Abstract:Focusing particles into a tight stream is usually a necessary step prior to counting, detecting and sorting them. Meanwhile, particle spacing control in microfluidic devices could also be applied in the field of accurate cell detection, material synthesis and chemical reaction. To achieve simultaneous particle focusing and spacing control, a novel microchannel composed by Dean and sheath flow section was proposed and fabricated according to the elaborated design principle with its manipulating performance in situ visualized. Using microspheres with a few microns as a template, the trajectory of the particles was discovered to follow lateral migration and reach certain equilibrium positions at the end of the designed Dean section. After being focused, the streamline was further concentrated and centralized with a controllable interparticle distance in sheath flow section. For sheath flow section, the angle between symmetrical tributaries and the mainstream channel and abrupt constriction/expansion structure of mainstream channel as important channel geometric features were investigated to minimize the focusing streamline width and optimize spacing control. An modified analytical model for sheath flow with different tributary angles was derived and proved to well describe the microsphere spacing control process.

Abstract:Using inertia effect of fluid to realize the high flux precise control of rigid particles is a novel microfluidic method, which has important applications in materials synthesis, biochemical reactions and medical diagnosis etc.In this study, we designed and fabricated an microfiuidic chip consisted of focusing units and the sheath liquid focusing structures based on the inertial focusing principle.Employing polystyrene microspheres with a diameter of 10 μm as solid particles model to study the focusing effect under certain flow conditions.The experimental results showed that when the flow rate of ethanol reached 710 μL/min, the solid microspheres began to be focusedto form a single flow pattern after flowed through the 60th focusing units.The sheath flow under certain conditions focusing on a single array of microspherescan enhance the focusing effect and controlthe spacing of the particles.The obtainedresults provide an important reference not only for the further study of the kinematic properties of rigid particles in microfluidics, but also for the development of novel microfluidic chips.

Abstract:A novel method based on gravity and electric force driving of cells was developed for flow cytometry and fluorescence activated cell sorting in a microfluidic chip system. In the experiments cells flowed spontaneously under their own gravity in a upright microchip, passed through the detection region and then entered into the sorting electric field one by one at an average velocity of 0.55 mm s(-1) and were fluorescence activated cell sorted (FACS) by a switch-off activation program. In order to study the dynamical and kinematic characteristics of single cells in gravity and electric field of microchannels a physical and numerical module based on Newton's Law of motion was established and optimized. Hydroxylpropylmethyl cellulose (HPMC) was used to minimize cell assembling, sedimentation and adsorption to microchannels. This system was applied to estimate the necrotic and apoptotic effects of ultraviolet (UV) light on HeLa cells by exposing them to UV radiation for 10, 20 or 40 min and the results showed that UV radiation induced membrane damage contributed to the apoptosis and necrosis of HeLa cells.

Abstract:Recently, studies on particle behavior under Newtonian and non-Newtonian fluids in microchannel have attracted considerable attention because particles and cells of interest can be manipulated and separated from biological samples without any external force. In this paper, two kinds of microchannels with non-rectangular cross-section were fabricated using basic MEMS processes (photolithography, reactive ion etching and anisotropy wet etching), plasma bonding and self-alignment between two PDMS structures. They were used to achieve the experiments for inertial and elasto-inertial particle focusing under Newtonian and non-Newtonian fluids. The particle behavior was compared and investigated for different flow rates and particle size in the microchannel with rhombic and equilateral hexagonal cross section. We also investigated the influence of Newtonian fluid and viscoelastic fluid on particle migration in both microchannels through the numerical simulation. The experimental results showed the multi-line particle focusing in Newtonian fluid over a wide range of flow rates, but the single-line particle focusing was formed in the centerline under non-Newtonian fluid. The tighter particle focusing appeared under non-Newtonian fluid in the microchannel with equilateral hexagonal cross-section than in the microchannel with rhombic cross section because of the effect of an obtuse angle. It revealed that particles suspended in the channel are likely to drift toward a channel center due to a negative net elasto-inertial force throughout the cross-sectional area. Simulation results support the present experimental observation that the viscoelastic fluid in the microchannel with rhombic and equilateral hexagonal cross-section significantly influences on the particle migration toward the channel center owing to coupled effect of inertia and elasticity.

Abstract:Viscoelastic focusing has emerged as a promising method for label-free and passive manipulation of micro and nanoscale bioparticles. However, the design of microfluidic devices for viscoelastic particle focusing requires a thorough comprehensive understanding of the flow condition and operational parameters that lead to the desired behavior of microparticles. While recent advancements have been made, viscoelastic focusing is not fully understood, particularly in straight microchannels with rectangular cross sections. In this work, we delve into inertial, elastic, and viscoelastic focusing of biological cells in rectangular cross-section microchannels. By systematically varying degrees of fluid elasticity and inertia, we investigate the underlying mechanisms behind cell focusing. Our approach involves injecting cells into devices with a fixed, non-unity aspect ratio and capturing their images from two orientations, enabling the extrapolation of cross-sectional equilibrium positions from two dimensional (2D) projections. We characterized the changes in hydrodynamic focusing behaviors of cells based on factors, such as cell size, flow rate, and fluid characteristics. These findings provide insights into the flow characteristics driving changes in equilibrium positions. Furthermore, they indicate that viscoelastic focusing can enhance the detection accuracy in flow cytometry and the sorting resolution for size-based particle sorting applications. By contributing to the advancement of understanding viscoelastic focusing in rectangular microchannels, this work provides valuable insight and design guidelines for the development of devices that harness viscoelastic focusing. The knowledge gained from this study can aid in the advancement of viscoelastic particle manipulation technique and their application in various fields.

Abstract:When Segré and Silberberg in 1961 witnessed particles in a laminar pipe flow congregating at an annulus in the pipe, scientists were perplexed and spent decades learning why such behavior occurred, finally understanding that it was caused by previously unknown forces on particles in an inertial flow. The advent of microfluidics opened a new realm of possibilities for inertial focusing in the processing of biological fluids and cellular suspensions and created a field that is now rapidly expanding. Over the past five years, inertial focusing has enabled high-throughput, simple, and precise manipulation of bodily fluids for a myriad of applications in point-of-care and clinical diagnostics. This review describes the theoretical developments that have made the field of inertial focusing what it is today and presents the key applications that will make inertial focusing a mainstream technology in the future.
Inertial Focusing of Spherical Particles: The Effects of Rotational Motion

Abstract:The identification of cells and particles based on their transport properties in microfluidic devices is crucial for numerous applications in biology and medicine. Neutrally buoyant particles transported in microfluidic channels, migrate laterally towards stable locations. However, the effect of the particle and flow properties on these focusing positions remain largely unknown. We conduct large scale simulations with dissipative particle dynamics, demonstrating that freely moving particles exhibit significant differences in their focusing patterns from particles that are prevented from rotation. In circular pipes, we observe drastic changes in rotating versus non-rotating focusing positions. We demonstrate that rotation-induced lateral lift force is significant, unlike previously believed, and is linearly dependent on the rotation magnitude. A simple phenomenological explanation extending existing theories is presented, that agrees well with our numerical findings. For flows in square ducts, we report four face-centered stable positions for the rotating particles, in accordance with experimental studies. However, the non-rotating particles stay scattered on a concentric one-dimensional annulus, thus revealing qualitatively different behavior with respect to the free ones. Our findings suggest new designs for micro-particle and cell sorting in microfluidics devices.

Abstract:Significance of single cell measurements stems from the substantial temporal fluctuations and cell-cell variability possessed by individual cells. A major difficulty in monitoring surface non-adherent cells such as bacteria and yeast is that these cells tend to aggregate into clumps during growth, obstructing the tracking or identification of single-cells over long time periods. Here, we developed a microfluidic platform for long term single-cell tracking and cultivation with continuous media refreshing and dynamic chemical perturbation capability. The design highlights a simple device-assembly process between PDMS microchannel and agar membrane through conformal contact, and can be easily adapted by microbiologists for their routine laboratory use. The device confines cell growth in monolayer between an agar membrane and a glass surface. Efficient nutrient diffusion through the membrane and reliable temperature maintenance provide optimal growth condition for the cells, which exhibited fast exponential growth and constant distribution of cell sizes. More than 24 h of single-cell tracking was demonstrated on a transcription-metabolism integrated synthetic biological model, the gene-metabolic oscillator. Single cell morphology study under alcohol toxicity allowed us to discover and characterize cell filamentation exhibited by different E. coli isobutanol tolerant strains. We believe this novel device will bring new capabilities to quantitative microbiology, providing a versatile platform for single cell dynamic studies.

Abstract:The transport of bio-particles in viscous flows exhibits a rich variety of dynamical behaviour, such as morphological transitions, complex orientation dynamics or deformations. Characterising such complex behaviour under well controlled flows is key to understanding the microscopic mechanical properties of biological particles as well as the rheological properties of their suspensions. While generating regions of simple shear flow in microfluidic devices is relatively straightforward, generating straining flows in which the strain rate is maintained constant for a sufficiently long time to observe the objects' morphologic evolution is far from trivial. In this work, we propose an innovative approach based on optimised design of microfluidic converging-diverging channels coupled with a microscope-based tracking method to characterise the dynamic behaviour of individual bio-particles under homogeneous straining flow. The tracking algorithm, combining a motorised stage and microscopy imaging system controlled by external signals, allows us to follow individual bio-particles transported over long-distances with high-quality images. We demonstrate experimentally the ability of the numerically optimised microchannels to provide linear velocity streamwise gradients along the centreline of the device, allowing for extended consecutive regions of homogeneous elongation and compression. We selected three test cases (DNA, actin filaments and protein aggregates) to highlight the ability of our approach for investigating the dynamics of objects with a wide range of sizes, characteristics and behaviours of relevance in the biological world.

Abstract:Microfluidic devices have been widely used since 1990s for diverse manipulations of particles (a general term of beads, cells, vesicles, drops, etc.) in a variety of applications. Compared to the active manipulation via an externally imposed force field, the passive manipulation of particles exploits the flow-induced intrinsic lift and/or drag to control particle motion with several advantages. Along this direction, inertial microfluidics has received tremendous interest in the past decade due to its capability to handle a large volume of samples at a high throughput. This inertial lift-based approach in Newtonian fluids, however, becomes ineffective and even fails for small particles and/or at low flow rates. Recent studies have demonstrated the potential of elastic lift in non-Newtonian fluids for manipulating particles with a much smaller size and over a much wider range of flow rates. The aim of this article is to provide an overview of the various passive manipulations, including focusing, separation, washing and stretching, of particles that have thus far been demonstrated in non-Newtonian microfluidics.
Oscillatory Viscoelastic Microfluidics for Efficient Focusing and Separation of Nanoscale Species

Abstract:Abstract The ability to precisely control particle migration within microfluidic systems is essential for focusing, separating, counting and detecting a wide range of biological species. To date, viscoelastic microfluidic systems have primarily been applied to the focusing, separation and isolation of micron-sized species, with their use in nanoparticle manipulations being underdeveloped and underexplored, due to issues related to nanoparticle diffusivity and a need for extended channel lengths. To overcome such issues, we herein present sheathless oscillatory viscoelastic microfluidics as a method for focusing and separating both micron and sub-micron species. To highlight the efficacy of our approach, we segment our study into three size regimes, namely micron (where characteristic particle dimensions are above 1 μm), sub-micron (where characteristic dimensions are between 1 μm and 100 nm) and nano (where characteristic dimensions are below 100 nm) regimes. Based on the ability to successfully manipulate particles in all these regimes, we demonstrate the successful isolation of p-bodies from biofluids (in the micron regime), the focusing of λ-DNA (in the sub-micron regime) and the focusing of extracellular vesicles (in the nano-regime). Finally, we characterize the physics underlying viscoelastic microflows using a dimensionless number that relates the lateral velocity (due to elastic effects) to the diffusion constant of the species within the viscoelastic carrier fluid. Based on the ability to precisely manipulate species in all three regimes, we expect that sheathless oscillatory viscoelastic microfluidics will provide for significant new opportunities in a range of biological and life science applications.
Influence of the internal structure of straight microchannels on inertial transport behavior of particles

Abstract:The rapid advancement of Micro-Electro-Mechanical Systems (MEMS) technology has established microfluidics as a pivotal field. This technology marks the onset of a new era in various applications, including drug testing, cell culture, and disease monitoring, underscoring its extensive practicality and potential for future exploration. This research delves into the intricate behavior of particle inertial migration within microchannels, particularly focusing on the impact of different channel structures and Reynolds numbers (Re). Our studies reveal that particles in microchannels with one-sided sharp-cornered microstructures migrate towards the sharp corner at a relative position of 0.4 under low flow rates, and towards the straight wall side at a relative position of 0.8 under high flow rates. The migration pattern of equilibrium positions varies with different arrangements of sharp-corner structures, achieving stability at the channel's center only when the sharp corners are symmetrically arranged on both sides. Our investigation into the shape of microstructures indicates that sharp-cornered structures generate a more stable secondary flow compared to rectangular and semi-circular structures, preventing particle aggregation at the outlet. To address the challenges associated with handling variable cross-section geometries and solid-wall boundaries in dissipative particle dynamics methods effectively, we have developed a dissipative particle dynamics model specifically for analyzing such microchannels. Building upon our previous research, this model introduces a conservative force coefficient for particles within the microstructured region and an interaction zone that only involves repulsive forces, aligning well with experimental outcomes. Through the study of microstructures' geometric shapes, this paper offers guidance for designing microchannels for particle enrichment. Furthermore, the dissipative particle dynamics model established for the particle flow and solid structure interaction within microstructured channels provides insights into the mesoscale dynamics of liquid-solid two-phase flow and particle motion. In conclusion, this paper aims to enhance particle motion sample preparation techniques, thereby broadening the scope of microfluidic applications in the biomedical field.

Abstract:Efficient and reliable manipulation of biological particles is crucial in medical diagnosis and chemical synthesis. Inertial microfluidic devices utilizing passive hydrodynamic forces in the secondary flow have drawn considerable attention for their high throughputs, low costs, and harmless particle manipulation. However, as the dominant mechanism, the inertial lift force is difficult to quantitatively analyze because of the uncertainties of its magnitude and direction. The equilibrium position of particles varies along the migration process, thus inducing the instabilities of particle separation. Herein, we present a designable inertial microfluidic chip combining a spiral channel with periodic expansion structures for the sheathless separation of particles with different sizes. The stable vortex-induced lift force arising from the periodic expansion and the Dean drag force significantly enhanced the focusing process and determined the final equilibrium position. The experimental results showed that over 99% of target particles could be isolated with the high target sample purity of 86.12%. In the biological experiment, 93.5% of the MCF-7, 89.5% of the Hela, and 88.6% of the A549 cells were steadily recovered with excellent viabilities to verify the potential of the device in dealing with biological particles over a broad range of throughputs. The device presented in this study can further serve as a lab-on-chip platform for liquid biopsy and diagnostic analysis.

Abstract:Hydrodynamic flow is an essential stimulus in many cellular functions, regulating many mechanical sensitive pathways and closely associating with human health status and diseases. The flow pattern of blood in vessels is the key factor in causing atherosclerosis. Hemodynamics has great effect on endothelial cells' gene expression and biological functions. There are various tools that can be used for studying flow-induced cellular responses but most of them are either bulky or lack precise controllability. We develop an integrated microfluidic device that can precisely generate different flow patterns to human endothelial cells cultured on-chip. We monitored cell morphology and used small-input RNA-seq technology to depict the transcriptome profiles of human umbilical vein endothelial cells under uni- or bidirectional flow. Such integrated and miniatured device has greatly facilitated our understanding of endothelial functions with shear stimulus, not only providing new data on the transcriptomic scale but also building the connection between cell phenotypic changes and expression alternations.

Abstract:The ability to manipulate and focus particles within microscale fluidic environments is crucial to advancing biological, chemical, and medical research. Precise and high-throughput particle focusing is an essential prerequisite for various applications, including cell counting, biomolecular detection, sample sorting, and enhancement of biosensor functionalities. Active and sheath-assisted focusing techniques offer accuracy but necessitate the introduction of external energy fields or additional sheath flows. In contrast, passive focusing methods exploit the inherent fluid dynamics in achieving high-throughput focusing without external actuation. This review analyzes the latest developments in strategies of sheathless inertial focusing, emphasizing inertial and elasto-inertial microfluidic focusing techniques from the channel structure classifications. These methodologies will serve as pivotal benchmarks for the broader application of microfluidic focusing technologies in biological sample manipulation. Then, prospects for future development are also predicted. This paper will assist in the understanding of the design of microfluidic particle focusing devices.

Abstract:Microfluidics-based cell assays offer high levels of automation and integration, and allow multiple assays to be run in parallel, based on reduced sample volumes. These characteristics make them attractive for studies associated with drug discovery. Controlled delivery of drug molecules or other exogenous materials into cells is a critical issue that needs to be addressed before microfluidics can serve as a viable platform for drug screening and studies. In this study, we report the application of hydrodynamic focusing for controlled delivery of small molecules into cells immobilized on the substrate of a microfluidic device. We delivered calcein AM which was permeant to the cell membrane into cells, and monitored its enzymatic conversion into fluorescent calcein during and after the delivery. Different ratios of the sample flow to the side flow were tested to determine how the conditions of hydrodynamic focusing affected the delivery. A 3D numerical model was developed to help understand the fluid flow, molecular diffusion due to hydrodynamic focusing in the microfluidic channel. The results from the simulation indicated that the calcein AM concentration on the outer surface of a cell was determined by the conditions of hydrodynamic focusing. By comparing the results from the simulation with those from the experiment, we found that the calcein AM concentration on the cell outer surface correlated very well with the amount of the molecules delivered into the cell. This suggests that hydrodynamic focusing provides an effective way for potentially quantitative delivery of exogenous molecules into cells at the single cell or subcellular level. We expect that our technique will pave the way to high-throughput drug screening and delivery on a microfluidic platform.
High throughput, automated prediction of focusing patterns for inertial microfluidics

Abstract:Visual inspections for identifying focusing points in inertial microfluidic flows are prone to misinterpreting stable locations and focusing shifts in the case of non-trivial focusing patterns. We develop and deploy an approach for automating the calculation of focusing patterns for a general channel geometry, and thereby reduce the dependence on empirical/visual procedures to confirm the presence of stable locations. We utilize concepts from interpolation theory (to represent continuous force-fields using discrete points), and stability theory to identify "basins of attraction" and quantitatively identify stable equilibrium points. Our computational experiments reveal that predicting equilibrium points accurately requires upto $\times$10-20 times more refined force-maps that conventionally used, which highlights the spatial resolution required for an accurate representation of cross-sectional forces. These focusing patterns are validated using experimental results for a rectangular channel, and triangular channel with an apex angle of $90^\circ$. We then apply the approach to predict and explain focusing patterns and shifts for a $90^\circ$-isosceles triangular channel across a range of Reynolds numbers for $\frac{a}{H} = 0.4$ (particle-to-channel size ratio). We observe that the predicted focusing patterns match experiments well. The force-maps also reveal certain "clouds" of localized stable points, which aid in explaining the onset of bifurcation observed in experiments. The current algorithm is agnostic to channel cross-sections and straight/curved channels, which could pave way to generating a library of focusing patterns as a function of channel geometry, and $Re$, to assist in design of novel devices for tailored particle-streams.

Hydrodynamic guiding for addressing subsets of immobilized cells and molecules in microfluidic systems

Abstract:BackgroundThe interest in microfluidics and surface patterning is increasing as the use of these technologies in diverse biomedical applications is substantiated. Controlled molecular and cellular surface patterning is a costly and time-consuming process. Methods for keeping multiple separate experimental conditions on a patterned area are, therefore, needed to amplify the amount of biological information that can be retrieved from a patterned surface area. We describe, in three examples of biomedical applications, how this can be achieved in an open microfluidic system, by hydrodynamically guiding sample fluid over biological molecules and living cells immobilized on a surface.ResultsA microfluidic format of a standard assay for cell-membrane integrity showed a fast and dose-dependent toxicity of saponin on mammalian cells. A model of the interactions of human mononuclear leukocytes and endothelial cells was established. By contrast to static adhesion assays, cell-cell adhesion in this dynamic model depended on cytokine-mediated activation of both endothelial and blood cells. The microfluidic system allowed the use of unprocessed blood as sample material, and a specific and fast immunoassay for measuring the concentration of C-reactive protein in whole blood was demonstrated.ConclusionThe use of hydrodynamic guiding made multiple and dynamic experimental conditions on a small surface area possible. The ability to change the direction of flow and produce two-dimensional grids can increase the number of reactions per surface area even further. The described microfluidic system is widely applicable, and can take advantage of surfaces produced by current and future techniques for patterning in the micro- and nanometer scale.

Elasto‐inertial particle focusing in sinusoidal microfluidic channels

Abstract:Dean flow existing in sinusoidal channels could enhance the throughput and efficiency for elasto‐inertial particle focusing. However, the fundamental mechanisms of elasto‐inertial focusing in sinusoidal channels are still unclear. This work employs four microfluidic devices with symmetric and asymmetric sinusoidal channels to explore the elasto‐inertial focusing mechanisms over a wide range of flow rates. The effects of rheological property, flow rate, sinusoidal channel curvature, particle size, and asymmetric geometry on particle focusing performance are investigated. It is intriguing to find that the Dean flow makes a substantial contribution to the particle elasto‐inertial focusing. The results illustrate that a better particle focusing performance and a faster focusing process are obtained in the sinusoidal channel with a small curvature radius due to stronger Dean flow. In addition, the particle focusing performance is also related to particle diameter and rheological properties, the larger particles show a better focusing performance than smaller particles, and the smaller flow rate is required for particles to achieve stable focusing at the outlet in the higher concentration of polyvinylpyrrolidone solutions. Our work offers an increased knowledge of the mechanisms of elasto‐inertial focusing in sinusoidal channels. Ultimately, these results provide supportive guidelines into the design and development of sinusoidal elasto‐inertial microfluidic devices for high‐performance focusing.
