Abstract:Radiofrequency (RF) hyperthermia has been widely used for tumor ablation since magnetic-fluid-hyperthermia (MFH) can be utilized for increasing temperature in tumor-region as a complementary-method for hyperthermia. In this study, the effectiveness of using the magnetite-nanoparticles (Fe 3 O 4 ) in RF hyperthermia for breast cancer (BC) treatment by determining 3D-temperature-distribution using bioheat-transfer-mapping was evaluated. A breast-phantom with a tumor region was placed in an RF-device with 13.56 MHz frequency in different states (with and without-nanomagnetite). Parallelly, the calculations of the RF-wave and bioheat-equation were accomplished by numerical-simulation and finite-element-method (FEM) in COMSOL-software. The temperature differences were experimentally measured at different points of the phantom with a precision of 0.1 °C, with temperature of 3.6 °C and 6.1 °C in without and with nanomagnetic conditions in tumor area, respectively, and also for normal area with temperature of 1.8 °C and 1.9 °C in non-presence and presence states of 0.05 gr magnetite for both conditions, respectively. Moreover, the difference between the simulation and the experimental results was 0.54–1.1 %. The conformity between temperature measurement in experimental and simulation studies in tumor and normal areas showed the effectiveness of the application of MNPs for RF hyperthermia in tissue equivalent breast phantom. Finally, the positive effect of 0.05 gr of MNPs on BC treatment was confirmed.

Abstract:We report the experimental characterization and finite element modeling of magnetic fluid hyperthermia (MFH) in tissue-like media using tetramethyl ammonium hydroxide coated superparamagnetic iron oxide magnetic nanoparticles (MNPs) of size ∼19.6 ± 1.2 nm, prepared using a co-precipitation technique. MFH properties are probed for the MNPs in ∼1 wt. % agar, resembling the tumor and surrounding normal tissues. The field-induced temperature rise (ΔT) is experimentally measured in real-time utilizing an infrared camera. A finite element model (FEM) is utilized to simulate the spatiotemporal variations in the thermal profiles, which are found to be in good agreement with the experimental data. FEM-based parametric studies reveal that the thermal conductivity of the medium is the most significant parameter influencing the thermal profiles. The spatiotemporal variations in the thermal profiles are numerically studied for seven different tissues, and the obtained results indicate the highest ΔT for the breast tissue in the tumor and the surrounding regions, which is due to the lowest volumetric specific heat and the highest thermal conductivity of the breast tissue, respectively. Numerical studies on the thermal profiles for sub-surface tumors with parametrically varying depths indicate a strong exponential correlation between the surface and tumor temperature, where the regression coefficients are found to be correlated with the thermo-physical properties of the tissues. The obtained findings are beneficial for developing a simplistic and easily deployable framework for a priori generation of the thermal profiles for various tissues during MFH, which is useful for appropriate planning and parameter selection for MFH-based therapy.

Abstract:The magnetic nanoparticles (MNPs) in ac alternating magnetic fields will produce a sufficient amount of heats owing to the Néel and Brown relaxations. Magnetic fluid hyperthermia (MFH), based on this mechanism, is a new promising approach for tumor treatments. The temperature field distribution in the cancer and its neighbor regions have a significant effect on the therapeutic effect of MFH. As a result, it is generally required to maintain the temperature in the cancer in the range of 42 °C–90 °C while guaranteeing a sharp temperature gradient in the neighbor regions of the cancer and healthy tissues. This paper provides an automated shape and MNP volume fraction solid design optimization methodology with finite element analysis, coupled field analysis, and an enhanced multiobjective quantum particle swarm optimization, to realize the aforementioned two ultimate goals. Moreover, an optimal radial basis function approximation technique is proposed to approximate the distribution of the optimized nanoparticles volume fraction solids to give a smooth and implementable nanoparticles distribution.

Abstract:In recent years, nanotechnology has made an invasion in biomedical applications which has gained much more attention because of its high efficiency and low side effects. One such application is magnetic hyperthermia (MHT) where magnetic nanoparticles (MNPs) are used to generate heat to kill the cancerous tissues over an alternating magnetic field. The heat generated by the nanoparticles is caused by hysteresis and Neel and Brownian relaxation. In this paper, we study the numerical modelling to obtain temperature distribution on different tumor tissues (liver, lungs, and kidney) embedded with nanoparticles during magnetic hyperthermia by varying different magnetic field strengths (4000–6000 A/m), different frequencies (100–500 kHz) and different volume fractions of nanoparticles (0.1––0.5 %). Results show that at 500 kHz frequency, the temperature at the centre of the tumor is elevated marginally by 8 %, 4 %, and 2 % for the lungs, liver, and kidney respectively. Similarly, at 6000 A/m magnetic field strength, the temperature at the given point of tumor enhances moderately by 25 %, 14 %, and 8 % in the case of lungs, liver, and kidney respectively. The temperature at the same location increases majorly by 81 %, 45 %, and 20 % for the case of lungs, liver, and kidney respectively at 0.5 % volume fraction of nanoparticles. In the study, the effect of the volume fraction of nanoparticles on temperature rise is found to be more significant compared to other cases. The heat transfer rate is maximum in the lungs followed by the liver and kidney respectively.

Abstract:A therapeutic breakthrough in cancer treatment has recently been made by using magnetic nanoparticles (MNPs) for heating in hyperthermia therapy. Unapproachable tumors are being effectively destroyed by focused heat produced by MNPs. The major challenges regarding practical application of this therapy include the control and improvement of induction heating ability of MNPs and hyperthermia temperature range between 42–47 °C, for secure treatment at targeted area. Here, in this study, we have reported the synthesis and characterization of core–shell structured silica-coated cobalt ferrite nanoparticles (SiO2 coated CoFe2O4 MNPs), which are potential candidates for use as heat source in magnetic hyperthermia therapy. The SiO2 coated CoFe2O4 MNPs were synthesized using the reverse micelle method, with the SiO2 coating performed simultaneously during MNP synthesis. Various analytical tools were utilized for the characterization. The structural measurements were probed by X-ray diffraction (XRD). Monodisperse MNPs with nearly spherical core–shell structure was revealed by Transmission electron microscopy (TEM) results. The average diameter of MNPs obtained from TEM analysis was 15 nm. The surface coating of cobalt ferrite MNPs with silica was verified by Fourier transform infrared spectrometry (FTIR). The saturation magnetization values obtained using vibrating sample magnetometer (VSM) measurements were 45.74 emu/g, revealing the superparamagnetic nature of the nanoparticles. The heating efficiency of different concentrations of synthesized nanoparticles was evaluated in an aqueous solution under an alternating magnetic field of strength 5.5 kA/m at a frequency of 260 kHz. A saturation temperature of 42 °C was attained at an optimum concentration of 1.7 mg/mL, while a magnetic field strength of 3.9 kA/m achieved 42 °C at a fixed concentration of 3.5 mg/ml. The obtained specific absorption rate (SAR) values for all the samples were between 56.23 to 100.1 W/g. The correlation between SAR values, MNP concentration, and applied magnetic field strength was also examined. The high saturation magnetization, efficient heating rates, and high SAR values make our synthesized silica-coated cobalt ferrite nanoparticles promising candidates for magnetic hyperthermia treatments, potentially improving therapeutic outcomes for cancer patients.

Abstract:Apoptosis or programmed cell death is one of the regulation mechanisms of cell homeostasis. Fas is a transmembrane receptor protein which transmits a cell death signal when cross linked with an antibody or with its physiological ligand—Fas ligand (Fas L). Fas and Fas L have a pivotal role in regulating lymphocyte apoptosis and maintaining lymphocyte homeostasis. Soluble forms of Fas and Fas L may be detectable and measured in the serum, and may reflect the activation of this pathway. Moreover, soluble forms of Fas regulate Fas/ Fas L mediated apoptosis. Raised levels of soluble Fas (sFas) have been shown in various chronic inflammatory rheumatic diseases, systemic lupus erythematosus, Sjögren’s syndrome, 4 5 and in the synovial fluid of rheumatoid arthritis. These diseases are autoimmune diseases with lymphocyte involvement. Ankylosing spondylitis (AS) is another chronic inflammatory rheumatic disorder, with less autoimmune background or lymphocyte involvement. Involvement of apoptosis in the pathogenesis of AS has not been discounted. This preliminary study aimed at evaluating the apoptotic Fas/Fas L system in AS by measuring the amount of the soluble forms of these proteins in the serum of patients with AS compared with controls. Forty nine consecutive inpatients and outpatients with AS according to the revised New York criteria were included. Forty healthy subjects without any inflammatory or autoimmune disease, with the same age and sex distribution, were used as controls. For the patients with AS the disease activity was assessed by clinical variables (Bath Ankylosing Spondylitis Disease Activity Index (BASDAI)) and biological variables (erythrocyte sedimentation rate (ESR), serum C reactive protein (CRP) levels). Soluble Fas (sFas) and soluble Fas Ligand (sFas L) levels were measured twice in the serum of the patients and controls using a sandwich enzyme linked immunosorbent assay (ELISA) (sFas ELISA kit and sFas L ELISA kit; Medical and Biological Laboratoires Co, Ltd, Nagoya Japan). Statistical analysis was by Student’s t test. Of the 49 patients (34 men, 15 women), with a mean (SD) disease duration of 8.6 (7.2) years, 40 (82%) were HLA-B27 positive. The mean BASDAI index was 5.34 (1.97), mean ESR 24.9 (22.7) mm/1st h, and mean CRP 16.0 (18.8) mg/l. There were no diVerences in the serum levels of sFas and sFas L between patients with AS and controls (table 1). Moreover, no diVerences were found in the sFas and sFas L serum levels between patients with active and inactive AS (table 2). For 26 patients the BASDAI was >5.3 and for 23 patients <5.2. Similarly, 19 patients had an ESR >25 mm/1st h and in 30 patients the ESR was <25 mm/1st h. CRP was >16 mg/l in 17 patients and < 16 mg/l in 32. The split between the groups was defined by the mean of the values. This study failed to show any modification in serum levels of sFas and sFas L in patients with AS compared with controls. There were no diVerences between patients with clinically or biologically active and inactive AS, and so there was no correlation with disease activity. These results are in contrast with the results found with other chronic inflammatory autoimmune rheumatic diseases. 4 5 However, Fas levels do not seem to correlate with the clinical diagnosis of autoimmune disease, or laboratory abnormalities. AS differs from these conditions as there is no lymphocyte activation, but involvement of polymorphonuclear cells and intracellular chronic infection. These mechanisms do not seem to interfere with apoptosis. For example, it has been shown that clearance of Chlamydia trachomatis (a micro-organism implicated in some spondyloarthropathies) from the genital mucosa does not require Fas mediated apoptosis. Our results suggest that there is no involvement of apoptosis via a Fas/Fas L pathway in AS.

Abstract:A Newtonian nanofluid containing suspended nanoparticles can substantially improve heat transfer due to enhanced energy transport mechanisms. This theoretical study investigates heat and mass transfer in biological tissues using such a nanofluid under a magnetic field. These properties have promising medical and engineering applications. The nonlinear governing equations were transformed into ordinary differential equations using similarity variables and numerically solved with MATLAB boundary value problem solver bvp4c, subject to appropriate boundary conditions. Results demonstrated increasing the heat source parameter dramatically raised tumor interstitial temperature. This heating, along with improved nanoparticle accumulation within the tumor due to the thermal effects, are together essential for effective hyperthermia treatment. The model provides new insights into tuning heat and mass transport mechanisms in biological tissues via nanofluids for therapeutic applications. Therefore, the findings of this study may improve the efficacy of thermal therapy in treating cancer.

Abstract:Diffusion of magnetic fluid and energy transport in the tumor are crucial processes for magnetic fluid hyperthermia and play an important role in tumor treatment.A coupled numerical simulation,including a fluid transport model,a solute transport model and a heat transfer model,was developed to investigate the diffusion process of magnetic fluid and the temperature rise in the tumor tissue in alternating magnetic field.The finite element method was applied to the calculations,and the pressure distribution,concentration distribution,and temperature distribution were obtained.Moreover,the effects of the diffusion time of the magnetic fluid,the number of injection points and the specific absorption rate(SAR)on temperature distributions were analyzed.The results showed that the increases of the diffusion time and the number of injection points caused the volume of tumor tissues with the desired necrosis temperature to increase,and decrease the highest heating temperature of the tumor tissue as well.An increase in the SAR always resulted in an obvious increase in the volume of tumor tissues with the desired necrosis temperature.However,the temperature of the normal tissue also increased with the increase of SAR,which might cause dangers in clinical treatments when it was higher than the safety temperature.

Abstract:Magnetic induction hyperthermia (MIH), based on the Neer relaxation heat generation mechanism of the magnetic medium in an alternating magnetic field, is an adjuvant treatment technology for tumor lesions. A precise temperature control of the hyperthermia region is essential in order to ensure a relatively desired therapeutic. In this paper, an automatic shape optimization procedure is implemented to acquire tumor lesion optimal temperature distribution produced by a uniform magnetic field generating device using magnetic-thermal coupled finite element analysis and multi-objective optimization algorithms. The effect of multi-injection and blood perfusion is considered. The simulation result shows that an appropriate device shape and injection point location can produce a relatively ideal temperature field distribution in the tumor hyperthermia region.

Abstract:Advancement of the recent micro/nanotechnology stimulates the renaissance of using magnetic micro/nanoparticles embedded in tissues for the target tumor hyperthermia. However, there is a strong lack of quantitative understanding of the temperature profiles thus induced by the applied external electromagnetic (EM) field, which may impede the successful operation of this therapy. In the current study, the three-dimensional quasi-steady-state EM field and transient tissue temperature behavior induced by two planar electrodes were numerically investigated. Detailed computations indicated that nanoparticles exhibit an extraordinary highly focused heating on target tumor tissue, which is much stronger than that in the surrounding areas. This heating effect depends heavily on the properties of the magnetic nanoparticles, which may vary appreciably for different samples depending on their particle sizes and microstructures. The effect of micro/nanoparticle concentration, heating area, and the frequency and strength of the external alternating EM field were also tested. Moreover, a criterion to determine the appropriate particle concentration thermally important for medical treatment was established. Given accurate thermal and EM parameters for cancerous tissue embedded with nanoparticles, the current model could possibly be applied in the hyperthermia treatment planning and help optimize the surgical procedures.

Abstract:Magnetic fluid hyperthermia (MFH) promises to be a viable alternative in the treatment of localized cancerous tumors. The treatment consists of introducing nanoparticles as energy absorbent agents in tumor tissue under an oscillating magnetic field, where nanoparticles dissipate energy in the form of heat, causing a localized rise in the temperature and tumor cell death. Traditional magnetic fluid under study is artificial magnetic nanoparticles. This work seeks to introduce the new natural biologic magnetic material bacterial magnetosomes (BMs) to be used in MFH. Properties of magnetosomes and chemically synthesized magnetic nanoparticles (MNPs), such as morphology, magnetic properties and their heating effects under magnetic field were compared. Cytotoxicity studies using human breast cancer cells MCF-7 indicated that cell viability could be significantly decreased by the heat derived from BMs and MNPs under alternative magnetic field. Biocompatibility of BMs and MNPs was compared in terms of evaluating their acute toxicity in mice and their decomposition abilities in vitro, and it showed that magnetosomes exhibit a lower toxicity. These findings provide evidence for beneficial activities of magnetosomes in MFH and support the continued investigation of it to be applied in biomedicine.
Simulation of magnetic hyperthermia cancer treatment near a blood vessel

Abstract:In this study, we conduct a study on magnetic hyperthermia treatment when a vessel is located near the tumor. The holistic framework is established to solve the process of tumor treatment. The interstitial tissue fluid, MNP distribution, temperature profile, and nanofluids are involved in the simulation. The study evaluates the cancer treatment efficacy by cumulative-equivalent-minutes-at-43 centigrade (CEM43), a widely accepted thermal dose. The influence of the nearby blood vessel is investigated, and parameter studies about the distance to the tumor, the width of the blood vessel, and the vessel direction are also conducted. After that, the effects of the fluid-structure interaction of moving vessel boundaries and blood rheology are discussed. The results demonstrate the cooling effect of a nearby blood vessel, and such effect reduces with the augment of the distance between the tumor and blood vessel. The combination of downward gravity and the cool effect from the lower horizontal vessel leads to the best performance with 97.77% ablation in the tumor and 0.87% injury in healthy tissue at distance d = 4 mm, but the cases of the vertical vessel are relatively poor. The vessel width and blood rheology affect the treatment by velocity gradient near the vessel wall. Additionally, the moving boundary almost has no impact on treatment efficacy. The simulation tool has been well-validated and the outcomes can provide a useful reference to magnetic hyperthermia treatment.

Abstract:The large blood vessels surrounding the tumor would significantly result in heat sink, and thus seriously limit the thermal ablative area during tumor hyperthermia. Magnetic nanoparticle (MNP) was recently identified as an important heating enhancer to improve the treatment efficiency. It will not only help to absorb more energy under the irradiation of external magnetic field, but also can block the blood flow and subsequently weaken the heat sink effect of large vessels. In this study, these two critical factors, reserved to be undisclosed before in theory, were comprehensively investigated through three-dimensional numerical simulation. The results suggested that concerning the contribution to temperature increase in the tissues surrounding large vessel, the factor of blood flow blocking is more effective than that of energy absorption. Therefore, selective loading of MNPs to the target sites is expected to serve as a promising method to perform successful hyperthermia treatment for tumor tissues embedded with large blood vessels.

Abstract:Magnetic fluid hyperthermia (MFH) is emerging as an alternate treatment therapy for cancer of different types, due to its potential low side effects and relatively less painless treatment option. However, the implementation of standalone therapy is still challenging and needs research to optimize the effective killing of 100% cancerous tissues, number of therapy sessions, time duration of the therapy, controlled hyperthermia window temperature, etc. We report here the potential of nanoclusters (nanoflowers) based dispersion and on an in vitro study on breast cancer cells (MDA-MB-231). The nanoclusters were characterized using XRD, TEM, DLS, VSM and MFH, before investigating them with breast cancer cells. The effect of MFH, before and after the treatment of induction heating on MDA-MB-231 cells, were also studied. Results show that the MFH is capable of killing almost 87% of cells within 30 min of treatment on cells in the presence of magnetic nanoclusters. This shows the potential of developing nanoflowers for the treatment of cancer over other single domain magnetic nanoparticles.

Abstract:Magnetic Particle Imaging (MPI)-guided Magnetic Fluid Hyperthermia (MFH) has the potential for widespread utilization, as it allows for the prediction of magnetothermal dosage, real-time visualization of the thermal therapy process, and precise localization of the lesion area. However, the existing MPI-guided MFH (MPI-MFH) method is insensitive to concentration gradients of magnetic nanoparticles (MNPs) and is susceptible to causing damage to normal tissues with high MNP concentrations during MFH treatment, while inadequately heating tumor tissues with lower MNP concentrations. In this work, we established a relationship between MNP concentration and heating efficiency through simulations and phantom measurements, enabling the optimal selection of MFH parameters guided by MPI. Based on these findings, we developed a high-gradient field MPI-MFH method using a fieldfree point (FFP) approach to achieve precise local heating. Phantom experiments and in vivo glioma model experiments were conducted to validate this proposed method. The results demonstrated that the proposed method of MPIMFH can improve the MNP concentration gradient sensitivity to ±1 mg/ml, thereby enabling more effective lesion-site heating without damaging normal tissues. This method not only reduced glioma size effectively but also holds promise for application in various other types of cancers.

Abstract:We developed a dedicated computational framework by coupling the lattice-Boltzmann-method (LBM) modeling and the particle-swarm-optimization (PSO) algorithm to search optimal strategies of magnetic nanoparticle (MNP) injection for hyperthermia-based cancer treatment. Two simplified tumor models were considered: a circular model representing geometrically regular tumors and an elliptic model representing geometrically irregular tumors, both sharing the same area. The temperature distribution in the tumor and its surrounding healthy tissue was predicted by solving the Pennes’ bio-heat transfer equation (PBHTE). Both single- and multi-site injection strategies were explored. The results suggest that the multi-site injection strategies generally work well, while the single-site injection strategy fails even on the simplest circular tumor model. The more the injection sites, the better the performance. In particular, when the number of injection sites reaches eight, all temperature requirements can be nearly 100% satisfied in both tumor models. Whether or not including the minimum dose requirement in the objective function only affects the optimization results by less than 2%. The thermal dose was also assessed by considering both temperature and heat exposure time. It was found that the optimal multi-site injection strategies perform reasonably well for both tumor models. Although the setting is only two dimensional and the optimization is on very simplified tumor models, the framework adopted in this present study works well and can provide useful insights into magnetic hyperthermia treatment.

Abstract:Hyperthermia treatment incorporating magnetic nanoparticles (MNPs) is a hopeful therapy for cancers. Acquiring information about the MNPs' deposition in tumor tissues and modeling magnetic heating in vivo are essential for successful treatment. In this paper, we discuss the inhomogeneous heat generation by MNPs distributed heterogeneously in a liver tumor during arterial embolization hyperthermia (AEH) treatments. In order to more accurately simulate the temperature elevation for an AEH treatment plan, we conducted the following experiments. First, we detected the distribution of magnetic field intensity in the aperture of a ferrite-core applicator. We found that attenuation of the magnetic field focuses mainly on the vertical distance of the aperture, which makes MNPs in tissues have different power loss along the lognitudinal axis. Second, we prepared 20 nm monodisperse lipiodol-soluble MNPs and injected super-selectively through the micro-catheter into the arteries of a rabbit with a VX-2 liver tumor. By histological cuts of the investigated specimen, as well as computed tomography (CT), we found MNPs mainly concentrated on the tumor periphery. Last, from the experimental information, we established a new model for simulating the increasing temperature in the liver tumor based on our inhomogeneous interior-heat-source analysis (IIA). We also compared the simulated results with the two types of homogeneous models. The results showed that IIA gives significantly different results from those for a homogeneous model and thus is preferable when an accurate treatment plan is required during AEH.
