Abstract:Selenium is an essential trace element for humans and obtained from diary diets. The consumption of selenium-rich agricultural food is an efficient way to obtain selenium, but the quality and safety of selenium-rich agro-food are always affected by their associated heavy metals, even poses a potential threaten to human health. In this research, a sampling survey of heavy metals contents in selenium-rich rice was conducted, 182 sets of selenium-rich rice samples were collected from five selenium-rich rice-producing areas of China, and the accumulation of selenium and cadmium were found to be associated in rice and soil. Subsequently, a pot experiment was performed in the greenhouse via treating the soil samples with 12 different concentrations of selenium and heavy metals, and the contents of selenium and cadmium in rice grain were confirmed to be significantly associated. Moreover, transcriptome analysis revealed that the up-regulation of transporter-coding may promote the absorption of selenium and cadmium. The expression of antioxidant-coding genes and cadmium chelator transporter coding-genes was up-regulated to reduce the toxicity of cadmium. Meanwhile, the up-regulation of key genes of the ascorbic acid-glutathione metabolic pathway were responsible for the association between selenium and cadmium in Se-rich rice. Our work suggested the correlation between selenium and cadmium accumulation in selenium-rich rice, clarified their accumulation mechanism, provides a direction for the scientific production of selenium-rich agro-foods.

Abstract:PurposeSilicon (Si) has inhibitory effects on cadmium (Cd) toxicity in rice, but the effects of Si-containing materials on the chemical properties of hydroponic media have been ignored. The increased pH value and introduction of excess cations when utilizing alkaline reagents may disguise the true function of the Si. Thus, the present study investigated Si-alleviated effect on Cd toxicity in rice seedlings by eliminating the aforementioned interferences.Materials and methodsThe hydroponic cultivation of rice seedlings was carried out to demonstrate the inhibiting effects of exogenous Si on Cd accumulation and distribution. The chlorophyll and carotenoid contents in leaf cells under Cd stress were also measured. Transmission electron microscopy was used to investigate ultrastructural changes in rice mesophyll cells, chloroplasts, and mitochondria under different Cd-stressed and Si-amended conditions. The alkalinity of the Si source was neutralized with nitric acid, and discrepancies in Na+ levels was eliminated by the addition of sodium nitrate to different treatments.Results and discussionToxicity symptoms were observed in rice seedlings under Cd (5 mg L−1) treatment after 35 days, and the biomass, and chlorophyll and carotenoid contents, were lowered. The levels of these indices increased after adding 120 mg L−1 Si. The bioconcentration and translocation factor values indicated that Si reduced Cd accumulation in rice plants and also decreased the ratio of Cd translocation from roots to shoots, which effectively alleviated the Cd toxicity. The positive effects of Si were mainly reflected in the recovery of grana lamellae and membrane structures in chloroplasts, and restored membrane morphology and increased numbers of cristae in mitochondria.ConclusionsExogenous Si reduced Cd uptake in rice seedlings, especially in shoots, and inhibited Cd transport into aboveground parts. In addition, Si restored the functions of chloroplasts owing to a decrease in lipid peroxidation. The application of Si indicated that Cd toxicity is alleviated partly because of the improvement in chloroplast biosynthesis and the diminution of Cd bioavailability.

Abstract:Cadmium (Cd), a highly toxic heavy metal for crops in China, poses a significant threat to rice cultivation. It is crucial to identify the genotypes with robust resistance to heavy metals, including Cd, in rice. The experiment was conducted to examine the mitigation effect of silicon (Si) on Cd toxicity levels in Se-enriched Z3055B and non-Se-enriched G46B rice genotypes. A basal dose of Si improved the growth and the quality of rice significantly by reducing the Cd content in rice roots, stems, leaves and grains and increased the yield, biomass and selenium (Se) content of brown rice in both genotypes. Additionally, Se content in brown rice and polished rice was notably higher in Se-enriched rice than in non-Se-enriched rice, with the highest amount at 0.129 mg/kg and 0.085 mg/kg, respectively. The results demonstrated that a basal fertilizer concentration of 30 mg/kg of Si was more effective in reducing Cd transport from roots to shoots in Se-enriched rice than in non-Se-enriched rice genotypes. Therefore, it can be concluded that Se-enriched rice genotypes are a viable option for food crop production in Cd-contaminated areas.

Abstract:Soil cadmium (Cd) pollution has emerged as a pressing concern due to its deleterious impacts on both plant physiology and human well-being. Silicon (Si) is renowned for its ability to mitigate excessive Cd accumulation within plant cells and reduce the mobility of Cd in soil, whereas Selenium (Se) augments plant antioxidant capabilities and promotes rhizosphere microbial activity. However, research focusing on the simultaneous utilization of Si and Se to ameliorate plant Cd toxicity through multiple mechanisms within the plant-rhizosphere remains comparatively limited. This study combined hydroponic and pot experiments to investigate the effects of the combined application of Si and Se on Cd absorption and accumulation, as well as the growth and rhizosphere of A. selengensis Turcz under Cd stress. The results revealed that a strong synergistic effect was observed between both Si and Se. The combination of Si and Se significantly increased the activity and content of enzymes and non-enzyme antioxidants within A. selengensis Turcz, reduced Cd accumulation and inhibiting its translocation from roots to shoots. Moreover, Si and Se application improved the levels of reducing sugar, soluble protein, and vitamin C, while reducing nitrite content and Cd bioavailability. Furthermore, the experimental results showed that the combination of Si and Se not only increased the abundance of core rhizosphere microorganisms, but also stimulated the activity of soil enzymes, which effectively limited the migration of Cd in the soil. These findings provided valuable insights into the effective mitigation of soil Cd toxicity to plants and also the potential applications in improving plant quality and safety.
Modulation of Key Physio-Biochemical and Ultrastructural Attributes after Synergistic Application of Zinc and Silicon on Rice under Cadmium Stress

Abstract:Excessive industrialization and the usage of pesticides plague the farming soils with heavy metals, reducing the quality of arable land. Assessing phytoavailability of cadmium (Cd) from growth medium to plant system is crucial and necessitates precise and timely monitoring of Cd to ensure food safety. Zinc (Zn) and silicon (Si) have singularly demonstrated the potential to ameliorate Cd toxicity and are important for agricultural production, human health, and environment in general. However, Zn-Si interaction on Cd toxicity alleviation, their effects and underlying mechanisms are still fragmentarily understood. Seven treatments were devised besides control to evaluate the single and combined effects of Zn and Si on the physio-biochemical attributes and ultrastructural fingerprints of Cd-treated rice genotypes, i.e., Cd tolerant “Xiushui-110” and Cd sensitive “HIPJ-1”. Supplementation of both Zn and Si promoted plant biomass, photosynthetic parameters, ionic balance, and improved chloroplast ultrastructure with minimized Cd uptake and malondialdehyde (MDA) content due to the activation of antioxidant enzymes in Cd stressed plants. The combined effects of 10 μM Zn and 15 μM Si on 15 μM Cd displayed a greater reduction in Cd uptake and root-leaf MDA content, while enhancing photosynthetic activity, superoxide dismutase (SOD) activity and root-leaf ultrastructure particularly in HIPJ-1, whilst Xiushui-110 had an overall higher leaf catalase (CAT) activity and a higher root length and shoot height was observed in both genotypes compared to the Cd 15 μM treatment. Alone and combined Zn and Si alleviation treatments reduced Cd translocation from the root to the stem for HIPJ-1 but not for Xiushui-110. Our results confer that Zn and Si singularly and in combination are highly effective in reducing tissue Cd content in both genotypes, the mechanism behind which could be the dilution effect of Cd due to improved biomass and competitive nature of Zn and Si, culminating in Cd toxicity alleviation. This study could open new avenues for characterizing interactive effects of simultaneously augmented nutrients in crops and provide a bench mark for crop scientists and farmers to improve Cd tolerance in rice.

Abstract:Cadmium (Cd) toxicity negatively impacts plant health and productivity. Nanosilica (SiO2NPs) and salicylic acid (SA) enhance plant performance and alleviate heavy metals stress. Yet, their combined effects against Cd-toxicity in rice remained less-explored. Thus, a hydroponic study investigated the individual and combined effects of SiO2NPs and SA on Cd-stress mitigation in rice at physio-biochemical, cellular, and molecular levels. Results indicated that Cd-alone treatment caused a significant reduction in rice growth and biomass and photosynthetic efficiency, which was associated with oxidative damage caused by enhanced Cd-accumulation in plant tissues. Cd-induction also potentiated its phytotoxicity by triggering enzymatic antioxidants against the extra production of reactive oxygen species (ROS). The addition of SiO2NPs and/or SA markedly minimized the Cd-induced toxicity by reducing Cd-bioaccumulation (42–56%), protecting photosynthetic efficiency, which were directly correlated with seedling biomass and restored cellular structures (leaf ultrastructure and surface morphology). The combined application of SiO2NPs and SA was more effective in activating antioxidant enzymes, phytohormones biosynthesis, and reducing oxidative damages caused by Cd than sole application. This was evident in the decreased production of ROS, malondialdehyde contents (29–37%), and recovered membrane stability. Moreover, SiO2NPs and/or SA relieved Cd-bioaccumulation (41–56%) by downregulating the Cd-related transporter genes (OsNramp1, OsNramp5, OsHMA2, and OsHMA3). Altogether, the cellular Cd-accumulation, photosynthesis, antioxidant defense, and phytohormones against oxidative stress can be ideal markers for cultivating rice in Cd-contaminated soils.

Abstract:Hydroponic experiments were performed to investigate physiological mechanisms of selenium (Se) mitigation of Cd toxicity in rice. Exogenous Se markedly reduced Cd concentration in leaves, roots, and stems. Addition or pretreatment of 3μM Se in 50μM Cd solution significantly addressed Cd-induced growth inhibition, recovered root cell viability, and dramatically depressed O2−, H2O2, and malondialdehyde (MDA) accumulation. Supplemental Se counteracted 50μM Cd-induced alterations of certain antioxidant enzymes, and uptake of nutrients, e.g. depressed Cd-induced increase in leaf and root superoxide dismutase (SOD) and leaf peroxidase (POD) activities, but elevated depressed catalase (CAT) activity; decreased Cd-induced high S and Cu concentrations in both leaves and roots. External Se counteracted the pattern of alterations in ATPase activities induced by Cd, e.g. significantly elevated the depressed root H+- and Ca2+-ATPase activities, but decreased the ascent root Na+K+-ATP activity. Results indicate that alleviated Cd toxicity by Se application is related to reduced Cd uptake and ROS accumulation, balanced nutrients, and increased H+- and Ca2+-ATPase activities in rice.
Silicon-Selenium Interplay Imparts Cadmium Resistance in Wheat through an Up-Regulating Antioxidant System

Abstract:Cadmium (Cd), being a highly toxic heavy metal, significantly impacts plant growth and development by altering nutrient uptake and causing oxidative and structural damage, resulting in reduced yield. To combat Cd toxicity and accumulation in wheat, it was hypothesized that co-application of Selenium (Se) and Silicon (Si) can reduce the adverse effect of Cd and regulate Cd resistance while improving Se fortification in wheat. Therefore, this study evaluated the comparative effect of Se and Si on the growth and antioxidant defense systems of wheat plants grown in a hydroponic setup. Briefly, the plants were acclimatized to the hydroponic solution for 1 week and then exposed to 10 µmol Cd. Afterwards, the treatments, including 0.2 mmol Si and 1.5 µmol Se, were applied as a root and foliar application, respectively. Plants supplemented with both Se and Si showed improved biomass and other physiological growth attributes, and this response was associated with improved activity/contents of antioxidants, including glutathione (GSH) content, glutathione reductase (GR), ascorbate peroxidase (APX), and catalase (CAT), with related lowering of hydrogen peroxide, malondialdehyde content, and structural damages. Moreover, by Se + Si supplementation, a decrease in total S levels in plant tissues was observed, whereas an increase in total protein concentration and GSH indicated a different and novel mechanism of Cd tolerance and S homeostasis in the plant. It was observed that Si was more involved in significantly reducing Cd translocation by stabilizing Cd in the root and reducing its content in the soluble fraction in both the root and shoot. Whereas Se was found to play the main role in reducing the oxidative damage caused by Cd, and the effect was more profound in the shoot. In addition, this study also observed a positive correlation between Si and Se for relative uptake, which had not been reported earlier. Our findings show that the Se and Si doses together benefit growth regulation and nutrient uptake; additionally, their combinations support the Cd resistance mechanism in wheat through upregulation of the antioxidant system and control of Cd translocation and subcellular distribution, ultimately contributing to the nutritional quality of wheat produced. Thus, it is concluded that the co-application of Se and Si has improved the nutritional quality while reducing the Cd risk in wheat and therefore needs to be employed as a potential strategy to ensure food safety in a Cd-contaminated environment.
Effect of selenium on the subcellular distribution of cadmium and oxidative stress induced by cadmium in rice (Oryza sativa L.)

Abstract:Cadmium (Cd) is absorbed readily by rice plants and is transferred to humans when contaminated rice is consumed. Adding selenium (Se) to the plant nutrient solutions reduces the accumulation of Cd in the rice (Oryza sativa L.) seedlings. However, as the relevant underlying mechanism remains unclear, the aim of our study was to improve our understanding of the Se-mediated resistance to Cd stress in rice. We conducted hydroponic experiments to study the effects of selenite or selenate on Cd subcellular distribution and xylem transport in rice seedlings under Cd stress, and we investigated the antioxidative defense responses in the rice plants. We found that the supplementation of both Se forms decreased the Cd accumulations in the roots and shoots of the rice plants. The selenite addition significantly decreased the Cd contents in different subcellular fractions of the rice roots, increased the proportion of Cd distributed to soluble cytosol by 23.41%, and decreased the Cd distribution in the organelle by 28.74% in contrast with the treatment with Cd only. As regards the selenate addition, only the Cd distribution ratio of cytosol was increased by 13.07%. After adding selenite, a decrease of 55.86% in the Cd concentration in xylem sap was observed, whereas little change was found after treatment co-applied with selenate. The hydrogen peroxide (H2O2) and malondialdehyde(MDA) contents in the rice roots were elevated under Cd stress, and the addition of selenite and selenate decreased the H2O2 levels by 77.78% and 59.26%, respectively. Co-exposure to Cd and Se elevated the glutathione (GSH) accumulations in the rice shoots and roots, with the degree of increase being the following: co-applied with selenite > co-applied with selenate > Cd alone treatment. Exposure to Cd increased the catalase (CAT) activity in the roots significantly, whereas it decreased in the shoots. After selenite or selenate supplementation, the CAT activity in the rice roots increased compared with applying only Cd. Compared with the control, the addition of Cd or Se had no significant effect on the activities of peroxidase (POD) or ascorbate peroxidase (APX). Our results showed that Se affected the Cd accumulation in rice seedlings by altering the Cd subcellular distribution and decreasing the ROS induced by Cd stress. Such effects were more significant in the selenite than in the selenate applied treatment.

Abstract:Cadmium (Cd) is a pollutant toxic to plants and a potential threat to human health. Selenium (Se), though not essential for plants, has beneficial effects on plants under abiotic stress. A hydroponic experiment was conducted to investigate the impact of different forms of Se (Nano-Se, selenite, selenate, and SeMet) on accumulation, subcellular distribution, and chemical forms of Cd, as well as oxidative stress in rice seedlings. Cd (20 μmol·L−1) treatment significantly decreased biomass accumulation and chlorophyll content. The application of all Se forms, except selenate, mitigated the adverse effects of Cd on growth and chlorophyll content. The application of selenite, Nano-Se, and SeMet decreased root and shoot Cd concentrations as well as root-to-shoot Cd translocation in rice seedlings. Selenate application decreased shoot Cd concentration and root-to-shoot Cd translocation with no effect on root Cd concentration. Accordingly, Se increased the sequestration of Cd in the cell wall and vacuoles and decreased the active chemical form of Cd in rice seedlings. SeMet was the most effective supplement that decreased Cd concentration and enhanced Se concentration in the roots and shoots of rice seedlings. All forms of Se further enhanced catalase (CAT) and glutathione peroxidase (GSH-Px) activities and inhibited MDA accumulation. To conclude, Se influenced Cd accumulation and translocation in rice seedlings by altering the subcellular distribution, chemical forms, and antioxidant defense system under Cd stress. These effects were highly significant with SeMet treatment, probably due to better absorption and utilization by the plant.

Abstract:Selenium (Se), iron (Fe), and humic acid (HA) are beneficial fertilizers that inhibit cadmium (Cd) uptake in crops and are crucial for agricultural yields as well as human health. However, the joined effect of Se, Fe, and HA on Cd uptake in rice are still poorly understood. Therefore, a hydroponic culture experiment was established to evaluate the combined effect of Se (Se4+ or Se6+), Fe, and HA on the biomass, Cd uptake, and Cd translocation of/in rice seedlings. Compared to Se6+ application, Se4+ application in most treatments resulted in lower Cd translocations from roots to shoots, leading to a significant decrease in shoot Cd concentrations. Compared to the treatments with Se4+ or Fe2+ application, joined application of Se4+ and Fe2+ inhibited Cd uptake in shoots by decreasing Cd adsorption onto (iron plaque) and uptake by roots, and alleviating Cd translocation from root to shoot. Compared to the treatments with Se6+ or Fe2+ application, joined application of Se6+ and Fe2+ inhibited Cd uptake in shoots by sequestering (retaining) Cd onto root surface (iron plaque). HA inhibited Cd uptake in all treatments by decreasing the bioavailability of Cd in the nutrient solution through complexation. The simultaneous application of Se, Fe, and HA decreased the shoot Cd concentrations the most, followed by the combined application of two fertilizers and their individual application; the mean shoot Cd concentration in the Fe-SeIV-HA2 treatment was the lowest among all the treatments, at only 11.39 % of those in the control treatments. The 3-way ANOVA results indicated that the Cd concentrations in shoots were significantly affected by Se, Fe, HA, and certain of their interactions (Fe×Se and Se×HA) (p< 0.05). The above findings suggest that the joined application of Se, Fe, and HA ameliorated Cd uptake mainly by inhibiting Cd adsorption onto (iron plaque) and uptake by roots and the translocation from roots to shoots (Fe×Se4+), retaining (sequestering) Cd in iron plaque (Fe×Se6+), and decreasing Cd availability in nutrient solution (HA).Graphical abstract

Abstract:Growth chamber studies were conducted to investigate the effects of silicon (Si) on the distribution of Cd in rice seedlings (Oryzasativa L., cv. Qiu Guang) grown hydroponically under toxic level of cadmium (Cd). Si added significantly alleviated the toxicity of Cd in aerobic rice seedlings. Si partly overcame the reduction in growth due to Cd. This amelioration was correlated with a reduction in Cd uptake. Si increased Cd accumulation in the roots and restricted the transport of Cd from roots to shoots, where the distribution of Cd in the shoots decreased by 33%. Si reduced the transport of Cd and the apoplastic fluorescence tracer PTS (tri-sodium-8-hydroxy-1, 3, 6-pyrenesulphonate) from roots to shoots by 23 and 36%, respectively. Energy-dispersive X-ray analysis (EDX) showed Cd was mainly deposited in the vicinity of the endodermis and epidermis, Si deposition was heavier in the vicinity of the endodermis than in the epidermis. Although the tracing result of fluorescein isothiocyanate-dextrans showed Si did not change epidermal wall porosity, the significant reduction of apoplastic PTS transport in +Si plants suggested that the heavy deposition of silica in the vicinity of endodermis might offer possible mechanisms by which silicon did at least partially physically block the apoplast bypass flow across the roots, and restrained the apoplastic transport of Cd. In addition, the effect of Si on the subcellular distribution and chemical form of Cd was investigated by fractionation. Si decreased the concentrations of Cd in shoots and roots, but did not remarkably change the distribution ratio of Cd in symplasm and apoplast. Mechanisms by which Si alleviates the toxicity of Cd in rice seedlings are discussed.

However, the effects of Si, Se and their interactions in reducing cadmium (Cd) toxicity and the related mechanisms require further elucidation. Hence, this study implemented a foliar application of Si and Se on soybean (Glycine max L.) that subjected to Cd-induced stress with four treatments (sole/combined application of Si, Se, no fertilizer treatment). The results demonstrated that Si and Se showed effective mitigation of Cd toxicity on soybeans mainly by promoting growth, enhancing photosynthesis, maintaining root vigor, improving antioxidant capacity, alleviating oxidative damage, altering the storage form, subcellular distribution of Cd in soybeans, and was more noticeable when combined overall (Si + Se＞Se＞Si). Si + Se increased root activity by 28% and CAT activity in leaves by 130.65%. Overall, the combined application of Si and Se exhibited a pronounced synergistic effect in enhancing the healthy growth of soybean plants under Cd pollution, with a more prominent impact observed following the second fertilization.

Abstract:Silicon (Si) and Zinc (Zn) have been frequently used to alleviate cadmium (Cd) toxicity, which are feasible strategies for crop safety production. However, the mechanisms underlying the interaction of Si and Zn on alleviating Cd toxicity are not well understood. A hydroponic system was adopted to evaluate morphological, physiological-biochemical responses, and related gene expression of wheat seedlings to Si (1 mM) and Zn (50 µM) addition under Cd stress (10 µM). Cd induced obvious inhibition of wheat growth by disturbing photosynthesis and chlorophyll synthesis, provoking generation of reactive oxygen species (ROS) and interfering ion homeostasis. Cd concentration was decreased by 68.3%, 43.1% and 73.3% in shoot, and 78.9%, 44.1% and 85.8% in root by Si, Zn, and combination of Si with Zn, relative to Cd only, respectively. Si and Zn effectively ameliorated Cd toxicity and enhanced wheat growth; but single Si or combination of Si with Zn had more efficient ability on alleviating Cd stress than only Zn, indicating Si and Zn have synergistic effect on Cd toxicity; Interaction of them alleviated oxidative stress by reducing ROS content, improving AsA-GSH cycle and antioxidant enzymes activities, and regulating Cd into vacuole through PC-Cd complexes transported by HMA3 transporter. Our results suggest that fertilizers including Si and Zn should be made to reduce Cd content, which will beneficial for food production and safety.
Exogenous selenium promotes cadmium reduction and selenium enrichment in rice: Evidence, mechanisms, and perspectives

Abstract:Cadmium (Cd) accumulation in rice, a global environmental issue, poses a significant threat to human health due to its widespread presence and potential transfer through the food chain. Selenium (Se), an essential micronutrient for humans and plants, can reduce Cd uptake in rice and alleviate Cd-induced toxicity. However, the effects and mechanisms of Se supplementation on rice performance in Cd-contaminated soil remain largely unknown. Here, a global meta-analysis was conducted to evaluate the existing knowledge on the effects and mechanisms by which Se supplementation impacts rice growth and Cd accumulation. The result showed that Se supplementation has a significant positive impact on rice growth in Cd-contaminated soil. Specifically, Se supplementation decreased Cd accumulation in rice roots by 16.3 % (11.8-20.6 %), shoots by 24.6 % (19.9-29.1 %), and grain by 37.3 % (33.4-40.9 %), respectively. The grain Cd reduction was associated with Se dose and soil Cd contamination level but not Se type or application method. Se influences Cd accumulation in rice by regulating the expression of Cd transporter genes (OSLCT1, OSHMA2, and OSHMA3), enhancing Cd sequestration in the cell walls, and reducing Cd bioavailability in the soil. Importantly, Se treatment promoted Se enrichment in rice and alleviated oxidative damage associated with Cd exposure by stimulating photosynthesis and activating antioxidant enzymes. Overall, Se treatment mitigated the health hazard associated with Cd in rice grains, particularly in lightly contaminated soil. These findings reveal that Se supplementation is a promising strategy for simultaneous Cd reduction and Se enrichment in rice.

Abstract:This study was carried out to investigate the effects of selenium (Se) on the uptake and translocation of cadmium (Cd) and essential elements in paddy rice (Oryza sativa L., Shuangyou 998). Selenium could alleviate/aggravate Cd toxicity in paddy rice, which depended on the dosages of Se and/or Cd. When Cd treatment level was as low as 35.6 μM, ≤12.7 μM Se could inhibit the uptake of Cd in paddy rice and increase the biomass of paddy rice; however, with Cd levels reaching 89–178 μM, the addition of Se resulted in increases in Cd uptake and exacerbated the growth of paddy rice. Cd always inhibited the uptake of Se. Cd alone suppressed the uptake of Ca, Mg, Mn, Cu, and Zn; however, Se reversed the decreases in the concentrations of the said elements, suggesting an element regulation mechanism to relieve Cd toxicity. Without Cd in the solution, low doses of Se increased the biomasses of shoots and roots at the expense of the more or less decreases in the concentrations of Ca, Mg, K, Fe, Mn, Cu, and shoot Zn, indicating an antagonistic effect of Se on these cations. The presence of Cd could also reverse these decreases especially at the highest treatment levels for both Se and Cd, also suggesting an element regulation mechanism responsible for the detoxification of high dosages of Se. Consequently, when Se is used to alleviate Cd toxicity, attention must be paid to the Cd pollution extent and doses of Se supplement.

Abstract:Cadmium (Cd) contamination has resulted in serious reduction of crop yields. Silicon (Si), as a beneficial element, regulates plant growth to heavy metal toxicity mainly through reducing metal uptake and protecting plants from oxidative injury. However, the molecular mechanism underlying Si-mediated Cd toxicity in wheat has not been well understood. This study aimed to reveal the beneficial role of Si (1 mM) in alleviating Cd-induced toxicity in wheat (Triticum aestivum) seedlings. The results showed that exogenous supply of Si decreased Cd concentration by 67.45% (root) and 70.34% (shoot), and maintained ionic homeostasis through the function of important transporters, such as Lsi, ZIP, Nramp5 and HIPP. Si ameliorated Cd-induced photosynthetic performance inhibition through up-regulating photosynthesis-related genes and light harvesting-related genes. Si minimized Cd-induced oxidative stress by decreasing MDA contents by 46.62% (leaf) and 75.09% (root), and helped re-establish redox homeostasis by regulating antioxidant enzymes activities, AsA-GSH cycle and expression of relevant genes through signal transduction pathway. The results revealed molecular mechanism of Si-mediated wheat tolerance to Cd toxicity. Si fertilizer is suggested to be applied in Cd contaminated soil for food safety production as a beneficial and eco-friendly element.

Abstract:Ghrelin, a 28-amino-acid acylated peptide, produced mainly by the stomach, displays strong growth hormone-(GH)-releasing activity mediated by the hypothalamus-pituitary growth hormone potential secretagogue (GHS) receptor which had been shown to be specific for a family of synthetic, orally active GHS. GHS are reliable provocative tests for the diagnosis of GH deficiency but, as orally active growth-promoting agents, they are not comparable with human recombinant GH in terms of efficacy. The usefulness of GHS in anabolic, anti-ageing drug intervention in the somatopause is still unclear. GHS also act on central and peripheral receptors and show other actions, including an orexigenic effect, an influence on gastroentero-pancreatic functions, and cardiovascular and anti-proliferative effects. Ghrelin mediates the neuroendocrine and metabolic response to starvation. Taking into account its orexigenic effect, GHS analogues acting as agonists or antagonists on appetite could represent a new drug intervention for eating disorders.
Cross-Talk between Cadmium and Selenium at Elevated Cadmium Stress Determines the Fate of Selenium Uptake in Rice

Abstract:Cadmium (Cd) is a well-known metal imposing threats to human health, and it can be accumulated in polished rice over the permitted range of 0.2 mg kg−1 (GB 2762-2017). It has been reported that selenium (Se) application decreases Cd uptake. Se-rich diets have gained attention recently, but the potential of Se-rich rice in mitigating Cd stress needs further investigation. In this study, a pot experiment in the field was conducted to assess the influence of environmental factors and exogenous split application of Se on the nutritional status of rice under Cd stress. The results indicated that the increased fertilizer treatment in soil bulk linearly increased the metal content in rice grains. Approximately 50–70% of metal was recovered in rice tissues, while 5–20% of the metal that was applied leached down into the soil. A Se concentration of 0.4 mg kg−1 could significantly improve the total Se content in grain and mitigate Cd toxicity (1 mg kg−1) below the permitted range. Panicles and roots were more active for total Se accumulation in Se-rich and non-Se-rich rice, respectively. Polishing and milling operations can significantly reduce the Cd content, as rice bran in rice tissues accumulated most of the metal’s residues. The late matured rice cultivars consumed more heat units, and more metal contents were found in them. Collectively, it was found that Se can mitigate Cd toxicity, but the rice cultivation at T2 (high Cd; 2 mg kg−1 and Se; 1 mg kg−1) increased the metal uptake capability and health-risk index in polished rice, with its Se content heightened over permitted range of 0.04 to 0.30 mg kg−1 (GB/T 22499-2008). However, further molecular studies are required, in order to completely access the inverted Se accumulation behavior in rice tissues at high Cd soil stress.

Abstract:Exogenous Si mitigates the mobility and bioavailability of Cd in the soil, thereby alleviating its phytotoxicity. This study focused on specific Si-induced immobilisation effects within the rhizosphere (S1), near-rhizosphere (S2), and far-rhizosphere (S3) zones. Based on the rhizobox experiment, we found that applying Si significantly elevated soil pH, and the variation amplitudes in the S3 soil exceeded those in the S1 and S2 soils. Si-induced changes in the rhizosphere also included enhanced dissolved organic carbon and diminished soil Eh, particularly in the Si400 treatment. Meanwhile, the introduction of Si greatly enhanced the Fe 2+ and Mn 2+ concentrations in the S1 soil, but reduced them in the S2 soil. The rhizosphere effect of Si which enriched Fe 2+ and Mn 2+ subsequently promoted the formation of Fe and Mn oxides/hydro-oxides near the rice roots. Consequently, the addition of Si significantly reduced the available Cd concentrations in S1, surpassing the reductions in S2 and S3. Moreover, Si-treated rice exhibited increased Fe plaque generation and fixation on soil Cd, resulting in decreased Cd concentrations in rice tissues, accompanied by reduced Cd translocation from roots to shoots and shoots to grains. Structural equation modelling further highlighted that Si is essential in Cd availability in S1 and Fe plaque development, ultimately mitigating Cd accumulation in rice. Si-treated rice also exhibited higher biomass and grain yield than those of control groups. These findings provide valuable insights into Si-based strategies for addressing the Cd contamination of agricultural soils.
