In this study, noninvasive magnetoencephalography (MEG) recordings are obtained from human subjects actively listening to spoken narratives, in both simple and cocktail party-like auditory scenes. By modeling how acoustic features of speech are encoded in ongoing MEG activity as a spectrotemporal response function, we demonstrate that the slow temporal modulations of speech in a broad spectral region are represented bilaterally in auditory cortex by a phase-locked temporal code. For speech presented monaurally to either ear, this phase-locked response is always more faithful in the right hemisphere, but with a shorter latency in the hemisphere contralateral to the stimulated ear. When different spoken narratives are presented to each ear simultaneously (dichotic listening), the resulting cortical neural activity precisely encodes the acoustic features of both of the spoken narratives, but slightly weakened and delayed compared with the monaural response. Critically, the early sensory response to the attended speech is considerably stronger than that to the unattended speech, demonstrating top-down attentional gain control. This attentional gain is substantial even during the subjects' very first exposure to the speech mixture and therefore largely independent of knowledge of the speech content. Together, these findings characterize how the spectrotemporal features of speech are encoded in human auditory cortex and establish a single-trial-based paradigm to study the neural basis underlying the cocktail party phenomenon.

Abstract:Human listeners can reliably recognize speech in complex listening environments. The underlying neural mechanisms, however, remain unclear and cannot yet be emulated by any artificial system. In this dissertation, we study how speech is represented in the human auditory cortex and how the neural representation contributes to reliable speech recognition. Cortical activity from normal hearing human subjects is noninvasively recorded using magnetoencephalography, during natural speech listening. It is first demonstrated that neural activity from auditory cortex is precisely synchronized to the slow temporal modulations of speech, when the speech signal is presented in a quiet listening environment. How this neural representation is affected by acoustic interference is then investigated. Acoustic interference degrades speech perception via two mechanisms, informational masking and energetic masking, which are addressed respectively by using a competing speech stream and a stationary noise as the interfering sound. When two speech streams are presented simultaneously, cortical activity is predominantly synchronized to the speech stream the listener attends to, even if the unattended, competing speech stream is 8 dB more intense. When speech is presented together with spectrally matched stationary noise, cortical activity remains precisely synchronized to the temporal modulations of speech until the noise is 9 dB more intense. Critically, the accuracy of neural synchronization to speech predicts how well individual listeners can understand speech in noise. Further analysis reveals that two neural sources contribute to speech synchronized cortical activity, one with a shorter response latency of about 50 ms and the other with a longer response latency of about 100 ms. The longer-latency component, but not the shorter-latency component, shows selectivity to the attended speech and invariance to background noise, indicating a transition from encoding the acoustic scene to encoding the behaviorally important auditory object, in auditory cortex. Taken together, we have demonstrated that during natural speech comprehension, neural activity in the human auditory cortex is precisely synchronized to the slow temporal modulations of speech. This neural synchronization is robust to acoustic interference, whether speech or noise, and therefore provides a strong candidate for the neural basis of acoustic background invariant speech recognition.
Adaptive Temporal Encoding Leads to a Background-Insensitive Cortical Representation of Speech

Abstract:Speech recognition is remarkably robust to the listening background, even when the energy of background sounds strongly overlaps with that of speech. How the brain transforms the corrupted acoustic signal into a reliable neural representation suitable for speech recognition, however, remains elusive. Here, we hypothesize that this transformation is performed at the level of auditory cortex through adaptive neural encoding, and we test the hypothesis by recording, using MEG, the neural responses of human subjects listening to a narrated story. Spectrally matched stationary noise, which has maximal acoustic overlap with the speech, is mixed in at various intensity levels. Despite the severe acoustic interference caused by this noise, it is here demonstrated that low-frequency auditory cortical activity is reliably synchronized to the slow temporal modulations of speech, even when the noise is twice as strong as the speech. Such a reliable neural representation is maintained by intensity contrast gain control and by adaptive processing of temporal modulations at different time scales, corresponding to the neural δ and θ bands. Critically, the precision of this neural synchronization predicts how well a listener can recognize speech in noise, indicating that the precision of the auditory cortical representation limits the performance of speech recognition in noise. Together, these results suggest that, in a complex listening environment, auditory cortex can selectively encode a speech stream in a background insensitive manner, and this stable neural representation of speech provides a plausible basis for background-invariant recognition of speech.
Robust cortical encoding of slow temporal modulations of speech.

Abstract:This study investigates the neural representation of speech in complex listening environments. Subjects listened to a narrated story, masked by either another speech stream or by stationary noise. Neural recordings were made using magnetoencephalography (MEG), which can measure cortical activity synchronized to the temporal envelope of speech. When two speech streams are presented simultaneously, cortical activity is predominantly synchronized to the speech stream the listener attends to, even if the unattended, competing-speech stream is more intense (up to 8 dB). When speech is presented together with spectrally matched stationary noise, cortical activity remains precisely synchronized to the temporal envelope of speech until the noise is 9 dB more intense. Critically, the precision of the neural synchronization to speech predicts subjectively rated speech intelligibility in noise. Further analysis reveals that it is longer-latency (similar to 100 ms) neural responses, but not shorter-latency (similar to 50 ms) neural responses, that show selectivity to the attended speech and invariance to background noise. This indicates a processing transition, from encoding the acoustic scene to encoding the behaviorally important auditory object, in auditory cortex. In sum, it is demonstrated that neural synchronization to the speech envelope is robust to acoustic interference, whether speech or noise, and therefore provides a strong candidate for the neural basis of acoustic-background invariant speech recognition.
Cortical Neural Coding of Speech in Simple and Complex Auditory Scenes.

Abstract:The neural representation of continuous speech in human auditory cortex was obtained noninvasively via magnetoencephalography. The neural response was recorded from human subjects listening to a spoken narrative, either in clean or in the presence of interfering speech. The cortical neural response to clean speech is demonstrated to precisely track the slow temporal modulations (<10 Hz) of speech in a broad spectral region between 400 Hz and 2 kHz. The neural code is sufficiently faithful to decode acoustic features of speech. To examine the robustness of, and the role of attention in, this neural code, another spoken narrative was presented simultaneously, either to a different ear (dichotically) or to the same ear (diotically), instructing the subjects to focus on only one of the two speech signals. The cortical representation of the attended speech is found to be substantially stronger than that of the unattended speech. This attentional effect is significant during the subjects’ first exposure to the spoken narratives. These results demonstrate that auditory cortex precisely represents the slow temporal modulations of speech and maintains separate neural representations for concurrent speech signals that can be individually and strongly modulated by attention. [The Neural Encoding of Continuous Speech - Recent Advances in EEG and MEG Studies].

Abstract:Speech comprehension is a central cognitive function of the human brain. In cognitive neuroscience, a fundamental question is to understand how neural activity encodes the acoustic properties of a continuous speech stream and resolves multiple levels of linguistic structures at the same time. This paper reviews the recently developed research paradigms that employ electroencephalography (EEG) or magnetoencephalography (MEG) to capture neural tracking of acoustic features or linguistic structures of continuous speech. This review focuses on two questions in speech processing: (1) The encoding of continuously changing acoustic properties of speech; (2) The representation of hierarchical linguistic units, including syllables, words, phrases and sentences. Studies have found that the low-frequency cortical activity tracks the speech envelope. In addition, the cortical activities on different time scales track multiple levels of linguistic units and constitute a representation of hierarchically organized linguistic units. The article reviewed these studies, which provided new insights into the processes of continuous speech in the human brain.
A hierarchy of processing complexity and timescales for natural sounds in human auditory cortex

Abstract:Efficient behavior is supported by humans' ability to rapidly recognize acoustically distinct sounds as members of a common category. Within auditory cortex, there are critical unanswered questions regarding the organization and dynamics of sound categorization. Here, we performed intracerebral recordings in the context of epilepsy surgery as 20 patient-participants listened to natural sounds. We built encoding models to predict neural responses using features of these sounds extracted from different layers within a sound-categorization deep neural network (DNN). This approach yielded highly accurate models of neural responses throughout auditory cortex. The complexity of a cortical site's representation (measured by the depth of the DNN layer that produced the best model) was closely related to its anatomical location, with shallow, middle, and deep layers of the DNN associated with core (primary auditory cortex), lateral belt, and parabelt regions, respectively. Smoothly varying gradients of representational complexity also existed within these regions, with complexity increasing along a posteromedial-to-anterolateral direction in core and lateral belt, and along posterior-to-anterior and dorsal-to-ventral dimensions in parabelt. When we estimated the time window over which each recording site integrates information, we found shorter integration windows in core relative to lateral belt and parabelt. Lastly, we found a relationship between the length of the integration window and the complexity of information processing within core (but not lateral belt or parabelt). These findings suggest hierarchies of timescales and processing complexity, and their interrelationship, represent a functional organizational principle of the auditory stream that underlies our perception of complex, abstract auditory information.

Naturalistic Audiovisual Stimulation Reveals the Topographic Organization of Human Auditory Cortex

Abstract:Abstract Despite the importance of audition in spatial, semantic, and social function, there is no consensus regarding the detailed organisation of human auditory cortex. Using a novel computational model to analyse a high-powered naturalistic audiovisual movie-watching dataset, we simultaneously estimate spectral tuning properties and category selectivity to reveal the modes of organisation and computational motifs that characterise human auditory cortex. We find that regions more remote from the auditory core exhibit more compressive, non-linear response properties and finely-tuned, speech-selective receptive fields in low frequency portions of the tonotopic map. These patterns of organisation mirror aspects of the visual cortical hierarchy, wherein tuning properties progress from a stimulus category-agnostic ‘front end’ towards more advanced regions increasingly optimised for behaviorally relevant stimulus categories.
Decoding of selective attention to continuous speech from the human auditory brainstem response

Abstract:Abstract Humans are highly skilled at analysing complex acoustic scenes. The segregation of different acoustic streams and the formation of corresponding neural representations is mostly attributed to the auditory cortex. Decoding of selective attention from neuroimaging has therefore focussed on cortical responses to sound. However, the auditory brainstem response to speech is modulated by selective attention as well, as recently shown through measuring the brainstem’s response to running speech. Although the response of the auditory brainstem has a smaller magnitude than that of the auditory cortex, it occurs at much higher frequencies and therefore has a higher information rate. Here we develop statistical models for extracting the brainstem response from multi-channel scalp recordings and for analysing the attentional modulation according to the focus of attention. We demonstrate that the attentional modulation of the brainstem response to speech can be employed to decode the attentional focus of a listener from short measurements of ten seconds or less in duration. The decoding remains accurate when obtained from three EEG channels only. We further show how out-of-the-box decoding that employs subject-independent models, as well as decoding that is independent of the specific attended speaker is capable of achieving similar accuracy. These results open up new avenues for investigating the neural mechanisms for selective attention in the brainstem and for developing efficient auditory brain-computer interfaces.
Neural Dynamics of the Processing of Speech Features: Evidence for a Progression of Features from Acoustic to Sentential Processing

Abstract:When we listen to speech, our brain’s neurophysiological responses “track” its acoustic features, but it is less well understood how these auditory responses are modulated by linguistic content. Here, we recorded magnetoencephalography (MEG) responses while subjects listened to four types of continuous-speech-like passages: speech-envelope modulated noise, English-like non-words, scrambled words, and narrative passage. Temporal response function (TRF) analysis provides strong neural evidence for the emergent features of speech processing in cortex, from acoustics to higher-level linguistics, as incremental steps in neural speech processing. Critically, we show a stepwise hierarchical progression of progressively higher order features over time, reflected in both bottom-up (early) and top-down (late) processing stages. Linguistically driven top-down mechanisms take the form of late N400-like responses, suggesting a central role of predictive coding mechanisms at multiple levels. As expected, the neural processing of lower-level acoustic feature responses is bilateral or right lateralized, with left lateralization emerging only for lexical-semantic features. Finally, our results identify potential neural markers of the computations underlying speech perception and comprehension.

Abstract:A core goal of auditory neuroscience is to build quantitative models that predict cortical responses to natural sounds. Reasoning that a complete model of auditory cortex must solve ecologically relevant tasks, we optimized hierarchical neural networks for speech and music recognition. The best-performing network contained separate music and speech pathways following early shared processing, potentially replicating human cortical organization. The network performed both tasks as well as humans and exhibited human-like errors despite not being optimized to do so, suggesting common constraints on network and human performance. The network predicted fMRI voxel responses substantially better than traditional spectrotemporal filter models throughout auditory cortex. It also provided a quantitative signature of cortical representational hierarchy-primary and non-primary responses were best predicted by intermediate and late network layers, respectively. The results suggest that task optimization provides a powerful set of tools for modeling sensory systems.

Abstract:Perception of sounds and speech involves structures in the auditory brainstem that rapidly process ongoing auditory stimuli. The role of these structures in speech processing can be investigated by measuring their electrical activity using scalp-mounted electrodes. However, typical analysis methods involve averaging neural responses to many short repetitive stimuli that bear little relevance to daily listening environments. Recently, subcortical responses to more ecologically relevant continuous speech were detected using linear encoding models. These methods estimate the temporal response function (TRF), which is a regression model that minimises the error between the measured neural signal and a predictor derived from the stimulus. Using predictors that model the highly non-linear peripheral auditory system may improve linear TRF estimation accuracy and peak detection. Here, we compare predictors from both simple and complex peripheral auditory models for estimating brainstem TRFs on electroencephalography (EEG) data from 24 participants listening to continuous speech. We also investigate the data length required for estimating subcortical TRFs, and find that around 12 minutes of data is sufficient for clear wave V peaks (>3 dB SNR) to be seen in nearly all participants. Interestingly, predictors derived from simple filterbank-based models of the peripheral auditory system yield TRF wave V peak SNRs that are not significantly different from those estimated using a complex model of the auditory nerve, provided that the nonlinear effects of adaptation in the auditory system are appropriately modelled. Crucially, computing predictors from these simpler models is more than 50 times faster compared to the complex model. This work paves the way for efficient modelling and detection of subcortical processing of continuous speech, which may lead to improved diagnosis metrics for hearing impairment and assistive hearing technology.

The neural dynamics of auditory word recognition and integration

Abstract:Listeners recognize and integrate words in rapid and noisy everyday speech by combining expectations about upcoming content with incremental sensory evidence. We present a computational model of word recognition which formalizes this perceptual process in Bayesian decision theory. We fit this model to explain scalp EEG signals recorded as subjects passively listened to a fictional story, revealing both the dynamics of the online auditory word recognition process and the neural correlates of the recognition and integration of words. The model reveals distinct neural processing of words depending on whether or not they can be quickly recognized. While all words trigger a neural response characteristic of probabilistic integration -- voltage modulations predicted by a word's surprisal in context -- these modulations are amplified for words which require more than roughly 150 ms of input to be recognized. We observe no difference in the latency of these neural responses according to words' recognition times. Our results are consistent with a two-part model of speech comprehension, combining an eager and rapid process of word recognition with a temporally independent process of word integration. However, we also developed alternative models of the scalp EEG signal not incorporating word recognition dynamics which showed similar performance improvements. We discuss potential future modeling steps which may help to separate these hypotheses.

Abstract:Human listeners have the ability to direct their attention to a single speaker in a multi-talker environment. The neural correlates of selective attention can be decoded from a single trial of electroencephalography (EEG) data. In this study, leveraging the source-reconstructed and anatomically-resolved EEG data as inputs, we sought to employ CNN as an interpretable model to uncover task-specific interactions between brain regions, rather than simply to utilize it as a black box decoder. To this end, our CNN model was specifically designed to learn pairwise interaction representations for 10 cortical regions from five-second inputs. By exclusively utilizing these features for decoding, our model was able to attain a median accuracy of 77.56% for within-participant and 65.14% for cross-participant classification. Through ablation analysis together with dissecting the features of the models and applying cluster analysis, we were able to discern the presence of alpha-band-dominated inter-hemisphere interactions, as well as alpha- and beta-band dominant interactions that were either hemisphere-specific or were characterized by a contrasting pattern between the right and left hemispheres. These interactions were more pronounced in parietal and central regions for within-participant decoding, but in parietal, central, and partly frontal regions for cross-participant decoding. These findings demonstrate that our CNN model can effectively utilize features known to be important in auditory attention tasks and suggest that the application of domain knowledge inspired CNNs on source-reconstructed EEG data can offer a novel computational framework for studying task-relevant brain interactions.
Sound reconstruction from human brain activity via a generative model with brain-like auditory features

Abstract:The successful reconstruction of perceptual experiences from human brain activity has provided insights into the neural representations of sensory experiences. However, reconstructing arbitrary sounds has been avoided due to the complexity of temporal sequences in sounds and the limited resolution of neuroimaging modalities. To overcome these challenges, leveraging the hierarchical nature of brain auditory processing could provide a path toward reconstructing arbitrary sounds. Previous studies have indicated a hierarchical homology between the human auditory system and deep neural network (DNN) models. Furthermore, advancements in audio-generative models enable to transform compressed representations back into high-resolution sounds. In this study, we introduce a novel sound reconstruction method that combines brain decoding of auditory features with an audio-generative model. Using fMRI responses to natural sounds, we found that the hierarchical sound features of a DNN model could be better decoded than spectrotemporal features. We then reconstructed the sound using an audio transformer that disentangled compressed temporal information in the decoded DNN features. Our method shows unconstrained sounds reconstruction capturing sound perceptual contents and quality and generalizability by reconstructing sound categories not included in the training dataset. Reconstructions from different auditory regions remain similar to actual sounds, highlighting the distributed nature of auditory representations. To see whether the reconstructions mirrored actual subjective perceptual experiences, we performed an experiment involving selective auditory attention to one of overlapping sounds. The results tended to resemble the attended sound than the unattended. These findings demonstrate that our proposed model provides a means to externalize experienced auditory contents from human brain activity.

Neural Processing of Naturalistic Audiovisual Events in Space and Time

Abstract:Our brain seamlessly integrates distinct sensory information to form a coherent percept. However, when real-world audiovisual events are perceived, the specific brain regions and timings for processing different levels of information remain less investigated. To address that, we curated naturalistic videos and recorded fMRI and EEG data when participants viewed videos with accompanying sounds. Our findings reveal early asymmetrical cross-modal interaction, with acoustic information represented in both early visual and auditory regions, while visual information only identified in visual cortices. The visual and auditory features were processed with similar onset but different temporal dynamics. High-level categorical and semantic information emerged in multi-modal association areas later in time, indicating late cross-modal integration and its distinct role in converging conceptual information. Comparing neural representations to a two-branch deep neural network model highlighted the necessity of early fusion to build a biologically plausible model of audiovisual perception. With EEG-fMRI fusion, we provided a spatiotemporally resolved account of neural activity during the processing of naturalistic audiovisual stimuli.

Abstract:The ability to recognize abstract features of voice during auditory perception is an intricate feat of human audition. For the listener, this occurs in near-automatic fashion to seamlessly extract complex cues from a highly variable auditory signal. Voice perception depends on specialized regions of auditory cortex, including superior temporal gyrus (STG) and superior temporal sulcus (STS). However, the nature of voice encoding at the cortical level remains poorly understood. We leverage intracerebral recordings across human auditory cortex during presentation of voice and nonvoice acoustic stimuli to examine voice encoding at the cortical level in 8 patient-participants undergoing epilepsy surgery evaluation. We show that voice selectivity increases along the auditory hierarchy from supratemporal plane (STP) to the STG and STS. Results show accurate decoding of vocalizations from human auditory cortical activity even in the complete absence of linguistic content. These findings show an early, less-selective temporal window of neural activity in the STG and STS followed by a sustained, strongly voice-selective window. Encoding models demonstrate divergence in the encoding of acoustic features along the auditory hierarchy, wherein STG/STS responses are best explained by voice category and acoustics, as opposed to acoustic features of voice stimuli alone. This is in contrast to neural activity recorded from STP, in which responses were accounted for by acoustic features. These findings support a model of voice perception that engages categorical encoding mechanisms within STG and STS to facilitate feature extraction.

Abstract:Processing auditory sequences involves multiple brain networks and is crucial to complex perception associated with music appreciation and speech comprehension. We used time-resolved cortical imaging in a pitch change detection task to detail the underlying nature of human brain network activity, at the rapid time scales of neurophysiology. In response to tone sequence presentation to the participants, we observed slow inter-regional signaling at the pace of tone presentations (2-4 Hz) that was directed from auditory cortex toward both inferior frontal and motor cortices. Symmetrically, motor cortex manifested directed influence onto auditory and inferior frontal cortices via bursts of faster (15-35 Hz) activity. These bursts occurred precisely at the expected latencies of each tone in a sequence. This expression of interdependency between slow/fast neurophysiological activity yielded a form of local cross-frequency phase-amplitude coupling in auditory cortex, which strength varied dynamically and peaked when pitch changes were anticipated. We clarified the mechanistic relevance of these observations in relation to behavior by including a group of individuals afflicted by congenital amusia, as a model of altered function in processing sound sequences. In amusia, we found a depression of inter-regional slow signaling toward motor and inferior frontal cortices, and a chronic overexpression of slow/fast phase-amplitude coupling in auditory cortex. These observations are compatible with a misalignment between the respective neurophysiological mechanisms of stimulus encoding and internal predictive signaling, which was absent in controls. In summary, our study provides a functional and mechanistic account of neurophysiological activity for predictive, sequential timing of auditory inputs.SIGNIFICANCE STATEMENT Auditory sequences are processed by extensive brain networks, involving multiple systems. In particular, fronto-temporal brain connections participate in the encoding of sequential auditory events, but so far, their study was limited to static depictions. This study details the nature of oscillatory brain activity involved in these inter-regional interactions in human participants. It demonstrates how directed, polyrhythmic oscillatory interactions between auditory and motor cortical regions provide a functional account for predictive timing of incoming items in an auditory sequence. In addition, we show the functional relevance of these observations in relation to behavior, with data from both normal hearing participants and a rare cohort of individuals afflicted by congenital amusia, which we considered here as a model of altered function in processing sound sequences.

Abstract:The patient had complete placenta previa and a giant placental tumor, and had a history of repair for cor triatriatum (CorT) 19 years previously. After anesthetic induction with remifentanil, thiamylal, and rocuronium, tachyarrhythmia occurred following multifocal ventricular premature contraction. Treatment with lidocaine was not effective. However, tachyarrhythmia converted naturally after delivery and removal of the placental tumor. We speculated that the surgical scar remaining after CorT repair acted as an arrhythmogenic substrate, and that these factors associated with hemodynamic change and hypomagnesaemia in this patient, including sympathetic instability of pregnancy, giant placental tumor, and general anesthesia, resulted in unexpected arrhythmia. In conclusion, in cases of caesarian section in grown-up congenital heart pregnant patients with complete placenta previa and giant placental tumor, careful consideration should be given to alterations in the hemodynamic condition.

Abstract:Natural sounds, including vocal communication sounds, contain critical information at multiple time scales. Two essential temporal modulation rates in speech have been argued to be in the low gamma band (similar to 20-80 ms duration information) and the theta band (similar to 150-300 ms), corresponding to segmental and diphonic versus syllabic modulation rates, respectively. It has been hypothesized that auditory cortex implements temporal integration using time constants closely related to these values. The neural correlates of a proposed dual temporal window mechanism in human auditory cortex remain poorly understood. We recorded MEG responses from participants listening to non-speech auditory stimuli with different temporal structures, created by concatenating frequency-modulated segments of varied segment durations. We show that such non-speech stimuli with temporal structure matching speech-relevant scales (similar to 25 and similar to 200 ms) elicit reliable phase tracking in the corresponding associated oscillatory frequencies (low gamma and theta bands). In contrast, stimuli with non-matching temporal structure do not. Furthermore, the topography of theta band phase tracking shows rightward lateralization while gamma band phase tracking occurs bilaterally. The results support the hypothesis that there exists multi-time resolution processing in cortex on discontinuous scales and provide evidence for an asymmetric organization of temporal analysis (asymmetrical sampling in time, AST). The data argue for a mesoscopic-level neural mechanism underlying multi-time resolution processing: the sliding and resetting of intrinsic temporal windows on privileged time scales.
