Abstract:A middle aged man was hospitalised in the state of acute renal failure with erythema nodosum lepromum. He was having progressive anaemia, oliguria, azotemia and impaired renal function. The outcome was fatal. Autopsy revealed smooth congested kidney with histopathological features of rapidly progressive - (crescentric) glomerulo-nephritis, presumably a result of immune-complex deposition from recurrent erythema nodosum leprosum episodes.

Abstract:This study was conducted to investigate the effectiveness of pulsed electric fields (PEFs), pulsed UV light (PL), and antimicrobial packaging (AP), either individually or combined, for inactivating bacteria and maintaining the quality of fruit juices. Apple juice samples were inoculated with Escherichia coli K-12 or native molds and yeasts (M&Y), treated with bench scale PEF and/or PL processing systems, and stored in glass jars with antimicrobial caps containing 10 mu L of carvacrol. The reductions in microbial populations and the physicochemical properties of juice samples were determined after treatments and during storage at 10 degrees C. The treatments were PL (5 to 50 s at 1.04 J/cm(2)/s), PEF (19, 23, and 30 kV/cm), PEF followed by PL (PEF+PL), PL followed by PEF (PL+PEF), and PEF+PL+AP. PEF treatments at 19 to 30 kV/cm (PEF19, PEF23, and PEF30) resulted in 2.0-, 2.6-, and 4.0-log reductions of E. coli, respectively, and PL treatments for 10 to 50 s (PL10, PL20, PL30, PL40, and PL50) resulted in 0.45-, 0.67-, 0.76-, 2.3-, and 4.0-log reductions, respectively. No significant differences (P > 0.05) were found between the combined PL20+PEF19 and PEF19+PL20 treatments; both treatments reduced E. coli K-12 populations to nondetectable levels (> 5-log reductions) after 7 days. Both PEF+PL and PEF+PL+AP treatments achieved > 5-log reductions of M & Y; however, juice samples subjected to PEF+PL+AP treatment had lower M&Y counts (2.9 log units) than did samples subjected to PEF+PL treatment (3.9 log units) after 7 days. No significant differences (P > 0.05) in pH, acidity, and total soluble solids were found among all samples after treatments. Increased PL treatment times reduced color a* and b* values, total phenolics, and carotenoids. This study provides information valuable to juice processors for consideration and design of nonthermal pasteurization of juice products. HIGHLIGHTS Combined treatments had additive or synergistic antimicrobial effects. Combined treatments achieved.5-log reductions of E. coli and molds and yeasts. Combined treatments did not significantly affect pH, acidity, or total soluble solids. Combined treatments slightly impacted color, carotenoids, and phenolic compounds.
Effect of an UV-C Light-Based Hurdle Strategy for Carrot-Orange Juice Processing on Candida parapsilosis Inactivation and Physiological State: Impact on Juice Sensory and Physicochemical Quality Parameters

Abstract:In the past two decades, novel preservation technologies have been investigated to achieve sufficient microbial reduction while improving the quality of food and beverages. The effect of UV-C light (UV-C, 1720 mJ/cm2) assisted by mild heat (H, 50 °C) on the inactivation and the physiological state of Candida parapsilosis ATCC 22019 in a carrot-orange juice was studied by flow cytometry. Additionally, the juice samples processed or not (control) by the single (UV-C; H) and combined (UV-C/H) treatments were analyzed for total polyphenol content (TPC), total antioxidant activity (TAA), color, turbidity, °Brix, pH, and pectin methylesterase units (PME) along 13–24-day storage (4 °C). Consumer-profiling studies were conducted on UV-C/H and a pasteurized juice. UV-C/H was highly effective in reducing C. parapsilosis by 5.5 log cycles, whereas the single UV-C and H treatments induced 2.9 and 3.9 log reductions, respectively. Flow cytometry revealed a shift with time from cells with intact membrane and esterase activity to those with permeabilized membrane and without esterase activity, which was significantly higher for UV-C/H samples (98.0%) compared with the single treatments (12.0–46.2%). UV-C/H preserved juice color, pH, °Brix, and turbidity of the juice, which exhibited equal TAA (0.7 mg/mL) and higher TPC (302.1 μg/mL) than the control (TAA = 0.7 mg/mL/TPC = 205.0 μg/mL), remaining constant throughout storage. PME decreased 42–45% for the H and UV-C/H treatments, respectively. A cluster sensory analysis revealed that one group of consumers showed a remarkable interest in the UV-C/H juice, while the conjoint study showed that it was perceived as very healthy. The CATA question determined that the UV-C/H processing prevented the juice from being perceived too particulate or with cooked or artificial taste, as happened with the pasteurized juice.

Abstract:Pasteurized sports drinks and other fruit-based beverages are susceptible to deterioration due to thermal processing ineffectiveness to inactivate certain spoilage microorganisms, like Alicyclobacillus acidoterrestris. This represents a major challenge for the beverage industry. The goals of this study were to: i) investigate the UV-C inactivation (annular thin film unit, actinometrical delivered fluence: 795-1270 mJ/cm2, 10-15 min, 20 °C, 1.8 L/h, Reh = 391-1067, recirculation mode operation) and the evolution during refrigerated storage of A. acidoterrestris ATCC 49025 spores and single or composite Escherichia coli ATCC 25922 in isotonic sports drinks (ISDs) made from orange (orange-ISD, UVT% = 81) or orange-banana-mango-kiwi-strawberry-lemon juices (multi-fruit-ISD, UVT% = 91), compared to a turbid orange-tangerine juice (OT juice, UVT% = 40); ii) assess the effect of pH, °Brix, A254nm, turbidity, colour and particle size of the ISDs and juice on microbial inactivation, iii) evaluate the evolution of native microbiota during cold storage, iv) investigate the Coroller, biphasic, Weibull, and Weibull-plus-tail models' ability to describe microbial inactivation and v) measure 5-hydroxymethylfurfural (HMF) formation. The modified biodosimetry method was used to calculate the germicidal UV-C fluences. Heat pasteurization (T-coil, 80 °C/6 min) was evaluated as the control treatment. UV-C was highly effective at inactivating E. coli as 4.1-5.1 and 4.5-5.6 log reductions were determined in the multi-fruit-ISD and orange-ISD, respectively, barely impacted by the background microbiota. No significant differences were recorded for the inactivation of E. coli in the UV-C and T-coil systems. Whereas, a significantly higher inactivation of A. acidoterrestris spores was achieved by UV-C (3.7-4.0 log reductions), compared to the negligible one achieved by the thermal treatment. Even though E. coli inactivation curves were similar in shape, UV-C was less effective when a cocktail of other E. coli strains was present. In comparison to the OT juice, the ISDs' inactivation kinetics were markedly different in shape, with a rapid decrease in population during the first minutes of treatment. The germicidal fluence (Hd biod) corresponding to A. acidoterrestris (19.1 mJ/cm2) was selected as it was higher than the one obtained for E. coli (11.0 mJ/cm2). UV-C induced 2.8- or 1.3 and 2.3- or 0.8 log-reductions of total aerobes or moulds and yeasts in the multi-fruit-ISD and orange-ISD, respectively. Compared to the other models, the Coroller and biphasic models showed a better fit and more accurate parameter estimates. UV-C-induced HMF production was not significant in the ISDs. The current study found that the UV-C treatment was more effective than typical heat pasteurization for inactivating A. acidoterrestris spores in isotonic drinks, following a similar trend for E. coli and native microbiota.
Stabilizing Methyl Ferulate with Nanosystem Enhances Its Antifungal Activity Against Alternaria Alternata in Vitro and in Vivo

Abstract:Postharvest fruit infection by Alternaria alternata results in significant food waste and economic losses. Methyl ferulate (MF) emerges as a safe and potent plant -derived agent against A. alternata , yet its effectiveness is hindered by poor water solubility, limiting its wide application in postharvest preservation. This study aimed to address this issue by utilizing naturally derived grapefruit essential oil (GEO) as the dissolution carrier for MF, followed by the preparation of a nanosystem to stabilize it through ultrasound -assisted methods. At 20 mg/mL MF loading, nanoemulsions (NEs) exhibited the smallest droplet size (15.66 nm), highest centrifugation stability (Ke of 2.32 %), and optimal storage stability (15.44 nm after 30 d at 4 degree celsius). Importantly, MF in the nanosystem (named MN) acts as an Ostwald ripening inhibitor and reduces the interfacial tension, which in turn benefits the stability of this system. In vitro , MN showed enhanced inhibition of A. alternata hyphal growth (an inhibitory rate in potato dextrose broth medium of 99.07 +/- 1.32 %) and a fourfold reduction in MIC compared to free MF. MN damaged A. alternata plasma membranes and induced reactive oxygen species (ROS) oxidative stress. In vivo , MN inhibited black spot disease and toxin production of A. alternata in pears. Additionally, MN incorporated into carboxymethylcellulose sodium (CMC) prolonged the shelf life of Korla pears and cherry tomatoes by delaying weight loss and maintaining their firmness and intrinsic nutrients. Therefore, this study broadened the application of MF, offering a viable approach for postharvest preservation of fruit infected by A. alternata .
A multidimensional evaluation of the effects of sweetener selection and UV-C treatment on orange juice and pectin-based confectionery gels

Abstract:Background: Consumers are seeking healthier alternatives to traditional confectioneries. They value the use of sugar replacers, more natural ingredients and/or environmentally friendly preservation technologies. UV-C light is considered an emerging alternative to thermal pasteurization that leaves no residue and requires minimal energy. The present study aimed to investigate the effect of novel sweetener combinations and juice UV-C assisted by mild heat treatment (UV-C/H) on the physicochemical, microbiological, morphological, rheological and sensory properties of orange juice pectin-based confectioneries stored at 5 ± 1 °C for 35 days. Results: For orange juice processing, UV-C/H (pilot-scale Dean-flow reactor; 892 mJ cm-2 ; 50 ± 1 °C) and thermal (T-coil, 80 °C; 6 min) treatments were used. Low-calorie confectionery gels were elaborated using the treated juices, low-methoxyl pectin and various sweetener combinations. UV-C/H and T-coil effectively inactivated juice native microbiota. The proposed formulations, derived from a previous Box-Behnken optimization study, included partial (F1: 3%-sucrose-S + 0.019%-rebaudioside-A-RA) or complete sucrose replacement (F2: 5.5%-erythritol-E + 0.019%-RA), and one control (C:10%-S). In general, the microbiota of the gels prepared with the UV-C/H or T-coil treated juices did not recover during storage. The physicochemical and mechanical parameters of the formulations were significantly influenced by the choice of sweetener and the duration of storage. The gel surface got smoother and had fewer holes when the sucrose level dropped, according to a scanning electron microscopy study. The UV-C/H-treated samples did not differ in acceptability, whereas the measured sensory attributes approached ideal levels. F1 and F2 showed distinctive temporal-dominance-of-sensations profiles, mainly dominated by sweetness and orange taste, respectively. However, consumers perceived sourness and astringency in C during consumption. Conclusion: The present study provides significant evidence in support of the development of confectionery gels F1 and F2 made from fruit juice treated by UV-C light assisted by mild heat and combinations of sucrose-alternative sweeteners. In terms of the properties investigated, these confectionery gels were comparable to, or even outperformed the full-sucrose option. © 2023 Society of Chemical Industry.

Abstract:The efficacy of combined UV-C radiation (39.6 J/L) and mild heat (65.0 ± 3.0 °C) on the inactivation of aerobic mesophilic and yeast/molds in grapefruit juice (GFJ) (pH 3.12, °Brix 9.6, 2500 NTU, absorption coefficient 42.9 cm−1) using a flow continuous reactor was investigated. Changes in microbial load, ascorbic acid, and total phenols content, antioxidant capacity, pH, acidity, °Brix, and color were evaluated during 28 days at 4.0 ± 1.0 °C. The spoilage microorganisms were inactivated due to the synergistic effect of both technologies, but the ascorbic acid (74%) and phenol (14%) contents were considerably reduced, with losses of antioxidant capacity (DPPH• = 19% and ABTS•+ = 16%). During storage, the ascorbic acid level and antioxidant capacity gradually decreased. On the other hand, the treated GFJ did not show microbial growth while the untreated GFJ reached more than 4 logs CFU/mL. Conversely, no changes (P < 0.05) in the physicochemical parameters were observed. Principal component analysis is discriminated among the antioxidant compounds and physicochemical properties of the untreated and treated GFJ during storage. Application of UV-C in a Teflon-coil reactor combined with mild heat resulted in longer microbial shelf stability in the GFJ and was able to maintain the physicochemical properties of the juice unchanged during 28 days of storage. Hence, this process can be effectively applied as a combined method to ensure microbial quality.

Abstract:The inhibition of Escherichia coli ATCC 25922 (E. coli), Staphylococcus aureus ATCC6538 (S. aureus), Salmonella Enteritidis ATCC 13076 (S. Enteritidis), and Listeria monocytogenes DSM12464 (L. monocytogenes) is one of the main aims of the food industry. This study was the first in which the use of ultrasound and microwave power were applied to optimize the values of the bioactive components, amino acids, and mineral compositions of tangerine juice and to inhibit Escherichia coli, Staphylococcus aureus, Salmonella Enteritidis, and Listeria monocytogenes. The response surface methodology (RSM) was used to describe the inactivation kinetics, and the effects of ultrasound treatment time (X1: 12–20 min), ultrasound amplitude (X2:60–100%), microwave treatment time (X3: 30–40 s), and microwave power (X4:200–700 W). The optimum parameters applied to a 5-log reduction in E. coli were determined as ultrasound (12 min, 60%) and microwave (34 s, 700 W). The optimum condition ultrasound–microwave treatment was highly effective in tangerine juice, achieving up to 5.27, 5.12, and 7.19 log reductions for S. aureus, S. Enteritidis, and L. monocytogenes, respectively. Ultrasound–microwave treatment increased the total phenolic compounds and total amino acids. While Cu, K, Mg, and Na contents were increased, Fe and Ca contents were lower in the UM-TJ (ultrasound–microwave-treated tangerine juice) sample. In this case, significant differences were detected in the color values of ultrasound–microwave-treated tangerine juice (UM-TJ) (p < 0.05). The results of this study showed that ultrasound–microwave treatment is a potential alternative processing and preservation technique for tangerine juice, resulting in no significant quality depreciation.

Abstract:This work explored the effect of ultraviolet-assisted ultrasound (US-UV) as an emerging non-thermal sterilization technology on mango juice in aspects of microbial growth and quality changes. The juice in the ice bath was subjected to US-UV treatment at different US powers (0-600 W) and times (0-40 min), and no pathogen bacteria could be detected after treatment, while the physicochemical features (particle size, suspension stability, color, content of total polyphenols, carotenoids, sugar, reducing sugar and protein) and antioxidant ability of treated juice was preserved or improved to some extent. Based on these results, we further validated its positive effects on the nutritional value (content of ascorbic acid and soluble dietary fiber, antioxidant ability) and quality parameters (titratable acid, sugar acidity, total soluble solids, rheological behavior, metal elements) of mango juice treated at the optimal US parameter (10 min, 600 W); Not only the inactivation of polyphenol oxidation enzyme, peroxidase and pectin methylesterase was achieved but also the treated juice has a significant different volatile profile compared with the fresh juice, which might offer the better color, texture, and smell. Importantly, through the HPLC-MSD-Trap-XCT (phenols) and UPLC-Q Exactive Orbitrap-MS (carotenoids) study, the US-UV treatment will not cause difference on compounds composition, but it was responsible for changes in content of individual compounds, especially the all-trans-β-carotene, became the main component of carotenoids in processed mango juice (increased from 43.72% to 75.15%, relative content), and the oxygenated carotenoids (xanthophylls) are highly sensitive to the US (reduced from 50.96% to 4.85%) while the carotenes show a strong resistance to the US (increased 49.04% to 95.15%). Thus, the overall safety and quality of mango juice were enhanced while the sensory characteristics remained stable, suggesting that this non-thermal combination sterilization processing may successfully be implemented in the commercial processing of mango juice.

Abstract:The inactivation of Escherichia coli is one of the major issues in the food industry. The present study focuses on the application of a combined microwave-ultrasound system for the optimization of the inactivation of Escherichia coli ATCC 25922 in an orange juice drink. Using response surface methodology (RSM), trials were planned with a Box–Behnken Design (BBD) to maximize the impact of microwave power (A: 300–900 W), microwave treatment time (B: 15–35 s), and time of ultrasound (C: 10–30 min) on E. coli inactivation. Analysis of variance (ANOVA) was carried out and E. coli inactivation was expressed with a mathematical equation depending on the factors. The results showed that both the microwave treatment time and the time of ultrasound were effective as independent variables in eliminating the E. coli strain. However, the effect of these two variables, ultrasound and microwave exposure time, in combination was significantly greater than when examined separately. RSM modeling determined that optimal treatment conditions include 900 W microwave power, 33 s microwave treatment time, and 20 min time of ultrasound to achieve an 8-log reduction of E. coli, constituting total inactivation. The results of this study showed that ultrasound-microwave treatment is a potential alternative processing method for an orange juice beverage.

Effect of Optimized UV-LED Technology on Modeling, Inactivation Kinetics and Microbiological Safety in Tomato Juice

Abstract:This research analyzed, optimized and modeled the inactivation kinetics of pathogenic bacteria (PB1: Escherichia coli O157:H7 and PB2: Listeria monocytogenes) and determined the microbiological safety of tomato juice processed by UV-LED irradiation and heat treatment. UV-LED processing conditions were optimized using response surface methodology (RSM) and were 90% power intensity, 21 min and 273–275 nm (251 mJ/cm2) with R2 > 0.96. Using the optimal conditions, levels of PB1 and PB2 resulted a log reduction of 2.89 and 2.74 CFU/mL, respectively. The Weibull model was efficient for estimating the log inactivation of PB1 and PB2 (CFU/mL). The kinetic parameter δ showed that 465.2 mJ/cm2 is needed to achieve a 90% log (CFU/mL) reduction in PB1 and 511.3 mJ/cm2 for PB2. With respect to the scale parameter p > 1, there is a descending concave curve. UV-LED-treated tomato juice had an 11.4% lower Listeria monocytogenes count than heat-treated juice on day 28 (4.0 ± 0.82 °C). Therefore, UV-LED technology could be used to inactivate Escherichia coli O157:H7 and Listeria monocytogenes, preserving tomato juice for microbiological safety, but studies are required to further improve the inactivation of these pathogens and analyze other fruit and vegetable juices.

Evaluation of ultrasound based sterilization approaches in terms of shelf life and quality parameters of fruit and vegetable juices

Abstract:The present work evaluates the performance of ultrasound based sterilization approaches for processing of different fruit and vegetable juices in terms of microbial growth and changes in the quality parameters during the storage. Comparison with the conventional thermal processing has also been presented. A novel approach based on combination of ultrasound with ultraviolet irradiation and crude extract of essential oil from orange peels has been used for the first time. Identification of the microbial growth (total bacteria and yeast content) in the juices during the subsequent storage and assessing the safety for human consumption along with the changes in the quality parameters (Brix, titratable acidity, pH, ORP, salt, conductivity, TSS and TDS) has been investigated in details. The optimized ultrasound parameters for juice sterilization were established as ultrasound power of 100 W and treatment time of 15 min for the constant frequency operation (20 kHz). It has been established that more than 5 log reduction was achieved using the novel combined approaches based on ultrasound. The treated juices using different approaches based on ultrasound also showed lower microbial growth and improved quality characteristics as compared to the thermally processed juice. Scale up studies were also performed using spinach juice as the test sample with processing at 5 L volume for the first time. The ultrasound treated juice satisfied the microbiological and physiochemical safety limits in refrigerated storage conditions for 20 days for the large scale processing. Overall the present work conclusively established the usefulness of combined treatment approaches based on ultrasound for maintaining the microbiological safety of beverages with enhanced shelf life and excellent quality parameters as compared to the untreated and thermally processed juices.

Abstract:Counteracting probiotic-induced physicochemical and sensory changes is a challenge in the development of probiotic beverages. The aim of the study is to apply ultrasound and microencapsulation for the attenuation of Limosilactobacillus reuteri DSM 17938 to avoid change in a probiotic tomato juice. Preliminarily, six ultrasound treatments were applied. Probiotic survival in acid environment (pH 2.5) and bile salts (1.5 g/l) after ultrasound treatment was also studied. The probiotic was inoculated in tomato juice in four forms: free cells (PRO-TJ), sonicated-free cells (US-TJ), untreated-microencapsulated (PRO-MC-TJ) and sonicated-microencapsulated cells (US-MC-TJ). Probiotic viability and pH were monitored during 28 days of storage at 4 and 20 °C. Sensory analysis was performed for PRO-TJ and US-MC-TJ sample (4 °C). Ultrasound (57 W for 6 min) did not affect cell survival and transitorily modulated probiotic acidifying capacity; it reduced probiotic survival in acidic environment but increased probiotic survival in bile salts solution. Ultrasound was effective in maintain pH value of tomato juice but only at 4 °C. Instead, microencapsulation with sodium-alginate leads to a more stable probiotic juice, particularly at 20 °C. Finally, probiotication slightly modified some sensory attributes of the juice. This study shows the potential of ultrasound and microencapsulation as attenuation strategies and highlights the need for process optimization to increase ultrasound efficacy.

Abstract:This study evaluated a combination of high-frequency ultrasound (HFU, 1 MHz, 1.6 W/cm2).and a food-grade antioxidant, propyl gallate (PG, 10 mM), to enhance inactivation rates of Listeria innocua and Escherichia coli O157:H7 in water and clarified apple juice. Treatment times ranged from 5 to 20 min. The study also assessed the potential mechanisms of synergistic interactions based on an evaluation of changes in bacterial permeability, morphology, and intracellular oxidative stress. Within 15 min of treatment time, HFU + PG significantly (reduced by 5.5 log CFU/mL, P 5 log CFU) and rapid inactivation (∼15 min) of inoculated model Gram positive and Gram negative bacteria in apple juice using a synergistic combination of propyl gallate and high frequency ultrasound. The synergistic interactions result in enhanced membrane damage and oxidative stress induction in bacteria. These results illustrate potential of the synergistic non-thermal thermal processing method for processing liquid beverages. Further studies are required for evaluating the scale up and optimization of the novel processing technology and enhancement in quality attributes of beverages.

Physiol. 22 l(1) : 246-250. 197 1 .-We studied spreading patterns of electric slow waves of circular muscle in longitudinal axis of cat colon. The colon, opened lengthwise, was pinned flat, serosa down, in a bath equilibrated with 957, OQ--5y0 CO2 at 36.5-37.5 C. Six volume-recording electrodes, put on circular muscle, exposed by removal of the mucosa, recorded slow waves simultaneously from six sites 5.8 mm apart. The first was just below the ileocecal junction. 1Ve determined proportion of time when slow waves were phase locked across all six electrodes, direction of spread and time delay while slow waves were phase locked, distance over which slow waves were phase locked, and oscillator frequency. During phase lock across all six points (67% of the time) slow waves spread orad 8.7y0, caudad 3.5y0, and split (orad proximally, caudad distally) 87.9y0 of the time. Apparent velocity orad, 2.6 & 1.1 (SD) mm/set, was less than caudad, 3.6 =t 2.1 (SD) mm/set, (P = 0.005). Distance over which phase lock occurred varied from 5.8 to 28.8 mm. Frequency was 3.12 =t 1.62 (SD) cycles/min. Distance of phase lock and direction of phase lag vary and may affect fecal flow through effects on integration and polarization of wall movements. Different apparent velocities in the two directions suggest rectification in coupling of slow-wave oscillators.
Sono-photodynamic inactivation of Escherichia coli and Staphylococcus aureus in orange juice

Abstract:Efficiency of blue (462 ± 3 nm) light emitting diode (LED) illumination to inactivate Escherichia coli and Staphylococcus aureus in the presence of exogenous photosensitizer (curcumin) was studied in freshly squeezed orange juice. Further, the combinational effect of ultrasound (US), photosensitizer (PS) and blue light (BL) on inactivation of microbes was evaluated. The effect of process parameters such as concentration of PS, US and volume of the juice on E. coli and S. aureus inactivation was also investigated. The US alone and PS + BL treatments resulted in 3.02 ± 0.52 and 1.06 ± 0.13 log reduction of E. coli; 0.18 ± 0.14 and 2.34 ± 0.13 log reduction of S. aureus, respectively. The combination of PS + US + BL treatment at optimized conditions resulted in 2.35 ± 0.16 log reduction of S. aureus. An additive effect on the inactivation of E. coli (4.26 ± 0.32 log reduction) was observed with PS + US + BL combination treatment. The US treatment showed significant change in cloud value, colour and browning index of orange juice. The combinational non-thermal processes (PS + BL and PS + US + BL) did not have any significant effect on total phenolic content, total flavonoid content, and hesperidin content of the orange juice. However, these processes affected ascorbic acid content and antioxidant activity negatively. Thus, this study indicated that photodynamic inactivation of E. coli and S. aureus using LED-based photosensitization in fruit juices could be a potential method for microbial inactivation. Nevertheless, the effect on quality parameters needs to be considered while optimizing the process.

Abstract:Penicillium digitatum is the causal agent for citrus green mold which generates 90% of post-harvest losses in Mediterranean countries. The use of a green seaweed extract to fight this fungal pathogen could represent an ecofriendly alternative to chemical fungicides. We propose to formulate an aqueous extract from Ulva lactuca (8.3%) in an emulsion for the preventive treatment of oranges. New biobased emulsifiers, alkyl aconitates, were synthesized according to a green process and were characterized by their hydrophilic and lipophilic balance (HLB) and their critical micelle concentration (CMC). These mono-, di and tri-dodecyl aconitates (8%) were effective in stabilizing an oil-in-water emulsion including the natural U. lactuca extract. The viscosities of resulting emulsions helped to form a film on the orange surface. Several in vitro microbiological tests towards Penicillium digitatum showed interesting anti-fungal activities for the new ingredients, as well as for the resulting emulsions. In-vivo tests also confirmed that mono-, di- and tri-docedyl aconitates could contribute to limit the development of P. digitatum on peel surface. Thus, the compositions of two ecofriendly emulsions combining a natural extract and a biobased emulsifier seem adapted to improve oranges preservation.

Inactivation of Bacillus Subtilis and Quality Assurance in Chinese Bayberry (myrica Rubra) Juice with Ultrasound and Mild Heat

Abstract:Combined ultrasound and mild heat (UH) and traditional thermal pasteurization (TP) were employed to inactivate thermoacidophilic B. subtilis suspended in Chinese bayberry juice. Flow cytometry was used to evaluate different action mechanisms of these two treatments. The changes of physicochemical properties, anthocyanin content, and volatile components were also studied to assess the effects on juice quality. Results showed a 5-log reduction was achieved in 36.91 min by TP, 23.06 min by UH55 degrees C (400W), 14.18 min by UH63 degrees C (200w), 9.59 min by UF63 degrees C (400W), and the inactivation date had a high goodness of fit with Weibull model, exhibiting a fast drop in first and a slow decline at last. Compared to the TP, UH treatment was not only able to cause lethal effect by compromising membrane integrity, inactivating esterase activity, but also destroying nucleic acids through the flow cytometry analysis. The pH and soluble solids content of bayberry juice had no significant changes with different treatments, while the light transmittance of UH groups during storage was higher, the color and anthocyanin retention were better than TP treatment. The GC-MS analysis indicated UH treatment can significantly inhibit the decline of key aromatic substances of bayberry juice, including 1-hexanol, (Z)-3-hexenol, (Z)-3-nonenol and dihydro-S-pentyl-2(3H)-furanone.
Biological and physical approaches to improve induced resistance against green mold of stored citrus fruit

Abstract:Health and environmental concerns have point out the need to improve or change several manufacturing steps in the food chain. In this context particular attention should be given to the technologies involved in fruits and vegetables production. Nearly all fresh fruit and vegetables are subjected to different periods of storage and/or shelf-life before of their consumption. This implies the need to protect the commodities from microbial spoilage. Some Citrus species (e.g. lemon and grapefruit) may be stored for several months before consumption and then post-harvest treatments are essential to contain green (Penicillium digitatum) and blue (P. italicum) moulds. Alternative approaches to chemicals usually have a lower efficacy in containing rots but fulfill the consumer's expectation. Among the alternative strategies, the improvement of host natural resistance is promising. In this regard, we report some results concerning the use of biotic (yeast) and abiotic agents as inducers of phytoalexin (i.e. scoparone and/or scopoletin) accumulation in Citrus rind and its importance in the control of fungal decay. In all experiments the inducers were applied on fruits before or 24 h after inoculation with P. digitatum and the rot severity was monitored 7 days later. The accumulation of phytoalexins was monitored according to a standard methodology by HPLC. In all experiments a positive correlation was found between increase of the phytoalexin scoparone in host tissue and reduction of decay.
