Abstract:Objective To develop a real-time fluorescence PCR assay rapidly to detect the genes encoding staphylococcal enterotoxins A and D and to investigate the clinical significance of staphylococcal enterotoxins.Methods The genes of SEA and SED were selected as target genes of staphylococcal enterotoxins A and D, and femB gene as a specific genomic marker for S. aureus; The primers and TaqMan probes were designed and synthesized. 487 stool samples of foodborne illness patients were investigated by real-time fluorescence PCR.Results The sensitivity of the assay for Staphylococcus aureus and enterotoxins A was 1.0×10~2copies, but the sensitivity of enterotoxins D was 1.0×10~3 copies. Among 487 samples, 72 S. aureus strains were found; 11 of these strains showed the production of enterotoxins A; 9 strains were positive for enterotoxins D, only one strain was positive for enterotoxins A and D.Conclusion The TaqMan PCR would be a rapid and sensitive method to detect the staphyloccal enterotoxins A and D, it would be well suited for screening large numbers of samples at the same time. Otherwise, the enterotoxins is one of causes of foodborne illness.

Abstract:The aims of this study were (i) to evaluate the performance of the Assurance ® GDS method combining immunomagnetic separation and real-time PCR for STEC detection in pooled samples (up to 375g) of vegetables and meat, and (ii) to compare its performances to that of the reference method ISO/TS-13136:2012 (25 g sample size) in artificially contaminated samples. The alternative Assurance ® GDS method includes enrichment in proprietary broth at 41.5°C for 10 h followed by primary screening of TOP 7 STEC containing eae , stx genes and O157:H7 markers using MPX-Top 7 (IMS+PCR), secondary screening for serogroup identification using MPX-ID/EHEC-ID (IMS+PCR). And finally, cultural confirmation from same IMS beads on two selective agars is set up. For reference method, the enrichment was performed in BPW at 41.5°C for 18 h. A total of 120 samples of meat and vegetables, including 20 uninoculated and 100 samples spiked with stressed cells (<12 CFU/25 or 375g) of STEC, were analyzed using both methods. Our results showed that the Assurance ® GDS method LOD 50 ranged from 0.12 CFU/375 to 0.522 CFU/375 g and the LOD 95 from 0.518 CFU/375 to 2.257 CFU/375 g. These data are similar to the LOD50 data of the reference method.

Abstract:ABSTRACT Shiga toxin-producing Escherichia coli (STEC) strains are commonly found in the intestine of ruminant species of wild and domestic animals. Excretion of STEC with animal feces results in a broad contamination of food and the environment. Humans get infected with STEC through ingestion of contaminated food, by contact with the environment, and from STEC-excreting animals and humans. STEC strains can behave as human pathogens, and some of them, called enterohemorrhagic E. coli (EHEC), may cause hemorrhagic colitis (HC) and hemolytic-uremic syndrome (HUS). Because of the diversity of STEC types, detection strategies for STEC and EHEC are based on the identification of Shiga toxins or the underlying genes. Cultural enrichment of STEC from test samples is needed for identification, and different protocols were developed for this purpose. Multiplex real-time PCR protocols (ISO/CEN TS13136 and USDA/FSIS MLG5B.01) have been developed to specifically identify EHEC by targeting the LEE (locus of enterocyte effacement)-encoded eae gene and genes for EHEC-associated O groups. The employment of more genetic markers ( nle and CRISPR) is a future challenge for better identification of EHEC from any kinds of samples. The isolation of STEC or EHEC from a sample is required for confirmation, and different cultivation protocols and media for this purpose have been developed. Most STEC strains present in food, animals, and the environment are eae negative, but some of these strains can cause HC and HUS in humans as well. Phenotypic assays and molecular tools for typing EHEC and STEC strains are used to detect and characterize human pathogenic strains among members of the STEC group.

Abstract:AIMS: The verotoxigenic Escherichia coli (VTEC) serotype most commonly associated with verotoxin (VT) production is O157:H7, but other serotypes have also been implicated in food-borne illness. These serotypes exhibit much greater genetic and biochemical diversity than E. coli O157:H7, making screening for all VTEC difficult. Here we describe development and testing of novel multi-analyte antibody-based dipstick methods for presumptive detection of VTEC cells and VTs, including non-O157 serotypes.METHODS AND RESULTS: The dipsticks are formatted as paddle-style and lateral flow devices. Test materials included raw milk, minced beef, apple juice and salami, spiked with VTEC. Prototype paddle dipsticks gave 47 of 48 E. coli O157-positive samples correct, and, simultaneously, 27 of 31 O26-positive samples correct, across the four food types. Prototype lateral flow dipsticks gave 12 of 12 E. coli O157-positive milk samples correct and, simultaneously, 28 of 28 positive VT samples correct.CONCLUSIONS: This work demonstrates that simple and rapid detection of more than one VTEC characteristic (toxin production and type, serogroup) is possible in a single dipstick test device, directly from a food enrichment culture.SIGNIFICANCE AND IMPACT OF THE STUDY: The development of simple easy-to-use rapid methods for simultaneous detection and preliminary characterization of VTEC will enable the risk presented by all VTEC to be more thoroughly assessed (e.g. in surveillance studies, outbreak investigations).

Abstract:Escherichia coli O157:H7 is an important foodborne pathogen and has been implicated in numerous food poisoning outbreaks worldwide. Although several microbiological and molecular methods have been developed to detect E. coli O157:H7, the difficulty to rapidly detect low levels of the foodborne bacteria persists. Here, the optimization of a filtration technique to concentrate and rapidly detect E. coli O157:H7 was conducted. Using homogenates prepared from freshly cut lettuce and cabbage samples, the E. coli O157:H7 concentration efficiencies of seven membrane filters were compared. Mixed cellulose ester (MCE) and polyvinylidene difluoride (PVDF) filters demonstrated the highest bacterial recoveries. In addition, the optimal E. coli O157:H7 detachment method from MCE filters after filtration was investigated. Tapping for 80 s was demonstrated to be the most effective method for detaching bacteria from the filters. Further, the possibility of the rapid detection of low levels of E. coli O157:H7 in lettuce and cabbage was evaluated using real-time polymerase chain reaction after bacterial concentration using MCE and PVDF filters. The use of MCE filters enabled the detection of 10° CFU/g (5 CFU/g) of E. coli O157:H7 within 2 hr without microbial enrichment culture. Therefore, concentration by filtration can be used for the rapid detection of low levels of foodborne pathogens. PRACTICAL APPLICATION: The modified method, which has been verified in this study, has been optimized to reduce the analysis time and to detect very low concentrations of E. coli O157:H7 within 2 hr. All these detection systems have a direct economic impact on the food analysis of producers, health authorities, or third-party laboratories.

Abstract:In this study, we describe the development of an automated immunomagnetic separation device combined with real-time polymerase chain reaction (PCR) for detecting foodborne bacteria. Immunomagnetic separation (IMS) is a well-known method for the separation and concentration of target bacteria from a large volume of food samples. Magnetic beads functionalized with an antibody provide selectivity for target bacteria such as Escherichia coli O157:H7. Moreover, compared to conventional methods, real-time PCR enables high-sensitivity detection of target bacteria. The method proposed in this study involves three steps: (1) pre-enrichment, (2) automated IMS and concentration of target bacteria, and (3) detection of target bacteria by real-time PCR. Using food samples with a working sample volume as large as 250 mL, the whole process only requires 3 h. As a result, target bacteria in the range of 101–102 colony-forming units per mg or g of sample can be detected in food samples, such as milk, ground beef, and cabbage, by using the proposed approach. We anticipate that the automated IMS system combined with real-time PCR will contribute to the development of a fully automated system for detecting foodborne bacteria and serve as a multi-tester for a variety of bacterial strains in the capacity of a sample-to-answer device in the near future.

Abstract:Enterotoxigenic E.coli (ETEC), enteropathogenic E.coli (EPEC), enterohemorrhagic E.coli (EHEC), and enteroinvasive E.coli (EIEC) are the major E.coli of causing diarrhea and threaten food safety and human health. Therefore, the development of a method to detect the four E.coli simultaneously is of great importance. In this study, four pairs of specific primers were designed according to the gene sequences of ETEC LT, EPEC bfpA, EHEC O-antigen, and EIEC invasion plasmid. A multi-PCR method to detect four kinds of diarrheogenic E.coli was established via optimizing the reaction conditions in single PCR process without cross reaction for each other was detected. The multi-PCR method has good specificity and sensitivity, and 24 bacteria strains were detected using the method, all four kinds of diarrheogenic E.coli were PCR positive result. In practice, 15 positive samples were detected from 124 pieces of meat, dairy products and artificially contaminated samples, which accorded with the testing result by GB (GB 4789.6-1994). The results indicated that the multi-PCR assay established in this study could be applied in rapid detection for major diarrheogenic ETEC, EPEC, EHEC, and EIEC singly and/or mixed infection diagnoses and food safety risk assessment, it has good practicality.

Abstract:In order to rapidly screen for the virulence factor that produces pathogenic Escherichia coli in food, we have developed multiplex polymerase chain reaction (PCR) assays. The multiplex PCR assays detect 4 pathogenic genes of enterohaemorrhagic E. coli (EHEC), enterotoxigenic E. coli (ETEC), enteropathogenic E. coli (EPEC), and enteroinvasive E. coli (EIEC). This allows for the generation of specific fragments 150, 179, 218, 401, 465, 584, and 881 bp for VT1, ST, LT, 16S rRNA, inV, VT2, and eaeA genes, respectively. The detection limit of 3 log CFU/mL for eaeA, LT, VT1, VT2, 4 log CFU/mL for inV, 6 log CFU/mL for ST by single PCR, while 5 log CFU/mL for VT1, VT2, 6 log CFU/mL for eaeA, LT, 7 log CFU/mL for ST, inV by multiplex PCR. This optimized detection method of pathogenic E. coli can be used as supportive data to revise the microbiological analytical manuals for the Korean Food Code.

Abstract:Ignition of explosive electron emission centers (ectons) is a great problem if operating voltages are rather low and vacuum conditions are clean. However, dense enough plasma adjoining directly to the cathode surface promotes new ectons to be ignited. The idea of the present work is an attempt of using laser ablation plasma to stimulate ignition of ectons. For tests, a gap formed by the ring anode and a single-wire cathode was located at the distance of about 1 cm from a target. High-voltage rectangular pulses of the amplitude 25–40 kV and the rise time about 3–4 ns were applied to the cathode synchronously with laser pulses. The paper presents results of experiments that are the ablation plasma parameters near the cathode and the statistics of successful ignition of ectons. As well, theoretical analysis of ecton initiation under actual experimental conditions is made and presented.

Abstract:Eighty strains of pathogenic Escherichia coli, representing each of the major diarrheal disease-causing groups, were examined by direct enzyme-linked immunosorbent assay (ELISA) for the presence of proteins associated with a 60-MDa plasmid from E. coli serotype O157:H7. Antiserum specific for plasmid-encoded proteins was prepared by immunizing a rabbit with a wild-type E. coli O157:H7 strain (strain 7785) and absorbing the serum with a plasmid-cured derivative (strain 2-45). Use of this antiserum in Western immunoblot analysis detected two proteins of 82 and 92 kDa in strain 7785 but not in strain 2-45. All 16 wild-type E. coli O157:H7 strains and all 10 Shiga-like toxin (SLT)-producing E. coli strains of serotypes other than O157 were ELISA positive. Thirteen of 14 enterotoxigenic and all of 24 enteroinvasive E. coli strains were ELISA negative, as were all of 16 E. coli strains isolated from healthy persons. Of 16 traditional enteropathogenic E. coli (EPEC) serotypes, 10 were ELISA positive, including 10 of 12 strains carrying the EPEC adherence factor gene. Absorption of the serum with an EPEC adherence factor-positive EPEC eliminated EPEC reactivity. This study demonstrates that two plasmid-mediated proteins are common to E. coli O157:H7 strains and to SLT-producing strains of other serotypes. Detection of these proteins by ELISA provides a sensitive and specific screening test for identifying SLT-producing E. coli of both O157 and non-O157 serotypes. Identification of the cross-reactive proteins found in EPEC could provide the basis for a single assay to detect both EPEC and SLT-producing E. coli.

Abstract:Shiga toxin (Stxs)-producing enterohaemorrhagic Escherichia coli (EHEC) and Shigella dysenteriae serotype 1 are major causative agents of severe bloody diarrhea (known as hemorrhagic colitis) and hemolytic uremic syndrome (HUS) associated with extraintestinal complications such as acute renal failure and neurologic impairment in infected patients under 9 years of age. Extreme nephrotoxicity of Stxs in HUS patients is associated with severe outcomes, highlighting the need to develop technologies to detect low levels of the toxin in environmental or food samples. Currently, the conventional polymerase chain reaction (PCR) or immunoassay is the most broadly used assay to detect the toxin. However, these assays are laborious, time-consuming, and costly. More recently, numerous studies have described novel, highly sensitive, and portable methods for detecting Stxs from EHEC. To contextualize newly emerging Stxs detection methods, we briefly explain the basic principles of these methods, including lateral flow assays, optical detection, and electrical detection. We subsequently describe existing and newly emerging rapid detection technologies to identify and measure Stxs.

Abstract:Background: Due to the diversity of Shiga toxin-producing Escherichia coli (STEC) isolates, detecting highly pathogenic strains in foodstuffs is challenging. Currently, reference protocols for STEC rely on the molecular detection of eae and the stx1 and/or stx2 genes, followed by the detection of serogroup-specific wzx or wzy genes related to the top 7 serogroups. However, these screening methods do not distinguish between samples in which a STEC possessing both determinants are present and those containing two or more organisms, each containing one of these genes. This study aimed to evaluate ecf1, Z2098, Z2099, and nleA genes as single markers and their combinations (ecf1/Z2098, ecf1/Z2099, ecf1/nleA, Z2098/Z2099, Z2098/nleA, and Z2099/nleA) as genetic markers to detect potentially pathogenic STEC by the polymerase chain reaction (PCR) in 96 animal samples, as well as in 52 whole genome sequences of human samples via in silico PCR analyses. Results: In animal isolates, Z2098 and Z2098/Z2099 showed a strong association with the detected top 7 isolates, with 100% and 69.2% of them testing positive, respectively. In human isolates, Z2099 was detected in 95% of the top 7 HUS isolates, while Z2098/Z2099 and ecf1/Z2099 were detected in 87.5% of the top 7 HUS isolates. Conclusions: Overall, using a single gene marker, Z2098, Z2099, and ecf1 are sensitive targets for screening the top 7 STEC isolates, and the combination of Z2098/Z2099 offers a more targeted initial screening method to detect the top 7 STEC isolates. Detecting non-top 7 STEC in both animal and human samples proved challenging due to inconsistent characteristics associated with the genetic markers studied.

Abstract:The polymerase chain reaction (PCR) was used to amplify DNA sequences from the malB operon of Escherichia coli. All E. coli strains tested yielded the specific DNA fragment. No amplification products were obtained with other Enterobacteriaceae. E. coli strains which produce enterotoxins were identified with additional primer pairs specific for the genes coding for the heat-labile toxin type I (LTI) and the heat-stable toxin type I (STI). Amplification products were identified by DNA-DNA hybridization. Alternatively, restriction endonuclease analysis was used for identification and to distinguish between different alleles of the enterotoxin genes. The detection limit was 10 bacteria. The PCR systems were validated by testing 27 E. coli of known enterotoxigenic properties. The PCR results were consistent with factual toxin production as determined by immunoassays. In addition, 58 E. coli strains isolated from soft cheese and mayonnaise were analyzed by PCR. One strain from a cheese sample was found to have the genetic information for STI production. This strain produced STI as determined by enzyme-linked immunosorbent assay.

Abstract:✓ Two cases of intracranial false aneurysm following penetrating orbitofacial wounds are reported. These were the only false aneurysms identified in a series of 2187 penetrating wounds of the brain from Vietnam. Three similar false aneurysms have been reported previously, and these also were associated with orbitofacial penetration. The unusual incidence of this type of arterial injury secondary to penetrating orbitofacial wounds suggests the value of early arteriographic evaluation.

Abstract:Escherichia coli (E. coli) O157: H7 is one of the most common foodborne pathogens that can be transmitted through contaminated food and water sources, posing a serious threat to food safety and public health worldwide. The rapid and accurate diagnosis of E. coli O157: H7 infection is essential for effective control and management of disease epidemics originating from food and water sources. In this study, we used immunomagnetic separation (IMS) technology to magnetically enrich E. coli O157: H7, further amplify the target gene rfbE using the method combining improved propidium monoazide (PMAxx) and loop-mediated isothermal amplification (LAMP) (PMAxx-LAMP), and detect viable E. coli O157: H7 in lettuce by the nucleic acid lateral flow strip (NALFS) method. We used IMS to capture and enrich the target bacteria, effectively eliminating the interference of the food matrix and improving the detection limit in lettuce samples. PMAxx treatment eliminated the false-positive results from dead bacteria and, in combination with LAMP-NALFS, detected viable E. coli O157: H7 accurately. The IMS-PMAxx-LAMP-NALFS assay allowed us to detect viable E. coli O157: H7 at concentrations as low as 81 CFU/g in artificially contaminated lettuce samples within 2 h without any heavy and costly instruments. Thus, this assay provides a simple, rapid, and low-cost tool for the on-site detection of pathogenic bacteria, especially with limited resources.

Abstract:Shiga-toxin producing Escherichia coli are considering as important emerging food-born bacterial pathogens. Among these kinds of Escherichia coli, serotype O157:H7 is the cause of sporadic and epidemic human cases of hemorrhagic colitis, which can lead to severe life threatening hemolytic uremic syndrome. Majority of cases are associated with ingestion of food or water contaminated with Escherichia coli O157:H7. This serotype is identified in 1982, and afterward the different pathogenic genes responsible for its virulence are sequenced. As the organism has been involved in many outbreaks of disease in human and animals, it is essential to develop a rapid, reliable and reproducible method of detection. In this review, different deoxyribonucleic acid based genotyping and detection methods are discussed. Genomic techniques like those that pulse field gel electrophoresis and restricted fragment length polymorphism are reviewed and Polymerase Chain Reaction variants are addressed along with some emerging technology like biosensor and microarray. The advantage and disadvantages of each described method are discussed throughout the review. Since each technique has its own merit and demerit, the decision for selection of E. coli O157:H7 detection technique will depend mainly on the balance between several factors such as the high speed, sensitivity, specificity, discrimination power, and availability of proper equipment’s and skilled man power. Thus, further research is recommended to improve molecular based detection techniques of E. coli O157:H7 in order to make the detection of this pathogen effective and specific in various regional and reference laboratory.

Abstract:Escherichia coli O157:H7 and certain non-O157 Shiga toxin-producing Escherichia coli (STEC) serogroups have emerged as important public health threats. The development of methods for rapid and reliable detection of this heterogeneous group of pathogens has been challenging. GeneDisc real-time PCR assays were evaluated for detection of the stx(1), stx(2), eae, and ehxA genes and a gene that identifies the O157 serogroup followed by a second GeneDisc assay targeting serogroup-specific genes of STEC O26, O45, O91, O103, O111, O113, O121, O145, and O157. The ability to detect the STEC serogroups in ground beef samples artificially inoculated at a level of ca. 2-20 CFU/25 g and subjected to enrichment in mTSB or buffered peptone water (BPW) was similar. Following enrichment, all inoculated ground beef samples showed amplification of the correct set of target genes carried by each strain. Samples inoculated with STEC serogroups O26, O45, O103, O111, O121, O145, and O157 were subjected to immunomagnetic separation (IMS), and isolation was achieved by plating onto Rainbow agar O157. Colonies were confirmed by PCR assays targeting stx(1), stx(2), eae, and serogroup-specific genes. Thus, this work demonstrated that GeneDisc assays are rapid, sensitive, and reliable and can be used for screening ground beef and potentially other foods for STEC serogroups that are important food-borne pathogens worldwide.

Abstract:Foodborne pathogens are a significant cause of illness, and infection with Shiga toxin-producing Escherichia coli (STEC) may lead to life-threatening complications. The current methods to identify STEC in meat involve culture-based, molecular, and proteomic assays and take at least four days to complete. This time could be reduced by using long-read whole-genome sequencing to identify foodborne pathogens. Therefore, the goal of this project was to evaluate the use of long-read sequencing to detect STEC in ground beef. The objectives of the project included establishing optimal sequencing parameters, determining the limit of detection of all STEC virulence genes of interest in pure cultures and spiked ground beef, and evaluating selective sequencing to enhance STEC detection in ground beef. Sequencing libraries were run on the Oxford Nanopore Technologies' MinION sequencer. Optimal sequencing output was obtained using the default parameters in MinKNOW, except for setting the minimum read length to 1 kb. All genes of interest (eae, stx1, stx2, fliC, wzx, wzy, and rrsC) were detected in DNA extracted from STEC pure cultures within 1 h of sequencing, and 30× coverage was obtained within 2 h. All virulence genes were confidently detected in STEC DNA quantities as low as 12.5 ng. In STEC-inoculated ground beef, software-controlled selective sequencing improved virulence gene detection; however, several virulence genes were not detected due to high bovine DNA concentrations in the samples. The growth enrichment of inoculated meat samples in mTSB resulted in a 100-fold increase in virulence gene detection as compared to the unenriched samples. The results of this project suggest that further development of long-read sequencing protocols may result in a faster, less labor-intensive method to detect STEC in ground beef.

Abstract:Investigation of the specificity of an enzyme-linked immunosorbent assay (ELISA) for detection of Escherichia coli O157:H7 in raw meats (EHEC-Tek; Organon Teknika Corp.) revealed that the target antigens of the detection reagent, monoclonal antibody 4E8C12, were present in numerous serotypes of E. coli and that their ELISA reactivity was influenced by bile salts, acriflavine, and heat. The specificity of the ELISA was improved by a modified test protocol incorporating immunocapture.
