Abstract:Polybrominated Diphenyl Ethers (PBDEs), recognized as persistent organic pollutants by the United Nations Environment Programme, are a class of brominated flame retardants that pose significant environmental and health risks. These compounds, consisting of two aromatic rings with up to 10 bromine atom substitutions, are categorized into penta-BDE, octa-BDE, and deca-BDE based on their bromine content, each exhibiting distinct environmental behaviors. PBDEs with fewer bromine atoms are more volatile and prone to bioaccumulation, raising significant health concerns. These compounds, often added physically to products, can leach into the environment, leading to pollution during production and after the parent polymer degrades. The transformation of higher brominated diphenyl ethers into less brominated forms in the environment further complicates their impact, with mono-brominated BDE-3 being particularly concerning due to its extended atmospheric photolysis lifetime and increased bioavailability. The management of PBDEs is challenging due to their persistence and transformation in the environment. As endocrine disruptors, they are linked to various acute and chronic toxicological effects, including neurodevelopmental toxicity, teratogenicity, and potential carcinogenicity. Their structural similarity to thyroid hormones allows them to disrupt thyroid hormone balance, leading to further health complications. The subject of this review is to summarize the current body of knowledge that is essential to understand their long-term effects on ecosystems and human health and to develop strategies to mitigate their adverse impacts.

Abstract:Polybrominated diphenyl ethers (PBDEs), especially commercial decabrominated diphenyl ethers (c-decaBDE), have been widely produced and applied to numerous materials because of their highly effective flame-retardant capabilities. The production of commercial pentaBDE (c-pentaBDE) and commercial octaBDE (c-octaBDE) ended in 2004 because they are persistent, bioaccumulative, and toxic to both humans and the environment, but decaBDE production and use continue. Furthermore, many congeners of PBDEs are still prevalent in consumer products and articles that they pose enormous threat to both the environment and human health. PBDEs have been detected in the casing of electrical and electronic equipment, textile materials, automotive interiors, polyurethane foam (PUF) in seat cushions, children’s toys, kitchenware, and other products. With increasing evidence about PBDE pollution and the adoption of international conventions, many developed countries have drawn more public attention to PBDEs and developed sound strategies for their management. This review summaries the utilization and management of PBDEs in a number of countries and reaches the conclusion that PBDEs are still prevalent in consumer articles, while specific regulations or policies for articles containing PBDEs are rare. Public awareness should be raised on the importance of sound management of articles containing PBDEs.

Abstract:Polybrominated diphenyl ethers (PBDEs) are brominated compounds connected by ester bonds between two benzene rings. There are 209 congeners of PBDEs, classified according to the number and position of the bromine atoms. Due to their low cost and superior flame retardant properties, PBDEs have been extensively used as flame retardants in electronic products, plastics, textiles, and other materials since the 1970s. PBDEs are classified as persistent organic pollutants (POPs) under the Stockholm Convention because of their environmental persistence, bioaccumulation, and toxicity to both humans and wildlife. Due to their extensive use and significant quantities, PBDEs have been detected across a range of environments and biological organisms. These compounds are known to cause damage to the metabolic system, exhibit neurotoxicity, and pose reproductive hazards. This review investigates the environmental distribution and human exposure pathways of PBDEs. Using China—a country with significant PBDE use—as an example, it highlights substantial regional and temporal variations in PBDE concentrations and notes that certain environmental levels may pose risks to human health. The article then examines the toxic effects and mechanisms of PBDEs on several major target organs, summarizing recent research and the specific mechanisms underlying these toxic effects from multiple toxicological perspectives. This review enhances our understanding of PBDEs' environmental distribution, exposure pathways, and toxic mechanisms, offering valuable insights for further research and management strategies.

Abstract:Because of their highly effective flame-retardant capability, polybrominated diphenyl ethers (PBDEs) have been extensively used as flame retardants in consumer goods. However, compelling evidence shows that many congeners of PBDEs have been accumulating in the environment, in biota and in human populations worldwide. In China, although octabrominated diphenyl ether (octaBDE) has never been produced or used, pentabrominated diphenyl ether (pentaBDE) and decabrominated diphenyl ether (decaBDE) have been produced and used in large quantities. In the face of increasing evidence about PBDE pollution and the adoption of international conventions, there is a growing push for China to develop more stringent methods of managing PBDE waste. This paper summarizes the information about PBDE production and application, describes the flame-retarding mechanism, and then reviews the toxicity and levels of PBDEs in China's environmental media and human tissues. Based on international regulations on PBDEs, the paper finally puts forward some suggestions for Chinese policy making and for self-regulation within the flame retardant industry.

Abstract:Polybrominated diphenyl ethers (PBDEs) are among the emerging contaminants that have been traced in almost all environmental compartments for the past 30 years. Their continued application as flame-retardant additives, persistence in nature due to fluorine groups, global atmospheric transport, and analytical challenges due to interferences and different properties of congeners indicate the urgent need of finding solutions to their use. The increasing level of PBDEs in the environment and especially human tissues is alarming due to their potential neurological effects, cancer proliferation, and thyroid hormone imbalance. Therefore, strict regulations need to be applied in all countries to control the PBDEs production consumption and disposal into the environment. Studies have shown that conventional wastewater treatment plants are unable to degrade PBDEs resulting in the transport of 60–90% of PBDEs to soil through biosolids application. On the other hand, advanced treatment processes, such as ultraviolet light, advanced oxidation, and photocatalytic degradation showed promising potential for removing PBDEs from wastewater (70–100% degradation efficiency). PBDEs can be replaced by natural flame retardants, such as nanoclay or new polymers, such as bishydroxydeoxybenzoin which have no environmental or health problems compared to PBDEs.

Abstract:Secretory leukoprotease inhibitor (SLPI) is a secreted glandular protein thought to regulate elastase activity and, more recently, to inhibit both mast cell chymase activity and histamine release from mast cells. To begin to examine the possible role of SLPI in humans, we determined the distribution of SLPI in the human nasal mucosa and quantitated the functional activity of SLPI in nasal lavage fluid. Immunochemical staining of the nasal mucosa revealed intense, selective immunoreactivity in the serous cells of the submucosal glands. The level of SLPI in nasal secretions was measured by enzyme immunoassay. In control subjects (n = 8), the level of SLPI in nasal lavage fluid (NLF) after saline challenge (baseline level) was 2.5 +/- 0.5 micrograms/ml, accounting for 3.3 +/- 0.6% of total protein in nasal secretions. After methacholine (MCh) and histamine (HIST) challenge, the level of SLPI increased to 7.0 +/- 1.4 and 6.1 +/- 1.6 micrograms/ml, respectively (both p < 0.05). In atopic patients (n = 8), the level of SLPI after MCh and HIST challenge increased from a baseline level of 7.6 +/- 2.0 micrograms/ml to 22.1 +/- 6.4 and 25.2 +/- 10.5 micrograms/ml, respectively. After allergen challenge, the concentration of SLPI increased significantly in atopic patients, whereas there was no increase in the level of SLPI in control subjects. Western blot analysis of MCh-induced nasal secretions revealed a single band with a molecular weight of 12 kD, the same as recombinant SLPI.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:Polybrominated diphenyl ethers (PBDEs) are a class of brominated flame retardants (BFRs), present in the environment, animals, and humans. Their levels, distribution, and human exposure have been studied extensively, and over the last decade, various legal measures have been taken to prohibit or minimize their production and use due to the increasing amount of evidence of their harmful effects on human and animal health.Our aim here was to make a comprehensive and up-to-date review of the levels and distribution of PBDEs in the aquatic environment, air, and soil, in indoor dust, and in humans. To fulfill this, we searched through Web of Science for literature data reported in the last five years (2015–2019) on levels of at least six key PBDE congeners in abovementioned matrices. According to our summarized data, significant PBDE mass concentrations/fractions are still being detected in various sample types across the world, which implies that PBDE contamination is an ongoing problem. Secondary sources of PBDEs like contaminated soils and landfills, especially those with electronic and electrical waste (e-waste), represent a particular risk to the future and therefore require a special attention of scientists.

Abstract:PBDEs are human-influenced chemicals utilized massively as flame retardants. They are environmentally persistent, not easily degraded, bioaccumulate in the biological tissue of organisms, and bio-magnify across the food web. They can travel over a long distance, with air and water being their possible transport media. They can be transferred to non-target organisms by inhalation, oral ingestion, breastfeeding, or dermal contact. These pollutants adsorb easily to solid matrices due to their lipophilicity and hydrophobicity; thus, sediments from rivers, lakes, estuaries, and ocean are becoming their major reservoirs aquatic environments. They have low acute toxicity, but the effects of interfering with the thyroid hormone metabolism in the endocrine system are long term. Many congeners of PBDEs are considered to pose a danger to humans and the aquatic environment. They have shown the possibility of causing many undesirable effects, together with neurologic, immunological, and reproductive disruptions and possible carcinogenicity in humans. PBDEs have been detected in small amounts in biological samples, including hair, human semen, blood, urine, and breastmilk, and environmental samples such as sediment, soil, sewage sludge, air, biota, fish, mussels, surface water, and wastewater. The congeners prevailing in environmental samples, with soil being the essential matrix, are BDE 47, 99, and 100. BDE 28, 47, 99, 100, 153, 154, and 183 are more frequently detected in human tissues, whereas in sediment and soil, BDE 100 and 183 predominate. Generally, BDE 153 and 154 appear very often across different matrices. However, BDE 209 seems not frequently determined, owing to its tendency to quickly breakdown into smaller congeners. This paper carried out an overview of PBDEs in the environmental, human, and biota niches with their characteristics, physicochemical properties, and fate in the environment, human exposure, and health effects.

Abstract:Polybrominated diphenyl ethers (PBDEs) used extensively over the past 3 decades as flame retardants in most types of polymers, all over the world, have been identified as global pollutants. PBDEs pose various health problems such as thyroid hormone disruption, permanent learning and memory impairment, behavioral changes, hearing deficits, delayed puberty onset, fetal malformations, and possibly cancer. Many measurements of PBDEs in various matrices from Sweden, Holland, Japan, the USA, and elsewhere have been reported, but few measurements are available for India. In this study, a preliminary screening of different congeners of PBDEs has been performed in different old electronic and consumer products with an objective to build capacity in order to analyze PBDEs and BFRs. Six different samples, foam from upholstery, motherboard of a computer, children toy composite sample, old vanishing window blind sample, electrical wire sample, and PVC flooring sample, were collected and analyzed for the presence of the following PBDE congeners: BDE-28, BDE-47, BDE-99, BDE-100, BDE-153, BDE-154, BDE-183, and BDE-209. It was found that three out of six samples were positive for the presence of PBDEs. Three congeners were detected in the samples, i.e., BDE-47, BDE-153, and BDE-209, of which, highest concentration was of BDE-209. Among the samples, motherboard of computer showed the highest concentration of BDE-209 followed by window blind and foam from upholstery. The results of this preliminary investigation indicate that PBDEs are still present in the old consumer products which can be an important additional source of exposure to the population.

Abstract:Polybrominated diphenyl ethers (PBDEs) are a group of flame retardants used in plastics, textiles, polyurethane foam, and other materials. They contain two halogenated aromatic rings bonded by an ester bond and are classified according to the number and position of bromine atoms. Due to their widespread use, PBDEs have been detected in soil, air, water, dust, and animal tissues. Besides, PBDEs have been found in various tissues, including liver, kidney, adipose, brain, breast milk and plasma. The continued accumulation of PBDEs has raised concerns about their potential toxicity, including hepatotoxicity, kidney toxicity, gut toxicity, thyroid toxicity, embryotoxicity, reproductive toxicity, neurotoxicity, and immunotoxicity. Previous studies have suggested that there may be various mechanisms contributing to PBDEs toxicity. The present study aimed to outline PBDEs' toxic effects and mechanisms on different organ systems. Given PBDEs' bioaccumulation and adverse impacts on human health and other living organisms, we summarize PBDEs' effects and potential toxicity mechanisms and tend to broaden the horizons to facilitate the design of new prevention strategies for PBDEs-induced toxicity.

Brominated flame retardants - Exposure and risk assessment for the general population

Abstract:Brominated flame retardants (BFRs) are a large group of different substances used in numerous products to prevent fire hazards. Some of them are persistent in the environment, accumulate in the food chain and are of toxicological concern, while for others current data are limited. Meanwhile, BFRs have been found in many environmental media, foods, and biota including humans. This review presents recent findings obtained from monitoring data in environmental media relevant for human exposure, as well as dietary exposure. In this context, concentrations in indoor and ambient air and in house dust are outlined. Furthermore, we summarize human biomonitoring data on BFR levels in blood and breast milk. Current estimates of the overall exposure of the general population using different relevant subsets are also addressed. All of these data are discussed in relation to currently available toxicological reference values used for risk assessment purposes. Obviously, the exposure of the general population varies considerably in different parts of the world and even within countries. Polybrominated diphenyl ethers (PBDEs) and hexabromocyclododecane (HBCD) show the highest intake during infancy. While the highest intake for BDE 47 for all groups was observed in the US, the total BDE 209 and HBCD intake was highest in the UK. For HBCD and all PBDEs except BDE 209, diet accounts for a large proportion of the total intake during infancy in all countries. With regard to toddlers and adults, the contribution of diet to total intake is high in Germany and the UK, while in the US, the high concentrations of PBDE in dust resulted in a notably smaller proportion of the intake being attributed to diet.

Abstract:With hexabromocyclododecane (HBCDs) and poly brominated diphenyl ethers (PBDEs) listed as persistent organic pollutants (POPs), tetrabromobisphenol A (TBBPA) and decabromodiphenyl ethane (DBDPE) have become two of the most widely produced and used brominated flame retardants (BFRs). The underlying ecological risk of TBBPA and DBDPE have received increasing attentions since these two typical BFRs are being detected at high concentrations in environmental media. This paper summarized the pollution status of TBBPA and DBDPE and their toxicological effects. TBBPA and DBDPE have been detected in multiple environmental media, such as atmosphere, waters, soil, sediment and organisms. In addition, more severe pollution could be found in industrial areas. TBBPA and DBDPE were even found in human body and breast milk. Overall, TBBPA presented developmental toxicity, hepatorenal toxicity, endocrine disruption effect, reproductive toxicity, and neurotoxicity, while DBDPE showed developmental toxicity, hepatorenal toxicity, and endocrine disruption effect. According to the limited reports on DBDPE toxicity, we concluded that DBDPE was of relatively low toxicity. The aim of this review is to help evaluate the environmental risk, analyze the environmental capacity, and governmentally control the production of TBBPA and DBDPE.

Abstract:Since 1970, polybrominated diphenyl ether (PBDE) has been widely used as additive flame retardants in everyday consumer products, including polyurethane foam and electronic products like mattresses and upholstered furniture. Thermoplastics, mixed in polymers, do not chemically bond with plastics, textiles, etc., and therefore can be separated from the product into the environment. Because of its high lipophilicity, accumulation, degradation-resistance and biochemical toxicity, PBDE can invade the human body in a variety of ways and is toxic to multiple systems in the human body. PBDE affects the male reproductive system in many aspects, as by causing sperm quality decline, epididymis, seminal vesicle and prostate dysplasia, sperm head deformity, and decreased levels of testosterone and other reproductive hormones. PBDE also affects male reproductive function from the genetic aspect, as by altering the sperm DNA methylation level, inducing sperm chromatin damage, etc. Some environmental factors, such as high-fat diet and indoor dust increase, can indirectly promote the reproductive toxicity of PBDE. This article reviews the impacts of PBDE on exposed populations and the animal reproductive system and the latest research progress at home and abroad.
Environmental release and behavior of brominated flame retardants

Abstract:Recently, environmental problems relating to brominated flame retardants (BFRs) have become a matter of greater concern than ever before, because of the recent marked increase in levels of polybrominated diphenyl ethers (PBDEs) found in human milk in Sweden and North America. The question that arises is whether environmental levels of PBDEs and other BFRs will continue to increase, causing toxic effects to humans. In an attempt to elucidate the current state of the science of BFRs, we review the consumer demand for BFRs (mainly in Japan), the characteristics of waste flame-retarded products, sources of emission, environmental behavior, routes of exposure of humans, temporal trends, and thermal-breakdown products of BFRs. At present, flame-retarded consumer products manufactured 10-20 years ago, when PBDEs were frequently used, are being dumped. The possible major sources of emission of BFRs into the environment are effluent and flue gases from BFR factories and other facilities processing BFRs. With respect to the environmental behavior of BFRs, the lower brominated compounds are, on the whole, predicted to be more volatile, more water soluble, and more bioaccumulative than the higher brominated compounds. The most probable route for exposure of the general human population to PBDEs, especially the lower brominated congeners, is through the diet. The release of BFRs from consumer products treated with these compounds could also lead to human exposure. Temporal trends in PBDE levels in the environment and in humans worldwide seem to vary considerably, depending on the regions or country, with possible reflections of the historic and current use of PBDEs. The environment and the general human population are also exposed to the thermal-breakdown products of PBDEs, such as polybrominated and mixed brominated/chlorinated dibenzo-p-dioxins and dibenzofurans (PBDDs/DFs and mixed PXDDs/DFs).

Abstract:Disruption of epigenetic regulation by environmental toxins is an emerging area of focus for understanding the latter’s impact on human health. Polybrominated diphenyl ethers (PBDEs), one such group of toxins, are an environmentally pervasive class of brominated flame retardants that have been extensively used as coatings on a wide range of consumer products. Their environmental stability, propensity for bioaccumulation, and known links to adverse health effects have evoked extensive research to characterize underlying biological mechanisms of toxicity. Of particular concern is the growing body of evidence correlating human exposure levels to behavioral deficits related to neurodevelopmental disorders. The developing nervous system is particularly sensitive to influence by environmental signals, including dysregulation by toxins. Several major modes of actions have been identified, but a clear understanding of how observed effects relate to negative impacts on human health has not been established. Here, we review the current body of evidence for PBDE-induced epigenetic disruptions, including DNA methylation, chromatin dynamics, and non-coding RNA expression while discussing the potential relationship between PBDEs and neurodevelopmental disorders.
Critical review of soil contamination by polybrominated diphenyl ethers (PBDEs) and novel brominated flame retardants (NBFRs); concentrations, sources and congener profiles

Abstract:Polybrominated diphenyl ethers (PBDEs) have been used in a broad array of polymeric materials such as plastics, foams, resins and adhesives to inhibit the spread of fires since the 1970s. The widespread environmental contamination and well documented toxic effects of PBDEs have led to bans and voluntary withdrawals in many jurisdictions. Replacement novel brominated flame retardants (NBFRs) have, however, exhibited many of the same toxic characteristics as PBDEs and appear to share similar environmental fate. This paper presents a critical review of the scientific literature regarding PBDE and NBFR contamination of surface soils internationally, with the secondary objective of identifying probable pollution sources. An evaluation of NBFR distribution in soil was also conducted to assess the suitability of the newer compounds as replacements for PBDEs, with respect to their land contamination potential. Principle production of PBDEs and NBFRs and their consequent use in secondary polymer manufacture appear to be processes with strong potential to contaminate surrounding soils. Evidence suggests that PBDEs and NBFRs are also released from flame retarded products during disposal via landfill, dumping, incineration and recycling. While the land application of sewage sludge represents another major pathway of soil contamination it is not considered in this review as it is extensively covered elsewhere. Both PBDEs and NBFRs were commonly detected at background locations including Antarctica and northern polar regions. PBDE congener profiles in soil were broadly representative of the major constituents in Penta-, Octa- and Deca-BDE commercial mixtures and related to predicted market place demand. BDE-209 dominated soil profiles, followed by BDE-99 and BDE-47. Although further research is required to gain baseline data on NBFRs in soil, the current state of scientific literature suggests that NBFRs pose a similar risk to land contamination as PBDEs.
Polybrominated diphenyl ethers (PBDEs) in household dust: A systematic review on spatio-temporal distribution, sources, and health risk assessment

Abstract:Much attention has been paid on polybrominated diphenyl ethers (PBDEs) in household dust due to their ubiquitous occurrences in the environment. Based on the data from 59 articles sampled from 2005 to 2020, we investigated the spatio-temporal distribution, sources, and health risk of 8 PBDE homologues in household dusts worldwide. BDE-209 is the predominant PBDE in household dusts, followed by BDE-99 and BDE-47. The total concentrations of PBDEs (∑ 8 PBDEs) are found to be high in household dusts sampled from 2005 to 2008 and show a significant decline trend from 2009 to 2016 ( p < 0.05) and a little upward tendency from 2017 to 2020. The concentrations of PBDEs in household dusts vary greatly in different countries of the world. The use of penta-BDE is the main source of three to five bromo-biphenyl ether monomers contributing 17.4% of ∑ 8 PBDEs, while BDE-209 and BDE-183 are derived from the use of household appliances contributing 82.6% of ∑ 8 PBDEs. Ingestion is the main exposure route for adults and toddlers, followed by dermal contact. The values of hazard index (HI) exposed to PBDEs in household dusts are all less than 1 for both adults and toddlers, indicating a low non-cancer risk. The incremental lifetime cancer risks (ILCRs) of BDE-209 are less than 10 −6 for both adults and toddlers, suggesting a negligible risk. However, the total carcinogenic risk of toddlers is higher than that of adults, indicating that much attention should be paid to toddlers exposed to BDE-209 in household dust.

Abstract:Polybrominated diphenyl ethers(PBDEs)as flame retardants in polymeric materials for fire safety of home products are new global environmental pollutants.Humans exposure to PBDEs mainly through foods,indoor air,etc.Levels of PBDEs in environmental medias and human body increased significantly in last 30 years.Laboratory data showed that PBDEs exposure could disturb endocrine function such as thyroid and sex hormone.Although mechanisms are not clear,PBDEs were found to induce toxicity to liver,nervous system,reproduction,and human development.However,limited data on human were reported.Environmental PBDEs pollution was found in China and more research are needed in China to evaluate environmental and human health risks from PBDEs exposure.
Legacy and novel brominated flame retardants in interior car dust - Implications for human exposure

Abstract:Brominated flame retardants (BFRs) are organobromine compounds with an inhibitory effect on combustion chemistry tending to reduce the flammability of products. Concerns about health effects and environmental threats have led to phase-out or restrictions in the use of Penta-, Octa- and Deca-BDE technical formulations, increasing the demand for Novel BFRs (NBFRs) as replacements for the banned formulations. This study examined the occurrence of legacy and NBFRs in the dust from the interior of private cars in Thessaloniki, Greece, aged from 1 to 19 years with variable origin and characteristics. The determinants included 20 Polybrominated Diphenyl Ethers (PBDEs) (Di-to Deca-BDEs), four NBFRs such as Decabromodiphenylethane (DBDPE), 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), 2-ethylhexyl-2,3,4,5-tetrabromobenzoate (TBB), and bis(2-ethylhexyl)-3,4,5,6-tetrabromophthalate (TBPH), three isomers of hexabromocyclododecane (HBCD), and tetrabromobisphenol A (TBBPA). The concentrations of ∑20PBDE ranged from 132 to 54,666 ng g-1 being dominated by BDE-209. The concentrations of ∑4NBFRs ranged from 48 to 7626 ng g-1 and were dominated by DBDPE, the major substitute of BDE-209. HBCDs ranged between <5 and 1745 ng g-1, with alpha-HBCD being the most prevalent isomer Finally, the concentrations of TBBPA varied from <10 to 1064 ng g-1. The concentration levels and composition profiles of BFRs were investigated in relation to the characteristics of cars, such as year of manufacture, country of origin, and interior equipment (type of car seats, electronic and electrical components, ventilation, etc.). The average daily intakes of selected BFRs (BDE-47, BDE-99, BDE-153, BDE-209, TBB, BTBPE, TBPH, DBDPE, HBCDs and TBBPA) via ingestion and dermal absorption were estimated for adults and toddlers. The potential health risk due to BFRs was found to be several orders of magnitude lower than their corresponding reference dose (RfD) values.
