Abstract:Stimulus-responsive microcapsules pose an opportunity to achieve controlled release of the entire load instantaneously upon exposure to an external stimulus. Core-shell microcapsules based on the polyanhydride poly(bis(2-carboxyphenyl)adipate) as a shell were formulated in this work to encapsulate the model active substance pyrene and enable a pH-controlled triggered release. A remarkably narrow triggering pH interval was found where a change in pH from 6.4 to 6.9 allowed for release of the entire core content within seconds. The degradation kinetics of the shell were measured by both spectrophotometric detection of degradation products and mass changes by quartz crystal microbalance with dissipation monitoring and were found to correlate excellently with diffusion coefficients fitted to release measurements at varying pH values. The microcapsules presented in this work allow for an almost instantaneous triggered release even under mild conditions, thanks to the designed core-shell morphology.

Abstract:Here, we report a novel thermo-triggered-releasing microcapsule for liposoluble drug delivery. Monodisperse microcapsules with a poly(N-isopropylacrylamide-co-methacrylic acid) hydrogel shell and an oil core were successfully fabricated by a double coaxial microfluidic device. Fluorescent dye Lumogen Red F300 as a model liposoluble drug was dissolved in the oil core with controllable loading capacity. The volume phase transition temperature (VPTT) of the microcapsule was adjusted by copolymerizing with the hydrophilic methacrylic acid. The in vitro release study demonstrates that the shells shrink, leading to the thermo-triggered release of the model drug from the microcapsules at the environmental temperature above the VPTT, while the swollen hydrogel shells can protect the encapsulated drug from leakage and contamination below the VPTT. The proposed microcapsule is a promising liposoluble drug delivery system with controllable loading and smart thermo-triggered release.

Photomechanically Controlled Encapsulation and Release from pH-Responsive and Photoresponsive Microcapsules

Abstract:Poly(acrylic acid)/azobenzene microcapsules were obtained through distillation precipitation polymerization and the selective removal of silica templates by hydrofluoric acid etching. The uniform, robust, and monodisperse microcapsules, confirmed by transmission electron microscopy and scanning electron microscopy, had reversible photoisomerization under ultraviolet (UV) and visible light. Under UV irradiation, azobenzene cross-linking sites in the main chain transformed from the trans to cis isomer, which induced the shrinkage of microcapsules. These photomechanical effects of azobenzene moieties were applied to the encapsulation and release of model molecules. After loading with rhodamine B (RhB), the release behaviors were completely distinct. Under steady UV irradiation, the shrinkage adjusted the permeability of the capsule, providing a novel way to encapsulate RhB molecules. Under alternate UV/visible light irradiation, a maximal release amount was reached due to the continual movement of shell networks by cyclic trans-cis photoisomerization. Also, microcapsules had absolute pH responsiveness. The diffusion rate and the final release percentage of RhB both increased with pH. The release behaviors under different irradiation modes and pH values were in excellent agreement with the Baker-Lonsdale model, indicating a diffusion-controlled release behavior. Important applications are expected in the development of photocontrolled encapsulation and release systems as well as in pH-sensitive materials and membranes.

Abstract:Light-responsive drug delivery systems are particularly appealing that are capable of releasing active molecules at the appropriate site and rate. We synthesized a series of photodegradable polymers that can form nanoparticles for drug encapsulation. These particles in aqueous solutions are stable in buffers with different pHs or at evaluated temperatures, while light can trigger the crash of particles and the release of encapsulated substances. The release efficiency can reach up to 90% based on Nile red fluorescence intensity upon 15 min light irradiation. Nanoparticle uptake by phagocytic cells and light-triggered release in cells were observed by fluorescence emission of the hydrolyzed fluorescein diacetate upon photoinduced degradation of these nanoparticles. No significant toxicity of these nanoparticles was found at the concentrations up to 1000 μg/mL before or after light irradiation. Further encapsulation and triggered release of a bioactive model drug (Tagalsin G) was evaluated for RAW 264.7 cells. Tagalsin G encapsulated in nanoparticles did not show cytotoxity to cells, while light triggered the release of Tagalsin G increasing cell death dramatically from 9% to 67%. Our model studies show a new promising strategy to trigger drug release in cells.

Abstract:The development of photo-responsive capsules to tune and control the sustained-release of encapsulated actives is a fascinating and challenging route to improve the performances and effectiveness of a wide range of delivery applications. In this work, we report the preparation of visible light-responsive capsules obtained via oil-in-water interfacial polycondensation between modified diacyl-chloride azobenzene moiety and diamine flexible spacer in the presence of cross-linkers with different structures and functionalities. The effect on the release profile of the encapsulated perfume oil was investigated using three flexible spacers with different lengths (1,8-diaminooctane; 1,6-diaminohexane and 1,4-diaminobutane) and two types of cross-linkers (1,3,5-benzenetricarbonyl trichloride and melamine). We analyzed how the properties of microcapsules can be tailored changing the design of the shell structure. Fine tuning of the perfume release profiles was obtained. The changes in capsules size and morphology due to visible light irradiation were monitored via light scattering, optical microscopy and atomic force microscopy. Perfume release was 50% faster in the systems prepared with melamine as the cross-linker. Modelling studies were carried out to support the discussion of the experimental results.

Abstract:In this study, dual-responsive polymeric nanocapsules, in which the state (swelling or collapse) can be repeatedly controlled by external stimuli (i.e., temperature and light), have been designed and prepared through distillation–precipitation polymerization. Temperature sensitive monomers of N-isopropylacrylamide are cross-linked by photoresponsive bis(methacryloylamino) onto a silica nanospherical template to form a core–shell (SiO2–PNIPAM/Azo) structure. The silica core is then removed by hydrofluoric acid to produce PNIPAM/Azo nanocapsules (P/ANCs) of diameter ∼238 nm at ∼25 °C. The size of the nanocapsule is temperature responsive and, as such, its diameter could be reduced to ∼182 nm on increasing the temperature to 40 °C. In addition, the permeability of nanocapsules can be adjusted by UV irradiation. The cis–trans transformation of modified azobenzene allowed us to perform both the encapsulation and controlled release of molecules. Rhodamine B (RhB) was successfully encapsulated using the photomechanical method. In controlled release experiments, after the majority of RhB (∼45%) was released from the P/ANCs using temperature (∼40 °C) and UV light, a second stage of release could be triggered by lowering the temperature (∼18.4%) and applying UV–visible lighting cycles (∼29.4%), respectively. We found that the diffusion coefficient, D, was 45% larger under alternate irradiation than UV light alone. Our results demonstrate considerable potential for customizable delivery systems for a variety of drugs.
Synthesis of Light-Induced Expandable Photoresponsive Polymeric Nanoparticles for Triggered Release

Abstract:High stability of drug-delivery nanocarriers during blood circulation is critical for effective drug delivery and low systematic toxicity, although destabilization of these nanocarriers is required for efficient release when they reach target sites. To develop efficient polymeric nanocarriers, we intended to synthesize and characterize a group of cross-linked, light-induced, expandable, polymeric nanoparticles through miniemulsion polymerization. These synthesized nanoparticles were stable in aqueous solutions, although light irradiation led to particle uncaging and further particle expansion up to 315-fold in volume. This resulted in the efficient release of the encapsulated contents in aqueous solutions and three cell lines (HeLa, RAW264.7, and MCF-7). Selective triggered release was also successfully achieved with spatial resolution in cell monolayers. In addition, curcumin encapsulation and photoregulation of its release were realized. Further cell viability of encapsulated curcumin was successfully achieved with light activation.

Abstract:We intend to form photosensitive block copolymer micelles for controllable release of encapsulated substances. Here, we designed and synthesized a new photocleavable cross-linker (2-nitrophenyl ethylene glycol dimethacrylate) for methyl methacrylate (MMA) atom transfer radical polymerization. Four different ratios (0:1, 1:26, 1:16, 1:8.8) of the photocleavable cross-linker to MMA monomer were used and four block copolymers (P0, P1, P2, P3) were synthesized with PEO-Br as the macroinitiator. Gel permeation chromatography and (1) H NMR studies showed that linear polymer molecules could be cross-linked by the photocleavable linker. The fluorescence studies of the encapsulated Nile Red (NR) showed that there were lower critical micelle concentrations for the polymer P1, P2 and P3 than polymer P0. And dynamic light scattering and SEM confirmed the formation of polymer micelles. Photolysis experiments demonstrated that NR encapsulated in the polymer micelles could be released upon UV irradiation (365 nm, 11 mW cm(-2)) due to the breakage of the photocleavable linker and the generation of more hydrophilic acid moieties, which destabilized polymer micelles. Our study shows a new strategy for the possibility of photocontrollable drug release for hydrophobic drugs.
One-Pot Synthesis of Redox-Labile Polymer Capsules Via Emulsion Droplet-Mediated Precipitation Polymerization

Abstract:Monodisperse poly(vinylcaprolactam) (PVCL)-based capsules are prepared by precipitation polymerization of vinylcaprolactam (VCL) onto dimethyldiethoxysilane (DMDES) emulsion droplets and removal of the DMDES templates by ethanol. Polymer chains in the shells can be cross-linked during the polymerization by disulfide-containing cross-linker N,N'-bis(acryloyl) cystamine, which endows the capsules with an excellent redox-labile property. Versatility of this technique to prepare capsules with diverse components is demonstrated by the copolymerization of methacrylic acid (MAA) and VCL in the shell to prepare poly(vinylcaprolactam-co-methacrylic acid) (P(VCL-co-MAA)) capsules. The disulfide-bonded capsules can degrade efficiently into low molecular weight species (ca. 1200 Da) when the capsules are incubated with 10 mM glutathione (GSH) as the reducing agent. Delivery of the anticancer drug (doxorubicin, DOX) was also investigated in the P(VCL-co-MAA) capsules. The cumulative in vitro release of DOX-loaded capsules allows a relatively low DOX release at pH 7.4. However, a burst release (ca. 90% in 6 h) of DOX was observed in the presence of 10 mM GSH. Cell viability assays show that the P(VCL-co-MAA) capsules have negligible cytotoxicity to HeLa cancer cells. In comparison, DOX-loaded P(VCL-co-MAA) capsules cause significant cell death following internalization. The reported capsules represent a novel and versatile class of stimuli-responsive carriers for controlled drug delivery.

Abstract:Stimuli-responsive/smart drug delivery systems (DDSs), particularly those that use temperature as a stimuli-response factor to activate drug release, are the subject of recent research. A phase change material (PCM) is a popular thermally responsive material that can be used as a drug carrier and only when the system temperature is above the phase change point is the drug released following the phase change material changing from solid to liquid. In this study, a novel NIR light-triggered temperature-sensitive drug delivery system is developed for controllable release of acyclovir (ACV). For this purpose, a mixture of a phase change material (T38) and an ACV compound is first emulsified with copper oxide nanoparticles (CuO NPs) as a Pickering stabilizer and a photothermal conversion material, and then encapsulated with SiO 2 to form a photothermal stimuli-responsive delivery system. This system shows a uniform spherical shape with a well-distinct core–shell structure, and is further experimentally proven to be able to controllably release drugs with solid–liquid transition of the phase change carrier upon temperature change. These results indicate that cumulative release of ACV can reach 51.2% at 40 °C within 20 hours, which is much higher than 27.3% release achieved below the melting point of T38. In addition, CuO NPs with excellent photothermal conversion ability endow the system with precisely controllable drug delivery via NIR light stimulation, where the cumulative drug release can reach 83.6% after 7 cycles of light stimulation, allowing controlled release at a specific time or location.

Abstract:Stimuli-responsive polymeric drug delivery nanocarriers potentially allow for drug release profiles in a spatiotemporal manner via multistimuli, either exogenous or endogenous. In this study, poly(N-isopropylacrylamide-co-N,N-dimethylacrylamide-b-glycidyl methacrylate) (P(NIPAM-co-DMA)-b-PGMA) was first synthesized through photoinduced electronic transfer reversible addition-fragmentation chain transfer (PET-RAFT) polymerization, using Eosin Y and 5,10,15,20-tetraphenyl-21H,23H-porphine zinc (ZnTPP) as photocatalysts under visible-light irradiation at room temperature. The polymer was then functionalized with 4-(phenylazo)phenol (Azo). Gel permeation chromatography characterization confirmed the efficient PET-RAFT polymerization of NIPAM with low dispersity (PDI, & Dstrok; < 1.35), temporal control, and chain-end fidelity, achieved under blue-light irradiation (lambda(max) = 420 nm, 24 mW/cm(2)). The subsequent blocking of PGMA on the P(NIPAM-co-DMA) chain was successfully accomplished with red-light irradiation (lambda(max) = 625 nm, 5.8 mW/cm(2)). After Azo grafting to the epoxy groups, a thermal, pH, and photo multistimuli responsive polymer was self-assembled in water, with a curcumin encapsulation efficiency of 90%, and particle morphologies observed with sizes ranging from 220 to 600 nm. The nanoparticles exhibited a lower critical solution temperature value (LCST) of 38.09 degrees C, and could undergo trans/cis isomerization of the azobenzene structure under UV irradiation. As a result, the nanoparticles demonstrated significant responsiveness to pH, UV, and heat stimuli. Curcumin was smoothly released under UV exposure and/or at 50 degrees C, with a release ratio of over 80% within 3 h. Additionally, rapid curcumin release was observed in an alkaline environment. Overall, the visible-light-induced PET-RAFT polymerization technique shows great potential in the controlled synthesis of multiresponsive intelligent drug delivery nanocarriers.

Abstract:The majority of current pH-triggered release systems is designed to respond to either low or high pH. Encapsulants based on polyampholytes are an example of materials that can respond to both acidic and basic pH. However, polyampholyte-based encapsulants generally possess a low loading capacity and have difficulty retaining their small-molecule cargo. The current work utilizes interfacial polymerization between polyamines and a pyromellitic diester diacid chloride to form high capacity "liquid core-shell" polyamide microcapsules that are stable in a dry or nonpolar environment but undergo steady, controlled release at pH 7.4 and accelerated release at pH 5 and pH 10. The rate of release can be tuned by adjusting the amine cross-linker feed ratio, which varies the degree of cross-linking in the polymer shell. The thin-shell microcapsule exhibited suitable barrier properties and tunable dual acid/base-triggered release, with applications in a wide range of pH environments.

Abstract:A controlled drug delivery system (DDS) was designed by integrating the thermoresponsive copolymer poly(N-isopropylacrylamide-co-methacrylic acid) (poly(NIPAAm-co-MAA)) with core-shell 1,6-hexanediol diacrylate (HDDA) microparticles. The monodisperse HDDA particles with a hollow core and a nanoporous shell were fabricated in a continuous manner by an initially proposed inkjet printing process combined with UV polymerization. The thermoresponsive poly(NIPAAm-co-MAA) copolymer was grafted onto the surface of HDDA microcapsules by free radical initiated polymerization. Particularly, the lower critical solution temperature (LCST) of the copolymer was adjusted to human physiological temperature by the optimal comonomer ratio of MAA. With temperature changes at around the LCST, the copolymer, which was modified on the internal nanopore, served as a "retractable gate" by virtue of its changes in conformation between swollen and collapsed structures. Thus, controlled drug release was achieved by the reversible "open-close" transition characteristics of the nanopores. Fluorescein as a hypothetical drug molecule was loaded in the microcapsules and used to investigate the controlled release of the material. The results confirmed that this system represents a promising candidate for use in preparing controlled DDSs.

Abstract:In this study, we incorporate capsaicin microcapsules into UV-curable coatings applied on PVC substrates to achieve sustained capsaicin release coatings. The microcapsules are synthesized using interfacial polycondensation, overcoming self-limitation nature of interfacial polycondensation through solvent evaporation and swelling effects, resulting in microcapsules with well-defined thick shells that can encapsulate capsaicin successfully. These capsaicin microcapsules exhibit enhanced thermal stability compared to pure capsaicin as revealed by thermogravimetry. When embedded into UV-curable coatings, the capsaicin microcapsules contribute to increased hardness, abrasion resistance, tensile strength with uncompromised adhesion strength. Cumulative releases in accelerated tests demonstrate that these coatings have the lowest release of 40% while pure capsaicin can reach cumulative release of 90% in the investigated time scale, indicating a coordinated regulation of capsaicin release by microcapsule shell. Importantly, these UV-curable coatings embedded with sustained release capsaicin microcapsules offer a promising solution for rodent repellency, particularly applicable to power cable surfaces in the power and telecommunication industries.

Abstract:To well adapt to the complicated physiological environments, it is necessary to engineer dual- and/or multi-stimuli responsive drug carriers for more effective drug release. For this, a novel temperature responsive lateral chain photosensitive block copolymer, poly[(N-isopropylacrylamide-co-N,N-dimethylacrylamide) -block-propyleneacylalkyl-4-azobenzoate] (P(NIPAM-co-DMAA)-b-PAzoHPA), is synthesized by atom transfer radical polymerization. The structure is characterized by 1 H nuclear magnetic resonance spectrometry and laser light scattering gel chromatography system. The self-assembly behavior, morphology, and sizes of micelles are investigated by fluorescence spectroscopy, transmission electron microscope, and laser particle analyzer. Dual responsiveness to light and temperature is explored by ultraviolet-visible absorption spectroscopy. The results show that the copolymer micelles take on apparent light and temperature dual responsiveness, and its lower critical solution temperature (LCST) is above 37 °C, and changes with the trans-/cis- isomerization of azobenzene structure under UV irradiation. The blank copolymers are nontoxic, whereas the paclitaxel (PTX)-loaded counterparts possessed comparable anticancer activities to free PTX, with entrapment efficiency of 83.7%. The PTX release from the PTX-loaded micelles can be mediated by changing temperature and/or light stimuli. The developed block copolymers can potentially be used for cancer therapy as drug controlled release carriers.

Abstract:Multifunctional triggered drug release systems have been one of the most intriguing and challenging topics. In this study, we synthesize amphiphilic copolymers composed of hydrophilic oligo ethylene glycol methyl ether methacrylate and hydrophobic azobenzene-containing methacrylate. These amphiphilic copolymers can easily self-assemble into micelles in aqueous media and show smart responses to temperatures, UV/vis lights and ions. Interestingly, the obtained copolymers can self-assemble onto hydrophobic upconversion nanoparticles in aqueous media via hydrophobic interaction to form hybrid colloidal clusters with the hydrophobic nanocrystal core and hydrophilic polymeric shell. The hybrid colloidal clusters exhibit a thermo-responsive and photosensitive performance. When served as a drug carrier, both the near-infrared light and temperature triggered drug release can be realized, which have potential applications in intelligent drug delivery.
Far-red light-triggered cargo release from liposomes bound to a photosensitizer-cellulose nanofiber hydrogel

Abstract:Light-triggered release from liposomes has been extensively researched. In virtually all the cases the photosensitizer has been encapsulated in liposomes core or liposomal membrane. However, the light-triggered drug release system, where the photosensitizer is located outside of the liposomes has never been reported before. The central concept of our research revolves around the far-red light-induced release of cargo from liposomes embedded in anionic nanofibrillar cellulose (ANFC), wherein the photosensitizer resides in the hydrogel, outside of liposomes. Both the sensitizer and the liposomes bind strongly to the anionic cellulose nanofibers by simple mixing, making the system easy to fabricate. Upon light activation, this photosensitizer generates reactive oxygen species, which, in turn, induce the liposomal bilayer disruption, ultimately resulting in cargo release. The release mechanism is the generation of reactive oxygen species and the subsequent oxidation of unsaturated lipids in the liposomal membrane. We were able to achieve ca. 70 % release of model hydrophilic cargo molecule calcein with a relatively low dose of light (262 J/cm2) while employing straightforward fabrication techniques for the drug delivery system. Our system was remarkably responsive to the far-red light (730nm, 0.875 W/cm²), enabling deep tissue penetration. Therefore, this very promising novel cellulose-immobilized photosensitizer liposomal complex could be used as a controlled drug delivery system, which can have potential applications in externally activated coatings or implants.

Abstract:Dynamic microcapsules are a highly sought-after class of encapsulant for advanced delivery applications with dynamically tunable release profiles, as actively manipulatable microreactors, or as selective microtraps for molecular separation and purification. Such dynamic microcapsules can only be realized with a nondestructive trigger-response mechanism that changes the permeability of the shell membrane reversibly, as found in hydrogels. However, the direct synthesis of a trigger-responsive hydrogel membrane around a water drop without the use of sacrificial templates remains elusive due to the incompatibility of the synthesis chemistry with aqueous emulsion processing. Here, we report on a facile approach to fabricate reversibly responsive hydrogel microcapsules utilizing reactive anhydride chemistry. Cross-linked and hydrophobic poly(methacrylic anhydride) microcapsules are obtained from microfluidic double emulsion drop templating that enables direct encapsulation of hydrophilic, water-suspended cargo within the aqueous core. Hydrolysis in aqueous environment yields microcapsules with a poly(acid) hydrogel shell that exhibit high mechanical and chemical stability for repeated cycling between its swollen and nonswollen states without rupture or fatigue. The permeability of the microcapsules is strongly dependent on the degree of swelling and hence can be actively and dynamically modified, enabling repeated capture, trap, and release of aqueous cargo over numerous cycles.

Abstract:Microcapsules containing water-soluble ammonium persulfate (APS) cores as gel breakers that offer controlled burst release, are become increasingly important in improving the gel breaking efficiency of fracturing fluids in oil fields. To date, microcapsules with various control slow release behaviors have been thoroughly investigated, but microcapsules with burst release properties are rarely studied. Here, we reported a novel inverse emulsion polymerization method to exploit this new type of microcapsule. The microcapsules were composed of APS cores and stimuli-responsive P(MMA-co-AA) copolymer shells. The microcapsules have spherical structures with diameters of 10 to 16 μm. The encapsulated cores offer controlled burst release from the microcapsules and their beginning time of burst release could be delayed up to 42 h. Moreover, all of the cores could be completely released within 1 h from the microcapsules. The temperature, pH, and salt concentration change could stimulate the release. These novel microcapsules might have promising applications in oil exploration, batteries, monomer polymerization, fiber printing, and grease.
