Abstract:The objective of this study was to evaluate the production (P) (g of fresh mushrooms /bag) and biological efficiency (BE) (g of fresh mushrooms per 100 g of dry substrate) of 17 fungal strains, namely Pleurotus ostreatus Po-IAP, Po-P38, Po-P14, Po-IE202, Po-Sfco, Po-JP, Po-Psma, and Po-POS, Pleurotus djamour Pd-PRO and Pd-UTMR, Pleurotus eryngii Pe-MB and Pe-PQ, Lentinula edodes L15, L9, L5, and LC, and Hericium erinaceus Heri, produced in corn stover (CS) and to assess the content of crude protein (CP), lignin (L), cellulose, hemicellulose, acid detergent fiber (ADF), and neutral detergent fiber (NDF) and in vitro digestibility of dry matter (IVDMD) of the residual substrate of CS, the so called spent mushroom substrate (SMS), in comparison to the non-inoculated substrate (C). The variables were analyzed as a completely randomized block design using R 4.0.3 software. Means were compared using Tukey's procedure. The Pleurotus spp. strains, compared to Lentinula spp. and Hericium spp., presented better BE and P. In comparison to C, the SMS increased the CP content (p < 0.05) from 10.8% (Po-JP) to 70.3% (LC), while NDF decreased (p < 0.05) from 11.5% (Pd-Pro) to 33.5% (L15) and IVDMD increased (p < 0.05) from 16.2% (Heri) to 47.7% (Pd-UTMR). In conclusion, of the 17 strains evaluated, the 3 strains of Lentinula edodes (L5, L15, and L9), one strain of Pleurotus djamour (Pd-UTMR), and one strain of Pleurotus ostreatus (Po-IAP) generated a SMS that, due to its nutritional improvement and increase in IVDMD, could be used as feed for ruminants. Our results also showed that corn stover is a suitable substrate to produce Pleurotus spp. fruiting bodies, with strain Po-IAP as the best yielding.
Development of a consortium-based microbial agent beneficial to composting of distilled grain waste for Pleurotus ostreatus cultivation

Abstract:Abstract Background Pleurotus ostreatus is an edible mushroom popularly cultivated worldwide. Distilled grain waste (DGW) is a potential substrate for P. ostreatus cultivation. However, components in DGW restrict P. ostreatus mycelial growth. Therefore, a cost-effective approach to facilitate rapid P. ostreatus colonization on DGW substrate will benefit P. ostreatus cultivation and DGW recycling. Results Five dominant indigenous bacteria, Sphingobacterium sp. X1, Ureibacillus sp. X2, Pseudoxanthomonas sp. X3, Geobacillus sp. X4, and Aeribacillus sp. X5, were isolated from DGW and selected to develop a consortium-based microbial agent to compost DGW for P. ostreatus cultivation. Microbial agent inoculation led to faster carbohydrate metabolism, a higher temperature (73.2 vs. 71.2 °C), a longer thermophilic phase (5 vs. 3 days), and significant dynamic changes in microbial community composition and diversity in composts than those of the controls. Metagenomic analysis showed the enhanced microbial metabolisms, such as xenobiotic biodegradation and metabolism and terpenoid and polyketide metabolism, during the mesophilic phase after microbial agent inoculation, which may facilitate the fungal colonization on the substrate. In accordance with the bioinformatic analysis, a faster colonization of P. ostreatus was observed in the composts with microbial inoculation than in control after composting for 48 h, as indicated from substantially higher fungal ergosterol content, faster lignocellulose degradation, and higher lignocellulase activities in the former than in the latter. The final mushroom yield shared no significant difference between composts with microbial inoculation and control, with 0.67 ± 0.05 and 0.60 ± 0.04 kg fresh mushroom/kg DGW, respectively ( p > 0.05). Conclusion The consortium-based microbial agent comprised indigenous microorganisms showing application potential in composting DGW for providing substrate for P. ostreatus cultivation and will provide an alternative to facilitate DGW recycling.

Abstract:The cultivation of edible mushrooms from fruit and vegetable residues presents itself as a promising solution for the sustainable utilization of these wastes and the production of nutritious food. Fruit and vegetable residues include crop residues, leaves, branches and unsold products, which contain a wide range of nutrients and bioactive compounds. These are used as substrate for the cultivation of edible mushrooms. Edible mushrooms, such as mushrooms, shiitake and pleurotus, are appreciated worldwide for their taste, texture and nutritional properties. They contain protein, fiber, vitamins, minerals and bioactive compounds with antioxidant, anti-cancer and anti-diabetic properties. In addition, the global market for edible mushrooms is experiencing significant growth due to their wide range of applications in the food, pharmaceutical and cosmetic industries. The method of cultivating edible mushrooms from fruit and vegetable waste involves the selection of suitable waste, its preparation by sterilization, addition of nutritional enrichment agents and inoculation of the desired mushroom. After a period of incubation and proper care, the mushrooms can be harvested and used in various food preparations. The cultivation of edible mushrooms by solid fermentation has great advantages, as it is the most efficient way of converting vegetable waste into high-value human food.
Effects of mixed agro-residues (corn crop waste) on lignin-degrading enzyme activities, growth, and quality of Lentinula edodes

Abstract:Using agro-residues in bioreactors to produce mushrooms is an important component of eco-agriculture. Here, we tested the cultivation of Lentinula edodes with corn cob (CC) and corn straw (CS), and then evaluated the resulting enzyme activities, agronomic traits, textures and nutrient compositions of mushrooms. Laccase (T1 formula, 3.26 g U-1) and carboxymethyl cellulase (T3 formula, 1.01 g U-1) activities were the highest at the time for a complete substrate colonization stage (TCSC), while acidic xylanase activity was the highest (CK formula, 4.05 g U-1) in the mushroom block to color-turned (TMBCT) stage. The biological efficiency of growth on the T6 formula was 8.82% higher than growth on the CK formula, wherein the low C/N ratio of the substrate had an obvious negative effect on yield while the mass ratio of pileus (MRP) of fruiting bodies did not change with mixed substrates. No significant differences were observed in mineral composition for CK formulas, but corn crop waste (CCW) formulas exhibited more optimal nutritional contents. A formula containing more corn cob and sawdust (SD) (sum of at least 70%) as the substrate can produce fruiting bodies with good hardness. These results indicate that the use of corn cobs as the main ingredient, mixed with sawdust and corn straw to grow L. edodes provides a more efficient use of agro-residues for growth. Thus, mixed agro-residue formulas have exceptional advantages in texture, nutrition of fruiting bodies, and yields.
From waste to protein: a new strategy of converting composted distilled grain wastes into animal feed

Abstract:Distilled grain waste (DGW) is rich in nutrients and can be a potential resource as animal feed. However, DGW contains as much as 14% lignin, dramatically reducing the feeding value. White-rot fungi such as Pleurotus ostreatus could preferentially degrade lignin with high efficiency. However, lignin derivatives generated during alcohol distillation inhibit P. ostreatus growth. Thus, finding a new strategy to adjust the DGW properties to facilitate P. ostreatus growth is critical for animal feed preparation and DGW recycling. In this study, three dominant indigenous bacteria, including Sphingobacterium thermophilum X1, Pseudoxanthomonas byssovorax X3, and Bacillus velezensis 15F were chosen to generate single and compound microbial inoculums for DGW composting to prepare substrates for P. ostreatus growth. Compared with non-inoculated control or single microbial inoculation, all composite inoculations, especially the three-microbial compound, led to faster organic metabolism, shorter composting process, and improved physicochemical properties of DGW. P. ostreatus growth assays showed the fastest mycelial colonization (20.43 μg·g −1 ergosterol) and extension (9 mm/d), the highest ligninolytic enzyme activities (Lac, 152.68 U·g −1 ; Lip, 15.56 U·g −1 ; MnP, 0.34 U·g −1 ; Xylanase, 10.98 U·g −1 ; FPase, 0.71 U·g −1 ), and the highest lignin degradation ratio (30.77%) in the DGW sample after 12 h of composting with the three-microbial compound inoculation when compared to other groups. This sample was relatively abundant in bacteria playing critical roles in amino acid, carbohydrate, energy metabolism, and xenobiotic biodegradation, as suggested by metagenomic analysis. The feed value analysis revealed that P. ostreatus mycelia full colonization in composted DGW led to high fiber content retention and decreased lignin content (final ratio of 5% lignin) but elevated protein concentrations (about 130 g·kg −1 DM). An additional daily weight gain of 0.4 kg/d was shown in cattle feeding experiments by replacing 60% of regular feed with it. These findings demonstrate that compound inoculant consisting of three indigenous microorganisms is efficient to compost DGW and facilitate P. ostreatus growth. P. ostreatus decreased the lignin content of composted DGW during its mycelial growth, improving the quality of DGW for feeding cattle.

Abstract:A large amount of agro-industrial residues are produced from the planting, production and processing of traditional Chinese herbs. As a tonic, edible, and economical herb, Codonopsis pilosula root has been extensively developed into medicine and functional food. However, thousands of tons of aerial parts (stems, leaves, flowers and fruits) have been directly discarded after harvest each year. To utilise agro-wastes, Pleurotus ostreatus was cultivated on a basal substrate supplemented with C. pilosula stems and leaves (CSL). Physicochemical analyses revealed that the basal substrate mixed with CSL was more abundant in cellulose, hemicellulose, and most of micronutrients such as K, Ca, Mg, S, Fe, Zn and Mo. After the first flush, the fruit bodies in CSL group exhibited a higher fresh weight, a wider average pileus diameter and a lower moisture level. Nutrition analyses presented a higher protein content and a lower fat content in mushrooms from CSL group compared with control group. Interestingly, 14 amino acids (glutamine, arginine, valine, leucine, and etc.) and 3 micronutrients (Se, Fe and Zn) were increased after CSL addition to the substrate. Based on untargeted metabolomics, a total of 710 metabolites were annotated. Compared with control group, there were 142 and 117 metabolites significantly increased and decreased in the CSL group. Most of them were grouped into classes of amino acids and peptids, fatty acids, carbohydrates, terpenoids, and etc. Moreover, an abundance of phytometabolites from Codonopsis were detected in P. ostreatus from CSL group, including polyacetylenes or polyenes, flavonoids, alkaloids, terpenoids, organic acids, and etc. UPLC-MS/MS results demonstrated that lobetyolin content in the CSL group samples was 0.0058%. In summary, the aerial parts of C. pilosula processed for use in the production of edible mushroom is an emerging strategy to converting agricultural waste into functional foods.

Abstract:The mushroom agroindustry generates a huge amount of waste from mushroom production (WMP). The composition of WMP is not standardized but differs mainly in terms of organic matter (OM) content and OM biodegradability. This makes WMP management, including anaerobic digestion (AD), a significant challenge. A potential solution could be co-digestion of WMP with municipal sewage sludge (SS), especially SS generated in small rural wastewater treatment plants (WWTPs). Therefore, this study investigated mesophilic methane production (MP) from WMP, SS, and mixtures of SS and WMP at ratios of 70:30, 50:50, and 30:70 (w/w OM). Even though the maximum cumulative MP from WMP was relatively low (approx. 60 NL/kg OM), co-digesting WMP with SS increased both MP and the methane content of the biogas: with 30%, 50%, and 70% shares of SS, MP increased almost 2, 2.5, and 3.3 times, and the methane content increased to 61%, 62%, and 64%, respectively. As the SS content was increased, the kinetic coefficients of MP and OM removal decreased (from 0.211 to 0.146 d−1 and from 0.215 to 0.152 d−1), whereas the initial rate of MP and of OM removal increased (from 12.5 to 36.8 NL/(kg OM·d) and from 0.51 kg OM/(m3·d) to 0.59 kg OM/(m3·d), respectively). The effectiveness of OM removal (EOMrem) was lowest with WMP only, at 46.6%. When the SS content of the mixtures was increased to 30%, 50%, and 70%, EOMrem also increased to 55.3%, 60.1%, and 64.9%, respectively. The relationship between maximal MP and the overall OM removed was such that both increased simultaneously. The higher values of EOMrem and, consequently, the lower final contents of OM with more effective MP indicate that the organics were degraded more efficiently. These results suggest that co-digestion may be a profitable solution for simultaneously utilizing both of these waste products, increasing the efficiency of biogas production to such an extent that it would be profitable to conduct AD on mushroom farms. This is a flexible approach that allows varying proportions of WMP and SS to be used, depending on the availability of both substrates and the energy needs of the mushroom farm. However, it should be borne in mind that a higher share of WMP results in lower gas productivity.

Enhanced mycelium biomass and polysaccharide production in genetically modified Pleurotus ostreatus using agricultural wastes

Abstract:Edible fungi, healthier for humans and sustainable for the planet, attract unprecedented attention. In the study, the genetically modified Pleurotus ostreatus overexpression phosphoglucomutase (PGM) was constructed. P. ostreatus overexpression PGM (Po::PGM) had 4.96-folds higher expression level of PGM. Po::PGM grew thicker mycelium and more mycelium branches. Additional Ca2+ can inhibit mycelium growth, and cyclic adenosine monophosphate completely inhibited their growth of Po::PGM. Secondly, Overexpression of PGM made P. ostreatus become more sensitive to cell wall disruptors, and caused 12.75 % reduction of β-1, 3-glucan and 40.53 % increase of chitin in cell wall. In submerged fermentation, the mycelia biomass yield and endopolysaccharide (IPS) production of Po::PGM in basic PDB can reach 11.18 g/l and 2.55 g/l, increasing by 20.86 % and 28.79 %, respectively. Whereas exopolysaccharide (EPS) reduced by 3.28 %. After replacing potato and glucose in PDB by wheat bran, mycelia biomass and EPS production of Po::PGM were all improved. The additional lactose in wheat bran did not only furtherly enhance mycelia biomass yield of Po::PGM to 27.78 g/l by 199.03 %, but IPS production also increased by 277.99 % to 6.07 g/l. The results provided us key ideas and important research directions that at least manipulating the PGM gene could obtain high-efficient use of agricultural wastes producing more fungus-based foods.
Characterization and nutritional valorization of agricultural waste corncobs from Italian maize landraces through the growth of medicinal mushrooms

Abstract:The research investigates the potential use of maize cobs (or corncobs) from five genotypes, including the B73 inbred line and four locally cultivated landraces from Northern Italy, as substrate for implementing Solid State fermentation processes with four Medicinal Mushrooms (MMs). The corncobs were characterized based on their proximate composition, lignin, phenolics content (both free and bound), and total antioxidant capacity. Among the MMs tested, Pleurotus ostreatus and Ganoderma annularis demonstrated the most robust performance. Their growth was parametrized using Image Analysis technique, and chemical composition of culture samples was characterized compared to that of corncobs alone. In all culture samples, the growth of MMs led to a significant reduction (averaging 40%) in the total phenolics contents compared to that measured in corncobs alone. However, the high content of free phenolics in the cobs negatively impacted the growth of P. ostreatus. The final MM-corncob matrix exhibited reduced levels of free sugars and starch (≤ 2.2% DW, as a sum) and increased levels of proteins (up to 5.9% DW) and soluble dietary fiber (up to 5.0% DW), with a notable trend toward higher levels of β-glucan compared to corncobs alone. This research paves the way for the use of this matrix as an active ingredient to enhance the nutritional value of food preparations.
Label-free comparative proteomic analysis of Pleurotus eryngii grown on sawdust, bagasse, and peanut shell substrates

Abstract:The white rot fungi Pleurotus eryngii are environmental microorganisms that can effectively break down lignocellulosic biomass. However, understanding of the mechanisms by which P. eryngii is effective in degrading lignocellulose is still limited. This work aimed to examine the extracellular secretory proteins implicated in the breakdown of lignocellulose in P. eryngii and identify degradation tactics across various cultivation substrates. Thus, a comparative analysis of the secretory proteins based on Nanoliquid chromatography combined with tandem mass spectrometry was conducted among P. eryngii cultivated on sawdusts, bagasse, peanut shells, and glucose. In total, 647, 616, 604, and 511 proteins were identified from the four samples, respectively. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis of protein expression differences identified pathways (hydrolytic enzymes, catalytic activity, metabolic processes, cellular processes, and response to stimuli) significantly enriched in proteins associated with lignocellulose degradation in P. eryngii. An integrated analysis of proteome data revealed specifically or differentially expressed genes secreted by P. eryngii in different cultivation substrates. The most prevalent carbohydrate-active enzymes involved in lignocellulose degradation in the secretome of the four samples were laccase (Lac), manganese peroxidase (MnP), aryl alcohol oxidase (AaO), and copper radical oxidase (CRO). Among them, Lac 2 mainly involved in the lignin degradation of sawdust peanut shells, and bagasse by P. eryngii, and Mnp 3 was mainly involved in the degradation of peanut shells. AaO and Lac 4 were mainly involved in glucose substrate defense and oxidative stress. It was found that exogenous addition of sawdust and peanut shells significantly increased lignolytic enzyme abundance. These findings provide insight and guidance for improving agricultural waste resource recovery. In this study, the secretomes of P. eryngii grown on four different carbon sources were compared. The findings revealed the extracellular enzymes implicated in the degradation of lignocellulose, offering avenues for further investigation into the biotransformation mechanisms of P. eryngii biomass and the potential utilization of agricultural wastes. SIGNIFICANCE: The cost of the substrate for mushroom cultivation has increased as the production of edible fungus has risen year after year. Therefore, the use of these locally available lignocellulosic wastes as substrates offers a cost-cutting option. Further, the overuse of wood for the cultivation of edible mushrooms is also detrimental to the conservation of forest resources or the ecological environment. Consequently, the use of other agricultural wastes as an alternative to sawdust or other woody substrates is a viable approach for cultivating P. eryngii. The distribution of extracellular lignocellulosic degrading enzymes, inferred in the present study could help improve the cultivation efficiency of P. eryngii vis-à-vis managing agricultural waste.
Degradation of edible mushroom waste by Hermetia illucens L. and consequent adaptation of its gut microbiota

Abstract:The edible fungus industry is one of the pillar industries in the Yunnan–Guizhou Plateau, China. The expansion of the planting scale has led to the release of various mushroom residues, such as mushroom feet, and other wastes, which are not treated adequately, resulting in environmental pollution. This study investigated the ability of black soldier fly ( Hermetia illucens L .) larvae (BSFL) to degrade mushroom waste. Moreover, this study analyzed changes in the intestinal bacterial community and gene expression of BSFL after feeding on mushroom waste. Under identical feeding conditions, the remaining amount of mushroom waste in Pleurotus ostreatus treatment group was reduced by 18.66%, whereas that in Flammulina velutipes treatment group was increased by 31.08%. Regarding gut microbial diversity, compared with wheat bran-treated control group, Dysgonomonas , Providencia , Enterococcus , Pseudochrobactrum , Actinomyces , Morganella , Ochrobactrum , Raoultella , and Ignatzschineria were the most abundant bacteria in the midgut of BSFL in F. velutipes treatment group. Furthermore, Dysgonomonas , Campylobacter , Providencia , Ignatzschineria , Actinomyces , Enterococcus , Morganella , Raoultella , and Pseudochrobactrum were the most abundant bacteria in the midgut of BSFL in P. ostreatus treatment group. Compared with wheat bran-treated control group, 501 upregulated and 285 downregulated genes were identified in F. velutipes treatment group, whereas 211 upregulated and 43 downregulated genes were identified in P. ostreatus treatment group. Using Kyoto Encyclopedia of Genes and Genomes and Gene Ontology enrichment analyses, we identified 14 differentially expressed genes (DEGs) related to amino sugar and nucleotide sugar metabolism in F. velutipes treatment group, followed by 12 DEGs related to protein digestion and absorption. Moreover, in P. ostreatus treatment group, two DEGs were detected for fructose and mannose metabolism, and two were noted for fatty acid metabolism. These results indicate that feeding on edible mushroom waste can alter the intestinal microbial community structure of BSFL; moreover, the larval intestine can generate a corresponding feedback. These changes contribute to the degradation of edible mushroom waste by BSFL and provide a reference for treating edible mushroom waste using BSFL.

Abstract:The edible mushroom industry produces massive amounts of spent mushroom substrate (SMS). Thus, there is an urgent need for high-value utilization technology to process the SMS, especially SMSs originating from woodchips. Protaetia brevitarsis larvae (PBL) can feed on various types of organic matter and can produce organic fertilizer and insect protein. In this study, we investigated the potential of PBL to utilize and convert SMSs from Auricularia auricula (SMS-AA) and Lentinula edodes (SMS-LE) cultivation. The results showed that the PBL were able to feed on SMS-AA and SMS-LE and form nutrient-enriched organic fertilizer with a low phytotoxicity and high humic acid content. Further analysis of the organic carbon dynamics suggested that PBL can efficiently digest and utilize lignin. Abstract:Ligninolytic enzymes from white-rot fungi, such as laccase (Lac) and Mn-peroxidase (MnP), are able to degrade aflatoxin B1 (AFB1), the most harmful among the known mycotoxins. The high cost of purification of these enzymes has limited their implementation into practical technologies. Every year, tons of spent mushroom substrate (SMS) are produced as a by-product of edible mushroom cultivation, such as Pleurotus spp., and disposed at a cost for farmers. SMS may still bea source of ligninolytic enzymes useful for AFB1 degradation. The in vitro AFB1-degradative activity of an SMS crude extract (SMSE) was investigated. Results show that: (1) in SMSE, high Lac activity (4 U g-1dry matter) and low MnP activity (0.4 U g-1dry matter) were present; (2) after 1 d of incubation at 25 °C, the SMSE was able to degrade more than 50% of AFB1, whereas after 3 and 7 d of incubation, the percentage of degradation reached the values of 75% and 90%, respectively; (3) with increasing pH values, the degradation percentage increased, reaching 90% after 3 d at pH 8. Based on these results, SMS proved to be a suitable source of AFB1 degrading enzymes and the use of SMSE to detoxify AFB1 contaminated commodities appears conceivable.
Optimization of White-Rot Fungi Mycelial Culture Components for Bioremediation of Pharmaceutical-Derived Pollutants

Abstract:White-rot fungi can degrade a wide spectrum of environmental pollutants, including pharmaceuticals, which are not efficiently removed from wastewater by conventional methods, e.g., the activated sludge method. However, the treatment of wastewater with the use of fungal cultures (mycoremediation) also has significant limitations: among others, the need to use appropriate, often-expensive culture media. We aimed to screen 18 media ingredients, including seven agrifood byproducts for Armillaria mellea, Phanerochaete chrysosporium and Pleurotus ostreatus in submerged cultures to select the low-cost medium optimal for biomass production and laccase activity. We screened nine mathematic models to describe the relation of fungal growth and the amount of the selected byproduct in media. Finally, we tested the ability of the strain with the highest mycelial growth and enzyme-producing ability in the selected medium to degrade eight drug contaminants. Three media variants composed of byproducts provided both efficient growth and laccase production: corn steep liquor + poplar, dried distillers grains with solubles + poplar and corn steep liquor 50%. Among the investigated growth models, the Han–Levenspiel equation described well the specific growth rate in function of the nominal substrate concentration in one-component media. Pleurotus ostreatus, the fungus with the highest ligninolytic enzyme activity, cultured in medium composed of corn steep liquor, removed six of eight drug contaminants with a removal degree of 20–90% in 48 h. The obtained data on the optimal culture media consisting of insoluble components provide initial data for upscaling the process and designing an appropriate type of bioreactor for the process of removing drug contaminants from water.

Abstract:A key aspect of sustainable bioeconomy is the recirculation of renewable, agricultural waste streams as substrates for microbial production of high-value compounds. One approach is the bioconversion of corn stover, an abundant maize crop byproduct, using the fungal maize pathogen Ustilago maydis. U. maydis is already used as a unicellular biocatalyst in the production of several industrially-relevant compounds using plant biomass hydrolysates. In this study, we demonstrate that U. maydis can grow using untreated corn stover as its sole carbon source. We developed a small-scale bioreactor platform to investigate U. maydis processing of corn stover, combining online monitoring of fungal growth and metabolic activity profiles with biochemical analyses of the pre- and post-fermentation residues. Our results reveal that U. maydis primarily utilizes soluble sugars i.e., glucose, sucrose and fructose present in corn stover, with only limited exploitation of the abundant lignocellulosic carbohydrates. Thus, we further explored the biotechnological potential of enhancing U. maydis ́ lignocellulosic utilization. Additive performance improvements of up to 120 % were achieved when using a maize mutant with increased biomass digestibility, co-fermentation with a commercial cellulolytic enzyme cocktail, and exploiting engineered fungal strains expressing diverse lignocellulose-degrading enzymes. This work represents a key step towards scaling up the production of sustainable compounds from corn stover using U. maydis and provides a tool for the detailed monitoring of the fungal processing of plant biomass substrates.

Abstract:Pollution made cancer a major disease threatening people's health. In the search for new cancer drug treatments, natural products perform an essential role. Escin, a mixture of triterpene saponins extracted from Aesculus Wilsonii Rehd., was used to analyze the cell growth inhibitory effects and the induction of apoptosis in human cervical carcinoma Hela cancer cell line in vitro. The results of MTT cell viability assay showed that escin could induce significant concentration-dependent and time-dependent inhibition of cell growth, and the cell cycle was arrested at G 1 /S phase in the escin-treated cells. Apoptosis were detected and assessed by Acridine Orange/Ethidium Bromide Staining and Flow Cytometry. The antitumor efficacy of escin on mice cervical carcinoma U_(14) in vivo was investigated, at dose of 2.8 mg/kg escin had rather high inhibition ratio(43.5%) on U_(14) tumor growth.

Abstract:Aflatoxin B1 (AFB1) is the most harmful mycotoxin that occurs as natural contaminant of agricultural commodities, particularly maize. Practical solutions for detoxification of contaminated staples and reduction of agricultural wastes are scarce. We investigated the capability of the white-rot and edible fungus Plerotus eryngii (king oyster mushroom) to degrade AFB1 both in vitro and in a laboratory-scale mushroom cultivation, using a substrate similar to that routinely used in mushroom farms. In malt extract broth, degradation of AFB1 (500 ng/mL) by nine isolates of P. eryngii ranged from 81 to 99% after 10 days growth, and reached 100% for all isolates after 30 days. The growth of P. eryngii on solid medium (malt extract-agar, MEA) was significantly reduced at concentrations of AFB1 500 ng/mL or higher. However, the addition of 5% wheat straw to the culture medium increased the tolerance of P. eryngii to AFB1 and no inhibition was observed at a AFB1 content of 500 ng/mL; degradation of AFB1 in MEA supplemented with 5% wheat straw and 2.5% (w/v) maize flour was 71-94% after 30 days of growth. Further, AFB1 degradation by P. eryngii strain ITEM 13681 was tested in a laboratory-scale mushroom cultivation. The mushroom growth medium contained 25% (w/w) of maize spiked with AFB1 to the final content of 128 μg/kg. Pleurotus eryngii degraded up to 86% of the AFB1 in 28 days, with no significant reduction of either biological efficiency or mushroom yield. Neither the biomass produced on the mushroom substrate nor the mature basidiocarps contained detectable levels of AFB1 or its metabolite aflatoxicol, thus ruling out the translocation of these toxins through the fungal thallus. These findings make a contribution towards the development of a novel technology for remediation of AFB1- contaminated corn through the exploitation of the degradative capability of P. eryngii and its bioconversion into high nutritional value material intended for feed production.
Reticuloendothelial Cell Function in a‐Methyldopa‐Induced Hemolytic Anemia

Abstract:Abstract. 2 patients having α‐methyldopa‐induced hemolytic anemia were followed sequentially using an in vitro assay of autologous monocyte‐macrophage activity to determine if their reticuloendothelial system (RES) function was abnormal and thus could be related to the mechanism of lysis. RES function was evaluated while the patients were actively hemolyzing and during remission, following discontinuance of the drug. The results indicated that RES activity is normal in patients having hemolytic anemia due to a‐methyldopa administration. Also, following cessation of drug therapy, the patients' IgG‐coated red cells interacted significantly for a prolonged period (4–5 months) with autologous or normal allogeneic monocyte‐macrophages. This was associated with a concurrent reti‐culocytosis and indicates a persistent low‐level hemolytic phase throughout this period, even though hemoglobin and hematocrit values remained within the normal ranges. Although levels of IgG sensitizing the patients' red cells were essentially constant during the hemolytic phase and when the patients were in complete remission, significant monocyte‐macrophage activity was only evident during the hemolytic period. In an attempt to explain this phenomenon, it is postulated that hemolysis in patients receiving α‐methyldopa is related to the interaction of drug with red cell membrane proteins which results in a variably expressed ‘altered’ antigen which is recognized by ‘autoantibody’. The proper expression of the Fc portion of the immunoglobin molecule to result in specific recognition by receptors on monocyte‐macrophages depends upon the extent of the antigen alteration by α‐methyldopa. If the drug does not result in appropriate antigen alteration, then, although ‘autoantibody’ may still bind to the red blood cell, its Fc region is not readily recognized by monocyte‐macrophages and little or no erythrophagocytosis occurs.

Abstract:The white button mushroom Agaricus bisporus is economically the most important commercially produced edible fungus. It is grown on carbon- and nitrogen-rich substrates, such as composted cereal straw and animal manure. The commercial mushroom production process is usually performed in buildings or tunnels under highly controlled environmental conditions. In nature, the basidiomycete A. bisporus has a significant impact on the carbon cycle in terrestrial ecosystems as a saprotrophic decayer of leaf litter. In this mini-review, the fate of the compost plant cell wall structures, xylan, cellulose and lignin, is discussed. A comparison is made from the structural changes observed to the occurrence and function of enzymes for lignocellulose degradation present, with a special focus on the extracellular enzymes produced by A. bisporus. In addition, recent advancements in whole genome level molecular studies in various growth stages of A. bisporus in compost are reviewed.
