Abstract:Background: Self-association of the HRD complex is important for its function in ER quality control, but the oligomeric state of the complex is still unclear. Results: The luminal component Yos9 dimerizes independently. Conclusion: Dimerization of Yos9 suggests a dimeric state of the HRD complex. Significance: The assembly of a functional HRD complex oligomer is further elucidated on a structural level. In yeast, the membrane-bound HMG-CoA reductase degradation (HRD) ubiquitin-ligase complex is a key player of the ER-associated protein degradation pathway that targets misfolded proteins for proteolysis. Yos9, a component of the luminal submodule of the ligase, scans proteins for specific oligosaccharide modifications, which constitute a critical determinant of the degradation signal. Here, we report the crystal structure of the Yos9 domain that was previously suggested to confer binding to Hrd3, another component of the HRD complex. We observe an αβ-roll domain architecture and a dimeric assembly which are confirmed by analytical ultracentrifugation of both the crystallized domain and full-length Yos9. Our binding studies indicate that, instead of this domain, the N-terminal part of Yos9 including the mannose 6-phosphate receptor homology domain mediates the association with Hrd3 in vitro. Our results support the model of a dimeric state of the HRD complex and provide first-time evidence of self-association on its luminal side.
Effects of dry-salting and brine-pickling processes on the physicochemical properties, nonvolatile flavour profiles and bacterial community during the fermentation of Chinese salted radishes

Abstract:The profiling of physicochemical properties, nonvolatile flavour compounds and bacterial community during the fermentation of Chinese dry-salted and brine-pickled radishes was performed. Dry-salted radishes matured on the 30th day, and the pH, total titratable acid, and water activity of the matured samples were 4.7, 0.43 g/100 g lactic acid, and 0.90, respectively. Brine-pickled radishes matured in only 5 days, and the pH, total titratable acid, and water activity were 3.4, 0.31 g/100 g lactic acid, and 0.96, respectively. The contents of reducing sugars, organic acids, and free amino acids generally increased with the fermentation of dry-salted radishes, reaching 15.8 g/100 g, 1009.1 mg/100 g, and 300.3 mg/100 g on the 30th day, respectively. Lactic acid was the characteristic flavour substance of brine-pickled radishes. Exiguobacterium, Glutamicibacter, and Leuconostoc were the dominant genera when the dry-salted radishes matured. Lactobacillus dominated brine-pickled radish fermentation and was correlated to lactic acid, Arg, Val, and His (P<0.05). This study could improve our understanding of the effects of dry salting and brine pickling on the flavour and bacterial characteristics of salted vegetables.

Abstract:The aim of this study was to investigate the effect of inoculated fermentation on the quality of Chinese sauerkraut. To this end we studied a co-culture system consisting of Lactobacillus plantarum Shanghai brewing 1.08 and Zygosaccharomyces rouxii CGMCC 3791 during inoculated sauerkraut fermentation. The nitrite concentrations in pickled cabbage and radish inoculated with starter cultures of L. plantarum and Z. rouxii were significantly lower than those in the spontaneous fermentation system during the whole fermentation process. In addition, co-culture of L. plantarum and Z. rouxii during the production of sauerkraut decreased the formation of biogenic amines in the pickled vegetables. Using gas chromatography–mass spectrometry we also compared the levels of volatile compounds in inoculated and naturally fermented Chinese sauerkraut. Sixty compounds were identified, with the sauerkraut inoculated with starter cultures containing overall higher contents of volatile compounds, including acids, alcohols, esters, and phenols. The structure of the microbial community during the production of sauerkraut was studied using phospholipid fatty-acid (PLFA) analysis. This analysis revealed that the brine of inoculated sauerkraut contained significantly higher contents of Gram-positive and fungal PLFAs and a lower content of Gram-negative PLFAs, suggesting that the improved quality of inoculated Chinese sauerkraut may be ascribed to the inhibition of the growth of Gram-positive during sauerkraut fermentation. These results may indicate a new strategy to enhance the quality of Chinese sauerkraut.

Abstract:Salt profoundly affects the physicochemical properties and microbial abundance of fermented foods such as suan cai, a popular traditional fermented food in China. It is vital to systematically investigate the effects of salt concentrations on fermented suan cai for high fermentation quality and large-scale production. We elucidated the effects of salt concentrations on Lactobacillus curvatus (LC-20) and suan cai during fermentation, and found that salt (0–1%) favoured an increase in LC-20 growth and a decrease in pH (salt: 0–2%). For suan cai fermentation, the results from sensory scoring judged 1% salt treatment the highest. Salt concentration also affected the nitrite content of the fermentation system with peak nitrite values in low salt treatments being significantly higher on the first day, and gradually decreasing to similar levels. After fermentation, the total titratable acid and lactic acid concentrations in the 0–1% treatments were higher (p < 0.05) than those in 2–5% treatments. The colony forming units of lactic acid bacteria increased initially and then decreased after 6 d of fermentation. At the phylum level, Firmicutes and Proteobacteria were predominant in all treatments, and at the genus level, Lactobacillus dominated the fermentation. Other lactic acid bacteria such as Lactococcus and Weissella were also detected. Quantitative PCR showed DNA concentration of LC-20 at 0.5–2% salt treatments were higher than that in other treatments and L. curvatus was the dominant microorganism during fermentation. Hence, we conclude that L. curvatus could be used for suan cai product at low salt concentrations.

Abstract:Mustard green paocai is a salted vegetable product that is fermented with low salt in the initial stage and preserved with high salt in the middle stage. To reveal the key flavor-producing potential of core bacterial and fungal communities during the 365-day fermentation of industrial-scaled paocai. In this study, microbiome and molecular sensory science were used to analyze potential relationships between microbial diversity and key flavor compounds. The results showed that bacteria were more diverse than fungi. High salinity improved the flavor of industrial paocai and simplified the microbial community. A total of 74 bacterial and yeast strains were isolated and identified throughout the fermentation process. Lactiplantibacillus, Halanaerobium, Halomonas, Pseudomonas, and Enterobacteriaceae were the dominant bacterial genera, and Debaryomyces hansenii was the core fungal genera in paocai. 4-Ethylphenol, anethole, butanoic acid, 4-ethylguaiacol, and 3-methyl-1-butanol were identified as key flavor compounds in industrial paocai with odor activity values greater than 1. Spearman correlation analysis indicated that bacteria have a greater impact on the flavor of paocai than fungi. Halanaerobium and Halomonas were correlated with flavor and spoilage in paocai. This study provides guidance for the production of desirable flavors by multi-bacteria synergistic fermentation of industrial paocai.

Abstract:The microbial community of lactic acid bacteria isolated from Chinese Paocai prepared with different salt concentrations (2％,moderate 5％ and 8％),was assessed by cultivable methods and various molecular biology methods.The results showed that lactic acid bacteria in Paocai brine exhibited faster growth rate at lower salt concentration during the fermentation process.A total of 563 lactic acid bacteria were isolated from Paocai brines fermented with different salt concentrations and identified by 16S rDNA sequencing analysis,multiplex-PCR method,and PCR-FRLP analyses.The isolates were identified to belong to 11 species of 5 genera respectively.Lactococcus lactis,Lactobacillus pentosus and Leuconostoc were found to be the dominant flora in Paocai samples during the early stage of fermentation with lower salt concentration (2％) while Lactobacillus pentosus dominated the late fermentation stage.Furthermore,Lactobacillus pentosus and Weissella were the most abundant isolates from Paocai brine during the early stage of fermentation with 5％ and 8％ salt concentrations,but the major microbes involved in the late fermentation stage were still Lactobacilluspentosus.

Abstract:Northeastern Chinese sauerkraut is a well-known traditional fermented vegetable in China. Incomplete identification of the microorganisms’ (bacteria in spontaneous fermentation) diversity and accumulation of nitrite make it difficult to normalize the fermentation process and product qualities of northeastern Chinese sauerkraut. Conventional culturing and polymerase chain reaction-denaturing gradient gel electrophoresis methods were combined to describe microbial structure and diversity. Lactobacillus, Leuconostoc, Enterobacter, Accumulibacter, Thermotoga, Pseudomonas, Clostridium, Rahnella and Citrobacter were predominant microorganisms in different fermentation periods. The pH value and nitrite concentration presented a certain relevance to the amount of lactic acid bacteria. Lactobacillus and Leuconostoc had the ability to decrease nitrite by inhibiting nitrate-reducing bacteria such as Enterobacter. Therefore, Northeastern Chinese sauerkraut should not be eaten until 4 weeks of fermentation for the safety and quality of fermented foods. Northeastern Chinese sauerkraut is rich in lactic acid bacteria, which demonstrate its ability as an excellent probiotic for applications in functional foods.

Sauerkraut with the Addition of Lactobacillus casei: Effects of Salt and Sugar Concentrations on Fermentation and Antioxidant Activity

Abstract:Background: Spontaneous fermentations have several disadvantages, and additions of lactic acid bacteria can improve sauerkraut fermentations and quality. Salt and sugar concentrations are important factors in sauerkraut fermentations, and the growth and activity of lactic acid bacteria can affect antioxidant activity of sauerkrauts. Objective: This study developed sauerkrauts with the addition of Lactobacillus casei and investigated how salt and sugar concentrations affected their fermentation and antioxidant activity. Methods: A fresh cabbage was washed and cut, before salt (1.0%; 1.5% and 2.0%) and sugar (0% and 2.0%) were added, prior to inoculation with a L.casei culture at 10% (v/w). The cabbage was then incubated at 28oC for 5 days. The controlled lactic fermentation of the cabbage without culture and sugar, but with salt at 2.5% was performed. The sauerkrauts were evaluated for total lactic acid bacteria, pH, total acidity, phenolic content and DPPH scavenging activity. Results: The fermentation increased the total lactic acid bacteria counts in the sauerkrauts from 1.38 x 109to 3.86 x 109 cfu/mL, and this demonstrated significant (p<0.05) direct salt and sugar effects. The high salt and sugar concentrations also increased the total acidity, antioxidant activity and phenolic content, but lowered the pH of the sauerkrauts. The additives (L.casei culture, salt and sugar), therefore, improved the sauerkraut fermentation and antioxidant activity compared to the control. Conclusion: With added sugar and salt, L. casei improved the sauerkraut fermentation and antioxidant activity, and both sugar and salt can each be added at a low (2%) concentration for maximum effects.

Abstract:The microbial compositions and metabolites of fermented sauerkraut with and without the addition of microorganisms have been compared. The OTU clustering, nonvolatile compounds, volatile compounds and associations between bacterial taxa and metabolites were analyzed by 16S rRNA high-throughput sequencing technology, ultra performance liquid chromatography (UPLC), gas chromatography ion mobility mass spectrometry (GC-IMS) and the O2PLS model studies. The results showed that at the phylum level, the microbial species in the four sauerkraut types consisted mainly of the phyla Firmicutes and Proteobacteria, but different modes of microbial addition formed their own unique microbial communities. There were significant differences in the microbial communities among different northeast China sauerkraut samples, and different microbial communities exerted similar effects to inhibit Firmicutes production. At the genus level, sauerkraut without added microorganisms had the lowest microbial diversity. A total of 26 amino acids and 11 organic acids were identified and were more abundant in nonmicrobially fermented sauerkraut; 88 volatile organic compounds were identified in the 4 types of sauerkraut, with the microbially fermented sauerkraut being richer in alcohols, esters and acids. Different brands of sauerkraut contain their own unique flavor compounds. Cystine and tyrosine, ascorbic acid and acetic acid, and alcohols and esters are closely related to a wide range of microorganisms in sauerkraut. Elucidating the correlations among microbiota and metabolites will help guide future improvements in sauerkraut fermentation processes.

Abstract:This work aimed to investigate the effect of salt concentration on the quality and microbial community of pickled peppers during fermentation, and the cross-correlation between microorganisms and quality was also revealed. The results showed that 9 volatile flavor compounds were unique to the low salt concentration group (D group), which also contained higher content of FAA, lactic acid and acetic acid than high salt concentration group (G group). Meanwhile, the samples of D2 group have a better texture properties. Firmicutes, Proteobacteria, Ascomycota, Lactobacillus, Pectobacterium, and Pseudomonas were detected as the main microbial community during the fermentation with different salt concentrations. Furthermore, the correlations analysis results indicated that the salt concentration has a significant effect on the microbial community of pickled peppers (p < 0.001), and Pediococcus, Lactobacillus, Cedecca, Issatchenkia, Pichia, Kazachstania, and Hanseniaspora were significantly correlated with flavors, which played crucial roles in the unique flavor formation of pickled peppers.
Adequacy of prenatal care of urban high school students.

Abstract:Relationships of five selected variables to the adequacy of prenatal care of pregnant students in urban public high schools were studied. These five variables were: the age of the baby's father, mother's education, parity, living arrangement, and functional areas of support. From 1986 to 1989, a convenience sample of 244 African American, unmarried, pregnant students completed a prenatal questionnaire. The Resource Size scale, a subscale in the questionnaire, was tested on pregnant students, and its coefficient alpha for internal consistency was .86. Additional data were obtained from the state's live birth certificate tape. Results showed that the age of the baby's father was significantly associated with adequacy of prenatal care. Because 51% of the babies' fathers are age 15-19 and most likely in school, the community and school health nurses may encourage them to get involved in the care of pregnant students. Other implications are also drawn.
The lactic acid bacteria and yeast community of home-made sauerkraut from three provinces in Southwest China

Abstract:The aim of this study was to investigate the lactic acid bacteria (LAB) and yeast community from home-made sauerkraut collected from Southwest China through culture-dependent and culture-independent technology. Forty-eight samples of home-made sauerkraut were collected from households at three different locations in Southwest China. The pH, total acidity and salt contents among these fermented vegetables were 3.69 ± 0.42, 0.86 ± 0.43 g/100 ml, and 3.86 ± 2.55 g/100 ml, respectively. The number of lactic acid bacteria (LAB) and yeasts were 7.25 ± 1.05 log10 colony-forming units (CFU)/ml and 3.74 ± 1.01 log CFU/ml, respectively. A total of 182 LAB and 81 yeast isolates were identified. The dominant isolates were Lactobacillus plantarum, L. brevis, Pediococcus ethanolidurans, Pichia membranifaciens, P. fermentans and Kazachstania bulderi. Denaturing gradient gel electrophoresis (DGGE) showed that L. plantarum, uncultured Lactobacillus sp, P. ethanolidurans, and K. exigua were the predominant microflora. Our studies demonstrated that the DGGE technique combined with a culture-dependent method is very effective for studying the LAB and yeast community in Chinese traditional fermentation vegetables. The results will give us an understanding of LAB and yeast community of Chinese sauerkraut and improve the knowledge of LAB and yeast community of Chinese sauerkraut.
Exploring potential correlations between bacterial communities, organic acids, and volatile metabolites of traditional fermented sauerkraut collected from different regions of Heilongjiang Province in Northeast China

Abstract:In this study, the bacterial communities and flavor metabolites of 27 traditional naturally fermented sauerkraut samples collected from nine regions of Heilongjiang Province in Northeast China were investigated. The dominant genera were Lactobacillus, Pseudomonas, Alcaligenes, Arcobacter, Pseudarcobacter, Lactococcus, Comamonas, Pediococcus, Prevotella, and Insolitispirillum. A total of 148 volatile compounds were detected in seven categories; esters and acids were the most abundant volatiles. Additionally, the highest content (15.96 mg/g) of lactic acid was detected in YC1. Acetic acid, oleic acid, palmitic acid, elaidic acid, and dehydroacetic acid were the key differential volatile compounds, which may be related to the bacterial communities. Spearman's correlation analysis revealed that Lactococcus and Lactobacillus were significantly positively correlated with flavor metabolites, suggesting that they may play a more significant role in flavor formation. The results of this study can help in the development of better quality of fermented vegetables.
Effect of Low Salt Concentrations on Microbial Changes During Kimchi Fermentation Monitored by PCR-DGGE and Their Sensory Acceptance

Abstract:Various salt concentrations (1.0%, 1.3%, 1.6%, 1.9%, and 2.1% labeled as sample A, B, C, D, and E, respectively) were investigated for microbial diversity, identification of Lactic Acid Bacteria (LAB) in salted kimchi cabbage, prepared under laboratory conditions. These samples were stored at 4°C for 5 weeks in proper aluminum-metalized pouch packaging with calcium hydroxide gas absorber. A culture-independent method known as polymerase chain reaction - denaturing gradient gel electrophoresis was carried out to identify LAB distributions among various salt concentration samples that had identified 2 Weissella (W. confusa and W. soli), 1 Lactobacillus (Lb. sakei), and 3 Leuconostoc (Lc. mesenteroides, Lc. lactis, and Lc. gelidum) in the overall kimchi samples. The pH, titratable acidity, viable cell counts, and coliform counts were not affected by salt variations. In order to assess sensory acceptance, the conducted sensory evaluation using a 9-point hedonic scale had revealed that samples with 1.3% salt concentration (lower than the manufacturer's regular salt concentration) was more preferred, indicating that the use of 1.3% salt concentration was acceptable in normal kimchi fermentation for its quality and safety. Despite similarities in pH, titratable acidity, viable cell counts, coliform counts, and LAB distributions among the various salt concentrations of kimchi samples, the sample with 1.3% salt concentration was shown to be the most preferred, indicating that this salt concentration was suitable in kimchi production in order to reduce salt intake through kimchi consumptions.
Bacterial profiles and volatile flavor compounds in commercial Suancai with varying salt concentration from Northeastern China

Abstract:Suancai, a popular traditional fermented cabbage in northeast China, is commonly prepared by the spontaneous fermentation process at a certain salt concentration. The salt can affect the metabolites by affecting the microorganisms during Suancai fermentation. The bacterial community and volatile flavor compounds in commercial Suancai from different regions of Northeastern China at different salt concentrations were investigated using next-generation sequencing and GC-MS. Firmicutes and Cyanobacteria were the dominant phyla in the commercial Suancai, and Lactobacillus, Pediococcus, and Leuconostoc were the dominant genera. Among them, Lactobacillus and Pediococcus were considered as the biomarkers of the low and high salt Suncai, respectively. Eighty-five volatile flavor compounds were detected, and HS exhibited higher contents of volatile flavor compounds than LS. Based on the results of correlation analysis, Pediococcus were highly correlated with the alcohols and nitriles in Suancai. The contents of alcohols and nitriles significantly increased in the Suancai, fermented by Pediococcus pentosaceus. The co-inoculated fermentation of Lactobacillus plantarum and P. pentosaceus could increase the concentrations of alcohols, esters, aldehydes, hydrocarbons, and nitriles in Suancai. This study provides a perspective for understanding the ecology of Suancai fermentation and facilitating the fermentation with multispecies inoculation fermentation at an appropriate salt concentration.
Effects of substrates and suppliers of ingredients on microbial community and metabolites of traditional non-salt Suancai

Abstract:Aim: Non-salt Suancai is an acidic fermented vegetable consumed by the Chinese Yi ethnic group. Traditionally, it is produced by fermentation without salt in a cold environment. The present study aimed to investigate the metabolite and microbial characteristics, and the effects of substrates/suppliers ingredients on non-salt Suancai. Methods: A simulated fermentation system of non-salt Suancai was constructed by using different substrates/suppliers' ingredients. The coherence and differential detection of the metabolite and microbial characteristics were done through non-target metabolomic and metagenomic analysis. Results: Lactic acid was the predominant organic acid across all samples. The enumeration of the Lactic acid bacteria showed no discernible differences between study groups, but that of yeast was highest in the mustard leaf stem (Brassica juncea var. latipa). The three major biological metabolic pathways were metabolism, environmental information, and genetic information processing based on the KEGG database. The metabolite diversity varied with the substrate/supplier of ingredients based on the PLS-DA plot. Lactiplantibacillus, Leuconostoc, and Lactococcus were prevalent in all samples but differentially. The microbial diversity and richness varied significantly, with 36~291 species being identified. Among the various substrates collected from the same supplier, 29, 59, and 29 differential species were identified based on LEfSe [linear discriminant analysis (LDA) > 2, P < 0.05]. Leuconostoc citreum, Leuconostoc mesenteroides, Leuconostoc pseudomesenteroides, Lactiplantibacillus plantarum, and Leuconostoc lactis were likely to be used as the species to discriminate samples collected from different suppliers. Conclusions: This research contributed to the exploration of microbial and metabolite characteristics behind the ingredient restriction of non-salt Suancai using traditional technology.
Effect of Different Salt Additions on the Flavor Profile of Fermented Ciba Pepper

Abstract:Salt is a key ingredient that can both enhance the taste and extend the shelf life of fermented vegetables. However, it is important to note that excessive salt levels can have adverse effects on consumer health. This study aimed to investigate the impact of various salt additions (2%, 4%, 6%, 8%, and 10% wt/wt) on the flavor profile of fermented ciba pepper, a traditional Chinese fermented chili sauce, using gas chromatography–ion mobility spectrometry (GC-IMS) in combination with an electronic nose (E-nose). Fermented ciba pepper samples were prepared with different salt additions: 2% (LJA), 4% (LJB), 6% (LJC), 8% (LJD), and 10% (LJE) (wt/wt). The physicochemical and sensory properties of the fermented ciba pepper samples were evaluated. Sensory evaluation indicated that LJC and LJD received higher scores compared to the other groups. The total acid and amino acid nitrogen contents displayed contrasting trends with the salt additions (p < 0.05). The E-nose analysis successfully differentiated the flavor profiles of the ciba pepper samples fermented with varying salt additions. Additionally, the GC-IMS analysis identified a total of 72 volatile compounds, including 14 alcohols, 21 esters, nine aldehydes, four acids, eight ketones, three terpenes, and eight other substances. Notably, the ciba pepper samples with lower salt additions exhibited higher levels of alcohols, aldehydes, and esters. In conclusion, the addition of salt during the fermentation process significantly influenced the formation of flavor compounds in ciba pepper. This study provides valuable insights into ciba pepper fermentation with different salt additions and offers prospects for the development of low-salt fermented ciba pepper products.

Abstract:The effect of salt concentration on soy sauce fermentation was studied. Four groups of soy sauce samples with 8%, 12%, 16%, and 20% salt concentrations were prepared and their physicochemical parameters, non-volatile metabolites (organic acids, free amino acids, and biogenic amines), volatile metabolites, and microbial abundance were analyzed during the whole fermentation. The sensory characteristics of the final products were also assessed. Salt concentration significantly affected the microbial abundance and metabolite profiles during soy sauce fermentation, resulting in different characteristics among samples. The RT-qPCR analysis showed that the microbial abundance in lower salted soy sauce was higher. Correspondingly, the accumulation of total acid and the decrease of reducing sugar were observed in lower salted soy sauce, aggravating the sourness of soy sauce. The levels of free amino acids and biogenic amines (detected by HPLC) in lower salted soy sauce were higher, resulting in a taste of umami but making unsafe soy sauce. Additionally, HS-SPME/GC-MS analysis showed that several distinctive volatile compounds were absent in lower salted soy sauce. Sensory evaluation revealed that the highest total score for the quality of soy sauce was obtained through fermentation under 16% salt concentration. This study helped deepen our understanding of the effect of salt on soy sauce fermentation and provide a theoretical data reference for low salt soy sauce development.

Abstract:To explore the effect of low-temperature and low-salt fermentation on the volatile flavor substances of Chinese kohlrabi, low-temperature and low-salt fermented Chinese kohlrabi (LSCK) and traditional high-salt fermented Chinese kohlrabi (HSCK) were produced. The physicochemical and texture properties of the two kinds of Chinese kohlrabies were evaluated. Headspace gas chromatography-ion mobility spectrometry (GC-IMS) and electronic nose (E-nose) were used to analyze the volatile flavor substances of the kohlrabi. The results showed that the total acid content significantly decreased (p < 0.05), while protein and reducing sugar contents significantly increased (p < 0.05) by low-temperature and low-salt fermentation. A total of 114 volatile flavor substances were identified. The alcohol, ketone, pyrazine, ether, and nitrile contents in LSCK were significantly higher than those in HSCK (p 1, including tetrahydrothiophene, ethyl 3-(methylthio)propanoate, 3-methylbutyric acid, hexanenitrile, and 3-methyl-3-buten-1-ol, could be used as potential biomarkers for identifying LSCK and HSCK. The E-nose analysis further demonstrated that there was a significant difference in overall flavor between the LSCK and HSCK. The present study provides support for the development of green processing technology and new low-salt Chinese kohlrabi products.
