Abstract:1. The hypothesis was that a diet with a high starch to fat ratio (HS) impairs nutrient digestibility and growth performance, as compared to a diet with a low starch to fat ratio (LS) in Eimeria-challenged broilers. From days 10 to 29, 12 replicate pens of birds were given isocaloric and isonitrogenous steam-pelleted diets with either HS or LS, by replacing the wheat starch in one diet by a mixture of rapeseed oil and inert sand in the other. On d 17, a 10-fold dose of live vaccine strains of Eimeria spp. was administered via drinking water. Ileal samples were collected on days 16 and 29. 2. Starch content in the ileum tended to be higher on d 16 and was significantly higher on d 29 in the HS group. 3. The HS diet did not induce exceedingly high levels of starch in the ileum, suggesting there was no starch overload in the gut. Ileal starch digestibility was improved with increasing dietary starch level from 23% to 45%. This demonstrated the capacity of the broiler chicken to digest high levels of starch regardless of Eimeria spp. infection. Ileal energy digestibility was not affected by the treatments. 4. Weight gain did not differ between treatments; however, birds fed the LS diet were less efficient in feed conversion as compared to those fed the HS diet. 5. The use of isolated starch and the unintended higher extent of starch gelatinisation in the HS diet may have contributed to the higher starch digestibility in birds given the HS diet. Thus, the hypothesis that high ratios of starch to fat in pelleted diets may impair starch digestibility and production performance in Eimeria-challenged broiler chickens was not verified. Further work is required to clarify this research question, taking into consideration the physical form of starch source and the potentially confounding role of feed processing on starch availability.
Effects of <i>Clostridium butyricum</i>- and <i>Bacillus</i> spp.-Based Potential Probiotics on the Growth Performance, Intestinal Morphology, Immune Responses, and Caecal Microbiota in Broilers

Abstract:We aimed to investigate the effects of Clostridium butyricum-, Bacillus subtilis-, and Bacillus licheniformis-based potential probiotics on the growth performance, intestinal morphology, immune responses, and caecal short chain fatty acids (SCFAs) and microbial structure in broiler chickens. Three treatment groups containing a total of 1200 one-day-old AA broilers were included: birds fed with a basal diet only (Con), birds fed with added 10(10) probiotics cfu/kg (ProL), and birds fed with added 10(11) probiotics cfu/kg (ProH). The dietary probiotics significantly improved the final and average body weights and serum immunoglobulins A, M, and Y. The probiotics also enhanced the ileal morphology and improved the caecal acetate, butyrate, and propionate contents. Furthermore, 16S rRNA sequencing revealed that dietary compound probiotics modulated the caecal microflora composition as follows: (1) all birds shared 2794 observed taxonomic units; (2) treatment groups were well separated in the PCA and PCoA analysis; (3) the relative abundance of Parabacteroides, Ruminococcaceae_UCG-014, Barnesiella, Odoribacter, [Eubacterium_coprostanoligenes_group], [Ruminococcus]_torques_group, and Butyricimonas significantly varied between treatments. The compound probiotics improved the growth performance, serum immune responses, the ratio of ileal villus height to crypt depth, and major caecal SCFAs in broiler chickens. The dietary C. butyricum-, B. subtilis-, and B. licheniformis-based probiotics improved overall broiler health and would benefit the poultry industry.

Abstract:The effects of feeding dehydrated Enterococcus faecium on growth performance, immune response, and cecal microflora in broiler chickens challenged with Escherichia coli K88 were investigated. Two hundred eighty-eight 1-d-old birds were randomly assigned to 4 treatments: negative control birds (N-con) fed a basal diet and not challenged with E. coli K88; positive control birds (P-con) fed a basal diet and challenged with E. coli K88; birds fed a basal diet including dehydrated E. faecium (Ef) at 1 × 10(9) cfu/kg of feed and challenged with E. coli K88; and birds fed a basal diet including the antibiotic colistine sulfate (Anti) at 10 mg/kg of feed and challenged with E. coli K88. Birds fed E. faecium had greater (P < 0.05) BW on d 14, 21, and 28 and greater (P < 0.05) jejunal villus height on d 21 and 28 compared with birds on the other treatments. Jejunal crypt depth was decreased (P < 0.05) in birds fed either E. faecium or antibiotic compared with P-con treatment birds on d 10, 21, and 28. Birds fed E. faecium had a greater (P < 0.05) concentration of IL-4 in their jejunal mucosa than did those in the N-con treatment group on d 10, 14, and 21. Infected birds, with or without E. faecium feeding, had a higher (P < 0.05) tumor necrosis factor-α and secreted IgA in their jejunal mucosa than did those in the N-con treatment group on d 10 and 14. Birds fed E. faecium had lower (P < 0.05) concentrations of E. coli on d 14 and 28, less (P < 0.05) Clostridium perfringens on d 28, greater Lactobacillus counts on d 14 and 21, and greater (P < 0.05) Bifidobacterium in their cecal contents on d 21 than did the P-con birds. These results suggest that E. faecium can promote growth performance, improve intestinal morphology, and beneficially manipulate the cecal microflora in broilers challenged with E. coli K88.
Altered chicken cecal microbial communities affect Salmonella colonization

Abstract:Altering the diets of chicken and other poultry shows promise as a mean for controlling and preventing Salmonellosis. However, it is not known whether alteration of the gut microbiota from changes in diet in turn changes the colonization of Salmonella. The current study describes an investigation of the effects of medium composition on chicken cecal microbiota and subsequent colonization of Salmonella in vitro. Both batch fermentation and chemostat systems were used to compare the effects on bacterial community by different culture media, which were either rich in starch and peptone (Veal Infusion medium, VI) or rich in simple sugars and hydrolyzed peptide (Viande-Levure medium, VL). Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) and quantitive real-time PCR (qPCR) were used to ascertain that the growth of chicken cecal microbiota in VL medium resulted in a Bacteroides-enriched community that was closer to the initial cecal inoculum. However, VI medium produced a microbiota represented mainly by Lactobacillus and Bifidobacterium. A time-dependent reduction of Salmonella was observed only in VI microbiota and the decrease was correlated with a marked increase in the proportion of Lactobacillus and Bifidobacterium relative to other species. Supernatant from lactic-acid bacteria (LAB) isolates from VI microbiota showed lactic acid-independent bacteriocidal activity on Salmonella. Animal challenge experiment also showed that LAB isolates have the function of decrease S. pullorum colonization in vivo. In summary, our results suggest that in vitro enriched microbiota with probiotic compositions can reduce colonization by Salmonella. [Yin Y, Lei F, Fang D, et al. Early Inoculation with Caecal Fermentation Broth Alters Small Intestine Morphology,gene Expression of Tight Junction Proteins in the Ileum, and the Caecal Metabolomic Profiling of Broilers

Abstract:Background The establishment of stable microbiota in early life is beneficial to the individual. Changes in the intestinal environment during early life play a crucial role in modulating the gut microbiota. Therefore, early intervention to change the intestinal environment can be regarded as a new regulation strategy for the growth and health of poultry. However, the effects of intestinal environmental changes on host physiology and metabolism are rarely reported. This study was conducted to investigate the effects of early inoculation with caecal fermentation broth on small intestine morphology, gene expression of tight junction proteins in the ileum, and cecum microbial metabolism of broilers. Results Our data showed that early inoculation with caecal fermentation broth could improve intestine morphology. The small intestine villus height was significantly increased ( P < 0.05) in the intervened broilers compared to the control group, especially on day 28. A similar result was observed in the ratio of villus height to crypt depth ( P < 0.05). Meanwhile, we found early inoculation significantly increased ( P < 0.05) the expression levels of zonula occludens-1 ( ZO1 ) on days 14 and 28, claudin-1 ( CLDN1 ) on day 28, whereas the gene expression of claudin-2 ( CLDN2 ) was significantly decreased ( P < 0.05) on days 14 and 28. Gas chromatography time-of-flight/mass spectrometry (GC-TOF/MS) technology was further implemented to systematically evaluate the microbial metabolite profiles. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) displayed a distinct trend towards separation between the fermentation broth group (F group) and the control group (C group). The differentially expressed metabolites were identified, and they were mainly functionally enriched in beta-alanine metabolism and biosynthesis of unsaturated fatty acids. In addition, 1,3-diaminopropane was selected as a key biomarker that responded to early inoculation with caecal fermentation broth. Conclusions These results provide insight into intestinal metabolomics and confirm that early inoculation with caecal fermentation broth can be used as a potential strategy to improve intestinal health of broilers.

Abstract:The present study investigated the effects of low protein diets with different starch sources and starch to protein ratio on growth, digestibility, intestinal health, caecal short chain fatty acids (SCFAs), serum cholesterol and triglycerides in broiler chickens. Eight hundred one-day-old male broiler chicks (Ross 308) were randomly allotted to one of 4 dietary treatments with 10 repeats and 20 birds in each repeat. The dietary treatments included 1) a standard protein corn-SBM based diet (SP), 2) a low protein corn-SBM based diet (LPI) without reduced starch: protein ratio, 3) a low protein corn-SBM based diet (LPII) with reduced starch: protein ratio, and 4) a low protein corn-SBM-peas based diet (LPP) and reduced starch: protein ratio. Soy hulls were added in the LPII and LPP diets to reduce starch: protein ratio. During the experiment period from 11 to 24 d, FI was not affected by the dietary treatments (P > 0.05). The BWG was significantly reduced in the LPI diet compared to the SP diet (P < 0.05). Likewise, FCR deteriorated in LPI and LPII but was better in the SP diet followed by the LPP diet (P < 0.05). The apparent total tract digestibility (ATTD) of dry matter (DM) varied significantly among the dietary treatments (P < 0.01). While ATTD of starch was similar for all the diets except the LPP diet wherein the ATTD of starch was significantly lower (P < 0.001). Ether extract digestibility was also significantly different between the SP and LPII dietary treatments (P < 0.01). The AME and AMEn values were significantly lower in the LPP diet compared with other dietary treatments (P < 0.001). Nitrogen retention (%) was increased in all the LP diets compared with the SP diet (P < 0.001), but it was significantly better in both LPII and LPP diets compared to the LPI diet. The data showed that cecal SCFAs production was increased in the LPII and LPP compared to the SP and LPI diets (P < 0.001). Further, the production of acetic, butyric, and propionic acids was substantially higher in the LPP diet (P < 0.001). There was no significant difference in gene expression of Claudin-1 and ZO-1 (P > 0.05). However, MUC-2 and GLUT-1 gene expression were significantly downregulated in the LPI diet (P < 0.05). The concentration of cholesterol and triglycerides was significantly increased in the LPI diet (P < 0.001). In conclusion, the addition of peas as a slowly digestible starch source combined with soy hulls in low protein diet helped to partly recover the growth performance and improved cecal SCFAs production compared to other low protein diets with and without reduced starch: protein ratio in broiler chickens.

Abstract:In a 21-d study, 480 Cobb 500 (off-sex) male broiler chicks were used to investigate the effects of feeding different sources and levels of resistant starches (RS) on growth performance, nutrient and energy utilization, and intestinal health in broiler chickens. The birds were allocated to 10 dietary treatments in a 3 × 3 + 1 factorial arrangement. The factors were 3 RS-sources (RSS): banana starch (BS), raw potato starch (RPS), and high-amylose corn starch (HCS); each at 3 levels (RSL) 25, 50, or 100 g/kg plus a corn-soybean meal control diet. Birds and feed were weighed on d 0, 8, and 21. On d 21, samples of jejunal tissue and digesta were collected for chemical analysis. Data were analyzed using the mixed model procedure of JMP with factor levels nested with the control. In the 0 to 21 phase, the birds fed the RPS diets had higher (P = 0.011) FI than those fed HCS or control diets, and FCR was greater (P = 0.030) in birds that received BS diets than in other diets. RSS × RSL was significant (P < 0.05) for total tract nutrient retention, AME, and AMEn on d 21. The starch digestibility was higher (P < 0.001) in birds that received the control diet than in RS diets, and decreased as RS levels increased, except for HCS. The apparent metabolizable energy (AME) and nitrogen-corrected AME (AMEn) were higher (P < 0.001) in birds fed 100 g/kg HCS diet, with both decreasing with increasing levels of BS and RPS, except for HCS. Relative ileal oligosaccharides profile showed significant (P < 0.05) RSS × RSL with a higher relative abundance of Hex(3) (P = 0.01) and Pent(3) (P = 0.001) in HCS diets. In conclusion, RS may influence gut health and growth performance in broiler chickens through modulation of cecal SCFA and nutrient digestion, but these depend largely on the botanical origin and concentrations of individual RS.
Influence of fermented feed additive on gut morphology, immune status, and microbiota in broilers

Abstract:Abstract Background This study examined the effects of a solid-state fermented feed additive (FFA) on the small intestine histology/morphology, immunity and microbiota of broilers. Two hundred eighty-eight day-old Arbor Acre chicks, were randomly assigned to one of four groups (each group has 6 replicates, with each replicate containing 12 chickens). The negative control (NC; basal diet), the positive control (PC; basal diet +antibiotic 15 ppm), the fermented feed additive low dose (FFL; basal diet + 0.3 kg/t FFA), and the fermented feed additive high dose (FFH; 3 kg/t FFA) with Lactobacillus casei ( L.casei ). Results The study found that the FFH and FFL groups gained more weight (1-21d) and the FFL and PC diets had better feed conversion ratio ( P < 0.05) than the NC from 0-42d. The FFH group had higher villus height ( P < 0.05) in the duodenum than the PC and villus height to crypt depth ratio VH/CD compared to PC and FFL groups. The FFL chickens had greater ( P < 0.05) jejunal and ileal villus height than PC and NC groups respectively. The FFL group had a higher ileal VH/CD ratio ( P < 0.05). Jejunum VH/CD was higher in FFL and FFH ( P < 0.05) than PC ( P < 0.05). FFH had a smaller thymus than NC ( P < 0.05). FFA diets also increased IL-10 expression ( P < 0.05). While IL-1 and TLR4 mRNA expression decreased ( P < 0.05) compared to NC. The microbiota analysis showed that the microorganisms that have pathogenic properties such as phylum Delsulfobacterota and class Desulfovibriona and Negativicutes was also significantly reduced in the group treated with FFH and PC while microorganisms having beneficial properties like Lactobacillaceae family, Lactobacillus aviarus genus and Lactobacillus spp were also tended to increase in the FFH and FFL fermented feed groups compared to the PC and NC groups. Conclusion These findings suggested that the FFA diet may modulate cecal microbiota by reducing pathogenic microorganisms such as phylum Delsulfobacterota and class Desulfovibriona and Negativicutes improve beneficial microorganisms like Lactobacillaceae family, Lactobacillus aviarus genus and Lactobacillus spp. While FFA diet also affect immunity, and gene expression related to immunity.

Ileal microbiota composition of broilers fed various commercial diet compositions

Abstract:Microbiota plays a role in the release and absorption of nutrients from feed components, thereby affecting digesta composition and moisture content of the excreta. The objective of the current study was to determine the effects of 5 different diets varying in ingredients (medium-chain fatty acids, nonstarch polysaccharides, and starch) on the microbiota composition of ileal digesta of broiler chickens and excreta DM content. Each treatment was repeated 6 times in cages each containing 18 Ross 308 broilers, with growth performance measured from 0 to 34 d of age and excreta DM and ileal microbiota composition analyzed at 34 d of age. Microbiota composition was evaluated using a novel ribosomal RNA microarray technology containing 370 different probes covering various genera, groups of microbial species, and individual species of the chicken gut microbiota, of which 321 had a signal above the background threshold. Replacing part of the animal fat and soybean oil in the wheat-based diet with medium-chain fatty acids (MCFA; 0.3% C10 and 2.7% C12) improved feed efficiency compared with the other dietary treatments. This coincided with a suppression of gram-positive bacteria belonging to the phylum of the Firmicutes, including Lactobacillus species, and species belonging to the family of the Enterococcaceae and Micrococcaceae, whereas the gram-negative bacteria belonging to the family of the Enterobacteriaceae were promoted. None of the other diets used in the present study notably changed the ileal digesta bacteria composition. Excreta DM content was not affected by dietary treatment. The variation between individual birds per dietary treatment was more pronounced than variation caused by feed composition, with the exception of the digesta microbiota of the birds fed the MCFA diet. It is concluded that a diet with MCFA significantly changes the ileal microbiota composition, whereas the effect of the other diets on the composition of the microbiota and excreta DM content is small in broiler chickens.

Abstract:This paper examines the relationship of performance with product and international diversification on Japanese multinational firms from 1977 to 1993. We show the relationships between diversification and performance change over time through the use of multiple time periods and accounting for keiretsu membership. Results show that while diversity strategies vary between keiretsu and non-keiretsu firms, performance is not much different. Across time periods, performance varies considerably, but strategies are less variable. Product diversity has weak effects on firm performance only in one time period, while international diversification has negative profitability and positive growth consequences in in some periods. These results suggest first that diversification strategies and their effects on performance vary across time periods and generally produce some unexpected findings. Diet-induced changes in the jejunal microbiota of developing broilers reduce the abundance of Enterococcus hirae and Enterococcus faecium

Abstract:Modern broiler breeds allow for high feed efficiency and rapid growth, which come at a cost of increased susceptibility to pathogens and disease. Broiler growth rate, feed efficiency, and health are affected by the composition of the gut microbiota, which in turn is influenced by diet. In this study, we therefore assessed how diet composition can affect the broiler jejunal gut microbiota. A total of 96 broiler chickens were divided into four diet groups: control, coated butyrate supplementation, medium-chain fatty acid supplementation, or a high-fibre low-protein content. Diet groups were sub-divided into age groups (4, 12 and 33 days of age) resulting in groups of 8 broilers per diet per age. The jejunum content was used for metagenomic shotgun sequencing to determine the microbiota taxonomic composition at species level. The composed diets resulted in a total of 104 differentially abundant bacterial species. Most notably were the butyrate-induced changes in the jejunal microbiota of broilers 4 days post-hatch, resulting in the reduced relative abundance of mainly Enterococcus faecium (-1.8 l2fc, P adj = 9.9E-05) and the opportunistic pathogen Enterococcus hirae (-2.9 l2fc, P adj = 2.7E-08), when compared to the control diet. This effect takes place during early broiler development, which is critical for broiler health, thus exemplifying the importance of how diet can influence the microbiota composition in relation to broiler health. Future studies should therefore elucidate how diet can be used to promote a beneficial microbiota in the early stages of broiler development.

Dietary strategies can increase cloacal endotoxin levels and modulate the resident microbiota in broiler chickens

Abstract:Endotoxins released from poultry feces have been associated with impaired human health. Because endotoxins are released from gram-negative intestinal bacteria, it was hypothesized that dietary strategies may influence endotoxin excretion via modulation of gut microbiota. We therefore tested dietary strategies that could potentially reduce cloacal endotoxin levels in broiler chickens. One-day-old male Ross 308 (N = 1,344) broilers were housed in 48 pens (N = 8 pens/treatment, 28 chickens per pen) and fed 1 of 6 diets for 35 days (d) in a 3-phase feeding program: a basic diet (CON) that served as the reference diet, or basic diet supplemented with butyrate (BUT), inulin (INU), medium-chain fatty acids (MCFA) or Original XPC™LS (XPC), or a high-fiber-low-protein (HF-LP) diet. A significant (P < 0.05) increase in cloacal endotoxin concentration at d 35 was observed in BUT as compared to CON. Analysis of cloacal microbiota showed a trend (P < 0.07) for a higher gram-negative/gram-positive ratio and for a higher relative abundance of gram-negative bacteria at d 35 (P ≤ 0.08) in BUT and HF-LP as compared to CON. A significant (P < 0.05) increase in average daily gain (ADG) and improved feed conversion ratio (P < 0.05) were observed in MCFA during the grower phase (d 14-28), and a significant (P < 0.05) increase in average daily feed intake (ADFI) was observed in MCFA during d 0 to 28. Broilers fed HF-LP had a significantly (P < 0.05) higher FCR and lower ADG throughout the rearing period. No treatment effects were found on footpad dermatitis, but BUT had worst hock burn scores at d 35 (P < 0.01) and MCFA had worst cleanliness scores at d 21 but not at d 35 (treatment*age P < 0.05), while INU had better cleanliness as compared to CON at d 35 (P < 0.05). In conclusion, especially BUT and HF-LP were able to modulate resident microbiota and BUT also increased cloacal endotoxin levels, which was opposite to our hypothesis. The present study indicates that cloacal endotoxin release can be affected by the diet but further study is needed to find dietary treatments that can reduce cloacal endotoxin release.
Interactions of Microbiota and Mucosal Immunity in the Ceca of Broiler Chickens Infected with Eimeria tenella

Abstract:The purpose of the study was to investigate the effects of Eimeria tenella infection on the cecal microbiome, the protein concentration of cecal content, cecal mucosal immunity, and serum endotoxin levels in broilers. Three hundred sixty 14-day-old broilers were allocated to five infection doses with six replicates. The five infection doses were: ID0: 0, ID1: 6250, ID2: 12,500, ID3: 25,000, and ID4: 50,000 Eimeria tenella oocysts. Eimeria tenella infection significantly increased the relative abundance of the phylum Proteobacteria, which includes diverse pathogenic bacteria, and significantly decreased the relative abundance of the phylum Firmicutes. Protein concentration of the cecal content was linearly increased (p < 0.05), and the concentration of secretory immunoglobulin A (sIgA) in the cecal content was linearly decreased by Eimeria tenella infection (p < 0.05). Goblet cell density was linearly reduced in the ceca by Eimeria tenella infection (p < 0.05). Eimeria tenella infection tended to linearly decrease the relative mRNA expression of antimicrobial peptide genes such as avian beta-defensin 9 (AvBD9; p = 0.10) and liver-expressed antimicrobial peptide 2 (LEAP2; p = 0.08) in the cecal tissue. Therefore, Eimeria tenella infection negatively modulated cecal microbiota via impairing cecal mucosal immunity and increasing protein concentration in the cecal content in broilers.

Abstract:Two 21-day experiments were conducted to investigate the effects of exogenous enzymes on growth performance, tight junctions, and nutrient transporters, jejunal oligosaccharides and cecal short-chain fatty acids (SCFA) of broiler chickens challenged with mixed Eimeria. Two different basal diets: high fiber-adequate protein (HFAP; Expt. 1) or low fiber-low protein (LFLP; Expt. 2) were used in the two experiments. In each experiment, birds were allocated to four treatments in a 2 × 2 factorial arrangement (with or without protease and xylanase combination; with or without Eimeria challenge). In Expt. 1, with HFAP diets, Eimeria upregulated (p < 0.05) the expression of claudin-1, but downregulated (p < 0.05) glucose transporters GLUT2/GLUT5. On the contrary, enzymes downregulated (p < 0.05) claudin-1 and alleviated the Eimeria-depressed GLUT2/GLUT5 expression. In both experiments, Eimeria decreased (p < 0.05) cecal saccharolytic SCFA and increased (p < 0.05) cecal branched-chain fatty acids. The challenge × enzyme interaction (p < 0.05) showed that enzymes reversed the Eimeria effects on fermentation pattern shift. In conclusion, Eimeria altered tight junctions and nutrient transporters expression promoted cecal proteolytic fermentation and inhibited saccharolytic fermentation. Exogenous enzymes showed the potential of alleviating the Eimeria-induced intestinal gene expression changes and reversing the unfavorable cecal fermentation pattern.

Impact of feed additives and host-related factors on bacterial metabolites, mucosal integrity and immune response in the ileum of broilers

Abstract:The present study aimed to investigate the effect of age, breed, and sex of broilers, as well as a probiotic or phytobiotic product on mucosal morphology, bacterial metabolites, and immune traits in the ileum of broilers. A total of 2,880 one-day-old male and female broiler chicks from two breeds (Ross308® and Cobb500®) were randomly assigned to 72 pens. Broilers were offered a wheat-soybean diet without (CO), or with either a probiotic (PO; 2.4 × 10 9 CFU/kg of Bacillus subtilis DSM32324 and DSM32325 and B. amyloliquefaciens DSM25840) or a phytobiotic (PY; grape extract, 165 ppm procyanidin and 585 ppm polyphenols of the diet) product. The trial was conducted with a 3 × 2 × 2 factorial arrangement of diet, breed, and sex in a completely randomized design (6 replicate-pens per treatment). At day 7, 21, and 35, one chicken per pen was slaughtered for collecting ileal tissue to evaluate of histomorphology and mRNA expression, as well as ileal digesta to measure bacterial metabolites. Data were subjected to ANOVA (the main factors; age, diet, breed, and sex) and Four-Way ANOVA (interactions) using GLM procedure. Overall, the concentration of acetate and total short chain fatty acids reached the peak and lactate decreased to its lowest on day 21, but their concentrations at day 7 and 35 were similar ( p > 0.05). Spermine, spermidine, and ammonia decreased after day 7, while putrescine and cadaverine increased after day 21 ( p < 0.05). mRNA expression of cytokines, mucin 2 ( MUC2 ) and claudin 5 ( CLDN5 ) was similar; increased from day 7 to 21 and decreased afterward ( p < 0.05). Villus height, crypt depth and villus surface area increased with age ( p < 0.05). Acidic goblet cells (GC) number and density increased after day 21 ( p < 0.05). Ross broilers showed higher D-lactate concentration and IFN -γ expression, while Cobb broilers had greater IL-4 , IL-6 and TNF-α expression and higher total GC number ( p < 0.05). Female displayed higher villus height and GC number and density (mixed and total GC) than male ( p < 0.05). The effect of dietary treatment was not found on any investigated variables ( p > 0.05). In conclusion, aging of broilers affected ileal histomorphology, cytokine expression, and barrier integrity, as well as bacterial activity. These observed impacts could be attributed to host-microbiota interaction and the direct effects of bacterial metabolites on intestinal cells and immune system.

At day 7, 21, and 35, one chicken per pen was slaughtered for caecal sampling to quantify bacterial metabolites (digesta) as well as evaluate mRNA abundance and histomorphology (tissue). Data were subjected to ANOVA using GLM procedure to evaluate age, diet, breed and sex and their interactions. Spearman's correlation (r) was analyzed between metabolite concentration and mRNA abundance. Overall, the concentration of short chain fatty acids increased with age, while lactate decreased from day 7 to 21 ( p < 0.05). The mRNA abundance of IL-2, IL-4, IL-6, IL-8, IL-10, IL-12, IL-17α, IL-18, IFN-γ and TGF-β2 increased with age but IL-1β and TNF-α increased in abundance from day 7 to 21 and then decreased ( p < 0.05). Abundance of MUC2 and CLDN5 increased after day 21 ( p < 0.05). Caecal crypt depth increased with age ( p < 0.05). Acidic goblet cell (GC) number peaked at day 21 ( p < 0.05), while mixed GC number was not affected by age. A few impacts of breed, diet and interactions on the investigated variables showed no meaningful biological pattern. Propionate positively correlated with all cytokines investigated (r = 0.150–0.548), except TNF-α. Lactate negatively correlated with pro-inflammatory cytokines like IL-1β (r = −0.324). Aging affected caecal histomorphology, bacterial activity and genes responsible for barrier integrity and inflammatory response. This effect could be attributed to the interaction between gut microbiota and immune system as well as the direct effect of metabolites on gut histomorphology and cytokine mRNA abundance.

Abstract:This paper presents an adaptive modulation and power allocation method for uplink multiuser multiple-input multiple-output systems under the assumption that there is perfect channel information at the receiver but not at the transmitter. The receiver jointly optimizes the power level and modulation order for all users under constraints of transmit power and error requirements, and returns these information to each user via a low-rate feedback channel. Power and modulation optimization maximizes the total throughput under a tolerable bit error ratio for each user. The ellipsoid method is used to design efficient algorithms for optimal power and modulation level.

Abstract:Introduction: Dietary nutrient content is crucial for energy metabolism and development of gut microbiota. Herein, this study aimed to explore the effects of fat-to-fiber ratios on nutrient transporter, energy metabolism and gut microbiota when ingredients composition was altered. Methods: A total of 240 as-hatched broiler chickens were randomly assigned into three groups including low fat-high dietary fiber (LF-HD), medium fat-medium dietary fiber (MF-MD) and high fat-low dietary fiber (HF-LD), with diets being iso-protein, and broilers were offered the same commercial diets from 21 to 42 d. The data were analyzed using one-way ANOVA of SPSS. Results and Discussion: Results showed that HF-LD diet significantly increased glucose content and decreased triglyceride in serum of broilers ( p < 0.05). The mRNA abundance of jejunal gene involved in glucose transporter and tricarboxylic acid (TCA) cycle was significantly increased in broilers fed with HF-LD diets. Compared with LF-HD, HF-LD had a lower abundance of Anaerofilum and CHKCI001 , and an increased proportion of beneficial bacteria such as Alistipes , Catenibacillus , Intestinimonas , Lactobacillus , and Peptococcus ( p < 0.05). Functional prediction of these microbial changes indicated that HF-LD diet drove caecal microbiota to participate in carbohydrate metabolism and TCA cycle ( p < 0.05). Dietary HF-LD-induced microbiota changes were positively correlated with growth performance of broilers ( p < 0.05). Therefore, HF-LD diet increased glucose transporters and energy metabolism in intestine and shaped microbial structure and metabolic pathways, which may benefit the growth performance of broilers.

Abstract:A 35-d study investigated the impact of dietary supplementation with Arginine (Arg) or branched-chain amino acids (BCAA) of broilers receiving low-protein diets whilst infected with mixed Eimeria species. All birds were given the same starter (d0-10) and finisher (d28-35) diets. The 4 grower diets used were a positive control (PC) with adequate protein (18.5%), a low protein diet (NC;16.5% CP), or the NC supplemented with Arg or BCAA. Supplemental AA was added at 50% above the recommended levels. The treatments were in a 4 × 2 factorial arrangement, with 4 diets, with or without Eimeria inoculation on d14. Birds and feed were weighed after inoculation in phases: prepatent (d14-17), acute (d18-21), recovery (d22-28), and compensatory (d29-35). Ileal digesta, jejunum, and breast tissue were collected on d21, 28, and 35. There was no diet × Eimeria inoculation on growth performance at any phase. Infected birds weighed less and consumed less feed (P < 0.05) in all phases. In the prepatent and acute phases, birds on the Arg diets had higher weight gain (P < 0.05) and lower FCR, similar to PC, when compared to NC and BCAA-fed ones. Infection reduced AA digestibility on d21 and 28 (Met and Cys). However, birds that received supplemental AA had higher digestibility (P < 0.05) of their respective supplemented AA on d 21 only. Infected birds had lower (P < 0.05) BO + AT and higher PEPT1 expression on d21. There was a diet × Eimeria interaction (P = 0.004) on gene expression at d28; 4EBP1 genes were significantly downwardly expressed (P < 0.05) in birds fed Arg diet, irrespective of infection. Infected birds exhibited an upward expression (P < 0.05) of Eef2 on d21 and d28 but experienced a downward expression on d35. Supplemental Arg and BCAA had variable effects on growth performance, apparent ileal AA digestibility, and genes of protein synthesis and degradation, but the effect of Arg on promoting weight gain, irrespective of the Eimeria challenge, was more consistent.

Abstract:OBJECTIVES To compare the volume of the maxillary sinus, dental factors, and craniofacial anatomical features between control subjects and patients with chronic rhinosinusitis (CRS) and to investigate critical factors for the volumetric change in the maxillary sinus in adults. DESIGN Retrospective case-control study. SETTING Tertiary referral center. PARTICIPANTS Ninety-nine individuals who visited an allergy and sinus center: 52 control subjects (septal deviation; mean age, 32.69 years) and 47 patients with CRS (mean age, 44.43 years). INTERVENTION Cephalometry and computed tomography were performed in all the participants. In blinded tests, dentists investigated the dental status of both groups. MAIN OUTCOME MEASURES Maxillary sinus: bone thickness and volume on computed tomography; craniofacial anatomical features: linear and angular variables in lateral cephalometry; and dental evaluation: malocclusion class, teeth status, and alveolar bone height. RESULTS Bony wall thickness of the maxillary sinus significantly increased in patients with CRS (P < .001) but showed no relationship with maxillary sinus volume. Maxillary sinus volume significantly decreased in patients with CRS (P = .001). Age and alveolar bone height had a negative effect on maxillary sinus volume in both groups. Abnormal teeth had no relationship with maxillary sinus volume in both groups but showed a negative effect on alveolar bone height in the CRS group (P = .02). Class II malocclusion associated with anterior movement of the maxilla significantly increased in the CRS group (P = .006). CONCLUSIONS Regardless of CRS, maxillary sinus volume decreased with older age and increased with alveolar bone loss. Regarding craniofacial anatomical features, CRS may have an effect on malocclusion in adults.
