Abstract:BACKGROUND Prolonged corticosteroid therapy increases the risk of osteoporosis and fracture. We studied whether corticosteroid-induced osteoporosis could be prevented by treatment with calcium, calcitriol (1,25-dihydroxyvitamin D3), and calcitonin. METHODS One hundred three patients starting long-term corticosteroid therapy were randomly assigned to receive 1000 mg of calcium per day orally and either calcitriol (0.5 to 1.0 microgram per day orally) plus salmon calcitonin (400 IU per day intranasally), calcitriol plus a placebo nasal spray, or double placebo for one year. Data on treatment efficacy were available for 92 of these patients. Bone density was measured every four months for two years by photon absorptiometry. There were no significant differences between groups with respect to age, underlying disease, initial bone density, or corticosteroid dose during the first year. RESULTS Calcitriol (mean dose, 0.6 microgram per day), with or without calcitonin, prevented more bone loss from the lumbar spine (mean rates of change, -0.2 and -1.3 percent per year, respectively) than calcium alone (-4.3 percent per year, P = 0.0035). Bone loss at the femoral neck and distal radius was not significantly affected by any treatment. In the second year, lumbar bone loss did not occur in the group previously treated with calcitonin plus calcitriol (+0.7 percent per year), but it did occur in the group given calcium alone (-2.3 percent per year). The calcitriol group also lost lumbar bone (-3.6 percent per year) but received more corticosteroid in the second year than the other two groups. CONCLUSIONS Calcitriol and calcium, used prophylactically with or without calcitonin, prevent corticosteroid-induced bone loss in the lumbar spine.
Grape Skin Extract Inhibits Mammalian Intestinal Α-Glucosidase Activity and Suppresses Postprandial Glycemic Response in Streptozocin-Treated Mice

Abstract:Intestinal α-glucosidases are the key enzymes responsible for starch digestion and absorption and their inhibition has been proven effective in both preventing and treating diabetes through improvement of postprandial hyperglycaemia. This study, for the first time, identified that a Norton grape skin extract (GSE) significantly inhibited mammalian intestinal α-glucosidases but not other digestive enzymes including structurally relevant pancreatic α-amylase. Norton GSE inhibited rat intestinal α-glucosidases through a competitive mode with an IC50 of 0.384mg/ml. Further animal study revealed that the oral intake of Norton GSE (400mg/kg) significantly reduced postprandial blood glucose by 30.9% in the streptozocin-treated male C57BL/6J mice following starch challenge. These findings suggest that Norton GSE may have a unique property of suppressing postprandial blood glucose through a mechanism involving the inhibition of α-glucosidases, thereby providing a novel dietary opportunity for diabetes management.
Enhancement of 1-deoxynojirimycin content and α-glucosidase inhibitory activity in mulberry leaf using various fermenting microorganisms isolated from Korean traditional fermented food

Abstract:Mulberry leaf is known to be effective in inhibiting increases in blood glucose levels in diabetic patients. 1-Deoxynojirimycin (DNJ), a α-glucosidase inhibitor, is the major effective component in mulberry leaf. The aim of this study was to evaluate the level of DNJ in mulberry leaves fermented by various microorganisms, including lactic acid bacteria, yeast, and the bacillus isolated from Korean traditional fermented soybean food, and to assess the inhibitory effect of fermented mulberry leaf powder extract (FMLE) on α-glucosidase activity. The DNJ content in unfermented mulberry leaf powder extract (UFMLE), the control, was determined to be 53.2 ± 1.5 mg/100 g (dry weight eq.). Lactobacillus plantarum, Zygosaccharomyces rouxii, Wickerhamomyces anomalus, and Bacillus subtilis remarkably increased the DNJ levels in mulberry leaves by 1.2, 1.5, 2.0, and 1.8-fold, respectively, compared to the control. The inhibitory effects of the FMLEs on α-glucosidase were also examined at various concentrations, and their α-glucosidase inhibitory effects tended to be higher than that in UFMLE. These findings suggest that Korean traditional fermenting microorganisms can be used as starter strains for the development of DNJ-enhanced food products, and that FMLEs have the potential to be used in various functional foods for the prevention of diabetes.

Abstract:This study identified vitexin as a potent inhibitor of α‐glucosidase (AG) in mulberry leaves, with an IC50 value of 105.50 ± 1.30 μg/mL. The inhibitory mechanism involved competitive inhibition and hydrogen bonding between vitexin and AG, with static quenching driven by entropy. Furthermore, the study found that vitexin and 1‐deoxynojirimycin (DNJ) synergistically inhibit AG activity. These findings provide insight into the potential use of vitexin‐DNJ complexes as functional AG inhibitors. Mulberry (Morus alba L.) leaf can effectively inhibit the digestion of starchy foods, and α‐glucosidase (AG) is its main target. This study employed an untargeted metabolomics approach combined with molecular docking to identify AG inhibitors. Subsequently, inhibition kinetics, fluorescence spectroscopy, and interaction force analysis were utilized to investigate the inhibitory mechanism. Results indicated that vitexin exhibited significant reversible inhibition of AG through competitive inhibition, with an IC50 value of 105.50 ± 1.30 μg/mL. Molecular docking revealed hydrogen bonding as the main interaction force between vitexin and AG, and quenching mechanism analysis showed that they underwent static quenching driven by entropy. The results of the combined experiment showed that 1‐deoxynojirimycin (DNJ), a known bioactive component in mulberry, has a synergistic effect of inhibiting AG activity with vitexin. This study elucidates the potential mechanism of vitexin in inhibiting AG activity and provides theoretical evidence for utilizing vitexin‐DNJ complexes as functional components of AG inhibitors.
Inhibition Mechanism of Α-Glucosidase by Three Geranylated Compounds: Kinetic, Spectroscopic and Molecular Docking

Abstract:alpha-Glucosidase inhibitors can effectively control postprandial hyperglycemia and play a crucial role in treating type II diabetes. Geranylated compounds have been demonstrated to have strong alpha-glucosidase inhibitory activity, while their interaction mechanism was still unclear. In this study, we investigated the inhibition mechanisms of three structurally different geranylated compounds with significant alpha-glucosidase inhibition (puerarol, 8-geranyl-7, 3 '-dihydroxy-4 '-methoxyisoflavone, and xanthoangelol) separated from Pueraria lobata. The IC50 values of these compounds were 6.14 +/- 0.20, 2.35 +/- 0.02, and 1.98 +/- 0.07 mu M, respectively. It was found that puerarol acted as a non-competitive inhibitor, while 8-geranyl-7, 3 '-dihydroxy-4 '-methoxyisoflavone and xanthoangelol acted as un-competitive inhibitors. Fluorescence and circular dichroism data indicated that all three geranylated compounds statically quenched the fluorescence of alpha-glucosidase, and bound to alpha-glucosidase through a single binding site, inducing rearrangement of alpha-glucosidase and leading to conformational changes. Thermodynamic analysis and molecular docking revealed that puerarol interacted with alpha-glucosidase through hydrogen bonds and van der Waals forces, while the other two compounds primarily interacted through hydrophobic forces. This study elucidated the binding mechanism between three different structural geranylated compounds and alpha-glucosidase for the first time, which could contribute to the development of functional additives for the prevention and treatment of type II diabetes.

Abstract:Mulberry ( Morus alba L.) leaves have long been considered beneficial in traditional Chinese medicine to treat infectious and internal diseases. Recently studies have discovered that the mulberry leaf's total flavonoids (MLF) display excellent hypoglycemia properties. However, the active ingredients and their molecular mechanisms are still uncharacterized. In this study, we explored the hypoglycemic effects of MLF and mulberry leaf polysaccharides (MLP) on ob/ob mice, an animal model of type 2 diabetes mellitus (T2DM), compared with Ramulus Mori (Sangzhi) alkaloid (RMA). Network pharmacology was employed to identify the potential available targets and active compounds of MLF and RMA against hyperglycemia. Molecular docking, an insulin-resistant cell model and qPCR were employed to verify the antidiabetic activity of the critical compounds and the gene expression profiles of the top molecular targets. Here, the results showed that MLF and MLP improved glucose uptake in insulin-resistant hepatocytes. MLF, MLP and RMA alleviated insulin resistance and glucose intolerance in ob/ob mice. Unlike MLF and MLP, RMA administration did not influence the accumulation of intrahepatic lipids. Network pharmacology analysis revealed that morusin, kuwanon C and morusyunnansin L are the main active compounds of MLF and that they amend insulin resistance and glycemia via the PI3K- Akt signaling pathway, lipid and atherosclerosis pathways, and the AGE-RAGE signaling pathway. Moreover, 1-deoxynojirimycin (DNJ), fagomine (FA), and N-methyl-1-deoxynojirimycin are the primary active ingredients of RMA and target carbohydrate metabolism and regulate alpha-glucosidase activity to produce a potent anti-diabetic effect. The molecular docking results indicated that morusin, kuwanon C and morusyunnansin L are the critical bioactive compounds of MLF. They had high affinities with the key targets adenosine A1 receptor (ADORA1), AKT serine/threonine kinase 1 (AKT1), peroxisome proliferator-activated receptor gamma (PPARγ), and glycogen synthase kinase 3 beta (GSK3β), which play crucial roles in the MLF-mediated glucose-lowering effect. Additionally, morusin plays a role in amending insulin resistance of hepatocytes by repressing the expression of the ADORA1 and PPARG genes. Our results shed light on the mechanism behind the glucose-lowering effects of MLF, suggesting that morusin, kuwanon C, and morusyunnansin L might be promising drug leads for the management of T2DM.

Abstract:Inhibition of α-glucosidase alleviates postprandial high glycemic levels in diabetic or prediabetic population. In Chinese bayberry fruit, myricetin, quercetin and kaempferol are main flavonols, which differ only in their hydroxylation on the B-ring. Kaempferol (4'-OH) showed high IC50 (65.36 ± 0.27 μmol L-1) against α-glucosidase, while quercetin (3',4'-OH) exhibited stronger inhibition (46.91 ± 0.54 μmol L-1) and myricetin (3',4',5'-OH) possessed the strongest inhibitory activity (33.20 ± 0.43 μmol L-1). Molecular docking analysis illustrated that these flavonols could insert to the active cavity of α-glucosidase. Adjacent hydroxyl groups at B-ring of myricetin and quercetin positively contributed to form hydrogen bonds that were important to the stability of flavonol-enzyme complex, while kaempferol had no adjacent hydroxyl groups. Such observation was further validated by molecular dynamics simulations, and in good consistency with in vitro kinetic analysis and fluorescence spectroscopy analysis. Among three flavonols tested, myricetin possessed the strongest inhibition effects on α-glucosidase with the lowest dissociation constant (K i = 15.56 μmol L-1) of myricetin-α-glucosidase, largest fluorescence quenching constant (K sv) of (14.26 ± 0.03) × 104 L mol-1 and highest binding constant (K a) of (1.38 ± 0.03) × 105 L mol-1 at 298 K with the enzyme. Bio-Layer Interferometry (BLI) and circular dichroism (CD) analysis further confirmed that myricetin had high affinity to α-glucosidase and induced conformational changes of enzyme. Therefore, myricetin, quercetin and kaempferol are all excellent dietary α-glucosidase inhibitors and their inhibitory activities are enhanced by increasing number of hydroxyl groups on B-ring.
Mulberry Anthocyanin Extract Regulates Glucose Metabolism by Promotion of Glycogen Synthesis and Reduction of Gluconeogenesis in Human HepG2 Cells.

Abstract:Mulberry has been demonstrated to possess important biological activities such as antioxidation and antiinflammation. However, research on the ability of mulberry for diabetes improvement mainly focuses on the leaves and less on the fruit. This study showed that a mulberry anthocyanin extract (MAE) had a significant effect on increasing the glucose consumption in HepG2 cells. The MAE enhanced the glycogen content and suppressed levels of glucose production. The enzyme activities of phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphatase (G6Pase) were decreased in HepG2 cells after MAE treatment due to PPARγ coactivator 1α (PGC-1α) and forkhead box protein O1 (FOXO1) inhibition. Moreover, the phosphorylation of protein kinase B (AKT) and glycogen synthase kinase-3β (GSK-3β) was increased by the MAE, leading to an expression enhancement of glycogen synthase 2 (GYS2). And this effect was blocked by the phosphoinositide 3-kinase (PI3K) inhibitor LY294002. In summary, our results suggested that the MAE regulates glucose metabolism by activating the PI3K/AKT pathway that relates to glycogen synthesis as well as through the inhibition of key molecules that promote gluconeogenesis.
Effect of Mulberry Leaves Extract on Mice Blood Glucose Level and the Mensuration of Its Active Components

Abstract:Using KM mouse as test animals,the effect of mulberry leaves extract(MLE) on blood glucose level was studied and the quantities of four components,named 1-deoxynojimycin(DNJ),flavone,polyphenol and polysaccharide were mensurated.The result showed that the MLE was able to inhibit the increase of blood glucose in diabetic model mice.When preparing MLE,there were remarkable differences among the mulberry varieties on the content of active components and the function of MLE in reducing the blood glucose level.Among the five tested mulberry varieties,the Shansang305 was the best one.Moreover,the optimum leaf order was the upper half of shoot from mulberry trees.There was a close relationship between the effect of MLE in reducing blood glucose and the content of DNJ.

Abstract:Chinese bayberry (Morella rubra Sieb. et Zucc.) fruit have a diverse flavonoid composition responsible for the various medicinal activities, including anti-diabetes. In the present study, efficient simultaneous purification of four flavonoid glycosides, i.e., cyanidin-3-O-glucoside (1), myricetin-3-O-rhamnoside (2), quercetin-3-O-galactoside (3), quercetin-3-O-rhamnoside (4), from Chinese bayberry pulp was established by the combination of solid phase extract (SPE) by C18 Sep-Pak((R)) cartridge column chromatography and semi-preparative HPLC (Prep-HPLC), which was followed by HPLC and LC-MS identification. The purified flavonoid glycosides, as well as different fractions of fruit extracts of six bayberry cultivars, were investigated for -glucosidase inhibitory activities. The flavonol extracts (50% methanol elution fraction) of six cultivars showed strong -glucosidase inhibitory activities (IC50 = 15.4-69.5 g/mL), which were higher than that of positive control acarbose (IC50 = 383.2 g/mL). Four purified compounds 1-4 exerted -glucosidase inhibitory activities, with IC50 values of 1444.3 g/mL, 418.8 g/mL, 556.4 g/mL, and 491.8 g/mL, respectively. Such results may provide important evidence for the potential anti-diabetic activity of different cultivars of Chinese bayberry fruit and the possible bioactive compounds involved.
Mulberry leaf extract reduces the glycemic indexes of four common dietary carbohydrates

Abstract:Background: 1-Deoxynojirimycin (DNJ), a component of mulberry leaf extract (MLE), reduces postprandial hyperglycemia by inhibiting intestinal a-glycosidase. The aim of this exploratory study was to investigate the effects of MLE on the glycemic indexes (GI) of common dietary carbohydrates. Methods: This single-center, randomized, open-label, 7-cycle self-controlled crossover study enrolled 15 healthy volunteers at the National Drug Clinical Trial Institution, Second Affiliated Hospital of Tianjin University of Traditional Chinese Medicine (June 2014 to December 2014). The participants were randomized to receive glucose (3 occasions), glucose+MLE, sucrose+MLE, maltose+MLE, and maltodextrin+MLE orally during 7 visits (every 3 days). Blood glucose level was tested at 15 minutes before and at 15, 30, 45, 60, 90, and 120 minutes after carbohydrate intake. The GI of each carbohydrate relative to glucose (GI = 100) was calculated using the incremental area under the curve method. Safety was assessed at each visit. Results: All participants completed the protocol. After carbohydrate ingestion, blood glucose level peaked at 30 minutes (glucose, glucose+MLE, sucrose+MLE, and maltose+MLE) or 45 minutes (maltodextrin+MLE) before returning to preprandial levels at 120 minutes. At 30 minutes, the change in blood glucose level was lower for sucrose+MLE, maltose+MLE, and maltodextrin+MLE than for glucose or glucose+MLE (P < .05). GI was lowest for sucrose+MLE (43.22 ± 17.47) and maltose+MLE (49.23 ± 22.39), intermediate for maltodextrin+MLE (75.90 ± 26.01), and higher for glucose+MLE (91.88 ± 27.24). MLE reduced the GIs for maltose, sucrose, maltodextrin, and glucose by 53.11%, 33.51%, 31.00%, and 8.12%, respectively. MLE was well tolerated. Conclusions: Coconsumption of MLE with sucrose, maltose, or maltodextrin can reduce the GI values of these carbohydrates. Trial registration: Chinese Clinical Trial Registry Platform, no. Isolation of Chalcomoracin as a Potential α-Glycosidase Inhibitor from Mulberry Leaves and Its Binding Mechanism

Abstract:Diabetes is a chronic metabolic disease, whereas α-glucosidases are key enzymes involved in the metabolism of starch and glycogen. There is a long history of the use of mulberry leaf (the leaf of Morus alba) as an antidiabetic herb in China, and we found that chalcomoracin, one of the specific Diels–Alder adducts in mulberry leaf, had prominent α-glucosidase inhibitory activity and has the potential to be a substitute for current hypoglycemic drugs such as acarbose, which have severe gastrointestinal side effects. In this study, chalcomoracin was effectively isolated from mulberry leaves, and its α-glucosidase inhibition was studied via enzymatic kinetics, isothermal titration (ITC) and molecular docking. The results showed that chalcomoracin inhibited α-glucosidase through both competitive and non-competitive manners, and its inhibitory activity was stronger than that of 1-doxymycin (1-DNJ) but slightly weaker than that of acarbose. ITC analysis revealed that the combination of chalcomoracin and α-glucosidase was an entropy-driven spontaneous reaction, and the molecular docking results also verified this conclusion. During the binding process, chalcomoracin went into the "pocket" of α-glucosidase via hydrophobic interactions, and it is linked with residues Val544, Asp95, Ala93, Gly119, Arg275 and Pro287 by hydrogen bonds. This study provided a potential compound for the prevention and treatment of diabetes and a theoretical basis for the discovery of novel candidates for α-glycosidase inhibitors.

Inhibition of α-amylase and α-glucosidase by Morus australis fruit extract and its components iminosugar, anthocyanin, and glucose

Abstract:The inhibition of α-amylase and α-glucosidase are important for the maintenance of blood glucose level. Mammalian α-glucosidase includes maltase-glucoamylase and sucrase-isomaltase complexes. In this study, we examined the inhibitory effects of Morus australis fruit extract and its components, that is, three iminosugars (1-deoxynojirimycin [1-DNJ], fagomine, and 2-O-α-D-galactopyranosyl deoxynojirimycin), two anthocyanins (cyanidin-3-glucoside and cyanidin-3-rutinoside), and glucose, against α-amylase and α-glucosidase. We also quantified the concentration of each component in M. australis fruit extract. The IC50 values of the fruit extracts of four M. australis subspecies were >10 mg/ml for α-amylase, 1.1-1.7 mg/ml for maltase, 6.9-8.6 mg/ml for glucoamylase, 0.13-1.0 mg/ml for sucrase, and 0.46-1.4 mg/ml for isomaltase. When the IC50 value of each component and the concentration of each component in the fruit extract were taken into consideration, our results indicated that glucose are involved in the inhibition of α-amylase, and 1-DNJ and glucose are involved in the inhibition of α-glucosidase. This is in contrast to that in M. australis leaf, neither anthocyanin nor glucose are contained, and 1-DNJ is a main inhibitor. PRACTICAL APPLICATION: It is widely accepted that inhibition of α-amylase and α-glucosidase is one of the strategies to treat type-2 diabetes. Today, acarbose, miglitol, and voglibose are clinically used for this purpose. Our results that 1-DNJ and anthocyanin are present in Morus australis fruit and are involved in the inhibition of α-amylase and α-glucosidase suggest that M. australis fruit is a healthy sweetener.
Identification of a Novel Α-Glucosidase Inhibitor from Melastoma Dodecandrum Lour. Fruits and Its Effect on Regulating Postprandial Blood Glucose.

Abstract:Melastoma dodecandrum Lour. (MDL) extracts have shown potent α-glucosidase inhibitory activity, suggesting MDL might be a good source of α-glucosidase inhibitors. The aim of the study was to identify compounds in MDL extracts with α-glucosidase inhibitory activities and evaluate their effect on postprandial blood glucose as well as elucidating the underlying mechanisms of inhibition. A total of 34 polyphenols were identified in MDL fruits, among which 10 anthocyanins and three proanthocyanidin derivatives were discovered for the first time. Dosing mice with MDL extracts (100 mg/kg body weight, by gavage) was associated with a significantly decrease in postprandial blood glucose concentrations after oral administration of maltose. The most potent α-glucosidase inhibitor was identified as casuarictin (IC50 of 0.21 μg/mL). Casuarictin bound competitively to α-glucosidase, occupying not only the catalytic site but also forming strong hydrogen bonds with α-glucosidase residues. Therefore, casuarictin derived from MDL fruits might be used as novel α-glucosidase inhibitor in functional foods or other dietary products.
Exploring synergistic inhibitory mechanisms of flavonoid mixtures on α-glucosidase by experimental analysis and molecular dynamics simulation

Abstract:The study was the first to evaluate the synergistic interaction of luteolin + quercetin, luteolin + 3-O-methylquercetin, and quercetin + 3-O-methylquercetin mixtures on α-glucosidase and the binding mechanisms were explored using both experimental and theoretical approaches. The results showed that three flavonoid mixtures exhibited a mixed type of inhibition and demonstrated the most potent synergistic effects on α-glucosidase inhibition at 6:4 ratio, with interaction index (γ) of 0.85, 0.78 and 0.73, respectively. The three mixtures had a great influence on α-glucosidase secondary structures. Molecular simulation further demonstrated that three flavonoid mixtures formed hydrophobic interactions and hydrogen bonds with amino acid residues at different sites of α-glucosidase. Collectively, luteolin + quercetin, luteolin + 3-O-methylquercetin and quercetin + 3-O-methylquercetin were found to inhibit α-glucosidase in a synergistic manner and can be potentially used for the development of hypoglycemic food products.

Abstract:1-Deoxynojirimycin(DNJ) is an effective α-glucosidase inhibitor,being able to lower blood glucose efficiently.Nojirimycin was firstly found in streptomyces,while DNJ was firstly isolated from the root bark of mulberry tree.DNJ has been isolated and identified from mulberry(Morus L.),dayflower(Commelina communis),hyacinth(Hyacinthus orientalis) and adenophora(Adenophora triphylla var.japonica),among which mulberry has the highest content of DNJ.DNJ content of mulberry varies greatly with mulberry varieties,medicament portions,seasons,climates,geographical and soil conditions,leaf positions,and development stages etc.In microorganisms,DNJ was isolated from various species of streptomycete and bacillus.It was also discovered that two endophytes from mulberry,Stenotrophomonas maltophilia and Micrococcus luteus,could produce DNJ stably.So far,fermentation conditions for many microorganisms with DNJ production have been investigated.In insect,besides silkworm(Bombyx mori) which has rich content of DNJ,other mulberry monophagous insect species or oligophagous ones including Bombyx mandarina,Rondotia menciana,Phthonandria atrineata,and Diaphani apyloalis also have high content of DNJ.DNJ in insect body comes from their food,and DNJ content in silkworm body varies with silkworm species,developmental periods,diet,tissue and organ etc.It was also noticed that absorption and concentration of DNJ in silkworm body changed periodically with the advance of larval instar.Researches aiming to elucidate biosynthetic pathway of DNJ in streptomycete,bacillus,and dayflower have been conducted.It was found that DNJ synthetic pathway is different in different species.Important progress has been made in chemosynthesis of DNJ.Three major synthetic methods have been developed and some synthesized DNJ derivatives have been used in clinic.
Effect of 1-Deoxynojirimycin Isolated from Mulberry Leaves on Glucose Metabolism and Gut Microbiota in a Streptozotocin-Induced Diabetic Mouse Model

Abstract:1-Deoxynojirimycin (DNJ) exerts hypoglycemic effects. However, the traditional method for DNJ extraction is inefficient, and the hypoglycemic mechanism of DNJ remains unclear. In this study, the mixed fermentation by Lactobacillus fermentum and Saccharomyces cerevisiae was used to enhance DNJ extraction efficiency. It was found that this strategy was more efficient than the traditional method as the yield improved from the original 3.24 mg/g to 5.97 mg/g. The purified DNJ significantly decreased serum glucose (P < 0.01) and insulin levels (P < 0.05), improved serum lipid levels (P < 0.05), and reversed insulin resistance (P < 0.05) in diabetic mice. These changes were caused by up-regulating the protein expression of insulin receptor and glycolysis enzymes (GK, PK, and PFK) (P < 0.05) and down-regulating the protein expression of insulin receptor substrate-1 and gluconeogenesis enzymes (PCB, PEPCK, FBPase, and G-6-Pase) (P < 0.05), thus alleviating glucose tolerance. Additionally, DNJ treatment relieved gut dysbiosis in diabetic mice by promoting the growth of Lactobacillus, Lachnospiraceae NK4A136 group, Oscillibacter, norank Lachnospiraceae, Alistipes, and Bifidobacterium (P < 0.05) and suppressing the growth of Ruminococcaceae UCG-014, Weissella, Ruminococcus, Prevotellaceae Ga6A1 group, Anaerostipes, Klebsiella, Prevotellaceae UCG-001, and Bacteroidales S24-7 group (P < 0.05).

Abstract:Green tea, green tea polyphenols and epigallocatechin gallate (EGCG) are confirmed to have beneficial effects in the treatment of diabetes mellitus, and a possible mechanism can be ascribed to their inhibitory effect against α-amylase and α-glucosidase in the digestive tract. In this paper, we first investigated the combined inhibitory effect of green tea extracts, green tea polyphenols or EGCG with acarbose on α-amylase and α-glucosidase in vitro. Our results indicated that the interaction between green tea extracts (green tea polyphenols or EGCG) and acarbose was complicated. The combination of green tea extracts, green tea polyphenols or EGCG with acarbose had a synergistic effect on α-amylase and α-glucosidase at low concentrations and the combined effect turned out to be antagonistic at high concentrations according to the Combination Index (CI) values. These findings not only provided some significant quantitative values, but also provide some valuable implications for the combined use of acarbose and GTE (GTP or EGCG) in the treatment of diabetes mellitus.
Beta-Glucuronidase Inhibition by Constituents of Mulberry Bark

Abstract:Abstract Intestinal bacterial β-glucuronidases, the key enzymes responsible for the hydrolysis of various glucuronides into free aglycone, have been recognized as key targets for treating various intestinal diseases. This study aimed to investigate the inhibitory effects and mechanisms of the Mulberry bark constituents on E. coli β-glucuronidase (EcGUS), the most abundant β-glucuronidases produced by intestinal bacteria. The results showed that the flavonoids isolated from Mulberry bark could strongly inhibit E. coli β-glucuronidase, with IC50 values ranging from 1.12 μM to 10.63 μM, which were more potent than D-glucaric acid-1,4-lactone. Furthermore, the mode of inhibition of 5 flavonoids with strong E. coli β-glucuronidase inhibitory activity (IC50 ≤ 5 μM) was carefully investigated by a set of kinetic assays and in silico analyses. The results demonstrated that these flavonoids were noncompetitive inhibitors against E. coli β-glucuronidase-catalyzed 4-nitrophenyl β-D-glucuronide hydrolysis, with Ki values of 0.97 μM, 2.71 μM, 3.74 μM, 3.35 μM, and 4.03 μM for morin (1), sanggenon C (2), kuwanon G (3), sanggenol A (4), and kuwanon C (5), respectively. Additionally, molecular docking simulations showed that all identified flavonoid-type E. coli β-glucuronidase inhibitors could be well-docked into E. coli β-glucuronidase at nonsubstrate binding sites, which were highly consistent with these agentsʼ noncompetitive inhibition mode. Collectively, our findings demonstrated that the flavonoids in Mulberry bark displayed strong E. coli β-glucuronidase inhibition activity, suggesting that Mulberry bark might be a promising dietary supplement for ameliorating β-glucuronidase-mediated intestinal toxicity.

Abstract:Diabetes is a kind of chronic disease that seriously threatens human health. α-glucosidase inhibitors are an effective way to inhibit the increase of postprandial blood glucose. Mulberry leaf flavonoids can be used as highly effective α-glucosidase inhibitors, but usually in low concentrations. Microbial fermentation is an effective method to increase the content of flavonoids in plants. In this paper, the fermentation of mulberry leaves with Ganoderma lucidum was conducted to investigate the cell growth of microbial, the content and α-glucosidase inhibitory efficiency of flavonoids in mulberry leaves. The results indicated that mulberry leaves can satisfy the nutritional needs of G. lucidum, and the maximum cell dry weight of 393.32 mg was obtained after a 10 d of fermentation. The flavonoid content in mulberry leaves was increased significantly with the fermentation time. Compared to unfermented mulberry leaves, the flavonoid content in mulberry leaves fermented for 8 d increased by 56.5%. The inhibitory efficiency of the fermented flavonoids was also significantly improved. The IC50 of αglucosidase of flavonoids extracted from 8 d fermented mulberry leaves and unfermented mulberry leaves were 0.041 mg/mL and 0.01 mg/mL, respectively. This article will provide a reference for the enrichment of functional active ingredients in plants through microbial fermentation technology.
