured. The results showed that the leaves of S. calendulacea turned yellow, photosynthetic pigment content (Chl a, Chl b, Car, Chl) decreased, gas exchange (Pn) and chlorophyll fluorescence parameters (Fv/Fm, ΦPSII) decreased under high temperature. It was also found that high temperature caused photoinhibition and a large amount of ROS accumulated, resulting in an increase in MDA and relative conductivity. Antioxidant enzymes (including SOD, POD, CAT, and APX) and antioxidants (including flavonoids, total phenols, and carotenoids) were decreased. The qPCR results further showed that the expression of the PsbP, PsbA, and RubiscoL, SOD, POD, CAT, and APX genes was downregulated, which was consistent with the results of physiological data. Otherwise, the resistance of S. trilobata to high temperature was better than that of S. calendulacea, which made it a superior plant in the invasion area. These results further indicated that the gradual warming of global temperature will greatly accelerate the invasion area of S. trilobata.

Abstract:Acclimation enables plants to adapt to immediate environmental fluctuations, supporting biodiversity and ecosystem services. However, global changes are altering conditions for plant disease outbreaks, increasing the risk of infections by pathogenic fungi and oomycetes, and often undermining plant immune responses. Understanding the molecular basis of plant acclimation is crucial for predicting climate change impacts on ecosystems and improving crop resilience. Here, we investigated how Arabidopsis thaliana quantitative immune responses acclimates to daily temperature fluctuations. We analyzed responses to the fungal pathogen Sclerotinia sclerotiorum following three acclimation regimes that reflect the distribution areas of both species. Mediterranean acclimation, characterized by broad diurnal temperature amplitudes, resulted in a loss of disease resistance in three natural A. thaliana accessions. Global gene expression analyses revealed that acclimation altered nearly half of the pathogen-responsive genes, many of which were down-regulated by inoculation and associated with disease susceptibility. Phenotypic analysis of A. thaliana mutants identified novel components of quantitative disease resistance following temperate acclimation. Several of these mutants were however more resistant than wild type following Mediterranean acclimation. Notably, mutant lines in the NAC42-like transcription factor did not show a loss of resistance under Mediterranean acclimation. This resistance was linked to an acclimation-mediated switch in the repertoire of NAC42-like targets differentially regulated by inoculation. These findings reveal the rewiring of immune gene regulatory networks by acclimation and suggest new strategies to maintain plant immune function in a warming climate.

High Temperature Alters Secondary Metabolites and Photosynthetic Efficiency in Heracleum sosnowskyi

Abstract:Due to global warming, invasive species have spread across the world. We therefore studied the impact of short-term (1 day or 2 days) and longer (7 days) heat stress on photosynthesis and secondary metabolites in Heracleum sosnowskyi, one of the important invasive species in the European Union. H. sosnowskyi leaves exposed to short-term heat stress (35 °C/1 d) showed a decrease in chlorophyll and maximum potential quantum efficiency of photosystem II (Fv/Fm) compared to control, 35 °C/2 d, or 30 °C/7 d treatments. In turn, the high level of lipid peroxidation and increased H2O2 accumulation indicated that the 30 °C/7 d stress induced oxidative damage. The contents of xanthotoxin and bergapten were elevated in the 2 d and 7 d treatments, while isopimpinellin was detected only in the heat-stressed plants. Additionally, the levels of free proline and anthocyanins significantly increased in response to high temperature, with a substantially higher increase in the 7 d (30 °C) treatment. The results indicate that the accumulation of proline, anthocyanins, and furanocoumarins, but not of phenolic acids or flavonols, contributes to protection of H. sosnowskyi plants against heat stress. Further studies could focus on the suppression of these metabolites to suppress the spread of this invasive species.

Abstract:Phenotypic plasticity occurs when plants acclimatize to contrasting conditions. Herein, we test the hypothesis that seedlings of a light-demanding species have greater phenotypic plasticity compared to seedlings of a shade-tolerant species under high irradiance. Thus, we investigate the growth, anatomical, and leaf gas exchange responses of Citharexylum myrianthum , a light-demanding species, and Poecilanthe parviflora , a shade-tolerant species, under full light and 60% shading. Under full light, the seedlings of both species were shorter, showed lower photosynthetic rates and specific leaf area, and thicker palisade parenchyma. In the same conditions, C. myrianthum showed increased number of leaves, and P. parviflora reduced leaf area and increased number of stomata and allocation of phloem and cortical parenchyma. Lower photosynthetic rates may negatively affect biomass allocation and growth, although C. myrianthum seems to show a higher tolerance to irradiance since it produced more leaves. P. parviflora seems to optimize heat dissipation, reduce water loss, and improve the allocation of photoassimilate transport and storage, which could increase performance during establishment in field conditions. The plasticity index of both species was similar. Thus, generalizations about the species plasticity and ecological group to which they belong should be avoided. Species-related responses of growth, anatomical, and gas exchange parameters were found, indicating that generalizations about the performance of functional groups should also be avoided. These findings may contribute to the success of forest restoration projects.

Strong responses of growth and photosynthesis of five C3 pasture species to elevated CO2 at low temperatures

Abstract:Five cool-season C3 pasture species (Bromus willdenowii Kunth, Festuca arundinacea Schreb, Lolium multiflorum Lam., Lolium perenne L. and Trifolium repens L.) were grown at a constant temperature of 12°C to assess effects of elevated CO2 on growth and photosynthesis. Plants were grown at either 350 or 700 μmol mol-1 CO2 for 42-56 d, after which they were destructively harvested to assess biomass. At the same time, leaf gas exchange was measured at 12°C for each species at both CO2 concentrations. Photosynthetic response to CO2, that is the A/Ci curve, was also measured. Results showed all species responded to elevated CO2 at low temperature by increasing total biomass, relative to that at ambient CO2, from 105 to 158%. T. repens and B. willdenowii had the smallest response, and the two Lolium species had the greatest response. Photosynthesis also responded strongly to elevated CO2 at low temperature, increasing on average by 40%. Only small growth CO2 effects occurred with the A/Ci curves, with lower CO2-saturated photosynthesis only for L. perenne and T. repens grown at elevated CO2. The photosynthetic response to CO2 was not correlated with the growth response. However, the product of photosynthesis and leaf area, that is carbon supply, was highly correlated with the growth rate, and growth CO2 had a marked effect. Results suggest T. repens responded poorly to elevated CO2 at low temperature because of a low capacity to fix carbon, while both Lolium species responded strongly at low temperature because of a high capacity to fix CO2. Changes to photosynthetic model parameters are now required to account for growth temperature effects on the CO2 response.

Abstract:Plants acclimate to temperature by adjusting their photosynthetic capacity over weeks to months. However, most evidence for photosynthetic acclimation derives from leaf-scale experiments. Here we address the scarcity of evidence for canopy-scale photosynthetic acclimation by examining the correlation between maximum photosynthetic rates ( A max,2,000 ) and growth temperature ( ) across a range of concurrent temperatures and canopy foliage quantity, using data from >200 eddy covariance sites. We detect widespread thermal acclimation of canopy-scale photosynthesis, demonstrated by enhanced A max,2,000 under higher , across flux sites with adequate water availability. A 14-day period is identified as the most relevant timescale for acclimation across all sites, with a range of 12–25 days for different plant functional types. The mean apparent thermal acclimation rate across all ecosystems is 0.41 (−0.38–1.04 for 5th–95th percentile range) μmol m −2 s −1 °C −1 , with croplands showing the largest acclimation rates and grasslands the lowest. Incorporating an optimality-based prediction of leaf photosynthetic capacities into a biochemical photosynthesis model is shown to improve the representation of thermal acclimation. Our results underscore the critical need for enhanced understanding and modelling of canopy-scale photosynthetic capacity to accurately predict plant responses to warmer growing seasons.

Abstract:Arctic ecosystems are important carbon sinks. Increasing temperatures in these regions might stimulate soil carbon release. Evidence suggests that deciduous shrubs might counteract these carbon losses because they positively respond to increasing temperature, but their role in ecosystem carbon budgets remains uncertain. Many studies dealing with large-scale tundra greening and carbon sequestration in relation to increasing temperature have usually based their estimations on the aboveground components, but very little is known about belowground growth. In this context, annual rings can provide a retrospective insight into intra-plant temperature responses and seasonal growth allocation. This study presents a 70-year-long and annually resolved intra-plant analysis of ring width and missing ring distribution from a comprehensive serial sectioning, including 142 cross-sections and the measurements of 471 radii from ten Salix polaris Wahlenb. dwarf shrubs growing in the high Arctic on Svalbard. Results indicate a high intra-plant and inter-annual growth variation, characterized by a high proportion of partially (13.6 %) and completely (11.2 %) missing rings. The annual growth and the frequency of completely missing rings were evenly distributed inside the plant and mainly controlled by summer temperatures. Radial growth in the belowground parts appeared to be proportionally higher during long and warm summers and lower in cold early growing seasons than in the aboveground parts. The results reveal a diverging allocation between aboveground and belowground growth depending on the climatic conditions. Favorable years promoted root allocation since root radial growth occurs after aboveground growth. The observed belowground responses suggest that shrub carbon allocation might be higher than estimated only from the aboveground compartments.

Abstract:The effects of increasing air temperature and changing daily temperature regime under conditions of elevated CO2 on the physiology, biomass, and reproduction of a C-3 plant species were investigated. Phytolacca americana L. ( Phytolaccaceae) was grown under either ambient ( 370 mu mol mol(-1)) or elevated ( 700 mu mol mol(-1)) CO2 at three air temperature regimes ( day/ night temperatures of 26 degrees/ 20 degrees C, T-1; 30 degrees/ 24 degrees C, T-2; and 28 degrees/ 24 degrees C, T-3). Length of day/ night temperature exposure was adjusted so that average daily temperature was 22 degrees C in T-1 and 26 degrees C in T-2 and T-3. Daily temperature regime was different for T-2 and T-3: plants in T-2 experienced a higher maximum daily temperature but for a shorter daily duration than plants in T-3. Elevated CO2 increased photosynthetic rate, total biomass, and root-to-shoot ratio (RSR) and decreased stomatal conductance and transpiration as well as allocation to reproduction. In contrast, elevated temperatures had no effect on photosynthetic rate, stomatal conductance, or total biomass, but they decreased RSR and increased transpiration, reproductive biomass, and allocation. Both elevated CO2 and increased temperatures advanced timing of flowering. The plant-level transpiration rate exhibited a unique response to each of the daily temperature regime treatments. These results indicate that elevated CO2 and increased temperatures elicit different responses at the physiological and whole-plant levels in P. americana, with little interaction between the CO2 and temperature effects. Furthermore, some evidence indicates that a changing daily temperature regime may be an important factor determining plant responses to warming temperatures and should be incorporated into predictions of plant and ecosystem responses to future climate change.

Abstract:The warming tolerances of understory plant species varied with macroclimatic differences among regions across Europe, as well as in response to forest microclimates, albeit to a lesser extent. Plant performance across species decreases in terms of growth and reproduction as local temperature conditions reach or exceed the warm limit of the focal species. Climate change is pushing species towards and potentially beyond their critical thermal limits. The extent to which species can cope with temperatures exceeding their critical thermal limits is still uncertain. To better assess species' responses to warming, we compute the warming tolerance (ΔTniche) as a thermal vulnerability index, using species' upper thermal limits (the temperature at the warm limit of their distribution range) minus the local habitat temperature actually experienced at a given location. This metric is useful to predict how much more warming species can tolerate before negative impacts are expected to occur. Here we set up a cross‐continental transplant experiment involving five regions distributed along a latitudinal gradient across Europe (43° N–61° N). Transplant sites were located in dense and open forests stands, and at forest edges and in interiors. We estimated the warming tolerance for 12 understory plant species common in European temperate forests. During 3 years, we examined the effects of the warming tolerance of each species across all transplanted locations on local plant performance, in terms of survival, height, ground cover, flowering probabilities and flower number. We found that the warming tolerance (ΔTniche) of the 12 studied understory species was significantly different across Europe and varied by up to 8°C. In general, ΔTniche were smaller (less positive) towards the forest edge and in open stands. Plant performance (growth and reproduction) increased with increasing ΔTniche across all 12 species. Our study demonstrated that ΔTniche of understory plant species varied with macroclimatic differences among regions across Europe, as well as in response to forest microclimates, albeit to a lesser extent. Our findings support the hypothesis that plant performance across species decreases in terms of growth and reproduction as local temperature conditions reach or exceed the warm limit of the focal species.

Abstract:The ability to acclimate to changing environmental conditions is essential for the fitness and survival of plants. Not only are seasonal differences challenging for plants growing in different habitats but, facing climate change, the likelihood of encountering extreme weather events increases. In order to better assess and respond to associated future challenges and risks it is important to understand the processes happening during acclimation. Previous studies of acclimation processes of Arabidopsis thaliana to changes in temperature and light conditions have revealed a multigenic trait comprising and affecting multiple layers of molecular organization. Here, a combination of experimental and computational methods was applied to study the effects of changing light intensities during cold acclimation on the central carbon metabolism of Arabidopsis thaliana leaves. Mathematical modeling, simulation and sensitivity analysis predicted an important role of hexose phosphate balance for stabilization of photosynthetic CO2 fixation. Experimental validation revealed a profound effect of temperature on the sensitivity of carbohydrate metabolism.

Abstract:This work examined the effects of elevated CO2 and temperature and water regimes, alone and in interaction, on the leaf characteristics [leaf area (LA), specific leaf weight (SLW), leaf nitrogen content (NL) based on LA], photosynthesis (light‐saturated net carbon fixation rate, Psat) and carbon storage in aboveground biomass of leaves (Cl) and stem (Cs) for a perennial reed canary grass (Phalaris arundinacea L., Finnish local cultivar). For this purpose, plants were grown under different water regimes (ranging from high to low soil moisture) in climate‐controlled growth chambers under the elevated CO2 and/or temperature (following a factorial design) over a whole growing season (May–September in 2009). The results showed that the elevated temperature increased the leaf growth, photosynthesis and carbon storage of aboveground biomass the most in the early growing periods, compared with ambient temperature. However, the plant growth declined rapidly thereafter with a lower carbon storage at the end of growing season. This was related to the accelerated phenology regulation and consequent earlier growth senescence. Consequently, the elevation of CO2 increased the Psat, LA and SLW during the growing season, with a significant concurrent increase in the carbon storage in aboveground biomass. Low soil moisture decreased the Psat, leaf stomatal conductance, LA and carbon storage in above ground biomass compared with high and normal soil moisture. This water stress effect was the largest under the elevated temperature. The elevated CO2 partially mitigated the adverse effects of high temperature and low soil moisture. However, the combination of elevated temperature and CO2 did not significantly increase the carbon storage in aboveground biomass of the plants.
