Abstract:To study the effects of Bacillus subtilis supplementation in a high-fat diet on the gut microbiota and nonalcoholic fatty liver disease in grass carp (Ctenopharyngodon idella), juveniles (60 ± 5 g) were fed three diets: (a) a control diet (CON), (b) a high-fat diet (HFD) and (c) a high-fat diet supplemented with B. subtilis (HFD + BS). After 8 weeks of feeding, fish growth, serum biochemical indices and total liver lipid content were measured, and gut microbiota analysis was performed using the MiSeq250 high-throughput sequencing platform. The results of this study showed that B. subtilis could improve growth and blood serum indices and reduce lipid deposition in the fish liver, preventing fatty liver disease. A grass carp model of fatty liver induced by a high-fat diet was successfully established. Moreover, B. subtilis altered the intestinal microbiota of HFD-fed grass carp, making it more similar to that of the control group. This study revealed the important effects of B. subtilis on grass carp with fatty liver induced by a high-fat diet and provides the foundation for the application of probiotics in grass carp farming.

Abstract:Bacillus subtilis is widely used in aquaculture as a probiotic. However, few studies have been conducted to examine the effect of B. subtilis on liver lipid metabolism. A total of 135 healthy grass carp (50.24 ± 1.38 g) were randomly divided into three groups: control (Con), high-fat diet (HF), and high-fat diet + B. subtilis (HF + B. subtilis), and fed for 8 weeks. The results showed that compared with the HF group, the weight gain rate (WGR) significantly increased (P < 0.05) and the hepatic lipid content, serum low-density lipoprotein cholesterol (LDL-C), and aspartate aminotransferase (AST) decreased in the group supplemented with B. subtilis (P < 0.05). Moreover, the hepatic mRNA expression of fatty acid synthase (FAS) was significantly down-regulated and the carnitine palmitoyl transferases (CPT1α1a) were up-regulated in the HF + B. subtilis group compared to the HF group (P < 0.05), respectively. Additionally, in the HF + B. subtilis group, glutathione (GSH) significantly increased (P < 0.05), while hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents significantly decreased compared to the HF group (P < 0.05). B. subtilis may reduce the hepatic lipid content by inhibiting its synthesis and promoting β-oxidation of fatty acids. B. subtilis may also alleviate dyslipidaemia and prevent oxidative damage in the liver caused by the high-fat diet of grass carp. Hence, dietary supplementation with B. subtilis shows promise as a therapeutic or preventive tool against fatty liver disease.

Several indicators of immunity and antioxidant activities improved in grass carp given a diet containing bacillus additive

Abstract:The effect of Bacillus additive on immunity and antioxidant activities of grass carp (Ctenopharyngodon idellus) was studied in this experiment. Animals of the control group were fed the basal diet and those of treatment group were fed the basal diet augmented with Bacillus additive at a dose of 10(5) Colony Forming Units per gram (CFU g(-1)) feed. Data showed that serum immunity parameters including IgM, complement C3 and Alkaline Phosphatase (AKP) significantly increased (p<0.05) by 30.77, 34.62 and 107.79% in the treatment group. However, there was no significant difference in serum total protein, globulin, albumin, A/G ratio, Lysozyme (LZM), Glutamic Oxaloacetic Transaminase (GOT) and Glutamic Pyruvic Transaminase (GPT) activities. Serum antioxidant parameters such as serum Gluthathione (GSH), Total Antioxidant Capacity (T-AOC), Superoxide Dismutase (SOD) and Gluthathione Peroxidase (GSH-Px) activities significantly (p<0.05) enhanced by 32.76, 41.75, 20.06 and 11.30%, respectively and hepatopancreatic antioxidant parameters including SOD activity and GSH increased significantly (p<0.05) by 33.83 and 25.86%. In addition, GSH-Px and Malondialdehyde (MDA) content in hepatopancreatic decreased by 20.69% (p<0.05) and 25.39% (p<0.05) individually and the anti-superoxide anion (anti-O-2(-)) activity in hepatopancreatic decreased by 32.21% (p<0.05) in the treatment group. These results indicate that the Bacillus addtive can promote the immunity and antioxidant function of grass carp.

Abstract:The effects of dietary Bacillus subtilis on growth performance,immunity and antioxidant function of grass carp(Ctenopharyngodon idellus) were studied in this experiment.Three hundred healthy grass carp with an average body weight of(51.0±2.3) g were randomly divided into 2 groups(control group and experimental group) with 3 replicates per group and 50 fish per replicate.The fish in the control group and experimental group were fed a basal diet and an experimental diet(the basal diet supplemented with B.subtilis at the dose of 1×105 CFU/g),respectively.The experiment lasted for 45 d.The results showed as follows: the ratio of weight to gain and special growth ratio of the experimental group were significantly higher than those of the control group(P<0.05).Compared with the control group,the contents of immunoglobulin M(IgM) and complement 3(C3) and the activity of serum alkaline phosphatase(AKP) of the experimental group were significantly increased(P<0.05),while the activities of myeloperoxidase(MPO) and glutamic pyruvic transaminase(GPT) of the experimental group were significantly decreased(P<0.05).However,the contents of serum total protein,globulin and albumin,albumin to globulin ratio,the activities of lysozyme(LZM) and glutamic oxalacetic transaminase(GOT) were not affected by B.subtilis supplementation(P>0.05).Compared with the control group,the total anti-oxidation capability(T-AOC),the activities of catalase(CAT) and superoxide dismutase(SOD) in serum and hepatopancreas,and the hepatopancreas GSH content of the experimental group were significantly increased(P<0.05),while the malondialdehyde(MDA) content and anti-O-2· activity in serum and hepatopancreas of the experimental group were significantly decreased(P<0.05).These results indicate that dietary B.subtilis can significantly promote the immunity and antioxidant function to increase the growth of grass carp.

Abstract:This study investigated whether Bacillus subtilis can provide protection for grass carp against oxidative stress damage induced by Aeromonas hydrophila. A total of 240 healthy grass carp (Ctenopharyngodon idellus) (average weight of 71.42 ± 4.36g) were randomly divided into four groups with three replicates: control group, A. hydrophila group, B. subtilis + A. hydrophila group, and A. hydrophila + B. subtilis group. After challenge with A. hydrophila, the lipid oxidative damage, antioxidant defenses, and the gene expression of inflammatory cytokines of the grass carp were investigated. Our results showed that A. hydrophila caused lipid oxidative damage, led to significant decreases in antioxidant defenses, and induced inflammatory responses of grass carp. However, the grass carp group fed the probiotic B. subtilis diet for 42 days before the challenge and the group fed the probiotic B. subtilis diet immediately after the challenge both showed (i) a reduced level of oxidative stress with a decrease in the level of MDA; (ii) an increase in antioxidant defenses, including an increase in total antioxidant capacity (T-AOC), increased activities of SOD and CAT, increased levels of GSH, and upregulated gene expression of antioxidant enzymes (SOD, CAT, and Gpx); and (iii) an improved immune response with the level of antiinflammatory cytokines IL-10 messenger RNA (mRNA) upregulated and the levels of pro-inflammatory cytokines TNF-α, IL-1β, and IL-8 mRNA downregulated. Based on this study, B. subtilis can provide effective protection of fish against oxidative stress damage induced by A. hydrophila infection.

Abstract:Gut microbiota of grass carp plays an important role in host. However, detailed information regarding the changes of microbiota after probiotics administration in relation to the gastrointestinal microbiota is absent. In the present study, dietary administration of putative probiotics Shewanella xiamenensis A-1, Aeromonas veronii A-7, and Bacillus subtilisstrains was conducted in grass carp to investigate if there is a discernible alteration in intestinal microbiota and whether the alteration is associated with previous study about the immunity regulation in grass carp. Bacterial 16S rRNA gene sequence-based comparisons of the bacterial communities in the grass carp intestine were detected after 28 days feeding by five diets, and results demonstrated the changes of microbial community composition at genus level. The abundance of Cetobacterium genus with potential immunity function increased. Potential pathogens and probiotics are important constitutions of the intestinal microbiota. Orally taken probiotics considerably reduced the abundance of the potential pathogenic bacteria (e.g., Pseudomonas and Flavobacterium genus) in the intestine. Meanwhile, putative probiotics used in this study were favorable to the reproduction of potential probiotics in THE intestine of grass carp (e.g., Vibrio, Streptococcus, and Enterococcus genus). Moreover, modulation of intestinal environment by the probiotics could impact the abundance of cellulose-degrading bacteria (e.g., Citrobacter genus). Those results suggested that oral probiotics administration can positively improve the composition of intestinal microbial community in grass carp, and this was associated with regulation of immunity in grass carp. Probiotics-induced alteration of microbiota may potentially lower the risk of disease outbreaks during cultivation stage of grass carp.

Abstract:A feeding trial was performanced at water temperature of 18-25 ℃ to investigate the effects of dietary bacterium Bacillus subtilis on digestive enzyme activity and intestinal microflora in grass carp Ctenopharyngodon idellus with initial body weight of 51.37 g±0.58 g.Three hundred individuals of grass carp were randomly divided into two treatments with three replicate tanks at a rate of 50 fish per tank,and fed the basal diet in the control group,and the basal diet supplemented with bacterium Bacillus subtilis in the treatment groups three times a day at 8:00 hour,12:00 hour and 19:00 hour at a feeding rate of about 3% for 45 days.There were very significantly 14.75% higher trypsin,13.03% higher lipase and 78.95% higher amylase activities in the intestinal chyme in the fish fed the diets containing Bacillus subtilis(P0.01)compared with those in the control group.The fish fed the diets containing Bacillus subtilis had 17.72% higher lipase and 13.81% higher trypsin activities in hepatopancreas(P0.01).There was significantly higher number of Bacillus sp.and Pseudomonas sp.in the intestine in the treated groups(P0.01),while there was significantly lower number of E.coli and Vibrios than that in the control group(P0.01),indicating that supplementation of Bacillus subtilis in diets led to increase in the digestive enzyme activities and improvement of the intestinal microflora in grass carp.
Bacillus Subtilis SC02 Supplementation Causes Alterations of the Microbial Diversity in Grass Carp Water

Abstract:This study was conducted to evaluate the effects of Bacillus subtilis SC02 supplement on water quality, microbial community diversity and structure in a grass carp (Ctenopharyngodon idellus) culture. Our selected strain, B. subtilis SC02, significantly reduced ammonia, nitrite and total nitrogen levels in water over an extended period compared with the control group. Pyrosequencing showed that the Shannon diversity index (Shannon) and species richness estimators (Chao) of the treatment group were higher, indicating that bacterial richness was significantly increased in the treatment group. The phyla Proteobacteria, Bacteroidetes and Actinobacteria were dominant in the treatment groups, accounting for 45, 21.9 and 21.9 % of the sequence reads, respectively. However, in sharp contrast, the control fishes were predominantly occupied by Proteobacteria (82.1 %) and Firmicutes (8.4 %). At the genus level, the microbial communities were different between the control and treatment groups, although the two groups shared similar genera. Additionally, some genera (such as Tepidimonas, Variovorax, Roseomonas, Rubritepida, Nitrobacter, etc.) only appeared in the treatment group, and many other genera only existed in the control group. Therefore, we conclude that the addition of the SC02 strain in water improves water quality, which may ultimately be a result of changes in microbial community diversity in grass carp cultures.
Effects of Bacillus Licheniformis on the Growth, Antioxidant Capacity, Intestinal Barrier and Disease Resistance of Grass Carp (ctenopharyngodon Idella).

Abstract:To study the effect of dietary supplementation of Bacillus licheniformis FA6 on the growth, survival and intestinal health of grass carp, we assessed the antioxidant capacity, intestinal barrier, expression levels of immune genes, and the resistance to Aeromonas hydrophila AH-1 infection. Experimental setup comprised three groups (90 specimens each; average initial weight = 16.5 g): the control group was fed the basal diet without B. licheniformis, the low-dose (LD) group was supplemented with B. licheniformis at the concentration of 1 × 105 cfu/g, and the high-dose (HD) group with 1 × 106 cfu/g. After 56 days of growth trial, the challenge test with A. hydrophila AH-1 was conducted for 14 days. The results revealed that the grass carp in LD group and HD group had significantly (p < 0.05) improved percent weight gain (PWG) and specific growth rate (SGR) parameters. Additionally, the antioxidant status was improved, which included increased superoxide dismutase (SOD) activity and decreased malondialdehyde (MDA) levels in the serum, and upregulated mRNA levels of antioxidant enzymes MnSOD and catalase (CAT) in the intestine. Meanwhile, B. licheniformis FA6 supplementation groups exhibited a decreased mRNA expression of proinflammatory cytokines (such as IL-1β and TNF-α) and increased the expression of anti-inflammatory cytokine IL-10. Histological (villi length was increased) and gene expression (qPCR: upregulated ZO-1, occludin, and claudin-c) analyses suggested improved functioning of the intestinal barrier. Post-challenge mortality rates in LD and HD groups were significantly lower (56.6% and 70% respectively) than in the control group (100%). Overall, these results indicated that dietary supplementation of B. licheniformis FA6 can improve growth, antioxidant capacity, intestinal barrier functions and disease resistance of grass carp.
Selenium-enriched Bacillus subtilis reduces the effects of mercury-induced on inflammation and intestinal microbes in carp (Cyprinus carpio var. specularis)

Abstract:Mercury (Hg) is a global pollutant that affects the health of humans and ecosystems. Selenium (Se) is an essential trace element for many organisms including humans. Bacillus subtilis is one of the main probiotics used in aquaculture, and has a certain adsorption effect on heavy metals. The interaction between Hg and Se was rigorously studied, especially due to the observation of the protective effect of Se on Hg toxicity. The objective of this study was to research the effects of Hg, Se, and B. subtilis on inflammation and intestinal microbes in common carp. The common carp was exposed to Hg (0.03 mg/L), and 105 cfu/g Se-rich B. subtilis was added to the feed. After 30 days of feeding, samples were taken to evaluate the growth performance, serological response, inflammatory response, and intestinal microbial changes. In this study, when fish were exposed to Hg, the growth performance of the Se-rich B. subtilis plus 0.03 mg/L Hg fish group was lower than that of the control group and higher than 0.03 mg/L Hg; The levels of serum immunoglobulin M (IgM) and lysozyme (LZM) decreased, but after supplementation with Se-rich B. subtilis, the levels of LZM and IgM increased; Hg treatment significantly upregulated the mRNA expression of interleukin-1β (IL-1β), interleukin-8 (IL-8), tumor necrosis factor-α (TNF-α), and nuclear factor-kB (NF-κB P65), but downregulated the mRNA expression of interleukin-10 (IL-10), transforming growth factor-β (TGF-β) and NF-kappa-B inhibitor alpha (IkBα). However, compared with the Hg group, the Se-rich B. subtilis plus Hg group can significantly increase the mRNA expression levels of IL-1β, IL-8, TNF-α, and NF-κB P65, but reduce the regulation of IL-10, TGF-β, and IkBα expression. Through the analysis of the microbiological, we found that the Hg group was mainly composed of Aeromonas sobria and Aeromonas hydrophila. However, in the Se-rich B. subtilis treatment group, we found that Aeromonas sobria was significantly less than the Hg group. Se-rich B. subtilis improves Hg-induced intestinal microbial changes, alleviates the abundance of Aeromonas, and alleviates the inflammation of the fish. The results of this study show that Se-rich B. subtilis dietary supplements can effectively protect common carp against Hg toxicity.

Abstract:This experiment was conducted to study the effects of Bacillus preparations on immunity and antioxidant activities in grass carp. A total of 315 grass carp, with similar initial weight (average weight of fish 45 g), were randomly divided into three groups with three replicates. The control group was fed the basal diet without Bacillus; treatment group 1 was added Bacillus preparation no. 1 with 1 × 10(8) cfu/m(3) per 7 days in culture water and also fed the basal diet; treatment group 2 was fed the basal diet mixed with 0.5 % Bacillus preparation no. 2, and the culture water was added 1 × 10(8) cfu/m(3) Bacillus preparation no. 1 per 7 days. After 4 weeks of culture, 12 grass carp from each replicate were taken randomly for the determination of immune response and oxidization resistance indices. The results showed that compared with control, the level of globulin and IgM of treatment group 2 was significantly increased (P < 0.05), which was also significantly higher (P < 0.05) than that of group 1. For the non-specific immunity index, compared with control, the lysozyme activity and complement (C3) content of treatment group 1 significantly increased (P < 0.05), and the level of myeloperoxidase and C3 of treatment group 2 was significantly higher (P < 0.05) than that of control, which was also significantly higher (P < 0.05) than that of group 1. In the serum, compared with the control, the level of total antioxidant activity (T-AOC), antisuperoxide anion free radical (ASAFR) and glutathione (GSH) of two treatment groups was significantly increased (P < 0.05); GSH content of treatment group 2 was significantly (P < 0.05) higher than that of treatment group 1 and control. There was no significant difference between T-AOC and ASAFR among the two treatment groups; no significant effect was found on glutathione peroxidase (GSH-Px) and maleic dialdehyde (MDA) among the three groups. In the liver, T-AOC, SOD, ASAFR, GSH-Px and GSH from two treatment groups were significantly higher (P < 0.05) than those of control; T-AOC, SOD and catalase from treatment group 2 were significantly higher (P < 0.05) than those of treatment group 1; and MDA of two treatment groups was significantly decreased (P < 0.05) as compared with the control. The results indicate that Bacillus preparations added into water can increase serum immunoglobulin levels and most of non-specific immune parameters content and enhance the antioxidant ability of grass carp, while adding Bacillus preparation into the water and feed is much better.
Gut Microbiota Modulation and Immunity Response Induced by Citrobacter Freundii Strain GC01 in Grass Carp (ctenopharyngodon Idellus)

Abstract:The effect of Citrobacter freundii, an opportunistic pathogen of fish, on the gut microbiota and immune responses in fish remains unknown. We fed grass carp (Ctenopharyngodon idellus) with two different concentrations of this bacterium (experimental Low and High groups, plus Control group). No signs of acute inflammatory response were found in either of the experimental groups using the intestinal HE staining method, but the number of intestinal goblet cells was much higher in the High group. Furthermore, we found that feeding C. freundii significantly decreased the alpha diversity and altered beta diversity of gut microbiota in both groups, and significantly promoted the relative abundance of Enterobacteriales, Pasteurellales, Neisseriales and Citrobacter in the High group. Spleen transcriptome showed that the expression pattern of immune system-related genes varied significantly between the Control group and High group. Several immunity-related pathways and two infectious diseases-related pathways were significantly enriched in the High group. Many complement system genes, including c3, c4, c5, c7, c8a, c8b and c1s, were also up-regulated in the High group. Overall, our results indicate that C. freundii did not trigger acute intestinal inflammation, but it changed the diversity and population structure of gut microbiota, and triggered the host's innate mucosal immune system. The enrichment of two infectious diseases-related pathways also suggested a higher risk of infection in the High group. These results contribute to our understanding of the effects of opportunistic fish pathogens on gut microbiota and host's immune responses.

Abstract:A 45d culture experiment was conducted to study the effect of Bacillus on growth, antioxidant activity of intestinal mucosa of grass carp （ Ctenopharyngodon idellus） and water quality. 450 grass carps with initial average weight of （51.0 ± 2.3） g were randomly divided into 3 groups （control group, treatment group 1 and 2 ; three rep- licates for each group and 50 fish per replicate）. Control group was fed with the basal diet, Group 1 were fed the basal diet supplemented with Bacillus complex （ proportion of mixture of B. subtilis and B. amyloliquefaciens ： 1 ： 1 ; 105 cfu/g） , and Group 2 were fed the basal diet supplemented with B. subtilis only （105efu/g）. The results indi- cated that compared with the control group, water nitrite nitrogen level in Group 1 and Group 2 significantly de- creased from the 21^st day to the end of the experiment （except on the 35th day for group 1, P 〈0.01 ） ; and ammo- nium nitrogen level in group 2 also decreased significantly on the 21^st and 35^th day. No significant differences were found for water pH and nitrate nitrogen level among the three groups. Supplementation of Bacillus also decreased the death rate （ P 〈 0.05 ） and improved the growth performance of grass carps. Compared with the control group, WGR of Group 1 and Group 2 increased by 52.93% （P 〈0.01 ） and 21.78% （P 〈0.05）, and ADFI increased by 44.44% （P 〈0.01 ） and 16.67% （P 〈0.05）, respectively; and compared with Group 2, supplementation of complex Bacillus increased both WGR and ADFI of grass carp significantly （25.58% and 23.81% , respectively, P 〈 0.01 ）. Further study showed that supplementation of Bacillus improved partial antioxidant capacity of intestinal mucosa of grass carp, as was evidenced by increased activities of superoxide dismutase （SOD） and catalase （CAT）. it can be concluded that, Bacillus supplementation in basal diets can improve water quality and antioxi- dant capacity of intestinal mucosa of grass carp, and further improve the growth performance and decrease the death rate of grass carp.
Novel insights into the intestinal immune regulatory effects of (2-Carboxyethyl) dimethylsulfonium Bromide (Br-DMPT) in on-growing grass carp (Ctenopharyngodon idella)

Abstract:The present study evaluated the effects of (2-Carboxyethyl)dimethylsulfonium Bromide (Br-DMPT) supplementation on the intestinal immune function and potential mechanisms of on-growing grass carp (Ctenopharyngodon idella) by feeding fish (initial weight 216.49 ± 0.29 g) five diets with gradational Br-DMPT (0-520 mg/kg diet) concentrations for 60 days and then infecting them with Aeromonas hydrophila for 14 days. Our results firstly indicated that compared with the control group, appropriate Br-DMPT supplementation increased the number of beneficial bacteria Lactobacillus and Bifidobacterium and enteritis resistance, decreased the number of detrimental bacteria Aeromonas and E. coli, and relieved the intestinal histopathological symptoms of fish. In addition, compared with the control group, appropriate Br-DMPT supplementation (1) increased lysozyme (LZ) and acid phosphatase (ACP) activities, as well as complement 3 (C3), C4 and immunoglobulin M (IgM) content; (2) upregulated the mRNA levels of anti-microbial substance: liver expressed anti-microbial peptide (LEAP) -2A, LEAP-2B, hepcidin, β-defensin-1 and Mucin2; (3) partially downregulated the mRNA levels of pro-inflammatory cytokines [interleukin 1β (IL-1β), IL-6, IL-8, IL-12p40, IL-15, IL-17D, tumour necrosis factor α (TNF-α) and interferon γ2 (IFN-γ2)] by inhibiting [IKKβ/IκBα/(NF-κBp65 and c-Rel)] signalling; and (4) partially upregulated the mRNA levels of anti-inflammatory cytokines [IL-4/13A, IL-10, IL-11, transforming growth factor (TGF)-β1] by activating [TOR/(S6K1 and 4E-BP)] signalling. The aforementioned results indicated that appropriate amount of Br-DMPT exerted a positive effect on the regulation of intestinal immune function in fish. Finally, based on enteritis morbidity, the IgM content and the lysozyme activity in the PI, the appropriate levels of Br-DMPT supplementation for on-growing grass carp were established as 295.43, 301.73 and 320.36 mg/kg diet, respectively.

Abstract:This study aimed to identify potential probiotic strains of Bacillus subtilis from healthy fish gut microbiota for application in aquaculture. The effects of dietary B. subtilis administration on growth performance, serum enzyme activity, immune gene expression, and disease resistance in darkbarbel catfish (Pelteobagrus fulvidraco) were investigated. The isolate, identified through gene sequencing and biochemical tests, demonstrated resilience to pH 3.0% and 6.0% bile, and exhibited extracellular protease, cellulose, lipase, and amylase production. Darkbarbel catfish were fed diets with varying B. subtilis concentrations (0 CFU/kg [T0], 107 CFU/kg [T1], 108 CFU/kg [T2], and 109 CFU/kg [T3]). After 8 weeks, significant increases (p < 0.05) were observed in final body weight, weight gain rate, specific growth rate, serum lysozyme, serum superoxide dismutase, alkaline phosphatase, and total antioxidant capacity, whereas malondialdehyde levels significantly decreased. Feeding darkbarbel catfish with B. subtilis diets increased immunoglobulin M (IgM) and C3 gene expression (p < 0.05), indicating a positive impact on the fish's immune system. The strain upregulated interleukin 10 (IL-10) and transforming growth factor-β (TGF-β) expression and downregulated TNF-α and IL-1β, suggesting potential anti-inflammatory effects. Following a 7-day challenge with Aeromonas hydrophila, fish fed with B. subtilis exhibited lower mortality, with higher survival rates in the T2 and T3 groups. In conclusion, supplementing darkbarbel catfish diets with B. subtilis effectively enhances growth performance, immune response, and disease resistance.

Abstract:The immunomodulatory effect of Astragalus polysaccharides (APS) has been revealed in aquatic animals. However, the fundamental mechanisms in grass carp (Ctenopharyngodon idellus) are not well characterized. This research has explored the impacts of APS on immune response, gut microbes and transcriptome profiles of grass carp. The eight week feeding trial was executed with the basal diet (Control) complemented with APS at diverse concentrations (0 and 1 g/kg). Firstly, the enzyme activity assay results indicated that dietary APS could considerably increase the activities of antioxidant and digestive enzymes. Furthermore, gut microbial analysis data showed that the intestinal microbial diversity and composition of grass carp were altered at the phylum level between the Control and APS groups. Additionally, beta-diversity revealed that APS had influenced the bacterial assembly in the foregut and hindgut of grass carp. The transcriptomic profiling was used to further explicate the mechanism fundamental to the effects of APS on molecular evidences in the intestines; the results revealed that the APS could activate immune related genes. Further study results revealed that the immune response stimulated by APS, the mRNA expression levels of TNF-α, IL-1β, CSF, ILF2, DCLK1 and TAGAP genes were all significantly up-regulated after dietary APS supplementation. Finally, histopathological changes of different tissues and bacterial load levels in the liver between the A. hydrophila and A. hydrophila + APS groups illustrated that APS protected the tissues under A. hydrophila challenge. Overall, the present study revealed that APS could modulate the intestinal microbiota and activate the immune response to protect the grass carp from bacterial infection.

Abstract:The goal of this study was to research the effects of Lactobacillus buchneri L3‐9 and its extracellular products on growth, immunity, intestinal microorganisms, and disease resistance to Aeromonas hydrophila in grass carp. A total of 120 fish (80–90 g) were distributed into 12 aquariums (10 fish/aquarium) in three replicates and fed 1% of their body weight for 21 days. The grass carp were fed four different diets: a control (group C), a diet supplemented with 1 × 106 cell g−1 of L. buchneri L3‐9 (group B), a diet supplemented with extracellular products of L. buchneri L3‐9 (group E), and a diet supplemented with L. buchneri L3‐9 and the extracellular products (group BE). Compared with the control diet, the B, E, and BE diets increased the weight gain rate by 4.49%, 2.59%, and 4.38%, respectively (p < 0.05), and significantly decreased the feed conversion ratio by 0.72, 0.57, and 0.76, respectively (p < 0.05). Groups B, BE, and E showed significant decreases in mortality, with values of 26.67%, 16.67%, and 23.33%, respectively (p < 0.05). Group B showed a significant increase in superoxide dismutase (SOD) activity and total antioxidant capacity (T‐AOC) and decrease in malondialdehyde (MDA) content compared to the control group (p < 0.05). The highest catalase (CAT) activity was found in group E (p < 0.05). In group B, the expression of the gut proinflammatory gene TNF-α was downregulated (p < 0.05), and the expression of anti‐inflammatory genes, including TGF-β1, IL-10, and Tlr-8, was upregulated compared to that in the control group (p < 0.05). Groups BE and E showed an increased abundance of Lactobacillaceae. Additionally, a drastic decrease in the abundance of pathogenic bacteria such as Aeromonadaceae and Enterobacteriaceae was in these groups compared to the control group. In conclusion, L. buchneri L3‐9 and its extracellular products could improve the growth performance, immune responses, and resistance to A. hydrophila in grass carp. This study provides insights for the development and application of microecological preparations.

Abstract:This experiment was designed to study the effects of Bacillus licheniformis BSK-4 on nitrogen removal and microbial community structure in a grass carp (Ctenopharyngodon idellus) culture. The selected strain Bacillus licheniformis BSK-4 significantly decreased nitrite, nitrate and total nitrogen levels in water over an extended, whereas increased ammonia level. Pyrosequencing showed that Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes were dominant in grass carp culture water. Compared with the control group, the number of Proteobacteria and Firmicutes were increased, while Actinobacteria and Bacteroidetes decreased in treatment group. At the genus level, some genera, such as Bacillus, Prosthecobacter, Enterococcus, etc., appear only in the treatment, while many other genera exist only in the control group; Lactobacillus, Luteolibacter, Phenylobacterium , etc. were increased in treatment group compared to those in control group. As above, the results suggested that supplementation with B. licheniformis BSK-4 could remove some nitrogen and cause alterations of the microbial composition in grass carp water.
Dection of Direct Versus Indirect Effects: Were Experiments Long Enough?

Abstract:To evaluate the hypothesis that indirect effects generally take much longer to become evident in manipulative studies of community regulation than do direct effects and thus may often be missed, I studied the effect of experiment duration in a survey of marine intertidal interaction webs. Contrary to expectation, indirect effects appeared either simultaneously with direct effects or shortly after direct effects were evident. While experiment durations varied greatly, on average most direct and indirect effects became statistically significant within the first 20%–40% of the total experiment duration. Further, the duration of most experiments appeared sufficient so that most indirect effects that would be generated by the manipulation could be observed. On average, a period of “constancy” (i.e., of no further change) lasting roughly 20%–60% of the total experiment duration occurred after the last indirect effect was observed. Experiment duration did not vary with web species richness, which suggests no tendency to perform manipulations for more (or less) time in more complex webs. The number of indirect effects per species did not increase with increasing experiment duration, nor did the number of longer interaction chains (four species vs. three species), which suggests no trends for increased complexity of indirect effects with longer experiments. Ecological theory states that, in interaction webs whose dynamics are imperfectly known, indirect effects may compromise the predictability of species manipulations. However, empirical results suggest that, despite incomplete knowledge of indirect effects, community dynamics may be more predictable than expected.

Abstract:Cottonseed meal and rapeseed meal exhibit a potential for fishmeal substitute in grass carp feed, while their excessive use contribute to growth decline and weakening immunity of aquatic animals. Clostridium butyricum metabolites (CBM) was recognized as a functional additive due to its antioxidant properties and maintenance of intestinal microbiota balance. CBM was added to a high of cottonseed and rapeseed meal diet to determine its effects on growth, immunity, and intestinal microbiota alterations of grass carp (Ctenopharyngodon idella) over 56 days. Eight hundred grass carp (mean weight, around 50 g) were randomized to five treatments and fed with the basic diet (CON), CBM0 diet (28 % cottonseed and 27 % rapeseed meal), and CBM diets (CBM0.5, CBM1, and CBM2, namely CBM0 diet supplemented with 500, 1000, and 2000 mg kg-1 CBM). The results indicated that compared to CBM0, The ingestion of 1000 mg kg-1 CBM diet by grass carp significantly promoted growth as measured by intestinal lipase activity, villus height, and muscle thickness. Moreover, accompanied by a decrease in intestine MDA content, and enhance antioxidant capacity by activating Keap1/Nrf2 signaling pathway to increase enzyme activities (SOD, CAT and T-AOC) and corresponding gene expression (mnsod, cat, gsto and gpx1) in the intestine of grass crap fed CBM1 diet. The dietary CBM1 diet increased serum levels of C3 and IgM, increased ACP activity and expression of the corresponding anti-inflammatory factors (tgf-β1 and il-15), and suppressed the expression of pro-inflammatory factors (tnf-α and il-12β), resulting in enhanced immunity. The dietary CBM1 diet up-regulates gene expression of tight junction proteins (zo-1, occludin, occludin7a and occludin-c), coupled with the decreases in DAO and D-lactate contents, implying that the decreased mucosal permeability could be observed in the gut. The dietary CBM1 diet largely altered the intestinal microbial community, especially reducing the relative abundance of intestinal pathogenic bacteria (Streptococcus and Actinomyces). And it significantly increased the content of short-chain fatty acids (acetic acid, butyric acid, isobutyric acid, propionic acid and isovaleric acid). Taken above, dietary CBM supplementation improved growth in grass carp and attenuated the intestinal oxidative stress, inflammation and microflora dysbacteriosis caused by high proportions of cottonseed and rapeseed meal diets.
