Abstract:Salt stress is a major abiotic stress factor affecting crop production, and understanding of the response mechanisms of seed germination to salt stress can help to improve crop tolerance and yield. The differences in regulatory pathways during germination in different salt-tolerant barley seeds are not clear. Therefore, this study investigated the responses of different salt-tolerant barley seeds during germination to salt stress at the proteomic and metabolic levels. To do so, the proteomics and metabolomics of two barley seeds with different salt tolerances were comprehensively examined. Through comparative proteomic analysis, 778 differentially expressed proteins were identified, of which 335 were upregulated and 443 were downregulated. These proteins, were mainly involved in signal transduction, propanoate metabolism, phenylpropanoid biosynthesis, plant hormones and cell wall stress. In addition, a total of 187 salt-regulated metabolites were identified in this research, which were mainly related to ABC transporters, amino acid metabolism, carbohydrate metabolism and lipid metabolism; 72 were increased and 112 were decreased. Compared with salt-sensitive materials, salt-tolerant materials responded more positively to salt stress at the protein and metabolic levels. Taken together, these results suggest that salt-tolerant germplasm may enhance resilience by repairing intracellular structures, promoting lipid metabolism and increasing osmotic metabolites. These data not only provide new ideas for how seeds respond to salt stress but also provide new directions for studying the molecular mechanisms and the metabolic homeostasis of seeds in the early stages of germination under abiotic stresses.
Exogenous Application of Salicylic Acid Improves Growth and Yield of Black Gram Vigna mungo L. by Improving Antioxidant Defense Mechanism under Saline Conditions

Abstract:The current experiment was designed to evaluate the effect of salicylic acid (SA) on the growth, physiological, biochemical, antioxidant and agronomic parameters of black gram Vigna mungo (L.) Hepper under salinity stress. One group of experimental plants of V. mungo varieties ADT-5 was treated with 75 mM NaCl, and the other group was kept without NaCl as control. Various concentrations of SA (0.5, 0.75, and 1 mM) were sprayed on the experimental plant to test its effect. Under salinity stress, the morpho-biochemical parameters were significantly reduced in the salt-treated plant. The results revealed that SA foliar spray mitigated the salinity-induced adverse effect on plant. The apparent increase in chlorophyll a by 81%, chlorophyll b by 63%, carotenoid by 25%, decreased malondialdehyde by 5% and electrolyte leakage by 29% was reported using SA (0.5 mM) foliar spray under salt stress. Foliar application of SA enhanced proline content and enzymatic antioxidants enzymes viz., catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), and glutathione reductase (GR) activity of V. mungo by 18, 29, 46, 37, and 38%, respectively, compared to the control group. It was revealed that foliar application of SA could enhance the yield of the V. mungo under salt stress conditions. Our results demonstrated that SA mitigated the deleterious impact of salt stress and may be used as a tool to enhance food production under global climate change.

Abstract:Soil salinity hampers the survival and productivity of crops. To minimize salt-associated damages in plant, better salt management practices in agriculture have become a prerequisite. Seed priming with different halo-agents is a technique, which improves the primed plant's endurance to tackle sodium. Salt tolerance is achieved in tolerant plants through fundamental physiological mechanisms– ion-exclusion and tissue tolerance, and salt-tolerant plants may (Na + accumulators) or may not (Na + excluders) allow sodium movement to leaves. While Na + excluders depend on ion exclusion in roots, Na + accumulators are proficient Na + managers that can compartmentalize Na + in leaves and use them beneficially as inexpensive osmoticum. Salt-sensitive plants are Na + accumulators, but their inherent tissue tolerance ability and ion-exclusion process are insufficient for tolerance. Seed priming with different halo-agents aids in 'rewiring' of the salt tolerance mechanisms of plants. The resetting of the salt tolerance mechanism is not universal for every halo-agent and might vary with halo-agents. Here, we review the physiological mechanisms that different halo-agents target to confer enhanced salt tolerance in primed plants. Calcium and potassium-specific halo-agents trigger Na + exclusion in roots, thus ensuring a low amount of Na + in leaves. In contrast, Na + -specific priming agents favour processes for Na + inclusion in leaves, improve plant tissue tolerance or vacuolar sequestration, and provide the greatest benefit to salt-sensitive and sodium accumulating plants. Overall, this review will help to understand the underlying mechanism behind plant's inherent nature towards salt management and its amelioration with different halo-agents, which helps to optimize crop stress performance.

Role of pH in fibroblast proliferation

Abstract:Secondary cultures of human diploid fibroblasts were used to study the effect of pH on cellular proliferation. In nonconfluent cultures, the growth rate at pH 7.1 was similar to that at pH 7.7 regardless of serum concentration. However, the saturation density achieved at pH 7.7 at any serum concentration was always 2–4 times that achieved at pH 7.1, although the greatest differences in saturation density were observed at the higher serum levels. The results suggest that the effect of pH on saturation density is due to two factors. One, cells at pH 7.1 seem to have a greater ability to undergo contact‐inhibition than at pH 7.7, independent of any serum functions; and, two, confluent cells in medium at pH 7.1 are somewhat less sensitive to growth stimulation by increasing serum concentration than are confluent cells raised in medium at pH 7.7.

Abstract:Soil salinity has come to be recognized as a global challenge to the sustainability of farming systems due to its adverse effects on crop quality and production, particularly in coastal regions of the world. Although maize (Zea mays L.), a C4 plant, has the capacity to withstand a medium degree of salinity, maize is vulnerable to salinity stress during its early growth phases. Therefore, in order to develop strategies to improve maize adaptability in saline settings, it is essential to increase our understanding of maize response to salt stress and tolerance mechanisms. According to reports, plants are known to be able to withstand salt stress when phytohormones are applied. Salicylic acid (SA), a widely-used plant hormone, has been shown to increase a plant's ability to withstand salt stress. In order to reduce the growth inhibition of maize caused by salt, the aim of this study was to clarify and assess the potential impact of priming and exogenous administration of salicylic acid (SA). The results revealed that salt stress decreased seed germination, seedling fresh and dry weight, leaf relative water content (RWC), and photosynthetic pigments of maize. Salicylic Acid (SA) treatment increased germination percentage (43-69%), shoot (24-56%) and root length (13-37%), dry weight of shoot and root (35-171%), RWC (34-39%), and chlorophyll contents (12-36%) of maize under salt stress. The findings highlight the need for additional research to fully understand the underlying mechanisms and explore SA's potential mitigating effects in lowering salt stress as well as its potential practical applications in agricultural practices.
Salt-tolerant bacteria enhance the growth of mung bean (Vigna radiata L.) and uptake of nutrients, and mobilize sodium ions under salt stress condition

Abstract:Salinity is one of the significant abiotic stresses that exert harmful effects on plant growth and crop production. It has been reported that the harmfulness of salinity can be mitigated by the use of salt-tolerant plant growth-promoting (PGP) bacteria. In this study, four bacteria were selected from a total of 30 cultures, based on salt-tolerant and PGP properties. The isolates were found to produce indole acetic acid (8.49-19.42 μg/ml), siderophore (36.04-61.77%), and solubilize potassium and inorganic phosphate. Identification based on 16S rRNA gene sequencing revealed that the isolates belonged to Cronobacter (two isolates) and Enterobacter (two isolates). Inoculation of PGP bacteria under 2 and 10% salinity stress showed enhanced plant growth parameters in Vigna radiata compared to both salinity and non-salinity control plants. The rate of germination (113.32-206.64%), root length (128.79-525.31%), shoot length (34.09-50.32%), fresh weight, and dry weight were 3-fold higher in bacteria-treated seeds than control plants. The estimation of chlorophyll (1-5-fold), carotenoids (1-4-fold), and proline content (3.65-14.45%) was also higher compared to control plants. Further, the bacterized seeds showed enhanced nitrogen and phosphorous uptake and mobilized sodium ions from roots to leaves. Overall the strains SS4 and SS5 performed well in both 2 and 10% salt-amended soils. These strains could be formulated as a bioinoculant to mitigate the salinity stress in salinized soils.

Abstract:Plants grow on soils that not only provide support for root anchorage but also act as a reservoir of water and nutrients important for plant growth and development. However, environmental factors, such as high salinity, hinder the uptake of nutrients and water from the soil and reduce the quality and productivity of plants. Under high salinity, plants attempt to maintain cellular homeostasis through the production of numerous stress-associated endogenous metabolites that can help mitigate the stress. Both primary and secondary metabolites can significantly contribute to survival and the maintenance of growth and development of plants on saline soils. Existing studies have suggested that seed/plant-priming with exogenous metabolites is a promising approach to increase crop tolerance to salt stress without manipulation of the genome. Recent advancements have also been made in genetic engineering of various metabolic genes involved in regulation of plant responses and protection of the cells during salinity, which have therefore resulted in many more basic and applied studies in both model and crop plants. In this review, we discuss the recent findings of metabolic reprogramming, exogenous treatments with metabolites and genetic engineering of metabolic genes for the improvement of plant salt tolerance.

Salicylic Acid Enhances Growth, Photosynthetic Performance and Antioxidant Defense Activity Under Salt Stress in Two Mungbean [Vigna radiata (L.) R. Wilczek] Variety

Abstract:Salt is regarded as a main cause for reduced yield under challenging conditions. Mungbean, a valuable protein crop, is sensitive to salt stress, leading to yield shortage. The growth hormone, salicylic acid (SA), enhances several processes necessary to confer salt tolerance and relieves poor agricultural yield. Seeds of mungbean were initially pretreated with SA (0.5 mM) for 4 h before sowing, while under a cumulative combination of SA + salt regimes: control, SA, 100 mM, SA +100 mM, 200 mM and SA +200 mM. Our study examined photosynthesis parameters such as photosynthetic pigment concentration, chlorophyll a fluorescence, protein, proline, and antioxidant enzymes in plants subjected to single and combined SA + salt stress concentrations. The result showed a greater decline in SPAD and photosynthetic quantum yield under 200 mM NaCl at 43% in Var. 145 than in Var. 155 at 32% compared to 11% in SA +100 mM and 34% in SA + 200 mM treatments in both varieties. Var. 145 was found to be more sensitive to 100 and 200 mM NaCl salt stress. In Var. 155, chlorophyll a and chlorophyll b concentrations were higher under control 52%, SA + 100 mM 49%, and SA +200 mM 42% than in Var. 145 at 51%, 38%, and 31%. Protein and proline revealed a higher content in Var. 155 in contrast to the lower activity in Var. 145. The enhanced performance of the Var. 155 exposed to SA + salt stress was followed by an increase in the activities of peroxidase (POD), CAT while the activity of MDA revealed a significant increase in Var. 145 under 100 mM 43% and 200 mM 48% NaCl treatment compared to Var. 155, which had 38% and 34%. The above results suggest that SA-treated Var. 155 confers tolerance to salt stress and is accompanied with a high osmoprotectant responses as provided by SA in Var. 155 than Var. 145. The potency of SA in providing salt tolerance capacity to plants is a future research interest to maintain sustainable yield in mungbean seedlings.

Abstract:Saline stress is one of the most critical abiotic stress factors that can lessen crops' productivity. However, emerging nanotechnology, nano-fertilizers, and developing knowledge of phytochromes can potentially mitigate the negative effects of saline stress on seed germination. Therefore, the aim of this study was to investigate the effects of seed priming either with zinc oxide nanoparticles (ZnO-NPs; 50 and 100 mg L−1) or 24-epibrassinolide (EBL; 0.2 and 0.4 μM) and their combinations on maize (Zea mays L.) grains sown in salt-stressed soil (50 and 100 mM NaCl). Saline stress treatments significantly affected all germination traits and chemical analysis of seeds as well as α-amylase activity. Compared to un-primed seeds, seed priming with ZnO-NPs or EBL and their combinations significantly increased the cumulative germination percentage, germination energy, imbibition rate, increase in grain weight, K+ content, and α-amylase activity, and significantly reduced germination time, days to 50% emergence, Na+ uptake, and Na+/K+ ratio of maize sown in salt-stressed-soil (50 or 100 mM NaCl). The combination of 100 mg ZnO-NPs L−1 + 0.2 μM EBL resulted in the highest improvements for most of the studied traits of maize seeds sown in salt-stressed soil in comparison to all other individual and combined treatments.

Abstract:Salinity stress represents a major threat to crop production by inhibiting seed germination, growth of seedlings, and final yield and, therefore, to the social and economic prosperity of developing countries. Recently, plant growth‐promoting substances have been widely used as a chemical strategy for improving plant resilience towards abiotic stresses. This study aimed to determine whether melatonin (MT) and glycine betaine (GB) alone or in combination could alleviate the salinity‐induced impacts on seed germination and growth of maize seedlings. Increasing NaCl concentration from 100 to 200 mM declined seed germination rate (4.6–37.7%), germination potential (24.5–46.7%), radical length (7.7–40.0%), plumule length (2.2–35.6%), seedling fresh (1.7–41.3%) and dry weight (23.0–56.1%) compared to control (CN) plants. However, MT and GB treatments lessened the adverse effects of 100 and 150 mM NaCl and enhanced germination comparable to control plants. In addition, results from the pot experiments show that 200 mM NaCl stress disrupted the osmotic balance and persuaded oxidative stress, presented by higher electrolyte leakage, hydrogen peroxide, superoxide radicals, and malondialdehyde compared to control plants. However, compared to the NaCl treatment, NaCl+MT+GB treatment decreased the accumulation of malondialdehyde (24.2–42.1%), hydrogen peroxide (36.2–44.0%), and superoxide radicals (20.1–50.9%) by up‐regulating the activity of superoxide dismutase (28.4–51.2%), catalase (82.2–111.5%), ascorbate peroxidase (40.3–59.2%), and peroxidase (62.2–117.9%), and by enhancing osmolytes accumulation, thereby reducing NaCl‐induced oxidative damages. Based on these findings, the application of MT+GB is an efficient chemical strategy for improving seed germination and growth of seedlings by improving the physiological and biochemical attributes of maize under 200 mM NaCl stress.

Metabolomics reveals the response of hydroprimed maize to mitigate the impact of soil salinization

Abstract:Soil salinization is a major environmental stressor hindering global crop production. Hydropriming has emerged as a promising approach to reduce salt stress and enhance crop yields on salinized land. However, a better mechanisitic understanding is required to improve salt stress tolerance. We used a biochemical and metabolomics approach to study the effect of salt stress of hydroprimed maize to identify the types and variation of differentially accumulated metabolites. Here we show that hydropriming significantly increased catalase (CAT) activity, soluble sugar and proline content, decreased superoxide dismutase (SOD) activity and peroxide (H2O2) content. Conversely, hydropriming had no significant effect on POD activity, soluble protein and MDA content under salt stress. The Metabolite analysis indicated that salt stress significantly increased the content of 1278 metabolites and decreased the content of 1044 metabolites. Ethisterone (progesterone) was the most important metabolite produced in the roots of unprimed samples in response to salt s tress. Pathway enrichment analysis indicated that flavone and flavonol biosynthesis, which relate to scavenging reactive oxygen species (ROS), was the most significant metabolic pathway related to salt stress. Hydropriming significantly increased the content of 873 metabolites and significantly decreased the content of 1313 metabolites. 5-Methyltetrahydrofolate, a methyl donor for methionine, was the most important metabolite produced in the roots of hydroprimed samples in response to salt stress. Plant growth regulator, such as melatonin, gibberellin A8, estrone, abscisic acid and brassinolide involved in both treatment. Our results not only verify the roles of key metabolites in resisting salt stress, but also further evidence that flavone and flavonol biosynthesis and plant growth regulator relate to salt tolerance.

Abstract:The process of soil salinization and the preponderance of saline water sources all over the world represent one of the most harmful abiotic stress to plant growth. This pointed to the importance of obtaining plants which are tolerant or resistant to salt, considering that projection of climate change for the coming years indicate an increase in temperature and rain scarcity. In the current study, the effect of NaCl was investigated on germinating seeds of Lathyrus sativus L., Vicia sativa L., Vigna radiata L. Wilczek and Vigna unguiculata L. Walp, by combining physiological, biochemical, biostatistical and ultrastructural analyses. Our results revealed that germination was not influenced by high NaCl concentrations, while seedling growth was affected even at low NaCl concentrations, probably due to an alteration in water uptake and in organic matter biosynthesis. Nevertheless, the synthesis of antioxidant enzymes, phenolic acids and flavonoids was registered in all species, which tended to cope with the increasing salt stress, allowing a response mechanism such as cytoplasm detoxification and cellular turgor maintenance. Besides, the ultrastructural analysis evidenced plasmolysed cells close to cells with a normal ultrastructure with no appreciable differences among the species. This research deeply investigates the mechanism of salt‐stress response focusing on species never tested before for their possible tolerance to salinity.

Salicylic Acid and Calcium Chloride Seed Priming: A Prominent Frontier in Inducing Mineral Nutrition Balance and Antioxidant System Capacity to Enhance the Tolerance of Barley Plants to Salinity

Abstract:The current investigation aims to underline the impact of salicylic acid or calcium chloride seed pre-treatments on mineral status and oxidative stress markers, namely levels of electrolyte leakage (EL) and lipid peroxidation levels, measured as thiobarbituric reactive substances (TBARS), and the activity of some antioxidant enzymes in roots and leaves of plants in two barley species grown under various salt treatments. Overall, our results revealed that salinity inhibits essential nutrient absorption such as iron, calcium, magnesium and potassium and stimulates the absorption of sodium. Also, this environmental constraint induced oxidative stress in plants in comparison with the control conditions. This state of oxidative stress is reflected by an increase in TBARS content as well as the stimulation of EL values. In addition, salinity induced disturbances in the activity of antioxidant enzymes, which were mainly dependent on the applied salt concentration and the species. In addition, Hordeum marinum maintained high antioxidant enzyme activity and low levels of oxidative stress parameters, which reinforces its salt-tolerant character. Importantly, salicylic acid or calcium chloride seed priming alleviated the mineral imbalance and the oxidative damage induced by salinity. Moreover, seed priming improves iron, calcium magnesium and potassium content and limitsthe accumulation of sodium. Also, both treatments not only decrease TBARS levels and limit EL, but they also stimulate the antioxidant enzyme activities in the leaves and roots of the stressed plants as compared with stressed plants grown from non-primed seeds. Interestingly, the beneficial effects of the mentioned treatments were more notable on Hordeum vulgare species.

Abstract:Plants are highly sensitive to various environmental stresses, which can hinder their growth and reduce yields. In this study, we investigated the potential of seed priming with salicylic acid (SA), gibberellic acid (GA3), and sodium chloride (NaCl) to mitigate the adverse effects of salinity stress in Hordeum vulgare at the germination and early seedling stages. Exposing H. vulgare seeds to salt stress reduced the final germination percentage and seedling shoot and root growth. Interestingly, all seed treatments significantly improved salt-induced responses, with GA3 being more effective in terms of germination performance, plant growth, and photosynthesis. SA priming exhibited promising effects on antioxidant defense mechanisms, proline, sugar, and ascorbic acid production. Notably, SA priming also suppressed reactive oxygen species accumulation and prevented lipid peroxidation. These findings highlight the ability of SA to manage crosstalk within the seed, coordinating many regulatory processes to support plant adaptation to salinity stress.

Abstract:Mungbean ( Vigna radiata ) sprouts are consumed globally as a healthy food with high nutritional values, having antioxidant and anticancer capacity. Under mild salinity stress, plants accumulate more secondary metabolites to alleviate oxidative stress. In this study, metabolomic and transcriptomic changes in mungbean sprouts were identified using a reference cultivar, sunhwa, to understand the regulatory mechanisms of secondary metabolites in response to salinity stress. Under salinity conditions, the contents of phenylpropanoid-derived metabolites, including catechin, chlorogenic acid, isovitexin, p -coumaric acid, syringic acid, ferulic acid, and vitexin, significantly increased. Through RNA sequencing, 728 differentially expressed genes (DEGs) were identified and 20 DEGs were detected in phenylpropanoid and flavonoid biosynthetic pathways. Among them, 11 DEGs encoding key enzymes involved in the biosynthesis of the secondary metabolites that increased after NaCl treatment were significantly upregulated, including dihydroflavonol 4-reductase (log 2 FC 1.46), caffeoyl-CoA O-methyltransferase (1.38), chalcone synthase (1.15), and chalcone isomerase (1.19). Transcription factor families, such as MYB , WRKY , and bHLH , were also identified as upregulated DEGs, which play a crucial role in stress responses in plants. Furthermore, this study showed that mild salinity stress can increase the contents of phenylpropanoids and flavonoids in mungbean sprouts through transcriptional regulation of the key enzymes involved in the biosynthetic pathways. Overall, these findings will provide valuable information for molecular breeders and scientists interested in improving the nutritional quality of sprout vegetables.

Abstract:Soil salinization poses a significant challenge to the sustainability and productivity of agriculture worldwide. This issue continues to hinder plant growth, requiring innovative solutions to alleviate salt stress. Moreover, climate change accelerates soil salinization, which may soon spread to previously unaffected agricultural areas. Therefore, the present study evaluated the potential role of different seed priming agents (hydro (H), salicylic acid (SA), proline (P), and melatonin (MEL)) on seedlings and leaf macro and micronutrients of sorghum grown under four (.27, 2.5, 5.0, and 8.0 dS m−1) soil salinity conditions. Soil salinity drastically reduced all the growth parameters of sorghum seedlings, primarily the reduction in growth traits, which was remarkable after 2.5 dS m−1 soil salinity. In addition, plant height, shoot fresh weight, and stomata were reduced by 40.8%, 74.6%, and 36.5%, respectively, at 8.0 dS m−1 compared to .27 dS m−1. SA‐ and MEL‐primed seeds mitigated the harmful effects of soil salinity by reducing Na+ accumulation in the leaves and increasing the K+/Na+ and Ca2+/Na+ ratios and photosynthetic activity under salt stress. However, the Zn2+, Mn2+, and Cu2+ contents of sorghum leaves increased with increasing soil salinity, and these nutrients also improved with seed priming by SA, MEL, and P. Considering all nutrients, MEL‐primed sorghum seeds had better macro‐ and micro‐nutrient uptake capacities than the H, SA, and P treatments under high soil salinity conditions. Finally, the present study showed that MEL‐induced improvement in salt tolerance in sorghum seedlings was related to enhanced nutritional status, photosynthetic activity, and biomass production in salinized areas.

Abstract:Salinity is a global challenge to agriculture production. Cicer arientinum L. grows under wide range of climatic conditions and is highly sensitive to salt stress. Salt stress causes osmotic, ionic and secondary stress, impairing various metabolic processes in plants and eventually results in a decline in the plant yield. The aim of the present study was to study the effect of salinity on morphological, anatomical and ionic responses of chickpea seedlings of the genotype PBG-7. Three NaCl concentrations (40 mM, 60 mM and 80 mM) with three replicates were investigated by conducting the experiments in petriplates and growth pouches. For the first 7 days, experiments were carried out in petriplates. Thereafter, the seedlings were transferred to the growth pouches of respective treatment and were maintained subsequently for 21 days. The results collectively suggest that salt stress negatively affected the xylem and phloem width along with the total biomass production. Further, the results indicated that salinity leads to an increase in the sodium ion content in the plants and was accompanied by a decline in K+/Na+ as well as Ca2+/Na+ ratio. Sodium ion content was found to be negatively correlated with all the measured parameters.

Abstract:Background: Increasing soil salinization has a detrimental effect on agricultural productivity.Therefore, strategies are needed to induce salinity-tolerance in crop species for sustainable foodproduction. γ-aminobutyric acid (GABA) plays a key role in regulating plant salinity stresstolerance. However, it remains largely unknown how mungbean plants (Vigna radiata L.) respondto exogenous GABA under salinity stress. Methods: Thus, we evaluated the effect of exogenous GABA (1.5 mM) on the growth and physiobiochemicalresponse mechanism of mungbean plants to saline stress (0-, 50-, and 100 mM [NaCland Na2SO4, at a 1:1 molar ratio]). Results: Increased saline stress adversely affected mungbean plants' growth and metabolism. Forinstance, leaf-stem-root biomass (34- and 56%, 31- and 53%, and 27- and 56% under 50- and 100mM, respectively]) and chlorophyll concentrations declined. The carotenoid level increased (10%)at 50 mM and remained unaffected at 100 mM. Hydrogen peroxide (H2O2), malondialdehyde(MDA), osmolytes (soluble sugars, soluble proteins, proline), total phenolic content, andenzymatic activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), peroxidase(POD), glutathione reductase (GTR), and polyphenol oxidation (PPO) were significantlyincreased. In leaves, salinity caused a significant increase in Na+ concentration but a decrease inK+ concentration, resulting in a low K+/Na+ concentration (51- and 71% under 50- and 100- mMstress). Additionally, nitrogen concentration and the activities of nitrate reductase (NR) andglutamine synthetase (GS) decreased significantly. The reduction in glutamate synthase (GOGAT)activity was only significant (65%) at 100 mM stress. Exogenous GABA decreased Na+, H2O2,and MDA concentrations but enhanced photosynthetic pigments, K+ and K+/Na+ ratio, Nmetabolism, osmolytes, and enzymatic antioxidant activities, thus reducing salinity-associatedstress damages, resulting in improved growth and biomass. Conclusion: Exogenous GABA may have improved the salinity tolerance of mungbean plants by maintaining their morpho-physiological responses and reducing the accumulation of harmfulsubstances under salinity. Future molecular studies can contribute to a better understanding of themolecular mechanisms by which GABA regulates mungbean salinity tolerance.
Transcriptome and Metabolome Analyses of Salt Stress Response in Cotton (Gossypium hirsutum) Seed Pretreated with NaCl

Abstract:Abiotic stresses adversely influence crop productivity and salt stress is one limiting factor. Plants need to evolve their defense mechanisms to survive in such fluctuating scenarios at either the biochemical, physiological, or molecular level. The analytical/critical investigations of cotton (Gossypium hirsutum) plants that involve looking into transcriptomic and metabolomic profiles could give a comprehensive picture of the response of the cotton plant to salt stress. This study was conducted on pre-treated cotton seeds by soaking them in a 3% sodium chloride (NaCl) solution at room temperature for 0.5, 1, and 1.5 h. In total, 3738 and 285 differentially expressed genes (DEGs) and metabolites, respectively, were discovered. The prominent DEGs included AtCCC1, EP1, NHE, AtpOMT, GAST1, CLC-c, ARP, AtKIN14, AtC3H2, COP9, AtHK-2, and EID1 to code for the regulation of seed growth, abscisic acid receptor PYR/PYL, a cellular response regarding stress tolerance (especially to salt) and germination, jasmonic acid, salicylic acid, and auxin-activated signaling pathways. A more significant amount of transcription factors, including the ethylene-responsive TFs ERF (205), bHLH (252), ZF-domains (167), bHLH (101), MYB (92), NAC (83), GATA (43), auxin-responsive proteins (30), MADs-box (23), bZIP (27), and HHO (13) were discovered in samples of NaCl-pretreated cotton seedlings under different treatments. The functional annotations of DEGs exposed their important roles in regulating different phytohormones and signal-transduction-mediated pathways in salt-treated seeds. The metabolites analysis revealed differential accumulation of flavonols, phenolic acid, amino acids, and derivatives in seedling samples treated for 0.5 h with NaCl. The conjoint analysis that showed most of the DEGs were associated with the production and regulation of glucose-1-phosphate, uridine 5′-diphospho-D-glucose, and 2-deoxyribose 1-phosphate under salt stress conditions. These results indicated positive effects of NaCl 0.5 h treatments on seedlings’ germination and growth, seemingly by activating specific growth-promoting enzymes and metabolites to alleviate adverse effects of salt stress. Hence, seed pre-treatment with NaCl can be beneficial in future cotton management and breeding programs to enhance growth and development under salt stress.

Abstract:The present study investigates a possible mechanism of 24-Epibrassinolide (EBL) mediated alleviation of salinity stress in Brassica juncea L. seedlings. EBL (10 –6 M and 10 –8 M) pretreated seedlings were grown up to ten-days either in alone and/or combination with defined levels of salinity treatments (100 mM and 200 mM NaCl) under laboratory growth conditions. Results showed that salinity stress significantly ( P ≤ 0.05 ) increased the Na + accumulation in a dose-dependent manner, which aggravated the nutrient(s) homeostasis and pigment contents by inducing oxidative damage through elevated hydrogen peroxide, malondialdehyde, and methyl glyoxal production. EBL mediated alleviation of salinity stress was achieved through decreased nuclear damage and cell death, coupled with increased membrane stability index and nutrients accumulation. The positive roles of EBL were also reflected through maintenance of AsA/DHA and GSH/GSSG redox ratios' that were maximally enhanced by 78% and 28%, respectively under 200 mM NaCl. EBL mediated up-regulation of γ-glutamyl cysteine synthetase activity increased the glutathione pool, which boosted the ascorbate–glutathione cycle and methyl glyoxal detoxification system. The nitrate reductase activity, endogenous nitric oxide and S-nitrosothiols pools were maximally increased by 45, 32, and 33% respectively in 200 mM NaCl treatment, over the salinity alone counterpart. Taken together, our findings suggested that EBL pretreatment alleviates salinity stress by regulating nitric oxide homeostasis, osmolytes synthesis, antioxidant defense systems, and increasing the nutrients and pigments contents in mustard seedlings under salinity stress. Therefore, seed-priming with EBL can be considered as an effective approach for improving the plant productivity under salinity stress.
