Abstract:BackgroundGlucomannans (GMs) are a group of hydrophilic polysaccharides, which have been produced from different parts of natural plants. Studies on plant-derived GMs contribute to wide applications of GMs or GMs-added products that have desirable quality and health characteristics. Among those popular GMs, konjac glucomannan (KGM) and acemannan are the most famous ones that have been widely reviewed from various points in recent years. However, there is a lack of a comprehensive review to introduce the sources, extraction and purification methods, structural features, and other aspects of GMs from common natural plants such as Aloe, Dendrobium, Bletilla, and so on.Scope and approachThis review firstly presents the extraction and purification technologies of GMs from different plant sources, aiming at optimizing more effective production methods with specific treatments for special plant origins. After that, structural features, biological properties, as well as structure-function relationship are summarized.Key findings and conclusionsGMs have been widely isolated from different sectors (leaf, stem, and roots) of many plant resources. A general extraction and purification procedure together with several suggestions is put forward to prepare GMs. Although the β-(1 → 4)-linked main chain has been confirmed, detailed structural characteristics of GMs such as side chain and functional group related properties need further studies. Molecular weights, monosaccharide composition, and acetyl group of GMs are key factors that have been identified to affect plentiful biological properties such as immunomodulatory, anti-tumor, anti-diabetic activities. However, the inherent action mechanisms remain unclear. Several research interests in the future are recommended.

Abstract:To explore greener modifier of konjac glucomannan, a new type of thermal irreversible gel using konjac glucomannan (KGM) and basic amino acids (L-lysine, and L-arginine) was prepared in the present study. The effects of different concentrations (1%, 2%, 3%, 4%, and 5%) of these two basic amino acids on the structure and properties of KGM hydrogels were mainly studied. The results showed that there was a better texture characteristic of composite gel as the concentration of amino acids was above 4%. Besides, with the increase in amino acid concentrations, the pore size of composite gel was denser and the syneresis of composite gel was more obvious, respectively. The rheological results indicated that the storage modulus (G ') was influenced by the concentration of amino acids and temperature. It was found that the two basic amino acids removed the acetyl group of KGM molecule and changed the original crystalline domain of KGM as evidenced by FT-IR and X-ray diffraction (XRD) analysis. Furthermore, compared to L-lysine, L-arginine could exert a greater effect on the KGM gel. In conclusion, the two basic amino acids (L-lysine, and L-arginine) could remarkedly improve the structure and properties of natural KGM and could be used as modifiers of the KGM hydrosol.
Protective effect of three glucomannans from different plants against DSS induced colitis in female BALB/c mice

Abstract:Glucomannans (GMs) from diverse natural plants have great potentiality in enhancing the host immune system. The protective effects of three GMs on the intestinal mucosal immunity in colitis mice were investigated and compared in this study. The three GMs (KGM, AGP, and DOP) were obtained from Amorphophallus rivieri, Aloe vera, and Dendrobium officinale, respectively, having different weight-averaged molecular weights (Mw), acetyl group content, and molar ratios of mannose to glucose (M/G). The three fractions were administered with or without dextran sodium sulfate (DSS) containing drinking water. Macroscopic observations (health state, crypt depth, and bowel thickness of colon tissue) were conducted. Furthermore, related cytokines and mRNA expressions of TLRs were measured by ELISA and RT-qPCR, respectively. Results showed that the administration of the three GMs improved the health state of colitis mice, such as the recovery of body weight, and the increase of the immune organ index, crypt depth, bowel wall thickness, and total number of immune cells. The integrity of intestinal mucosa was maintained by the increased number of goblet cells and mucin protein production. Further studies showed that GMs kept the balance between pro- and anti-inflammatory cytokines and also regulated the expressions of TLR-2, TLR-4, TLR-6, and TLR-9. The above results suggested that GMs could attenuate the intestinal epithelial injury and regulate the intestinal mucosal immunity. Structural features including the M/G ratio, Mw, and the content of acetyl groups jointly influence the protective effects of GMs on the colitis mice.

Abstract:Konjac glucomannan (KGM) is a water-soluble natural plant polysaccharide from Konjac tubers, a special resource in China. It has good water absorption and thickening properties. Due to the complex structure, large molecular weight, and strong hydrophilicity of the characteristic acetyl group, KGM-based materials have poor moisture barrier properties and poor mechanical properties, further limiting their applications. In the early stage, it was found that the rigidity and flexibility of Konjac glucomannan particle molecular chain may affect the mechanical stability of its grain state. This strange molecular rigidity and flexibility may be due to the dynamic change of molecular chain to form stable KGM particles. It is speculated that the change of molecular rigid and flexible structure can improve the stability of molecular chain of KGM particles and obtain KGM particles with good mechanical properties. This paper will deeply analyze the research process of "KGM particle molecular rigidity and flexible structure → effect of molecular rigidity and flexible structure on KGM particle mechanical stability → molecular dynamics simulation interactive visual analysis → experimental verification and model evaluation → strengthening the mechanical properties of KGM based materials". The mechanical stability mechanism of Konjac glucomannan particle state structure formation and its molecular rigidity and flexible regulation behavior were revealed. The dynamic regulation was carried out by computer simulation to explore the relationship between them. The purpose is to reveal the mechanical stability mechanism of Konjac glucomannan particle state and lay a theoretical foundation for the bottleneck problem faced by Konjac refined powder in the processing process and its molecular stiffness and flexibility regulation.
Molecular Dynamics Simulations of the Interactions between Konjac Glucomannan and Carrageenan

Abstract:The interactions between konjac glucomannan and carrageenan were studied with the method of molecular dynamics simulation. Part representative structure segments of KGM and two unit structures of K-carrageenan (Fig. 2) were used as mode, and the force-field was AMBER2. The stability and sites of konjac glucomannan/carrageenan interactions in water were researched at 373 K with the following results: the potential energy (EPOT) of the mixed gel was dropped, while those of single-konjac glucomannan gel and single carrageenan were increased. The surface area (SA) of KGM in the mixed system was decreased to 1002.2 angstrom(2), and that of carrageenan to 800.9 angstrom(2). The variations of two parameters showed that the stability of compound gel konjac glucomannan/carrageenan was improved, which is consistent with the previous studies. The sites of interactions in the mixed gel were the -OH groups on C(2), C(4) and C(6), the acetyl group in KGM mannose, and the -OH group on C(6) in carrageenan. The hydrogen bond was formed directly or indirectly by the bridge of waters.
The galactomannan-EGCG physical complex: Effect of branching degree and molecular weight on structural and physiological properties

Abstract:Polysaccharides and polyphenols are bioactive components that co-exist in many plant foods. Their binary interaction in terms of the structure-function relationships, however, has not been well clarified. This study elucidated the correlation between the structural and physiological properties of galactomannan (GM) -catechin monomer complexes and GM with different branching or molecular weight (Mw). Results indicated that locus bean gum with lower branching degree (Gal/Man is 0.259) bound more readily to EGCG with adsorption rate of 19.42 %. EGCG and ECG containing galloyl groups were more inclined to form hydrogen bonds with GMs, significantly improving the adsorption by GMs. The introduction of EGCG could enhance the antioxidant activity and starch digestion inhibition of GM, which positively correlated with the adsorption capacity of EGCG. The guar gum (GG) with higher Mw (7384.3 kDa) could transport 71.51 % EGCG into the colon, while the retention rate of EGCG reaching the colon alone was only 46.33 %. Conversely, GM-EGCG complex with lower Mw (6.9 kDa) could be readily utilized by gut microbiota, and increased production of short-chain fatty acids (SCFAs). This study elucidated the structure-properties relationship of GM-EGCG complexes, and provide a new idea for the development and precision nutrition of polysaccharides-polyphenol complexes fortified functional foods.
Effects of soluble glucomannan and insoluble cellulose treatment on mucin secretion and mucin glycosylation-related gene expression in the colons of mice

Abstract:Background: Fiber added to the diet can promote intestinal mucin secretion, relieve intestinal inflammation, and enhance the intestinal barrier function. Glycosylation is the key to mucin function. However, there are few studies on the correlation between dietary fiber and mucin glycosylation, especially two kinds of dietary fiber with different solubility. The aim of this study was to investigate the effects of soluble glucomannan (GM) and insoluble cellulose (CL) treatment on mucin secretion and mucin glycosylation-related gene expression in the colons of mice. Results: The GM group significantly increased the goblet cell number, crypt depth, and the expression of mucin 2 (Muc2) and mucin 3a (Muc3a) genes in the colon. At the same time, the analysis of the colon transcriptome showed that the GM group changed the expression of genes related to the mucin glycosylation process, and the GM group up-regulated the expression of Gcnt3, Gcnt4, St3gal1, Galnt13, and B3gnt6 genes involved in the O-glycosylation process. Similarly, Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that differentially glycosylated genes in the GM group were mainly related to the biosynthesis of mucin type O-glycans, while the genes in the CL group were related to the biosynthesis of various types of N-glycans. The correlation analysis between colonic microbes and differentially glycosylated genes also showed that the abundance of Alistipes in the GM group was significantly associated with the expression of Gcnt3, a key glycosylation gene. Conclusion: Glucomannan treatment was more favorable for colonic Muc2 and Muc3a secretion and mucin O-glycosylation gene expression. © 2023 Society of Chemical Industry.
Effects of galactomannans of varied structural features on the functional characteristics and in vitro digestibility of wheat starch

Abstract:This study explores the effects of four natural galactomannans (GMs) with varying degrees of branching (fenugreek gum, guar gum, tara gum and locust bean gum) on the functional properties and in vitro digestibility of wheat starch (WS). Results from rapid viscosity analysis (RVA) and low-field nuclear magnetic resonance (LF-NMR) analysis revealed that GMs with lower branching degrees were correlated with higher paste viscosity, peak viscosity, and greater water-holding capacity in the WS-GM mixtures. Additionally, these lower branching GMs more effectively inhibited amylose leaching during starch gelatinization, leading to a softer gel texture and increased transparency of the mixtures. X-ray diffraction (XRD) and differential scanning calorimetry (DSC) analysis demonstrated that starch mixtures containing lower branching GMs exhibited reduced relative crystallinity and enthalpy values during aging. Furthermore, the incorporation of lower branching GMs resulted in decreased starch digestibility in vitro, thereby enhanced resistant starch content. These findings highlight the potential of selectively branched GMs to modulate the functional properties and nutritional profile of WS, providing a promising approach for the development of starch-based products with improved health benefits.

Abstract:The hypoglycemic effects of konjac glucomannans (KGMs) are well recognized, and our previous study showed KGMs with different molecular weight have different hypoglycemic effects on diabetes rats, but the detailed mechanisms still remain unclear. In this study, KGMs with medium molecular weight (KGM-M, 757.1 kDa) and low molecular weight (KGM-L, 87.3 kDa) were utilized to investigate the possible mechanism on hypoglycemic effects of type 2 diabetic (T2DM) rats. The results revealed that KGM-M had better effects than KGM-L on decreasing fasting blood glucose, mitigating insulin resistance and improving inflammation. Further mechanism analysis showed that KGM-M better enriched gut flora diversity and the abundance of Ruminococcus and Lachnoclostridium , which was accompanied by increased short chain fatty acids (SCFAs) production and expression of G protein-coupled receptors (GPCRs), and improved regulation on bile acid synthesis. Antibiotics treatment eliminated the beneficial effects of KGMs on gut flora, SCFAs, GPCRs and bile acid synthesis. By contrast, fecal microbiota transplantation (FMT) treatment restored the structure of intestinal microbiota. And after FMT treatment, KGM-M displayed higher hypoglycemic activity than KGM-L, probably due to the better effects on intestinal microbiota, SCFAs production, GPCRs expression and bile acid synthesis inhibition.

Abstract:Deacetylated Konjac Glucomannan (DKGM) and Konjac Oligo-glucomannan (KOGM) as two most widely used derivatives in food industry are structurally and physiologically distinct from Konjac glucomannan (KGM). However, the roles of their distinct structures and physicochemical properties in directing microbiota community and the following outcomes are not fully understood. This paper aims to build links between structural complexity of KGM and its derivatives and microbial metabolism. Results showed that structural alterations changed molecular chain aggregation and water binding ability, thus affected the susceptibility to enzymatic degradation, leading to the distinct microbial composition and outcomes profile. Explicitly, KOGM was distinctive in higher abundances of Catenibacterium and Megasphaera , and lacking Prevotella, which was additionally enriched by KGM and DKGM. KOGM, owned the same butyrate-dominant profile with KGM, was utilized fast. However, KGM possessed the highest fermentability. Severe deacetylation reduced fermentability and led DKGM to a propionate-dominant pattern.

Impact of konjac glucomannan with varied deacetylation degrees on plant-based meat analogue: Study on the gel properties, digestibility, and microstructure

Abstract:Polysaccharides are widely used to improve the quality of plant-based meat analogue (PMA). In this study, four kinds of konjac glucomannan (KG) with different deacetylation degrees (DD) were prepared, namely KG1 (native KG, DD = 0.00 %), KG2 (DD = 41.40 %), KG3 (DD = 80.01 %) and KG4 (DD = 89.07 %), and their effects on the quality of PMA were studied. Results manifested that KG3 improved the hardness (from 3017.16 g to 3307.16 g) and protein digestibility (from 49.65 % to 53.01 %) of PMA without reducing the P21, KG2 and KG4 were less effective than KG3, while KG1 led to a significant decline in the hardness and protein digestibility of PMA. The rheological properties and intermolecular force analysis showed that the partially deacetylated KG was more conducive to improving the G' of the protein system during heating and the proportion of covalent bonds in PMA. These findings suggested that partially deacetylated KG was more promising than native or highly deacetylated KG in PMA. Furthermore, scanning electron microscopy revealed that the morphology of KG gradually changed from fine filaments, to coarse filaments, short filaments and granules as DD increased. This study provides a theoretical basis for the application of partially deacetylated KG in PMA.

Abstract:The effect of structural difference for konjac glucomannan (KGM) and guar galactomannan (GGM) on their physicochemical properties including selective carboxylation, biodegradation and scale inhibition was firstly investigated. Compared with GGM, KGM can be specially modified by amino acid to prepare carboxyl-functionalized polysaccharides. The structure-activity relationship explaining the difference in carboxylation activity and the anti-scaling abilities of polysaccharides and carboxylated derivatives were explored by static anti-scaling, iron oxide dispersion and biodegradation tests coupled with structural and morphological characterizations. KGM with linear structure was preferred for carboxylated modification by glutamic acid (KGMG) and aspartic acid (KGMA) while GGM with branched structure failed to accomplish that due to steric hindrance. GGM and KGM showed limited scale inhibition performance, which was probably attributed to the moderate adsorption and isolation effect of macromolecular stereoscopic structure. While KGMA and KGMG performed as effective and degradable inhibitors for CaCO3 scale with inhibitory efficiencies > 90 %.

Abstract:Konjac glucomannan (KGM) is a dietary fiber supplement that exhibits multiple biological activities, including weight control and regulation of glucose and lipid metabolism. Currently, KGM intake patterns in practical applications include KGM sol, thermal irreversible gel, and frozen thermal irreversible gel. In this study, four intake patterns of KGM, namely KGM sol (KS), deacetylated KGM (DK), KGM gel (KG) and frozen KGM gel (FKG), were used as materials to explore the effects of KGM intake patterns on glucose and lipid metabolism and intestinal flora of obese mice induced by high fat diet under the same dose. Results showed that any type of KGM intake could reduce body weight, fat mass, lipid levels, and insulin resistance in obese mice, as well as partially alleviating liver damage and inflammation caused by obesity. However, KS has the most significant effect on controlling blood glucose and blood lipid in obese mice. Additionally, it was found that KS, DK, KG and FKG can increase the α-diversity of intestinal microflora in high-fat mice, and improve the microflora disorder in high-fat mice. Finally, KS may increase the levels of fasting appetite hormones GLP-1 and PYY in mice, up-regulate the expression of LDLR, GCK and G-6-pase mRNA, and increase the production of short-chain fatty acids (SCFAs) in the intestinal flora of mice, thus regulating glucose and lipid metabolism . This study systematically investigated the effects of different intake forms of KGM on metabolism and intestinal flora in obese mice, which is of great significance for further understanding the role of KGM in the prevention and treatment of obesity metabolic diseases, and for developing targeted dietary interventions.

Abstract:Konjac glucomannan (KGM) is a naturally occurring macromolecular polysaccharide that exhibits remarkable film–forming and gel–forming properties, and a high degree of biocompatibility and biodegradability. The helical structure of KGM is maintained by the acetyl group, which plays a crucial role in preserving its structural integrity. Various degradation methods, including the topological structure, can enhance the stability of KGM and improve its biological activity. Recent research has focused on modifying KGM to enhance its properties, utilizing multi–scale simulation, mechanical experiments, and biosensor research. This review presents a comprehensive overview of the structure and properties of KGM, recent advancements in non–alkali thermally irreversible gel research, and its applications in biomedical materials and related areas of research. Additionally, this review outlines prospects for future KGM research, providing valuable research ideas for follow–up experiments.

Abstract:In this study, the effect of konjac glucomannan (KGM, 0-5%) on the structure, physicochemical properties, and in vitro digestibility of extruded yam starch (EYS) was investigated. The EYS became rougher on the surface and the particle size increased as observed using scanning electron microscopy and particle size analysis. X-ray diffraction and Raman results revealed that the relative crystallinity (18.30% to 22.30%) of EYS increased, and the full width at half maxima at 480 cm-1 decreased with increasing KGM content, indicating the increment of long-range and short-range ordered structure. Differential scanning calorimetry and rheological results demonstrated that KGM enhanced thermal stability and the gel strength of EYS due to enhanced interaction between KGM and YS molecules. Additionally, a decrease in the swelling power and viscosity of EYS was observed with increased KGM content. The inclusion of KGM in the EYS increased the resistant starch content from 11.89% to 43.51%. This study provides a dual-modified method using extrusion and KGM for modified YS with high thermal stability, gel strength, and resistance to digestion.
In vitro fermentation characteristics of oxidized konjac glucomannan and its modulation effects on gut microbiota

Abstract:Depolymerized konjac glucomannan (KGM) had better health benefits than natural KGM. Oxidation and enzyme-hydrolysis were two effective ways to prepare depolymerized KGM. Oxidized KGM (OKGM) and enzyme-hydrolyzed KGM (EKGM) varied in molecular structures, verified by nuclear magnetic resonance and atomic force microscope. The differences in chemical structure definitely influenced the fermentability of depolymerized KGM. Herein, effects of OKGM on in vitro fecal fermentation was investigated, comparing with EKGM at approximately same molecular weights. During fermentation, OKGM groups displayed slightly lower reducing sugar concentrations and higher residual carbohydrate contents than EKGM groups. After fermentation, OKGM groups produced more acetic acids and propionic acids than EKGM groups, while EKGM groups generated more butyric acids than OKGM groups. Meanwhile, relative abundances of the Firmicutes and Lactobacillus in OKGM groups were higher, while relative abundances of the Actinobacteria , Fusobacteria and Bacteroides was higher in EKGM groups. OKGM and EKGM differed in modulating gut microbiota.

Lesionalized Therapy beyond Personalized Therapy in Cancer Management

Abstract:Personalized therapy which specifically focuses on targeting cancer cells is now considered as a standard therapy in cancer management. Though personalized therapy can reduce side effects of normal cell damages by specific targeting, most tumors eventually acquire resistance to target specific drugs by genetic or epigenetic changes. Thus, additional efforts have been made to overcome the limitations of personalized therapy. Tumor heterogeneity induces characteristic changes in their therapeutic responses. Local factors such as blood supply and tissue acidosis differently influence the tumor cells and cells in the tumor microenvironment. Previous therapies have focused on only tumor cells, but tumor cells and their microenvironmental cells should be considered as therapeutic targets (1). Since malignant progression followed by distal dissemination can reflect either genetic or epigenetic cellular heterogeneity or both, cellular heterogeneity between primary and metastatic lesions could be also considered in the design of more efficient cancer therapies (2). Cell-to-cell copy number variation of oncogenes such as MET and PDGFR can confer altered characteristics to subspecialized cell populations. Epigenetic heterogeneity can also contribute to differentiation-related or drug resistance-related changes in cancer cells, in which DNA is hypomethylated to generate genetic instability and disturb gene expression. Cellular heterogeneity can even occur within one tumor lesion. In deeper area of the tumor, most cells exit from the cell cycle, and only cancer cells located on the exterior of the tumor mass undergo cell cycling. Most conventional chemotherapies affect the dividing cells, and non-dividing cells are relatively resistant to chemo- and/or radiotherapy. Because different clinical presentations subsequently affect therapeutic response and prognosis, successful tumor therapy requires deep understanding of tumor heterogeneity. Thyroid carcinoma is a good example to discuss tumor heterogeneity and different clinical presentations in each patient, or even in the different lesions from same patient. Thyroid cancer is the most common cancer in Korean women, and almost cases are differentiated thyroid cancer (DTC). Radioiodines have been widely used for the management of DTC and the molecular mechanism of iodine selectivity is related to the sodium iodide symporter (NIS). Accumulation of radioiodine is dependent on the expression of membrane NIS, which is a molecular target of radioiodine whole body scans and 131I therapy. NIS expression is quite variable among tumors, and is an important factor in 131I therapy response of residual DTC. Response rate of 131I treatment was 80% and 33% in NIS positive and negative tumor, respectively (3). Moreover, different NIS expression was observed in different tumor lesions even in one patient. Only 40% of DTC patients showed similar NIS activity between primary tumors and metastatic lymph nodes (4). Half of patients with multiple lymph node metastases showed different NIS expression between the different lesions. We found that tumor detectability of radioiodine scan is higher in relatively well differentiated tumor than in less differentiated tumor, but tumor detectability of 18F-FDG PET is reversed (5). Radioiodine scan and 18F-FDG PET can visualize the functional level of NIS molecule in DTC tissues (6). This kind of molecular imaging can characterize the heterogeneity of DTC tumor in different lesions, and guide the most proper management. Though in vitro molecular diagnostics can analyze the differences in tumor lesions, most of them require tissue sample and are difficult for direct application in clinic. However, molecular imaging modalities easily visualize the heterogeneous features of different tumor lesions. According to different characteristics among tumor lesions in one patient, more efficient treatment and suitable management should be taken. We would like to name it as "lesionalized therapy." Lesionalized therapy beyond personalized therapy should be applied for more effective and successful cancer management.

Abstract:Galactomannan (GM) has been widely applied in food and other fields due to its appealing physicochemical properties. In this work, considering the changes in structural and physicochemical properties of Sophora japonica f. pendula (SJ-GM) with very high mannose to galactose (M/G) ratio in the late deposition stage, extensive exploration is conducted. The core of structural change is the change of M/G ratio (4.94–5.68), which is caused by the loss of galactose side residues modulated by α- d -galactosidase during seed maturation. Afterwards, the more compact conformation, the higher molecular weight, the increased hydrophobicity, and the greater solution viscosity of SJ-GM can be caused. Notably, the gel strength of SJ-GM with the highest M/G surpasses other GMs, including fenugreek gum (M/G = 1.20), guar gum (M/G = 1.80), Gleditsia microphylla gum (M/G = 2.77), and LBG (M/G = 4.00). Finally, SJ-GM is proven to be an attractive alternative to other GMs.

Abstract:Konjac glucomannan (KGM) is a high-molecular-weight polysaccharide that was originally extracted from the corms (underground storage organs) of Amorphophallus konjac. KGM and its oligomers have been reported as dietary fibers that exhibit an array of health benefits. The depolymerization of KGM via enzymatic hydrolysis at different conditions gives products of low viscosity and can be used for coating materials in microencapsulation. In the present study, konjac glucomannan hydrolysates (KGMHs) were produced by enzymatic hydrolysis using commercial mannanase at pH 4.5 at 70 °C for 5–120 min, then KGMHs' molecular weight (Mw), Degree of Polymerization (DP) and their bioactivities were determined. A longer hydrolysis time resulted in KGMH of a lower DP. Oligoglucomannans (Mw < 10,000) could be obtained after hydrolysis for 20 min. The DP of KGMH rapidly decreased during an early stage of the hydrolysis (first 40 min); DP reached around 7 at the end of the hydrolysis. Antioxidant activities were determined by the DPPH radical scavenging and FRAP assays of KGMHs prepared at pH 4.5 and evaluated at pH 2.0–8.0 depending on pH. KGMH having lower Mw exhibited higher antioxidant activities. KGMHs having the smallest molecular weight (Mw = 419) exhibited the highest DPPH radical scavenging activity. Mw and pH have a greater impact on KGMHs' bioactivities which can be useful information for KGMHs as functional ingredients.
