Abstract:Many insects use memories of their visual environment to adaptively drive spatial behaviours. In ants, visual memories are fundamental for navigation, whereby foragers follow long visually guided routes to foraging sites and return to the location of their nest. Whilst we understand the basic visual pathway to the memory centres (Optic Lobes to Mushroom Bodies) involved in the storage of visual information, it is still largely unknown what type of representation of visual scenes underpins view-based navigation in ants. Several experimental studies have shown ants using “higher-order” visual information – that is features extracted across the whole extent of a visual scene – which raises the question as to where these features are computed. One such experimental study showed that ants can use the proportion of a shape experienced left of their visual centre to learn and recapitulate a route, a feature referred to as “fractional position of mass” (FPM). In this work, we use a simple model constrained by the known neuroanatomy and information processing properties of the Mushroom Bodies to explore whether the use of the FPM could be a resulting factor of the bilateral organisation of the insect brain, all the whilst assuming a “retinotopic” view representation. We demonstrate that such bilaterally organised memory models can implicitly encode the FPM learned during training. We find that balancing the “quality” of the memory match across left and right hemispheres allows a trained model to retrieve the FPM defined direction, even when the model is tested with other shapes, as demonstrated by ants. The result is shown to be largely independent of model parameter values, therefore suggesting that some aspects of higher-order processing of a visual scene may be emergent from the structure of the neural circuits, rather than computed in discrete processing modules.

Investigating visual navigation using spiking neural network models of the insect mushroom bodies

Abstract:Ants are capable of learning long visually guided foraging routes with limited neural resources. The visual scene memory needed for this behaviour is mediated by the mushroom bodies; an insect brain region important for learning and memory. In a visual navigation context, the mushroom bodies are theorised to act as familiarity detectors, guiding ants to views that are similar to those previously learned when first travelling along a foraging route. Evidence from behavioural experiments, computational studies and brain lesions all support this idea. Here we further investigate the role of mushroom bodies in visual navigation with a spiking neural network model learning complex natural scenes. By implementing these networks in GeNN–a library for building GPU accelerated spiking neural networks–we were able to test these models offline on an image database representing navigation through a complex outdoor natural environment, and also online embodied on a robot. The mushroom body model successfully learnt a large series of visual scenes (400 scenes corresponding to a 27 m route) and used these memories to choose accurate heading directions during route recapitulation in both complex environments. Through analysing our model's Kenyon cell (KC) activity, we were able to demonstrate that KC activity is directly related to the respective novelty of input images. Through conducting a parameter search we found that there is a non-linear dependence between optimal KC to visual projection neuron (VPN) connection sparsity and the length of time the model is presented with an image stimulus. The parameter search also showed training the model on lower proportions of a route generally produced better accuracy when testing on the entire route. We embodied the mushroom body model and comparator visual navigation algorithms on a Quanser Q-car robot with all processing running on an Nvidia Jetson TX2. On a 6.5 m route, the mushroom body model had a mean distance to training route (error) of 0.144 ± 0.088 m over 5 trials, which was performance comparable to standard visual-only navigation algorithms. Thus, we have demonstrated that a biologically plausible model of the ant mushroom body can navigate complex environments both in simulation and the real world. Understanding the neural basis of this behaviour will provide insight into how neural circuits are tuned to rapidly learn behaviourally relevant information from complex environments and provide inspiration for creating bio-mimetic computer/robotic systems that can learn rapidly with low energy requirements.

Abstract:Insects can navigate efficiently in both novel and familiar environments, and this requires flexiblity in how they are guided by sensory cues. A prominent landmark, for example, can elicit strong innate behaviours (attraction or menotaxis) but can also be used, after learning, as a specific directional cue as part of a navigation memory. However, the mechanisms that allow both pathways to co-exist, interact or override each other are largely unknown. Here we propose a model for the behavioural integration of innate and learned guidance based on the neuroanatomy of the central complex (CX), adapted to control landmark guided behaviours. We consider a reward signal provided either by an innate attraction to landmarks or a long-term visual memory in the mushroom bodies (MB) that modulates the formation of a local vector memory in the CX. Using an operant strategy for a simulated agent exploring a simple world containing a single visual cue, we show how the generated short-term memory can support both innate and learned steering behaviour. In addition, we show how this architecture is consistent with the observed effects of unilateral MB lesions in ants that cause a reversion to innate behaviour. We suggest the formation of a directional memory in the CX can be interpreted as transforming rewarding (positive or negative) sensory signals into a mapping of the environment that describes the geometrical attractiveness (or repulsion). We discuss how this scheme might represent an ideal way to combine multisensory information gathered during the exploration of an environment and support optimal cue integration.

Abstract:Navigation mechanisms include path integration and landmark‐based navigation, and we show the first evidence for molecular pathways required for navigation in desert ants. Transcriptional activity in the central brain but not in the optic lobes might be necessary for the estimation of distance traveled, which is crucial for path integration. Correlation in gene expression and correctness in the optic lobes may indicate the importance of visual processing in route recognition. Navigation is crucial for central‐place foragers to locate food and return to the nest. Cataglyphis ants are renowned for their advanced navigation abilities, relying on landmark cues and path integration. This study aims to uncover the transcriptomic basis of exceptional spatial learning in the central nervous system of Cataglyphis niger. Ants navigated a maze with a food reward, and we examined expression changes linked to correct decisions in subsequent runs. Correct decisions correlated with expression changes in the optic lobes, but not the central brain, showing a downregulation of genes associated with sucrose response and Creb3l1. The latter gene is homologous to Drosophila crebA, which is essential for long‐term memory formation. To understand how ants use distance information during path integration, we analyzed expression shifts associated with the last distance traveled. We uncovered a transcriptomic footprint in the central brain, but not in the optic lobes, with genes enriched for energy consumption and neurological functions, including neuronal projection development, synaptic target inhibition, and recognition processes. This suggests that transcriptional activity in the central brain is necessary for estimating distance traveled, which is crucial for path integration. Our study supports the distinct roles of different brain parts for navigation in Cataglyphis ants.

Abstract:The Mushroom Bodies, a prominent and evolutionary conserved structure of the insect brain, are known to be the support of olfactory memory. There is now evidence that this structure is also required for visual learning, but the hypotheses about how the view memories are encoded are still largely based on what is known of the encoding of olfactory information. The different processing steps happening along the several relays before the Mushroom Bodies is still unclear, and how the visual memories actually may allow navigation is entirely unknown. Existing models of visual learning in the Mushroom Bodies quickly fall short when used in a navigational context. We discuss how the visual world differs from the olfactory world and what processing steps are likely needed in order to form memories useful for navigation, and demonstrate it using a computational model of the Mushroom Bodies embedded in an agent moving in through a virtual 3D world.

The role of learning-walk related multisensory experience in rewiring visual circuits in the desert ant brain

Abstract:Efficient spatial orientation in the natural environment is crucial for the survival of most animal species. Cataglyphis desert ants possess excellent navigational skills. After far-ranging foraging excursions, the ants return to their inconspicuous nest entrance using celestial and panoramic cues. This review focuses on the question about how naïve ants acquire the necessary spatial information and adjust their visual compass systems. Naïve ants perform structured learning walks during their transition from the dark nest interior to foraging under bright sunlight. During initial learning walks, the ants perform rotational movements with nest-directed views using the earth's magnetic field as an earthbound compass reference. Experimental manipulations demonstrate that specific sky compass cues trigger structural neuronal plasticity in visual circuits to integration centers in the central complex and mushroom bodies. During learning walks, rotation of the sky-polarization pattern is required for an increase in volume and synaptic complexes in both integration centers. In contrast, passive light exposure triggers light-spectrum (especially UV light) dependent changes in synaptic complexes upstream of the central complex. We discuss a multisensory circuit model in the ant brain for pathways mediating structural neuroplasticity at different levels following passive light exposure and multisensory experience during the performance of learning walks.
A neural correlate of learning fails to predict foraging efficiency in

Abstract:The mushroom bodies (MB) are integrative structures in the insect brain that, in social bees, contribute to both visual and olfactory learning. Changes in the density of presynaptic boutons (or microglomeruli) within the calyx region of the MB have been repeatedly linked to various aspects of foraging, including forms of learning that are thought to be key to support foraging efficiency. Here we set out to directly test the relationship between foraging efficiency and microglomerulus density in a bumblebee model, We found no evidence that microglomerulus density predicted real-world foraging performance, nor any relationship with foraging experience. Instead, our data suggest a potential non-linear relationship between individual age, independent of foraging experience, and microglomerulus density in the lip region of the calyx, which is associated with olfactory processing. Our findings suggest that in real-world scenarios there is no simple direct relationship between microglomerular density, learning ability and foraging efficiency in bumblebees, highlighting the gap in knowledge regarding the relationships between learning abilities, neuroanatomy and foraging efficiency.

Abstract:Mushroom bodies (MBs), one of the central brain structures in Drosophila melanogaster, are involved in several cognitive behaviors, such as olfactory learning and memory, visual context generalization, choice behavior facing conflicting cues. Attention is a cognitive behavior, and it facilitates a focus on the attended event while filtering out irrelevant events, thereby allowing more rapid and accurate reactions at a lower threshold in primates. Using the visual orientation paradigm in a flight simulator, we observed that MBs modulate salience-based selective fixation behavior, which resembles attention in primates to a certain degree. We found that the fixation ability of MB-deficient flies was significantly reduced when the contrast levels were lowered as well as when a certain amount of background noise was applied. Moreover, MB-deficient flies exhibited poor object fixation ability in the presence of an olfactory 'distracter'. Furthermore, during visual selection among multiple objects of different contrast, flies with MBs were able to 'pop-out' of the most salient object in a three-object selection paradigm. Finally, we determined that flies exhibited cross-modal synergistic integration between olfactory and visual signals during object-fixation behavior, which was independent of MBs. Taken together, our findings suggest that MBs do not contribute to cross-modal synergetic integration between olfactory and visual signals; instead, they confer sensory gain control and inhibitory gating in flies, this property allows entry of the salient signal as well as filters out background noise and irrelevant signals.

Abstract:Summary The prevailing account of visually controlled routes is that an ant learns views as it follows a route, while guided by other path-setting mechanisms. Once a set of route views is memorised, the insect follows the route by turning and moving forwards when the view on the retina matches a stored view. We have engineered a situation in which this account cannot suffice in order to discover whether there may be additional components to the performance of routes. One-eyed wood ants were trained to navigate a short route in the laboratory guided by a single black, vertical bar placed in the blinded visual field. Ants thus had to turn away from the route to see the bar. They often turned to look at or beyond the bar and then turned to face in the direction of the goal. Tests in which the bar was shifted to be more peripheral or more frontal than in training produced a corresponding change in the ants’ paths, demonstrating that they were guided by the bar, presumably obtaining information during scanning turns towards the bar. Examination of the endpoints of turns away from the bar suggest that ants use the bar for guidance by learning how large a turn-back is needed to face the goal. We suggest that the ants’ zigzag paths are an integral part of visually guided route following. In addition, on some runs in which ants did not take a direct path to the goal, they still turned to face and sometimes approach the goal for a short stretch. This off-route goal facing indicates that they store a vector from start to goal and use path integration to track their position relative to the endpoint of the vector.
The Use of Edges in Visual Navigation by the Ant Leptothorax albipennis

Abstract:Certain navigating insects home in on their goal by moving so that currently viewed images of landmarks fall on the same retinal locations memorized during previous visits. Here we show that ants can use similar retinotopic learning to guide lengthy routes, by memorizing and walking parallel to a distinct visual edge. We induced workers of the ant Leptothorax albipennis to travel parallel to a prominent wall. When the wall’s height was changed, the ants’ paths consistently shifted toward a lowered wall and away from a raised wall, as would be expected if they attempt to keep the wall’s image at a constant retinal position. These path shifts were smaller than would be expected if the wall was the only guide to navigation, suggesting that other cues are also important. Significantly larger shifts were seen when edge guidance was enhanced by using two walls, one on each side of the path.

Importance of magnetic information for neuronal plasticity in desert ants

Abstract:Many animal species rely on the Earth's magnetic field during navigation, but where in the brain magnetic information is processed is still unknown. To unravel this, we manipulated the natural magnetic field at the nest entrance of Cataglyphis desert ants and investigated how this affects relevant brain regions during early compass calibration. We found that manipulating the Earth's magnetic field has profound effects on neuronal plasticity in two sensory integration centers. Magnetic field manipulations interfere with a typical look-back behavior during learning walks of naive ants. Most importantly, structural analyses in the ants' neuronal compass (central complex) and memory centers (mushroom bodies) demonstrate that magnetic information affects neuronal plasticity during early visual learning. This suggests that magnetic information does not only serve as a compass cue for navigation but also as a global reference system crucial for spatial memory formation. We propose a neural circuit for integration of magnetic information into visual guidance networks in the ant brain. Taken together, our results provide an insight into the neural substrate for magnetic navigation in insects.
A Method for Visual Psychophysics based on the Navigational Behaviour of Desert Ants (Melophorus bagoti)

Abstract:The Australian desert ant Melophorus bagoti is known to navigate in their complex visual environment relying on path integration and landmark learning. We investigated the navigational behaviour of desert ants in response to visual stimulus changes along their route to a food source. Ants were trained to self navigate a track between their nest and a feeder, with visual stimuli introduced near the feeder. After three days of training, we altered the visual scene by changing colours and observed the ants reactions. The results showed that ants in the test conditions, when visual changes were introduced, displayed more meandering paths, increased scanning behaviour, and slower speeds compared to control conditions. The sinuosity of their paths increased, and their orientation became less efficient. Additionally, changes in colour led to significant alterations in path characteristics, including a higher frequency of head oscillations and deviations from straight-line paths. These findings suggest that disruption to visual cues used for navigation causes noticeable changes in their movement patterns. A subset of these changes, easy to measure and calculate, can provide a signature to indicate that ants have detected a change in the visual stimuli. This proof of concept study thus highlights a method for studying visual psychophysics in ants in their natural habitat.

Abstract:Cataglyphis desert ants are charismatic central place foragers. After long-ranging foraging trips, individual workers navigate back to their nest relying mostly on visual cues. The reproductive caste faces other orientation challenges, i.e. mate finding and colony foundation. Here we compare brain structures involved in spatial orientation of Cataglyphis nodus males, gynes, and foragers by quantifying relative neuropil volumes associated with two visual pathways, and numbers and volumes of antennal lobe (AL) olfactory glomeruli. Furthermore, we determined absolute numbers of synaptic complexes in visual and olfactory regions of the mushroom bodies (MB) and a major relay station of the sky-compass pathway to the central complex (CX). Both female castes possess enlarged brain centers for sensory integration, learning, and memory, reflected in voluminous MBs containing about twice the numbers of synaptic complexes compared to males. Overall, male brains are smaller compared to both female castes, but the relative volumes of the optic lobes and CX are enlarged indicating the importance of visual guidance during innate behaviors. Male ALs contain greatly enlarged glomeruli, presumably involved in sex-pheromone detection. Adaptations at both the neuropil and synaptic levels clearly reflect differences in sex- and caste-specific demands for sensory processing and behavioral plasticity underlying spatial orientation.This article is protected by copyright. Mental navigation in the primate entorhinal cortex

Abstract:A cognitive map is a suitably structured representation that enables novel computations using previous experience; for example, planning a new route in a familiar space 1 . Work in mammals has found direct evidence for such representations in the presence of exogenous sensory inputs in both spatial 2,3 and non-spatial domains 4,5,6,7,8,9,10 . Here we tested a foundational postulate of the original cognitive map theory 1,11 : that cognitive maps support endogenous computations without external input. We recorded from the entorhinal cortex of monkeys in a mental navigation task that required the monkeys to use a joystick to produce one-dimensional vectors between pairs of visual landmarks without seeing the intermediate landmarks. The ability of the monkeys to perform the task and generalize to new pairs indicated that they relied on a structured representation of the landmarks. Task-modulated neurons exhibited periodicity and ramping that matched the temporal structure of the landmarks and showed signatures of continuous attractor networks 12,13 . A continuous attractor network model of path integration 14 augmented with a Hebbian-like learning mechanism provided an explanation of how the system could endogenously recall landmarks. The model also made an unexpected prediction that endogenous landmarks transiently slow path integration, reset the dynamics and thereby reduce variability. This prediction was borne out in a reanalysis of firing rate variability and behaviour. Our findings link the structured patterns of activity in the entorhinal cortex to the endogenous recruitment of a cognitive map during mental navigation.

Abstract:Solitary foraging insects like desert ants rely heavily on vision for navigation. While ants can learn visual scenes, it is unclear what cues they use to decide if a scene is worth exploring at the first place. To investigate this, we recorded the motor behavior of Cataglyphis velox ants navigating in a virtual reality set-up and measured their lateral oscillations in response to various unfamiliar visual scenes under both closed-loop and open-loop conditions. In naturalistic-looking panorama, ants display regular oscillations as observed outdoors, allowing them to efficiently scan the scenery. Manipulations of the virtual environment revealed distinct functions served by dynamic and static cues. Dynamic cues, mainly rotational optic flow, regulated the amplitude of oscillations but not their regularity. Conversely, static cues had little impact on the amplitude but were essential for producing regular oscillations. Regularity of oscillations decreased in scenes with only horizontal, only vertical or no edges but was restored in scenes with both edge types together. The actual number of edges, the visual pattern heterogeneity across azimuths, the light intensity or the relative elevation of brighter regions did not affect oscillations. We conclude that ants use a simple but functional heuristic to determine if the visual world is worth exploring, relying on the presence of at least two different edge orientations in the scene.

A comparative study of navigation behaviours in ants

Abstract:Ants inhabit a vast range of ecosystems and exhibit wide morphology. They are expert navigators employing a handful of well-understood navigational strategies. However, the specific relationships among ant navigation behaviours, ecology, and morphology remain unclear, highlighting the need for comparative studies across diverse species. Here, we conducted field displacement experiments with 15 ant species across different habitats, assessing the prevalence of path integration, view-based navigation, olfactory trail following, and backtracking. We further tested whether use of particular navigation strategies was correlated with variation in morphological traits that could affect navigation efficiency, namely body size, eye size (view-based, path integration) and scape length (olfactory). There was a negative correlation between path integration and olfaction across different ant species, and no other clear trade-offs were identified between navigational strategies. Olfactory navigation emerged as the most dominant strategy. Path integration was also prevalent but limited to arboreal ants. View-based navigation was observed in both ground-foraging and tree-climbing ant species, and, unexpectedly, backtracking was also widespread. Species with larger eyes and body size showed a stronger preference for view-based navigation. However, no significant relationship was found between eye size or antennal scape length with preference for either path integration or olfaction. These results highlight the diversity and specialization of navigational strategies in ants, which appear to depend on the species' ecological niche and morphological traits. Our study confirmed that path integration performs better in open sky environments, while view-based navigation appears more effective in cluttered habitats. We also showed the importance of plasticity in both foraging strategies and navigational profile at individual and colony levels, demonstrating the adaptability of ants' navigation strategies to their environment.

Abstract:Decades of research into intelligent animal navigation posits that organisms build and maintain internal spatial representations (or maps) 1 of their environment, that enables the organism to determine and follow task-appropriate paths (Epstein, Patai, Julian, & Spiers, 2017; O'keefe & Nadel, 1978; Tollman, 1948). Hamsters, wolves, chimpanzees, and bats leverage prior exploration to determine and follow shortcuts they may never have taken before (Chapuis & Scardigli, 1993; Harten, Katz, Goldshtein, Handel, & Yovel, 2020; Menzel, 1973; Peters, 1976; Toledo et al., 2020). Even blind mole rats and animals rendered situationally-blind in dark environments demonstrate shortcut behaviors (Avni, Tzvaigrach, & Eilam, 2008; Kimchi, Etienne, & Terkel, 2004; Maaswinkel & Whishaw, 1999). Ants forage for food along meandering paths but take near-optimal return trips (Müller & Wehner, 1988), though there is some controversy about whether insects like ants and bees are capable of forming maps (Cheung et al., 2014; Cruse & Wehner, 2011).

While this species seems to be sensitive to handling and reacts to visual changes in their surroundings, not much is known about how added olfactory stimuli impact their route navigation on a vertical surface. In the current study, we added one of five different invisible odours on the trees on which foragers normally forage. We found that the bull ants showed neophobic responses to all the odours. The Teatree and Lavender odours showed the strongest impact on the bull ants navigation by causing detours, U-turns, and avoidance of the sensory stimuli, with the ants meandering more and scanning more frequently. The odours of Olive oil, Flax-seed oil, and Eucalyptus oil had a moderate impact on the ants navigation. These findings showed the widespread influence of non-visual chemical cues in shaping bull ant navigation and highlight the induction of neophobic responses stemming from chemical alterations on learned routes. Overall, this study contributes to the understanding of the effects of foreign odours, adding to our understanding of the complex learning processes of bull ants in their vertical navigation.

Abstract:The ant Myrmica sabuleti uses essentially odors for navigating. It has also several skills which presume that it could learn, memorize a sequence of odors and afterwards use it for navigating. By training (operative conditioning) then testing forager workers, we demonstrate that these ants are able to soon learn and memorize a sequence of four odors, i.e., thyme, lavender, basilica, and orange zest, linearly set during training between the nest entrance and the food site, and a simple additional experiment showed that the trained ants effectively used the learned sequence of odors for moving between the nest and the food. Moreover, as workers of this ant can associate odors with the time of the day during which they perceive them, they should be able to use a memorized sequence of odors at appropriate time periods in order to optimize a safe navigation. As this ant is known to use visual cues, odometry, area marking, and deposition of the trail pheromone to navigate, it is also liable to combine these strategies with that of following odor sequences.

Abstract:Cockroaches are scavengers that forage through dark, maze-like environments. Like other foraging animals, for instance rats, they must continually asses their situation to keep track of targets and negotiate barriers. While navigating a complex environment, all animals need to integrate sensory information in order to produce appropriate motor commands. The integrated sensory cues can be used to provide the animal with an environmental and contextual reference frame for the behavior. To successfully reach a goal location, navigational cues continuously derived from sensory inputs have to be utilized in the spatial guidance of motor commands. The sensory processes, contextual and spatial mechanisms, and motor outputs contributing to navigation have been heavily studied in rats. In contrast, many insect studies focused on the sensory and/or motor components of navigation, and our knowledge of the abstract representation of environmental context and spatial information in the insect brain is relatively limited. Recent reports from several laboratories have explored the role of the central complex (CX), a sensorimotor region of the insect brain, in navigational processes by recording the activity of CX neurons in freely-moving insects and in more constrained, experimenter-controlled situations. The results of these studies indicate that the CX participates in processing the temporal and spatial components of sensory cues, and utilizes these cues in creating an internal representation of orientation and context, while also directing motor control. Although these studies led to a better understanding of the CX's role in insect navigation, there are still major voids in the literature regarding the underlying mechanisms and brain regions involved in spatial navigation. The main goal of this review is to place the above listed findings in the wider context of animal navigation by providing an overview of the neural mechanisms of navigation in rats and summarizing and comparing our current knowledge on the CX's role in insect navigation to these processes. By doing so, we aimed to highlight some of the missing puzzle pieces in insect navigation and provide a different perspective for future directions.
