Abstract:Three-dimensional (3D) bioprinting is a promising and innovative biomanufacturing technology, which can achieve precise position controlling of cells and extracellular matrix components, and further create complex and functional multi-cellular tissues or organs in a 3D environment. Bioink in the form of the cell-loaded hydrogel is most commonly used in bioprinting, and it is vital to the process of bioprinting. The bionic scaffold should possess suitable mechanical strength, biocompatibility, cell proliferation, survival, and other biological characteristics. The disadvantages of natural polymer hydrogel materials include poor mechanical properties as well as low printing performance and shape fidelity. Over the past years, a series of synthetic, modified, and nanocomposite hydrogels have been developed, which can interact through physical interactions, chemical covalent bond crosslinking, and bioconjugation reactions to change the characteristics to satisfy the requirements. In this review, a comprehensive summary is provided on recent research regarding the unique properties of hydrogel bioinks for bioprinting, with optimized methods and technologies highlighted, which have both high-value research significance and potential clinical applications. A critical analysis of the strengths and weaknesses of each hydrogel-based biomaterial ink is presented at the beginning or end of each section, alongside the latest improvement strategies employed by current researchers to address their respective shortcomings. Furthermore, we propose potential repair sites for each hydrogel-based ink based on their distinctive repair features, while reflecting on current research limitations. Finally, we synthesize and analyze expert opinions on the future of these hydrogel-based bioinks in the broader context of tissue engineering and regenerative medicine, offering valuable insights for future investigations.

Abstract:3D bioprinting is transforming tissue engineering in medicine by providing novel methods that are precise and highly customizable to create biological tissues. The selection of a "cell ink", a printable formulation, is an integral part of adapting 3D bioprinting processes to allow for process optimization and customization related to the target tissue. Bioprinting hydrogels allows for tailorable material, physical, chemical, and biological properties of the cell ink and is suited for biomedical applications. Hydrogel-based cell ink formulations are a promising option for the variety of techniques with which bioprinting can be achieved. In this review, we will examine some of the current hydrogel-based cell inks used in bioprinting, as well as their use in current and proposed future bioprinting methods. We will highlight some of the biological applications and discuss the development of new hydrogels and methods that can incorporate the completed print into the tissue or organ of interest.

Abstract:As the biocompatible materials, hydrogels have been widely used in three- dimensional (3D) bioprinting/organ printing to load cell for tissue engineering. It is important to precisely control hydrogels deposition during printing the mimic organ structures. However, the printability of hydrogels about printing parameters is seldom addressed. In this paper, we systemically investigated the printability of hydrogels from printing lines (one dimensional, 1D structures) to printing lattices/films (two dimensional, 2D structures) and printing 3D structures with a special attention to the accurate printing. After a series of experiments, we discovered the relationships between the important factors such as air pressure, feedrate, or even printing distance and the printing quality of the expected structures. Dumbbell shape was observed in the lattice structures printing due to the hydrogel diffuses at the intersection. Collapses and fusion of adjacent layer would result in the error accumulation at Z direction which was an important fact that could cause printing failure. Finally, we successfully demonstrated a 3D printing hydrogel scaffold through harmonize with all the parameters. The cell viability after printing was compared with the casting and the results showed that our bioprinting method almost had no extra damage to the cells.

Abstract:<p>3D printing alias additive manufacturing can transform 3D virtual models created by computer-aided design (CAD) into physical 3D objects in a layer-by-layer manner dispensing with conventional molding or machining. Since the incipiency, significant advancements have been achieved in understanding the process of 3D printing and the relationship of component, structure, property and application of the created objects. Because hydrogels are one of the most feasible classes of ink materials for 3D printing and this field has been rapidly advancing, this Review focuses on hydrogel designs and development of advanced hydrogel-based biomaterial inks and bioinks for 3D printing. It covers 3D printing techniques including laser printing (stereolithography, two-photon polymerization), extrusion printing (3D plotting, direct ink writing), inkjet printing, 3D bioprinting, 4D printing and 4D bioprinting. It provides a comprehensive overview and discussion of the tailorability of material, mechanical, physical, chemical and biological properties of hydrogels to enable advanced hydrogel designs for 3D printing. The range of hydrogel-forming polymers covered encompasses biopolymers, synthetic polymers, polymer blends, nanocomposites, functional polymers, and cell-laden systems. The representative biomedical applications selected demonstrate how hydrogel-based 3D printing is being exploited in tissue engineering, regenerative medicine, cancer research, in vitro disease modeling, high-throughput drug screening, surgical preparation, soft robotics and flexible wearable electronics. Incomparable by thermoplastics, thermosets, ceramics and metals, hydrogel-based 3D printing is playing a pivotal role in the design and creation of advanced functional (bio)systems in a customizable way. An outlook on future directions of hydrogel-based 3D printing is presented.</p>

Abstract:Recently, tissue engineering (TE) is one of the fast growing research fields due the accessibility of extra-molecular matrix (ECM) at cellular and molecular level with valuable potential prospective of hydrogels. The enhancement in the production of hydrogel-based cellular scaffolds with the structural composition of ECM has been accelerated with involvement of rapid prototyping techniques. Basically, the recreation of ECM has been derived from naturally existed or synthetic hydrogel-based polymers. The rapid utilization of hydrogels in TE puts forward the scope of bioprinting for the fabrication of the functional biological tissues, cartilage, skin and artificial organs. The main focus of the researchers is on biofabrication of the biomaterials with maintaining the biocompatibility, biodegradability and increasing growth efficiency. In this review, biological development in the structure and cross-linking connections of natural or synthetic hydrogels are discussed. The methods and design criteria that influence the chemical and mechanical properties and interaction of seeding cells before and after the implantations are also demonstrated. The methodology of bioprinting techniques along with recent development has also been reviewed. In the end, some capabilities and shortcomings are pointed out for further development of hydrogels-based scaffolds and selection of bioprinting technology depending on their application.

Abstract:The use of three-dimensional bioprinting technology combined with the principle of tissue engineering is important for the construction of tissue or organ regeneration microenvironments. As a three-dimensional bioprinting ink, hydrogels need to be highly printable and provide a stiff and cell-friendly microenvironment. At present, hydrogels are used as bioprinting inks in tissue engineering. However, there is still a lack of summary of the latest 3D printing technology and the properties of hydrogel materials. In this paper, the materials commonly used as hydrogel bioinks; the advanced technologies including inkjet bioprinting, extrusion bioprinting, laser-assisted bioprinting, stereolithography bioprinting, suspension bioprinting, and digital 3D bioprinting technologies; printing characterization including printability and fidelity; biological properties, and the application fields of bioprinting hydrogels in bone tissue engineering, skin tissue engineering, cardiovascular tissue engineering are reviewed, and the current problems and future directions are prospected.

Abstract:Hydrogels are a promising material for a variety of applications after appropriate functional and structural design, which alters the physicochemical properties and cell signaling pathways of the hydrogels. Over the past few decades, considerable scientific research has made breakthroughs in a variety of applications such as pharmaceuticals, biotechnology, agriculture, biosensors, bioseparation, defense, and cosmetics. In the present review, different classifications of hydrogels and their limitations have been discussed. In addition, techniques involved in improving the physical, mechanical, and biological properties of hydrogels by admixing various organic and inorganic materials are explored. Future 3D printing technology will substantially advance the ability to pattern molecules, cells, and organs. With significant potential for producing living tissue structures or organs, hydrogels can successfully print mammalian cells and retain their functionalities. Furthermore, recent advances in functional hydrogels such as photo- and pH-responsive hydrogels and drug-delivery hydrogels are discussed in detail for biomedical applications.

Abstract:Three-dimensional (3D) printing is well acknowledged to constitute an important technology in tissue engineering, largely due to the increasing global demand for organ replacement and tissue regeneration. In 3D bioprinting, which is a step ahead of 3D biomaterial printing, the ink employed is impregnated with cells, without compromising ink printability. This allows for immediate scaffold cellularization and generation of complex structures. The use of cell-laden inks or bio-inks provides the opportunity for enhanced cell differentiation for organ fabrication and regeneration. Recognizing the importance of such bio-inks, the current study comprehensively explores the state of the art of the utilization of bio-inks based on natural polymers (biopolymers), such as cellulose, agarose, alginate, decellularized matrix, in 3D bioprinting. Discussions regarding progress in bioprinting, techniques and approaches employed in the bioprinting of natural polymers, and limitations and prospects concerning future trends in human-scale tissue and organ fabrication are also presented.

Abstract:Granular hydrogels composed of hydrogel microparticles are promising candidates for 3D bioprinting due to their ability to protect encapsulated cells. However, to achieve high print fidelity, hydrogel microparticles need to jam to exhibit shear‐thinning characteristics, which is crucial for 3D printing. Unfortunately, this overpacking can significantly impact cell viability, thereby negating the primary advantage of using hydrogel microparticles to shield cells from shear forces. To overcome this challenge, we introduce a novel solution: a biphasic, granular colloidal bioink designed to optimize cell viability and printing fidelity. The biphasic ink consists of cell‐laden polyethylene glycol (PEG) hydrogel microparticles embedded in a continuous gelatin methacryloyl (GelMA)‐nanosilicate colloidal network. Here, we demonstrate that this biphasic bioink offers outstanding rheological properties, print fidelity, and structural stability. Furthermore, we demonstrate its utility for engineering complex tissues with multiple cell types and heterogeneous microenvironments, which we demonstrate by incorporating β‐islet cells into the PEG microparticles and endothelial cells in the GelMA‐nanosilicate colloidal network. Using this approach, it is possible to induce cell patterning, enhance vascularization, and direct cellular function. The proposed biphasic bioink holds significant potential for numerous emerging biomedical applications, including tissue engineering and disease modeling. This article is protected by copyright. Abstract:Advancement in additive manufacturing has opened new avenues to serve the biomedical field, including tissue engineering. The major focus of tissue engineering is to replicate or restore the lost functions of failed organs/damaged tissues by integrating cells and biomaterials. The ease in fabrication of customized scaffolds with high precision and low material wastage are the key features of additive manufacturing over conventional methods. Recently, hydrogel materials as bio ink have shown their potential in biomedical field owing to their resemblance to extracellular matrix and biocompatibility. The application of hydrogels can be exploited for regenerative medicine, wound healing, and drug delivery. Various hydrogels find their application in tissue engineering that can be fabricated into scaffolds using a wide range of processes of additive manufacturing. However, due to the lack of structural strength, the hydrogel matrix is incorporated with different additives in order to improve overall mechanical properties. This review article gives a comprehensive overview on the use of hydrogels of different kinds, such as naturally occurring, synthetic, and composite hydrogels, as prospective bio ink materials for tissue engineering. Different sources of hydrogels, their fabrication and tissue engineering specific properties are discussed in detail. Further, the fundamental and technological challenges for additive manufacturing of hydrogel bioink material for tissue engineering are reviewed. This article will facilitate the reader with recent advancements and the current challenges in bioprinting of hydrogels for tissue engineering.

Review of alginate-based hydrogel bioprinting for application in tissue engineering

Abstract:The dawn of 3D printing in medicine has given the field the hope of vitality in many patients fighting a multitude of diseases. Also entitled bioprinting, this appertains to its sequential printing of precursor ink, embodying cells and polymer/composite in a predetermined trajectory. The precursor ink, in addition to cells, is predominantly constituted of hydrogels due to its biodegradability and ability to mimic the body's anatomy and mechanical features, e.g. bones, etc. This review paper is devoted to explicating the bioprinting (3D/4D) of alginate hydrogels, which are extracts from brown algae, through extrusion additive manufacturing. Alginates are salt derivatives of alginic acid and constitute long chains of polysaccharides, which provides pliability and gelling adeptness to their structure. Alginate hydrogel (employed for extrusion) can be pristine or composite relying on the requisite properties (target application controlled or in vivo environment), e.g. alginate-natural (gelatin/agarose/collagen/hyaluronic acid/etc) and alginate-synthetic (polyethylene glycol (PEG)/pluronic F-127/etc). Extrusion additive manufacturing of alginate is preponderate among others with its uncomplicated processing, material efficiency (cut down on wastage), and outspread adaptability for viscosities (0.03-6 * 104 Pa.s), but the procedure is limited by resolution (200 μm) in addition to accuracy. Abstract:Abstract3D printing technologies, also referred to as additive manufacturing or rapid prototyping have gained attention over the past few years for tissue engineering, the printing of artificial organs and the bioprinting of 3D hydrogels for wound dressings applications. 3D printing can transform the virtual 3D model created by computer-aided design into layer-by-layer physical 3D objects by using different 3D printing technologies. Wounds are a major health fear which affects the lives of people. So, delay in the treatment can lead to the development of chronic wounds which may even lead to mortality. Recently, 3D printing technology is an emerging platform in the development the smart wound dressings which can be loaded with a variety of materials (antibiotics, antibacterial drugs and nanoparticles) that can help accelerate the wound healing rate. Hydrogels have been widely used in wound healing, as they are biocompatible, non-toxic materials. This review mainly focuses on the hydrogel-based biomaterial used in 3D bioprinting and their potential use as a wound healing material. The materials for 3D printing of hydrogels, such as synthetic polymers and natural polymers used as wound dressings are mainly discussed. It covers various 3D bioprinting technologies which include laser printing, stereolithography, extrusion printing, fused disposition printing, and inkjet printing. It also provides a comprehensive overview of the physical, mechanical and biological properties of biomaterial, enabling them as an ideal biomaterial for 3D printing. Abstract:The advent of 3D printing technology in the 1980s enabled the fabrication of patient-specific products with precise shape, size, and complexity. A variety of 3D printing techniques, such as direct ink writing by extrusion, particle fusion printing, light-induced printing, volumetric bioprinting, and inkjet printing, have been explored recently for the purpose of developing desired 3D printed objects. Among these techniques, direct ink writing has been used extensively due to its affordability, ease of use, potential for scaling up, and faster speed. This approach saves time and resources while allowing for the flexible use of various materials. The lack of a variety of varied printing inks, however, continues to be an impediment to its widespread commercial use. Owing to their quick gelling behaviour, extrudability, and shape fidelity, injectable hydrogels have become interesting alternatives for printing inks. Hydrogel inks can be developed by either natural or synthetic polymers, or a combination of the both, to achieve the desired rheological and mechanical properties utilizing various strategies of crosslinking and incorporation of nanoparticles like nanoclay, bioactive glass, graphene oxide and silica nanoparticles. Further, various stimuli responsive hydrogel inks responding to various external stimuli (such as temperature, pH, magnetic field) can be developed to prepare engineered biostructures that can be used in healthcare sector. This review provides a spotlight on recent advancement in developing hydrogel inks and their different physicochemical aspects for developing desired engineered biostructures. Finally, we discuss the opportunities of 4D printing, current difficulties and future prospects for hydrogel inks-based 3D printing to emphasize the significant scope for future innovations.

Abstract:The two fields of continuing professional development (CPD) and knowledge translation (KT) within the health care sector, and their related research have developed as somewhat parallel paths with limited points of overlap or intersection. This is slowly beginning to change. The purpose of this paper is to describe and compare the dominant conceptual models informing each field with the view of increasing understanding and appreciation of the two fields, how they are similar and where they differ, and the current and potential points of intersection. The models include the "knowledge-to-action" (KTA) cycle informing KT, models informing CPD curriculum design and individual self-directed learning, and the Kirkpatrick model for evaluating educational outcomes. When compared through the perspectives of conceptual designs, processes, and outcomes, the models overlap. THE EFFECT OF ZERO HLA CLASS I AND II MISMATCHING IN CYCLOSPORINE‐TREATED KIDNEY TRANSPLANT PATIENTS

Abstract:The effect of HLA matching on one-year first cadaver donor graft survival rates between best and worst matches was 6% (P < 0.001) for A, B; 7% (P < 0.001) for DR; 9% (P < 0.001) for A, DR; 15% (P < 0.001) for B, DR; and 17% (P < 0.001) for A, B, DE. For second cadaver donor grafts, the differences were comparable. Analysis of the cyclosporine-treated patients separately yielded similar results: 5% (NS) for A, B; 7% (P < 0.001) for DR; 13% (P < 0.001) for A, DR; 16% (P < 0.001) for B, DR; and 18% (P < 0.001) for A, B, DR. The most significant effect of matching was achieved by zero mismatching B and DR antigens. The one-year graft survival for patients with zero A, B, DR mismatch was 88% with cyclosporine. Without cyclosporine, zero mismatched A, B, DR grafts survive at 84%; this difference is not statistically significant. Zero mismatching for class I and II antigens (that is, A, DR or B, DR with cyclosporine) gives one-year graft survivals of 84% and 87%, respectively. The zero mismatching HLA class I and II antigen effect is lost ‘when even one antigen is mismatched. Transfusions improved the one-year graft survival 10% in cyclosporine-treated patients, but not in those “who were not treated with cyclosporine. Seventy-one patients transfused with more than 4 units of blood, zero B, DR mismatched, and treated with cyclosporine had a 91% one-year graft survival. Recipient pool sizes for obtaining zero A, B, DR or B, DR mismatched donors are calculated. Zero A, B, DR mismatched patients can be transplanted at a 19% frequency with a 10,000 recipient pool. The success rate for zero mismatching of class I and class II antigens indicates that kidney sharing and large recipient pool sizes are a reasonable policy.

Abstract:Three-dimensional (3D) bioprinting is a rapidly growing technology that has been widely used in tissue engineering, disease studies, and drug screening. It provides the unprecedented capacity of depositing various types of biomaterials, cells, and biomolecules in a layer-by-layer fashion, with precisely controlled spatial distribution. This technology is expected to address the organ-shortage issue in the future. In this review, we first introduce three categories of 3D bioprinting strategies: inkjet-based printing (IBP), extrusion-based printing (EBP), and light-based printing (LBP). Biomaterials and cells, which are normally referred to as “bioinks,” are then discussed. We also systematically describe the recent advancements of 3D bioprinting in fabricating cell-laden artificial tissues and organs with solid or hollow structures, including cartilage, bone, skin, muscle, vascular network, and so on. The development of organs-on-chips utilizing 3D bioprinting technology for drug discovery and toxicity testing is reviewed as well. Finally, the main challenges in current studies and an outlook of the future research of 3D bioprinting are discussed.
